13. Heilmittel, dadurch gekennzeichnet, dass die Verbindungen der Formel (I) bzw. ihre physiologisch verträglichen Salze enthalten.

worin Y für Halogen, vorzugsweise Chlor oder Brom, oder für eine Gruppe Ry - 503 - 0-, worin Ry Phenyl, Tolyl oder niederes Alkyl bedeutet, steht. Y steht insbesondere für Chlor. Die Umsetzung wird worzugsweise in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z.B. in einem Äther wie Dioxan durchgeführt. Gegebenenfalls verwendet man als Lösungsmittel die Verbindung der Formel 111 im Überschuss.
Zweckmässig wird die Umsetzung auch in der Schmelze durchgeführt. Die Reaktionstemperatur liegt z.B. zwischen etwa 20 und etwa 200"C, vorzugsweise bei Siede- temperatur des Reaktionsgemisches, ausser falls in der Schmelze gearbeitet wird.
Die Verbindungen der Formel I können in freier Form oder in Salzform, z.B. in Säureadditionssalzform vorliegen. Aus den Verbindungen der Formel I in freier Form lassen sich in bekannter Weise Säureadditionssalze, beispielsweise das Bis [base] fumarat, das Hydrogenmalonat oder das Hydrogenmaleinat oder, falls R3 Hy droxy oder Re Wasserstoff bedeuten, Salze mit starken Basen, beispielsweise mit Natronlauge, gewinnen und umgekehrt.
In den Verbindungen der Formel list das Kohlenstoffatom der Hydroxypropoxyseitenkette, das die Hydroxygruppe trägt, asymmetrisch substituiert; sie können daher in Form von Racematen oder optisch aktiven Verbindungen auftreten. Von den Verbindungen der Formel I in optisch aktiver Form sind diejenigen bevorzugt, in denen die (S)-Konfiguration am asymmetrisch substituierten Kohlenstoffatom der Hydroxypropoxyseitenkette besteht.
Die Enantiomeren der Verbindungen der Formel I können auf an sich bekannte Weise erhalten werden, z.B. durch Aufspaltung mit optisch aktiven Säuren oder durch Durchführung der erfindungsgemässen Umsetzung ausgehend von entsprechenden, optisch aktiven Ausgangsprodukten.
Soweit die Herstellung der benötigten Ausgangsmaterialien nicht beschrieben wird, sind diese bekannt oder können nach bekannten Verfahren bzw. analog zu an sich bekannten Verfahren hergestellt und gereinigt werden. Das 4-Hydroxyindol-2-carbonitril bzw. 4-Hydroxy-3-methylindol-2-carbonitril erhält man durch Wasserentziehung mit z.B. Titantetrachlorid aus dem entsprechenden 2-Carboxamidderivat.
In den nachfolgenden Beispielen erfolgen alle Temperaturangaben in Celsiusgraden und sind unkorrigiert. Das Beispiel 1, das von Formel I nicht mehr umfasst wird, dient der Erläuterung der bei der Durchführung des erfindungsgemässen Verfahrens verwendbaren Reaktionsbedingungen.
Beispiel 1: 1- (3- Chlor-2-methylindol-4-yloxy) -3- (2-phenoxyäthylamino) -2propanol
5 g 3-Chlor-4-(2,3-epoxypropoxy)-2-methylindol, 4,3 g Phenoxy äthylamin und 75 ml Dioxan werden 15 h in einem Autoklaven auf 130"C erhitzt. Nach Abkühlen wird das Dioxan am Wasserstrahlvakuum und der Überschuss an Phenoxyäthylamin am Hoch- vakuum bei 80 abdestilliert. Den Rückstand schüttelt man zwischen Weinsäurelösung und Methylenchlorid aus, stellt die saure Phase mit konz. Ammoniak alkalisch und extrahiert mit Methylenchlorid.
Nach Abdampfen des Lösungsmittels kristallisiert der Rückstand mit 0,5 mol Malonsäure aus Methanol (Smp. des Hydrogenmalonats 135-137").
Analog zu Beispiel 1 erhält man durch Umsetzung der entsprechenden Verbindungen der Formel II, in denen Ro für - CHCH2 - Cl steht, mit den entsprechenden Verbindungen der

Die Verbindungen der Formel 1, d.h.
die Verbindungen der Formel 1 in freier Form oder in Forme ihrer physiologisch verträglichen Salze, besitzen interessante pharmakologische Eigenschaften. Sie können daher als Heilmittel verwendet werden.
Sie besitzen eine Blockerwirkung auf die adrenergischen u- und p-Rezeptoren und können daher u.a. zur Prophylaxe und Therapie von Krankheitszuständen, die mit einer adrenergischen Vasokonstriktion verbunden sind, insbesondere von myokardialen und peripheren Durchblutungsstörungen, sowie von Koronarerkrankungen, insbesondere von Angina pectoris, Verwendung finden. Ausserdem eignen sie sich zur Behandlung von Krankheitszuständen, die mit einer Lähmung der Darmmotilität einhergehen, z.B. vom paralytischen Ileus. Aufgrund ihrer antiarrhythmischen Wirkung können sie zudem zur Behandlung von Herzrhythmusstörungen eingesetzt werden.
Besonders interessant als a- und ss-Blocker ist die Verbindungen des Beispiels 19.

Die Verbindungen der Formel I besitzen ausserdem antihypertensive Eigenschaften. Aufgrund ihrer blutdrucksenkenden Wirkung können sie als Antihypertensiva, z.B. in der Hochdrucktherapie Anwendung finden.
Die Verbindungen der Formel I zeigen ausserdem interessante Stoffwechselwirkungen, indem sie die durch psychischen Stress induziert Mobilisation von Glucose und Fettsäure im Blut hemmen.
Aufgrund ihrer metabolischen Wirkung können sie bei Zuständen, die zu einer durch psychischen Stress verursachten, unerwünschten Stoffwechselmobilisation führen, eingesetzt werden.
Besonders interessant in dieser Indikation sind die Verbindungen der Beispiele 2, 5 und 19, insbesondere die Verbindung des Beispiels 5.

Insofar as the production of the required starting materials is not described, they are known or can be produced and purified by known processes or analogously to processes known per se. The 4-hydroxyindole-2-carbonitrile or 4-hydroxy-3-methylindole-2-carbonitrile is obtained by dehydration with e.g. Titanium tetrachloride from the corresponding 2-carboxamide derivative.
In the following examples, all temperatures are given in degrees Celsius and are uncorrected. Example 1, which is no longer included in formula I, serves to explain the reaction conditions which can be used when carrying out the process according to the invention.
Example 1: 1- (3-chloro-2-methylindol-4-yloxy) -3- (2-phenoxyethylamino) -2 propanol
5 g of 3-chloro-4- (2,3-epoxypropoxy) -2-methylindole, 4.3 g of phenoxyethylamine and 75 ml of dioxane are heated in an autoclave at 130 ° C. for 15 hours. After cooling, the dioxane is removed in a water jet vacuum and the excess of phenoxyethylamine was distilled off under high vacuum at 80. The residue was shaken out between tartaric acid solution and methylene chloride, the acidic phase was made alkaline with concentrated ammonia and extracted with methylene chloride.

The compounds of formula 1, i.e.
the compounds of formula 1 in free form or in the form of their physiologically tolerable salts have interesting pharmacological properties. They can therefore be used as a remedy.
They have a blocker effect on the adrenergic u and p receptors and can therefore, among other things. for the prophylaxis and therapy of disease states associated with an adrenergic vasoconstriction, in particular of myocardial and peripheral circulatory disorders, and of coronary diseases, in particular of angina pectoris.
