Die Erfindung betrifft neue Tetrahydrobenz[g]isochinoline der Formel I, worin
R1 Wasserstoff, Halogen mit einer Atomzahl von 9-35, Alkyl mit 14 Kohlenstoffatomen oder Alkoxy mit 14 Kohlenstoffatomen bedeutet,
R3 Wasserstoff, Alkyl mit 14 Kohlenstoffatomen oder eine PhenylalkyIgruppe mit 7-1OKohlenstoffatomen, deren Phenylring gegebenenfalls durch Halogen mit einer Atomzahl von 9-35, Alkyl mit 14 Kohlenstoffatomen oder Alkoxy mit 14 Kohlenstoffatomen monosubstituiert ist, bedeutet und
R4 für Wasserstoff steht oder
R3 und R4 zusammen mit dem Stickstoffatom, an welches sie gebunden sind, einen l-Pyrrolylrest bilden, und ihre Säureadditionssalze sowie Verfahren zu ihrer Herstellung.
In den Verbindungen der Formel I enthaltene vorstehend definierte Alkyl- oder Alkoxygruppen enthalten vorzugsweise 1 oder 2 Kohlenstoffatome und stellen insbesondere Methyl oder Methoxy dar.
Der Substituent R1 steht vorzugsweise für Wasserstoff.
Ein sonstiger Substituent R1 ist vorzugsweise in 7- oder 8 Stellung des BenzLg]isochinolingerüstes angeordnet.
Der Substituent R3 bedeutet vorzugsweise Wasserstoff oder auch Methyl oder bildet zusammen mit dem Substituen ten R4 und dem Stickstoffatom, an welches sie gebunden sind, einen l-Pyrrolylrest. Eine vorstehend definierte Phenylalkylgruppe R3 stellt vorzugsweise eine gegebenenfalls substituierte Benzyl- oder Phenäthylgruppe dar.
Erfindungsgemäss gelangt man zu den neuen Verbindungen der Formel I und ihren Säureadditionssalzen, indem man aus Verbindungen der Formel II, worin R1, R3 und R4 obige Bedeutung besitzen und R2 einen solvolytisch abspaltbaren Rest bedeutet, den Rest R2 durch Solvolyse abspaltet und die erhaltenen Verbindungen der Formel I gewünschtenfalls in ihre Säureadditionssalze überführt.
Die Abspaltung des Restes R2 aus den Verbindungen der Formel II kann auf an sich zur Abspaltung von Aminoschutzgruppen aus heterocyclischen Aminen übliche Weise solvolytisch, insbesondere hydrolytisch erfolgen, beispielsweise nach zur Urethan- oder Amidspaltung bekannten Methoden. Geeig nete Schutzgruppen R2 sind beispielsweise Acylgruppen, vorzugsweise Alkoxy- oder Aryloxycarbonylgruppen oder Alka- noyl- oder Aroylgruppen, beispielsweise Alkanoylgruppen mit 24 Kohlenstoffatomen, gegebenenfalls durch Halogen, Alkyl oder Alkoxy monosubstituierte Benzoyl- oder Phenylalkanoylgruppen mit 7-10 Kohlenstoffatomen oder niedere Alkoxycarbonyl- oder Aryloxycarbonylgruppen.
Die Spaltung der Verbindungen der Formel II kann je nach Art des Restes R2 bevorzugt in saurem Medium, beispielsweise in Gegenwart starker Mineralsäuren, oder in alkalischem Medium, beispielsweise in Gegenwart anorganischer Basen durchgeführt werden.
Die erfindungsgemäss erhaltenen Verbindungen der Formel I können in Form der freien Basen oder ihrer Säureadditionssalze vorliegen. Die freien Basen können auf an sich bekannte Weise in ihre Säureadditionssalze überführt werden und umgekehrt. So können die erfindungsgemässen Verbin dungen der Formel I z. B. mit anorganischen Säuren wie Chlor wasserstoff oder mit organischen Säuren wie Fumarsäure
Säureadditionssalze bilden.
Die Ausgangsverbindungen können wie folgt erhalten werden: a') Verbindungen der Formel II, worin R3 nicht für Was serstoff steht, können beispielsweise durch Substitution der
5-Aminogruppe in Verbindungen der Formel IIa, worin R1 und R2 obige Bedeutung besitzen, auf an sich bekannte Weise erhalten werden. Beispielsweise können die Verbindungen der Formel IIa mit entsprechenden Alkyl- oder Phenylalkylhalogeniden in Gegenwart von tert. Kaliumbutylat oder, falls Rs Methyl bedeuten soll, auch mit Dimethylsulfat/Pyridin alkyliert werden.
Verbindungen der Formel II, worin R3 und R4 zusammen mit dem Stickstoffatom, an welches sie gebun- den sind, einen 1-Pyrrolylrest bedeuten, können durch Umsetzung von Verbindungen der Formel IIa mit 2,5-Dimethoxytetrahydrofuran und anschliessende Abspaltung von Methanol aus dem Reaktionsprodukt erhalten werden.
b') Verbindungen der Formel IIa können beispielsweise ausgehend von 3-Benzyl-4-piperidonderivaten der Formel III, worin R1 und R2 obige Bedeutung besitzen, erhalten werden, indem man aus diesen durch Anlagerung von Blausäure nach zur Cyanhydrinherstellung üblichen Verfahren, vorzugsweise durch Umsetzung mit Acetoncyanhydrin die entsprechenden 3-Benzyl-4-hydroxy-4-piperidinonitrilderivate herstellt, diese durch Wasserabspaltung, beispielsweise mittels Thionylchlorid/ Pyridin in 5-Benzyl-4-cyan-1,2,3,6-tetrahydropyridinderivate der Formel IV, worin R1 und R2 obige Bedeutung besitzen, überführt und diese, welche als Nebenprodukt auch entsprechende isomere 3-Benzyl-4-cyan-1,2,3,6-tetrahydropyridin- derivate enthalten können, nach an sich bekannten Methoden,
beispielsweise durch Behandlung mit 90 %iger Schwefelsäure cyclisiert.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
Die Verbindungen der Formel I und ihre physiologisch verträglichen Säureadditionssalze zeichnen sich durch interessante pharmakologische Eigenschaften aus und können daher als Heilmittel verwendet werden.
Insbesondere zeigen die Verbindungen in Tierversuchen antiaggressive Wirkungen und vermögen z. B. durch Isolation erzeugtes aggressives Verhalten von Mäusen zu dämpfen.
Aufgrund ihrer aggressionshemmenden Eigenschaften können die Substanzen zur Behandlung von aggressiven Erregungszuständen, beispielsweise zur Dämpfung von aggressivem Verhalten von Psychopathen Verwendung finden.
Ausserdem besitzen die Verbindungen in Tierversuchen zentraldämpfende Wirkungen, welche sich beispielsweise an Mäusen im Klettertest durch Hemmung der Motilität zeigen.
Aufgrund ihrer zentraldämpfenden Wirkungen können die Substanzen in der Psychiatrie zur Behandlung von Erregungszuständen Verwendung finden.
Als Heilmittel können die Verbindungen der Formel I und ihre physiologisch verträglichen Säureadditionssalze zusammen mit üblichen pharmazeutischen Hilfsstoffen in galenischen Zubereitungen wie z. B. Tabletten, Kapseln oder Lösungen enthalten sein. Diese galenischen Zubereitungen können nach an sich bekannten Methoden dargestellt werden.
In den nachfolgenden Beispielen, die die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel 1 5-Amino-i,2,3,4-tetrahydrobenz[gjisochinolin
Eine Lösung von 9,6 g 5-Amino-1,2,3,4-tetrahydrobenz [g]isochinolin-2-ylcarbonsäureäthylester in 80 ml Eisessig wird bei Raumtemperatur tropfenweise mit 80 ml 48 %iger Bromwasserstoffsäure versetzt und 3,5 Stunden zum Sieden erwärmt. Das abgekühlte Reaktionsgemisch wird auf Eis gegossen, durch Zugabe von 40 %iger Natronlauge auf pH 12-13 gestellt und mit Methylenchlorid ausgeschüttelt. Die organische Lösung wird mit Wasser neutral gewaschen, über Natriumsulfat getrocknet, eingedampft und der feste Rückstand wird aus Benzol 2mal umkristallisiert. Smp. der Titelverbindung: 150-152.
Das Ausgangsmaterial kann wie folgt hergestellt werden: a) 47,5 g 3 -Benzyl-4-oxopiperidin- 1-carbonsäureäthylester werden mit 15,6 g Acetoncyanhydrin versetzt und nach Zugabe einer Spatelspitze mit Methanol angefeuchtetem Kaliumcarbonat wird das Reaktionsgemisch 5-8 Stunden bei Raumtemperatur weitergerührt. Nach 5-10 Stunden Stehen bei Raumtemperatur wird das breiige Reaktionsgemisch mit 20 ml Äther und 20 ml Petroläther versetzt, das feste Produkt abgesaugt, nachgewaschen und bei 50 getrocknet. Smp. des erhaltenen 3-Benzyl-4-cyan-4-hydroxypiperidin-1-carbonsäure äthylesters: 120-121 (aus Äther/Petroläther).
b) Eine Lösung von 16,0 g des vorstehenden Produktes in 20 mi wasserfreiem Pyridin und 14 ml wasserfreiem Benzol wird bei 5" mit 6,5 ml Thionylchlorid tropfenweise versetzt.
Das Reaktionsgemisch wird 5 Stunden bei 0-5" und 12 Stunden bei Raumtemperatur stehen gelassen, auf 100 ml Eiswasser gegossen, die organische Phase wird abgetrennt, mit Wasser neutral gewaschen, über Magnesiumsulfat getrocknet und eingedampft. Das als Öl erhaltene 5-Benzyl-4-cyan-1,2,3, 6-tetrahydropyridin- 1 - carbonsäureäthylester (IR: CN-Bande bei 2230 com') wird roh weiterverarbeitet.
c) Unter starkem Rühren werden 9,8 g des vorstehenden Produktes mit 70 ml 90%iger Schwefelsäure so versetzt, dass die Temperatur 5" nicht übersteigt. Das Gemisch wird 1 Stunde bei 5" und 3-4 Stunden bei Raumtemperatur unter gelegentlichem Umschütteln stehen gelassen. Die erhaltene einheitliche Lösung wird nun auf 500 g Eis gegossen, mit Methylenchlorid extrahiert, die wässrige Lösung durch Zugabe von 40 %iger Natronlauge auf pH 9-10 gestellt und wieder mit Methylenchlorid ausgeschüttelt. Die vereinigten Extrakte werden mit Wasser gewaschen, über Magnesiumsulfat getrocknet und unter Stickstoffatmosphäre eingedampft.
Der als Eindampfrückstand verbleibende feste 5-Amino-1,2,3,4-tetra- hydrobenz[g]isochinolin-2-ylcarbonsäureäthylester wird aus Essigester umkristallisiert. Smp.: 198" unter Zers.
Beispiel 2 1,2,3 ,4-Tetrahydro-5-(1-pyrrolyl)benz[g]isochinolin
Ein Gemisch von 2,0 g 1,2,3,4-Tetrahydro-5-(1-pyrrolyl)benz[g]isochinolin-2-carbonsäureäthylester und 4,0 g Kaliumhydroxid in 20 ml n-Butanol wird 2 Stunden zum Sieden erhitzt. Nach dem Abkühlen auf Raumtemperatur wird es mit Benzol verdünnt, mit Wasser neutral gewaschen und 3mal mit 10 %iger Weinsäurelösung ausgeschüttelt. Die saure Lösung wird mit Äther gewaschen, durch Zugabe von 40 %iger Natronlauge alkalisch gestellt und mit Benzol extrahiert. Die Benzollösungen werden mit Wasser gewaschen, über Magnesiumsulfat getrocknet. Die nach Abdampfen des Lösungsmittels als Rückstand verbleibende rohe Titelverbindung wird in Äthanol gelöst und in ihr Hydrochlorid übergeführt. Smp.: Zers. ab 240".
Das Ausgangsmaterial wird wie folgt hergestellt: a) Ein Gemisch von 30,0 g 5-Amino-1,2,3,4-tetrahydrobenz[g]isochinolin-2-carbonsäureäthylester und 14,7 g 2,5 Dimethoxytetrahydrofuran in 150 ml Eisessig wird 30 Minuten zum Sieden erhitzt und auf Raumtemperatur abgekühlt.
Zum Reaktionsgemisch wird nun eine Lösung von 11,1 g Kaliumhydroxid in 75 ml Methanol und 75 ml Wasser zugegeben und das Gemisch wird 5 Stunden unter Rühren zum Sieden erhitzt. Die erhaltene Reaktionslösung wird nach Abkühlen auf 20 auf Eis gegossen, mit konz. Salzsäure auf pH 2-3 gestellt und mit Äther ausgeschüttelt. Die Ätherextrakte werden mit Wasser und mit gesättigter Kochsalzlösung gewaschen, über Natriumsulfat getrocknet und eingedampft.
Der als Rückstand verbleibende 1,2,3,4-Tetrahydro-5-(1 pyrruWl)benz[gisochinolin-2-carbonsäureäthylester wird aus Äther/Petroläther 2mal umkristallisiert. Smp.: 109-111".
Analog Beispiel 1 oder 2 können die folgenden Verbindungen der Formel I durch Abspaltung der R2-Gruppe aus entsprechenden Verbindungen der Formel II hergestellt werden: Beispiel Endprodukt Ausgangsprodukt Nr. R, R3 R4 Smp. W Rs R4 R2 Smp.

Das Ausgangsmaterial für Beispiel 2B kann wie folgt hergestellt werden: a) Ein Gemisch von 27,0 g 5-Amino-1,2,3,4-tetrahydro benz[g]isochinolin-2-ylcarbonsäureäthylester in 50 ml Wasser wird mit 3 ml Pyridin versetzt. Zu diesem Gemisch wird innert 10 Minuten 50,4 g Dimethylsulfat zugetropft und das Gemisch wird 8 Stunden bei 55-60 gerührt.
Die erhaltene klare Lösung wird mit 100 ml Wasser verdünnt, mit Chloroform mehrmals ausgeschüttelt, die Extrakte werden mit Wasser neutral gewaschen, über Natriumsulfat getrocknet und eingedampft. Der ölige Rückstand wird in Äther aufgenommen, mit ätherischer Chlorwasserstofflösung angesäuert und das ausgefallene Produkt wird abfiltriert und aus Äthanol/Äther umkristallisiert. Smp. vom 1,2,3,4-Tetrahydro-5-methylamino.

The invention relates to new tetrahydrobenz [g] isoquinolines of the formula I, in which

According to the invention, the new compounds of the formula I and their acid addition salts are obtained by splitting off the radical R2 by solvolysis from compounds of the formula II in which R1, R3 and R4 have the above meaning and R2 is a radical which can be cleaved off by solvolysis, and the resulting compounds of Formula I, if desired, converted into their acid addition salts.

The compounds of the formula I and their physiologically acceptable acid addition salts are distinguished by interesting pharmacological properties and can therefore be used as medicaments.
In particular, the compounds show anti-aggressive effects in animal experiments and are able, for. B. to dampen aggressive behavior of mice caused by isolation.

Example 2 1,2,3,4-Tetrahydro-5- (1-pyrrolyl) benz [g] isoquinoline
