Description
The present invention relates to a number of new compounds with inhibitory effects against the biosynthesis of cholesterol. These new compounds are derivatives of compounds referred to as ML-263A and MB-530A. The invention also relates to a process for the preparation of these compounds.
As is known, hyperlipemia, especially hypercholesteremia, is one of the main causes of cardiopathy, e.g. Myocardial infarction or arteriosclerosis. Accordingly, considerable attempts have been made to develop compounds which are capable of lowering the lipid level and in particular the cholesterol level in the blood. A group of compounds of this type is found in U.S. Patent No. 3,983,140 and has been isolated from microorganisms of the genus Pénicillium. This group of compounds is commonly referred to as ML-236.
Another such compound, designated Monacolin-K or MB-530B, is disclosed in U.S. Patent Applications No. 121,515 of February 14, 1980 and No. 137,821 of April 4, 1980. This connection can be established by cultivating microorganisms of the genus Monascus, in particular strains of Monascus ruber.
A number of new compounds have now been developed which are related to ML-236 and MB-530B and which develop better activities because they are easier to reabsorb by oral administration and are more accessible as precursors to drugs, i.e. as an active ingredient that is converted into an active or more active form in the body after administration by means of chemical or biochemical reactions.
ML-236A und ML-236B, zwei der in der US-PS Nr. 3 983 140 geoffenbarten Verbindungen des ML-236-Komplexes, entsprechen den beiden folgenden Formeln: ML-236A and ML-236B, two of the compounds of the ML-236 complex disclosed in U.S. Patent No. 3,983,140, correspond to the following two formulas:

The alkanoyl group of this phenylalkanoyl group is preferably an alkanoyl group having 2 to 4 carbon atoms and preferably an acetyl, propionyl or butyryl group, while the substituents are preferably alkyl groups with 1 to 4 carbon atoms, alkoxy groups with 1 to 4 carbon atoms, hydroxyl groups, methylenedioxy groups, halogen atoms or trifluoromethyl groups. These substituents can be in the ortho, meta or para positions. If two or more substituents are present, they can be the same or different substituents.
Examples of such araliphatic acyl groups are the cinnamoyl, chlorcinnamoyl, bromcinnamoyl, methoxy-cinnamoyl, methylcinnamoyl, phenylacetyl, phenoxy-acetyl and phenylpropionyl groups, the substituents present on the cinnamoyl groups in the ortho, meta or can stand para positions.
On the other hand, R1 and / or R2, although less preferred, may contain a heterocyclic acyl group, e.g. 2-furoyl, 2-thenoyl, 3-thenoyl, nicotinoyl or iso-nicotinoyl groups, or a heterocyclically substituted, aliphatic acyl group, e.g. the 2-thienylacetyl, 3-thie-nylacetyl, 2-furylacetyl, 3-furylacetyl, 2-thienylacryloyl, 3-thienylacryloyl, 2-furylacryloyl or 3-furylacryloyl groups, or an alicyclic acyl group. In the latter case, it is preferably a cyclo-propane carbonyl, cyclobutane carbonyl, cyclopentane carbonyl, cyclohexane carbonyl, cycloheptane carbonyl, cyclooctane carbonyl, cyclohexane acetyl, 3-cyclohexane propionyl, 4-cyclohexane butyryl carbonate or 1 -io group.

April 1979 unter No. FERM 4956 beim The Fermentation Research Institute, Agency of Industriai Science and Technology, Ministry of International Trade and Industry, Japan, und am 25. April 1980 beschrieben. MB-530A, one of the compounds according to the invention, can be produced by cultivating an MB-530A-producing microorganism of the genus Monascus, preferably a strain of Monascus ruber and particularly preferably of the strain Monascus ruber SANK 15 177. This strain was released on April 27, 1979 under No. FERM 4956 at The Fermentation Research Institute, Agency of Industriai Science and Technology, Ministry of International Trade and Industry, Japan, and on January 25, 1980 under No. NRRL12 081 deposited with the Agricultural Research Culture Collection, Northern Regional Research Center, Peoria, Illinois, USA. The morphology and physiology of this strain are described in greater detail in U.S. Patent Application Serial No. 137,821 of April 4, 1980.
The desired compound MB-530A can be obtained by culturing the selected microorganism in a culture broth under aerobic conditions, using the usual working methods for the cultivation of fungi and other microorganisms. For example, you can first grow the selected strain of Monascus on a suitable medium and then collect the microorganisms generated and inoculate and grow on another culture medium in order to obtain the desired MB-530A. The culture medium used for the propagation of the microorganism and the culture medium used for the production of MB-530A can be the same or different media.
Any culture media as is well known for the cultivation of mushrooms can be used for this purpose, provided that, as is also well known, they contain the necessary nutrients, in particular an assimilable carbon source and an assimilable nitrogen source. Examples of suitable sources of assimilable carbon are glucose, maltose, dextrin, starch, lactose, sucrose and glycerin. From these sources, glucose, glycerin and starch will preferably be used to produce MB-530A. Examples of suitable assimilable nitrogen sources are peptone, meat extract, yeast, yeast extract, soybean meal, peanut meal, corn steep liquor, rice bran and inorganic nitrogen sources. Among these nitrogen sources, corn steep liquor and peptone will be particularly preferred. When producing MB-530A, an inorganic salt and / or a metal salt can be added to the culture medium. A small amount of heavy metal can also be added if necessary.
The microorganism is preferably grown under aerobic conditions using cultivation methods well known from the literature, e.g. solid cultures, shake cultures or cultivation with aeration and shaking, grown. The microorganism grows within a wide temperature range of, for example, 7 to 40 ° C. For the production of MB-530A, a growth temperature in the range of 20 to 30 ° C will preferably be used.
During the growth of the microorganism, the production of MB-530A can be monitored by taking samples from the culture medium, measuring the physiological activity of the medium with known tests. The cultivation can then be carried out until a substantial accumulation of MB-530A has taken place in the culture medium, whereupon the MB-530A can be isolated and obtained from the culture medium and the tissues of the microorganism, using any suitable combination of isolation methods depending on its physical and chemical properties. One or all of the following isolation techniques can be used, namely extraction of the liquid from the culture broth with the aid of a hydrophilic solvent, e.g. Diethyl ether, ethyl acetate, chloroform or benzene, extraction of the organism with a hydrophilic solvent, e.g.

ML-236A, ML-236B, MB-530A or MB-530B is reacted with an acid chloride or acid anhydride which corresponds to the acyl derivative to be produced. The reaction is preferably carried out in the presence of a base which acts as an acid-binding agent, an organic amine, e.g. Pyridine, triethylamine, N, N-dimethylaminopyridine, N-methylpyrrolidine or N-methylmorpholine used. The reaction is preferably carried out in the presence of a solvent, and any solvent can be used, provided that it does not adversely affect the reaction. Suitable solvents are chloroform, methylene chloride and diethyl ether. In certain cases it is possible to carry out the reaction using an excess of one of the reactants or the base as a solvent. The reaction takes place over a wide range of temperatures, but to control the reaction accurately, one will work at a relatively low temperature, typically from -20 ° C to room temperature, and more preferably between -20 ° C and 0 ° C. However, higher temperatures can also be used if desired.
A carboxylic acid is treated with a chlorocarbonic acid ester or with a sulfonic acid chloride in the presence of a base, for example one of the above-mentioned organic amines, in order to obtain a mixed acid anhydride. This connection is then implemented with ML-236A, ML-236B, MB-530A or MB-530B. The reaction is preferably carried out in the presence of a solvent, the nature of which is not important, provided that it does not adversely affect the reaction. Suitable solvents are, for example, diethyl ether, benzene, chloroform and methylene chloride. The reaction takes place in a wide temperature range from, for example, -20 ° C to room temperature and preferably from -20 ° C to 0 ° C.
Methode 3 Method 3
ML-236A, ML-236B, MB-530A oder MB-530B wird mit einer Carbonsäure und einem Diazoalkyldicarboxylat in Gegenwart von beispielsweise Dicyclohexylcarbodiimid, Tri- ML-236A, ML-236B, MB-530A or MB-530B is mixed with a carboxylic acid and a diazoalkyl dicarboxylate in the presence of, for example, dicyclohexylcarbodiimide, tri-

The reaction is preferably carried out in a solvent, the nature of which is not important, provided that it does not adversely affect the reaction. Suitable 5 solvents are chloroform, methylene chloride, benzene and diethyl ether.
Each of the above reactions is usually completed within 30 minutes to 5 hours, although the exact time required for the reaction will depend on the reactants and the reaction temperature. After the reaction has ended, the desired product can be obtained in a manner known per se, for example by evaporating the solvent from a solution containing the desired product, which can simply be the reaction mixture or can be a solution obtained by extracting the reaction mixture with an organic solvent was separated from the reaction mixture, the solution optionally being washed and dried beforehand, whereupon the product can be purified by known means, for example by column chromatography, thin-layer chromatography or recrystallization or by a combination of these measures.
Im allgemeinen kann dies in folgender Weise geschehen: In those cases where the starting material used for the acylation reaction 25 contains two hydroxyl groups, i.e. if it is ML-530A or MB-530A, the reactions described above can effect the generation of the 3-monoester, the 3,8'-diester or the 8'-monoester or a mixture thereof. The nature of the product obtained can be affected by varying the amounts of reactants, the reaction temperature and other reaction conditions. Although the reasons are not known, it is believed that the 3-monoester is preferably formed, whereupon the 3,8'-diester and the 8'-monoester are formed. In the case of special reaction conditions, however, the person skilled in the art can easily determine which are the optimal conditions for producing the desired product. In general, this can be done in the following ways:
The 3-monoester and / or 8-monoester are obtained in predominant amounts if the reaction is carried out using an acid anhydride or halide as acylating agent in the presence of an organic base at a temperature between -20 ° C and 0 ° C . The amount of acylating agent to be used, preferably acid halide, is preferably about 1 equivalent per equivalent of ML-236A, ML-236B, MB-530A or MB-530B.
The 3,8'-diester is mainly produced when the acylating agent used is an acid anhydride or halide and in an amount of at least 2 equivalents per equivalent of ML-236A, ML-236B, MB-530A or MB-530B is used. The acylation then takes place in the presence of an organic base at a temperature 55 above room temperature.

Japan) and containing 0.5% ammonium chloride were placed in a 600 liter fermentation tank and inoculated with a culture of Monascus ruber SANK 15 177 (FERM 4956, NRRL 12 081). The cultivation of the microorganism was continued for 120 hours at 27 ° C. with an aeration speed of 300 liters / minute and with stirring at 190 revolutions per minute.
At the end of this period, the culture broth was filtered through a filter press to obtain a filtrate and a filter cake, the latter containing wet cells of the microorganism.
Dieses Produkt hatte die folgenden Eigenschaften: The filtrate was adjusted to a pH of 3.0 by adding a 6N hydrochloric acid solution and then extracted with 400 liters of ethyl acetate. The approximately 400 liter extract was concentrated by evaporation under reduced pressure and then dehydrated over anhydrous sodium sulfate, after which it was evaporated to dryness. In this way, about 60 g of an oily product were obtained. This oily product was washed with ethylcyclohexane and with hexane, and the residue, consisting of 20 g, was chromatographed using a liquid chromatography device for processing large volumes (“System 500 liquid chromatography” from Waters Co., USA) using a 60 volume product. separated aqueous methanol solution as eluent. The fractions, which had a chromatographic retention time of 6 minutes, were collected and concentrated by evaporation under reduced pressure to give 100 mg of the desired MB-530A as an oily product. This oily MB-530A was recrystallized from a mixture of acetone and diethyl ether to give 57 mg of the desired product in the form of colorless needles. This product had the following characteristics:

ML-236A-8 ' -butyrat ML-236A-8 'butyrate
The resulting solution was cooled to 0 ° C. and 0.35 ml of butyryl chloride was then added dropwise. When the reaction had ended, water was added and the separated methylene chloride layer was washed with water and then dried over anhydrous sodium sulfate. The residue formed by evaporation of the methylene chloride was subjected to a separation process by column chromatography over 10 g of silica gel, after which it was recrystallized from diethyl ether. In this way, 395 mg of the desired product with a melting point of 124 to 125 ° C. were obtained.
