Die Abspaltung der Benzoylgruppe entsprechend Verfahren a kann auf an sich bekannte Weise erfolgen. Vorzugsweise lässt sich die Benzoylgruppe hydrolytisch, insbesondere unter sauren Bedingungen, abspalten.

Insoweit die Herstellung der Ausgangsverbindungen nicht besonders beschrieben wurde, sind diese bekannt oder können nach an sich bekannten Methoden oder in Analogie zu den vorliegend beschriebenen Verfahren oder zu bekannten Verfahren hergestellt und gereinigt werden.
Die Verbindungen der Formel I können in racemischer oder optisch aktiver Form vorliegen. Die optisch aktiven Formen können auf an sich bekannte Weise wie fraktionierte Kristallisation der dia- stereomeren Salze hergestellt werden.
Die Verbindungen der Formel I können in freier Form als Base oder in Form ihrer Additionssalze mit Säuren vorliegen. Aus den freien Basen lassen sich in bekannter Weise Säureadditionssalze herstellen oder umgekehrt. Eine geeignete Säure für die Salzbildung ist die Fumarsäure.
Die Verbindungen der Formel I und die physiologisch verträgli- chen Säureadditionssalze dieser Verbindungen weisen im Tierversuch interessante pharmakodynamische Eigenschaften auf. Sie kön- nen daher als Heilmittel verwendet werden.
Insbesondere besitzen sie analgetische Eigenschaften. Überdies besitzen sie auch eine Wirkung auf das Zentralnervensystem. Aufgrund ihrer analgetischen Wirksamkeit können sie als strong (non narcotic) oder mild Analgetika zur Behandlung von Schmerzen verschiedenster Genese verwendet werden.
Zum Beispiel kann an der Maus mit der Phenylbenzochinonwrithing Testmethode bei per-os-Verabreichung von 1,7 bis 56 mg/kg Tiergewicht Analgesie und ZNS-Aktivität festgestellt werden. Weiterhin kann an der Maus mit der tail-flick-Testmethode bei per-os Verabreichung von 36 bis > 100 mg/kg Tiergewicht Analgesie festgestellt werden. Schliesslich kann an der entzündeten Rattenpfote mit der Randall-Selitto-Testmethode bei subkutaner Verabreichung von 3,5 bis 56 mg/kg Tiergewicht Analgesie festgestellt werden.
Die Dosierung wird für die oben angeführten Verwendungen von der Art der Verabreichung sowie von der gewünschten Behandlung abhängen.
Im allgemeinen werden zufriedenstellende Resultate erreicht bei Verabreichung einer täglichen Dose von I bis ungefähr 100 mg/kg Tiergewicht, wobei diese zweckmässig in geteilten Dosen 2- bis 4mal täglich oder in einer geeigneter Verabreichungsform gegeben wird.
Für grössere Säugetiere liegt die totale tägliche Dose zwischen unge fähr 100 und 300 mg. Eine geeignete Dosierungsform für die orale Verabreichung liegt zwischen ungefähr 20 und 150 mg einer Verbindung der Formel 1, vermischt mit einem festen oder flüssigen phar mazeutischen Träger oder Verdünnungsmittel.
Die Verbindungen der Formel I können allein oder in geeigneter Dosierungsform verabreicht werden. Die Arzneiformen, beispielsweise eine Tablette, können analog zu bekannten Methoden hergestellt werden.
Gegenstand der Erfindung sind auch Heilmittel, die die Verbindungen der Formel I enthalten, sowie die Herstellung dieser Heilmittel auf an sich bekannte Weise. Für ihre Herstellung können die in der Pharmazie gebräuchlichen Hilfs- und Trägerstoffe verwendet werden.
In den folgenden Beispielen erfolgen alle Temperaturangaben in Celsiusgraden und sind nicht korrigiert.
Beispiel 1: (4aRS,6RS,8aRS)-bzw.(4aRS,6SR,8aRS)-6-Azidodecahydro
4a-(3-methoxyphenyl)cisisochinolin (4aRS,6RS,8aRS)-bzw.(4aRS,6SR,8aRS)-6-Azido-2-benzoyloxydecahydro-4a-(3-methoxyphenyl)cisisochinolin wird in 40 ml n-Butanol gelöst und nach Zugabe von 40 ml 4N-Salzsäure während 72 h bei 90 gerührt. Nach Abkühlen wird das Reaktionsgemisch zuerst mit Hexan extrahiert, mit 10%ig. Sodalösung alkalisch gestellt und mit Methylenchlorid/Äthanol (8:2) extrahiert. Zur Reinigung wurde an der 40fachen Menge Kieselgel mit einem Gemisch von Methylen chlorid/Methanol/konz. Ammoniak (95:4,5:0,5) chromatographiert.
(4aRS,6RS,8aRS)-6-Azidodecahydro-4a-(3-m cisisochinolin wird als farbloser Schaum erhalten, der als Hydrochlorid mit Smp. 113-115' aus Aceton-Äther kristallisiert.
(4aRS,6SR,8aRS)-6-Azidodecahydro-4a-(3-m cisisochinolin wird in analoger Weise erhalten.
Das als Ausgangsmaterial verwendete (4aRS,6RS,8aRS)- bzw.
(4aRS,6S R,8aRS)-6-Azido-2-benzyloxydecahydro-4a- phenyl)cisisochinolin wird wie folgt hergestellt: 1. 64aRS,6aRS) -2-Benzoyloctahydro-4a- 03-methoxYPhenyl) cis-6- [2H]-isochinolinon
Ein Gemisch von 600 ml abs.
Tetrahydrofuran, 112,2 g (0,6 mol) 3-Bromanisol und 661 mg (6 mmol) Hydrochinon wird unter Stick- stoffbei652C mit 278 ml 2,2 N-Butyllithiumlösung (612 mmol) versetzt, 30 min bei50' gehalten, mit 57,13 g (0,3 mol) Kupfer(l)- jodid versetzt, 60 min bei43 gerührt, mit 51,06 g (0,2 mol) ( 1,3,4,7,8,8a-Hexahydro-2-methyl-6-[2H]-i vermischt, während 2 h auf 0 aufgewärmt und während 16 h bei 0 ausreagieren gelassen.
Nach Versetzen des erhaltenen Reaktionsgemisches mit 1,2 I Wasser und 79,28 g (0,6 mol) Ammoniumsulfat wird das Produkt z 1it Toluol extrahiert und der erhaltene ölige Eindampf rückstand an Kieselgel mit Äthylacetat chromatographiert und anschliessend aus Methanol kristallisiert. Man erhält farblose Kristalle von 2-Benzoyloctahydro-4a-(3-methoxyphenyl)-cis-6-[2 chinolinon (Smp. 78 ).
2. 64aRS,65R,8aRS)- bzw. (4aRS,6RS,8aRS)-2-Benzoyldecahydro- 6-hydroxy-4a-(3-methoxyphenyl)cisisochinolin a) 72,6 g (0,2 mol) 2-Benzoyloctahydro-4a-(3-methoxyphenyl) cis-6-[2H]-isochinolin werden in 200 ml Methanol suspendiert und bei 0 unter Rühren portionenweise mit 2,84 g (0,075 mol) Natriumborhydrid versetzt. Nach 2 h werden 20 ml Aceton und 200 ml Was- ser zugefügt und die Reaktionslösung mit Methylenchlorid erschöp- fend extrahiert.
b) Das erhaltene Rohprodukt wird zur Trennung der Epimeren mit 100 ml Pyridin und 190 ml Acetanhydrid in üblicher Weise ace- tyliert. Nach schonender Aufarbeitung des Reaktionsgemisches wird der ölige Rückstand in 80 ml Aceton aufgenommen und mit 50 ml Äther versetzt, wobei (4aRS,6SR,8aRS)-6-Acetoxy-2-benzoyl- decahydro-4a-(3-methoxyphenyl)cisisochinolin als farblose Prismen vom Smp. 133-135- erhalten wird.
Nach Chromatographie der Mutterlauge an der 10fachen Menge Kieselgel, Elution mit Methylenchlorid unter Zusatz von 0,51% Methanol und Kristalllisation aus Essigester/Äther wird (4aRS,6RS,8aRS)-6-Acetoxy-2-benzoyldecahydro-4a-(3-meth phenyl)cisisochinolin als farblose Kristalle vom Smp. 124-125- erhalten.
c) Eine Suspension von 40,8 g (0, I mol) (4aRS,6SR,8aRS)-6- Acetoxy-2-bezoyldecahydro-4a(3-methoxyphenyl)cis-6-[2 chinolin in 200 ml Methanol und 50 ml Methylenchlorid wird bei Raumtemperatur unter Rühren mit 100 ml 2N-Kalilauge [Methanol/Wasser (9:1)] versetzt. Nach 90 min engt man das Reaktionsgemisch im Vakuum ein, nimmt in Methylenchlorid/Äthanol (9: 1) auf, wäscht in üblicher Weise mit Wasser, trocknet und dampft ein: (4aRS,6SR,8aRS)-2-Benzyloxydecahydro-6-hydroxy-4a-(3- methoxyphenyl)cisisochinolin wird als farbloser Schaum erhalten und direkt weiterverarbeitet.
Ausgehend von (4aRS,6RS,8aRS)-6-Acetoxy-2-benzyloxy- decahydro-4a-(3-methoxyphenyl)cisisochinolin wird in analoger Weise (4aRS,6RS,8aRS)-2-Benzoyloxydecahydro-6-hydr methoxyphenyl)cisisochinolin als farbloser Schaum erhalten und direkt weiterverarbeitet.
3. (4aRS,6RS,8aRS)- b-w. (4aRS,6SR,8aRS)-Azido-2- benzoyloxydecahydro-4a-(3-methoxyphenyl)cisisochinolin a) 44,3 g (0,12 mol) (4aRS,6SR,8aRS)-2-Benzyloxydecahydro-6- hydroxy-4a-(3-methoxyphenyl)cisisochinlin, gelöst in 300 ml abs.
Tetrahydrofuran und 60 ml Triäthylamin werden bei Raum temperatur unter Riihren und Kiihlen mit 11,4 ml (0,14 mol) Methansulfonsäurechlorid in 100 ml abs. Tetrahydrofuran versetzt.
Nach 1 h wird das Reaktionsgemisch auf Eis gegossen und mit Methylenchlorid extrahiert. Der nach dem Eindampfen verbleibende Riickstand kristallisiert aus Methyläthylketon/Petroläther in farblosen Prismen vom Smp. 142-143 .
In analoger Weise wird das Epimere (4aRS,6RS,8aRS)-2 Benzoyloxydecahydro-6-hydroxy-4a-(3-methoxyphenyl)cisisochinolin umgesetzt und ergibt nach Krisallisation aus Methyläthylketon/ ither farblose Kristalle vom Smp. 148-150 .
b) Vom erhaltenen Mesylat werden 12,8 g (29 mmol), gelöst in 60 ml Dimethylsulfoxyd, mit 3,8 g (58 mmol) Natriumazid versetzt, und 6 h unter Überleiten von Stickstoff und Riihren auf 80 erhitzt.
Anschliessend wird das Reaktionsgemisch zwischen Toluol und Wasser verteilt, die organischen Phasen vereinigt, getrocknet und eingedampft und das Rohprodukt ohne weitere Reinigung direkt weiterverarbeitet.
Beispiel 2: (4aRS,6RS,8aRS)-bzw.(4aRS,6SR,8aRS)-6-Azidodecahydro
2-methyl-4a-(3-methoxyphenyl)cisisochinolin
28,6 g (0,1 mol) (4aRS,6RS,8aRS)-6-Azidodecahydro-4a-(3methoxyphenyl)cisisochinolin und 100 ml 35%ig.-wasseriger Formaldehydlösung in 300 ml Ethanol werden bei Raumtemperatur unter Rtihren und Kiihlen mit 19 g (0,5 mol) Natriumborhydrid versetzt. Nach 30 min wird mit Eiswasser versetzt und mit Methylenchlorid erschöpfend extrahiert. Das gesuchte Produkt kristallisiert als Hydrogenmaleinat in farblosen Prismen vom Smp. 155 aus Aceton/Äther.
Ausgehend von (4aRS,6SR,8aRS)-6-Azidodecahydro-4a-(3methoxyphenyl)cisisochinolin wird analog das Epimere Azid hergestellt: Hydrogenmaleinat als farblose Kristalle vom Smp. 138-140 .
aus Methyläthylketon/Äther.
Beispiel 3: (4aRS,6aRS,8aRS)-6-Azidodecahydro-2-isopropyl-4a-(3 methoxyphenyl) cisisochinolin
1,43 g (5 mmol) (4aRS,6RS,8aRS)-6-Azidodecahydro-4a-(3 metboxyphenyl)cisisochinolin in 25 ml Aceton und 25 ml Methanol werden mit 0,95 ml (5,5 mmol) N-Athyldiisopropylamin, 0,91 g (5,5 mmol) Kalizimjodid und 1,0 ml (10 mmol) Isopropyljodid versetzt und unter Rühren und Überleiten von Stickstoff auf 50 erhitzt. Nach 20 h wird im Vakuum eingeengt, der Ruckstand zwischen Methylenchlorid und Eiswasser verteilt, die organische Phase gewaschen, getrocknet und eingedampft. Das Rohprodukt wird zur Reinigung an der 10fachen Menge Kieselgel chromatographiert und mit Methylenchlorid/Methanol (99:1) eluiert. Das gesuchte Produkt kristallisiert als Hydrochlorid vom Smp. 192-194 aus Methanol/Aceton.

The benzoyl group can be split off in accordance with process a in a manner known per se. The benzoyl group can preferably be split off hydrolytically, in particular under acidic conditions.

Insofar as the preparation of the starting compounds has not been particularly described, they are known or can be prepared and purified by methods known per se or in analogy to the processes described here or to known processes.
The compounds of the formula I can be present in racemic or optically active form. The optically active forms can be prepared in a manner known per se, such as fractional crystallization of the diastereomeric salts.
The compounds of formula I can exist in free form as a base or in the form of their addition salts with acids. Acid addition salts can be prepared in a known manner from the free bases or vice versa. A suitable acid for salt formation is fumaric acid.
The compounds of the formula I and the physiologically tolerable acid addition salts of these compounds have interesting pharmacodynamic properties in animal experiments. They can therefore be used as a remedy.
In particular, they have analgesic properties. They also have an effect on the central nervous system. Due to their analgesic effectiveness, they can be used as strong (non narcotic) or mild analgesics for the treatment of pain of various origins.
For example, analgesia and CNS activity can be determined on the mouse using the phenylbenzoquinone writing test method when administered per os from 1.7 to 56 mg / kg animal weight. In addition, analgesia can be determined on the mouse using the tail flick test method with per-os administration of 36 to> 100 mg / kg animal weight. Finally, analgesia can be determined on the inflamed rat paw using the Randall-Selitto test method with subcutaneous administration of 3.5 to 56 mg / kg animal weight.
The dosage for the above uses will depend on the mode of administration and the treatment desired.
In general, satisfactory results are achieved when a daily dose of I to about 100 mg / kg of animal weight is administered, this being conveniently given in divided doses 2-4 times a day or in a suitable administration form.
For larger mammals, the total daily dose is between about 100 and 300 mg. A suitable dosage form for oral administration is between about 20 and 150 mg of a compound of Formula 1 mixed with a solid or liquid pharmaceutical carrier or diluent.
The compounds of formula I can be administered alone or in a suitable dosage form. The dosage forms, for example a tablet, can be produced analogously to known methods.
The invention also relates to medicaments which contain the compounds of the formula I, and to the preparation of these medicaments in a manner known per se. The auxiliaries and carriers customary in pharmacy can be used for their production.
In the following examples, all temperatures are given in degrees Celsius and are not corrected.
Example 1: (4aRS, 6RS, 8aRS) or (4aRS, 6SR, 8aRS) -6-azidodecahydro
4a- (3-methoxyphenyl) cisisoquinoline (4aRS, 6RS, 8aRS) or (4aRS, 6SR, 8aRS) -6-azido-2-benzoyloxydecahydro-4a- (3-methoxyphenyl) cisisoquinoline is dissolved in 40 ml of n-butanol and, after adding 40 ml of 4N hydrochloric acid, stirred at 90 for 72 h. After cooling, the reaction mixture is first extracted with hexane, with 10%. Soda solution made alkaline and extracted with methylene chloride / ethanol (8: 2). For cleaning was 40 times the amount of silica gel with a mixture of methylene chloride / methanol / conc. Ammonia (95: 4.5: 0.5) chromatographed.
(4aRS, 6RS, 8aRS) -6-azidodecahydro-4a- (3-m cisisoquinoline is obtained as a colorless foam which crystallizes as a hydrochloride with mp. 113-115 'from acetone ether.
(4aRS, 6SR, 8aRS) -6-azidodecahydro-4a- (3-m cisisoquinoline is obtained in an analogous manner.
The (4aRS, 6RS, 8aRS) or
(4aRS, 6S R, 8aRS) -6-azido-2-benzyloxydecahydro-4-phenyl) cisisoquinoline is prepared as follows: 1. 64aRS, 6aRS) -2-benzoyloctahydro-4a- 03-methoxYPhenyl) cis-6- [2H ] isoquinolinone
A mixture of 600 ml abs.
Tetrahydrofuran, 112.2 g (0.6 mol) of 3-bromoanisole and 661 mg (6 mmol) of hydroquinone are mixed with 278 ml of 2.2 N-butyllithium solution (612 mmol) under nitrogen at 652C, kept at 50 'for 30 min 57.13 g (0.3 mol) of copper (l) - iodide added, stirred for 60 min at 43, with 51.06 g (0.2 mol) (1,3,4,7,8,8a-hexahydro-2 -methyl-6- [2H] -i mixed, warmed to 0 for 2 h and left to react at 0 for 16 h.
After the reaction mixture obtained has been mixed with 1.2 l of water and 79.28 g (0.6 mol) of ammonium sulfate, the product is extracted with toluene and the oily evaporation obtained is chromatographed on silica gel with ethyl acetate and then crystallized from methanol. Colorless crystals of 2-benzoyloctahydro-4a- (3-methoxyphenyl) -cis-6- [2 quinolinone (mp 78) are obtained.
2. 64aRS, 65R, 8aRS) - or (4aRS, 6RS, 8aRS) -2-benzoyldecahydro-6-hydroxy-4a- (3-methoxyphenyl) cisisoquinoline a) 72.6 g (0.2 mol) 2-benzoyl octahydro -4a- (3-methoxyphenyl) cis-6- [2H] -isoquinoline are suspended in 200 ml of methanol and 2.84 g (0.075 mol) of sodium borohydride are added in portions at 0 with stirring. After 2 h, 20 ml of acetone and 200 ml of water are added and the reaction solution is extracted exhaustively with methylene chloride.
b) The crude product obtained is acetylated in a conventional manner with 100 ml of pyridine and 190 ml of acetic anhydride to separate the epimers. After careful processing of the reaction mixture, the oily residue is taken up in 80 ml of acetone and mixed with 50 ml of ether, where (4aRS, 6SR, 8aRS) -6-acetoxy-2-benzoyl-decahydro-4a- (3-methoxyphenyl) cisisoquinoline as colorless Prisms from mp. 133-135- is obtained.
After chromatography of the mother liquor on a 10-fold amount of silica gel, elution with methylene chloride with addition of 0.51% methanol and crystallization from ethyl acetate / ether, (4aRS, 6RS, 8aRS) -6-acetoxy-2-benzoyldecahydro-4a- (3-meth phenyl) cisisoquinoline as colorless crystals of mp 124-125- obtained.
c) A suspension of 40.8 g (0.1 mol) (4aRS, 6SR, 8aRS) -6-acetoxy-2-bezoyldecahydro-4a (3-methoxyphenyl) cis-6- [2 quinoline in 200 ml methanol and 50 ml of methylene chloride is mixed with 100 ml of 2N potassium hydroxide solution [methanol / water (9: 1)] at room temperature with stirring. After 90 min, the reaction mixture is concentrated in vacuo, taken up in methylene chloride / ethanol (9: 1), washed in the usual way with water, dried and evaporated: (4aRS, 6SR, 8aRS) -2-benzyloxydecahydro-6-hydroxy -4a- (3-methoxyphenyl) cisisoquinoline is obtained as a colorless foam and processed directly.
Starting from (4aRS, 6RS, 8aRS) -6-acetoxy-2-benzyloxy-decahydro-4a- (3-methoxyphenyl) cisisoquinoline, (4aRS, 6RS, 8aRS) -2-benzoyloxydecahydro-6-hydr methoxyphenyl) cisisoquinoline is produced in an analogous manner obtained as a colorless foam and processed directly.
3. (4aRS, 6RS, 8aRS) - b-w. (4aRS, 6SR, 8aRS) -azido-2-benzoyloxydecahydro-4a- (3-methoxyphenyl) cisisoquinoline a) 44.3 g (0.12 mol) (4aRS, 6SR, 8aRS) -2-benzyloxydecahydro-6-hydroxy- 4a- (3-methoxyphenyl) cisisoquinline, dissolved in 300 ml abs.
Tetrahydrofuran and 60 ml of triethylamine are at room temperature with stirring and cooling with 11.4 ml (0.14 mol) of methanesulfonic acid chloride in 100 ml of abs. Tetrahydrofuran added.
After 1 h the reaction mixture is poured onto ice and extracted with methylene chloride. The residue remaining after evaporation crystallizes from methyl ethyl ketone / petroleum ether in colorless prisms of mp 142-143.
The epimer (4aRS, 6RS, 8aRS) -2 benzoyloxydecahydro-6-hydroxy-4a- (3-methoxyphenyl) cisisoquinoline is reacted in an analogous manner and, after crystallization from methyl ethyl ketone / ether, gives colorless crystals of mp 148-150.
b) 3.8 g (58 mmol) of sodium azide are added to 12.8 g (29 mmol) of the mesylate obtained, dissolved in 60 ml of dimethyl sulfoxide, and the mixture is heated to 80 for 6 h while passing nitrogen and stirring.
The reaction mixture is then partitioned between toluene and water, the organic phases are combined, dried and evaporated and the crude product is directly processed without further purification.
Example 2: (4aRS, 6RS, 8aRS) or (4aRS, 6SR, 8aRS) -6-azidodecahydro
2-methyl-4a- (3-methoxyphenyl) cisisoquinoline
28.6 g (0.1 mol) (4aRS, 6RS, 8aRS) -6-azidodecahydro-4a- (3-methoxyphenyl) cisisoquinoline and 100 ml of 35% aqueous formaldehyde solution in 300 ml of ethanol are added at room temperature with stirring and cooling 19 g (0.5 mol) of sodium borohydride were added. After 30 min, ice water is added and the mixture is extracted exhaustively with methylene chloride. The product sought crystallized as hydrogen maleinate in colorless prisms of mp 155 from acetone / ether.
Starting from (4aRS, 6SR, 8aRS) -6-azidodecahydro-4a- (3methoxyphenyl) cisisoquinoline, the epimer azide is prepared analogously: hydrogen maleinate as colorless crystals of mp. 138-140.
from methyl ethyl ketone / ether.
Example 3: (4aRS, 6aRS, 8aRS) -6-azidodecahydro-2-isopropyl-4a- (3 methoxyphenyl) cisisoquinoline
1.43 g (5 mmol) (4aRS, 6RS, 8aRS) -6-azidodecahydro-4a- (3 metboxyphenyl) cisisoquinoline in 25 ml acetone and 25 ml methanol are mixed with 0.95 ml (5.5 mmol) N-ethyldiisopropylamine , 0.91 g (5.5 mmol) of potassium iodide and 1.0 ml (10 mmol) of isopropyl iodide are added and the mixture is heated to 50 while stirring and passing over nitrogen. After 20 h, the mixture is concentrated in vacuo, the residue is partitioned between methylene chloride and ice water, the organic phase is washed, dried and evaporated. The crude product is purified by chromatography on 10 times the amount of silica gel and eluted with methylene chloride / methanol (99: 1). The product sought crystallized as a hydrochloride of mp 192-194 from methanol / acetone.
