Abstract
本发明公开了一种制备软骨细胞的方法,包括步骤:(a)在适合生长的条件下,将成纤维细胞在含软骨源性形态发生蛋白的成纤维细胞培养液中培养3-30天,从而使成纤维细胞诱导形成软骨细胞;(b)从培养物中分离出软骨细胞。实验研究证明成纤维细胞在CDMP1生长因子诱导下能向成软骨细胞表型分化,并能分泌软骨细胞特异性基质,因而可成为软骨组织工程的种子细胞来源。

技术领域
本发明涉及生物学和组织工程学领域,更具体地涉及诱导成纤维细胞形成软骨细胞的方法。
背景技术
软骨组织作为一种***主要起支撑、保护、分散应力等作用。组织学上可分为透明软骨(关节软骨、肋软骨、鼻软骨、气管软骨)、弹性软骨(会厌软骨、耳软骨)、纤维软骨(半月板)三类。它们在组织学上都是只含有软骨细胞的单一组织,内部无血管,营养主要依靠组织液的渗透。
从结构上看,软骨细胞被包裹在软骨陷窝内,其周围厚厚的软骨囊作为一道天然屏障,将软骨细胞与周围组织分离开来。因此,软骨组织具有抗原性弱,不易被机体免疫***识别和攻击而具有较轻免疫排斥反应的特点。早期的研究结果表明,同种异体软骨细胞可以在受体体内存活并保持分泌基质的功能。
软骨病损是临床常见疾病之一,如由创伤、感染、自身免疫、肿瘤等因素引起的各种关节表面疾患;耳廓软骨、鼻、喉软骨支架的缺损、畸形;以及儿童骨骺生长板的创伤、早闭、坏死等。由于软骨组织几乎没有自我修复能力,所以必须利用软骨或其它替代材料进行修复。
自体和异体软骨移植是目前的常用方法。自体软骨组织移植疗效可靠,但必须以牺牲人体正常组织为代价,且患者要多遭受一次手术的痛苦。此外,自体组织来源有限,小块移植物还有易变形的问题。异体和异种移植物又具有免疫原性。
目前,虽然有一些人工合成的软骨组织代用品,还存在着组织相容性、材料机械特性等方面的缺陷,尚不能满足临床上的各项要求。人工合成替代材料只具备充填、支持及维持美观的作用,无生物学功能,尚不能达到真正意义上的结构与功能重建。
组织工程学为软骨组织缺损的治疗开辟了新的途径。组织工程用少量细胞在体外扩增,然后接种到生物可降解材料上,构建成细胞-材料复合物,用于移植,修复缺损。
构建组织工程化软骨组织的重要条件之一是必须获得大量的有功能和活力的软骨细胞。在高等大型哺乳动物甚至人体内也可以利用组织工程技术再生软骨组织,但这些研究所应用的种子细胞均为软骨细胞,且只能用原代或早期传代的软骨细胞,并且体外大量扩增后的软骨细胞易发生老化及去分化,难以在体内形成软骨组织。自体软骨细胞来源极为有限,同种异体及异种软骨细胞又无法杜绝免疫排斥反应,生长因子虽可大量扩增软骨细胞,但其费用昂贵,所形成组织在体内的长期效果尚不肯定,这些都使软骨组织工程难以在临床推广应用。
软骨组织工程种子细胞应具备来源广泛、取材方便并对机体创伤小、体外增殖能力强不易老化等特点。而目前主要取自自体软骨组织的软骨种子细胞来源非常有限,终末分化的软骨细胞体外培养过程中极易去分化,扩增数量不能满足组织构建的需要(VonderMark K,Gauss V,Vonder Mark H,et al.Relationship between cell shape and type of collagen synthesized aschondrocytes lose their cartilage phenotype in culture.Nature,1997,267:531-532)。结合发育生物学研究,体内分布广泛的真皮成纤维细胞与软骨细胞都来源于中胚层***(Mesenchymal cells),与软骨细胞不同的是成纤维细胞保留了中胚层***增殖能力强的生物学特性,多次传代仍保持较好的增殖能力。研究表明,成纤维细胞在特定的体外环境中可表达特异的骨基质(Inaba K,Matsunaga S,Ishidou Y,et al.Effect oftransforming growth factor-beta on fibroblasts in ossification of theposterior longitudinal ligament.In Vivo.1996 10:445-449.)。然而到目前为止,尚没有以成纤维细胞系为种子细胞制得软骨细胞的报道。
综上所述,种子细胞来源不足是限制软骨组织工程临床推广应用的主要困难,因此本领域迫切需要开发新的制备软骨的种子细胞,以及用其制备的软骨细胞和软骨移植物。
发明内容
本发明的目的就是提供一种来源广泛的软骨移植物的种子细胞,以及用该种子细胞制备软骨细胞和移植物的方法。
在本发明的第一方面,提供了一种制备软骨细胞的方法,包括步骤:
(a)在适合生长的条件下,将成纤维细胞在含10-1000纳克/毫升软骨源性形态发生蛋白(尤其是CDMP-1)的成纤维细胞培养液中培养3-30天,从而使成纤维细胞诱导形成软骨细胞;
(b)从培养物中分离出软骨细胞。
在另一优选例中,所述的生长条件是37±2℃,CO2 5±3%。
在另一优选例中,所述的培养液是含0-20%(更佳地3-15%)胎牛血清的F-12,DMEM或1640培养液。
在另一优选例中,所述的成纤维细胞是人的成纤维细胞。
在另一优选例中,步骤(a)中所述的成纤维细胞的密度为0.5×104-5×105个/ml。
在另一优选例中,所述的成纤维细胞是原代的或传代的成纤维细胞。
在本发明的第二方面,提供了用本发明所述的方法制备的软骨细胞。
在本发明的第三方面,提供了一种组织工程化软骨移植物,它包括:
(a)药学上可接受的生物可降解材料;
(b)用本发明所述方法制备的软骨细胞,其含量为1×105-5×107个细胞/毫升,或1×105-5×107个细胞/cm3。
在另一优选例中,所述的药学上可接受的生物可降解材料选自下组:聚乳酸、聚羟基乙酸、聚乳酸-聚羟基乙酸共聚物,Pluronics、聚羟基丁酸、聚酸酐、聚偶磷氮、聚氨基酸、假聚氨基酸、聚原酸酯、聚酯尿烷、聚碳酸酯、聚乙二醇、聚环氧乙烷、聚对二氧六环酮、胶原、明胶、糖氨聚糖、壳聚糖、甲壳素、海藻酸盐、藻酸钙凝胶、脱细胞基质,及其混合物。
在本发明的第四方面,提供了一种成纤维细胞的用途,它用于制备软骨细胞。
在本发明的第五方面,提供了一种制备组织工程化软骨移植物的方法,它包括步骤:将本发明所述方法制备的软骨细胞与药学上可接受的生物可降解材料混合,形成组织工程化软骨移植物。

具体实施方式
本发明人经过广泛而深入的研究发现,软骨源性形态发生蛋白(cartilage-derived morphorgentic protein,CDMP)特别适合作为诱导成纤维细胞分化形成软骨细胞的诱导因子,可以有效地诱导成纤维细胞在体外形成软骨细胞。在此基础上完成了本发明。
术语
术语“纯化的或分离的”指纯化的或分离的物质基本上不含有其他细胞、蛋白质或多肽,例如纯化的细胞因子,或分离的软骨细胞。
术语“异种移植”指将所需生物活体材料(如成纤维细胞)从某一物种中取出并再施用于另一物种对象的方法。
术语“自体移植”指将所需生物活体材料(如成纤维细胞)从某病人中取出并再施用于同一病人的方法。
术语“异体移植”指将所需生物活体材料(如软骨细胞)从某个体中取出并再施用于同物种另一不同个体的方法。
成纤维细胞
本发明中成纤维细胞的来源没有特别限制,一种优选的来源是来自自体的真皮。通常,本发明的成纤维细胞是来自自体或同种异体。成纤维细胞可以是原代的或传代的成纤维细胞。
分离和培养获得成纤维细胞的方法都是本领域中已知的。一种优选的方法是取皮肤真皮组织,剪碎移入离心管中,加入约10倍于组织体积的0.1%的I型胶原酶消化,37℃,2小时,100目细胞滤器过滤后离心,1000转5分钟,弃去上清,加入含10% FBS DMEM培养液混匀细胞,以2×104/cm2接种于100mm培养皿中培养,待细胞融合至80%-90%时,进行传代,传代密度为1×104/cm2。优选第2~18代细胞用于制备人工软骨。
一类优选的成纤维细胞是体外培养的第二代~第六代的成纤维细胞。此时的成纤维细胞有较强的增殖能力和向软骨细胞分化的潜能。
诱导因子
如本文所用,“诱导因子”指软骨源性形态发生蛋白(Cartilage-DerivedMorphogenetic Proteins,CDMPs)。这是BMP家族中一类特异生长因子,它在软骨发育过程中发挥重要的调控作用,其中软骨源性形态发生蛋白-1(CDMP1)和软骨源性形态发生蛋白-2(CDMP2)是优选的。更佳地是CDMP1。
如本文所用,“细胞诱导液”指含有10-1000纳克/毫升(较佳地20-500ng/ml,更佳地50-200ng/ml)软骨源性形态发生蛋白的成纤维细胞培养液。
除了CDMP之外,在本发明的方法中还可联用其他因子,例如转化生长因子β(transforming growth factor beta,TGF β)1、2、3、骨形态发生蛋白(bonemorphorgenetic protein,BMP)2、6、7。联用其他诱导因素如:低氧环境培养、在特定的软骨细胞外基质成分中培养、应用阻滞或激活特定的细胞信号传导通路、与软骨细胞共同培养等
诱导成纤维细胞形成软骨细胞
诱导步骤是本发明方法的关键。通常,在适合生长的条件下,将成纤维细胞在添加了10-1000纳克/毫升(较佳地20-500ng/ml,更佳地50-200ng/ml)软骨源性形态发生蛋白的成纤维细胞培养液中培养3-30天(较佳地5-15天),就可使成纤维细胞诱导形成软骨细胞。
一种优选的方法是单层培养细胞诱导,在含约10%FBS和约10-1000ng/ml的CDMP成纤维细胞培养基(如F-12培养基)中进行细胞诱导,每三天换一次培养液,诱导3-30天。
另一种优选方法是离心管培养细胞诱导。将成纤维细胞在约1500转离心,弃上清后加入细胞诱导液,将细胞团块摇起,悬浮于培养液中,诱导3-30天。
经过诱导培养后,可以通过Masson’s Trichrome,Alcian Blue与免疫组化染色,RT-PCR,Western印迹分析等进行定性和半定量鉴定,然后从培养物中分离出软骨细胞。
生物可降解材料
可用于本发明的组织工程化软骨的材料是医学上可接受的生物可降解材料,包括(但并不限于):
(a)可降解性合成高分子材料,例如聚α-羟基酸(如聚乳酸PLA、聚羟基乙酸PGA、聚羟基丁酸PHB等)、聚酸酐(polyanhydrides)、聚偶磷氮(polyphosphazenes)、聚氨基酸(polyamino acid)、假聚氨基酸(pesudo-polyamino acid)、聚原酸酯(polyorthoesters)、聚酯尿烷(polyesterurethane)、聚碳酸酯(polycarbonate)、聚乙二醇、聚对二氧六环酮(polydioxanone)等;
(b)天然可降解材料,例如胶原(collagen)、明胶(gelatin)、糖氨聚糖(glycosaminoglycan,GAGs)、壳聚糖(chitosan)、甲壳素(chitin)、海藻酸盐、藻酸钙凝胶等;各种脱细胞基质;
(c)可注射性材料,如Pluronic(一种市售的聚环氧乙丙烯商品)、藻酸钙凝胶等;
(d)上述材料的混合物或复合材料或共聚物,尤其是高分子材料与天然材料的复合材料,以及固体材料与可注射性材料的复合材料。共聚物:聚羟基乙酸—聚乳酸共聚物(lactic-co-glycolic acid,PLGA)
优选的医学上可接受的生物可降解材料是可注射性的材料,例如藻酸钙凝胶、Pluronic凝胶等。
本发明组织工程化软骨中的软骨细胞的细胞浓度通常约为2×107/ml至5×107/ml或更高。当材料为可注射性材料时,以该液体材料调整软骨细胞浓度;当材料为固体性材料时,以培养液调整软骨细胞浓度,然后与固体性材料混合,其中混合时培养液与固体性材料的比例没有特别限制,但是以该固体材料所能够吸附培养液的最大量为准。
在本发明的组织工程化软骨移植物中,还可添加或复合其他各种细胞因子、生长因子、各种转基因成分,从而保持软骨细胞表型、促进软骨细胞生长或基质合成能力等。
组织工程化软骨移植物
用本发明方法制备的软骨细胞可用于构建组织工程化软骨移植物,即将一定数量的所述软骨细胞与药学上可接受的生物可降解材料混合即可。
用本发明方法形成的组织工程化软骨移植物,分为两类,一类是软骨细胞与可注射性生物材料构成的复合物,该复合物可直接注射到皮下、软骨缺损处等体内各部位。另一类是软骨细胞与非可注射性生物材料构成的复合物,该复合物可直接植入体内皮下、软骨缺损处等体内各部位。
在一优选例中生物可降解材料为固态。此时,可用上述细胞与培养液制成浓度为约5×107/ml的细胞悬液,然后与生物材料聚羟基乙酸/聚乳酸支架(PGA/PLA)形成复合物,该复合物大小、形状依照软骨缺损的大小及形状确定。体外培养一周至三周,每二至三天换液一次,以保证细胞营养,当体外组织工程化软骨初步形成时植入体内皮下、软骨缺损处等体内各部位。
在另一优选例中,生物可降解材料是可注射性材料(Pluronic凝胶、藻酸钙凝胶等)。以该液体材料调整细胞浓度为约5×107/ml,直接用于皮下注射或体内缺损的修复。
应用方法
当生物可降解材料是可注射性材料时,细胞与生物材料复合物可直接植入皮下或体内软骨缺损处。当生物可降解材料为固态时,细胞与生物材料在体外复合,在生物反应器内形成适合各种软骨缺损大小和形状的组织工程化软骨后,再植入体内软骨缺损处。
本发明的主要优点在于:
(1)应用自体细胞,且一次取材既可获得足够的细胞量;
(2)成纤维细胞的采集操作简单,对病人损伤极小,无需住院,且可以重复取材;
(3)体外培养方法简单易学,便于推广应用。
下面结合具体实施例,进一步阐述本发明。应理解,这些实施例仅用于说明本发明而不用于限制本发明的范围。下列实施例中未注明具体条件的实验方法,通常按照常规条件,例如Sambrook等人,分子克隆:实验室手册(NewYork:Cold Spring Harbor Laboratory Press,1989)中所述的条件,或按照制造厂商所建议的条件。
实施例1
成纤维细胞培养与传代
无菌条件下取***环切术中切除的皮肤真皮组织,剪碎移入离心管中,加入约10倍于组织体积的0.1%的I型胶原酶(WORTHINGTON公司)消化,37℃,2小时,100目细胞滤器过滤后离心,1000转5分钟,弃去上清,加入含10% FBS DMEM培养液(GIBCO)混匀细胞,以2×104/cm2接种于100mm培养皿(FALCON,FranklinLakes NJ)中培养,待细胞融合至80%-90%时,进行传代,传代密度为1×104/cm2。体外扩增到第二代后,进行实验分组。
实施例2
单层培养细胞诱导法诱导形成软骨细胞
在本实施例中,用单层培养细胞诱导法诱导形成软骨细胞。方法如下:
用0.1%BSA稀释CDMP1生长因子(Research Diagnostics)至100μg/ml,加入含10%FBS的F-12(Gibco公司)培养液使其终浓度为100ng/ml,配制成细胞诱导液。取第二代成纤维细胞接种6-12小时,细胞诱导,每三天换一次培养液,诱导7天(在37℃,5%CO2,饱和湿度),分离获得软骨细胞。
实施例3
离心管培养细胞诱导
在本实施例中,用单层培养细胞诱导法诱导形成软骨细胞。方法如下:
诱导液配制同实施例2。收取传代至第6代的成纤维细胞1500转离心5分钟,弃上清后加入细胞诱导液,将细胞团块摇起,诱导7天,获得软骨细胞团。
石蜡包埋后切片,供Masson′s Trichrome,Alcian Blue与免疫组化染色等鉴定使用。
实施例4
诱导效果的检测
一、检测方法
(a)细胞激光共聚焦检测
将实施例2中对照组和实验组细胞分别在培养和诱导7天后,95%丙酮固定10分钟,PBS漂洗,10%羊血清封闭30分钟,兔抗I、II、III型胶原抗体(1:100,Chemicon公司)分别孵育1小时,PBS漂洗,FITC山羊抗兔IgG(1:1000稀释,Oncogene公司)孵育40分钟,PBS漂洗,PI(Propidiumiodide碘化丙啶)(1:1000稀释,Sigma公司)室温孵育10分钟,PBS漂洗后封片。将标记后的标本置于双光子激光共聚焦显微镜(购自Zeiss公司,型号LSN510),激光激发波长488nm,镜下观察。所得图像通过镜载摄像***输入电脑保存,自动拍照。
(b)细胞团块II型胶原免疫组织化学检测
将实施例3中制备的切片脱蜡入水后,PBS漂洗,3%H2O2封闭内源性过氧化物酶,滴加0.4%胃蛋白酶37℃孵育30min,加入兔抗人II型胶原单克隆抗体(1:100,Chemicon公司),置湿盒4℃过夜,滴加山羊抗兔二抗(1:100,DAKO),37℃孵育30min,PBS漂洗后,DAB显色。苏木素复染,中性树脂封片。以不加一抗样本作为空白对照。
(c)细胞胶原表达RT-PCR检测
分别收集对照组和实验组在培养和诱导7天后细胞,检测II型胶原与聚集蛋白聚糖(Aggrecan)表达。Trizol(Invitrogen公司)提取总mRNA,检测I、II型胶原和聚集蛋白聚糖的mRNA表达。根据RT-PCR试剂盒(Takara)操作说明,进行反转录和PCR扩增反应。相应引物(由上海申能***生物科技有限公司合成)序列见表1。反应条件:94℃变性,55℃退火,35个循环。

(d)细胞II型胶原分泌Western印迹检测
分别收集对照组和实验组在培养和诱导7天后细胞,细胞总数为均1×106个,加入三去污裂解液1ml,裂解细胞后移入1.5ml Eppendorf tube内,10000rpm,4℃离心5分钟。取上清,加入等体积2×SDS Loading buffer,β-巯基乙醇2μl/100μl。100℃沸水变性10分钟,瞬离心2秒钟,待冷至室温即上样。聚丙烯酰胺凝胶电泳10mA电泳3-4小时。电泳结束后,90v,4℃转膜2小时,封闭缓冲液(PIERCE公司)4℃封闭过夜。37℃依次加入II型胶原鼠抗一抗(1:500,Sigma公司)、生物素化二抗(1:500,Vector Lab公司)、链亲和素-AP(1:3400,Pierce公司)进行杂交,逐级杂交放大后,滴加1-Step NBT/BCIP(Pierce公司)显色液,显色后进行结果分析。
二、结果
细胞形态变化:应用CDMP1诱导3天后,成纤维细胞形态未见无明显改变;诱导7天后时,可见长梭形的成纤维细胞开始向多角形、多边形转变,与软骨细胞形态非常相似。对照组细胞未见明显形态改变(图1)。
细胞胶原表达观察:激光共聚焦显微镜观察发现,单层培养的正常成纤维细胞(未诱导组)仅I、III型胶原荧光表达,未见II型胶原荧光阳性细胞。应用CDMP诱导后,II型胶原荧光阳性细胞明显增多,同时可观察细胞仍表达I、III型胶原荧光阳性,荧光强度略有减弱(图2)。
细胞团诱导后基质合成检测:应用离心管法培养的细胞团块,应用CDMP1诱导7天后,Masson′s Trichrome染色可见基质呈弥漫性亮绿色,表明胶原合成旺盛(图3A)。Alcian Blue染色可见基质呈均一性天青色蓝染,表明GAG大量沉积(图3B)。细胞团块II型胶原免疫组化染色发现,棕黄色的颗粒分布于细胞内与胞外基质中;II型胶原表达在细胞密度较高、基质合成致密的区域染色较强(图4)。对照组检测为阴性。
II型胶原和Aggrecan表达RT-PCR检测:单层培养的成纤维细胞CDMP1诱导后7天,RT-PCR检测到大小为510bp的II型胶原和Aggrecan扩增产物,与阳性对照软骨细胞扩增结果相符。为诱导组未见扩增阳性条带(图5A)。诱导成纤维细胞仍可检测到I、III型胶原mRNA表达(图5B,C)。
II型胶原表达Western印迹分析检测:单层培养的成纤维细胞CDMP1诱导后7天,可检测到II型胶原蛋白表达,与阳性对照组软骨细胞表达条带相同。未诱导对照组成纤维细胞未见II型胶原蛋白(图6)。
实施例5
组织工程软骨移植物的制备
将实施例2或3中制备的软骨细胞,分别以5×107个细胞/ml的终浓度与30%Pluronic F-127(一种市售的聚环氧乙丙烯商品)/DMEM混合,从而制得组织工程软骨移植物。
讨论
作为TGF-β超家族中的一类生长因子,人CDMP首先由关节软骨中克隆分离出,主要包括CDMP1与CDMP2两个亚型,分别对应BMP家族中BMP14与BMP13;同时对应鼠BMP家族中生长分化因子(Growth and Differentiation Factor,GDF)。
在目前所发现的BMP全部家族成员中,CDMP是最为特异的与软骨形态发生与发育相关的一类生长因子,在四肢骨的图式形成(patterning)、软骨发生与长骨生长过程中发挥重要的调控作用。
研究发现,CDMP1特异性表达于胚胎发育过程中关节及关节软骨形成部位(TsumakiN,Tanaka K,Arikawa-Hirasawa E,et al.Role of CDMP-1 inskeletal morphogenesis:Promotion of mesenchymal cell recruitment andchondrocyte differentiation.J.Cell Biol.,1999.144:161-173);体内发育模型(Storm EE and Kingsley DM.GDF5 coordinates bone and jointformation during digit development.Dev.Bio.1999,209:11-27)及转基因实验均证实,CDMP1主要通过促进间充质细胞的聚集(consendation),提高局部软骨前体细胞的数量,加速关节软骨形成、软骨细胞的分化并调节骨骼组织的发育。出生后及成年关节软骨组织中均可检测到CDMP的表达,提示CDMP在出生后可继续维持正常软骨组织的生长;体外器官培养研究发现,损伤的关节软骨中有大量CDMP表达,在外源性CDMP因子作用下,软骨基质合成明显增加,提示CDMP可促进关节软骨的损伤修复过程(Ludwig E,Chee KNG,Robert U,et al.Presence of cartelage-derived morphogenetic proteins in articularcartelage and enhancement of matrix replacement in vitro.Arthriti.Rheumat.1998,41:263-273)。上述研究表明,CDMP主要通过调节间质前体细胞的分化,参与了软骨组织发生、生长与损伤修复的几乎全部生物学过程,是重要的调节软骨细胞分化的生长因子。
一般认为,真皮成纤维细胞是分化完全的终末体细胞,虽然取材与培养简单、体外扩增能力强,但不再具备干细胞样的向其他类型细胞分化的能力,只作为真皮组织工程种子细胞来源。然而,本发明中应用CDMP1生长因子,作用于体外培养的真皮成纤维细胞,却发现7天后细胞可表达特异性软骨细胞基质:II型胶原与聚集蛋白聚糖,同时伴有细胞形态的软骨细胞样改变,这证实了终末分化的二倍体成纤维细胞仍然具有作为构建其他组织的种子细胞来源的潜力。
软骨细胞外基质主要由II型胶原纤维(COLII)与蛋白聚糖组成,其中蛋白聚糖的主要成分为聚集蛋白聚糖。胶原纤维形成网络结构具有很高的抗张强度,存在于网络中的聚集蛋白聚糖分子的毛刷样结构能使其更好地对抗压力与II型胶原一起共同支持软骨组织的结构,维持着软骨组织的生物学特征,因此COLII与聚集蛋白聚糖的表达目前是软骨细胞最特异的标记(Micky D.Tortorella,Michael A.,et al.The Interglobular Domain of CartilageAggrecan Is Cleaved by Hemorrhagic Metalloproteinase HT-d(AtrolysinC)at the Matrix Metalloproteinase and Aggrecanase Sites.J.Bio.Chem.1998,273:5846-5850)。真皮成纤维细胞本身不表达COLII与聚集蛋白聚糖,在本发明的培养体系作用下,单层培养的成纤维细胞经CDMP诱导7天后,细胞形态发生了由梭形向软骨细胞样多角形形态的转变,免疫荧光、Western-Blot与RT-PCR均证实了II胶原的表达;RT-PCR结果也观察到成纤维细胞诱导后表达聚集蛋白聚糖。离心管培养是模拟软骨形态发生过程中间充质前体细胞聚集、分化过程,研究干细胞向软骨细胞分化的体外培养方法。为了进一步证实上述研究结果,本发明人采用离心管培养技术研究成纤维细胞在聚集状态下的诱导分化过程。研究发现,在CDMP1作用下,诱导7天的细胞团经阿丽新兰染色证实有大量酸性粘多糖沉积,Masson’s染色也观察到胶原与粘多糖的合成。免疫组化、Western-Blot与RT-PCR均检测到离心管诱导培养后COLII与聚集蛋白聚糖的表达。上述研究结果与骨髓基质干细胞向软骨细胞诱导分化的报道相符(Johnstone B,Hering TM,Caplan AI,et al.In vitro chondrogenesisof bone marrow derived mesenchymal progenitor cells.Exp cell res,1998,238:265—272.)。
本研究中发现,在CDMP1作用下成纤维细胞虽然表达COLII与聚集蛋白聚糖,但I、III型胶原的表达并未消失,表明经诱导的成纤维细胞与自然的软骨细胞还有一定的差距。Lennon(Lennon D,Haynesworth SE,ArmDM,et al.Dilution of human mesenchymal stem cells with dermal fibroblasts andthe effects on in vitro and in vivo osteochondrogenesis.Dev.Dyn.2000,219:50-62)等将人真皮成纤维细胞与骨髓基质干细胞以25-50%的比例混合,离心管培养诱导后仍可见软骨组织形成,显著降低了骨髓基质干细胞的用量。本发明人认为,通过CDMP的诱导,在软骨组织构建过程中有可能进一步降低骨髓基质干细胞的比例。同时由于软骨形成常为软骨内化骨过程中的早期阶段,因此本发明人认为本发明中成纤维细胞表达特异性软骨基质,不能排除是成纤维细胞向成骨表型分化的一个阶段。在骨髓基质细胞的诱导分化研究中发现(GruberR,Mayer C,Schulz W,et al.St imulatory effects ofcarti lage-derived morphogenetic proteins 1 and 2 on osteogenicdifferentiation of bone marrow stromal cells.Cytokine.2000,12:1630-1638),CDMP1、2均可促进骨髓基质干细胞向成骨细胞表型分化,但其成骨作用明显低于BMP-6与OP-1;虽然有IIA型前胶原表达,但未检测到作为成熟软骨细胞基质的IIB型前胶原与聚集蛋白聚糖分泌,该研究中生长因子作用时间仅为4天。在另一项研究中,当CDMP诱导时间延长至7天时,骨膜源性细胞表现了成骨与成软骨的多向分化,可见明显II型胶原与聚集蛋白聚糖表达,成软骨作用明显强于成骨作用。因此,CDMP在不同作用时间下、针对不同细胞表现出成骨与成软骨的双能作用,有必要在同一作用条件下继续观察其成骨或成软骨的功能。
本发明的实验证明了,真皮成纤维细胞通过CDMP-1诱导,在单层培养与离心管培养的条件下,均出现了向软骨细胞表型分化的现象,成纤维细胞可合成特异性软骨细胞基质:II型前胶原与聚集蛋白聚糖。这一研究结果目前尚未见文献报道,提示通过CDMP的诱导作用,成纤维细胞具有作为软骨组织工程新的种子细胞来源的可能。

Claims (7)
1.一种制备软骨细胞的方法,其特征在于,包括步骤:
(a)在适合生长的条件下,将成纤维细胞在含10-1000纳克/毫升软骨源性形态发生蛋白的成纤维细胞培养液中培养5-15天,从而使成纤维细胞诱导形成软骨细胞;
(b)从培养物中分离出软骨细胞。
2.如权利要求1所述的方法,其特征在于,所述的生长条件是37±2℃,CO25±3%。
3.如权利要求1所述的方法,其特征在于,所述的培养液是含0-20%胎牛血清的F-12,DMEM或1640培养液。
4.如权利要求1所述的方法,其特征在于,所述的成纤维细胞是人的成纤维细胞。
5.如权利要求1所述的方法,其特征在于,步骤(a)中所述的成纤维细胞的密度为0.5×104-5×105个/ml。
6.如权利要求1所述的方法,其特征在于,所述的成纤维细胞是原代的或传代的成纤维细胞。
7.一种制备软骨移植物的方法,其特征在于,包括步骤:
(a)在适合生长的条件下,将成纤维细胞在含10-1000纳克/毫升软骨源性形态发生蛋白的成纤维细胞培养液中培养5-15天,从而使成纤维细胞诱导形成软骨细胞;
(b)从培养物中分离出软骨细胞;
(c)将步骤(b)中获得的软骨细胞与药学上可接受的生物可降解材料混合,形成软骨移植物,其中所述移植物中所述软骨细胞的含量为1×105-5×107个细胞/毫升,或1×105-5×107个细胞/cm3。
