Erfindungsgemäss gelangt man zu den Verbindungen der Formel I, indem man eine Verbindung der Formel II,

worin R1 und R2 obige Bedeutung besitzen, mit einer Verbindung der Formel III, R3-o-CH=CH2 III worin R3 obige Bedeutung besitzt, umsetzt.
Das erfindungsgemässe Verfahren wird vorteilhafterweise in einer Inertgasatmosphäre, z. B. unter Stickstoff oder Argon, in einem abgeschlossenen Gefäss bei erhöhter Temperatur, z. B. bei 160 bis 200 C, durchgeführt. Das Molverhältnis der Verbindung der Formel II zur Verbindung der Formel III beträgt vorzugsweise 1:1 bis 1:4, die Reaktion ist z. B. nach 10 bis 70 Stunden beendet. Aus dem Reaktionsgemisch können die Verbindungen der Formel I nach an sich bekannten Methoden isoliert und gegebenenfalls gereinigt werden.
Stehen R1 und/oder R2 für eine substituierte Phenylgruppe, so ist diese vorzugsweise ein- oder zweifach substituiert, als Substituenten werden vorzugsweise Halogen und niedere Alkoxygruppen, insbesondere Chlor und Methoxy, verwendet. Bedeuten R1 und/oder R2 einen heterocyclischen Rest, so besitzt dieser vorzugsweise fünf oder sechs Ringglieder und Sauerstoff, Stickstoff oder Schwefel als Heteroatom, ein Pyridylrest kann auch quaternisiert sein. Die durch R3 symbolisierte niedere Alkylgruppe kann geradkettig oder verzweigt sein und besitzt vorzugsweise 1 bis 6 Kohlenstoffatome, sie bedeutet insbesondere die n-Butylgruppe.
Die Ausgangsprodukte der Formel II und III sind bekannt.
Die Verbindungen der Formel I und ihre pharmakologisch verträglichen Säureadditionssalze besitzen bei geringer Toxizität wertvolle pharmakodynamische Eigenschaften und können daher als Heilmittel verwendet werden. Sie entfalten eine antivirale Wirksamkeit, insbesondere gegen Influenzaviren, wie sich in vivo im Mäuseschutzversuch zeigen lässt.
Diese antivirale Wirksamkeit zeigt sich durch eine Erhöhung der Überlebensrate.
Für die Dauer der Infektionsgefährdung beträgt die Tagesdosis für Erwachsene etwa 100 bis 400 mg Wirkstoff, vorzugsweise verabreicht in Teilmengen zwischen 35 bis 200 mg 2- bis 3mal täglich.
Die nach dem erfindungsgemässen Verfahren erhältlichen Verbindungen können bei der antiviralen Behandlung auf jede Art verabreicht werden, welche einen Kontakt der aktiven Verbindung mit dem Sitz der Virusinfektion im Körper bewirkt. Damit ist die Behandlung der Infektionsstelle sowohl vor der Ausbildung der Infektion als auch nachher zu verstehen. Beispielsweise kann die Verabreichung parenteral, das ist subcutan, intravenös, intramuskulär oder intraperitonal, erfolgen.
Alternativ oder gleichzeitig können die Verbindungen wirksam oral verabreicht werden. Da die Verbindungen besonders gegen Infektionen der Atmungsorgane, wie virale Influenza oder virale Pneumonie, wirksam sind, kann die Verabreichung mittels Dampf oder Spray durch die Mund- oder Nasengänge erfolgen.
Die aktive Verbindung kann in entsprechenden Zusammensetzungen in Form von Tabletten, Kapseln, Pulverpackungen oder flüssigen Lösungen, Suspensionen oder Elixieren für die orale Verabreichung, oder in Form flüssiger Lösungen für die parenterale Verwendung, und in gewissen Fällen in Form von Suspensionen für parenteralen Gebrauch (ausgenommen intravenös) angewendet werden. In solchen Zusammensetzungen ist der aktive Bestandteil gewöhnlich in Mengen von wenigstens 0,0001 Gew. %, bezogen auf das Gesamtgewicht der Zusammensetzung, und in Mengen von nicht mehr als 99 Gew. % zugegen.
Neben dem aktiven Bestandteil enthält die antivirale Zu sammensetzung einen festen oder flüssigen, nichttoxischen pharmazeutischen Träger für den aktiven Bestandteil.
Der feste Träger kann eine Kapsel sein, z. B. eine übliche
Gelatinetype. Ferner kann der aktive Bestandteil mit oder ohne Hilfsmittel tablettiert oder in Pulverpackungen eingebracht werden.
Wie bereits erwähnt wurde, kann der pharmazeutische Träger eine sterile Flüssigkeit wie Wasser oder ein Öl sein. Gewöhnlich stellen Wasser, Salzlösungen sowie wässrige Dex troselösungen (Glucose) und ähnliche Zuckerlösungen vor zugsweise flüssige Träger dar, insbesondere für Injektions lösungen.
Die orale Verabreichung kann in Form einer geeigneten
Suspension oder eines Sirups erfolgen, wobei der aktive Be standteil gewöhnlich in einer Menge von etwa 0,5 bis 10 Gew. %, vorzugsweise etwa 2 bis 5 Gew. %, vorhanden ist.
In den nachfolgenden Beispielen, die die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel 1
2-n-Butoxy-3,4-dihydro-5-phenyl-4 (2-thienyl )-2H-pyran
3,48 g 2-Phenyl-3-(2-thienyl)-acrolein und 4 g n-Butylvinyläther werden nach Zugabe von einigen Kristallen Hydrochinon 38 Stunden im Stahlautoklav unter Argon auf 180 bis 185 erhitzt. Nach dem Ausfüllen werden die leicht flüchtigen Anteile im Vakuum (12 Torr Badtemperatur bis 100 ) abgezogen. Der Rückstand wird im Vakuum (10-3 Torr) destilliert. Man erhält die Titelverbindung als schwach gelbes Öl, Kp10-3 : 150-165 .
Beispiel 2 2-n-Butoxy-4,5-di-(2-furyl)-3 ,4-dihydro-2H-pyran Aus 3,18 g n-Butylvinyläther und 2 g 2,3-Di-(2-furyl)acrolein erhält man analog wie in Beispiel 1 beschrieben nach 24 Stunden Erhitzen auf 170-180 die Titelverbindung als gelbes Öl, Kpl : 140-150 .
Beispiel 3
2-n-Butoxy-5- (2,4-dichlorphenyl)-3 ,4-dihydro-4- (3-pyridyl)-2H-pyran
Aus 15,7 g 2-(2,4-Dichlorphenyl)-3-(3-pyridyl)-acrolein und 17,4 g n-Butylvinyläther erhält man analog wie in Beispiel 1 beschrieben nach 48 Stunden Erhitzen bei 1800 die Titelverbindung als schwach gelbes Öl, Kplo : 180-195 .
Beispiel 4
4-[(1-Benzyl)-pyridinium-3-yl]-2-n-butoxy-5-(2,4- dichlorphenyl)-3 ,4-dihydro-2H-pyranchlorid
1,5 g 2-n-Butoxy-5- (2,4-dichlorphenyl)-3 ,4-dihydro-4- (3-pyridyl)-2H-pyran und 5 ml Benzylchlorid werden unter Stickstoff 3 Stunden auf 100 erhitzt. Man erhält weisse Kristalle, Fp. 1200 (Zers.).

The inventive method is advantageously carried out in an inert gas atmosphere, e.g. B. under nitrogen or argon, in a closed vessel at elevated temperature, e.g. B. at 160 to 200 C carried out. The molar ratio of the compound of the formula II to the compound of the formula III is preferably 1: 1 to 1: 4; B. ended after 10 to 70 hours. The compounds of the formula I and their pharmacologically acceptable acid addition salts have valuable pharmacodynamic properties with low toxicity and can therefore be used as medicaments. They develop an antiviral activity, especially against influenza viruses, as can be shown in vivo in the mouse protection test.
This antiviral effectiveness is shown by an increase in the survival rate.
For the duration of the risk of infection, the daily dose for adults is about 100 to 400 mg of active ingredient, preferably administered in portions between 35 to 200 mg 2 to 3 times a day.
The compounds obtainable by the process according to the invention can be administered in the antiviral treatment in any way which brings about contact of the active compound with the location of the viral infection in the body. This means that the treatment of the infection site is to be understood both before the infection develops and afterwards. For example, administration can be parenteral, that is, subcutaneous, intravenous, intramuscular or intraperitoneal.
Alternatively, or simultaneously, the compounds can be effectively administered orally. Since the compounds are particularly effective against infections of the respiratory organs, such as viral influenza or viral pneumonia, they can be administered by means of steam or spray through the mouth or nasal passages.
The active compound may be in appropriate compositions in the form of tablets, capsules, powder packs or liquid solutions, suspensions or elixirs for oral administration, or in the form of liquid solutions for parenteral use, and in certain cases in the form of suspensions for parenteral use (except intravenous). In such compositions, the active ingredient is usually present in amounts of at least 0.0001% by weight based on the total weight of the composition and in amounts of no more than 99% by weight.
In addition to the active ingredient, the antiviral composition contains a solid or liquid, non-toxic pharmaceutical carrier for the active ingredient.
The solid support can be a capsule, e.g. B. a usual
Gelatin type. In addition, the active ingredient can be tabletted with or without an adjuvant or incorporated in powder packs.

3.48 g of 2-phenyl-3- (2-thienyl) -acrolein and 4 g of n-butyl vinyl ether are heated to 180-185 for 38 hours in a steel autoclave under argon after adding a few crystals of hydroquinone. After filling, the volatile components are drawn off in vacuo (12 Torr bath temperature up to 100). The residue is distilled in vacuo (10-3 Torr). The title compound is obtained as a pale yellow oil, boiling point 10-3: 150-165.

4 - [(1-Benzyl) -pyridinium-3-yl] -2-n-butoxy-5- (2,4-dichlorophenyl) -3, 4-dihydro-2H-pyran chloride
1.5 g of 2-n-butoxy-5- (2,4-dichlorophenyl) -3, 4-dihydro-4- (3-pyridyl) -2H-pyran and 5 ml of benzyl chloride are heated to 100 for 3 hours under nitrogen. White crystals are obtained, melting point 1200 (decomp.).
