Description
BESCHREIBUNG DESCRIPTION
The present invention relates to a stable pharmaceutical preparation containing a granulocyte colony stimulating factor. In particular, the present invention relates to a stabilized pharmaceutical preparation containing a granulocyte colony stimulating factor which is protected against the loss or inactivation of the active component (ie granulocyte colony stimulating factor) due to adsorption on the wall of a container in which the Preparation has been given, or due to the association, polymerization or oxidation of the component mentioned.
Chemotherapy has been used as a method for treating a variety of infectious diseases, but it has recently been found that chemotherapy causes some serious clinical problems, such as the production of drug-resistant organisms, alteration in the causative organisms, and high side effects. In order to avoid these problems associated with chemotherapy, including the use of therapeutic agents such as antibiotics and bactericides, efforts have been made to use a substance which activates the host's prophylactic abilities against an organism causing infection and thereby provides a complete solution to the chemotherapy problems mentioned above. From the variety of host prophylactic abilities, it is believed that the phagocytic bactericidal activity of leucocytes has the greatest impact on the initial period of bacterial infection, and it has therefore been believed that it is important to promote the host's infection protective abilities Promoting the growth of neutrophils and their differentiation in the mature stage. Granulocyte colony stimulating factor (G-CSF) is one of the very useful substances showing such effects, and the applicants of the present invention have previously filed a patent application for an anti-infection agent using G-CFS (Japanese Patent Application No. 23,777 / 1985).
As mentioned above, chemotherapy as it is currently practiced involves various unavoidable problems and intensive efforts are made to use a drug substance which5

che fähig ist zur Aktivierung der prophylaktischen Funktionen des Wirtes oder der Person, welche infisziert worden ist. is capable of activating the prophylactic functions of the host or the person who has been infected.
Needless to say, G-CSF itself has the ability to activate the host's prophylactic functions, and it has also been found that G-CSF exhibits greater therapeutic effects in clinical applications when used in combination with a substance, which activates the prophylactic abilities of the host.
G-CSF is used in a very small amount and a pharmaceutical preparation containing 0.1-100 (ig (preferably 5-50 (ig) of G-CSF) is usually administered at a dose of 1-7 times a week to an adult, however, G-CSF tends to be absorbed on the wall of a container such as an ampoule for injection or a syringe when the drug is used as an injection in such a form as an aqueous solution , it is therefore adsorbed onto the wall of the container, such as an ampoule or a syringe, resulting in either G-CSF failing to fully develop its activity as a pharmaceutical agent, or it incorporates G-CSF in an amount more than necessary, taking into account the possible loss due to adsorption.
G-CSF is also unstable and highly sensitive to environmental factors such as temperature, humidity, oxygen and ultraviolet rays. Through the action of such factors, G-CSF undergoes physical or chemical changes, such as association, polymerization and oxidation, and suffers a great loss of activity. These phenomena make it difficult to ensure complete fulfillment of a therapeutic act by administering a very small amount of G-CSF in a very precise manner.
It is therefore necessary to develop a stable pharmaceutical preparation of G-CSF which is completely protected against a drop in the activity of its active component. This is the main object of the present invention, which provides a stable pharmaceutical preparation of G-CSF.
The inventors of the present invention conducted intensive studies to favor the stability of a pharmaceutical preparation containing G-CSF, and found that this object can be achieved effectively by adding a pharmaceutically acceptable surfactant, saccharide, protein or high molecular weight compound.
Accordingly, the stable G-CSF-containing pharmaceutical preparation of the present invention is characterized in that it contains both G-CSF and at least one substance selected from the group of a pharmaceutically acceptable surface agent, saccharides, protein and high molecular weight compound.
The G-CSF contained in the pharmaceutical preparation of the present invention can be obtained by any method such as those described in the descriptions of Japanese Patent Application Nos. 153 273/1984, 169 455/1985, 269 456 / 1985, 270 838/1985 and 270 839/1985. For example, a human G-CSF can be produced either by culturing a cell strain (CNCM deposit number 1-315 or 1-483) collected from tumor cells from patients with oral cavity cancer (oral cavity cancer) or by input (expressing) a recombined DNA (which has been produced by the action of a human G-CSF encoding gene [encoding gene]) in a suitable host cell (eg E. coli, C 127 cell or Chinese hamster ovary cells).
Some human G-CSF. which has been highly purified can be used as G-CSF to contain 20 in the pharmaceutical preparation of the present invention. Preferably, the human G-CSFs are those obtained by isolation from the floating part of the culture of a human G-CSF producing cell, and a polypeptide or glycoprotein which has human G-CSF activity by transforming a host with a recombined vector that has incorporated therein a gene that co-30s a polypeptide that has human G-CSF activity.

For details of the process for producing these two types of G-CSF, see the descriptions of Japanese Patent Application Nos. 153 273/1984, 269 455/1985, 269 456/1985, 270 838/1985 and 270 839/1985, all of which have been filed by the applicants of the present invention.
Another method that can be used is to merge a G-CSF producing cell with a self-proliferating malignant tumor cell and cultivate the resulting hybridoma in the presence or absence of mytogen.
The resulting solution, which contains human G-CSF, can be stored in a frozen state after being further purified and concentrated, if necessary, by known techniques. Alternatively, the solution can be stored after it has been dehydrated, for example by means of freeze drying.
All of the human G-CSFs thus produced can be processed as indicated in the present invention to obtain stable pharmaceutical preparations containing G-CSF.

The stabilized G-CSF pharmaceutical preparation of the present invention may be formulated for either oral or parenteral administration, such as by injection, used in various ways, and a variety of modes of administration may be used, depending on the specific one Method of administration. Typical dosage forms include: those intended for oral administration, such as tablets, pills, capsules, granules and suspensions; Solutions, suspensions and freeze-dried preparations mainly intended for intravenous injection, intramuscular injection, subcutaneous injection and intracutaneous injection; and those intended for transmucosal administration, such as rectal suppositories, nasal drugs, and vaginal suppositories.
According to the present invention, at least one substance selected from the group consisting of a surface agent, a saccharide, a protein or a high molecular weight compound is added to a pharmaceutical preparation containing G-CSF so as to prevent it from being attached to the wall of its container or a syringe while remaining stable over an extended period of time.
The detailed mechanism by which the substances mentioned above stabilize G-CSF or prevent it from being absorbed has yet to be clarified. In the presence of a surface agent, the surface of the G-CSF, which is a hydrophobic protein, would be covered with the surface agent to be solubilized, so that the G-CSF, which is present in a trace amount, is effectively prevented from doing so Wall of his container or a syringe to be absorbed. A saccharide or a hydrophilic, high molecular weight compound would form a hydrated layer between G-CSF and the adsorptive surface of the wall of the container or a syringe, preventing the adsorption of G-CSF in an effective manner. A protein would compete with the G-CSF for adsorption on the wall of its container or syringe, effectively preventing the adsorption of G-CSF.
In addition to preventing G-CSF adsorption, the substances mentioned above also help to prevent the association or polymerization of the G-CSF molecules. In the presence of a surfactant, saccharide, protein, or high molecular weight compound, the individual molecules of G-CSF are dispersed in these substances and the interaction between the G-CSF molecules is reduced enough to significantly decrease the likelihood of them Association or polymerization to effect. In addition, these substances delay the autoxidation of G-CSF, which is accelerated at high temperatures or humidity, or prevent G-CSF from being associated or polymerized as a result of the autoxidation. These effects of delaying the autoxidation of G-CSF or preventing association or polymerization are further enhanced by the addition of an amino acid, a sulfur reducing agent, or an antioxidant.
The problems described above occur specifically in solutions for injections and in suspensions, but they also occur during the process of formulating G-CSF in other dosage forms, such as tablets. The addition of surface agents, saccharides, proteins or high molecular weight compounds is also effective in the latter case.
By adding at least one substance selected from the group consisting of a surfactant, saccharide, protein and a high molecular weight compound, G-CSF is strongly stabilized and retains its activity for an extended period of time, as shown in the following examples . To achieve these results, the amount of each of these substances, especially their lower limit, is critical and the following ranges are desirable: 1 - 10,000 parts by weight of surface agent, 1 - 10,000 parts by weight of saccharide, 1 - 20,000 parts by weight of protein, and 1-20,000 parts by weight of high molecular weight compound, per 1 part by weight of G-CSF.
According to the present invention, a surfactant, a saccharide, a protein and / or a high molecular weight compound is used in a specified concentration, and this is not only effective in preventing G-CSF from being absorbed on the wall of its container or a syringe , but also in favor of the stability of a pharmaceutical preparation containing G-CSF. As a result, it becomes possible to ensure the administration of a small but highly precise dose of G-CSF to patients. Because G-CSF is expensive, its efficient use leads to low costs in the manufacture of pharmaceutical preparations containing G-CSF.
The following examples are for the purpose of further illustrating the present invention and are not to be considered as limiting in any sense. To 5 pg of G-CSF, one of the stabilizing agents listed in Table 1 was added and the mixture was aseptically dissolved in a 20 mM buffer solution (containing 100 mM sodium chloride; pH 7.4) to prepare a pharmaceutical preparation , containing 5 \ xg G-20 CSF per ml, which was then freeze-dried. To 10 ig G-CSF was added one of the stabilizing agents listed in Table 3, and the mixture was dissolved in a 20 mM phosphate buffer solution (containing 100 mM sodium chloride; pH 7.4) to prepare a pharmaceutical preparation containing 10 hours of G-CSF per ml. One milliliter of the preparation was placed in a sulfa-treated, silicone-coated glass vial and left at a temperature of 4 CC. The effectiveness of that stabilizing agent in preventing G-CSF adsorption was evaluated by measuring the remaining activity of G-CSF in the solution after 0.5, 2 and 24 hours. The measurement was carried out according to method (b) using reverse phase HPLC (high-performance liquid chromatography). The results are shown in Table 3.
