worin Ro, Rl, R2, R3, R4 und Rs wie oben definiert sind und R' eine Alkylgruppe ist, die wie oben definiert ist, zusammen mit Titantetrachlorid auf eine Temperatur zwischen etwa 40 und 160"C erhitzt.
Die Produkte der Formel I als auch die erfindungsgemäss erhaltenen Produkte der Formel II zeigen eine Aktivität als Tranquilizer. Die bekannten Verbindungen der Formel II dieser Patentschrift wurden in der US-Patentanmeldung Serien Nr. 169 091, eingereicht am 4. August 1971, beschrieben.
Der Beruhigungseffekt von erfindungsgemäss hergestellten Verbindungen wird anhand der folgenden Tests mit Mäusen gezeigt.
Kamintest (Chimney test):
Der Test bestimmt die Fähigkeit von Mäusen aus einem vertikalen Glaszylinder innerhalb 30 Sekunden nach oben herauszukriechen. Der EDso-Wert entspricht derjenigen Dosierung, bei welcher dies 50% der Mäuse nicht gelingt.
Schalentest (Dish test):
Mäuse, die in Petri-Schalen von 10 cm Durchmesser und 5 cm Randhöhe, die teilweise in Holzspänen oder Sägespänen eingebettet waren, gesetzt wurden, kletterten aus diesen innerhalb sehr kurzer Zeit heraus, wenn sie unbehandelt waren. Mäuse die während mehr als 3 Minuten in der Petrischale verblieben, zeigten Tranquilizator-Wirkung. Die EDso entspricht derjenigen Dosis der Testverbindung, bei welcher 50% der Mäuse in der Petrischale blieben.
Podest-Test (Pedestal test):
Unbehandelte Mäuse verlassen den Podest in weniger als 1 Minute, um auf den Boden des Standard-Mauskäfigs zurückzukehren. Mäuse, die unter Tranquilizator-Wirkung stehen, bleiben auf dem Podest während mehr als 1 Minute.
Nikotin-Antagonismus-Test :
Eine Gruppe von 6 Mäusen wurde mit der Testverbindung behandelt. Nach 30 Minuten wurden diese Mäuse und darüber hinaus unbehandelte Mäuse des Kontrollversuches mit Nikotinsalicylat in einer Dosis von 2 mg/kg Körpergewicht durch Injektion behandelt. Die Mäuse des Kontrollversuches zeigten Übererregbarkeit (Hyperstimulation) d. h. (1) durchlaufende Konvulsionen und nachfolgende (2) tonische Streckanfälle, die (3) vom Tod gefolgt wurden.

Die pharmazeutischen Formen, die hier in Frage stehen, sind pharmazeutische Zusammensetzungen, die geeignet sind für die orale, parenterale und rektale Anwendung, wie z. B.
Tabletten, Pulverpakete, Arzneikapseln, Dragees, andere Behälterformen, Lösungen, Suspensionen, sterile injizierbare Formen, Suppositorien, in Körperhöhlungen einführbare arzneimittelhaltige Verabreichungsformen (Bougies) und ähnliche. Geeignete Verdünnungsmittel oder Trägermittel sind z. B. Kohlehydrate, Lactose, Proteine, Lipide, Kalziumphosphat, Maisstärke, Stearinsäure, Methylcellulose und ähnliche, die als Trägermaterialien oder zu Überzugszwecken verwendet werden können. Darüberhinaus sind Öle, wie z. B. Kokosnussöl, Sesamöl, Saffloröl, Baumwollsamenöl, Erdnussöl und Wasser zur Herstellung von Lösungen oder Suspensionen des aktiven Materiales anwendbar. Es können auch süssungs-, färbe- und geschmackgebende Mittel zugesetzt werden.
Für Säugetiere und Vögel sind Futtervormischungen mit Stärke, Hafermehl, getrocknetem Fischfleisch, Fischmehl, Mehl und ähnlichen anwendbar.
Bei Anwendung der Verbindungen II oder deren pharmakologisch akzeptablen Säureadditionssalze als Tranquilizer können Dosierungen von 0,1 bis 10,0 mg/kg Körpergewicht bei der oralen oder injizierbaren Verabreichungsform wie oben beschrieben angewandt werden, um Spannungszustände und Angstzustände bei Säugetieren oder Vögeln auszugleichen, wie sie z. B. entstehen, wenn Tiere über weitere Strekken transportiert werden (Reisekrankheit).
Andere Säureadditionssalze der Verbindungen der allgemeinen Formel II können ebenso hergestellt werden, wie z. B. die Fluorsiliciumsäureadditionssalze, welche als Verbindungen für die mottensichere Ausrüstung anwendbar sind, oder die Trichloracetate, die wirksam sind als Herbizide, z. B. gegen Johnson Gras , Bermuda Gras , gelben und grünen Fuchsschwanz und Quäcken.
Die Ausgangsmaterialien der Formel I können nach Verfahrensweisen hergestellt werden, die im allgemeinen nach dem Stand der Technik bekannt sind wie z. B. von R. A. Baxter et al., J. Chem. Soc., (London) 370 (1947) beschrieben oder wie es im folgenden bei den Präparationen gezeigt wird.
Bei der Ausführung des neuartigen erfindungsgemässen Verfahrens wird ein gewähltes [5-Phenyl-[1,2-dihydro 3H-1,4-benzodiazepin-2-yliden]amino]säurealdehyddialkyl- acetal I mit Titantetrachlorid auf Temperaturen zwischen etwa 40 und etwa 1600C in einem geeigneten organischen Lösungsmittel erhitzt. In einer bevorzugten Ausführungsform des erfindungsgemässen Verfahrens wird die Reaktion zuerst bei Zimmertemperatur während bis zu 5 Minuten ablaufen gelassen und sodann wird Hitze angewandt. Am günstigsten wird die gewählte Reaktionstemperatur diejenige sein, welche derjenigen Temperatur entspricht, bei welcher die Reaktionsmischung unter Rückfluss siedet. Als Lösungsmittel werden organische Lösungsmittel angewandt, wie z. B.
Methylenchlorid, Chloroform, Äthylenchlorid, Tetrahydrofuran, 1,2 Dimethoxyäthan, Glycolmonoäthyläther (Monoglyme), Diäthylenglycol-dimethyläther, Diglycoldimethyläther (Diglyme) und ähnliche.
Die Heizdauer beträgt gewöhnlich 1 bis 36 Stunden und im Falle von Chloroform und Mono- oder Diglyme ist im allgemeinen eine Erhitzung von 3 bis 8 Stunden ausreichend. Nach der Beendigung der Reaktion kann das Produkt (II) mittels üblicher Verfahrensweisen isoliert werden, wie z. B. durch Kühlen der Mischung und Ablöschen in wässriger Natronlauge oder Kalilauge, Entfernung der Titansalze durch Filtration, Extraktion mit wasserunmischbarem Lösungsmittel, beispielsweise mit Chloroform, Konzentrieren des Extraktes und Reinigung mittels Chromatographie und/oder Kristallisation, um die entsprechende Verbindung der Formel II zu erhalten. Die Erfindung sei nun anhand der folgenden Präparationen und Beispiele näher erläutert.
Präparat 1
Herstellung von a-Brompropianaldehyd-dimethylacetal
Zu einer Lösung von 0,5 Mol (29 g) Propionaldehyd in 400 ml wasserfreiem Methanol wurde unter Rühren Brom tropfenweise zugegeben, während man mit einer 250 Watt-Lampe weisses Licht in den Reaktionsbehälter einstrahlte. Die Wärme, die von der Lampe geliefert wurde, war ausreichend, um eine Temperatur der Lösung von 50 bis 55OC aufrechtzuerhalten. In dieser Weise konnten sämtliche 81 g (0,50 Mol) Brom tropfenweise während eines Zeitraumes von 8 Stunden zugesetzt werden, so dass nur eine schwach gelbe Färbung, die von überschüssigem Brom herrührte, während der Reaktion auftrat. Die farblose Reaktionslösung wurde sodann auf 15 C gekühlt und wurde vorsichtig in 400 ml Hexan, das 80 g wasserfreies Natriumcarbonat enthielt, eingegossen.
Nachdem die Gasentwicklung aufgehört hatte, wurde 500 ml kaltes Wasser zugesetzt und die Temperatur wurde bei oder unter Zimmertemperatur (etwa 23 C) gehalten. Das Produkt wurde mit Hexan extrahiert, über wasserfreiem Magnesiumsulfat getrocknet und man engte auf dem Wasserbad unter Anwendung einer Vigreux-Kolonne ein. Die Destillation ergab drei Fraktionen einer farblosen Flüssigkeit, die einen Siedepunkt von 55 bis 56"C bei 13 Torr aufwies, in einer Ausbeute von 46 g mit einem Brechungsindex von N25 = 1,6588.
Die Elementaranalyse auf Basis der Summenformel CsHiiBrO ergab die folgenden Werte:

Herstellung von a-Azidopropionaldehyd-dimethylacetal a-Brompropionaldehyd-dimethylacetal (34,8 g, 0,19 Mol) wurde in 150 ml Dimethylformamid gelöst und 32,5 g (0,50 Mol) Natriumazid behandelt. Die Mischung wurde auf 20OC während 3 vollen Tagen erwärmt. Die Reaktionsmischung wurde dadurch aufgearbeitet, dass man zuerst auf Raumtemperatur abkühlte, in kaltes Wasser (mit einem pH-Wert von über 9) eingoss und mit Benzol extrahierte. Die organische Schicht wurde über wasserfreiem Magnesiumsulfat getrocknet und aus dem Wasserbad unter Anwendung einer Vigreux Kolonne eingeengt.
Das zurückbleibende gelbe Ol wurde bei 300 Torr destilliert und ergab 27,5 g einer farblosen Flüssigkeit mit einem Siedepunkt von 67 bis 70oC, welche das Azid und eine kleine Menge des Ausgangsmaterials enthielt.
Die Elementaranalyse auf Basis der Summenformel C5HllN302 ergab die folgenden Werte:

Herstellung von a-Aminopropionaldehyd a-Azidopropionaldehyd-dimethylacetal wurde in 150 ml absolutem Äthanol gelöst und mit Wasserstoff reduziert, wobei man 0,50 g eines Katalysators von 10% Palladium auf Aktivkohle in einer Parr-Apparatur bei Raumtemperatur und 3,52 Atmosphären-Anfangsdruck anwandte. Da ein Äquivalent Stickstoff pro Äquivalent Wasserstoff entsteht, entstand kein Druckabfall während der Reaktion. Nach der über Nacht geführten Reaktion wurde der Katalysator abfiltriert und mit absolutem Äthanol gewaschen. Das Äthanol wurde auf dem Wasserbad über eine Vigreux-Kolonne entfernt. Das zurückbleibende Öl wurde in eine kalte wässrige Natronlaugelösung eingegossen (150C; pH 10) und mit Hexan extrahiert.
Das Hexanextrakt entfernte das gesamte Bromacetal, aber nicht das Aminprodukt aus der Wasserschicht. Die wässrige Schicht wurde auf einen pH-Wert von grösser als 13 (unter Verwendung von Natronlauge) eingestellt und mit Chloroform extrahiert. Die Chloroformschicht wurde über wasserfreiem Magnesiumsulfat getrocknet und ergab 11,8 g von a-Aminopropionaldehyd in Form eines farblosen Öles, das einen Siedepunkt von 78 C bei 67 Torr zeigte.

Herstellung von [[7-Chlor-5-phenyl- 1 ,2-dihydro- 3H-1,4-benzodiazepin-2-yliden]amino]-acetaldehyd- dimethylacetal
Eine Lösung aus 27,1 g (0,1 Mol) 7-Chlor-1,2-dihydro 5-phenyl-3H-1,4-benzodiazepin-2-on und 100 ml Aminoacetaldehyd-dimethylacetal (Aldrich) in 700 ml Tetrahydrofuran wurde auf OoC mittels eines Eiskochsalzbades gekühlt. Der Titantetrachloridkomplex wurde dadurch hergestellt, dass man langsam unter heftigem Rühren 10,57 g (0,55 Mole) Titantetrachlorid zu 330 ml Tetrahydrofuran zusetzt. Diese Lösung wurde langsam unter Rühren während 10 Minuten zur oben angeführten Lösung bei einer Temperatur von -5 bis O"C zugesetzt.
Nach einer Verweilzeit von 0,5 Stunden bei OoC wurde die Temperatur auf Raumtemperatur ansteigen gelassen und die Lösung wurde weiter über Nacht gerührt. Sodann wurden weitere 5 ml Titantetrachlorid tropfenweise zugesetzt und die Lösung wurde während 7,5 Stunden gerührt und sodann über Nacht stehen gelassen. Schliesslich wurden 65 ml Wasser langsam unter heftigem Rühren zugegeben. Die Mischung wurde filtriert und das Material wurde im Vakuum auf ein kleines Volumen eingeengt und mit Wasser verdünnt, wodurch man 34,8 g [[7-Chlor-5-phenyl-1,2-dihydro-3H-1,4-benzodiaz- epin-2-ylidenjarnino]-acetaldehyd-dimethylacetal mit einem Schmelzpunkt von 142-154"C erhielt.
Dieses Material wurde in 400 ml 2-Propanol gelöst, heiss filtriert und auf 350 ml eingeengt und sodann gekühlt wodurch man 26,94 g (75,4%) [[7-Chlor-5-phenyl- 1,2-dihydro-3H-1,4- benzodiazepin-2-yliden]amino]acetaldehyd-dimethylacetal in Form eines weissen kristallinen Feststoffes mit einem Schmelzpunkt von 158,5 bis 160,5 C erhielt.
Die Elementaranalyse auf Basis der Summenformel C19H20C1N302 ergab die folgenden Werte:

Herstellung von [[5-(o-Chlorphenyl)-1,2-dihydro-3H-1,4 benzodiazepin-2-yliden]amino]acetaldehyd-dimethylacetal
Eine Lösung aus 11,4 g (40 mMol) 1,2-Dihydro-5-(o-chlorphenyl)-3H-1,4-benzodiazepin-2-thion in 400 ml N-Butanol wurde zusammen mit 0,12 Mol (12,6 g) Aminoacetaldehyddimethylacetal während 7 Stunden unter Rückfluss erhitzt und sodann über das Wochenende stehen und abkühlen gelassen.
Das Butanol wurde im Vakuum entfernt und man erhielt einen Feststoff, der in heissem Essigester gelöst, filtriert und abgekühlt wurde, wodurch man 14,08 g [[5-(o Chlorphenyl)-1 ,2-dihydro-2H-1 ,4-benzodiazepin-2-yliden] amino]acetaldehyd-dimethylacetal in zwei Fraktionen erhielt, welche einen Schmelzpunkt von 179 bis 182"C zeigten.
Eine analytische Probe kleiner weisser Prismen zeigte einen Schmelzpunkt von 181 bis 1820C.
Die Elementaranalyse auf Basis der Summenformel C19H20C1N302 ergab die folgenden Werte:

Herstellung von [[7-Chlor-5-(o-chlorphenyl)-1,2-dihydro- 3H- 1 ,4-benzodiazepin-2-yliden]aminojacetaldehyd- dimethylacetal
Eine Lösung von 7-Chlor-5-(o-chlorphenyl)-1,2-dihydro3H-1,4-benzodiazepin-2-thion (6,40 g, 0,020 Mol) und Aminoacetaldehyd-dimethylacetal (6,30 g, 0,060 Mol) wurde in 200 ml N-Butanol hergestellt und während 3 Stunden unter Rüclcfluss erhitzt. Die Reaktionsmischung wurde sodann auf Zimmertemperatur über Nacht abkühlen gelassen.
Der so entstandene Niederschlag wurde abfiltriert, mit Essigester und Hexan gewaschen und man erhielt 66,7 g (85,4%) eines weissen Pulvers mit einem Schmelzpunkt von 176 bis 177,50C. Die Mutterlauge wurde im Vakuum eingeengt und man erhielt 1,6 g eines braunen Feststoffes, der nicht weiter untersucht wurde. Eine analytische Probe weisser Prismen, die aus Essigester-Hexan erhalten wurde, zeigte einen Schmelzpunkt von 175,5 bis 176,50C.
Die Elementaranalyse auf Basis der Summenformel C19H19C12N302 ergab die folgenden Werte:

Herstellung von [[7-Chlor-5-(o-chlorphenyl)-1,2-dihydro-
3H- 1,4-benzodiazepin-2-yliden] a-amino]propion- aldehyd-dimethylacetal
Eine Lösung aus 7-Chlor-1 ,2-dihydro-2H-5-(o-chlorphen- yl)-1,4-benzodiazepin-2-thion (6,40 g, 0,020 Mol) und a-Amino-propionaldehyd-dimethylacetal (6,30 g, 0,050 Mol) in 200 ml N-Butanol wurde während 20 Stunden unter Rückfluss erhitzt. Die Lösung wurde sodann auf Zimmertemperatur gekühlt und das N-Butanol wurde im Vakuum entfernt wodurch man ein rot-oranges Öl erhielt. Das erwünschte Produkt wurde durch Behandlung mit Essigester-Hexan erhalten.
Die Ausbeute betrug 4,80 g festes [[7-Chlor-5 (o-chlorphenyl)-1,2-dihydrn-2H- 1,4-benzodiazepin-2-y1- iden]-a-amino]propionaldehyd-dimethylacetal mit einem Schmelzpunkt von 153 bis 155 C. Eine analytische Probe zeigte einen Schmelzpunkt von 157 C.
Die Elementaranalyse auf Basis der Summenformel C20H21ClN23Ol ergab die folgenden Werte:

Beispiel 1
Herstellung von 8-Chlor-6-phenyl-4H imidazo [I,2-a] [1,4]benzodiazepin [[7-Chlor-5-phenyl- 1,2-dihydro-2H-l ,4-benzodiazepin 2-yliden]amino]acetaldehyd-dimethylacetal (5,4 g, 0,015 Mol) wurde in 180 ml Monoglyme gelöst und vorsichtig mit 2,85 ml (4,90 g, 0,026 Mol) Titantetrachlorid behandelt. Die schwarze Lösung wurde während 4 Stunden unter Rückfluss erhitzt, während welcher Zeit das gesamte Ausgangsmaterial und das unidentifizierte Zwischenprodukt reagiert hatten. (Das Ausgangsmaterial hatte innerhalb einer halben Stunde reagiert). Die Reaktionsmischung wurde sodann auf Zimmertemperatur abgekühlt, in 450 ml 5 %iger kalter wässriger Natriumhydroxydlösung eingegossen und mit Methylenchlorid extrahiert. Die Titansalze wurden durch Filtration entfernt.
Der organische Extrakt wurde mit gesättigter wässriger Natriumchloridlösung gewaschen, über wasserfreiem Magnesiumsulfat getrocknet und im Vakuum eingeengt und das Produkt aus Essigester umkristallisiert, wodurch man 2,8 g (63,6%) 8 Chlor-6-phenyl-4H-imidazo[1 ,2-a] [1 ,4]benzodiazepin mit einem Schmelzpunkt von 147-148,5"C erhielt. Eine analytische Probe zeigte einen Schmelzpunkt von 147,5 bis 149 C.
Die Elementaranalyse auf Basis der Summenformel C17Hl2CIN3 zeigte die folgenden Werte:

Beispiel 2
Herstellung von 6-(o-Chlorphenyl).4H- imidazo [1,2-aj[1,4]benzodiazepin
Eine Lösung aus 8,1 g (22,5 mMol) [[5-(o-Chlorphenyl)1,2-dihydro-3H- 1,4-benzodiazepin-2-yliden]amino]acetal- dehyd-dimethylacetal in 270 ml Monoglyme wurde mit 3,38 ml Titantetrachlorid behandelt. Nachdem die anfänglich heftige Reaktion aufgehört hatte, wurde die Mischung auf dem Wasserbad während 5 Stunden zum Rückfluss erhitzt. Die Reaktionsmischung wurde auf Zimmertemperatur gekühlt, in 800 ml kalte (10oC) 5 %ige wässrige Natriumhydroxydlösung eingegossen und mit 1 Liter Chloroform extrahiert.
Die Chloroformschicht wurde mit gesättigter Kochsalzlösung gewaschen, über wasserfreiem Magnesiumsulfat getrocknet und zu einem braunen Öl konzentriert. Das Öl wurde über 900 g Siliciumdioxyd-Gel chromatographiert und mit Essigester und 5% und 10% Methanol-Essigester eluiert. Das Produkt wurde im polaren Lösungsmittel eluiert und gab eine totale Ausbeute von 5,37 g eines Öles. Im gesamten wurde 214 g eines leicht braunen kristallinen Feststoffes erhalten durch Umkristallisieren aus Essigester/Skellysolve-B Hexan, welcher einen Schmelzpunkt von 148,5-150"C aufwies.
Die Elementaranalyse auf Basis der Summenformel C17Hl2CIN3 ergab die folgenden Werte:

Beispiel 3
Herstellung von 8-Chlor-6-(o-chlorphenyl)
4H-imidazo[1,2-a][1,4]benzodiazepin [[7-Chlor-5-(o-chlorphenyl)-1,2-dihydro-2H-1,4-benzo- diazepin-2-yliden]amino]acetaldehyd-dimethylacetal (7,84 g, 0,020 Mol) wurde in 100 ml Chloroform gelöst. Zu der Lösung, die gerührt wurde, wurden in 2 Portionen 3,0 ml Titantetrachlorid (5,20 g, 0,027 Mol) zugesetzt. Die Lösung bekam eine orange-braune Farbe und wurde handwarm. Ein kleiner Anteil eines braunen öligen Feststoffes schied sich von der Lösung ab. Die Mischung wurde während 5 Stunden unter Rückfluss gehalten, während welcher Zeit 30 ml Monoglym zugesetzt wurden.
Man erhitzte unter Rückfluss über Nacht weiter. Die Aufarbeitung bestand darin, dass man die Reaktionsmischung in eine kalte (0 bis 5 C) Mischung aus einer 5 %igen wässrigen Natriumhydroxydlösung und Chloroform eingoss. Die Mischung wurde filtriert, um die Titansalze zu entfernen. Der Feststoff wurde gründlich mit Chloroform und 5 %iger wässriger Natriumhydroxydlösung gewaschen. Die Chloroformphasen wurden vereinigt, über wasserfreiem Magnesiumsulfat getrocknet und im Vakuum eingeengt, wodurch man 6,6 g eines braunen Feststoffes erhielt.
Dieses Material wurde aus Essigester-Hexan umkristallisiert und man erhielt 5,3 g (81%) 8-Chlor-6-(o-chlorphenyl) -4H-imidazo[1,2-a][1,4]benzodiazepin mit einem Schmelzpunkt von 177,5 bis 179 C. Eine zweite Kristallfraktion (0,53 g, 8%) wurde ebenfalls gesammelt. Eine analytische Probe zeigte einen Schmelzpunkt von 179 bis 1800C.

Herstellung von 8-Chlor-2-methyl-6-(o-chlor phenyl)-4H-imidazolo[1,2-a][1,4]benzodiazepin
Eine Probe von [[7-Chlor-5-(o-chlorphenyl)-1,2-di- hydro-3H-1,4-benzodiazepin-2-yliden]-a-amino]propionaldehyd-dimethylacetal (2,03 g, 5,000 mMol) wurde in 60 ml Monoglyme gelöst und mit 0,75 ml (1,30, 6,85 mMol) Titantetrachlorid behandelt. Es trat eine heftige Reaktion auf und es schied sich im Reaktionsbehälter ein brauner Feststoff. Die Mischung wurde unter eine Stickstoffatmosphäre während 4 Stunden unter Rückfluss erwärmt.
Sodann wurde die Reaktionsmischung auf Zimmertemperatur gekühlt, in 250 ml kalter 5 %iger wässriger Natriumlauge eingegossen und mit 400 ml Chloroform extrahiert. Die Chloroformschicht wurde mit konzentrierter Kochsalzlösung gewaschen, über wasserfreiem Magnesiumsulfat getrocknet und im Vakuum eingeengt, wodurch man einen Feststoff erhielt. Das 8 -Chlor-2-methyl-6-(o-chlorphen yl)-4H-imidazolo[1,2-a][1,4]benzodiazepin, das so erhalten worden war, wurde in zwei Anteilen aus Essigester umkristallisiert, wodurch man 960 mg Substanz in Form von Prismen erhielt, die einen Schmelzpunkt von 169,5 bis 170,5cm zeigten.

Herstellung von 8-Chlor-6- (2,6-difluorphenyl) -4H imidazo[1,2-aj[1,4]benzodiazepin
In der gleichen Weise wie in Beispiel 1 beschrieben, wird [[7-Chlor-5-(2,6-difluorphenyl)-1,2-dihydro-3H- 1 ,4-benzodiazepin-2-yliden]amino]acetaldehyd-dimethyl- acetal in Tetrahydrofuran zusammen mit Titantetrachlorid erhitzt, wodurch man 8 -Chlor-6-(2,6-difluorpheayl)-4H- imidazo[1,2-a][1,4jbenzodiazepin erhält.
Beispiel 7
Herstellung von 8 -Nitro-6-(o-chlorphenyl-4H- imidazo[1,2-a] [1,4]benzodiazepin
In der gleichen Weise wie in Beispiel 1 beschrieben, wird [[7-Nitro-5-(o-chlorphenyl)-1 ,2-dihydro-3H-1,4- benzodiazepin-2-yliden]amino]acetaldehyd.dimethylacetal in Monoglyme zusammen mit Titantetrachlorid erhitzt und man erhält 8-Nitro-6-(o-chlorphenyl)-4H-imidazo[1 ,2-a) [1,4]benzodiazepin.

Herstellung von 10-Isopropyl-4-methyl-6-(o methoxyphenyl)-4H-imidazo[1,2-a][1,4]benzodiazepin

In der gleichen Weise wie in den vorhergehenden Beispielen wurde durch Behandlung eines bestimmten [5-Phenyl- 1,2-dihydro-3H- 1,4-benzodiazepin-2-yliden[-c-amino]- 2-aldehyd-dialkylacetal oder -keton-dialkylketals (I) mit Titantetrachlorid in einem geeigneten organischen Lösungsmittel, vorzugsweise bei der Temperatur, bei welcher Rückfluss stattfindet, ein 6-Phenyl-4H-imidazo[1,2-a] [1,4]benzodiazepin der allgemeinen Formel II erhalten.

Preparation of α-Azidopropionaldehyde Dimethylacetal α-Bromopropionaldehyde dimethylacetal (34.8 g, 0.19 mol) was dissolved in 150 ml of dimethylformamide and 32.5 g (0.50 mol) of sodium azide was treated. The mixture was warmed to 20OC for 3 full days. The reaction mixture was worked up by first cooling to room temperature, pouring it into cold water (with a pH of over 9) and extracting with benzene. The organic layer was dried over anhydrous magnesium sulfate and concentrated from the water bath using a Vigreux column.
The remaining yellow oil was distilled at 300 torr to give 27.5 g of a colorless liquid with a boiling point of 67 to 70 ° C. which contained the azide and a small amount of the starting material.
The elemental analysis based on the empirical formula C5HllN302 resulted in the following values:

Preparation 3
Preparation of α-aminopropionaldehyde α-azidopropionaldehyde dimethylacetal was dissolved in 150 ml of absolute ethanol and reduced with hydrogen, 0.50 g of a catalyst of 10% palladium on activated carbon in a Parr apparatus at room temperature and 3.52 atmospheres initial pressure applied. Since there is one equivalent of nitrogen per equivalent of hydrogen, there was no pressure drop during the reaction. After the reaction carried out overnight, the catalyst was filtered off and washed with absolute ethanol. The ethanol was removed on the water bath via a Vigreux column. The oil that remained was poured into a cold aqueous sodium hydroxide solution (150 ° C.; pH 10) and extracted with hexane.
The hexane extract removed all of the bromo acetal but not the amine product from the water layer. The aqueous layer was adjusted to a pH greater than 13 (using sodium hydroxide solution) and extracted with chloroform. The chloroform layer was dried over anhydrous magnesium sulfate and gave 11.8 g of α-aminopropionaldehyde in the form of a colorless oil, which had a boiling point of 78 ° C. at 67 torr.

Preparation of [[7-chloro-5-phenyl-1, 2-dihydro-3H-1,4-benzodiazepin-2-ylidene] amino] acetaldehyde dimethyl acetal
A solution of 27.1 g (0.1 mol) of 7-chloro-1,2-dihydro 5-phenyl-3H-1,4-benzodiazepin-2-one and 100 ml of aminoacetaldehyde dimethylacetal (Aldrich) in 700 ml of tetrahydrofuran was cooled to OoC using an ice-salt bath. The titanium tetrachloride complex was prepared by slowly adding 10.57 g (0.55 moles) of titanium tetrachloride to 330 ml of tetrahydrofuran with vigorous stirring. This solution was slowly added to the above solution at a temperature of -5 to 0 "C over 10 minutes with stirring.
After a residence time of 0.5 hours at OoC, the temperature was allowed to rise to room temperature and the solution was stirred further overnight. An additional 5 ml of titanium tetrachloride was then added dropwise and the solution was stirred for 7.5 hours and then allowed to stand overnight. Finally, 65 ml of water were slowly added with vigorous stirring. The mixture was filtered and the material was concentrated in vacuo to a small volume and diluted with water to give 34.8 g of [[7-chloro-5-phenyl-1,2-dihydro-3H-1,4-benzodiaz- epin-2-ylidenjarnino] acetaldehyde dimethylacetal with a melting point of 142-154 "C.
This material was dissolved in 400 ml of 2-propanol, filtered hot and concentrated to 350 ml and then cooled, whereby 26.94 g (75.4%) of [[7-chloro-5-phenyl-1,2-dihydro-3H -1,4-benzodiazepin-2-ylidene] amino] acetaldehyde dimethylacetal in the form of a white crystalline solid with a melting point of 158.5 to 160.5 ° C.

Preparation of [[5- (o-chlorophenyl) -1,2-dihydro-3H-1,4 benzodiazepin-2-ylidene] amino] acetaldehyde dimethyl acetal
A solution of 11.4 g (40 mmol) of 1,2-dihydro-5- (o-chlorophenyl) -3H-1,4-benzodiazepine-2-thione in 400 ml of n-butanol was added together with 0.12 mol ( 12.6 g) Aminoacetaldehyde dimethyl acetal heated under reflux for 7 hours and then left to stand and cool over the weekend.
The butanol was removed in vacuo and a solid was obtained which was dissolved in hot ethyl acetate, filtered and cooled, whereby 14.08 g of [[5- (o chlorophenyl) -1, 2-dihydro-2H-1, 4- benzodiazepin-2-ylidene] amino] acetaldehyde dimethylacetal was obtained in two fractions which had a melting point of 179 to 182 "C.
An analytical sample of small white prisms showed a melting point of 181 to 1820C.

Preparation of [[7-chloro-5- (o-chlorophenyl) -1,2-dihydro-3H-1,4-benzodiazepin-2-ylidene] aminojacetaldehyde dimethylacetal
A solution of 7-chloro-5- (o-chlorophenyl) -1,2-dihydro3H-1,4-benzodiazepine-2-thione (6.40 g, 0.020 mol) and aminoacetaldehyde dimethyl acetal (6.30 g, 0.060 Mol) was prepared in 200 ml of n-butanol and heated under reflux for 3 hours. The reaction mixture was then allowed to cool to room temperature overnight.
The resulting precipitate was filtered off, washed with ethyl acetate and hexane, and 66.7 g (85.4%) of a white powder with a melting point of 176 to 177.50 ° C. were obtained. The mother liquor was concentrated in vacuo and 1.6 g of a brown solid were obtained, which was not investigated further. An analytical sample of white prisms obtained from ethyl acetate-hexane showed a melting point of 175.5 to 176.50C.

Preparation of [[7-chloro-5- (o-chlorophenyl) -1,2-dihydro-
3H-1,4-benzodiazepin-2-ylidene] α-amino] propionaldehyde dimethyl acetal
A solution of 7-chloro-1, 2-dihydro-2H-5- (o-chlorophenyl) -1,4-benzodiazepine-2-thione (6.40 g, 0.020 mol) and a-amino-propionaldehyde dimethylacetal (6.30 g, 0.050 mol) in 200 ml of n-butanol was refluxed for 20 hours. The solution was then cooled to room temperature and the n-butanol was removed in vacuo to give a red-orange oil. The desired product was obtained by treatment with ethyl acetate-hexane.
The yield was 4.80 g of solid [[7-chloro-5 (o-chlorophenyl) -1,2-dihydric-2H-1,4-benzodiazepin-2-y1-iden] -a-amino] propionaldehyde dimethyl acetal with a melting point of 153 to 155 C. An analytical sample showed a melting point of 157 C.

Preparation of 8-chloro-6-phenyl-4H imidazo [I, 2-a] [1,4] benzodiazepine [[7-chloro-5-phenyl-1,2-dihydro-2H-l, 4-benzodiazepine 2- ylidene] amino] acetaldehyde dimethylacetal (5.4 g, 0.015 mol) was dissolved in 180 ml of monoglyme and carefully treated with 2.85 ml (4.90 g, 0.026 mol) of titanium tetrachloride. The black solution was refluxed for 4 hours, during which time all of the starting material and the unidentified intermediate had reacted. (The starting material had reacted within half an hour). The reaction mixture was then cooled to room temperature, poured into 450 ml of 5% cold aqueous sodium hydroxide solution and extracted with methylene chloride. The titanium salts were removed by filtration.
The organic extract was washed with saturated aqueous sodium chloride solution, dried over anhydrous magnesium sulfate and concentrated in vacuo, and the product was recrystallized from ethyl acetate, giving 2.8 g (63.6%) of 8-chloro-6-phenyl-4H-imidazo [1, 2-a] [1, 4] benzodiazepine with a melting point of 147-148.5 "C. An analytical sample showed a melting point of 147.5 to 149 C.

Preparation of 6- (o-chlorophenyl) .4H-imidazo [1,2-aj [1,4] benzodiazepine
A solution of 8.1 g (22.5 mmol) [[5- (o-chlorophenyl) 1,2-dihydro-3H-1,4-benzodiazepin-2-ylidene] amino] acetaldehyde dimethylacetal in 270 ml Monoglyme was treated with 3.38 ml of titanium tetrachloride. After the initially violent reaction had ceased, the mixture was refluxed on the water bath for 5 hours. The reaction mixture was cooled to room temperature, poured into 800 ml of cold (10 ° C.) 5% aqueous sodium hydroxide solution and extracted with 1 liter of chloroform.
The chloroform layer was washed with saturated saline, dried over anhydrous magnesium sulfate and concentrated to a brown oil. The oil was chromatographed over 900 g of silicon dioxide gel and eluted with ethyl acetate and 5% and 10% methanol-ethyl acetate. The product was eluted in the polar solvent and gave a total yield of 5.37 g of an oil. Overall, 214 g of a slightly brown crystalline solid were obtained by recrystallization from ethyl acetate / Skellysolve-B hexane, which had a melting point of 148.5-150 ° C.

Production of 8-chloro-6- (o-chlorophenyl)
4H-imidazo [1,2-a] [1,4] benzodiazepine [[7-chloro-5- (o-chlorophenyl) -1,2-dihydro-2H-1,4-benzodiazepin-2-ylidene] amino] acetaldehyde dimethyl acetal (7.84 g, 0.020 mol) was dissolved in 100 ml of chloroform. To the solution, which was being stirred, was added 3.0 ml of titanium tetrachloride (5.20 g, 0.027 mol) in 2 portions. The solution turned orange-brown in color and became lukewarm. A small portion of a brown oily solid separated from the solution. The mixture was refluxed for 5 hours during which time 30 ml of monoglyme was added.
The reflux was continued overnight. The work-up consisted of pouring the reaction mixture into a cold (0 to 5 ° C.) mixture of a 5% strength aqueous sodium hydroxide solution and chloroform. The mixture was filtered to remove the titanium salts. The solid was washed thoroughly with chloroform and 5% aqueous sodium hydroxide solution. The chloroform phases were combined, dried over anhydrous magnesium sulfate and concentrated in vacuo to give 6.6 g of a brown solid.
This material was recrystallized from ethyl acetate-hexane to give 5.3 g (81%) of 8-chloro-6- (o-chlorophenyl) -4H-imidazo [1,2-a] [1,4] benzodiazepine having a melting point from 177.5 to 179 C. A second crystal fraction (0.53 g, 8%) was also collected. An analytical sample showed a melting point of 179 to 1800C.

Preparation of 8-chloro-2-methyl-6- (o-chlorophenyl) -4H-imidazolo [1,2-a] [1,4] benzodiazepine
A sample of [[7-chloro-5- (o-chlorophenyl) -1,2-di-hydro-3H-1,4-benzodiazepin-2-ylidene] -a-amino] propionaldehyde dimethylacetal (2.03 g , 5,000 mmol) was dissolved in 60 ml of monoglyme and treated with 0.75 ml (1.30, 6.85 mmol) of titanium tetrachloride. A vigorous reaction occurred and a brown solid separated out in the reaction vessel. The mixture was refluxed under a nitrogen atmosphere for 4 hours.
The reaction mixture was then cooled to room temperature, poured into 250 ml of cold 5% aqueous sodium hydroxide solution and extracted with 400 ml of chloroform. The chloroform layer was washed with concentrated saline, dried over anhydrous magnesium sulfate and concentrated in vacuo to give a solid. The 8-chloro-2-methyl-6- (o-chlorophen yl) -4H-imidazolo [1,2-a] [1,4] benzodiazepine thus obtained was recrystallized in two portions from ethyl acetate, whereby 960 mg of substance were obtained in the form of prisms which had a melting point of 169.5 to 170.5 cm.

In the same manner as described in Example 1, [[7-chloro-5- (2,6-difluorophenyl) -1,2-dihydro-3H-1,4-benzodiazepin-2-ylidene] amino] acetaldehyde-dimethyl acetal heated in tetrahydrofuran together with titanium tetrachloride to give 8-chloro-6- (2,6-difluoropheayl) -4H-imidazo [1,2-a] [1,4-benzodiazepine.

Preparation of 8-nitro-6- (o-chlorophenyl-4H-imidazo [1,2-a] [1,4] benzodiazepine
In the same manner as described in Example 1, [[7-nitro-5- (o-chlorophenyl) -1, 2-dihydro-3H-1,4-benzodiazepin-2-ylidene] amino] acetaldehyde dimethylacetal in monoglyme heated together with titanium tetrachloride and 8-nitro-6- (o-chlorophenyl) -4H-imidazo [1, 2-a) [1,4] benzodiazepine is obtained.

Preparation of 10-isopropyl-4-methyl-6- (o methoxyphenyl) -4H-imidazo [1,2-a] [1,4] benzodiazepine
In the same manner as described in Example 1, [9-isopropyl-3-methyl-5- (o-methoxyphenyl) -1,2-di-hydro-3H-1,4-benzodiazepin-2-ylideneamino] acetaldehyde Dimethylacetal in diglyme is heated together with titanium tetrachloride to give 10-isopropyl-4-methyl-6- (o-methoxyphenyl) -4H-imidazo- [1,2-a] [1,4] benzodiazepine.

Preparation of 9-propylsulfonyl-2-methyl-6-phenyl 4H-imidazo [1,2-al [1,4] benzodiazepine
In the same manner as described in Example 1, [[8-propylsulfonyl-5-phenyl-1,2-dihydro-3H-1,4-benzodiazepin-2-ylidene] -a-amino] propionaldehyde diethyl acetal is combined in diglyme heated with titanium tetrachloride, giving 9-propylsulfonyl-2-methyl-5-phenyl) -4H imidazo [1,2-a] [1,4] benzodiazepine.

Preparation of 2-methyl-8-nitro-6- (o-chloro-phenyl) 4H-imidazo [1,2-a] [1,4] benzodiazepine

Preparation of 8-chloro-1-propyl-6- (o-chlorophenyl) -4H-imidazo [1, j-aj [, 4] benzodiazepine
In the same way as described in Example 1, [[7-chloro-5- (o-chlorophenyl) -1,2-dihydro-3H-1,4-benzodiazepin-2-ylidene] -a-amino] pentanaldehyde heated dimethylacetal in diglyme together with titanium tetrachloride and 8-chloro-1-propyl-6- (o-chlorophenyl) -4H-imidazo [1, ja] [1,4] benzodiazepine was obtained.
In the same way as in the previous examples, treatment of a certain [5-phenyl-1,2-dihydro-3H-1,4-benzodiazepin-2-ylidene [-c-amino] -2-aldehyde-dialkyl acetal or - ketone dialkyl ketals (I) with titanium tetrachloride in a suitable organic solvent, preferably at the temperature at which reflux takes place, a 6-phenyl-4H-imidazo [1,2-a] [1,4] benzodiazepine of the general formula II is obtained .

Treatment of compounds of formulas I and II with pharmacologically acceptable acids, such as e.g. Hydrochloric acid, hydrobromic acid, phosphoric acid, sulfuric acid, acetic acid, propionic acid, toluenesulfonic acid, methanesulfonic acid, tartaric acid, citric acid, lactic acid, malic acid, maleic acid or cyclohexanesulfamic acid, leads to pharmacologically acceptable salts of these compounds of the formula I and II. The salts can be used for the same areas of application like the free bases of the compounds of formula II.
