Les composés, préparés conformément au procédé selon
l'invention, possèdent au moins l'une des activités pharmaceutiques 'suivantes: anti-inflammatoire, antiulcère, analgésique, anti
pyrétique, antihistaminique et dépressive du système nerveux
central (SNC), ainsi qu'une faible toxicité; la plupart d'entre
eux possèdent même plus d'une de ces activités. C'est pourquoi
il est à remarquer que certains des composés entrant dans le
cadre de la présente invention, sont intéressants comme agents
analgésiques, anti-inflammatoires et dépresseurs du SNC.
Les composés, particulièrement intéressants, préparés par le
procédé selon la présente invention, peuvent être représentés par
la formule (I) ci-dessus, dans laquelle le groupe NO2 est en
position 3, c'est-Åa-dire en méta par rapport au carbone relié au
noyau pyrimidinique; R est hydrogène, alkyle en C1 à C6, halogéno
alkyle ou trihalogénoalkyle en C1 à C3, alkényle en C3 à C5, halo
génoallyle, propargyle, cyclopropylméthyle, acétoxyéthyle,
hydroxyalkyle à 2 ou 3 carbone, alkoxyalkyle comportant de 2 à
4 carbone, vinyloxyéthyle, hydroxyéthoxyéthyle, carboxyméthyle,
éthoxycarbonylméthyle, époxy-2,3 propyle, diéthylaminoéthyle,
méthyl-4 pipérazinoéthyle, benzyle, phénétyle, ou cinnamyle,

Conviennent par exemple comme composés de formule générale (III-2): urée, diéthylurée, N-propyluréthanne et thio-urée.
Les réactions selon la présente invention s'effectuent généralement dans un solvant organique, tel que diméthylformamide,
diglyme, tétrahydrofuranne ou alcool. Dans une forme préférée de l'invention, on opère en présence d'un composé métallique,
notamment sodium métallique, amidure ou hydrure de sodium,
ou bien d'une base organique telle que trialkylamine ou pyridine, ou bien encore d'une base minérale. par exemple carbonate ou hydroxyde alcalin. Les composés métalliques mentionnés en premier se révèlent les plus efficaces pour l'obtention de rendements élevés. La température souhaitable n'est pas critique, et peut être la température ambiante ou une température plus élevée. Cependant, lorsqu'on utilise comme réactif le phosgène ou le thiophosgène, on opère sous refroidissement par de la glace.
Le solvant de réaction est éliminé du mélange réactionnel par distillation sous pression réduite, et le résidu, obtenu ainsi, est mélangé avec de l'eau pour précipiter le produit brut. Par recristallisation de ce dernier à partir d'un solvant organique, tel que méthanol ou acétone, on obtient des cristaux purs.
La substance de départ, représentée par la formule générale III, peut être préparée avec des rendements élevés par réaction d'un dérivé d'acide anilino-2 nicotinique ou de chlorure d'anilino-2 nicotinoyle, avec une amine ou un alcool, mais sa préparation ne se limite pas à ce procédé.
Dans le tableau I sont réunis à titre d'exemples quelques composés obtenus par le procédé selon la présente invention, accompagnés de leur point de fusion.

Pour de nombreux composés préparés conformément à l'invention, on a étudié la toxicité aiguë, afin de s'assurer de leur inocuité, ainsi que leurs effets analgésique,
anti-inflammatoire et de dépression du SNC, pour montrer leurs excellentes activités à ces égards. Les résultats de chaque examen sont rassemblés dans le tableau II. Chaque essai est conduit de la manière suivante.
1. Toxicité aiguë
Le composé à essayer, mis en suspension dans une solution saline de gomme adragante à 0,5%, est administré par voie intrapéritonéale ou orale à des souris mâles de souche dd (16-24 g).
La dose léthale est déterminée à partir du nombre d'animaux morts 72 h après l'administration.
2. Activité anti-inflammatoire
On administre oralement à un groupe de 5 rats mâles de souche Wistar (100-150 g) chaque composé essayé, mis en suspension dans une solution saline de gomme adragante à 0,5%.
Au bout de 30 mn, on injecte dans la patte arrière, par voie
sous-cutanée, une suspension dans l'eau à 0,5 à 1% de carraghénine.
Au bout de 3 h, on mesure le volume de l'oedème provoqué par la
carraghénine, et l'on détermine le pourcentage d'inhibition par
comparaison avec les résultats obtenus chez les animaux témoins.
Ces pourcentages sont représentés par les notations suivantes: moins de 15%: +
de 16 à 30%: +
de31 à45%: ++
de46à60%: +++ plus de 61%: ++++ 3. Activité analgésique
Chaque composé essayé, mis en suspension dans une solution saline de gomme adragante à 0,5%, est administré oralement à des souris mâles de souche dd (18-20 g). Au bout de 1 h, on injecte par voie intrapéritonéale une solution d'acide acétique à 0,6% à raison de 0,1 ml/10 g. Le syndrome de crispation est observé pendant 10 mn au bout de 30 mn après l'injection; on calcule par la méthode de Litchfield-Wilcoxon, la dose analgésique active à 50% (DA5o) et ses limites de fiabilité à 95%.
4. Activité dépressive sur le SNC
Chaque composé essayé, mis en suspension dans une solution saline de gomme adragante à 0,5%, est injecté par voie intrapéritonéale à des souris mâles de souche dd (16-24 g). La disparition du réflexe de sursaut est étudiée en milieu insonorisé. La dose nécessaire, pour obtenir cette disparition, est indiquée par les notations suivantes: plus de 1000 mg/kg: - de 300 à 1000 mg/kg: f de 100 à 300 mg/kg:

Dans le tableau II sont rassemblés les résultats des activités analgésiques, anti-inflammatoires et dépressives'du SNC, ainsi que
la toxicité des composés préparés selon l'invention, et, à titre de comparaison, de composés déjà connus en thérapeutique.

La présente invention est illustrée de façon non limitative par les exemples ci-après.
Exemple 1:
A une solution de 3 g de (m-nitroanilino)-2 N-(n-propyl)nicotinamide dans 25 ml de tétrahydrofuranne, on ajoute 0,5 g d'hydrure de sodium à 50%, et l'on agite le tout pendant 15 mn.
Puis on y ajoute goutte à goutte sous refroidissement 5,4 g de chlorocarbonate d'éthyle. On laisse reposer le mélange pendant 1 h et l'on porte ensuite au reflux pendant 10 h. Lorsque la réaction est achevée, le solvant est chassé du mélange résultant par distillation sous pression réduite. Le résidu ainsi obtenu est additionné d'eau pour précipiter le produit brut. Celui-ci est recristallisé à partir de méthanol et donne 2,5 g de m-nitrophényl-l n-propyl-3 pyrido[2,3-d](l H,3H)pyrimidinedione-2,4, sous forme de prismes jaune pâle.
Point de fusion: 184-185 C

Exemple 2:
On ajoute à une solution de 3 g de (m-nitroanilino)-2 N-allylnicotinamide dans 25 ml de tétrahydrofuranne, 1 g d'hydrure de sodium à 50% environ, et l'on agite pendant 30 mn. Puis, I'on additionne le tout sous refroidissement, et goutte à goutte, de 16 g d'une solution à 30% de phosgène dans le toluène. Le mélange est agité pendant 1 h, et puis porté à reflux pendant 2 h. Lorsque la
réaction est achevée, le solvant est chassé du mélange résultant par distillation sous pression réduite. On ajoute de l'eau au
résidu ainsi obtenu pour précipiter le produit brut.
Celui-ci est
recristallisé à partir d'acétate d'éthyle pour donner 2,8 g de
m-nitrophényl- 1 allyl-3 pyrido[2,3-d]( I H,3H)pyrîmidinedione-2,4,
sous forme d'aiguilles incolores.
Point de fusion: 188-189"C

Exemple 3:
2,7 g de (m-nitroanilino)-2 N-méthylnicotinamide dans
20 ml de diglyme sec sont additionnés de 0,6 g d'hydrure de sodium
à 50% et de 5,9 g de carbonate de diéthyle, et l'on porte à reflux
pendant 12 h. Lorsque la réaction est achevée, le solvant est
chassé du mélange par distillation sous pression réduite, laissant
un résidu qui, par addition d'eau, précipite le produit brut. Par
recristallisation de celui-ci à partir de méthanol, on obtient 2,6 g

Le résidu ainsi obtenu est additionné d'eau glacée, et l'on obtient ainsi un précipité qui, par recristallisation à partir de méthanol, donne 2,6 g de m-nitrophényl-l éthyl-3 pyrido[2,3-d](l H,3H)- pyrimidinedione-2,4, sous forme d'aiguilles incolores.

On ajoute à une solution de 1,25 g de (m-nitroanilino)-2
N-méthylnicotinamide dans 20 ml de tétrahydrofuranne, 0,48 g d'hydrure de sodium à 50% environ, et l'on agite le mélange pendant 30 mn. On y ajoute ensuite goutte à goutte 2,3 g de thiophosgène sous refroidissement avec de la glace, et le mélange résultant est abandonné à température ambiante pendant 30 mn. L'excès de thiophosgène est décomposé par addition d'une solution méthanolammoniaque. Le solvant est chassé du mélange par distillation, abandonnant un résidu, qui, par addition d'eau, précipite le produit brut.
Par recristallisation de ce dernier à partir d'acétone, on obtient 1,2 g de m-nitrophényl-l méthyl-3 thio-2 oxo-4 tétrahydro-1,2,3,4 pyrido[2,3-d]pyrimidine pure, sous forme de prismes jaune pâle.

Lorsque la réaction est achevée, le précipité solide est éliminé par filtration. Le filtrat est concentré sous pression réduite et abandonne un résidu qui, par addition d'eau, donne le produit brut. The compounds, prepared according to the process according to
invention, have at least one of the following pharmaceutical activities: anti-inflammatory, antiulcer, analgesic, anti
pyretic, antihistamine and nervous system depressant
central (CNS), as well as low toxicity; most of
they even have more than one of these activities. That is why
it should be noted that some of the compounds entering into the
context of the present invention, are useful as agents
analgesics, anti-inflammatories and CNS depressants.
The particularly interesting compounds prepared by the
method according to the present invention, can be represented by
formula (I) above, in which the NO2 group is in
position 3, that is to say in meta with respect to the carbon linked to the
pyrimidine nucleus; R is hydrogen, C1 to C6 alkyl, halo
C1 to C3 alkyl or trihaloalkyl, C3 to C5 alkenyl, halo
genoallyl, propargyl, cyclopropylmethyl, acetoxyethyl,
2 or 3 carbon hydroxyalkyl, alkoxyalkyl having from 2 to
4 carbon, vinyloxyethyl, hydroxyethoxyethyl, carboxymethyl,
ethoxycarbonylmethyl, epoxy-2,3 propyl, diethylaminoethyl,
4-methyl piperazinoethyl, benzyl, phenethyl, or cinnamyl,

The reactions according to the present invention are generally carried out in an organic solvent, such as dimethylformamide,
diglyme, tetrahydrofuran or alcohol. In a preferred form of the invention, the operation is carried out in the presence of a metal compound,
in particular metallic sodium, sodium amide or hydride,
or else an organic base such as trialkylamine or pyridine, or else an inorganic base. for example carbonate or alkali hydroxide. The metal compounds mentioned first are found to be the most effective in obtaining high yields. The desirable temperature is not critical, and can be room temperature or a higher temperature. However, when phosgene or thiophosgene are used as reagent, the operation is carried out under cooling with ice.
The reaction solvent is removed from the reaction mixture by distillation under reduced pressure, and the residue thus obtained is mixed with water to precipitate the crude product. By recrystallization of the latter from an organic solvent, such as methanol or acetone, pure crystals are obtained.
The starting material, represented by the general formula III, can be prepared in high yields by reacting a derivative of 2-anilino-nicotinic acid or 2-anilino-nicotinoyl chloride, with an amine or an alcohol, but its preparation is not limited to this process.
Some compounds obtained by the process according to the present invention, together with their melting point, are combined by way of example.

For many compounds prepared in accordance with the invention, the acute toxicity was studied in order to ensure their harmlessness, as well as their analgesic effects,
anti-inflammatory and CNS depression, to show their excellent activities in these respects. The results of each examination are collated in Table II. Each test is carried out as follows.
1. Acute toxicity
The test compound, suspended in 0.5% tragacanth saline, is administered intraperitoneally or orally to male dd strain mice (16-24 g).
The lethal dose is determined from the number of animals that died 72 hours after administration.
2. Anti-inflammatory activity
To a group of 5 male rats of the Wistar strain (100-150 g), each test compound, suspended in 0.5% saline tragacanth solution, was administered orally.
After 30 minutes, we inject into the hind paw, by
subcutaneously, a suspension in water containing 0.5 to 1% carrageenan.
After 3 h, the volume of the edema caused by the
carrageenan, and the percentage inhibition is determined by
comparison with the results obtained in control animals.
These percentages are represented by the following notations: less than 15%: +
from 16 to 30%: +
from 31 to 45%: ++
from 46 to 60%: +++ over 61%: ++++ 3. Analgesic activity
Each test compound, suspended in 0.5% tragacanth saline, was administered orally to male dd strain mice (18-20 g). After 1 h, a 0.6% acetic acid solution is injected intraperitoneally at a rate of 0.1 ml / 10 g. The crispation syndrome is observed for 10 min after 30 min after the injection; the 50% active analgesic dose (DA50) and its 95% reliability limits are calculated by the Litchfield-Wilcoxon method.
4. CNS depressive activity
Each test compound, suspended in 0.5% tragacanth saline, is injected intraperitoneally into male mice of the dd strain (16-24 g). The disappearance of the startle reflex is studied in a soundproof environment. The dose necessary to obtain this disappearance is indicated by the following notations: more than 1000 mg / kg: - from 300 to 1000 mg / kg: f from 100 to 300 mg / kg:

The present invention is illustrated in a nonlimiting manner by the examples below.
Example 1:
To a solution of 3 g of (m-nitroanilino) -2 N- (n-propyl) nicotinamide in 25 ml of tetrahydrofuran, 0.5 g of 50% sodium hydride is added, and the whole is stirred. for 15 minutes.
5.4 g of ethyl chlorocarbonate are then added dropwise thereto under cooling. The mixture is left to stand for 1 h and then refluxed for 10 h. When the reaction is complete, the solvent is removed from the resulting mixture by distillation under reduced pressure. The residue thus obtained is added with water to precipitate the crude product. This is recrystallized from methanol and gives 2.5 g of m-nitrophenyl-l n-propyl-3 pyrido [2,3-d] (1 H, 3H) pyrimidinedione-2,4, in the form of prisms pale yellow.
Melting point: 184-185 C

Example 2:
Is added to a solution of 3 g of (m-nitroanilino) -2 N-allylnicotinamide in 25 ml of tetrahydrofuran, 1 g of approximately 50% sodium hydride, and the mixture is stirred for 30 min. in the form of colorless needles.

Example 3:
2.7 g of (m-nitroanilino) -2 N-methylnicotinamide in
20 ml of dry diglyme are added with 0.6 g of sodium hydride
at 50% and 5.9 g of diethyl carbonate, and the mixture is brought to reflux
for 12 h. When the reaction is complete, the solvent is
distilled off from the mixture under reduced pressure, leaving
a residue which, on addition of water, precipitates the crude product. By
recrystallization of the latter from methanol, 2.6 g are obtained
m-nitrophenyl-1 methyl-3 pyrido [2,3-d] (1 H, 3H) pyrimidine
2,4-dione, in the form of colorless prisms.

Example 4:
Has a solution of 2.9 g of (m-nitroanilino) -2 N-ethyl
nicotinamide in 50 ml of tetrahydrofuran, 1 g of 50% sodium hydride and 4.9 g of carbonyl-1, diimidazole are added, and this mixture is stirred for 1 h before bringing it to reflux for 5 h. When the reaction is complete, the solvent is removed from the mixture under reduced pressure.
To the residue thus obtained is added ice-water, and a precipitate is thus obtained which, on recrystallization from methanol, gives 2.6 g of m-nitrophenyl-1 ethyl-3 pyrido [2,3-d] (1H, 3H) - pyrimidinedione-2,4, in the form of colorless needles.

Example 5:
Is added to a solution of 1.25 g of (m-nitroanilino) -2
N-methylnicotinamide in 20 ml of tetrahydrofuran, 0.48 g of approximately 50% sodium hydride, and the mixture is stirred for 30 min. 2.3 g of thiophosgene are then added dropwise thereto under cooling with ice, and the resulting mixture is left at room temperature for 30 min. The excess thiophosgene is decomposed by adding a methanolammonia solution. The solvent is removed from the mixture by distillation, leaving a residue, which, on addition of water, precipitates the crude product.
By recrystallization of the latter from acetone, 1.2 g of m-nitrophenyl-1 methyl-3-thio-2-oxo-4-tetrahydro-1,2,3,4 pyrido [2,3-d] pyrimidine are obtained. pure, in the form of pale yellow prisms.

Example 6:
2.9 g of (m-nitroanilino) -2 ethyl nicotinate dissolved in 20 ml of dimethylformamide are added 0.48 g of 55% sodium hydride, and the whole is stirred for 1 h at room temperature. Then 10.3 g of N-methylurethane was added thereto and this mixture was reacted at 100 ° C for 20 h.
When the reaction is complete, the solid precipitate is removed by filtration. The filtrate is concentrated under reduced pressure and leaves a residue which, on addition of water, gives the crude product. This is collected by filtration, and then recrystallized from methanol, which gives 1.4 g of m-nitrophenyl-1 methyl-3 pyrido [2,3-d] (1 H, 3H) pyrimidinedione-2, 4, in the form of pale yellow prisms.

The filtrate is concentrated under reduced pressure and leaves a residue which, on addition of water, gives the crude product. The latter, by recrystallization from dimethylformamide, gives 1.5 g of m-nitrophenyl-I pyrido [2,3-d] (III, 3H) pyrimidinedione-2,4, in the form of colorless prisms.
