Beschreibung description
The present invention relates to a process for the preparation of alkali metal or alkaline earth metal salts of (6R) or (6S) N (5) -methyl-5,6,7,8-tetrahydrofolic acid.
N5-methyl-5,6,7,8-tetrahydrofolic acid, sometimes abbreviated herein as N5-methyl-THF, is the predominant circulating form of reduced folates in mammals.
There is increasing interest in the use of this natural metabolite as at least one active compound in a therapeutic agent, for example as a vitamin in folate deficiency states. Therapeutic agents containing N5-methyl-THF can also be used for synergistically influencing a cancer-fighting compound and / or for reducing the toxicity of a cancer-fighting compound and / or for protecting human and / or animal cells. Such an agent usually contains N5-methyl-THF in an amount of 1 mg to 500 mg, in particular 5 mg to 150 mg, per unit dose. The medicament is preferably in the form of a parenteral and / or oral pharmaceutical preparation. Reference is made to references 1 to 5 mentioned at the end of the description.
Die absolute Konfiguration der natürlichen Form von N5-Methyl-THF entspricht der folgenden Formel A The absolute configuration of the natural form of N5-methyl-THF corresponds to Formula A below
und wird als (6S,L) definiert, wobei sich «6S» auf die Konfiguration am C6 des Pterin-Ringsystems und «L» auf die Konfiguration des a-Kohlenstoffatoms der Glutamat-Seitenkette beziehen. and is defined as (6S, L), where “6S” refers to the configuration at C6 of the pterin ring system and “L” refers to the configuration of the a-carbon atom of the glutamate side chain.
Commercially available forms of N5-methyl-THF obtained from the chemical reduction of folic acid are a (6RS) -diastereoisomeric mixture with the «L» configuration in the glutamate side chain.
There is no agreement on the function of the (6R) diastereoisomer of N5-methyl-THF. The unnatural (6R) diastereoisomer of N5-methyl-THF has been assumed to be inert and, as such, is excreted unchanged; see, for example, reference No. 13. However, it has also been postulated that the unnatural (6R) diastereoisomer could interfere with the folate transport system across the cell membrane in mammals, including humans; see for example references 6 to 8.
The individual (6S) and (6R) diastereoisomers of N5-methyl-THF have been produced in milligram quantities of questionable chemical purity, either by complex enzymatic reactions (see, for example, reference No. 6), or indirectly by reducing the corresponding N5, Nio-methyl-THF isomer, which was obtained by chromatographic separation of the (6RS) mixture (see references Nos. 7 and 9).
5,6,7,8-tetrahydrofolic acid is sometimes abbreviated as THF.
Diese Verfahren basieren auf der fraktionierten Kristallisation von N5, N1°-Methenyl-THF.CIe.HCI der Formel Indirect processes for the preparation of the individual diastereoisomers of Ns-methyl-THF have recently been published; see references Nos. 10 and 11. These methods are based on the fractional crystallization of N5, N1 ° -methenyl-THF.CIe.HCI of the formula

H - H • Cl coou followed by chemical conversion of the separate, individual (6R) and (6S) diastereoisomers into N5-methyl-THF. The present invention is characterized by the features in the independent claim. Preferred embodiments of this invention are defined in the dependent claims.
Various possible embodiments are described below. Embodiments as defined in the claims are normally no longer repeated.
In the first process for the preparation of the new ammonium salts, N5-methyl-THF is preferably introduced in the form of the calcium salt in water. The nitrogen-containing base, which is then added, can be a primary or secondary alkyl or arylamine. The calcium cation present is expediently precipitated as calcium oxalate by the addition of oxalic acid, which is then filtered off. The aqueous filtrate contains the dissolved ammonium salt. After evaporation of the water under reduced pressure or after lyophilization, the ammonium salt is usually obtained as an amorphous glass.
In the second process for the preparation of the new ammonium salts, N5-methyl-THF, ie in the form of the carboxylic acid, is advantageously introduced in water or in a mixture of water and ethanol and / or methanol. This process eliminates the steps of precipitating and separating the alkali metal or alkaline earth metal cation, as described in the first process. The remaining steps of the second method correspond to the corresponding steps in the first method.
The new ammonium salts can also be produced by ion exchange on a corresponding column, on which, for example, a calcium cation is exchanged for the desired ammonium ion.
The new ammonium salts, for example prepared according to a method described above, are now subjected to a fractional crystallization in which the (6RS) -diastereoisomer mixture is separated into the individual, pure (6R) and (6S) diastereoisomers. For this purpose, preferably one part by weight of a new ammonium salt is mixed with 5 parts by weight, based on the above part by weight of ammonium salt, of a mixture of 85% by volume of ethanol and 15% by volume of water. Depending on the ammonium salt, heating to a temperature of 40 ° C to 60 ° C may be necessary to obtain a clear solution.
A diastereoisomer is allowed to crystallize out of this clear solution and then filtered off. Crystallization can take place at ice water temperature or in the refrigerator or at a temperature of 10 ° C to 25 ° C. If necessary, the clear solution can be seeded with seed crystals and then treated with ultrasound if necessary. The mother liquor obtained is preferably diluted with ethanol with a low water content. The other diastereoisomer is allowed to crystallize out of this dilute solution and is then also filtered off. This second mother liquor can be concentrated and subjected to another fractional crystallization.
Which diastereoisomer crystallizes first depends, among other things, on the nature of the ammonium salt to be separated. In the case of the NS-methyl-THF-cyclohexylammonium salt, the (6R) salt crystallizes out first. The (6S) salt is crystallized out of the mother liquor as described above. In the case of the N5-methyl-THF-diisopropylammonium salt, the (6S) salt crystallizes out first. The (6R) salt is then crystallized out of the mother liquor.
The diastereoisomerically pure ammonium salts of N5-methyl-THF thus separated are converted into the alkali metal or alkaline earth metal salts, in particular into the calcium salt or the magnesium salt. For this purpose, a corresponding ammonium salt is preferably dissolved in a volume of water. Then an aqueous solution of the desired alkali metal or alkaline earth metal salt is added, for example calcium chloride. The pH in this mixture can be adjusted to a value of 6.0 to 7.0. This solution is cooled to a temperature of 0 ° C to 10 ° C and preferably stirred for about an hour. Then 1.5 to 2.5 parts by volume, based on the above part by volume of water, of, for example, ethanol or acetone are added, preferably while stirring

2.03 g of diisopropylamine were added to a suspension of 5.0 g of (6RS) -N5-methyl-THF calcium salt in 40 ml of water, which was stirred at a temperature of 40 ° C. under a nitrogen atmosphere. The temperature was raised to about 55 ° C and a clear solution was obtained. After cooling to room temperature, a solution of 1.2 g of oxalic acid dihydrate in 10 ml of water was added, and this mixture was stirred at a temperature of 0 ° C for 30 minutes. After the calcium oxalate formed had been removed by centrifugation, the supernatant solution was concentrated at a temperature of 70 ° C. under reduced pressure. Crude (6RS) -N5-methyl-THF-diisopropylammonium salt was obtained in the form of a honey-like resp. vitreous solid. HPLC analysis of this product showed a single peak.

Man erhielt 1,07 g an (6S)-N5-MethyI-THF-diisopropylammoniumsalz. The crude (6RS) -N5-methyl-THF-diisopropylammonium salt prepared according to Example 5 was dissolved in 27 ml of a mixture of 85 parts by volume of ethanol and 15 parts by volume of water at a temperature of 60 ° C. The resulting clear solution was cooled to room temperature and stirred at this temperature for 48 hours. The solid obtained was isolated by centrifugation. This solid was washed once with 85% ethanol and dried under reduced pressure at a temperature of 50 ° C for 2 hours. 1.07 g of (6S) -N5-methyl-THF-diisopropylammonium salt were obtained.
Analysis on a RESOLVOSIL column showed a diastereoisomeric ratio of (6S) / (6R) = 90/10 (see reference no. 12).
