Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von C-5 ethylenisch-ungesättigten Hydantoinderivaten, welche nützlich sind als Zwischenprodukte bei der Synthese von 1,3-Dihydro-2H-imidazo[4,5-b] chinolin-2-onen, welche potente antithrombotische Mittel sind.
Die Kondensation von Hydantoin (Eis) mit aromatischen Aldehyden (2) zur Herstellung von C-5-ungesättigten Hydantoinderivaten (3) ist in der Literatur beschrieben: E. Ware, Chemical Reviews, 1950, 46, Seiten 403-470; H.L. Wheeler et al., Am. Chem. J., 1911, 45, Seiten 368-383 und US-A 4 345 042 und US-A 4 582 903. Diese Reaktion wird folgendermassen illustriert:

worin R Wasserstoff oder einen Substituenten darstellt. Die Addukte (3) sind von synthetischem Nutzen als Vorläufer für Derivate von Phenylalanin und Phenylbrenztraubensäure. Es ist eine Anzahl verschiedener Katalysatoren und Lösungsmittelsysteme beschrieben worden, um diese Umwandlung durchzuführen. Im allgemeinen werden die Aldehyde, das Hydantoin und der Ka talysator in einem zweckmässigen Lösungsmittel auf eine Temperatur im Bereich von 70 bis 170 DEG C während 0,5 bis 24 h erwärmt. Einige der meist verwendeten Bedingungen sind folgende: NaOAc/AcOH bei 160 bis 170 DEG C während 3 h; in NH4OAc/AcOH unter Rückfluss während 4 h; in NaOAc/Ac2O unter Rückfluss während 1 h; in Piperidin bei 130 bis 140 DEG C während 0,5 h; und in Dimethylamin bei 100 DEG C während 22 h in einem geschlossenen Reaktionsgefäss.
Diese Verfahren sind jedoch nicht ganz befriedigend, werden nämlich einige Aldehyde nicht mit Hydantoin umgesetzt oder bilden nur Produkte in niederer Ausbeute. Es wurde nicht berichtet, dass einfache Ketone in dieser Reaktion umgesetzt werden.
Die Reaktion von Phosphonatcarbanionen mit Ketonen und Aldehyden ist allgemein bekannt: Wadsworth et al., J. Amer. Chem. Soc., 1961, 83, Seiten 1733-1738; Wadsworth, Org. Reactions, 1978, 25, 73-253.
Die vorliegende Erfindung stellt das im Patentanspruch 1 definierte Verfahren für die Herstellung von C-5-ethylenisch ungesättigten Hydantoinderivaten zur Verfügung, welche als Zwischenprodukte bei der Synthese von 1,3-Dihydro-2H-imidazo-[4,5-b]chinolin-2-onen nützlich sind, wie dies in den folgenden parallelen amerikanischen Patentanmeldungen beschrieben ist: Serial No. 726 869 am 25. April 1985 eingereicht; Serial No. 832 212, am 26. Februar 1986 eingereicht; Serial No. 866 813 am 23. Mai 1986 eingereicht; und Serial No. 883 258 am 8. Juli 1986 eingereicht. Die 5-ethylenisch ungesättigten Hydantoinderivate, welche nach dem erfindungsgemässen Verfahren hergestellt werden können, sind ebenfalls nützlich als Zwischenprodukte bei der Herstellung von anderen Verbindungen, z.B. von Phenylalanin und Phenylbrenztraubensäure.
Das im erfindungsgemässen Verfahren eingesetzte Dialkyl-5-phos-phonathydantoin besitzt vorzugsweise folgende Formel:

worin R1 und R2 Wasserstoff, Alkyl oder Aralkyl mit bis zu 10 C-Atomen sind, z.B. Benzyl, Phenethyl, Phenylpropyl und R3 Niederalkyl ist, wie beispielsweise Methyl, Ethyl oder Propyl.
Die Erfindung stellt ebenfalls die neue Verbindung, Diethyl-1-methyl-2,4-dioxoimidazolidin-5-phosphonat zur Verfügung. Das entsprechende Diethyl-2,4-dioxoimidazolidin-5-phosphonat ist in der Literatur in Chem. Abstracts, 1985, 102, Nr. 204 029a beschrieben.
Die Phosphonate der Formel I werden durch Bromierung eines zweckmässigen R1- und R2-Imidazolidin-2,4-dion in Essigsäure hergestellt, wobei das entsprechende 5-Bromhydantoin erhalten wird und das Produkt anschliessend mit Trialkylphosphit umgesetzt wird.
Es kann jedes aliphatische oder aromatische Aldehyd oder Keton oder jede alpha-Dicarbonylverbindung im erfindungsgemässen Verfahren verwendet werden. Die aromatischen Aldehyde, die in der US-A 4 345 072 offenbart sind und die aromatischen und aliphatischen Aldehyde, welche in der US-A 4 582 903 offenbart sind, können im erfindungsgemässen Verfahren verwendet werden. Die in der US-A 4 345 072 beschriebenen aromatischen Aldehyde umfassen Benzaldehyd, Tolylaldehyd, 4-Isopropylbenzaldehyd, 4-Hydroxybenzaldehyd, 3,4,5-Trimethoxybenzaldehyd, 3-Brom-4-methoxybenzaldehyd, 3,4-Methylendioxybenzaldehyd, 2-Hydroxy-4-nitrobenzaldehyd, 4,5-dimethoxy-2-nitrobenzaldehyd, Salicylaldehyd, Vanillin, 4-Phenylbenzaldehyd, 4-Benzylbenzaldehyd, 4-Fluorbenzaldehyd, 4-Dimethylaminobenzaldehyd, 4-Acetoxybenzaldehyd, 4-Acetaminobenzaldehyd, 4-Methylthiobenzaldehyd und 3,5-Dichlor-4-hydroxybenzaldehyd.

Eine grosse Anzahl von aliphatischen und aromatischen Aldehyden und Ketonen und alpha-Dicarbonyl-Verbindungen, welche für die Praxis der vorliegenden Erfindung nützlich sind, werden in den folgenden Beispielen erläutert. Eine bevorzugte Klasse von Aldehyden besitzt die Formel RCHO, worin R ausgewählt ist aus der Gruppe:
a) substituiertes oder unsubstituiertes Alkyl, worin die Alkylgruppe 2-6 Kohlenstoffatomen besitzt,
b) substituiertes oder unsubstituiertes Alkenyl, worin die Alkenylgruppe 3-5 Kohlenstoffatome besitzt,
c) substituiertes oder unsubstituiertes Phenyl, worin einer oder mehrere Substituenten des Phenyles ausgewählt sind aus der Gruppe Halogen, Hydroxy, Alkoxy oder substituiertes Alkoxy, worin Alkoxy 1-6 Kohlenstoffatome besitzt, Nitro, Alkyl mit 1-6 Kohlenstoffatomen und substituiertes oder unsubstituiertes Amino, 2-(2-Methyldioxalano), 1-Piperidino, 1-Morpholino, 1-Pyrrolidino oder 1-Piperazino,
d) Thienyl,
e) Pyridinyl und
f) PhC(O).
Die Substituenten, welche an der Alkoxygruppe vorhanden sein können, umfassen Mono- und Dialkylaminogruppen, welche 1-6 Kohlenstoffatome aufweisen, eine Arylsulfonylgruppe mit 6-10 Kohlenstoffatomen, eine Carbonsäureestergruppe mit 2- 6 Kohlenstoffatomen und eine stickstoffhaltige, heterocyclische Gruppe mit 1-4 Stickstoffatomen, welche selbst weiter mit Cycloalkylgruppen, die 6-10 Kohlenstoffatome enthalten, substituiert sein kann. Die Substituenten, welche an den Aminogruppen vorhanden sein können, umfassen Mono- und Dialkylgruppen, die 1-6 Kohlenstoffatome enthalten, Alkylcarbonylgruppen, die 2-6 Kohlenstoffatome enthalten und Carboxylestergruppen, die 2-6 Kohlenstoffatome enthalten.
Die Substituenten, welche an der 1-Piperazingruppe vorhanden sein können, umfassen eine Aroylgruppe, die 6-10 Kohlenstoffatome aufweist und gegebenenfalls mit einer oder mehreren Alkoxygruppen mit 1-6 Kohlenstoffatomen oder Carbonsäureestergruppen mit 2-6 Kohlenstoffatomen substituiert sein kann. Die an der 1-Piperidinogruppe gegebenenfalls vorhandenen Substituenten umfassen Carbonsäureestergruppen mit 2-6 Kohlenstoffatomen und N-Cycloalkyl, N-Alkylcarboxamid mit 7-10 Kohlenstoffatomen.
Bevorzugte Ketone sind ausgewählt aus der folgenden Gruppe: C1-3-Alkyl-2-ketophenylacetat, C1-3-Alkyl-2-keto C1-5-Alkylacetat, substituiertes oder unsubstituiertes C4-8-Cycloalkylketon, worin einer oder mehrere Substituenten ausgewählt sind aus: Halogen, Hydroxy, Alkoxy und substituiertem Alkoxy, worin die Alkoxygruppe 1-6 C-Atome aufweist, und Alkyl mit 1-6 Kohlenstoffatomen, 1,2-Dioxocyclohexan, substituierte oder unsubstituierte C8-10-Alkylphenone, worin das substituierte Phenyl einen Substituenten trägt, der ausgewählt ist aus der Gruppe: Halogen, Hydroxy, C1-6-Alkoxy oder C1-6-Alkyl, 3-Oxoindol und N-C1-4-Alkyl-3-oxoindol. Obschon gewisse bevorzugte Klassen von Aldehyden und Ketonen definiert wurden, ist es möglich, dass sämtliche aliphatische oder aromatische Aldehyde oder Ketone oder alpha-Dicarbonylverbindungen im erfindungsgemässen Verfahren verwendet werden können.
Vorzugsweise werden 0,5 bis 1,5 Mole eines aliphatischen oder aromatischen Aldehydes oder Ketones oder einer alpha-Dicarbonylverbindung pro Mol Phosphonat bei einer Temperatur von 0 bis 50 DEG C unter basischen Bedingungen während 15 min bis 24 h umgesetzt. Die nachstehend beschriebenen Reaktionsbedingungen werden stärker bevorzugt.
Die Zugabe von aromatischen Aldehyden zu einem schwachen Überschuss von Phosphonaten und Natriumethoxid in Ethanol bei Zimmertemperatur führt im allgemeinen zu einer raschen Ausscheidung von 5-Benzylidinhydantoinderivaten in einer ausgezeichneten Ausbeute. Da die Phosphonat-Ausgangsmaterialien und die Diethylphosphat-Nebenprodukte wasserlöslich sind, besteht die Isolierung des Produktes einfach aus einer Verdünnung mit Wasser oder 2N-Salzsäurelösung und Wasser mit anschliessender Filtration. In einigen Fällen erleichtert die vorherige Entfernung des Lösungsmittels dieses Verfahren. Die aus der Reaktionsmischung isolierten Rohprodukte sind im allgemeinen von hoher Reinheit, wie dies aufgrund der NMR-Spektraldaten geschlossen werden kann, obschon häufig eine Mischung von geometrischen Isomeren erhalten wird.
Es werden, ungeachtet der sterischen oder elektronischen Natur der Aldehyde, hohe Ausbeuten erhalten, obschon sterisch gehinderte und deaktivierte Aldehyde mit einer etwas kleineren Reaktionsgeschwindigkeit umgesetzt werden. Die milden Reaktionsbedingungen sind für einen grossen Bereich von funktionellen Gruppen zweckmässig. Die oben beschriebenen, wasserfreien Bedingungen sind für den Erfolg der Reaktion nicht kritisch. Wässrige Natriumhydroxid oder Lithiumhydroxydlösungen in Ethanol sind äquivalent wirksam und ein Zweiphasen-System, welches Dichlormethan und wässriges Natriumhydroxyd mit oder ohne Phasentransferkatalysator enthält, führt ebenfalls zu einer hohen Ausbeute von Addukten. Andere zweckmässige Reaktionsbedingungen sind beispielsweise Triethanol in Acetonitril, Triethylamin und Lithiumbromid in Acetonitril und Lithiumbromid in Pyridin.
Aliphatische Aldehyde reagieren unter Bildung von 5-Alkylidenhydantoinderivaten mit Hydantoinphosphonaten in synthetisch nützlichen Ausbeuten. Phenylacetaldehyd reagiert exotherm mit Phosphonaten, wobei ausgezeichnete Ausbeuten an Addukten in 20 min erhalten werden können, obschon gemäss der Literatur Phenylacetaldehyd und 4-Methoxyphenylacetaldehyd als unter verschiedenen Bedingungen nicht mit Hydantoin kondensierbar gilt.
Alpha-Dicarbonylverbindungen reagieren im allgemeinen unter den üblichen Bedingungen exotherm, wobei hohe Ausbeuten von Addukten erhalten werden.
Cyclohexanon reagiert in Gegenwart von Lithium, im Gegensatz zu Natrium, mit den Hydantoinphosphonaten unter Bildung von hohen Ausbeuten. Die Lithiumsalze von Triethylphosphonoacetat hat sich als hoch chelatbildend erwiesen und das Lithiumatom kann eine ähnliche Rolle bei der Reaktion der Hy-dantoinphosphonate mit den Ketonen spielen. Obschon ein fest gebundenes Lithiumsalz stabiler sein muss und demzufolge weniger reaktiv ist, kann es die erhöhte Stabilität des Salzes ermöglichen, dass das Kupplungsverfahren erfolgreich mit alternativen Reaktionswegen, die hauptsächlich durch Protonentransfer gesteuert werden dürften, konkurrieren kann.
Die erfindungsgemäss hergestellten C-5-ungesättigten Hydantoinderivate können in einigen Fällen als Mischungen von (Z) und (E)-Isomeren gemäss den nachstehenden Formeln isoliert werden, worin R min eine Gruppe darstellt, welche von aliphatischen oder aromatischen Aldehyden abgeleitet ist und R1 und R2 wie oben definiert sind:

Das geometrische Isomer, worin R min bezüglich der N-R2-Gruppe des Hydantoinringes in cis-Stellung vorliegt, wird als (Z) bezeichnet, während dasjenige Isomer, worin R min für N-R2-Gruppe in trans-Stellung vorliegt, wird als (E) bezeichnet. Falls ein Vinylproton vorhanden ist, sind Signale im unteren Gebiet des NMR-Spektrums üblicher Weise vorherrschend. Wenn das Proton der Doppelbindung durch einen anderen Rest, wie beispielsweise durch eine vom Keton oder einer alpha-Dicarbonylgruppe abgeleiteten Gruppe, ersetzt wird, dann wird die (E) oder (Z)-Bezeichnung aufgrund der Cahn-Wright-Prelog-Prioritätsordnung vorgenommen (vgl. Advanced Organic Chemistry, 2. Auflage, Jerry Merch, McGraw-Hill, 1977, Seiten 113-117 und Seiten 99-101).
Die folgenden Beispiele dienen zur Erläuterung der folgenden Erfindung. Sämtliche Temperaturen sind in DEG C angegeben und die Schmelzpunkte, welche mit einem Thomas Hoover-Kapillarapparat aufgenommen wurden, sind unkorrigiert. Bei der Angabe der Kernresonanz-Spektraldaten wurden übliche Abkürzungen verwendet, wobei Tetramethylsilan als interne Referenz verwendet wurde, und die chemischen Verschiebungen in ppm (Teile pro Million) angegeben sind. Sämtliche neuen Verbindungen ergaben befriedigende Elementaranalysedaten und wiesen Spektralcharakteristiken auf, welche mit den angenommen Strukturen in Einklang standen.
Beispiel 1
Diethyl-2,4-dioxoimidazolidin-5-phosphonat
Eine Mischung von Imidazolidin-2,4-dion (200 g, 2M) und Essigsäure (800 ml) wurden in einem \lbad auf 85 DEG C erwärmt. In einem Tropftrichter wurde Brom (352 g, 112,8 ml, 2,2 M) und ein kleiner Anteil Brom ( &tilde& 5 ml) gegeben und der Inhalt wurde unter heftigem Rühren in die Reaktionsmischung gegeben. War einmal die orange Farbe verschwunden wurde der Rest des Broms schnell während ungefähr 7 min zugegeben, wobei eine klare Lösung erhalten wurde. Nach Rühren bei 85 DEG C während 30 min wurde die Reaktionsmischung in einem Eisbad auf 30 DEG C abgekühlt und Triethylphosphit (465 g, 479 ml, 2,8 M) wurde in einer solchen Geschwindigkeit zugegeben, dass die Reaktionstemperatur im Bereich von 45 bis 50 DEG C blieb. Nach der Beendigung der Zugabe wurde das Eisbad entfernt und die Reaktionsmischung wurde bei Zimmertemperatur während 90 min gerührt.
Das Lösungsmittel wurde im Vakuum entfernt und der Rückstand mit Diethylether (800 ml) unter Aufwirbelung verdünnt, um eine Ausfällung eines weissen Feststoffes zu bewirken. Die Mischung wurde unter kräftigem Rühren in Diethylether (2 l) gegossen. Nach 30 min wurde der Feststoff abfiltriert, mit Diethylether gewaschen und an der Luft getrocknet, wobei Diethyl-2,4-Dioxoimidazolidin-5-phosphonat (337,0 g, 71%) erhalten wurde, welches ohne weitere Reinigung weiterverwendet wurde. Es wurde eine analytische Probe hergestellt, unter Umkristallisierung aus Ethanol, welches einen Schmelzpunkt von 161 bis 163 DEG C aufwies.

Beispiel 2
Diethyl-1-methyl-2,4-dioxoimidazolidin-5-phosphonat
Eine Mischung von 1-Methylimidazolidin-2,4-dion (202,5 g, 1,8 M) und Eisessig (1 l) wurden in einem \lbad auf 90 DEG C erwärmt. Ein Tropftrichter wurde mit Brom beschickt (311,5 g, 100 ml, 1,95 M) und ein kleiner Anteil Brom wurde in die Reaktionsmischung eingegeben. Nach dem Verschwinden der orangen Farbe wurde der Rest des Broms tropfenweise mit einer solchen Geschwindigkeit zugegeben, dass eine augenblickliche Entfärbung stattfand. Nach der Beendigung der Zugabe wurde die Mischung während 60 min auf 90 DEG C erwärmt, auf Zimmertemperatur abgekühlt und über Nacht gerührt. Der Eisessig wurde vom weissen Niederschlag dekantiert, im Vakuum konzentriert und der Rückstand wurde mit dem Niederschlag vereinigt und in Diethylether (etwa 2 l) suspendiert. Es wurde Triethylphosphit (295 g, 320 ml, 1,8 M) portionenweise und unter Rühren zugegeben.
Es fand eine exotherme Reaktion statt, welche mit Leitungswasserkühlung am Reaktionsgefäss unter Kontrolle gehalten wurde. Es wurde eine Lösung erhalten, welche unter fortgesetztem Rühren einen weissen Niederschlag bildete. Nachdem die Mischung 60 min stehengelassen wurde, wurde sie in Diethylether (4 l) gegossen und über Nacht stehengelassen. Die Filtration ergab Diethyl-1-methyl-2,4-dioxoimidazolidin-5-phosphonat (331,7 g, 75%), Schmelzpunkt 95-96 DEG C. Eine aus MeOH/Et2O umkristallisierte analytische Probe besass einen Schmelzpunkt von 95-96 DEG C. <1>H-NMR (CDCl3) delta 1,26 (6H, m, OCH2CH3), 2,90 (3H, s, N-CH3), 4,12 (4H, m, OCH2CH3), 4,72 (1H, d, J = 14Hz, P(0).CH.CO), und 11,06 (1H, bs, NH). 1R (KBr) 1025, 1050 (P-OC2H5), 1275, 1250 (P=O), 1725 (>C=O) und 1775 (>C=O) cm<-><1> . m/e 251 (M<+>).

Beispiel 3
5-[(2,3-Dimethyl-5-nitrophenyl)methylen]-2,3-imidazolidindion (R1=R4=H, R2=R3=CH3)
Natrium (0,41 g, 0,018 Grammatom) wurde in Ethanol (40 ml) aufgelöst und Diethyl-2,4-dioxoimidazolidin-5-phosphonat (4,21 g, 18 mmol) wurde zugegeben. Nach 5 min wurde 2,3-Dimethyl-6- nitrobenzaldehyd (1,66 g, 15 mmol) auf einmal zugegeben und die Mischung während 90 min bei Zimmertemperatur gerührt. Die Mischung wurde mit Wasser verdünnt, filtriert und der Feststoff wurde mit Wasser gewaschen und luftgetrocknet, wobei 5-[(2,3-Dimethyl-5-nitrophenyl)methylen]-2,4-imidazolidindion als einziges geometrisches Isomer (3,35 g, 86%) erhalten wurde. Durch Umkristallisation aus Methanol wurde eine analytische Probe erhalten, die einen Schmelzpunkt von 293-295 DEG C aufwies.

Beispiel 4
5-[(2-Methyl-6-nitrophenyl)methylen]-2,4-imidazolidindion (R1=R2=R4=H; R3=CH3)
Die Umsetzung von 2-Methyl-6-nitrobenzaldehyd mit Diethyl-2,4-dioxoimidazolidin-5-phosphat gemäss den in Beispiel 3 beschriebenen Verfahren ergab die Titelverbindung als 84:1 Mischung der geometrischen Isomere (F. 238-239 DEG C, Zersetzung) in einer Ausbeute von 81%.

Die Umsetzung von 2,3-Dimethyl-6-nitrobenzaldehyd mit Diethyl-1-methyl-2,4-dioxoimidazolidin-5-phosphonat gemäss den in Beispiel 3 beschriebenen Verfahren ergab die Titelverbindung (partielles Hydrat) als Mischung von geometrischen Isomeren, F. 195-198 DEG C, Ausbeute 88%.

Die Umsetzung von 2-Methyl-6-nitrobenzaldehyd mit Diethyl-1-methyl-2,4-dioxoimidazolidin-5-phosphonat gemäss dem in Beispiel 3 beschriebenen Verfahren ergab die Titelverbindung (partielles Hydrat) als Mischung von geometrischen Isomeren, F. 261-262 DEG C, Ausbeute 66%.

Die Umsetzung von 2,3,4-Trimethoxy-6-nitrobenzaldehyd mit Diethyl-2,4-dioxoimidazolidin-5-phosphonat gemäss den in Beispiel 3 beschriebenen Verfahren ergab die Titelverbindung als einziges geometrisches Isomer, F. 206-208 DEG C, Ausbeute 91% nach Auskristallisierung aus Methanol.

Beispiel 9

Die Umsetzung von 2-Methyl-6-nitrobenzaldehyd mit Diethyl-1-methyl-2,4-dioxoimidazolidin-5-phosphonat gemäss den in Beispiel 3 beschriebenen Verfahren ergab die Titelverbindung als Mischung von geometrischen Isomeren, F. 194-197 DEG C, Ausbeute 80%.

Die folgenden Beispiele 10 bis 27 erläutern die Herstellung von zusätzlichen Arylidenhydantoinen.
Beispiel 10
Ethyl-4-[3-[(2,4-dioxoimidazolidin-5-yl-iden)-methyl]-4-nitrophenoxy]butanoat
Natrium (4,92 g, 0,21 Grammatom) wurde in absolutem Ethanol (600 ml) aufgelöst und Diethyl-2,4-dioxoimidazolidin-5-phosphonat (50,5 g, 0,21 mol) wurde zugegeben. Nach 10 Minuten wurde eine Lösung von Ethyl-4-(3-formyl-4-nitrophenoxy)-butanoat (50,0 g, 0,18 Mol) in Ethanol (100 ml) miteinander zugegeben. Die Mischung wurde während 2 Std. gerührt, im Vakuum auf etwa 250 ml eingeengt und mit Wasser verdünnt. Nach 20 Min. wurde der Niederschlag abfiltriert. Zwei weitere Ausbeuten wurden anschliessend aus den Mutterlaugen gesammelt. Die vereinigten Feststoffe wurden im Vakuum über P2O5 getrocknet, wobei Ethyl-4-[3 [(2,4-dioxoimidazolidin-5-yliden)methyl] 4-nitrophenoxy]butanoat (61,3 g, 95%) erhalten wurde, welches gemäss NMR als 4:1-Mischung von geometrischen Isomeren nachgewiesen wurde.
Eine analytische Probe des Hauptisomeren wurde durch Umkristallisation aus wässrigem Ethanol erhalten und besass einen Schmelzpunkt von 131 bis 134 DEG C.

Die Herstellung erfolgte aus 2,4-Dioxoimidazolidin-5-phosphonat und Ethyl-4-(3-formyl-4-nitrophenoxy)acetat in analoger Weise, wie in dem in Beispiel 10 beschriebenen Verfahren; F. 268-270 DEG C.

Beispiel 15
N-Cyclohexyl-N-methyl-4-[3-[(1-methyl-2,4-dioxoimidazolidin-5-yliden)methyl-4-nitrophenoxy]butanamid
Natrium (0,079 g, 0,003 Grammatom) wurde in Ethanol (20 ml) aufgelöst und Diethyl-1-methyl-2,4-dioxoimidazolidin-5-phosphonat (0,86 g, 3,4 mmol) wurden zugegeben. Nach 5 min wurde N-Cyclohexy-4-(3-formyl-4-nitrophenoxy)-N-methyl-butanamid (1 g, 2,9 mmol) zugegeben und die Mischung wurde bei Zimmertemperatur während 90 min gerührt. Das Lösungsmittel wurde abgedampft und der Rückstand wurde mit Wasser verdünnt und mit Dichlormethan extrahiert, wobei ein Schaum erhalten wurde. Die Kristallisation auf Hexan/Dichlormethan ergab N-Cyclohexy-N-methyl-4-[3-[(1-methyl-2,4-dioxoimidazolidin-5-yliden)methyl-4-nitrophenoxy]butanamid (0,96 g, 75%) als eine 3:1-Mischung von geometrischen Isomeren, F. 149-154 DEG C.

Beispiel 16
5[[5[4(1-Cyclohexyl-1H-tetrazol-5-yl)-butoxy]-2-nitrophenyl]methylen]-2,4-imidazolidindion-Hydrat
Natrium, (1,38 g, 0,06 Grammatom) wurde in Ethanol (250 ml) aufgelöst und Diethyl-2,4-dioxoimidazolidin-5-phosphonat (14,22 g, 60 mmol) wurde zugegeben. Nach 5 min wurde eine Lösung von 5-[4-(1-Cyclohexyl-1H-tetrazol-5-yl)butoxy]-2-nitrobenzaldehyd (17,30 g, 46 mmol), welches gemäss T. Nishi, et al., Chem. Pharm. Bull., 33, 1140-1147 (1985) erhalten wurde, in Ethanol (50 ml) und Dichlormethan (50 ml) auf einmal zugegeben. Diese Mischung wurde während 10 min gerührt das Lösungsmittel abgedampft und der Rückstand mit Wasser und 2N-Salzsäurelösung verdünnt. Der gelbe Niederschlag wurde abfiltriert, mit Wasser gewaschen und luftgetrocknet, wobei 5-[[5-[4-(1-Cyclohexyl-1H-tetrazol-5-yl)-butoxy]-2-nitrophenyl] methylen]-2,4-imidazolidindion (18,38 g, 87%) erhalten wurde.
Eine analytische Probe (als partielles Hydrat) wurde durch Auskristallisation aus wässrigem Dimethylformamid hergestellt und hatte einen unbestimmten Schmelzpunkt.

Beispiel 17
5-[[2-Nitro-5-4-(phenylsulfonyl)butoxy]-phenyl]methylen]-2,4-imidazolidindion
Natrium (0,386 g, 0,017 Grammatom) wurde in Ethanol (70 ml) aufgelöst und Diethyl-2,4-dioxoimidazolidin-5-phosphonat (3,96 g, 17 mmol) wurde zugegeben. Nach 1 h wurde eine Lösung von 2-Nitro-5-[4-(phenylsulfonyl)butoxy]benzaldehyd (4,70 g, 13 mmol) (hergestellt durch Alkylierung von 5-Hydroxy-2-nitrobenzaldehyd mit 4-Phenylsulfonylbutyl-bromid) in Ethanol und Chloroform zugegeben. Nach 20 min wurde das Lösungsmittel abgedampft und der Rückstand mit Chloroform extrahiert, wobei ein schaumartiger Feststoff erhalten wurde, welcher in Acetonitril aufgelöst wurde. Es wurde bis zum Punkt der Ausfällung Diethylether zugegeben und die Mischung wurde über Nacht stehengelassen.
Ein Feststoff (0,86 g) wurde gesammelt und weiter durch Auskristallisation aus Acetonitril-diethylether gereinigt, wobei 5-[[2-nitro-5-[4-(phenylsulfonyl)butoxy]phenyl]-methylen]-2,4-imidazolidindion (0,5 g) erhalten wurde. Die Konzentration der kombinierten Mutterlaugen ergaben 4,1 g Material, welches ohne Reinigung weiterverwendet wurde. Ausbeute: 4,6 g (61%). Das kristallisierte Material hatte einen Schmelzpunkt von 150-152 DEG C.

Beispiel 18
5-[[5-[(2-Diethylamino)ethoxy]-2-nitrophenyl]-methylen]-2,4-imidazolidindion
Eine Mischung von 5-Hydroxy-2-nitrobenzaldehyd (10 g, 60 mmol), 2-Diethylaminoethylchlorid-hydrochlorid (13,4 g, 78 mmol), gepulvertes Natriumcarbonat (24,8 g, 180 mmol) und Dimethylformamid (200 ml) wurde auf 100 DEG C auf einem \lbad erwärmt. Nach 2 h wurde die Mischung abgekühlt, mit Wasser verdünnt und mit Diethylether extrahiert. Die kombinierten Extrakte wurden zweimal mit Wasser gewaschen, getrocknet und das Lösungsmittel abgedampft, wobei ein \l zurückblieb (14,6 g, 92%), welches alles auf einmal zu einer ethanolischen Lösung des Natriumsalzes von Diethyl-2,4-dioxoimidazolidin-5-phosphonat (hergestellt durch Auflösen von Natrium (1,46 g, 0,06 Grammatom) in Ethanol (200 ml) und Zugabe von Diethyl-2,4-dioxoimidazolidin-5-phosphonat (13,00 g, 49 mmol) erhalten wurde) gegeben wurde.
Nach 30 min wurde die Mischung mit Wasser verdünnt, filtriert, der Feststoff wurde mit Wasser gewaschen und an der Luft getrocknet, wobei 5-[[5-[(2-Diethyl-amino)ethoxy]2-nitrophenyl]-methylen]-2,4-imidazolidindion (11,0 g, 65%) erhalten wurde. Eine analytische Probe wurde durch Umkristallisation aus wässrigem Dimethylformamid hergestellt und besass einen Schmelzpunkt von 208 bis 211 DEG C (Zersetzung).

Beispiel 19
5-[[5-[[[3-(1-Methylethyl)-2-oxooxazolidin-5-yl]-methyl]oxy]-2-nitrophenyl]methylen]-2,4-imidazolidindion
Stufe 1: 2-[2-Nitro-5-(oxiranylmethoxy)phenyl]-1,3-dioxolan
Eine Mischung von 2-(2-Nitro-5-hydroxyphenyl)-1,3, dioxolan (29,5 g, 0,14 Mol), Epibromhydrin (29,03 g, 18,15 ml, 0,21 Mol), Natriumcarbonat (48,67 g, 0,35 Mol) und Dimethylformamid (150 ml) wurde unter Rühren auf 100 DEG C erwärmt. Nach 30 min wurde die Mischung abgekühlt, mit Wasser verdünnt und mit Diethylether extrahiert. Die kombinierten organischen Extrakte wurden mit Wasser (dreimal) gewaschen, über Natriumsulfat getrocknet und konzentriert, wobei ein kristalliner Feststoff (34,50 g, 92%) erhalten wurde, welcher ohne weitere Reinigung eingesetzt wurde. Eine analytische Probe wurde durch Auflösen von 1 g in Dichlormethan (15 ml) hergestellt. Die Zugabe von Hexan (etwa 50 ml) führte zur Ausfällung eines gelben Feststoffes, welcher durch Filtration abgetrennt wurde.
Ohne weitere Verdünnung mit Hexan fällte 2-[2-Nitro-5-(oxiranylmethoxy)phenyl]-1,3-dioxolan (0,8 g), F. 78-79,5 DEG C.

Stufe 2: 1-[3-(1,3-Dioxolan-2-yl)-4-nitrophenyl]-3-[(1-methylethyl)amino]-2-propanol
Eine Mischung von 2-[2-Nitro-5-(oxiranylmethoxy)phenyl]-1,3-dioxolan (2 g, 7,5 mmol) und Isopropylamin (10 ml) wurde während 23 Std. unter Rückfluss erwärmt. Das Isopropyl amin wurde im Vakuum entfernt und der Rückstand wurde in Dichlormethan aufgelöst, in Wasser gewaschen, getrocknet und konzentriert, wobei ein Feststoff erhalten wurde. Es wurde eine Reinigung erzielt, indem in Dichlormethan aufgelöst und durch einen Stopfen aus Silikagel filtriert wurde, wobei 10% Methanol/Chloroform als Elutionsmittel verwendet wurde. Der isolierte Feststoff wurde in Dichlormethan aufgelöst und mit Hexan verdünnt, wobei 1-[3-(1,3-Dioxolan-2-yl)-4-nitrophenyl]-3-[(1-methylethyl)amino]-2-propanol (0,8 g, 32%) erhalten wurde, F. 97-99 DEG C.
Anal. Ber. für C15H22N2O6: C, 55,20; H, 6,80; N, 8,59. Gefunden: C, 54,80; H, 6,69; N, 8,54.
Stufe 3: 5-[[[3-(1-Methylethyl)-2-oxooxazolidin-5-yl]-methyl]-oxo]-2-nitrobenzaldehyd
Phosgen (11,12 g, 0,11 Mol) in Toluol (50 ml) wurde tropfenweise zu einer gerührten Lösung von 1-[3-(1,3-dioxolan-2-yl)-4-nitrophenyl]-3-[(1-methylethyl]amino]-2-propanol (14,65 g, 0,05 Mol) und Pyridin (8,88 g, 9,1 ml, 0,11 Mol) in Dichlormethan (150 ml) gegeben bei 0 DEG C in einem Eisbad. Nach der Beendigung der Zugabe wurde das Eisbad entfernt und die Mischung auf Zimmertemperatur aufgewärmt und während 15 min gerührt, bevor mit Wasser verdünnt wurde. Die Mischung wurde mit Dichlormethan extrahiert, der kombinierte Extrakt wurde getrocknet und konzentriert, wobei ein \l erhalten wurde, welches in Tetrahydrofuran (300 ml) aufgelöst wurde. Verdünnte Salzsäurelösung (75 ml) wurde zugegeben und die Mischung wurde unter Rückfluss erwärmt. Nach 90 Min. wurde das Tetrahydrofuran abgedampft und der Rückstand mit Dichlormethan extrahiert.
Die kombinierten Extrakte wurden über Natriumsulfat getrocknet und das Lösungsmittel wurde abgedampft, wobei ein \l zurückblieb, welches unter Bildung eines gelben Feststoffes kristallierte (12,0 g, 86%). Eine analytische Probe wurde hergestellt, indem ein Teil (1 g) in Dichlormethan aufgelöst und Diethylether zur Ausfällung zugegeben wurde, wobei eine klebrige Substanz erhalten wurde. Nach der Dekantierung, wurde die Lösung mit Diethylether verdünnt und Hexan zugegeben, wobei 5[[[3-(1-Methylethyl)-2-oxooxazolidin-5-yl]methyl]oxy]-2-nitrobenzaldehyd (0,7 g, 74%) erhalten wurde, F. 94-97 DEG C.

Stufe 4: 5-[[5-[[[3-(1-Methylethyl)-2-oxooxazolidin-5-yl]-methyl]-oxy]-2-nitrophenyl]methylen]-
2,4-imidazolidindion
Natrium (0,9 g, 0,04 Grammatom) wurde in Ethanol (150 ml) aufgelöst und anschliessend Diethyl-2,4-dioxoimidazolidin-5-phosphonat (9,19 g, 40 mmol) zugegeben. Nach 30 min wurde festes 5-[[[3-(1-Methylethyl)-2-oxooxazolidin-5-yl]methyl]-oxy]-2-nitrobenzaldehyd (10,0 g, 32 mmol) zugegeben und die Mischung wurde heftig gerührt. Nach 30 Min. wurde die Mischung mit Wasser verdünnt (150 ml), filtriert und der Feststoff mit Wasser gewaschen und luftgetrocknet, wobei 5-[[5-[[[-3-(1-Methylethyl)-2-oxooxazolidin-5-yl]methyl]oxy]-2-nitrophenyl]methylen]-2,4-imidazolidindion (9,80 g, 77%) erhalten wurde. Eine analytische Probe wurde durch Umkristallisieren auf Dimethylformamid und Wasser erhalten und besass einen Schmelzpunkt von 285-287 DEG C (Zersetzung).
Anal. Ber. für C17H18N4O7: C, 52,31; H, 4,65; N, 14,35. Gefunden: C, 51,84; H, 4,64; N, 14,25.
Beispiel 20
5-[[5-[[3-(1,1-Dimethylethyl)-2-oxooxazolidin-5-yl]-methoxy]-2-nitrophenyl]methylen]imidazolidin-2,4-dion
Diese Verbindung wurde aus 5-[[[3-(1,1-Dimethylethyl)-2-oxooxazolidin-5-yl]methyl]oxo-2-nitrobenzaldehyd und Diethyl-2,4-dioxoimidazolidin-5-phosphonat in analoger Weise wie in dem in Beispiel 19 (Stufe 4) beschriebenen Verfahren erhalten; F. 273-275 DEG C (Zersetzung).
Anal. Ber. für C18H20N4O7: C, 53,46; H, 4,99; N, 13,86. Gefunden: C, 53,35; H, 5,08; N, 13,86.
Beispiel 21
5-[[[2-Nitro-5-[(3-tetrahydro-2H-pyran-2-yl)-oxy]-propoxy]phenyl]methylen]-2,4-imidazolidindion
Eine Mischung von 5-Hydroxy-2-nitrobenzaldehyd (8,18 g, 49 mmol), 1-Brom-3-(tetrahydro-2H-pyran-2-yl)oxypropan (11,50 g, 51 mmol), Kaliumcarbonat (7,16 g, 51 mmol), Kaliumiodid (katalytische Menge) und Dimethylformamid (800 ml) wurden unter Rühren während 30 min auf 110 DEG C erwärmt. Die Mischung wurde abgekühlt, mit Wasser verdünnt (150 ml) und mit Dichlorethan extrahiert. Die kombinierten Extrakte wurden mit Wasser gewaschen (dreimal), über Natriumsulfat getrocknet und im Vakuum konzentriert, wobei ein \l zurückblieb, welches in Ethanol (15 ml) aufgelöst wurde und zu einer Lösung von Natriumethoxid (3,99 g, 58 mmol) und Diethyl-2,4-dioxoimidazolidin- 5-phosphonat (13,87 g, 59 mmol) in Ethanol (200 ml) zugegeben. Nach 90 min wurde das Ethanol abgedampft und der Rückstand mit Wasser verdünnt und mit Dichlormethan extrahiert.
Die kombinierten Extrakte wurden über Natriumsulfat getrocknet und im Vakuum konzentriert, wobei ein \l erhalten wurde, welches in Dichlormethan aufgelöst und durch Silikagelstopfen filtriert wurde, wobei Diethylether als Elutionsmittels verwendet wurden. Die Verdampfung des Lösungsmittels ergab 5-[[2-Nitro-5-[(3-tetrahydro-2H-pyran-2-yl]propoxy]phenyl]methylen]-2,4-imidazolidindion als viskoses \l, welches ohne weitere Reinigung verwendet wurde. Eine analytische Probe von 2,4-Imidazolidin als partielles Hydrat wurde durch Ausfällung aus Dichlormethan mit Hexan hergestellt und besass einen Schmelzpunkt von 128-134 DEG C.

Beispiel 22
5[[5-(2-Ethoxyethoxy)-2-nitrophenyl]-methylen]-2,4-imidazolidindion
Eine Mischung von 5-Hydroxy-2-nitrobenzaldehyd (1,00 g, 6 mmol), 2-Bromethyl-ethylether (1,00 g, 0,74 ml, 6,5 mmol), gepulvertes Kaliumcarbonat (0,91 g, 6,5 mmol), Kaliumiodid (katalytische Menge) und Dimethylformamid (10 ml) wurde unter Rühren auf 110 DEG C erwärmt. Nach 30 min wurde die Mischung abgekühlt, mit Wasser verdünnt und mit Dichlormethan extrahiert (dreimal). Die kombinierten Extrakte wurden mit Wasser gewa schen (zweimal), über Natriumthiosulfat getrocknet und im Vakuum konzentriert, wobei ein \l erhalten wurde, welches mit Ethanol (3 ml) verdünnt wurde und zu einer gerührten Lösung von Natriumethoxid (0,53 g, 78 mmol) und Diethyl-2,4-dioxoimidazolidin-5-phosphonat (1,83 g, 77 mmol) in Ethanol (15 ml) gegeben.
Nach 10 Min. wurde die Mischung mit 2N-Salzsäurelösung verdünnt und der Feststoff abfiltiert und luftgetrocknet, wobei 5-[[5-(2-Ethoxyethoxy)-2-nitrophenyl]methylen]-2,4-imidazolidindion als ein partielles Hydrat erhalten wurde (1,40 g, 73%), F. 228-233 DEG C (Zersetzung).
Anal. Ber. für C14H15N3O6 0.12H2O: C, 51,99; H, 4,75; H, 13,00; H2O, 0,67. Gefunden: C, 51,59; H, 4,72; N, 12,82; H2O, 0,25. Anschliessend wurde eine zweite Ausbeute (0,2 g, 10%) gesammelt.
Beispiel 23
5-[[5-[3-(2-Methyl-1,3-dioxolan-2-yl)-propoxy]-2-nitrophenyl]methylen]2,4-imidazolidindion
Eine Mischung von 5-Hydroxy-2-nitrobenzaldehyd (20,00 g, 0,12 mol), 5-Chlor-2-pentanon-ethylenketal (21,7 g, 0,132 mol), Kaliumcarbonat (20,00 g, 0,14 mol), Kaliumiodid (0,5 g) und Dimethylformamid (200 ml) wurden unter Rühren auf 120 DEG C erwärmt. Nach 4 min wurde die Mischung abgekühlt, mit Wasser verdünnt und mit Diethylether extrahiert. Die kombinierten, etherischen Extrakte wurden mit Wasser gewaschen, über Magnesiumsulfat getrocknet und das Lösungsmittel abgedampft. Das zurückbleibende \l enthielt 5-[3-(2-methyl-1,3-dioxolan-2-yl) propoxy]-2-nitrobenzaldehyd (ohne Reinigung verwendet), wur de in Ethanol (200 ml) aufgelöst und in einem Teil zu einer Lösung von Natriumethoxid (10,05 g, 0,15 mol) und Diethyl-2,4-dioxoimidazolidin-5-phosphonat (35,00 g, 0,15 Mol) in Ethanol (300 ml) gegeben.
Die Mischung wurde bei Zimmertemperatur während 90 min gerührt, auf ein Volumen von etwa 90 ml eingeengt und mit Wasser verdünnt. Der gelbe Niederschlag wurde abfiltriert, mit Wasser gewaschen und im Vakuum bei 70 DEG C getrocknet, wobei 5-[[5-[3-(2-methyl-1,3-dioxolan-2-yl)-propoxy]-2-nitrophenyl]methylen]-2,4-imidazolidindion (38,17 g, 84%) erhalten wurde. Eine analytische Probe wurde durch Umkristallisation aus Ethanol erhalten und besass einen Schmelzpunkt von 175-180 DEG C.
Anal. Ber. für C17H19N3O7: C, 54,11; H, 5.08; N, 11,14. Gefunden: C, 54,34; H, 5,08; N, 10,85.
Beispiel 24
5-[[2-Nitro-5-[3-(phenylsulfonyl)propoxy]-phenyl]-methylen]-2,4-imidazolidindion
Die Verbindung wurde durch Umsetzungen von 2-Nitro-5-3-(phenylsulfonyl)propoxy benzaldehyd (erhalten durch Alkylierung von 5-Hydroxy-2-nitrobenzaldehyd mit 3-Phenyl-sulfonylpropylbromid) mit Diethyl-2,4-dioxoimidazolidin-5-phosphonat in analoger Weise, wie in den in Beispiel 17 beschriebenen Verfahren erhalten und besass einen Schmelzpunkt von 125-157 DEG C.
Anal. Ber. für C19H17N3O7S: C, 52,90; H, 3,97; N, 9,74. Gefunden: C, 52,81; H, 4,10; N, 9,71.
Beispiel 25
1-Methyl-5-[[2-nitro-5-[3-(phenylsulfonyl)-propoxy]-phenyl]methylen]-2,4-imidazolidindion
Die Verbindung wurde hergestellt durch Umsetzen von 2-Nitro-5-[3-(phenylsulfonyl)propoxy]benzaldehyd mit Diethyl-1-methyl-2,4-dioxoimidazolidin-5-phosphonat in analoger Weise, wie in den in Beispiel 17 beschriebenen Verfahren; F. 147-158 DEG C.

Beispiel 26
2-[3-[(2,4-Dioxoimidazolidin-5-yliden)-methyl]-4-nitrophenoxy]ethyl-acetat
Diethyl-2,4-dioxoimidazolidin-5-phosphonat wurde zu Triethylamin (äquimolar) in Acetonitril gegeben. Nach 1 Std. wurde eine äquimolare Menge von 4-[3-Formyl-4-nitrophenoxy]ethyl-acetat (erhalten durch Alkylierung von 5-Hydroxy-2-nitrobenzaldehyd mit 2-Bromethylacetat) zugegeben und die Mischung wurde während 2 h gerührt. Die Isolierung des Produktes gemäss dem Verfahren nach Beispiel 10 ergab 2-[3-[(2,4-Dioxoimidazolidin-5-yliden)methyl]-4-nitrophenoxy]ethylacetat; F. unbestimmt.

3-[3-[(2,4-Dioxoimidazolidin-5-yliden)-methyl]-4-nitrophenoxy]propyl-acetat
Die Verbindung wurde erhalten durch Umsetzen von Diethyl-2,4-dioxoimidazolidin-5-phosphonat mit Ethyl-4-[3-formyl-4-nitrophenoxy]propyl-acetat (erhalten durch Alkylierung mit 5-Hydroxy-2-nitrobenzaldehyd mit 3-Brompropyl-acetat) in analoger Weise wie in den in Beispiel 26 beschriebenen Verfahren, F. 94-130 DEG C.

Die folgenden zusätzlichen substituierten Hydantoine können nach den Verfahren gemäss den Beispielen 10-27 hergestellt werden, indem zweckmässige 2-Nitrobenzaldehyde und Hydantoin-5-phosphonate verwendet werden:
5-[[5-[3-[[(Cyclohexyl)methylamino]-carbonyl]propoxy]-2-nitrophenyl]methylen]imidazolidin-2,4-dion; 4-[[4-Nitro-3-[(2,4-dioxoimidazolidin-5-yliden)methyl]phenyl]-oxy]butansäure; 5-[[4-Nitro-3-[(2,4-dioxoimidazolidin-5-yliden)methyl]phenyl]oxy]pentansäure; N-Cyclohexyl-N-methyl-5-[3-[(2,4-dioxoimidazolidin-5-yliden)methyl]-4-nitrophenoxy]pentanamid; und N-Cycloheptyl-N-methyl-5-[[4-nitro-3-(2,4-dioxoimidazolidin-5-yl)methylen]-phenoxy]pentanamid.
Die folgenden Beispiele 30-42 illustrieren die Herstellung von zusätzlichen Arylidenhydantoinen.
Beispiel 30
5-[[3-Nitro-5-[(1-piperidinyl)phenyl]-methylen]-2,4-imidazolidindion
Natrium (0,6 g, 0,026 Grammatom) wurde in absolutem Ethanol (200 ml) aufgelöst und Diethyl-2,4-dioxoimidazolidin-5-phosphonat (6,0 g, 25 mmol) zugegeben. Nach 10 min wurde 2-Nitro-5-(1-piperidinyl)benzaldehyd (5,0 g, 21 mmol) in einem Teil zugegeben und die Mischung bei Zimmertemperatur während 5 h gerührt. Der gelbe Niederschlag abfiltriert, mit Wasser gewaschen und luftgetrocknet, wobei 5-[[2-Nitro-5-(1-piperidinyl)phenyl]methylen]-2,4-imidazolidindion (6,17 g, 92%) erhalten wurde; F. 273-276 DEG C. Eine bei 110 DEG C vakuumgetrocknete Probe hatte einen Schmelzpunkt von 280 DEG C (Zersetzung).

Beispiel 31
5-[[2-Nitro-5-(1-pyrrolidinyl)phenyl]-methylen]-2,4-imidazolidindion
Hergestellt aus Diethyl-2,4-dioxoimidazolidin-5-phosphonat und 2-Nitro-5-(1-pyrrolidinyl)benzaldehyd, analog zum Verfahren gemäss Beispiel 30, F. 289 DEG C (Zersetzung), kristallisiert aus DMF-H2O.

Die entsprechenden Hydantoine, worin R1 Methyl ist, wurden durch Verwendung des zweckmässigen Benzaldehydes und Diethyl-1-methyl-2,4-dioxoimidazolidin-5-phosphonat erhalten.
Die nachfolgenden Beispiele 43-45 erläutern die Kupplung eines aromatischen Aldehydes mit einem Dialkyl-5-phosphonat-Hydantoin.
Beispiel 43
5-[(2-Nitrophenyl)methylen]-2,4-imidazolidindion
Lithiumhydroxid-monohydrat (0,334 g, 8 mmol) wurde in Wasser (10 ml) verdünnt und die Lösung wurde mit Ethanol (20 ml) verdünnt. Diethyl-2,4-dioxoimidazolidin-5-phosphonat (1,875 g, 8 mmol) wurde zugegeben und die Mischung wurde während 5 Min. gerührt, bevor festes 2-Nitrobenzaldehyd (1 g, 6,6 mmol) zugegeben wurde. Innerhalb von wenigen Minuten wurde ein schwerer gelber Niederschlag gebildet. Nach 15 min wurde die Mischung mit 1N-Salzsäurelösung verdünnt und der Feststoff mit Wasser gewaschen und luftgetrocknet, wobei eine 2,5:1 Mischung von (Z)- und (E)-5-[(2-Nitrophenyl)methylen]-2,4-imidazolidindion erhalten wurde; Ausbeute 1,48 g, 96%, F. 300-302 DEG C (Zersetzung).

Beispiel 44
5-(Phenylmethylen)-2,4-imidazolidindion
Benzaldehyd (0,5 g, 5 mmol) in Dichlormethan (10 ml) wurde zu einer Lösung von Diethyl-2,4-dioxoimidazolidin-5-phosphonat (1,45 g, 6 mmol) und 4N-Natriumhydroxidlösung (0,15 ml) in Wasser (10 ml) gegeben. Die Mischung wurde über Nacht heftig gerührt, der Feststoff abfiltriert und mit Wasser gewaschen und luftgetrocknet, wobei 5-(Phenylmethylen)-2,4-imidazolidindion (0,73 g, 82%) erhalten wurde.
Die Wiederholung der obigen Reaktion in Gegenwart von Tetrabutylammonium-hydrogensulfat (Spatelspitze) resultierte die Isolierung von 5-(Phenylmethylen)-2,4-imidazolidindion (0,75 g, 84% Ausbeute).
Beispiel 45
Die Reaktion des zweckmässigen aromatischen Aldehyds mit einem Dialkyl-5-phosphonat-hydantoin ergab die folgenden Verbindungen:
a) (Z)-1-Methyl-5-[(2-nitrophenyl)methylen]-2,4-imidazolidindion, F. 273-275 DEG C.

Beispiel 46
5-(2-Phenylethyliden)-2,4-imidazolidindion
Natrium (0,460 g, 0,02 Grammatom) wurde in Ethanol (30 ml) aufgelöst und Diethyl-2,4-dioxoimidazolidin-5-phosphonat (4,72 g, 20 mmol) wurde zugegeben. Nach 5 min wurde frisch destilliertes Phenylacetaldehyd (2,0 g, 16 mmol) zugegeben, wobei eine schwach exotherme Reaktion erhalten wurde. Nach 15 min wurde die Reaktionsmischung in 1N-Salzsäurelösung (ca. 180 ml) gegossen, der ausgefällte Feststoff wurde abfiltriert und mit Wasser gewaschen und luftgetrocknet, wobei 5-(2-Phenylethyliden)-2,4-imidazolidindion (2,74 g, 81%) erhalten wurde, F. 180-186 DEG C, 2:1-Mischung der (Z):(E)-Isomere.

5-[1-(Phenylmethyl)-4-piperidinyliden]-2,4-imidazolidindion
Lithiumhydroxid-monohydrat (0,288 g, 7 mmol) wurde in Wasser verdünnt (10 ml). Die Lösung wurde mit Ethanol (20 ml) verdünnt und Diethyl-2,4-dioxoimidazolidin-5-phosphonat (1,62 g, 7 mmol) wurde zugegeben. Nach 5 min wurde 1-Phenylmethyl-4-piperidon (1,00 g, 5 mmol) zugegeben und die Mischung bei Zimmertemperatur während 21 h gerührt. Die Reaktionsmischung wurde mit Wasser (etwa 30 ml) und Essigsäure (etwa 10 ml) verdünnt, im Vakuum konzentriert und die Mischung wurde mit gesättigter Natriumcarbonatlösung verdünnt, wobei ein weisser Feststoff ausgefällt wurde, welcher abfiltriert, luftgetrocknet und auf wässrigem Methanol umkristallisiert wurde, wobei 5-[1-(Phenylmethyl)-4-piperidinyliden]-2,4-imidazolidindion (1,20 g, 83%) erhalten wurde, F. 243-245 DEG C (Zersetzung).

5-(2-Oxo-2-phenylethyliden)-2,4-imidazolidindion-hydrat (BMY K-21588-3)
Natrium (0,393 g, 0,017 Grammatom) wurde in Ethanol (0,25 ml) aufgelöst und Diethyl-2,4-dioxoimidazolidin-5-phosphonat (4,04 g, 17 mmol) zugegeben. Nach 5 min wurde Phenylglyoxal-monohydrat (2 g, 13 mmol) als Feststoff in einer Portion zugegeben. Nahezu unmittelbar wurde ein gelber Niederschlag ausgeschieden. Die Mischung wurde während 30 min verrührt, mit Wasser verdünnt, filtriert und der Feststoff mit Wasser gewaschen. Die Ansäuerung der Mutterlauge ergab eine zweite Ausbeute, welche mit der ersten Ausbeute kombiniert und luftgetrocknet wurde, wobei 5-(2-Oxo-2-phenylethyliden)-2,4-imidazolidindion-hydrat (2,85 g, 100%) erhalten wurde; F. 264-266 DEG C (Zersetzung). <1>H-NMR (DMSO-d6) delta 6,85 (1H, s, Vinyl H), 7,50 bis 7,80 (4H, m, aromatisches H) und 8,08 (1H, d, J=6,5Hz aromatisches H). lR (KBr) 1675, 1740 und 1785 (C=O) cm<-><1> . m/e 217 (MH<+> ).

The present invention relates to a process for the preparation of C-5 ethylenically unsaturated hydantoin derivatives which are useful as intermediates in the synthesis of 1,3-dihydro-2H-imidazo [4,5-b] quinolin-2-ones, which are potent antithrombotic agents.
The condensation of hydantoin (ice) with aromatic aldehydes (2) for the production of C-5-unsaturated hydantoin derivatives (3) is described in the literature: E. Ware, Chemical Reviews, 1950, 46, pages 403-470; H.L. Wheeler et al., Am. Chem. J., 1911, 45, pages 368-383 and US-A 4,345,042 and US-A 4,582,903. This reaction is illustrated as follows:

wherein R represents hydrogen or a substituent. The adducts (3) are of synthetic use as precursors for derivatives of phenylalanine and phenylpyruvic acid. A number of different catalysts and solvent systems have been described to accomplish this conversion. In general, the aldehydes, the hydantoin and the catalyst are heated in a suitable solvent to a temperature in the range from 70 to 170 ° C. for 0.5 to 24 hours. Some of the most commonly used conditions are as follows: NaOAc / AcOH at 160 to 170 ° C for 3 h; in NH4OAc / AcOH under reflux for 4 h; in NaOAc / Ac2O under reflux for 1 h; in piperidine at 130 to 140 ° C. for 0.5 h; and in dimethylamine at 100 ° C. for 22 hours in a closed reaction vessel.
However, these processes are not entirely satisfactory, namely that some aldehydes are not reacted with hydantoin or only form products in low yield. Simple ketones have not been reported to be used in this reaction.
The reaction of phosphonate carbanions with ketones and aldehydes is generally known: Wadsworth et al., J. Amer. Chem. Soc., 1961, 83, pages 1733-1738; Wadsworth, Org. Reactions, 1978, 25, 73-253.
The present invention provides the process defined in claim 1 for the preparation of C-5-ethylenically unsaturated hydantoin derivatives which are used as intermediates in the synthesis of 1,3-dihydro-2H-imidazo [4,5-b] quinoline 2-ones are useful, as described in the following parallel U.S. patent applications: Serial No. 726,869 filed April 25, 1985; Serial No. 832,212, filed February 26, 1986; Serial No. 866,813 filed on May 23, 1986; and serial no. 883,258 filed on July 8, 1986. The 5-ethylenically unsaturated hydantoin derivatives which can be prepared by the process of the invention are also useful as intermediates in the preparation of other compounds, e.g. of phenylalanine and phenylpyruvic acid.
The dialkyl-5-phos-phonathydantoin used in the process according to the invention preferably has the following formula:

wherein R1 and R2 are hydrogen, alkyl or aralkyl with up to 10 carbon atoms, e.g. Benzyl, phenethyl, phenylpropyl and R3 is lower alkyl, such as methyl, ethyl or propyl.
The invention also provides the new compound, diethyl 1-methyl-2,4-dioxoimidazolidine-5-phosphonate. The corresponding diethyl 2,4-dioxoimidazolidine 5-phosphonate is described in the literature in Chem. Abstracts, 1985, 102, No. 204 029a.
The phosphonates of the formula I are prepared by bromination of an appropriate R1- and R2-imidazolidine-2,4-dione in acetic acid, the corresponding 5-bromohydantoin being obtained and the product then being reacted with trialkyl phosphite.
Any aliphatic or aromatic aldehyde or ketone or any alpha-dicarbonyl compound can be used in the process according to the invention. The aromatic aldehydes disclosed in US Pat. No. 4,345,072 and the aromatic and aliphatic aldehydes disclosed in US Pat. No. 4,582,903 can be used in the process according to the invention. The aromatic aldehydes described in US-A 4,345,072 include benzaldehyde, tolylaldehyde, 4-isopropylbenzaldehyde, 4-hydroxybenzaldehyde, 3,4,5-trimethoxybenzaldehyde, 3-bromo-4-methoxybenzaldehyde, 3,4-methylenedioxybenzaldehyde, 2-hydroxy -4-nitrobenzaldehyde, 4,5-dimethoxy-2-nitrobenzaldehyde, salicylaldehyde, vanillin, 4-phenylbenzaldehyde, 4-benzylbenzaldehyde, 4-fluorobenzaldehyde, 4-dimethylaminobenzaldehyde, 4-acetoxybenzaldehyde, 4-acetaminobenzaldehyde, 4-methylthiobaldehyde -Dichlor-4-hydroxybenzaldehyde.

A large number of aliphatic and aromatic aldehydes and ketones and alpha-dicarbonyl compounds useful in the practice of the present invention are illustrated in the following examples. A preferred class of aldehydes has the formula RCHO, where R is selected from the group:
a) substituted or unsubstituted alkyl, in which the alkyl group has 2-6 carbon atoms,
b) substituted or unsubstituted alkenyl, in which the alkenyl group has 3-5 carbon atoms,
c) substituted or unsubstituted phenyl, in which one or more substituents of phenyl are selected from the group halogen, hydroxy, alkoxy or substituted alkoxy, in which alkoxy has 1-6 carbon atoms, nitro, alkyl with 1-6 carbon atoms and substituted or unsubstituted amino, 2- (2-methyldioxalano), 1-piperidino, 1-morpholino, 1-pyrrolidino or 1-piperazino,
d) thienyl,
e) pyridinyl and
f) PhC (O).
The substituents which may be present on the alkoxy group include mono- and dialkylamino groups which have 1-6 carbon atoms, an arylsulfonyl group with 6-10 carbon atoms, a carboxylic ester group with 2-6 carbon atoms and a nitrogen-containing heterocyclic group with 1-4 nitrogen atoms , which itself may be further substituted with cycloalkyl groups containing 6-10 carbon atoms. The substituents which may be present on the amino groups include mono- and dialkyl groups containing 1-6 carbon atoms, alkylcarbonyl groups containing 2-6 carbon atoms and carboxyl ester groups containing 2-6 carbon atoms.
The substituents which may be present on the 1-piperazine group include an aroyl group which has 6-10 carbon atoms and may optionally be substituted with one or more alkoxy groups with 1-6 carbon atoms or carboxylic acid ester groups with 2-6 carbon atoms. The substituents optionally present on the 1-piperidino group include carboxylic ester groups with 2-6 carbon atoms and N-cycloalkyl, N-alkylcarboxamide with 7-10 carbon atoms.
Preferred ketones are selected from the following group: C1-3-alkyl-2-ketophenylacetate, C1-3-alkyl-2-keto C1-5-alkyl acetate, substituted or unsubstituted C4-8 cycloalkyl ketone, in which one or more substituents are selected from: halogen, hydroxy, alkoxy and substituted alkoxy, in which the alkoxy group has 1-6 C atoms, and alkyl with 1-6 carbon atoms, 1,2-dioxocyclohexane, substituted or unsubstituted C8-10 alkylphenones, in which the substituted phenyl is a Carries substituents, which is selected from the group: halogen, hydroxy, C1-6-alkoxy or C1-6-alkyl, 3-oxoindole and N-C1-4-alkyl-3-oxoindole. Although certain preferred classes of aldehydes and ketones have been defined, it is possible that all aliphatic or aromatic aldehydes or ketones or alpha-dicarbonyl compounds can be used in the process according to the invention.
Preferably 0.5 to 1.5 moles of an aliphatic or aromatic aldehyde or ketone or an alpha-dicarbonyl compound per mole of phosphonate are reacted at a temperature of 0 to 50 ° C. under basic conditions for 15 minutes to 24 hours. The reaction conditions described below are more preferred.
The addition of aromatic aldehydes to a slight excess of phosphonates and sodium ethoxide in ethanol at room temperature generally leads to a rapid excretion of 5-benzylidine hydantoin derivatives in an excellent yield. Since the phosphonate starting materials and the diethylphosphate by-products are water-soluble, the isolation of the product simply consists of a dilution with water or 2N hydrochloric acid solution and water with subsequent filtration. In some cases, prior removal of the solvent will facilitate this procedure. The crude products isolated from the reaction mixture are generally of high purity, as can be concluded from the NMR spectral data, although a mixture of geometric isomers is often obtained.
Regardless of the steric or electronic nature of the aldehydes, high yields are obtained, although sterically hindered and deactivated aldehydes are reacted with a somewhat slower reaction rate. The mild reaction conditions are expedient for a wide range of functional groups. The anhydrous conditions described above are not critical to the success of the reaction. Aqueous sodium hydroxide or lithium hydroxide solutions in ethanol have an equivalent effect and a two-phase system which contains dichloromethane and aqueous sodium hydroxide with or without a phase transfer catalyst also leads to a high yield of adducts. Other suitable reaction conditions are, for example, triethanol in acetonitrile, triethylamine and lithium bromide in acetonitrile and lithium bromide in pyridine.
Aliphatic aldehydes react with hydantoinphosphonates to produce 5-alkylidene hydantoin derivatives in synthetically useful yields. Phenylacetaldehyde reacts exothermically with phosphonates, and excellent yields of adducts can be obtained in 20 min, although according to the literature phenylacetaldehyde and 4-methoxyphenylacetaldehyde are considered to be non-condensable with hydantoin under various conditions.
Alpha-dicarbonyl compounds generally react exothermically under the usual conditions, high yields of adducts being obtained.
Cyclohexanone reacts in the presence of lithium, in contrast to sodium, with the hydantoin phosphonates to form high yields. The lithium salts of triethylphosphonoacetate have been shown to be highly chelating and the lithium atom can play a similar role in the reaction of the hydantoinphosphonates with the ketones. Although a tightly bound lithium salt must be more stable and therefore less reactive, the increased stability of the salt may allow the coupling process to successfully compete with alternative pathways that are likely to be controlled primarily by proton transfer.
The C-5-unsaturated hydantoin derivatives prepared according to the invention can in some cases be isolated as mixtures of (Z) and (E) isomers according to the formulas below, in which R min represents a group which is derived from aliphatic or aromatic aldehydes and R1 and R2 are as defined above:

The geometric isomer in which R min is in the cis position with respect to the N-R2 group of the hydantoin ring is designated as (Z), while the isomer in which R min is in the trans position for N-R2 group is referred to as (E). If a vinyl proton is present, signals in the lower region of the NMR spectrum are usually predominant. When the proton of the double bond is replaced by another residue, such as a group derived from the ketone or an alpha-dicarbonyl group, the (E) or (Z) designation is made on the basis of the Cahn-Wright-Prelog order of priority ( see Advanced Organic Chemistry, 2nd edition, Jerry Merch, McGraw-Hill, 1977, pages 113-117 and pages 99-101).
The following examples serve to illustrate the following invention. All temperatures are given in DEG C and the melting points, which were recorded with a Thomas Hoover capillary, are uncorrected. Common abbreviations were used to indicate the nuclear magnetic resonance spectral data, with tetramethylsilane being used as the internal reference, and the chemical shifts in ppm (parts per million). All new compounds gave satisfactory elemental analysis data and showed spectral characteristics that were in agreement with the assumed structures.

Diethyl 2,4-dioxoimidazolidine 5-phosphonate
A mixture of imidazolidine-2,4-dione (200 g, 2M) and acetic acid (800 ml) were heated to 85 ° C. in a \ l bath. Bromine (352 g, 112.8 ml, 2.2 M) and a small portion of bromine (& tilde & 5 ml) were added to a dropping funnel and the contents were added to the reaction mixture with vigorous stirring. Once the orange color disappeared, the rest of the bromine was quickly added over about 7 minutes to give a clear solution. After stirring at 85 ° C. for 30 minutes, the reaction mixture was cooled to 30 ° C. in an ice bath and triethyl phosphite (465 g, 479 ml, 2.8 M) was added at a rate such that the reaction temperature ranged from 45 to 50 DEG C stayed. Diethyl 1-methyl-2,4-dioxoimidazolidine-5-phosphonate
A mixture of 1-methylimidazolidine-2,4-dione (202.5 g, 1.8 M) and glacial acetic acid (1 l) was heated to 90 ° C. in a \ l bath. A dropping funnel was charged with bromine (311.5 g, 100 ml, 1.95 M) and a small portion of bromine was added to the reaction mixture. After the orange color disappeared, the rest of the bromine was added dropwise at a rate such that instant decolorization occurred. After the addition was complete, the mixture was heated to 90 ° C. for 60 min, cooled to room temperature and stirred overnight. Ethyl 4- [3 - [(2,4-dioxoimidazolidin-5-yl-idene) methyl] -4-nitrophenoxy] butanoate
Sodium (4.92 g, 0.21 gram atom) was dissolved in absolute ethanol (600 ml) and diethyl 2,4-dioxoimidazolidine 5-phosphonate (50.5 g, 0.21 mol) was added. Sodium (0.386 g, 0.017 gram atom) was dissolved in ethanol (70 ml) and diethyl 2,4-dioxoimidazolidine 5-phosphonate (3.96 g, 17 mmol) was added. A mixture of 5-hydroxy-2-nitrobenzaldehyde (8.18 g, 49 mmol), 1-bromo-3- (tetrahydro-2H-pyran-2-yl) oxypropane (11.50 g, 51 mmol), potassium carbonate ( 7.16 g, 51 mmol), potassium iodide (catalytic amount) and dimethylformamide (800 ml) were heated to 110 ° C. with stirring for 30 min. The mixture was cooled, diluted with water (150 ml) and extracted with dichloroethane. The combined extracts were washed with water (three times), dried over sodium sulfate and concentrated in vacuo to leave a residue which was dissolved in ethanol (15 ml) and a solution of sodium ethoxide (3.99 g, 58 mmol) and Diethyl 2,4-dioxoimidazolidine 5-phosphonate (13.87 g, 59 mmol) in ethanol (200 ml) was added. After 90 min the ethanol was evaporated and the residue diluted with water and extracted with dichloromethane.
The combined extracts were dried over sodium sulfate and concentrated in vacuo to give one which was dissolved in dichloromethane and filtered through silica gel plugs using diethyl ether as the eluent. Evaporation of the solvent gave 5 - [[2-nitro-5 - [(3-tetrahydro-2H-pyran-2-yl] propoxy] phenyl] methylene] -2,4-imidazolidinedione as a viscous oil, which was obtained without further purification An analytical sample of 2,4-imidazolidine as the partial hydrate was prepared by precipitation from dichloromethane with hexane and had a melting point of 128-134 ° C.

A mixture of 5-hydroxy-2-nitrobenzaldehyde (1.00 g, 6 mmol), 2-bromoethyl ethyl ether (1.00 g, 0.74 ml, 6.5 mmol), powdered potassium carbonate (0.91 g, 6.5 mmol), potassium iodide (catalytic amount) and dimethylformamide (10 ml) were heated to 110 ° C. with stirring. After 30 min the mixture was cooled, diluted with water and extracted with dichloromethane (three times). The combined extracts were washed with water (twice), dried over sodium thiosulfate and concentrated in vacuo to give a \ l, which was diluted with ethanol (3 ml) and to a stirred solution of sodium ethoxide (0.53 g, 78 mmol) and diethyl 2,4-dioxoimidazolidine 5-phosphonate (1.83 g, 77 mmol) in ethanol (15 ml).
After 10 min the mixture was diluted with 2N hydrochloric acid solution and the solid filtered off and air dried to give 5 - [[5- (2-ethoxyethoxy) -2-nitrophenyl] methylene] -2,4-imidazolidinedione as a partial hydrate (1.40 g, 73%), mp 228-233 ° C (decomposed).
Anal. Ber. for C14H15N3O6 0.12H2O: C, 51.99; H, 4.75; H, 13.00; H2O, 0.67. Found: C, 51.59; H, 4.72; N, 12.82; H2O, 0.25. A second crop (0.2 g, 10%) was then collected.
Example 23
5 - [[5- [3- (2-Methyl-1,3-dioxolan-2-yl) propoxy] -2-nitrophenyl] methylene] 2,4-imidazolidinedione
A mixture of 5-hydroxy-2-nitrobenzaldehyde (20.00 g, 0.12 mol), 5-chloro-2-pentanone ethylene ketal (21.7 g, 0.132 mol), potassium carbonate (20.00 g, 14 mol), potassium iodide (0.5 g) and dimethylformamide (200 ml) were heated to 120 ° C. with stirring. After 4 min the mixture was cooled, diluted with water and extracted with diethyl ether. The combined ethereal extracts were washed with water, dried over magnesium sulfate and the solvent was evaporated. The remaining \ l contained 5- [3- (2-methyl-1,3-dioxolan-2-yl) propoxy] -2-nitrobenzaldehyde (used without purification), was dissolved in ethanol (200 ml) and in one portion added to a solution of sodium ethoxide (10.05 g, 0.15 mol) and diethyl 2,4-dioxoimidazolidine 5-phosphonate (35.00 g, 0.15 mol) in ethanol (300 ml).
The mixture was stirred at room temperature for 90 min, concentrated to a volume of about 90 ml and diluted with water. The yellow precipitate was filtered off, washed with water and dried in vacuo at 70 ° C., 5 - [[5- [3- (2-methyl-1,3-dioxolan-2-yl) propoxy] -2-nitrophenyl ] methylene] -2,4-imidazolidinedione (38.17 g, 84%) was obtained. An analytical sample was obtained by recrystallization from ethanol and had a melting point of 175-180 ° C.

The compound was obtained by reacting 2-nitro-5-3- (phenylsulfonyl) propoxy benzaldehyde (obtained by alkylating 5-hydroxy-2-nitrobenzaldehyde with 3-phenylsulfonylpropyl bromide) with diethyl 2,4-dioxoimidazolidine-5-phosphonate in a manner analogous to that obtained in the processes described in Example 17 and had a melting point of 125-157 ° C.

Diethyl 2,4-dioxoimidazolidine 5-phosphonate was added to triethylamine (equimolar) in acetonitrile. The compound was obtained by reacting diethyl 2,4-dioxoimidazolidine 5-phosphonate with ethyl 4- [3-formyl-4-nitrophenoxy] propyl acetate (obtained by alkylation with 5-hydroxy-2-nitrobenzaldehyde with 3- Bromopropyl acetate) in a manner analogous to that described in Example 26, F. 94-130 ° C.

Example 30
5 - [[3-Nitro-5 - [(1-piperidinyl) phenyl] methylene] -2,4-imidazolidinedione
Sodium (0.6 g, 0.026 gram atom) was dissolved in absolute ethanol (200 ml) and diethyl 2,4-dioxoimidazolidine 5-phosphonate (6.0 g, 25 mmol) was added. After 10 min, 2-nitro-5- (1-piperidinyl) benzaldehyde (5.0 g, 21 mmol) was added in one portion and the mixture was stirred at room temperature for 5 h. Example 43
5 - [(2-nitrophenyl) methylene] -2,4-imidazolidinedione
Lithium hydroxide monohydrate (0.334 g, 8 mmol) was diluted in water (10 ml) and the solution was diluted with ethanol (20 ml). Diethyl 2,4-dioxoimidazolidine 5-phosphonate (1.875 g, 8 mmol) was added and the mixture was stirred for 5 min before adding solid 2-nitrobenzaldehyde (1 g, 6.6 mmol). A heavy yellow precipitate was formed within a few minutes. After 15 min the mixture was diluted with 1N hydrochloric acid solution and the solid washed with water and air dried using a 2.5: 1 mixture of (Z) - and (E) -5 - [(2-nitrophenyl) methylene] -2 , 4-imidazolidinedione was obtained; Yield 1.48 g, 96%, mp 300-302 ° C (decomposition).

5- (phenylmethylene) -2,4-imidazolidinedione
Benzaldehyde (0.5 g, 5 mmol) in dichloromethane (10 ml) became a solution of diethyl 2,4-dioxoimidazolidine 5-phosphonate (1.45 g, 6 mmol) and 4N sodium hydroxide solution (0.15 ml ) in water (10 ml). Sodium (0.393 g, 0.017 gram atom) was dissolved in ethanol (0.25 ml) and diethyl 2,4-dioxoimidazolidine 5-phosphonate (4.04 g, 17 mmol) was added.
