Die Standard Methode zur Bestimmung von Immunlagen in vivo durch Zell vermittelte Immunantworten besteht darin, einem Individuum Antigene parenteral zuzuführen und nach Ablauf einer gewissen Zeitdauer nach der Zuführung die Auswirkung letzterer klinisch zu beurteilen. Werden bei dieser Methode Antigene in die Haut gebracht und werden zur Bestimmung der Immunlage Hautreaktionen beurteilt, spricht man von der Messung von Zellimmunantworten mittels Hautreaktionen vom verzögerten Typ (auch: delayed type hypersensitivity, DTH, nur englisch gebräuchlich, genannt). Sind Zellimmunantworten positiv, indem beispielsweise eine verzögerte Schwellung der Haut nach sub- oder intrakutaner Verabfolgung eines aus einem Protein bestehenden Antigenes (oder anderer) klinisch festgestellt wird, so liegt eine Immunität gegen das Antigen und die durch dieses ausgelöste Krankheit (z.B. Infektionskrankheit) vor. Sind Zellimmunantworten negativ, indem keine Schwellungen, Reizungen oder dergleichen feststellbar sind, so liegt keine nachweisbare Immunität vor.
