5. Verfahren zur quantitativen Bestimmung des Enzyms Cl-Esterase in einem Medium, welches das genannte Enzym enthält oder in welchem das letztere entsteht oder verbraucht wird, dadurch gekennzeichnet, dass man das genannte Medium mit einem Peptidderivat gemäss Patentanspruch 1 zur Reaktion bringt und die Menge des durch die katalytische hydrolytische Wirkung des genannten Enzyms auf das Peptidderivat pro Zeiteinheit freigesetzten Spaltproduktes Rl-H durch photometrische spektrophotometrische, fluoreszenzspektrophotometrische oder elektrochemische Methoden misst.
Das menschliche Blut enthält einen unter dem Namen Cl-Esteraseproenzym bekannten Wirkstoff, der unter der Einwirkung der Verbindung eines Antikörpers mit einem Antigen zu dem aktiven Enzym Cl-Esterase aktiviert wird.
Dieses Enzym aktiviert kaskadenartig weitere Proenzyme zu aktiven Enzymen im Komplementsystem. Diese aktivierten Enzyme lysieren ihrerseits Zellmembranen von Bakterien oder abgestorbenen Erythrozyten und spielen somit eine wichtige Rolle bei der immunologischen Abwehr. Das Plasma enthält auch einen wichtigen Inhibitor, der die Cl-Este- rase hemmt und den Namen Cl-Esteraseinhibitor trägt. Bei inflammatorischen Prozessen wird C1-Esterase aktiviert, wobei je nach dem Cl-Esteraseinhibitorspiegel des Blutes das Komplementsystem schneller oder langsamer aktiviert wird.
Es ist vom klinischen Standpunkt wünschenswert, in solchen Fällen sowohl den Cl-Esterasespiegel als auch den Cl-Este- raseinhibitorspiegel im Blut zu bestimmen. Diese Bestimmungen werden gegenwärtig nach umständlichen und wenig genauen immunologischen und titrimetrischen Methoden durchgeführt [siehe W.J. Canady et al., Immunochemistry 1976, Bd. 13, 229-233, und D. Ogston et al., Thrombosis Research, Bd. 9, 217-222 (1976)1.
Es wurde nun gefunden, dass die Bestimmung der C3- Esterase wesentlich rascher und genauer durchgeführt werden kann, wenn man als Substrate gewisse einfache Peptidderivate, welche Gegenstand der vorliegenden Erfindung sind, verwendet.

Die nachfolgende Tabelle enthält Zahlenangaben, welche die Spaltbarkeit einiger erfindungsgemässer Peptidderivate durch Cl-Esterase betreffen. Die angegebenen Werte wurden wie folgt ermittelt: Einer Mischung von 1,8 ml Trisimidazolpuffer und 0,015 ml einer Lösung, die 800 Tosyltyrosinäthylester-Einheiten Cl-Esterase pro ml enthielt, wurde bei 37 C 0,2 ml einer 2 x 10-3-molaren Peptidderivatlösung zugesetzt.
Dann wurde die durch die Spaltung des Peptidderivates in der Mischung bewirkte Änderung der optischen Dichte tOD pro 5 Min. bei 405 nm gemessen.

Die Bestimmung des Cl-Esteraseinhibitorspiegels in Blutplasma kann wie folgt durchgeführt werden:
Eine Mischung von 1,6 ml Trisimidazolpuffer von pH 7,4, Ionenstärke 0,2, und 0,1 ml Citratplasma wird bei 37 C mit 0,1 ml gereinigter Cl-Esterase 4 Minuten inkubiert. Dem Inkubat wird 0,2 ml einer 2 x 10-3-molaren wässrigen Lösung des erfindungsgemässen Substrates zugesetzt.
Wenn das Substrat als chromogene Gruppe (Rl) eine p-Nitroanilinogruppe ist, wird die pro Minute freigesetzte Menge des Spaltproduktes p-Nitroanilin (Rl-H) bei 405 nm spektrophotometrisch gemessen. In einem Testsystem, das kein Plasma enthält, sonst aber gleich zusammengesetzt ist, wird die pro Minute freigesetzte Menge des p-Nitroanilins wie oben beschrieben gemessen. Aus der Differenz zwischen den beiden Messwerten wird der Cl-Esteraseinhibitorspiegel des Blutplasmas ermittelt.

The determination of the Cl-esterase inhibitor level in blood plasma can be carried out as follows:
A mixture of 1.6 ml of trisimidazole buffer of pH 7.4, ionic strength 0.2, and 0.1 ml of citrated plasma is incubated at 37 C with 0.1 ml of purified Cl esterase for 4 minutes. 0.2 ml of a 2 × 10-3 molar aqueous solution of the substrate according to the invention is added to the incubate.
If the substrate as a chromogenic group (Rl) is a p-nitroanilino group, the amount of the cleavage product p-nitroaniline (Rl-H) released per minute is measured spectrophotometrically at 405 nm. The amount of p-nitroaniline released per minute is measured in a test system which contains no plasma but is otherwise composed in the same way as described above. The Cl-esterase inhibitor level of the blood plasma is determined from the difference between the two measured values.
