本发明涉及一种对食品中细菌进行检测的检测装置,特别涉及一种快速检测食品中产志贺毒素大肠杆菌的试剂盒。主要解决现有检测方法存在的检测时间过长的技术问题,一种快速检测食品中产志贺毒素大肠杆菌的试剂盒,由以下试剂组成:试剂A:DNA提取液;试剂B:PCR反应液,其中含有特定四对引物stx1-F和stx1-R、stx2-F和stx2-R、eaeA-F和eaeA-R、hlyA-F和hlyA-R,其序列分别为:5’ACACTGGATGATCTCAGTGG 3’和5’CTGAATCCCCCTCCATTATG 3’;5’CCATGACAACGGACAGCAGTT3’和5’CCTGTCAACTGAGCACTTTG 3’;5’AAGCGACTGAGGTCACT3’和5’ACGCTGCTCACTAGATGT 3’;5’CACACGGAGCTTATAATATTCTGTCA和AATGTTATCCCATTGACATCATTTGACT 3’;试剂C:STEC分型血清O157、O26、O111、O103和O145。用于对样品中STEC检测。

Description

产志贺毒素大肠杆菌核酸纯化血清鉴定试剂盒
技术领域:本发明涉及一种对食品中细菌进行检测的检测装置,特别涉及一种快速检测食品中产志贺毒素大肠杆菌的试剂盒。
背景技术:产志贺毒素大肠杆菌(Shiga Toxin-producing Escherichiacoli,简称STEC)为食源性致病菌,革兰氏阴性,有周鞭毛,并有菌毛,不产生芽孢。STEC是世界上人类食品中毒的重要的致病因子,也是引起大规模食源性食物中毒的主要病原菌之一。STEC中有100余个血清型的致病性大肠杆菌可以致病,其中0157:H7是主要的血清型。可引起非出血性腹泻、出血性结肠炎(haemorrhagic colitis,HC)、溶血性尿毒综合症(haemolytic uraemicsyndrome,HUS)、血栓性血小板减少性紫癜(thrombotic throbocytopenicpurpura,TTP)。而非0157的STEC主要的血清型有026、0111、0103和0145也严重威胁着人类的健康。
近年来STEC引起的大规模食源性食物中毒越来越受到高度重视,而且非0157:H7 STEC引起的HC、HUC在逐年增加。由于0157:H7不发酵山梨醇比非0157:H7 STEC容易鉴定,所以非0157:H7常被人们所忽视。
STEC的毒力是由多因子决定的。其致病的特征性毒力因子是I型志贺毒素(shiga toxin type1,stx1)和2型志贺毒素(shiga toxin type2,stx2),分别由stx1基因和stx2基因编码。其他毒力因子如由eaeA基因编码的粘附因子(attac hment and effacement factor);由hlyA基因编码的溶血素(hemolysin)等也是重要的毒力因素。采用多重PCR技术对样品进行STEC的毒力基因鉴定,不仅可作特征性鉴定,而且还可对其潜在的致病性作出前瞻性判断,可快速检测出样品中STEC。
近年来,国内外的研究人员检测食品中STEC研究的较多,包括李咏梅等采用的PCR方法、Paton AW等采用的PCR方法。上述鉴定方法包括营养培养、菌落选择、生化反应特性和志贺毒素的鉴定,需要几天的时间才能完成STEC的鉴定。还没有解决快速、准确检出该菌的问题。
发明内容:本发明的目的是提供一种快速检测样品中STEC的试剂盒和使用方法,主要解决现有检测方法存在的检测时间过长的技术问题,采用复合PCR可以快速、特异地筛选标本中的STEC,以便可以快速、灵敏、准确地检出食品中STEC,该试剂盒适合于各种样品的检测。
本发明的试剂盒主要由以下几种试剂配方组成:
试剂A:DNA提取液;
试剂B:PCR反应液,其中含有特定四对引物stx1-F和stx1-R、stx2-F和stx2-R、eaeA-F和eaeA-R、hlyA-F和hlyA-R,其序列分别为:5’ACACTGGATGATCTCAGTGG3’和5’CTGAATCCCCCTCCATTATG3’;5’CCATGACAACGGACAGCAGTT3’和5’CCTGTCAACTGAGCACTTTG3’;5’AAGCGACTGAGGTCACT3’和5’ACGCTGCTCACTAGATGT3’;5’CACACGGAGCTTATAATATTCTGTCA和AATGTTATCCCATTGACATCATTTGACT3’
试剂B中所含的特定引物stx1-F和stx1-R、stx2-F和stx2-R、eaeA-F和eaeA-R、hlyA-F和hlyA-R,每种含量应不少于5pm。
试剂C:STEC分型血清0157、026、0111、0103和0145。
本发明所提供的快速检测样品中STEC的试剂盒的使用方法如下:
一、样品中DNA的提取
1.取1mL样品增菌液,12000g离心1分钟,弃尽上清液;
2.加入20μL溶菌酶,室温放置5分钟。
3.加入SDS(十二烷基硫酸钠)12μL,4μL蛋白酶K,37℃水浴30分钟。裂解过程中上下混匀几次,降解完全的样品应为半透明粘稠液体,否则应继续保温处理。
4.加入等体积苯酚:氯仿:异戊醇=25:24:1236μL;混匀10分钟,13800g离心5分钟。
5.弃去有机层,加入等体积的氯仿:异戊醇=24:1,轻轻混匀,弃去有机层。
6.加入1/10体积3mol/L醋酸钠,2倍体积的冷无水乙醇(—20℃隔夜),离心沉淀DNA。
7.600μL 70%乙醇洗一次,倒置于干净的滤纸上,室温干燥。
8.待检样品DNA用50μL milliQ水溶解。
二、PCR反应
取10μL待检样品DNA加入反应管B中,混匀后在PCR仪中进行以下反应:

4℃   停止
反应结束后,取20μL PCR产物,加2μL溴酚蓝混匀后进行确定扩增产物。如出现614bp和/或779bp的反应产物,说明待检样品中有产毒的STEC。
三、鉴定:用STEC分型血清对二中产毒的STEC进行血清凝集。
本发明的有益效果是,使用本发明的试剂盒,采用复合PCR可以快速、特异地筛选标本中的STEC,并能在24小时内快速、灵敏、准确地检出样品中的STEC,灵敏度可达15CFU/mL。
具体实施方式:
猪肉糜和果汁用快速检测样品中STEC的试剂盒检测。试剂盒组成为:
试剂A:DNA提取液。其组成为20%SDS,苯酚:氯仿:异戊醇=25:24:1;氯仿:异戊醇=24:1;
试剂B:PCR反应液,每管STEC的50μL扩增反应体系组成中含5μL0.025mol l-1MgCl2;5pmol 4对引物stx1-F和stx1-R、stx2-F和stx2-R、eaeA-F和eaeA-R、hlyA-F和hlyA-R各为2μL;PCR buffre 5μL;200μmol dNTP 1μL;Taq酶1.0μL;DNA 5μL(终浓度为2ng/ml);ddH2O 22.0μL;
试剂C:STEC分型血清0157、026、0111、0103和0145。
按本发明所提供的快速检测样品中STEC试剂盒的使用方法,先用试剂A提取样品中的DNA,之后进行PCR反应,反应结束后取20μL PCR反应产物加2μL溴酚蓝混匀后进行2%琼脂糖凝胶电泳,即可发现样品中是否含有产毒的STEC,最后用STEC分型血清进行鉴定。
本发明所提供的快速检测样品中STEC试剂盒特异性强,灵敏度高,能准确检测出食品中的产志贺毒素大肠杆菌,灵敏度达到15CFU/mL。

1.一种产志贺毒素大肠杆菌核酸纯化血清鉴定试剂盒,其特征在于该试剂盒由以下试剂组成:
试剂A:DNA提取液;
试剂B:PCR反应液,其中含有特定四对引物stxl-F和stx1-R、stx2-F和stx2-R、eaeA-F和eaeA-R、hlyA-F和hlyA-R,其序列分别为:5’ACACTGGATGATCTCAGTGG3’和5’CTGAATCCCCCTCCATTATG3’;5’CCATGACAACGGACAGCAGTT3’和5’CCTGTCAACTGAGCACTTTG3’;5’AAGCGACTGAGGTCACT3’和5’ACGCTGCTCACTAGATGT3’;5’CACACGGAGCTTATAATATTCTGTCA和AATGTTATCCCATTGACATCATTTGACT3’;
试剂C:STEC分型血清0157、026、0111、0103和0145。
2.根据权利要求1所述的产志贺毒素大肠杆菌核酸纯化血清鉴定试剂盒,其特征在于所说的试剂B:PCR反应液,每管组成为每管STEC的50μL扩增反应体系组成中含5μL0.025mol 1-1MgCl2;5pmol4对引物stx1-F和stx1-R、stx2-F和stx2-R、eaeA-F和eaeA-R、hlyA-F和hlyA-R各为2μL;PCR buffre5μL;200μmol dNTP1μL;Taq酶1.0μL;DNA5μL;ddH2O22.0μL。
