Beschreibung description
In biological and biochemical laboratory technology, especially in genetic research, one is often faced with the task of using a preferably aqueous and buffered suspension of various cell components, such as is obtained, for example, by mechanical or enzymatic disintegration of tissues or from cell cultures. to sort out and isolate certain components. In particular, this involves sorting out protoplasts and cell organelles, e.g. Cell nuclei, mitochondria or chloroplasts.
Such sorting and isolation tasks have so far been based on relatively lengthy and complex methods, such as centrifugation by means of a density gradient. The usual working method with sucrose solutions requires e.g. a refrigerated centrifuge; in addition, the osmotic influences due to the variable sugar concentration are often not beneficial to the cell components, and there is always the risk of enzymatic contamination.
For example, also classifiers that optically and / or electronically assess the components according to their size or other criteria and sort them out automatically, e.g. using a switch built into a liquid flow. If the components are only classified based on their size, the easiest way to sort them out is to use a filter-like filter. By vertically stacking screen elements of different hole sizes, several components can be separated and sorted out in one work step.
As sieve elements for biological components, e.g. individual cells, protoplasts, and organelles, such as cell nuclei, chloroplasts or mitochondria, are excellently suitable for gauze tissue, such as those e.g. are known from the milling industry. Such fabrics are made in various mesh sizes between 1 and 500 µm from various natural and synthetic fibers, e.g. made of cotton, natural silk or preferably synthetic monofilament fibers made of polyamides, polypropylene and polyesters, and offered commercially. The synthetic gauze fabrics in particular are mechanically stable, water-resistant, easy to clean and can be easily sterilized using the methods customary in biology, e.g. in an autoclave at 120 ° C and 1 atm pressure.
The present invention relates to a filtering device designed for the biological-genetic laboratory, in particular for the separation of cell components, which can be assembled from similar module-like elements to form a column with any number of stages. The module-like elements each consist of two hollow cylindrical pieces, which are put together with the interposition of a piece of filter fabric.
By plugging together a number of such two-part elements, a filtration column of any size can be built. The individual elements are preferably fitted with filter fabrics of decreasing mesh size from top to bottom. With this e.g. a suspension of biological particles is • separated into fractions of different particle sizes.
A particular advantage of the device according to the invention is that the pieces of filter fabric do not have to be specially cut. A simple, e.g. square section is sufficient; Due to the special design features of the individual elements, the filter fabric is stretched smoothly and wrinkle-free over the open cross-section when the parts are put together. The part of the fabric that is not required protrudes freely from the place where the elements are put together. Since the fabric also receives sufficient tension when it is put together, support by perforated plates, wire mesh, glass frits or the like is not necessary. Problems due to filter leakage or clogging are also largely avoided.
Another advantage, which mainly affects the time and preparation costs, is that the assembled device including the clamped filter tissue as a whole according to the methods used in biology under heat and steam, e.g. can be sterilized as often as required in an autoclave at 120 ° C and 1 atm pressure. The relatively costly pre-sterilized disposable filter can therefore be dispensed with thanks to the device according to the invention.
Der Aufbau des Geräts ist aus den Figuren 1 bis 4 ersichtlich: The structure of the device can be seen from Figures 1 to 4:

The lower short cylinder piece (2), with an inner diameter that is slightly larger than the outer diameter at the lower edge of the larger cylinder piece (1), has an annular bead (4) on its inner surface below its upper edge, through which the filter fabric at Put together tight and held in place. The outer diameter of the piece (2) is tapered in a step-like manner at its lower end (10) and fits into the inner diameter of another piece (1) when several filter elements are plugged together, so that filter columns of any length can be constructed from several elements.
In Fig. 3 two plate-shaped end pieces (5) and (7) are shown, each with a central hole into which the cylindrical piece (2) can be inserted with its lower end. The outer edge (11) of the two end pieces is slightly cut so that the plates can be placed on a petri dish (6), (8).
In Fig. 4, the cylindrical pieces (1) and (2) and the two end plates (5) and (7) are shown in section, whereby the bead (4) and the diameter ratios of the pluggable pieces are clearly visible.
In a special embodiment of the invention, which is not shown in the figures, in the uppermost filter element above the uppermost filter fabric, a plate essentially parallel to the filter can be arranged, e.g. is caused to vibrate vertically by an AC magnet. Such a vibrating plate facilitates and accelerates the filtration in many cases, especially wherever very fine filters are used, or with materials that tend to form a compact, clogging layer on the filter.

The suspension obtained in this way, in addition to undegraded leaf constituents and cells with undigested cell walls, a large number of protoplasts, i.e. cells freed from their cell wall were added to the filtration column. After the filtration, mostly remaining aggregates from the plasmolysis as well as some larger protoplasts were found in the uppermost stage and protoplasts were found almost exclusively in the petri dish serving as the collecting vessel. The protoplasts prepared and isolated using this method were at least 95% fully viable and capable of division.

0.1 mmol N, N'-bis (3-aminopropyl-1,4-diaminobutane (spermine) cut into small pieces at 40 ° C. and then shaken gently for 5 minutes.
The suspension was then placed in a step filtration device according to the invention, which was equipped in four steps with different nylon fabrics, with mesh sizes, counting from top to bottom, of 100, 40, 20 and 10 μm. The fragments of leaf tissue created during disintegration collected in the various filtration stages of the device, while a suspension containing the cell nuclei and smaller organelles such as mitochondria and chloropiaes left the filter column downwards. The cell nuclei could finally be separated from the smaller organelles by a further filtration stage with a nylon fabric of 5 µm mesh size.
By washing the finest nylon mesh with a small volume of the buffer solution described above, an almost exclusively suspension containing cell nuclei could finally be obtained and made available for cell biological, genetic and biochemical investigations.
