REVENDICATIONS
1. Procédé de N-acylation de l'homocystéine ou de l'un de ses esters, thiolesters, sels ou un composé S-acylé d'homocys téïne, caractérisé en ce que l'on soumet à une N-acylation l'homocystéine ou l'un de ses esters, thiolesters, sels ou un composé S-acylé d'homocystélne.
2. Procédé selon la revendication 1, caractérisé en ce que ledit N-acyl homocystéine obtenu est soumis à une S-acylation pour obtenir le N,S-acyl homocystéïne.
3. Procédé selon la revendication 1, caractérisé en ce que l'on emploie pour la N-acylation un anhydride d'acide ou un halogénure d'acide, en présence d'eau et d'un agent alcalin.
4. Procédé selon la revendication 1, caractérisé en ce que l'on emploie pour la N-acylation l'anhydride de l'acide propionique ou butyrique ou le chlorure d'acétyle, de propionyle, de butyryle, de benzoyle, de nicotinoyle ou de cinnamoyle substitué.
5. Procédé selon la revendication 1, caractérisé en ce que l'on soumet un ester ou un thiolester d'homocystéïne à une Nacylation.
6. Procédé selon la revendication 5, caractérisé en ce que l'on soumet à une S-acylation les composés obtenus après Nacylation.
7. Procédé selon la revendication 1, caractérisé en ce que l'on soumet à une estérification avec un alcool en présence d'acide.
8. Procédé selon la revendication 1, caractérisé en ce que l'on soumet à une thiolestérification des homocystéines, Nacylées et S-acylées en les faisant réagir avec un mercaptan en présence d'acide.
9. Procédé selon la revendication 1 pour la préparation de
N,S-diacylhomocystéïnes à partir de l'homocystéine ou de l'un de ses sels, caractérisé en ce que l'on commence par N-acyler l'homocystéine en présence d'un agent alcalin puis en ce que l'on S-acyle en milieu aqueux alcalin avec des anhydrides d'acides ou des halogénures d'acides identiques ou différents, sans isoler le N-acyl-homocystéïne intermédiaire.
10. Procédé selon la revendication 1, caractérisé en ce que l'on prépare par N-acylation un composé de formule (A) SCH2- CH2-CH(NHR CO, dans laquelle R représente un groupe acyle renfermant plus de 4 atomes de carbone qui est substitué quand il signifie benzoyl ou nicotinoyl et non substitué quand il signifie cinnamoyl, ou un composé de formule (B) R'-S-CH2 CH2-CH(NHR)-CO-X-Ri', dans laquelle X représente un oxygène ou un soufre, R' représente un hydrogène ou un radical acyle et R' un hydrogène ou un alcoyle éventuellement substitué, tels que R" porte au moins deux carbones quand X est de l'oxygène, R un acyl inférieur et R' de l'hydrogène, et ses sels d'acides ou de bases organiques ou minérales.
11. Procédé selon la revendication 1 pour la préparation de
N-acétylhomocystéïnenate de méthyle.
12. Procédé selon la revendication 6 pour la préparation de
N,S-diacétyl benzoyl ou m-fluorobenzoyl homocystéine; Nacétyl-S-benzoyl, m-fluorobenzoyl, thénoyl-2, p-chlorophénoxyacétyl, salicyloyl, pivaloyl, succinyl, homocystéine ou Nacétyl-S-benzoyl homocystéinates de méthyle, éthyle, propyle-2 ou butyle-2.
13. Application du procédé selon la revendication 1 en ce que l'on soumet à la N-acylation un composé préparé d'un dérivé S-alcoyle de l'homocystéine que l'on désalcoyle et que l'on fait ensuite réagir avec un et éventuellement deux anhy
drides ou halogénures d'acides en présence d'eau et d'agents
alcalins, sans isoler aucun composé intermédiaire.
14. Application selon la revendication 13 pour la préparation de N,S-diacétyl benzoyl ou m-fluorobenzoyl homocys téine; N-acétyl-S-benzoyl, m-fluorobenzoyl, théonyl-2, p-chlorophénoxyacétyl, salicyloyl, pivaloyl, succinyl, homocystéine ou N-acétyl-S-benzoyl homocystéinate de méthyle, éthyle, propyle-2 ou butyle-2.
L'amino-3 dihydrothiophénone-2 ou homocystélne thiolactone, appelée ADT ci après, fut décrite par du Vigneaud en
1935. On la prépare par désalcoylation de la méthionine ou autre alcoylhomocystéine par réduction au sodium dans l'ammoniac ou l'acide iodhydrique, suivie d'une cyclisation. Encore peu développée, elle est relativement onéreuse car soluble dans l'eau, son isolement est délicat. On en connaît quelques dérivés N-acylés, mais les S-acyl homocystéines sont inconnues. Les nouvelles réactions de l'invention donnent accès, parfois très économiquement à de nombreux dérivés utiles en thérapeutique, cosmétologie, photographie, etc., et permettant un accès plus économique aux dérivés connus.
L'invention concerne donc un procédé de N-acylation de l'homocystélne ou de l'un de ses dérivés, caractérisé en ce que l'on soumet à une N-acylation l'homocystéine ou l'un de ses esters, thiolesters, sels ou un composé S-acylé d'homocystélne.
Les composés sont préparés à l'état libre ou de sels d'acides ou bases organiques ou minéraux, des dérivés de l'ADT dont la formule comporte le groupement -S-CH2-
CH2-CH(NHR)-CO- dans lequel R représente un hydrogène ou un acyle. La structure des com osés est soit cyclique suivant la formule (A) S-CH2-CH2CH(NHR)-CO où R porte alors plus de 4 carbones et est substitué quand il est un benzoyl ou nicotinoyl et non substitué quand il est un cinnamoyl, soit
ouvert suivant la formule (B) R'-SCH2-CH2-CH(NHR
CO-X-R'', où X représente un soufre ou oxygène, R et R', identiques ou différents, de l'hydrogène ou des acyles, R un hydrogène ou un alcoyle éventuellement substitué, tels que R" porte au moins deux carbones si X est de l'oxygène,
R un benzoyl ou acyl inférieur et R' de l'hydrogène.
D'une part, les composés ont la structure cyclique (A) et R est un radical de la liste suivante: p.chlorophénoxy-2 isobutyryl, pivaloyl, m.fluorobenzoyl, triméthoxy-3, 4benzoylsuccinoyl, p.chlorophénoxy-acétyl, salicyloyl, acétylsalicyloyl, thénoyl-2, chromone-2carbonyl.
D'autre part, les composés ont la structure ouverte (B) où X est de l'oxygène, R' et R" de l'hydrogène et R l'un des radicaux de l'alinéa précédent, plus les radicaux nicotinoyl et propionyl; ou -XR;' est un -OH et R et R, identiques ou différents, l'un des radicaux de la liste suivante: acétyl, propionyl, pivaloyl, succinoyl, benzoyl, m fluorobenzoyl, triméthoxy-3,4,5benzoyl, salicyloyl, acétysalicyloyl, thénoyl-2, p.chlorophénoxyacétyl, p.chlorophénoxy-2isobutyryl, nicotinoyl, chromone-2carbonyl.
En outre on peut préparer des homologues des produits de l'alinéa précédent où R'' devient un radical méthyle, éthyle, propyl ou butyle.
Les N-acyl ADT ont été obtenus uniquement par acylation de l'ADT en milieux anhydres, et les N-acy ADT de l'invention peuvent être ainsi obtenus. Une seule référence, Yoshitomi, Brevet Japonais 16712/62', fait état de l'acétylation de l'ADT dans une solution de bicarbonate par l'anhydride acétique.
Le procédé selon l'invention peut être exécuté en ce qu'on dissout l'ADT dans un solvant aqueux, préférentiellement l'eau. On ajoute une base faible comme un bicarbonate, ou plus forte, telle que la soude, mais en quantité calculée pour éviter l'hydrolyse de la thiolactone. On ajoute à cette solution un anhydride ou un halogénure d'acide à une température spécifique du réactif, souvent optimale entre - 10 et l'am- biante, l'anhydride acétique étant exclu des réactifs utilisés.
Une S-acylation des ADT N-acylées ou non, de préférence
dans l'eau peut être effectuée en deux étapes, hydrolyse de la thiolactone puis acylation. L'hydrolyse s'accélère quand le pH et la température s'élèvent. Ensuite, on abaisse ces paramètres et ajoute un halogénure ou anhydride d'acide. De préférence on utilise la soude en excès et acyle entre - 10 et l'ambiante.
En fin de réaction, on acidifie et isole la S-acylhomocystéine.
Cette technique permet d'accéder à des composés identiques ou différemment acylés sur le soufre et l'azote.
Pour préparer les esters ou thiolesters d'homocystéïnes Sacylés on sature d'acide chlorhydrique anhydre par exemple, un solvant qui peut être l'alcool ou mercaptan réactif., on dissout l'homocystéîne et abandonne le mélange souvent à température ambiante. On isole l'ester en évaporant le solvant et
rajoutant au résidu une solution de bicarbonate pour éliminer
l'homocystéine résiduelle.
Pour la N et S-acylation des esters et thiolesters d'homocys
téines on additionne au substrat un anhydride ou halogénure d'acide en milieu alcalin très doux, soit anhydre, soit aqueux à
pH modéré et à froid.
Pour préparer les S-acyl homocystéïnes on peut S-acyler un
ester préalablement préparé, puis l'hydrolyser par action d'un
réactif alcalin, généralement à froid.
Pour les préparations des N-acyl ADT et homocystéines, N et/ou S acylées ou non directement à partir des S-alcoylhomo
cystéines N-acylées ou non, de préférence la méthionine ou
l'acétyl-méthionine, on commence par désalcoyler le substrat suivant les procédés connus, on évapore le solvant et sans
isoler le produit intermédiaire, on acyle successivement sur l'azote et sur le soufre suivant les procédés précédemment
décrits. Si la déalcoylation a lieu dans l'acide iodhydrique, le
résidu d'évaporation est dissous dans l'eau. On N-acyle à pH
doux, puis augmente le pH pour S-acyler. Si la déalcoylation se
fait par le sodium dans l'ammoniac, on évapore ce derbier et dissout le résidu dans l'eau. Pour N- et S-acyler identiquement,
on acyle directement.
Pour N- et S-acyler différemment, il faut
recycliser en ADT en revenant en milieu acide, retourner à pH
alcalin doux pour N-acyler puis à pH plus élevé pour S-acyler.
Ces moyens sont très économiques car ils évitent le délicat isolement de l'ADT; seul le produit final est à isoler, et sou
vent, insoluble, il précipite directement.
Pour préparer des N,S-diacylhomocystéïnes on peut sou
mettre les produits N-acylés obtenus selon le procédé de l'in
vention à une S-acylation sans isoler le N-acyl ADT intermé
diaire.
Avec les produits obtenus on peut préparer des médica
ments pour la thérapeutique humaine ou vétérinaire qui
contiennent un ou plusieurs de ces nouveaux dérivés obtenus.
On a vérifié que certains présentent peu de toxicité et diverses
activités telles que des actions hépatoprotectrices, cicatrisantes,
mucolytiques, hypolipémiantes, hypocholestérolémiantes,
vasoprotectrices, calmantes, antiagrégante plaquettaire. . .. Le
support homocystéïne est par lui-même atoxique et possède
d'intéressantes propriétés thérapeutiques qui peuvent être
complétées, synergisées ou modifiées par des restes d'acides,
alcools et mercaptans qui possèdent en propre des propriétés
thérapeutiques et dont les produits de l'invention peuvent
servir de vecteur d'introduction pour améliorer leurs qualités
pharmacologiques ou résoudre des problèmes de stabilité,
solubilité, effet retars, passage de parois ou autres.
Les compositions préparées et les formes pharmaceutiques pour l'administration par voies locale, orale, parenterale, rectale, en otorhinolaryngologie et ophtalmologie telles que
pommades, solutions, laits, aérosols, pilules, cachets, dragées,
gélules, chronules, sirop, capsules, gouttes, solutés à boire ou à
injecter, tout prêts ou à préparer à l'emploi, cônes, supposi
toires, lavements, qui renferment à l'état pur ou en mélange un
ou plusieurs dérivés de l'invention.
Les exemples et tableaux suivants illustrent l'invention dont on peut imaginer de nombreuses autres variantes.
Exemple I
Procédé de synthèse A-N-acétyl ADT
On dissout 1,535 g d'ADT,HCl dans 10,1 ml de soude N et ajoute 1,1 ml d'anhydride acétique. La température croît de 22 à 290, 10 minutes après, on ajoute 4 ml d'acide acétique et évapore à sec. On reprend par l'acétone filtre le sel et évapore le filtrat. En ajoutant de l'éther, le produit cristallise.
Exemple 2
Procédé B N-propionyl ADT
Suivant l'exemple 1, en remplaçant la quantité calculée de soude par un excès de bicarbonate de sodium, et l'anhydride acétique par l'anhydride propionique.
Exemple 3
Procédé C N-p.chlorophénoxy-2isobutyryl ADT
Dissoudre 3,07 g d'ADT, HCl dans 20 ml de diméthylformamide. Chauffer jusqu'à dissolution, refroidir à 50O et ajouter 5,8 ml de triéthylamine dont le chlorhydrate précipite, puis 4,70 g de chlorure de clofibryle. La température monte à 70 .
Chauffer 2 minutes à 1000 et refroidir, filtrer et ajouter 80 ml d'eau au filtrat; une huile précipite et cristallise rapidement.
Exemple 4
Procédé D N-pivaloyl ADT
Suivant l'exemple 2, avec le chlorure de pivaloyle au lieu de l'anhydride propionique.
Exemple 5
Procédé N,S-diacétyl homocystéine
On dissout 11,2 g de N-acétyl ADT dans 40 ml de soude à 14%, et à 10o on ajoute doucement 7,15 g d'anhydride acétique. Après quelques minutes, on acidifie avec HCI 10%, sature de sel dont on filtre l'excès, On extrait avec du chlorure de méthylène, évapore à secet reprend à l'éther. Une huile précipite et cristallise par la suite.
Exemple 6
Procédé F N,S-diacétylhomocystéïne
Même procédé que dans l'exemple 5, mais en utilisant l'ADT, HCI dont la quantité molaire est triplée en soude et doublée en anhydride acétique.
Exemple 7
Procédé G N,S-diacétylhomocystéïne
Suivant l'exemple 5, en remplaçant l'anhydride par le chlorure d'acétyle et opérant à 0".
Exemple 8
Procédé H N,S-diacétylhomocystéïne
Suivant l'exemple 6, en remplaçant l'anhydride par le chlorure d'acétyle et opérant à 00.
Exemple 9
Procédé I N,S-dibenzoylhomocystéine
Dissoudre 1,54 de ADT,HCI dans 20 ml de soude 2N et, à
100, ajouter doucement sous agitation 2,9 g de chlorure de benzoyle. Quand le mélange est limpide, ajouter HCI 10% jusqu'à pH 2: le produit précipite et cristallise en glaçant.
Exemple 10
Procédé J N-acétyl,S-benzoylhomocystéine
Dissoudre 1,59 g de N-acétyl ADT dans 15 ml de soude 2N.
A 100, ajouter doucement 1,45 g de chlorure de benzoyle.
Revenir à 200 et lorsque le mélange est limpide, verser HCI
10% jusqu'à pH 2. Le produit précipite et cristallise rapidement.
Exemple il
Procédé K N-acétyl,S-benzoyl homocystéinate de méthyle
Introduire 3,28 g de N-acétylhompcystéïnate de méthyle dans 30 ml de benzène sec et 2,06 g de triéthylamine. A 10 , introduire 3,10 g de chlorure de benzoyle, abandonner 2 jours
et filtrer le sel. Laver le filtrat avec HCl dilué, du bicarbonate, puis de l'eau. Sécher la phase benzénique et évaporer à sec, ajouter une solution de bicarbonate: le produit cristallise.
Exemple 12
Procédé L N-acétyl,S-benzoylhomocystéïnate de méthyle
Les réactifs de l'exemple précédent sont traités dans la soude normale. Le précipité formé est immédiatement essoré et lavé à l'eau, ne donnant qu'un faible rendement à cause de l'hydrolyse.
Exemple 13
Procédé M N-acétyl,S-benzoylhomocystéinate de méthyle
Dans l'exemple précédent, en remplaçant la soude par du bicarbonate en excès et en laissant revenir à 20 , on complète la réaction et évite l'hydrolyse, ce qui conduit à un excellent rendement.
Exemple 14
Procédé N N-acétyl,Sp.chlorophénoxy 2isobutyrylhomocystéine
On agite une heure l'ester méthylique correspondant dans de la soude normale, puis on acidifie à pH 2 avec HCl 10%. Le produit précipite et cristallise rapidement.
Exemple 15
Procédé O N-acétyl,S-benzoylhomocystéinate de méthyle
Dissoudre 1,41 g de N-acétyl,S-benzoylhomocystéine dans 15 ml d'alcool méthylique saturé d'acide chlorhydrique anhydre. Abandonner 12 heures et évaporer à sec. Reprendre le résidu par du bicarbonate à 10%. En glaçant, le produit cristallise.
Exemple 16
Procédé P N-acétyl-S-benzoylhomocystéinate
de N-acétylhomocystéïnate de méthyle
Des quantités équimolaires de N-acétyl,S-benzoylhomocys téine et de N-acétyl homocystéïnate de méthyle sont dissous dans du dioxanne chlorhydrique. Abandonner une journée et traiter comme dans l'exemple précédent.
Exemple 17
Procédé R N-acétyl,S-benzoylhomocystéïne
Dissoudre 1,69 g d'ADT,HCl dans 22 ml de bicarbonate
10% et à 20' ajouter 1,3 ml d'anhydride acétique. On laisse
réagir 10 minutes refrigère à 15 et ajoute en agitant 1,41 g de chlorure de benzoyle. Quand la solution est limpide, on acidifie à pH 2 avec HCl 10%: le produit précipite et cristallise.
Exemple 18
Procédé Q N-thénoyl-2ADT
On déméthyle 15 g de méthionine dans l'acide iodhydrique
à 40% suivant les procédés connus, évapore l'acide à sec sous vide et reprend le résidu dans 100 ml de bicarbonate à 10%, et en s'assurant que le milieu est toujours alcalin, ajouter doucement 14,65 g de chlorure de thénoyle à 15 . Le produit se
concrétise en 1/2 heure et on le filtre.
Exemple 19
Procédé S N-acétylADT
On déméthyle 7,5 g de méthionine par réduction au sodium dans l'ammoniac et évapore ce dernier. Par précaution on détruit avec 5 ml d'alcool puis neutralise à pH 2 avec HCI
10%. On chauffe 'heure à 80 pour cycliser et neutralise avec
du bicarbonate en excès et ajoute à 200 6 ml d'anhydride
acétique et procède à l'isolement comme dans l'exemple 1.
Exemple 20
Procédé T N,S-diaeétylhomocystéine
On déméthyle de même 7,5 g de méthionine par le sodium dans l'ammoniac que l'on évapore. Après destruction du résidu à l'alcool, on étend à l'eau, refroidit à 15-20 et ajoute 12 ml d'anhydride acétique. Après quelques minutes, acidifier et isoler ainsi que dans l'exemple 5.
Exemple 21
Procédé V N,S-dibenzoylhomocystéïne
Suivant l'exemple 20, en remplaçant l'anhydride acétique par le chlorure de benzoyle. En fin de réaction, dès que l'on acidifie, le produit précipite et cristallise.
Exemple 22
Procédé V N-acétyl,S-thénoyl-2homocystéïne
On procède comme dans l'exemple 19 mais n'isole pas la Nacétyl ADT. On ajoute à ce moment au mélange 5 g de soude en pastilles, refroidit à 15o et ajoute 7,35 g de chlorure de thénoyle. Quand le mélange est redevenu limpide, on acidifie à pH 2 et le produit précipite et cristallise.
Tous les produits obtenus suivant les procédés A à V sont cristallisés et blancs, bien que ceci ne soit pas caractéristique de l'invention. Ils sont consignés dans les tableaux suivants.

Amino-3-2-dihydrothiophenone or homocystelin thiolactone, called ADT hereafter, was described by du Vigneaud in
1935. It is prepared by dealkylation of methionine or other alkyl homocysteine by reduction with sodium in ammonia or hydriodic acid, followed by cyclization. Still little developed, it is relatively expensive because soluble in water, its isolation is difficult. Some N-acyl derivatives are known, but the S-acyl homocysteines are unknown. The new reactions of the invention provide access, sometimes very economically, to numerous derivatives useful in therapy, cosmetology, photography, etc., and allowing more economical access to known derivatives.
The invention therefore relates to a process for the N-acylation of homocystelin or of one of its derivatives, characterized in that the homocysteine or one of its esters, thiolesters, is subjected to N-acylation, salts or an S-acyl compound of homocystel.
The compounds are prepared in the free state or from salts of organic or inorganic acids or bases, derivatives of ADT whose formula comprises the group -S-CH2-
CH2-CH (NHR) -CO- in which R represents hydrogen or acyl. The structure of the compounds is either cyclic according to the formula (A) S-CH2-CH2CH (NHR) -CO where R then carries more than 4 carbons and is substituted when it is a benzoyl or nicotinoyl and unsubstituted when it is a cinnamoyl , is

The N-acyl ADTs were obtained only by acylation of ADT in anhydrous media, and the N-acyl ADTs of the invention can thus be obtained. A single reference, Yoshitomi, Japanese Patent 16712/62 ', discloses the acetylation of ADT in a bicarbonate solution by acetic anhydride.
The process according to the invention can be carried out in that the ADT is dissolved in an aqueous solvent, preferably water. A weak base such as a bicarbonate, or a stronger base, such as sodium hydroxide, is added, but in an amount calculated to avoid hydrolysis of the thiolactone. To this solution is added an anhydride or an acid halide at a specific temperature of the reagent, often optimum between -10 and ambient, acetic anhydride being excluded from the reagents used.
S-acylation of N-acylated or unacylated ADTs, preferably
in water can be carried out in two stages, hydrolysis of the thiolactone followed by acylation. Hydrolysis accelerates with increasing pH and temperature. Then these parameters are lowered and an acid halide or anhydride is added. Preferably, excess sodium hydroxide is used and acylated between -10 and ambient.
At the end of the reaction, the S-acylhomocysteine is acidified and isolated.
This technique gives access to compounds which are identical or differently acylated on sulfur and nitrogen.
To prepare the esters or thiolesters of sacylated homocysteins, anhydrous hydrochloric acid is saturated, for example, a solvent which may be the alcohol or reactive mercaptan, the homocysteine is dissolved and the mixture is often left at room temperature. The ester is isolated by evaporating the solvent and
adding a bicarbonate solution to the residue to remove
residual homocysteine.
For the N and S-acylation of homocys esters and thiolesters

acetyl-methionine, the substrate is first dealkylated according to known methods, the solvent is evaporated off and without
isolate the intermediate product, one acylates successively on nitrogen and on sulfur according to the above processes

evaporation residue is dissolved in water. N-acyl at pH
mild, then increases the pH for S-acyler. If the dealkylation is

For N- and S-acylate differently, it is necessary

To prepare N, S-diacylhomocysteins, it is possible to
put the N-acylated products obtained according to the method of in

alcohols and mercaptans which have specific properties

serve as an introduction vector to improve their qualities

solubility, delayed effect, passage of walls or others.
The compositions prepared and the pharmaceutical forms for local, oral, parenteral, rectal, otorhinolaryngology and ophthalmology administration such as
ointments, solutions, milks, aerosols, pills, tablets, dragees,
capsules, chronules, syrup, capsules, drops, solutions to drink or to

Example I
A-N-acetyl ADT synthesis process
1.535 g of ADT, HCl are dissolved in 10.1 ml of N sodium hydroxide and 1.1 ml of acetic anhydride are added.
