Die erfindungsgemässe Umsetzung stellt eine Kondensationsreaktion für Amide dar. Sie kann analog zu bekannten Methoden durchgeführt werden.
Sie kann in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel oder Lösungsmittelgemisch in Gegenwart eines säurebindenden Agens erfolgen.
Beispielsweise geht man so vor, dass man als reaktionsfähiges, funktionelles Derivat einer Säure der Formel II das bei der Umsetzung einer Säure der Formel II mit einem Chlorierungsmittel und einem N-di(nieder)alkylsubstituierten Säu
reamid einer aliphatischen Carbonsäure mit 1 bis 3 Kohlenstoffatomen, wie Dimethylformamid oder Dimethylacetamid, entstehende Additionsprodukt verwendet. Ebenso können auch andere reaktionsfähige Derivate einer Säure der Formel II eingesetzt werden, die analog zu bekannten Methoden herstellbar sind, beispielsweise das Säurechlorid-hydrochlorid, das Säureazid oder gemischte Anhydride einer Säure der Formel II mit Schwefelsäure oder Trifluoressigsäure.
Als organische Lösungsmittel sind beispielsweise Chloroform, Methylenchlorid, Acetonitril oder Dimethylformamid geeignet, als säurebindende Agentien tertiäre organische Basen, wie z. B. Pyridin oder dessen Homologe. Als Chlorierungsmittel können beispielsweise Thionylchlorid, Phosgen oder Oxalchlorid eingesetzt werden. Die Umsetzung kann bei Temperaturen zwischen - 30 bis 0 und unter Normaldruck durchgeführt werden.
Zweckmässig verwendet man auf 1 Mol der Verbindung der Formel III in Salzform 1,2 bis 2,4 Mol einer Säure der Formel II. Für die Verbindung der Formel III ist die bevorzugte Salzform das Hydrochlorid. Der Reaktionsablauf ist unabhängig von der Reihenfolge der Zugabe von Reagentien.
Die Aufarbeitung des Reaktionsgemisches und Isolierung der Verbindungen der Formel I kann auf an sich bekannte Weise erfolgen.
Aus den freien Basen lassen sich in bekannter Weise Säureadditionssalze gewinnen und umgekehrt.
Die als Ausgangsprodukt benötigten Verbindungen der Formel II sind neu und können analog zu bekannten Methoden erhalten werden. Beispielsweise geht man so vor, dass man Verbindungen der Formel IV,

worin R1 obige Bedeutung besitzt, unter milden alkalischen Bedingungen, vorteilhaft durch Behandlung mit Natronlauge in einem organischen Lösungsmittel oder Lösungsmittelgemisch, verseift und anschliessend schwach ansäuert.
Die Verbindungen der Formel IV können durch Alkylierung von 6-nor-9,10-Dihydrolysergsäuremethylester erhalten werden.
Die Verbindnung der Formel III ist bekannt.
Die Verbindungen der Formel I in freier Form oder in Form von physiologisch verträglichen Additionssalzen mit Säuren zeichnen sich durch interessante pharmakodynamische Eigenschaften aus. Sie können als Heilmittel verwendet werden. So können sie aufgrund ihrer Vasoaktivität und ihren arteriellen, gefässtonisierenden Eigenschaften zur Anfallscoupierung von Migräneattacken eingesetzt werden. Aufgrund ihrer venentonisierenden Wirkung können sie auch bei der Behandlung orthostatischer Beschwerden Verwendung finden.
Für obige Verwendungen kommen insbesondere die Verbindungen der Formel I, in denen R1 für Isopropyl steht, in Betracht.
Die Verbindungen der Formel I können in freier Form oder in Form ihrer physiologisch verträglichen Additionssalze mit Säuren zur Herstellung von Heilmitteln verwendet werden. Diese Heilmittel, beispielsweise eine Lösung oder eine Tablette, können nach bekannten Methoden, unter Verwendung der üblichen Hilfs- und Trägerstoffe, hergestellt werden.
In dem nachfolgenden Beispiel, welches die Erfindung näher erläutert, ihren Umfang aber in keiner Weise einschränken soll, erfolgen alle Temperaturangaben in Celsiusgraden, ohne Korrekturen. Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten oder analog zu an sich bekannten Verfahren herstellbar.
Die Bezeichnung der Verbindungen der Formel I ist vom Grundgerüst der Formel V

Beispiel
6-nor-6-Isopropyl-9,10-dihydro-2'ss-äthyl 5'a-isopropyl-ergopeptin
Zu einer Lösung von 300 ml Dimethylformamid und 150 ml Acetonitril werden bei -10 bis 150 8,6 ml Oxalylchlorid gelöst in 20 ml Acetonitril innert 10 Minuten zugetropft und dann weitere 10 Minuten gerührt. Darauf werden bei - 200 29,8 g wasserfreie 6-nor-6-Isopropyl-9,10-dihydro-lysergsäure eingestreut und während 30 Minuten bei 100 gerührt. Nach Abkühlung auf 200 werden 200 ml Pyridin und 16,6 g (2R,5S,10aS,10bS)-2-Amino-2-äthyl-5-isopropyl- 3,6-dioxo-l Ob-hydroxy-octahydro-8H-oxazolo- [3,2-a]pyrrolo- ,1-c]pyrazin-hydrochlond zugegeben und während 2 Stunden bei 0 gerührt.
Zur Aufarbeitung werden 100 ml Pufferlösung pH = 4 zugefügt und das Reaktionsgemisch zwischen Methylenchlorid und 2N Sodalösung verteilt. Die organischen Phasen werden zweimal mit Wasser gewaschen, mit Natriumsulfat getrocknet und am Rotavapor zur Trockne eingeengt.
Die erhaltene Rohbase wird nach dem Trocknen am Hochvakuum in etwa 150 ml Äthanol gelöst und die Lösung angeimpft. Die Titelverbindung schmilzt bei 176-1780 unter Zer 20 setzung; [a] 2D0= 230 (c = 0,5 in Methylenchlorid).
Zu der als Ausgangsmaterial eingesetzten 6-nor-6-Isopropyl-9,10-dihydrolysergsäure kann man z. B. durch Alkylierung von 6-nor-9,10-Dihydrolysergsäuremethylester mit Isopropylbromid und Verseifung des so erhaltenen 6-nor-6-Iso propyl-9, 10-dihydrolysergsäuremethylesters (Smp. 1940) gelangen.
6-nor-6-Isopropyl-9, 10-dihydrolysergsäure schmilzt bei 20 290 unter Zersetzung; [a] 2D0= - 1010 (c = 0,6 in Methanol).
