1. Verfahren zur Herstellung von Verbindungen der Formel:

2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man die Umsetzung in Gegenwart eines Alkalis durchführt.
3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man die Umsetzung bei einer Temperatur im Bereich von Raumtemperatur bis zum Siedepunkt des verwendeten Lösungsmittels durchführt.
4. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man als inertes Lösungsmittel Methanol, Äthanol, n-Propanol, Isopropanol, n-Butanol, tert.-Butanol, Methoxy äthanol, Äthoxyäthanol, Tetrahydrofuran, Dioxan, Wasser, Chloroform, Tetrachlorkohlenstoff, 1,2-Dichloräthan, 1,1,1 Trichloräthan, Benzol oder Toluol oder eine Mischung davon verwendet.
5. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man Chlor oder Brom in wenigstens einer äquimolaren Menge, bezogen auf das 3,4-Dihydro-2(1 H)-chinazolinon der Formel II, einsetzt.
Die Erfindung betrifft ein neues Verfahren zur Herstellung von 2(1H)-Chinazolinon-Derivaten.
Bislang war ein Verfahren zur Überführung eines 3,4-Dihydro-2(1H)-chinazolinon-Derivats in das entsprechende 2(1 H)-Chinazolinon-Derivat mit hervorragenden pharmakologischen Eigenschaften, wie entzündungshemmender, analgetischer, uricosurischer und/oder antivireller Wirksamkeit, bekannt, wobei ein Schwermetall-Oxydationsmittel, wie eine Manganverbindung, Chromverbindung oder ein Salz davon (z.B. Kaliumpermanganat, Mangandioxid oder Chromtri- oxid), verwendet wurde, wie es in der kanadischen Patentschrift Nr. 949 570 beschrieben ist.
Diese im früheren Verfahren verwendeten Oxydationsmittel sind verhältnismässig teuer und können ferner die Ursache schwieriger Probleme bei der Abfallbehandlung und -verwertung im Hinblick auf die Umweltverschmutzung sein, insbesondere im Falle des Überganges auf die Erzeugung im grossen Massstab.
Bei den Untersuchungen zur Überwindung der Schwierigkeiten des bekannten Verfahrens wurde festgestellt, dass 3,4 -Dihydro-2( 1 H)-chinazolinon-Derivate durch Einwirkung von Chlor oder Brom oder einem Derivat der unterchlorigen bzw.
unterbromigen Säure leicht in die entsprechenden 2(1 H)- -Chinazolin-Derivate mit hoher Reinheit übergeführt werden können.
Die vorliegende Erfindung schafft ein neues Verfahren zur Herstellung von Verbindungen der Formel (I)

Bei den Verbindungen der Formel (I) und in dieser Be
schreibung bedeutet der-Ausdruck Alkyl sowohl geradwie verzweigtkettige, gesättigte, aliphatische, Kohlenwasserstoff-Reste und niedriges Alkyl kann Methyl, Äthyl, n-Propyl, Isopropyl, n-Butyl oder Isobutyl sein; der Ausdruck niedriges Alkoxy kann Methoxy, Äthoxy, n-Propoxy, Isopropoxy, n-Butoxy oder Isobutoxy, niedriges Cycloalkyl , kann Cyclopropyl, Cyclobutyl, Cyclopentyl, Cyclohexyl, Cycloheptyl oder Cyclooctyl bedeuten, Aralkyl kann z.B. Bei der Ausführung des erfindungsgemässen Verfahrens wird die Umsetzung vorzugsweise bei einer Temperatur im Bereich von Raumtemperatur bis zum Siedepunkt des verwendeten Lösungsmittels ausgeführt.
Zu Beispielen von Alkali gehören Natriumhydroxid, Kaliumhydroxid, Natriumcarbonat, Kaliumcarbonat, Natriumbicarbonat, Kaliumbicarbonat, Natriumacetat, Kaliumacetat, Natriummethoxid, Natriumäthoxid, Kalium-tert. -Butoxid u. dgl.
Zu geeigneten inerten Lösungsmitteln gehören beispielsweise Methanol, Äthanol, n-Propanol, Isopropanol, n-Butanol, tert.-Butanol, Methoxyäthanol, Äthoxyäthanol, Tetrahydrofuran, Dioxan, Wasser, Chloroform, Tetrachlorkohlenstoff, 1,2-Dichloräthan, 1,1,1-Trichloräthan, Benzol, Toluol oder Mischungen davon.
Chlor oder Brom können in wenigstens äquimolaren Mengen, insbesondere in äquimolarer Menge bis zum mehrfachen Gewicht des 3,4-Dihydro-2(1 H)-chinazolinons der Formel (II) eingesetzt werden. Für die Umsetzung wird Chlor oder Brom auch in Form eines Derivats der unterchlorigen bzw. unterbromigen Säure, wie Natriumhypochlorit, Natriumhypobromit, Kaliumhypobromit oder tert.-Butylhypochlorit, verwendet, das nach einem bekannten Verfahren einfach durch Zugabe von Chlor oder Brom zu einer gekühlten Lösung von Natriumhydroxid oder Kaliumhydroxid hergestellt werden kann.
Unter bestimmten Bedingungen kann man ein 3-halogensubstituiertes 3 ,4-Dihydro-2( I H)-chinazolinon-Derivat als Zwischenprodukt der Umsetzung erhalten. Das Zwischenprodukt ist verhältnismässig instabil und kann leicht durch Erhitzen oder Behandeln mit einer anorganischen oder organischen Base, wie Alkalihydroxid, Alkalialkoxid, Ammoniak, Triäthylamin, Pyridin. od. dgl. zum Endprodukt umgewandelt werden.
Die Umsetzung wird üblicherweise ohne Isolierung des Zwischenproduktes ausgeführt. Bei dieser Umsetzung können die Verbindungen der Formel (I) im wesentlichen als Hydrochlorid oder Hydrobromid hergestellt werden, welches durch Behandlung mit einem Alkali leicht das Hydrohalogenid freisetzt.
Die Erfindung wird durch die nachfolgenden Beispiele, welche der Erläuterung dienen, ohne den Rahmen der Erfindung zu beschränken, an bevorzugten Ausführungsformen näher beschrieben.
Beispiel 1
Einer Lösung von 3,13 g 1-Cyclopropylmethyl-4-phenyl -6-chlor-3,4-dihydro-2(1H)-chinazolinon in 60 ml heissem Methanol fügt man bei 60 bis 65"C tropfenweise 3 g Brom zu, hält das Gemisch unter Rühren während einer Stunde bei milden Rückflussbedingungen und entfernt dann das Lösungsmittel unter vermindertem Druck, um das Hydrobromid des Produktes als orangefarbenen Rückstand zu erhalten. Diesem Rückstand werden 50 ml Ammoniakwasser zugefügt und die erhaltene Suspension wird gut gerührt.
Den farblosen Niederschlag sammelt man durch Filtration, wäscht mit Wasser, trocknet und erhält 3,06 g l-Cyclopro- pylmethyl-4-phenyl-6-chlor-2( 1 H)-chinazolinon, Fp. 174 bis
1750C.
Beispiel 2
Einer Suspension von 3,13 g 1 -Cyclopropyl-methyl-4- -phenyl-6-chlor-3,4-dihydro-2( 1 H)-chinazolinon und 3,2 g Natriummethoxid in 60 ml Methanol fügt man tropfenweise eine Lösung von 5 g Brom in 20 ml Methanol zu, hält das Gemisch unter Rühren während 6 Stunden auf Rückflussbedingungen, entfernt dann das Lösungsmittel unter vermindertem Druck und verreibt den Rückstand mit wässeriger Natriumbicarbonat-Lösung. Der Niederschlag wird durch Filtration gesammelt, mit Wasser gewaschen und getrocknet und ergibt 3,06 g 1-Cyclopropyl-methyl-4-phenyl-6 -chlor-2(1H)-chinazolinon, Fp. 172- 173"C.
Beispiel 3
Einer Lösung von 3,13 g 1 -Cyclopropylmethyl-4-phenyl -6-chlor-3 ,4-dihydro-2( 1 H)-chinazolinon in 20 ml Chloroform fügt man tropfenweise 3,8 g Brom zu, rührt das Gemisch während 10 Stunden bei Raumtemperatur, entfernt dann das Lösungsmittel unter vermindertem Druck, wäscht die zurückbleibenden orangefarbenen Kristalle mit Isopropanol-Isopropyläther und'trocknet, um 3,94 g l-Cyclopro pylmethyl-4-phenyl-6-chlor-2( 1 H)-chinazolinon-Hydrobromid, Fp. 187 - 1890C zu erhalten.
Beispiel 4
Einer Lösung von 3,08 g 1-Cyclopropylmethyl-4-phenyl -6-methoxy-3,4-dihydro-2(1H)-chinazolinon in 15 ml heissen Dioxans fügt man tropfenweise 2,3 g Brom zu und rührt das Gemisch während 6 Stunden bei 75 bis 80"C. Nach dem Abkühlen wird der Niederschlag abfiltriert, mit Äthanol-n -Hexan gewaschen und getrocknet und ergibt 3,8 g 1-Cyclo propylmethyl-4-phenyl-6.methoxy-2( 1 H)-chinazolinon-Hy- drobromid als orangefarbene Kristalle, welche aus Methanol Isopropanol umkristallisiert werden, um orangefarbene Nadeln, Fp. 196,5 - 1970C (Zers.) zu ergeben.
Beispiel 5
Einer Lösung von 3,08 g 1-Cyclopropylmethyl-4-phenyl -6-methoxy-3,4-dihydro-2( 1 H)-chinazolinon in 30 ml Äthanol fügt man bei 75"C tropfenweise eine Lösung von 4,8 g Brom in 15 ml Äthanol zu und rührt das erhaltene Gemisch während 2 Stunden bei 75"C. Dann fügt man dem Reaktionsgemisch anteilsweise bei 60 - 709C unter Rühren 3 g Natriumäthoxid zu, destilliert danach den Hauptteil des Äthanols ab und setzt dem Rückstand 50 ml Wasser zu, um das anorganische Salz aufzulösen und das Produkt zu kristallisieren. Der erhaltene gelbe Niederschlag wird durch Filtration gesammelt, mit Wasser gewaschen und getrocknet und ergibt 3,02 g 1-Cyclopropylmethyl-4-phenyl-6-methoxy-2(1H)- -chinazolinon, Fp. 114 - 1 150C.
Beispiel 6
Einer Lösung von 3,08 g 1-Cyclopropylmethyl-4-phenyl -6-methoxy-3 ,4-dihydro-2(1 H)-chinazolinon und 0,8 g Natriumhydroxid in 30 ml heissem Methanol fügt man bei 65 bis 70"C anteilsweise 15 g Natriumhypochlorit-Lösung (mit 12% verfügbarem Chlor) zu, welche aus Chlor und wässerigem Natriumhydroxid nach einem bekannten Verfahren hergestellt war. Das Gemisch wird während einer weiteren Stunde unter Rühren bei 70"C erhitzt. Nach dem Abkühlen fügt man dem Reaktionsgemisch unter Rühren 30 ml Wasser zu, kühlt das erhaltene Gemisch in Eiswasser, saugt den Niederschlag am Filter ab, wäscht mit Wasser und trocknet, um 2,9 g 1-Cyclopropylmethyl-4-phenyl-6-methoxy-2(1H)-china- zolinon, Fp. 114-1 150C zu erhalten.
Beispiel 7
Einer Lösung von 3,41 g 1-(2,2,2-Trifluoräthyl)-4-phe- nyl-6-chlor-3,4-dihydro-2(1H)-chinazolinon in 20 ml heissem Methanol fügt man tropfenweise bei etwa 60 C eine Lösung von 4,0 g Brom in 5 ml Methanol zu, erhitzt das Gemisch unter Rühren während 2 Stunden bei 60 C und fügt dann dem Reaktionsgemisch bei 50 - 60 C anteilsweise 40 g einer 10%igen wässerigen Natriumcarbonatlösung zu.
Nach dem Abkühlen wird der farblose Niederschlag durch Absaugen filtriert, mit Wasser gewaschen und getrocknet und ergibt 3,35 g 1-(2,2,2-Trifluoräthyl)-4-phenyl-6-chlor -2(1H)-chinazolinon, Fp. 183,5 - 184,50C.
Beispiel 8
Unter Anwendung von Verfahren, die den in den Beispielen 1 bis 7 beschriebenen ähnlich sind, wobei man von einem entsprechenden 3,4-Dihydro-2(1H)-chinazolinon-De- rivat ausgeht, werden die nachstehenden Verbindungen in ausgezeichneten Ausbeuten erhalten.

Example 2
A solution is added dropwise to a suspension of 3.13 g of 1-cyclopropyl-methyl-4-phenyl-6-chloro-3,4-dihydro-2 (1 H) -quinazolinone and 3.2 g of sodium methoxide in 60 ml of methanol 5 g of bromine in 20 ml of methanol are added, the mixture is kept at reflux conditions with stirring for 6 hours, the solvent is then removed under reduced pressure and the residue is triturated with aqueous sodium bicarbonate solution. Example 3
3.8 g of bromine are added dropwise to a solution of 3.13 g of 1-cyclopropylmethyl-4-phenyl-6-chloro-3, 4-dihydro-2 (1 H) -quinazolinone in 20 ml of chloroform, and the mixture is stirred while 10 hours at room temperature, then the solvent is removed under reduced pressure, the remaining orange-colored crystals are washed with isopropanol-isopropyl ether and dried to obtain 3.94 g of l-cyclopropylmethyl-4-phenyl-6-chloro-2 (1 H) - quinazolinone hydrobromide, m.p. 187-1890C.
Example 4
2.3 g of bromine are added dropwise to a solution of 3.08 g of 1-cyclopropylmethyl-4-phenyl -6-methoxy-3,4-dihydro-2 (1H) -quinazolinone in 15 ml of hot dioxane and the mixture is stirred while 6 hours at 75 to 80 "C. After cooling, the precipitate is filtered off, washed with ethanol-n-hexane and dried, giving 3.8 g of 1-cyclopropylmethyl-4-phenyl-6th methoxy-2 (1H) -quinazolinone-hydrobromide as orange crystals, which are recrystallized from methanol isopropanol to give orange needles, mp 196.5-1970C (decomp.).
Example 5
A solution of 3.08 g of 1-cyclopropylmethyl-4-phenyl-6-methoxy-3,4-dihydro-2 (1H) -quinazolinone in 30 ml of ethanol is added dropwise at 75 ° C. to a solution of 4.8 g Bromine in 15 ml of ethanol is added and the mixture obtained is stirred at 75 ° C. for 2 hours. Then 3 g of sodium ethoxide are added to the reaction mixture at 60-709C with stirring, the main part of the ethanol is then distilled off and 50 ml of water are added to the residue to dissolve the inorganic salt and crystallize the product. Example 6
A solution of 3.08 g of 1-cyclopropylmethyl-4-phenyl-6-methoxy-3, 4-dihydro-2 (1 H) -quinazolinone and 0.8 g of sodium hydroxide in 30 ml of hot methanol is added at 65 to 70 " C proportionally 15 g of sodium hypochlorite solution (with 12% available chlorine), which was prepared from chlorine and aqueous sodium hydroxide by a known process. The mixture is heated at 70 ° C. for a further hour while stirring. A solution of 3.41 g of 1- (2,2,2-trifluoroethyl) -4-phenyl-6-chloro-3,4-dihydro-2 (1H) -quinazolinone in 20 ml of hot methanol is added dropwise about 60 ° C., a solution of 4.0 g of bromine in 5 ml of methanol is added, the mixture is heated at 60 ° C. with stirring for 2 hours and then 40 g of a 10% aqueous sodium carbonate solution are added to the reaction mixture at 50 ° -60 ° C.
