PATENTANSPRÜCHE 1. Verfahren zur Herstellung von pharmazeutisch verwendbaren Salzen von Oxothiaverbindungen der Formel

Verfahren gemäss Anspruch 1, dadurch gekennzeichnet, dass man Verbindungen der Formel 1, worin Ph gegebenenfalls durch Niederalkyl, Niederalkoxy, Halogen, Trifluormethyl und/oder Nitro substituiertes 1,2-Phenylen darstellt, X für Sauerstoff oder Schwefel steht, Rl Niederalkyl, Niederalkenyl, Cycloalkyl, Cycloalkenyl oder Cycloalkylniederalkyl, gegebenenfalls im Phenylteil durch Niederalkyl, Niederalkoxy, Halogen, Trifluormethyl und/oder Nitro substituiertes Phenylniederalkyl, gegebenenfalls durch Niederalkyl, Niederalkoxy, Halogen, Trifluormethyl und/oder Nitro substituiertes Phenyl, gegebenenfalls durch Niederalkyl, Niederalkoxy, Halogen, Trifluormethyl und/oder Nitro substituiertes, gegebenenfalls Benzo-enthaltendes Mono- oder Diazaaryl mit 6 Ringgliedern im heterocyclischen Teil oder gegebenenfalls durch Niederalkyl, Niederalkoxy,
Halogen, Trifluormethyl und/oder Nitro substituiertes, gegebenenfalls Benzo-enthaltendes Oxaza- oder Thiazaaryl mit 5 Ringgliedern im heterocyclischen Teil darstellt, und R2 für Wasserstoff oder Niederalkyl steht, in Form ihrer pharmazeutisch verwendbaren Salze mit Basen herstellt.
3. Verfahren gemäss Anspruch 1 und 2, dadurch gekennzeichnet, dass man Verbindungen der Formel I, worin Ph gegebenenfalls durch Niederalkyl, Niederalkoxy, Halogen mit Atomnummer bis zu 35 oder Trifluormethyl substituiertes 1,2-Phenylen darstellt, X für Sauerstoff steht, Rl gegebenenfalls durch Niederalkyl, Niederalkoxy, Halogen mit Atomnummer bis 35, Trifluormethyl und/oder Nitro substituiertes Phenyl oder gegebenenfalls durch Niederalkyl substituiertes Pyridyl, Oxazolyl, Isoxazolyl, Thiazolyl oder Isothiazolyl bedeutet, und R2 für Wasserstoff oder Niederalkyl steht, in Form ihrer pharmazeutisch verwendbaren Salze mit Basen herstellt.

Verfahren gemäss Anspruch 1 bis 3, dadurch gekennzeichnet, dass man Verbindungen der Formel I, worin Ph 1,2-Phenylen darstellt, das gegebenenfalls durch Niederalkyl, Niederalkoxy und/oder Halogen mit Atomnummer bis 35 substituiert sein kann, X für Sauerstoff steht, Rl gegebenenfalls durch Niederalkyl, Niederalkoxy, und/oder Halogen mit Atomnummer bis 35 substituiertes Phenyl oder gegebenenfalls durch Niederalkyl substituiertes Pyridyl, Oxazolyl, Isoxazolyl, Thiazolyl oder Isothiazolyl bedeutet, und R2 für Wasserstoff steht, in Form ihrer pharmazeutisch verwendbaren Salze mit Basen herstellt.
5. Verfahren gemäss den Ansprüchen 1 bis 4, dadurch gekennzeichnet, dass man Verbindungen der Formel I in Form ihrer pharmazeutisch verwendbaren Metallsalze herstellt.
6. Verfahren gemäss den Ansprüchen 1 bis 5, dadurch gekennzeichnet, dass man Verbindungen der Formel 1 in Form ihrer pharmazeutisch verwendbaren Alkalimetallsalze herstellt.
7. Verfahren gemäss den Ansprüchen 1 bis 5, dadurch gekennzeichnet, dass man Verbindungen der Formel I in Form ihrer pharmazeutisch verwendbaren Zinksalze herstellt.
8. Verfahren gemäss den Ansprüchen 1 bis 4, dadurch gekennzeichnet, dass man Verbindungen der Formel I in Form ihrer pharmazeutisch verwendbaren Ammoniumsalze mit sekundären organischen Aminen herstellt.
9. Verfahren gemäss den Ansprüchen 1 bis 4 und 8, dadurch gekennzeichnet, dass man Verbindungen der Formel I in Form ihrer pharmazeutisch verwendbaren Mor pholinsalze herstellt
10. Verfahren gemäss den Ansprüchen 1 bis 9, dadurch gekennzeichnet, dass man N-(3-Chlorphenyl)-2-oxo-2,3dihydro-3-benzo[b]thiophencarboxamid in Form eines pharmazeutisch verwendbaren Salzes mit Basen herstellt.
Die Erfindung betrifft ein Verfahren zur Herstellung von Salzen von Oxothiaverbindungen, nämlich 2-Oxo-2,3-dihy drobenzo[bjthiophen-Verbindungen der Formel

Die pharmazeutisch verwendbaren Salze der obigen 2-Oxo-2,3-dihydro-benzo[b]thiophen-Verbindungen mit Basen leiten sich von den entsprechenden Tautomeren, d.h.
den 2-Hydroxy-benzo[b]thiophen-Verbindungen ab.
Im Zusammenhang mit der vorliegenden Beschreibung enthalten mit nieder bezeichnete organische Reste und Verbindungen bis zu 7, vorzugsweise bis zu 4 Kohlenstoff.- atome.
Ein über ein Kohlenstoffatom gebundener organischer Rest ist z.B. ein gegebenenfalls substituierter aliphatischer Kohlenwasserstoffrest, z.B. ein aliphatischer, cycloaliphatischer, cycloaliphatisch-aliphatischer oder araliphatischer Kohlenwasserstoffrest, oder insbesondere aromatischer Kohlenwasserstoffrest, d.h. ein über ein Kohlenstoffatom gebundener, gegebenenfalls substituierter aromatischer oder heteroaromatischer Rest.
Ein aliphatischer Kohlenwasserstoffrest ist insbesondere Niederallcyl, ferner Niederalkenyl oder Niederalkinyl, ein cycloaliphatischer Kohlenwasserstoffrest z.B. Cycloalkyl oder Cycloalkenyl, vorzugsweise mit bis zu 8 Ringgliedern, und ein cycloaliphatisch-aliphatischer Kohlenwasserstoffrest, z.B. Cycloalkylniederalkyl, vorzugsweise mit bis zu 8 Ringgliedern im cycloaliphatischen Teil, während ein araliphatischer Kohlenwasserstoffrest insbesondere monocyclisches Arylniederalkyl, d.h. Ein heteroaromatischer Rest ist insbesondere ein monocyclischer heteroaromatischer Rest mit 5 oder 6 Ringgliedern, wobei mindestens ein Ringglied ein Stickstoffatom ist und ein gegebenenfalls zusätzlich vorhandenes Heteroringatom ein Stickstoff-, Sauerstoff- oder Schwefelatom sein kann.
Substituenten der obgenannten organischen Reste, insbesondere eines aromatischen oder heteroaromatischen Restes Rl, wie auch des 1,2-Phenylenrestes Ph sind u.a. Niederalkyl, Niederalkoxy, Halogen, Trifluormethyl oder Nitro, wobei zwei benachbarte Kohlenstoffatome, insbesondere eines heteroaromatischen Restes, auch durch gegebenenfalls, z.B.
wie ein aromatischer Rest, substituiertes 1,3-Butadien-1,4ylen, d.h. unter Ankondensation eines aromatischen Restes und unter Bildung eines bicyclischen, insbesondere eines benzoheteroaromatischen Restes, substituiert sein kann.
Niederalkyl ist z.B. Methyl, Äthyl, n-Propyl, Isopropyl, n-Butyl oder tert.-Butyl, während Niederalkenyl z.B. Allyl oder Methallyl, und Niederalkinyl z.B. Propargyl, sind.
Cycloalkyl ist z.B. Cyclopropyl, Cyclo- pentyl, Cyclohexyl oder Cycloheptyl, und Cycloalkenyl z.B.
in irgendeiner geeigneten Stellung die Doppelbindung aufweisendes Cyclopentenyl, Cyclohexenyl oder Cycloheptenyl, während Cycloalkylniederalkyl z.B. Cyclopropylmethyl, Cyclopentylmethyl, Cyclohexylmethyl oder 2-Cyclohexyl äthyl darstellt.
Phenylniederalkyl ist z.B. Benzyl oder 1 - oder 2-Phenyl äthyl.
Ein heteroaromatischer Rest mit einem Ringstickstoffatom ist u.a. monocyclisches Monoazaaryl, z.B. Pyridyl, wie 2-, 3oder 4-Pyridyl, während ein entsprechender Rest mit zwei Ringstickstoffatomen u.a. monocyclisches Diazaryl, z.B.
Pyridazinyl, wie 3- oder 4-Pyridazinyl, Pyrimidinyl, wie 2oder 4-Pyrimidinyl, oder Pyrazinyl, wie 2-Pyrazinyl, sein kann. Ein heteroaromatischer Rest mit einem Ringstickstoffatom und einem Ringsauerstoff- oder Ringschwefelatom ist u.a. monocyclisches Oxazaaryl oder Thiazaaryl, z.B. Oxazolyl, z.B. 2-Oxazolyl, Isoxazolyl, z.B. 3-Isoxazolyl, Thiazolyl, z.B. 2-Thiazolyl, oder Isothiazolyl, z.B. 2-Isothiazolyl.
Entsprechende heteroaromatische Reste, worin zwei benachbarte Ringkohlenstoffatome durch 1 ,3-Butadien-l ,4-ylen substituierte sind, d.h. benzoheteroaromatische Reste, sind u.a. bicyclisches Benzoazaaryl, z.B. Chinolinyl, wie 4-Chinolinyl, oder Isochinolinyl, oder bicyclisches Benzooxaaryl oder Benzothiazaaryl, z.B. Benzoxazolyl, z.B. 2-Benzoxa- zolyl, oder Benzthiazolyl, z.B. 2-Benzthiazolyl.
Niederalkoxy ist z.B. Methoxy, Äthoxy, n-Propyloxy, Isopropyloxy, n-Butyloxy oder Isobutyl.
Halogen ist in erster Linie Halogen mit einer Atomnummer bis 39, d.h. Fluor, Chlor oder Brom.
Salze von Verbindungen der Formel I mit Basen sind vor allem Metall- oder Ammoniumsalze. Metallsalze sind dabei vor allem von Metallen der Gruppen Ia, Ib, IIa und IIb des Periodischen Elementsystems abgeleitete Metallsalze, wie Alkalimetall- oder Erdalkalimetallsalze, z.B. Natrium-, Kalium-, Magnesium-, Calcium-, Zink- oder Kupfersalze.
Ammoniumsalze sind vor allem Salze mit sekundären oder tertiären organischen Basen, z.B. mit Morpholin, Thiomorpholin, Piperidin, Pyrrolidin, Dimethyl- bzw. Diäthylamin oder Triäthylamin, in zweiter Linie aber auch Salze mit Ammoniak. Die Salzbildung mit Verbindungen der Formel I erfolgt dabei wahrscheinlich aus der tautomeren 2-Hydroxybenzo[b]thiophen-Form heraus.
Die neuen Verbindungen zeigen wertvolle pharmakologische Eigenschaften. So weisen sie antiinflammatorische Wirkungen auf, die z.B. im Kaolinödemtest an der Ratte analog der von Menasse und Krupp, Toxicol. Appl. Pharmacol. Bd. 29, S. 389 (1974) beschriebenen Methode in Dosen von etwa 30 mg/kg bis etwa 100 mg/kg p.o. nachgewiesen werden können. Vor allem zeigen sie analgetische Wirkungen, die z.B. an der Maus im Phenyl-p-Benzochinon-Writingtest analog der von Krupp et al., Schweiz. Med. Wschr., Bd. 105, 5.646(1975), beschriebenen Methode in Dosen von etwa 3 mg/kg bis etwa 100 mg/kg p.o. nachgewiesen werden können. Ferner weisen sie uricosurische Wirkungen auf, die z.B. im Phenolrot-Ausscheidungstest an der Ratte, analog der von Swindleetal., Arch. Int.
Pharmacodyn., Bd. 189,5. 129 (1971), beschriebenen Methode in Dosen von etwa 100 mg/kg p.o. nachgewiesen werden. Die Verbindungen werden deshalb als Antiphlogistika, z.B. zur Behandlung von arthritischen Entzündungen, oder als Analgetika, z.B. zur Behandlung von Schmerzzuständen, sowie als Uricosurika, z.B. zur Behandlung von Gicht verwendet.
Die neuen Verbindungen zeigen ebenfalls antithrombotische Wirkungen, die am Kaninchen in der experimentellen Lungenembolie analog der von Silver et al., Science, Bd. 183, S. 1085 (1974), beschriebenen Methode in Dosen von etwa 3 mg/kg bis etwa 30 mg/kg p.o. nachgewiesen werden können.
Sie können deshalb auch als Thrombolytika verwendet werden.
Die Erfindung betrifft in erster Linie ein Verfahren zur Herstellung von Verbindungen der Formel I, worin Ph gegebenenfalls durch Niederalkyl, Niederalkoxy, Halogen, Trifluormethyl und/oder Nitro substituiertes 1,2-Phenylen darstellt, X für Sauerstoff oder in zweiter Linie für Schwefel steht, Rl Niederalkyl, Niederalkenyl, Cycloalkyl, Cycxloalkenyl oder Cycloalkylniederalkyl, oder gegebenenfalls im Phenylteil durch Niederalkyl, Niederalkoxy, Halogen, Trifluormethyl und/oder Nitro substituiertes Phenylniederalkyl, oder in erster Linie gegebenenfalls durch Niederalkyl, Niederalkoxy, Halogen, Trifluormethyl und/oder Nitro substituiertes Phenyl oder gegebenenfalls durch Niederalkyl, Niederalkoxy, Halogen, Trifluormethyl und/oder Nitro substituiertes, gegebenenfalls Benzo-enthaltendes Mono- oder Diazaaryl mit 6 Ringgliedern im heterocyclischen Teil,
insbesondere entsprechendes Pyridyl oder Chinolinyl, ferner Pyrimidinyl, oder gegebenenfalls durch Niederalkyl, Niederalkoxy, Halogen, Trifluormethyl und/oder Nitro substituiertes, gegebenenfalls Benzo-enthaltendes Oxaza- oder Thiazaaryl mit 5 Ringgliedern im heterocyclischen Teil, insbesondere entsprechendes Oxazolyl, Benzoxazolyl, Isoxazolyl, Thiazolyl, Benzthiazolyl oder Isoxazolyl, darstellt, und R2 für Wasserstoff oder Niederalkyl steht, in Form ihrer pharmazeutischen anwendbaren Salze mit Basen.
Die Erfindung betrifft in erster Linie ein Verfahren zur Herstellung von Verbindungen der Formel I, worin Ph gegebenenfalls durch Niederalkyl, z.B. Methyl, Niederalkoxy, z.B. Methyl substituiertes Pyridyl, z.B. 2-oder 4-Pyridyl, Oxazolyl, z.B. 2-Oxa- zolyl, Isoxazolyl, z.B. 3-Isoxazolyl, Thiazolyl, z.B. 2-Thia- zolyl, oder Isothiazolyl, z.B. 3-Isothiazolyl, bedeutet, und R2 für Wasserstoff oder in zweiter Linie für Niederalkyl, z.B.
Methyl, steht, in Form ihrer pharmazeutisch verwendbaren Salze mit Basen.
Die Erfindung betrifft in erster Linie ein Verfahren zur Herstellung von Verbindungen der Formel I, worin Ph 1,2-Phenylen ist, das gegebenenfalls durch Niederalkyl, z.B.
Methyl, Niederalkoxy, z.B. Methoxy, und/oder Halogen mit Atomnummer bis 35, z.B. Fluor oder Chlor, substituiert sein kann, wobei sich solche Substituenten in erster Linie in 5 und/oder 6-Stellung des 2,3-Dihydro-benzo[b]-thiophenrings befinden, X für Sauerstoff steht, Rt gegebenenfalls durch Niederalkyl, z.B. Methyl, Niederalkoxy, z.B. Methoxy, und/oder Halogen mit Atomnummer bis 35, z.B. Fluor oder Chlor, substituiertes Phenyl oder gegebenenfalls durch Niederalkyl, z.B. Methyl, substituiertes Pyridyl, z.B. 2- oder 4-Pyridyl, Oxazolyl, z.B. 2-Oxazolyl, Isoxazolyl, z.B. 3-Iso- xazolyl, Thiazolyl, z.B. 2-Thiazolyl, oder Isothiazolyl, z.B.
3-Isothiazolyl, bedeutet, und R2 für Wasserstoff steht, in Form ihrer pharmazeutisch verwendbaren Salze mit Basen.
Die Verbindungen der Formel I werden erfindungsgemäss in Form ihrer pharmazeutisch verwendbaren Salze erhalten, indem man freie Verbindungen der Formel I mit Basen umsetzt. Die in freier Form vorliegenden Ausgangsstoffe der Formel 1 erhält man z.B., indem man in eine Verbindung der Formel

Die 2-Oxo-2,3-dihydro-benzo[b]thiophen-Verbindung der Formel II kann auch in tautomerer Form, d.h. als 2-Hydroxybenzo[b]thiophen-Verbindung, vorliegen.
Die Einführung der Gruppe der Formel Ia kann direkt oder schrittweise erfolgen. So kann man diese direkt einführen, in dem man eine Verbindung der Formel II mit einer Verbindung der Formel I

Eine verätherte Hydroxygruppe Ro ist vorzugsweise durch einen, gegebenenfalls substituierten Kohlenwasserstoffrest, wie Niederalkyl, z.B. Methyl oder Äthyl, oder Halogenniederalkyl, z.B. 2,2,2-Trichloräthyl, und in erster Linie durch gegebenenfalls substituiertes, wie Niederalkyl, Niederalkoxy, Halogen und/oder Nitro enthaltendes Phenyl ver äthertes Hydroxy, und stellt z.B. Die obige Reaktion wird üblicherweise in Gegenwart eines basischen Mittels, wie eines entsprechenden anorganischen oder organischen Mittels vorgenommen. Als anorganische Basen kommen in erster Linie Salz-, insbesondere Alkalimetallsalz-bildende Mittel, wie Alkalimetallhydride oder -amide, sowie Alkalimetallorganische Verbindungen, wie entsprechende Niederalkanolate, ferner entsprechende Niederalkyl- oder Phenylverbindungen, z.B. Natriummethylat, Natriumäthylat, Kalium-tert.-butylat, n-Butyllithium oder Phenyllithium, in Frage. Geeignete organische Basen sind in erster Linie Amine, wie tertiäre Amine, vorzugsweise Triniederalkylamine, z.B. Triäthylamin, heterocyclische tertiäre Basen, insbesondere vom Pyridintyp, z.B.
Pyridin, oder quaternäre Basen, wie Tetraniederalkylammonium- oder Triniederalkyl-phenylniederalkyl-ammoniumhydroxide. In Gegenwart der Base kommt die Verbindung der Formel II in anionischer, d.h. in Salzform, zur Umsetzung mit der Verbindung der Formel III.
Letztere sind der Formel III entsprechende Carbaminsäureester, Carbaminsäurehalogenide, Harnstoffe und Isocyanate, sowie die entsprechenden Schwefelverbindungen.
Die Reaktion wird in An- oder Abwesenheit eines Lösungs- oder Verdünnungsmittels, und, wenn notwendig, unter Kühlen oder Erhitzen, z.B. in einem Temperaturbereich von etwa - 10"C bis etwa + 120"C, in einem geschlossenen Gefäss und/oder in einer Inertgas-, z.B. Stickstoffatmosphäre durchgeführt.
Die schrittweise Einführung einer Gruppe der Formel Ia in eine Verbindung der Formel II, kann so durchgeführt werden, dass man eine Verbindung der Formel II mit einer Verbindung der Formel Rb -C(=X)-R (IV), worin Ro und Rb unabhängig voneinander für eine verätherte oder veresterte Hydroxygruppe stehen, umsetzt, und eine als Zwischenprodukt erhältliche Verbindung der Formel

Verätherte oder veresterte Hydroxygruppen Ro und Rb haben z.B. die oben für den entsprechenden Rest Ro gegebenen Bedeutungen und sind z.B. Niederalkoxy, wie Methoxy oder;2fhoxy, ferner gegebenenfalls substituiertes Phenyloxy, oder Halogen, z.B. Chlor. Geeignete Verbindungen der Formel IV sind z.B. Diniederalkylcarbonate, z.B.
Diäthyl- oder Diphenylcarbonat, Phosgen oder Halogenameisensäure-niederalkylester, z.B. Chlorameisensäureisobu- tylester, sowie die entsprechenden Schwefelverbindungen.
Die Reaktion der Verbindung der Formel IIa mit einer Verbindung der Formel IV wird üblicherweise in Gegenwart einer Base, wie einer der obgenannten, z.B. eines Alkalimetallhydrids oder eines Triniederalkylamins durchgeführt.
Üblicherweise wird ein Zwischenprodukt der Formel IIa nicht isoliert, sondern direkt mit dem Amin der Formel V umgesetzt.
Die obigen Verfahrensschritte werden in Ab- oder Anwesenheit eines Lösungs- oder Verdünnungsmittels, und, wenn notwendig, unter Kühlen oder Erhitzen, z.B. in einem Temperaturbereich von etwa - 10"C bis etwa + I20 C, in einem geschlossenen Gefäss und/oder in einer Inertgas-, z.B. Stickstoffatmosphäre durchgeführt. Die Verbindungen der Formeln II und III sind bekannt oder können in an sich bekannter Weise hergestellt werden.
Verbindungen der Formel II können beispielsweise erhalten werden, indem man ein von einem gegebenenfalls wie für Ph angegeben substituierten Cyclohexanon abgelei tetes Enamin mit einem Cyanessigsäureester umsetzt, den erhaltenen 2-Amino-4,5,6,7-tetrahydrobenzothiophen-3-car- bonsäureester an der Aminogruppe axyliert, das Reaktionsprodukt mit Schwefel dehydriert und den erhaltenen 2-Acylamino-benzothiophen-3-carbonsäureester mit Natronlauge behandelt, oder indem man ein entsprechendes Benzothiophenyl mit Butyllithium in die 2-Lithiumverbindung überführt, diese mit Borsäuretributylester umsetzt und das Reaktionsprodukt mit Wasserstoffperoxid oxydiert.
Ein insbesondere zur Herstellung von halogensubstituierten Verbindungen der Formel II geeignetes Verfahren besteht darin, dass man einen entsprechenden Benzothiophen-2-carbonsäureester mit Hydrazin in das Säurehydrazid überführt, dieses mit salpetriger Säure zum Azid umsetzt, dieses zum Isocyanat umlagert, das Isocyanat durch Alkoholyse in das Urethan überführt, dieses zur Carbaminsäure hydrolysiert, diese decarboxyliert und dies erhaltene 2-Iminobenzothiophen hydrolysiert.
Verbindungen der Formel III können z.B. hergestellt werden durch Umsetzung von Verbindungen der Formeln Rb0 -C(=X)- Ro (IV) undHNRIR2(V).
Die Ausgangstoffe der Formel I in freier Form können ebenfalls erhalten werden, wenn man eine Verbindung der Formel I

Alkalimetallsalz-bildenden Mittels, u.a. auch eines Alkalimetallniederalkanolates, z.B. Natriummethylat, Natriumäthylat oder Kalium-tert.-butylat, vorgenommen werden. Dabei arbeitet man in Ab- oder Anwesenheit eines Lösungs- oder Verdünnungsmittels, wenn notwendig, unter Kühlen oder Erwärmen, z.B. in einem Temperaturbereich von etwa 0 C bis etwa 150"C, in einem geschlossenen Gefäss und/oder in einer Inertgas-, z.B. Beim Verfahren der vorliegenden Erfindung werden vorzugsweise solche Ausgangsstoffe verwendet, welche zu den eingangs als besonders wertvoll geschilderten Verbindungen führen.
Bei pharmazeutischen Präparaten, welche Verbindungen der Formel I in Form ihrer pharmazeutisch verwendbaren Salze mit Basen enthalten, handelt es sich um solche zur enteralen, wie oralen, rektalen oder parenteralen Verabreichung oder zur topischen bzw. lokalen Verwendung an Warmblüter, welche den pharmakologischen Wirkstoff allein oder zusammen mit einem pharmazeutisch anwendbaren Trägermaterial enthalten. Die Dosierung des Wirkstoffs hängt von der Warmblüter-Spezies, dem Alter und dem individuellen Zustand, sowie von der Applikationsweise ab.
Die neuen pharmazeutischen Präparate enthalten von etwa 10% bis etwa 95%, vorzugsweise von etwa 20% bis etwa 90% des Wirkstoffs. Pharmazeutische Präparate sind beispielsweise solche in Elixier-, Aerosol- oder Sprayform oder in Dosiseinheitsform, wie Dragees, Tabletten, Kapseln, Suppositorien oder Ampullen.
Die pharmazeutischen Präparate werden in an sich bekannter Weise, z.B. mittels konventioneller Misch-, Granulier-, Dragier-, Lösungs- oder Lyophilisierungsverfahren hergestellt.
Präparate zur oralen Anwendung kann man z.B. erhalten, indem man den Wirkstoff mit festen Trägerstoffen kombiniert, ein erhaltenes Gemisch gegebenenfalls granuliert, und das Gemisch bzw. Granulat, wenn erwünscht oder notwendig nach Zugabe von geeigneten Hilfsstoffen, zu Tabletten oder Dragée-Kerner verarbeitet. Geeignete Trägerstoffe sind insbesondere Füllstoffe, wie Zucker, z.B. Lactose, Saccharose, Mannit oder Sorbit, Cellulosepräparate und/oder Calciumphosphate, z.B.
Tricalciumphosphat oder Calciumhydrogenphosphat, ferner Bindemittel, wie Stärkekleister unter Verwendung z.B. von Mais-, Weizen-, Reis- oder Kartoffelstärke, Gelatine, Traganth, Methylcellulose, Hydroxypropylmethylcellulose, Natriumcarboxymethylcellulose und/oder Polyvinylpyrrolidon, und/oder, wenn erwünscht, Sprengmittel, wie die obgenannten Stärken, ferner Carboxymethylstärke, quervernetztes Polyvinylpyrrolidon, Agar, Alginsäure oder ein Salz davon, wie Natriumalginat. Hilfsmittel sind in erster Linie Fliessregulier- und Schmiermittel, z.B. Kieselsäure, Talk, Stearinsäure oder Salze davon, wie Magnesiumoder Calciumstearat, und/oder Polyäthylenglykol.
Dragee Kerne werden mit geeigneten, gegebenenfalls Magensaftresistenten Überzügen versehen, wobei man u.a. konzentrierte Zuckerlösungen, welche gegebenenfalls arabischen Gummi, Talk, Polyvinylpyrrolidon, Polyäthylenglycol und/ oder Titandioxid enthalten, Lacklösungen in geeigneten organischen Lösungsmitteln oder Lösungsmittelgemischen oder, zur Herstellung von Magensaft-resistenten Überzügen, Lösungen von geeigneten Cellulosepräparaten, wie Acetylcellulosephthalat oder Hydroxypropylmethylcellulosephthalat, verwendet. Den Tabletten oder Dragee-Überzügen können Farbstoffe oder Pigmente, z.B. zur Identifizierung oder zur Kennzeichnung verschiedener Wirkstoffdosen, beigefügt werden.
Weitere, oral anwendbare pharmazeutische Präparate sind Steckkapseln aus Gelatine, sowie weiche, geschlossene Kapseln aus Gelatine und einem Weichmacher, wie Glycerin oder Sorbitol. Die Steckkapseln können den Wirkstoff in Form eines Granulats, z.B. im Gemisch mit Füllstoffen, wie Lactose, Bindemitteln, wie Stärken, und/oder Gleitmitteln, wie Talk oder Magnesiumstearat, und gegebenenfalls von Stabilisatoren, enthalten. In weichen Kapseln ist der Wirkstoff vorzugsweise in geeigneten Flüssigkeiten, wie fetten Ölen, Paraffinöl oder flüssigen Polyäthylenglykolen, gelöst oder suspendiert, wobei ebenfalls Stabilisatoren zugefügt sein können.
Als rektal anwendbare pharmazeutische Präparate kommen z.B. Suppositorien in Betracht, welche aus einer Kombination des Wirkstoffs mit einer Suppositoriengrundmasse bestehen. Als Suppositoriengrundmasse eignen sich z.B. natürliche oder synthetische Triglyceride, Paraffinkohlenwasserstoffe, Polyäthylenglykole oder höhere Alkanole.
Ferner können auch Gelatine-Rektalkapseln verwendet werden, die aus einer Kombination des Wirkstoffs mit einer Grundmasse bestehen; als Grundmassenstoffe kommen z.B.
flüssige Triglyceride, Polyäthylenglykole oder Paraffinkohlenwasserstoffe in Frage.
Zur parenteralen Verabreichung eignen sich in erster Linie wässrige Lösungen eines Wirkstoffs in wasserlöslicher Form, z.B. eines wasserlöslichen Salzes, ferner Suspensionen des Wirkstoffs, wie entsprechende ölige Injektionssuspensionen, wobei man geeignete lipophile Lösungsmittel oder Vehikel, wie fette Öle, z.B. Sesamöl, oder synthetische Fettsäureester, z.B. Äthyloleat oder Triglyceride, verwendet, oder wässrige Injektionssuspensionen, welche viskositätserhöhende Stoffe, z.B. Natrium-Carboxymethylcellulose, Sorbit und/oder Dextran und gegebenenfalls auch Stabilisatoren enthalten.
Pharmazeutische Präparate für topische und lokale Verwendung sind z.B. für die Behandlung der Haut Lotionen und Cremen, die eine flüssige oder semifeste Öl-in-Wasseroder Wasser-in-Öl-Emulsion enthalten, und Salben (wobei solche vorzugsweise ein Konservierungsmittel enthalten), für die Behandlung der Augen Augentropfen, welche die aktive Verbindung in wässriger oder öliger Lösung enthalten und Augensalben, die vorzugsweise in steriler Form hergestellt werden, für die Behandlung der Nase Puder, Aerosole und Sprays (ähnlich den oben beschriebenen für die Behandlung der Atemwege), sowie grobe Puder, die durch schnelles Inhalieren durch die Nasenlöcher verabreicht werden, und Nasentropfen, welche die aktive Verbindung in wässriger oder öliger Lösung enthalten, oder für die lokale Behandlung des Mundes Lutschbonbons,
welche die aktive Verbindung in einer im allgemeinen aus Zucker und Gummiarabikum oder Trafakanth gebildeten Masse enthalten, welcher Geschmacksstoffe beigegeben sein können, sowie Pastillen, die den Aktivstoff in einer inerten Masse, z.B. aus Gelatine und Glycerin oder Zucker und Gummiarabikum, enthalten.
Die nach der Erfindung erhältlichen neuen Verbindungen werden als pharmakologisch wirksame Stoffe verwendet, insbesondere als Antiinflammatorika, Analgetika, Uricosurika, Antiallergika und/oder Thrombolytika, vorzugsweise in Form von pharmazeutischen Präparaten. Die Tagesdosis, die in erster Linie vom Zustand des zu behandelnden Organismus und/oder von der Indikation abhängt, beträgt für einen Warmblüter von etwa 70 kg von etwa 300 mg bis etwa 1 g.
Die nachfolgenden Beispiele illustrieren die oben beschriebene Erfindung. Temperaturen werden in Celsiusgraden angegeben.
Das Ausgangsmaterial wird wie folgt hergestellt:
Eine Suspension von 5,9 g einer 50%igen Natriumhydrid Mineralöl-Suspension in 180 ml Hexamethylphosphorsäuretriamid wird unter Kühlen tropfenweise mit einer Lösung von 18,2 g 2,3-Dihydro-2-oxo-benzo[b]thiophen in 60 ml Hexamethylphosphorsäuretriamid versetzt, wobei die Reaktionstemperatur unter 15 gehalten wird. Nach einstündigem Rühren bei Raumtemperatur werden unter Aussenkühlung portionsweise 30,4 g N-(3-Chlorphenyl)-carbaminsäurephe- nylester zugegeben. Man rührt bei Raumtemperatur 16 Stunden nach und giesst auf ein Gemisch von 100 ml 2n-Salzsäure und 1000 g Eis, wobei sich ein Öl abscheidet, das nach einigen Stunden Stehenlassen kristallisiert. Die Kristalle werden gesammelt und in 300 ml Diäthyläther gelöst.
Die Lösung wird mit Wasser gewaschen, die organische Phase abgetrennt, über Natriumsulfat getrocknet und zur Trockne eingedampft. Der Rückstand wird aus Äther umkristallisiert und ergibt das N-(3-Chlorphenyl)-2-oxo-2,3-dihydro-3benzo[b]thiophencarboxamid vom F. 175-177".
Beispiel I
Zu einer siedenden Suspension von 7 g N-(3-Chlorphenyl) 2-oxo-2,3-dihydro-3-benzo[b]thiophencarboxamid in 200 ml Aceton gibt man 2 ml Morpholin hinzu, wobei alles in Lösung geht. Die nunmehr klare Lösung wird abgekühlt und mit 250 ml Petroläther verdünnt, wobei das Morpholinsalz des N-(3-Chlorphenyl)-2-oxo-2,3-dihydro-3-benzotb]thiophen carboxamides auskristallisiert, das abfiltriert und getrocknet wird. Es schmilzt bei 172,5-173,5'.
Beispiel 2
2,2 g N-(3-Chlorphenyl)-2-oxo-2,3-dihydro-3-benzo[b]- thiophencarboxamid werden in einem Gemisch aus 7,5 ml n-Natronlauge und 30 ml Wasser gelinde erwärmt, wobei bei etwa 50 alles in Lösung geht. Man versetzt mit einer Lösung von 1,1 g Zinksulfat-Heptahydrat in 5 ml Wasser, filtriert nach etwa 30 Minuten den kristallinen Niederschlag des Zinksalzes des N-(3-Chlorphenyl)-2-oxo-2,3-dihydro-3benzo[b]thiophen-carboxamids ab und trocknet diesen. Das Salz schmilzt bei etwa 172" (unter Gasentwicklung).
Beispiel 3
20 g N-(3-Chlorphenyl)-2-oxo-2,3-dihydro-3-benzolbjthio- phen-carboxamid werden in 250 ml Aceton suspendiert und mit 66 ml n-Natronlauge versetzt, worauf Lösung eintritt.
Man dampft zur Trockne ein, verrührt den Eindampfrückstand zunächst mit Toluol und dann mit Diäthyläther, saugt ab und trocknet. Man erhält das Natriumsalz des N-(3-Chlorphenyl)-2-oxo-2,3-dihydro-3-benzo[b]thiophen-carboxamides, F. > 255 .
Tabletten enthaltend 0,1 g N-(3-Chlorphenyl)-2-oxo-2,3 dihydro-3-benzo[bjthiophencarboxamid-Natriumsalz werden wie folgt hergestellt: Zusammensetzung (für 1000 Tabletten): N-(3-chlorphenyl)-2-oxo-2,3-dihydro-3- benzo[b]thiophencarboxamid-Natriumsalz 100,00 g Lactose 50,00 g Weizenstärke 73,00 g Kolloidale Kieselsäure 13,00 g Magnesiumstearat 2,00 g Talk 12,00 g Wasser q.s.
Das N-(2-Fluorophenyl)-2-oxo-2,3-dihydro-3benzo[b]thiophencarboxamid-Natriumsalz wird mit einem Teil der Weizenstärke, mit der Lactose und der kolloidalen Kieselsäure vermischt und das Gemisch durch ein Sieb getrieben. Ein weiterer Teil der Weizenstärke wird mit der fünffachen Menge Wasser auf dem Wasserbad verkleistert und die obige Pulvermischung mit diesem Kleister angeknetet, bis eine schwach plastische Masse entstanden ist. Die plastische Masse wird durch ein Sieb von etwa 3 mm Maschenweite gedrückt, getrocknet und das trockene Granulat nochmals durch ein Sieb getrieben. Darauf werden die restliche Weizenstärke, der Talk und das Magnesiumstearat zugemischt und die erhaltene Mischung zu Tabletten von 0,25 g verpresst.
In analoger Weise können Tabletten enthaltend jeweils 0,1 g N-(3-Chlorphenyl)-2-oxo-2,3-dihydro-3-benzo[b]thiophen- carboxamid als Zink- oder Morpholinsalz hergestellt werden.

which contain the active compound in a mass generally formed from sugar and gum arabic or trafakanth, to which flavorings can be added, and pastilles which contain the active substance in an inert mass, e.g. made of gelatin and glycerin or sugar and gum arabic.
The new compounds obtainable according to the invention are used as pharmacologically active substances, in particular as anti-inflammatory agents, analgesics, uricosurics, antiallergics and / or thrombolytics, preferably in the form of pharmaceutical preparations. The daily dose, which depends primarily on the condition of the organism to be treated and / or on the indication, is from about 300 mg to about 1 g for a warm-blooded animal of about 70 kg.
The following examples illustrate the invention described above. Temperatures are given in degrees Celsius.
The starting material is produced as follows:
A suspension of 5.9 g of a 50% sodium hydride mineral oil suspension in 180 ml of hexamethylphosphoric triamide is added dropwise with cooling with a solution of 18.2 g of 2,3-dihydro-2-oxo-benzo [b] thiophene in 60 ml of hexamethylphosphoric triamide added, the reaction temperature being kept below 15. After stirring for one hour at room temperature, 30.4 g of phenyl N- (3-chlorophenyl) carbamate are added in portions with external cooling. The mixture is stirred at room temperature for 16 hours and poured onto a mixture of 100 ml of 2N hydrochloric acid and 1000 g of ice, an oil separating out which crystallizes after standing for a few hours. The crystals are collected and dissolved in 300 ml of diethyl ether.
The solution is washed with water, the organic phase is separated off, dried over sodium sulfate and evaporated to dryness. The residue is recrystallized from ether and gives the N- (3-chlorophenyl) -2-oxo-2,3-dihydro-3benzo [b] thiophenecarboxamide of F. 175-177 ".
Example I
2 ml of morpholine are added to a boiling suspension of 7 g of N- (3-chlorophenyl) 2-oxo-2,3-dihydro-3-benzo [b] thiophenecarboxamide in 200 ml of acetone, everything dissolving. The now clear solution is cooled and diluted with 250 ml of petroleum ether, the morpholine salt of N- (3-chlorophenyl) -2-oxo-2,3-dihydro-3-benzotb] thiophene carboxamides crystallizing out, which is filtered off and dried. It melts at 172.5-173.5 '.
Example 2
2.2 g of N- (3-chlorophenyl) -2-oxo-2,3-dihydro-3-benzo [b] - thiophenecarboxamide are gently warmed in a mixture of 7.5 ml of n-sodium hydroxide solution and 30 ml of water, whereby at about 50 everything goes into solution. A solution of 1.1 g of zinc sulfate heptahydrate in 5 ml of water is added, and after about 30 minutes the crystalline precipitate of the zinc salt of N- (3-chlorophenyl) -2-oxo-2,3-dihydro-3benzo [b ] thiophene-carboxamids and dries it. The salt melts at about 172 "(with gas evolution).
Example 3
20 g of N- (3-chlorophenyl) -2-oxo-2,3-dihydro-3-benzolbthio-phen-carboxamide are suspended in 250 ml of acetone and mixed with 66 ml of n-sodium hydroxide solution, whereupon solution occurs.
It is evaporated to dryness, the evaporation residue is stirred first with toluene and then with diethyl ether, filtered off with suction and dried. The sodium salt of N- (3-chlorophenyl) -2-oxo-2,3-dihydro-3-benzo [b] thiophene-carboxamides, mp> 255, is obtained.
Tablets containing 0.1 g of N- (3-chlorophenyl) -2-oxo-2,3 dihydro-3-benzo [bjthiophencarboxamide sodium salt are prepared as follows: Composition (for 1000 tablets): N- (3-chlorophenyl) - 2-oxo-2,3-dihydro-3-benzo [b] thiophenecarboxamide sodium salt 100.00 g lactose 50.00 g wheat starch 73.00 g colloidal silica 13.00 g magnesium stearate 2.00 g talc 12.00 g water qs
The N- (2-fluorophenyl) -2-oxo-2,3-dihydro-3benzo [b] thiophencarboxamide sodium salt is mixed with part of the wheat starch, with the lactose and the colloidal silica and the mixture is passed through a sieve. Another part of the wheat starch is gelatinized with five times the amount of water on the water bath and the above powder mixture is kneaded with this paste until a weak plastic mass has formed. The plastic mass is pressed through a sieve with a mesh size of approximately 3 mm, dried and the dry granules are again passed through a sieve. Then the remaining wheat starch, the talc and the magnesium stearate are mixed in and the mixture obtained is compressed into tablets of 0.25 g.
Tablets containing 0.1 g of N- (3-chlorophenyl) -2-oxo-2,3-dihydro-3-benzo [b] thiophene carboxamide as zinc or morpholine salt can be prepared in an analogous manner.
