Die Erfindung betrifft ein Verfahren zur Herstellung von Carbamaten, wobei Alkohole in die entsprechenden Carbamoyl- oxyderivate überführt werden.
Die Carbamate sind wertvolle Derivate von Alkoholen, die für deren Identifikation und Kennzeichnung verwendbar sind. Ausserdem haben sich Carbamate, wie Meprobamat, Carbachol und Novobiocin, als wertvoll auf dem Gebiet der Medizin erwiesen.
Kürzlich wurde gefunden, dass 3-Carbamoyloxymethylcephalosporine, die durch Fermentation erhalten werden, wertvolle Antibiotica sind. Das erfindugsgemässe Verfahren ist von beson r derem Wert für die Herstellung solcher Cephalosporine sowie anderer Carbamate.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man einen Alkohol der allgemeinen Formel ROH, in der R den organischen Rest des Alkohols bedeutet, mit einem Isocyanat der Formel ,SiNCO, in der =Si einen Triorganosilylrest bedeutet, zu dem entsprechenden N-substituierten Carbamat der For mel ROCONHSi-- umsetzt und dieses Zwischenprodukt zu dem Carbamat hydrolysiert.
Das vorliegende erfindungsgemässe Verfahren kann durch die folgende Reaktionsgleichung veranschaulicht werden:

Die als SL- angeführte Gruppe ist eine Triorganosilylgruppe, die leicht abgespalten und durch Wasserstoff ersetzt werden kann.
Es können zwar eine Anzahl verschiedener Triorganosilylisocyanate verwendet werden, das bevorzugte Triorganosilylisocyanat ist jedoch N-Trimethylsilylisocyanat, da es am leichtesten erhält lichist.
In diesem Verfahren wird der Alkohol I, in dem R' den organischen Rest des Alkohols bedeutet, mit dem Silylisocyanat II zu dem N-substituierten Zwischenprodukt III umgesetzt, und dieses wird dann zu dem gewünschten Carbamat IV hydrolysiert.
Die erste Stufe des oben beschriebenen Verfahrens, nämlich die Herstellung des Imidodicarboxylats oder der N-Triorganosilylcarbamoyloxyverbindung III wird vorzugsweise durchgeführt, indem man den Alkohol innig mit dem Isocyanat, insbesondere in einem nicht-protischen Lösungsmittel, wie Methylenchlorid, Tetrahydrofuran, Dimethylformamid und dgl., in Kontakt bringt.
Im allgemeinen wird die Umsetzung vorzugsweise unter Wasserausschluss und unter Verwendung wenigstens einer äquimolaren Menge an dem Isocyanat durchgeführt, damit maximale Ausbeuten an dem gewünschten Zwischenprodukt erhalten werden. Die Umsetzung kann im allgemeinen bei Temperaturen zwischen etwa 0 und 30 C durchgeführt werden, wird im allgemeinen aber vorzugsweise bei etwa Raumtemperatur durchgeführt. Die bei diesem Verfahren anzuwendenden Bedingungen hängen teilweise von dem umzusetzenden Alkohol ab. Die Abtrennung der schützenden Gruppe und ihr Ersatz durch Wasserstoff lassen sich leicht durch Hydrolyse unter leicht sauren Bedingungen durchführen.
Das erfindungsgemässe Verfahren ist besonders wertvoll für die Herstellung von Cephalosporinen mit einem 3-Carbamoyloxymethylsubstituenten. Diese bevorzugte Durchführungsform des erfindungsgemässen Verfahrens kann durch die folgende Reaktionsgleichung veranschaulicht werden:

worin Si die oben angegebene Bedeutung hat, R" ein Acylrest ist, R3 Wasserstoff oder eine Gruppe wie eine Methoxygruppe ist, und R Wasserstoff oder ein blockierender Substituent ist. In diesem Verfahren wird z.B. das 3-Hydroxymethylcephalosporin V mit dem Isocyanat II zu der N-substituierten Carbamoyloxyver bindung VI umgesetzt, und diese wird dann zu dem 3-Carbamoyloxymethylester VII deblockiert. Dann wird die blockierende Carboxygruppe abgetrennt, so dass die freie Säure VIII erhalten wird.
Der Acylrest R" kann eine Acylgruppe einer Carbonsäure oder ein substituierter Sulfonylrest, wie Phenylsulfonyl, Äthylsulfonyl, Benzylsulfonyl, 2,5-Dimethylphenylsulfonyl, 4-Chlorphenylsulfonyl, 4-Methoxyphenylsulfonyl und dgl., sein. D.h. R" kann ein aliphatischer, aromatischer oder heterocyclischer, araliphatischer oder heterocyclylaliphatischer Carbonsäurerest, wie der Acylrest der bekannten Cephalosporine und Penicilline, sein.
Acylgruppen der allgemeinen Formel RIIRloCHCO, in der Rlo und R11 die weiter unten angegebene Bedeutung haben, stellen wegen ihrer antibiotischen Wirkung eine bevorzugte Gruppe von Substituenten dar. Rlo ist Wasserstoff, Halogen, Amino, Guanidino, Phosphono, Hydroxy, Tetrazolyl, Carboxy, Sulfo oder Sulfamino. R11 steht für Phenyl, einen monocyclischen heterocyclischen 5- oder 6-gliedrigen Ring, der ein oder mehrere Sauerstoff-, Schwefel- oder Stickstoffatome im Ring enthält, substituierte Heterocyclen, Phenylthio, heterocyclische oder substituierte heterocyclische Thiogruppen; oder Cyano. Die Substituenten können Halogen, Carboxymethyl, Guanidino, Guanidinomethyl, Carboxamidomethyl, Aminomethyl, Nitro, Methoxy oder Methyl sein.
Als Beispiele für diese bevorzugten Substituenten können erwähnt werden: Phenacetyl, 3-Bromphenylacetyl, p-Aminomethylphenylacetyl, 4-Carboxylmethylphenylacetyl, 4-Carboxamidomethylphenylacetyl, 2-Furylacetyl, 5-Nitrofurylacetyl, 3-Furylacetyl, 2 Thienylacetyl, 5-Chlorthienylacetyl, 5-Methoxythienylacetyl, a- Guanidino-2-thienylacetyl, 3-Thienylacetyl, 4-Methylthienylace- tyl, 3-lsothiazolylacetyl, 4-Methoxyisothiazolylacetyl, 4-lsothiazolylacetyl, 3-Methylisothiazolylacetyl, 5-lsothiazolylacetyl, 3 Chlorisothiazolylacetyl, 3-Methyl- 1 ,2,5-oxadiazolylacetyl, 1,2,5 Thiadiazolyl-4-acetyl, 3-Methyl-1,2,5-thiadiazolyl-4-acetyl, 3 Chlor-l ,2,5-thiadiazolyl-4-acetyl, 3-Methoxy- 1 ,2,5-thiadiazolyl-4acetyl,

Besonders bevorzugte 3-Hydroxymethylcephalosporine, die nach dem erfindungsgemässen Verfahren in die entsprechenden 3 Carbamoyloxymethylcephalosporine übergeführt werden können, sind diejenigen, in denen Rlo Wasserstoff, Amino oder Carboxy und R11 Phenyl oder ein 5-gliedriger heterocyclischer Ring mit einem Sauerstoff- oder Schwefelheteroatom ist, und insbesondere diejenigen, in denen R" Benzylcarbonyl, 2- und 3-Thienylmethylcarbonyl oder 2- und 3-Furylmethylcarbonyl ist.
Die Cephalosporincarbamate werden in der Regel hergestellt, indem man Cephalosporine mit einem 3-Hydroxymethylsubstituenten oder ein Salz oder einen Ester eines solchen Cephalosporins mit dem Isocyanat umsetzt und das Produkt dieser Umsetzung dann wie oben beschrieben spaltet. Wenn das umzusetzende Cephalosporin andere Substituenten, die mit dem Isocyanat reagieren, wie weitere Hydroxy- oder Aminogruppen, enthält, so werden diese Substituenten gewöhnlich durch Gruppen, wie Trityl, tertiär Butyloxycarbonyl, N-Trichlororäthoxycarbonyl und dgl., blockiert oder abgeschirmt und dann nach Einführen der Carbamoyloxygruppe entfernt.
Beispielsweise wird 7-Tritylaminocephalosporansäure innig mit Citrusacetylesterase in Kontakt gebracht, wobei 7-Tritylaminodesacetylcephalosporansäure gebildet wird, aus der durch Umsetzen mit Isocyanat und Hydrolyse des Reaktionsproduktes die 3-Carbamoyloxymethylverbindung erhalten wird. Durch Abtrennen der schützenden Trityl gruppe nach an sich bekannten Methoden kann die 3-carbamoyloxymethyl-7-aminodesacetylcephalosporansäure erhalten werden, die dann nach an sich bekannten Methoden zu 3-Carbamoyloxy methyldesacetyl cephalosporinen acyliert werden kann.
Alternativ können 3-Hydroxymethyl-7-acylamido-3-cephem-4carbonsäure und die entsprechenden Verbindungen mit einem Substituenten in der 7-Stellung, wie Methoxy, anstelle von Wasserstoffnach dem Verfahren gemäss der Erfindung in die entsprechenden 3-Carbamoyloxymethylcephalosporine übergeführt werden.
Die nach dem erfindungsgemässen Verfahren erhaltenen 3 Carbamoyloxymethylcephalosporine sind wertvolle Antibiotica, die in niedrigen Dosierungen gegen verschiedene gram-positive und gram-negative Pathogene, wie Staphylococcus aureus Staphylococcus pyogenes, Proteus vulgaris, Escherichia coli und dgl., wirksam sind. D.h. diese neuen Cephalosporine sind wertvoll für die Behandlung von Infektionen bei Menschen und Tieren. Sie können auch in verdünnten wässrigen Lösungen, die weniger als 100 ppm Antibioticum enthalten, zur Entfernung von darauf ansprechenden Organismen von pharmazeutischen, medizinischen und zahnmedizinischen Geräten und zur Isolierung von Mikroorganismen aus Gemischen von Mikroorganismen verwendet werden.
Bei einer bevorzugten Ausführungsform des oben beschriebenen Verfahrens wird die Carboxygruppe des Cephalosporins blockiert oder abgeschirmt, indem man ein geeignetes Derivat bildet, das anschliessend leicht gespalten werden kann, ohne dass der ss-Lactamring angegriffen wird. Im allgemeinen wird der Carboxysubstituent vorzugsweise durch Bildung eines geeigneten Esters blockiert. Als Beispiele für solche Ester können erwähnt werden die Benzyl-, Benzhydryl-, Methoxymethyl-, p Nitrophenyl-, Trimethylsilyl-, Trichloräthoxy-, p-Methoxybenzyl-, Phthalimidomethyl- und Succinimidomethylester. Diese Ester können nach an sich bekannten Methoden gespalten werden, so dass die freie Säure erhalten wird.
Die folgenden Beispiele veranschaulichen das oben beschriebene Verfahren gemäss der Erfindung.
Beispiel 1: 3-Carbamoyloxymethyl-7-methoxy-7ss- (2-thienylacetamido) -
3-cephem-4-carbonsäure.
Zu 10 ml einer Lösung von 650 mg Benzhydryl-3-hydroxyme thyl-7-methoxy-7ss-(2-thienylacetamido)-3-cephem-4-carboxylat in Methylenchlorid werden tropfenweise 115 mg N-Trimethylsilylisocyanat zugesetzt. Das Reaktionsgemisch wird 1 Stunde bei Raumtemperatur gerührt und dann auf Eis abgekühlt und mit verdünnter HCI auf pH 5 angesäuert. Die Schichten werden getrennt, und die organische Phase wird abgetrennt und über Magnesiumsulfat getrocknet. Durch Abtrennen des Lösungsmittels unter vermindertem Druck wird das Benzhydryl-3-carbamoyloxymethyl-7-methoxy-7 t3-(2-thienylacetamido)-3-cephem-4- carboxylat erhalten.
Eine kalte Lösung von 136 mg des Benzhydrylesters von 3 Carbamoyloxymethyl-7-methoxy-7ss-(2-thienylacetamido)-3- cephem-4-carbonsäure in 1,1 ml Anisol wird V2 Stunde mit 0,55 ml Trifluoressigsäure bei 0 C verrührt. Die flüchtigen Materialien werden im Hochvakuum abgetrennt, und das Produkt wird aus Äthylacetat umkristallisiert; F. 165 bis 167"C.
Nach den oben beschriebenen Verfahren werden Benzhydryl
3-hydroxymethyl-7-methoxy-7 ss-acetamido-3-cephem-4-carboxylat und Benzhydryl-3-hydroxymethyl-7-methoxy-7ss-(2-furylacet- amido)-3-cephem-4--carboxylat in 3-Carbamoyloxymethyl-7 methoxy-7ss-acetamido-3-cephem4-carbonsäure bzw. 3-Carba moyloxymethyl-7-methoxy-7 p-(2-furvlacetamido)-3-cephem-4 carbonsäure (F 156 bis 161ob) übergeführt.
Die Ausgangsmaterialien für das obige Beispiel werden hergestellt, indem man die Benzhydrylester von 7-Methoxy-7ss-(2- thienylacetamido)-, -acetamido- oder -(2-furylacetamido)-cephalosporanat nach an sich bekannten Methoden mit Citrusacetylesterase bebrütet.
Die Benzhydrylester von 7-Methoxy-7ss-(2-thienylacetamido)- cephalosporansäure, 7-Methoxy-7ss-acetamidocephalosporansäure und 7-Methoxy-7ss-(2-furylacetamido)-cephalosporans ure werden hergestellt, wie in der USA-Patentanmeldung Serial Nr. 149364 vom 2. Juni 1971 beschrieben.
Beispiel 2: 3-Carbamoyloxymethyl-7ss- (2-thienylacetamido)3-cephem 4-carbonsäure.
Nach den Verfahren von Beispiel 1 wird eine äquivalente Menge an Methoxymethyl-3-hydroxymethyl-7ss-(2-thienylaceta- mido)-3-cephem-4-carbonsäure mit N-Trimethylsilylisocyanat zu Methoxymethyl-3-carbamoyloxymethyl-7 ss-(2-thienylacetamido)- 3-cephem-4-carboxylat umgesetzt.
Eine Suspension von 920 mg des Methoxymethylesters in 9,2 ml Methanol, das HCI in einer Konzentration von 10% enthält, wird 90 Minuten bei Raumtemperatur gerührt. Dann wird eine kalte Lösung von 1,65 g Natriumbicarbonat in 40 ml Wasser zugesetzt, und die klare Lösung wird bei Raumtemperatur unter vermindertem Druck auf ein Volumen von 30 bis 35 ml eingeengt. Dann wird die wässrige Lösung mit Äthylacetat extrahiert, und die organische Schicht wird verworfen. Die wässrige Schicht wird auf 0 bis 5O C gekühlt, mit 40 1 Äthylacetat bedeckt, und das pH wird mit kalter 10%der Salzsäure unter Rühren auf 1,8 eingestellt. Die Schichten werden getrennt, und die Äthylacetatschicht wird mit Eiswasser gewaschen.
Die wässrige Schicht und die Waschflüssigkeiten werden vereinigt und erneut zweimal mit je 20 ml Äthylacetat extrahiert. Der zweite Äthylacetatextrakt wird zweimal mit kaltem Wasser gewaschen und dann mit dem ersten Extrakt vereinigt. Die Lösung wird zur Trockne eingedampft und bis zur Gewichtskonstanz getrocknet, wobei die 3 Carbamoyloxymethyl-7ss-(2-thienylacetamido)-3-cephem-4- carbonsäure erhalten wird. Das Ausgangsmaterial für dieses Beispiel wird hergestellt, indem man den Methoxymethylester von 7ss-(2-Thienylacetamido)-cephalosporansäure nach an sich bekannten Methoden mit Citrusacetylesterase bebrütet. Der Methoxymethylester wird hergestellt, indem man das Di-(cyclohexyl)aminsalz von 7ss-(2-Thienylacetamido)-cephalosporansäure mit Chlormethylmethyläther in Lösung in wasserfreiem Methylenchlorid verestert.
Das gewünschte Produkt wird durch Abdampfen des Lösungsmittels erhalten.
In dem erfindugsgemässen Verfahren kann anstelle des Esters des Cephalosporins auch die freie Säure oder ein Salz davon verwendet werden. Gewöhnlich wird jedoch vorzugsweise der Ester verwendet, da mit diesem unter optimalen Bedingungen maximale Ausbeuten an dem gewünschten Produkt erhalten werden.

The 3 carbamoyloxymethylcephalosporins obtained by the process according to the invention are valuable antibiotics which, in low doses, are effective against various gram-positive and gram-negative pathogens such as Staphylococcus aureus, Staphylococcus pyogenes, Proteus vulgaris, Escherichia coli and the like. They can also be used in dilute aqueous solutions containing less than 100 ppm antibiotic to remove responsive organisms from pharmaceutical, medical and dental devices and to isolate microorganisms from mixtures of microorganisms.
In a preferred embodiment of the method described above, the carboxy group of the cephalosporin is blocked or shielded by forming a suitable derivative which can then easily be cleaved without the β-lactam ring being attacked. In general, the carboxy substituent is preferably blocked by formation of an appropriate ester. Examples of such esters include the benzyl, benzhydryl, methoxymethyl, p-nitrophenyl, trimethylsilyl, trichloroethoxy, p-methoxybenzyl, phthalimidomethyl and succinimidomethyl esters. These esters can be cleaved by methods known per se, so that the free acid is obtained.
The following examples illustrate the above-described method according to the invention.
Example 1: 3-Carbamoyloxymethyl-7-methoxy-7ss- (2-thienylacetamido) -
3-cephem-4-carboxylic acid.
115 mg of N-trimethylsilyl isocyanate are added dropwise to 10 ml of a solution of 650 mg of benzhydryl-3-hydroxymethyl-7-methoxy-7ss- (2-thienylacetamido) -3-cephem-4-carboxylate in methylene chloride. The reaction mixture is stirred for 1 hour at room temperature and then cooled on ice and acidified to pH 5 with dilute HCl. The layers are separated and the organic phase is separated and dried over magnesium sulfate. The benzhydryl-3-carbamoyloxymethyl-7-methoxy-7-3- (2-thienylacetamido) -3-cephem-4-carboxylate is obtained by separating off the solvent under reduced pressure.
A cold solution of 136 mg of the benzhydryl ester of 3-carbamoyloxymethyl-7-methoxy-7ss- (2-thienylacetamido) -3-cephem-4-carboxylic acid in 1.1 ml of anisole is stirred with 0.55 ml of trifluoroacetic acid at 0 ° C. for V2 hour . The volatile materials are separated in a high vacuum and the product is recrystallized from ethyl acetate; F. 165 to 167 "C.
Following the procedures described above, benzhydryl
3-hydroxymethyl-7-methoxy-7ss-acetamido-3-cephem-4-carboxylate and benzhydryl-3-hydroxymethyl-7-methoxy-7ss- (2-furylacet-amido) -3-cephem-4-carboxylate in 3-Carbamoyloxymethyl-7-methoxy-7ss-acetamido-3-cephem-4-carboxylic acid or 3-Carba moyloxymethyl-7-methoxy-7-p- (2-furvlacetamido) -3-cephem-4-carboxylic acid (F 156 to 161ob) transferred.
The starting materials for the above example are prepared by incubating the benzhydryl esters of 7-methoxy-7ss- (2-thienylacetamido), -acetamido- or - (2-furylacetamido) -cephalosporanate with citrus acetylesterase according to methods known per se.
The benzhydryl esters of 7-methoxy-7ss- (2-thienylacetamido) -cephalosporanic acid, 7-methoxy-7ss-acetamidocephalosporanic acid, and 7-methoxy-7ss- (2-furylacetamido) -cephalosporanic acid are prepared as described in U.S. Patent Application Serial No. 149364 of June 2, 1971.
Example 2: 3-Carbamoyloxymethyl-7ss- (2-thienylacetamido) 3-cephem 4-carboxylic acid.
Following the procedure of Example 1, an equivalent amount of methoxymethyl-3-hydroxymethyl-7ss- (2-thienylacetamido) -3-cephem-4-carboxylic acid is added with N-trimethylsilyl isocyanate to methoxymethyl-3-carbamoyloxymethyl-7ss- (2 -thienylacetamido) - 3-cephem-4-carboxylate implemented.
A suspension of 920 mg of the methoxymethyl ester in 9.2 ml of methanol containing HCl in a concentration of 10% is stirred for 90 minutes at room temperature. A cold solution of 1.65 g of sodium bicarbonate in 40 ml of water is then added and the clear solution is concentrated to a volume of 30 to 35 ml at room temperature under reduced pressure. Then the aqueous solution is extracted with ethyl acetate and the organic layer is discarded. The aqueous layer is cooled to 0-5O C, covered with 40 liters of ethyl acetate, and the pH is adjusted to 1.8 with cold 10% hydrochloric acid with stirring. The layers are separated and the ethyl acetate layer is washed with ice water.
The aqueous layer and the washing liquids are combined and extracted again twice with 20 ml of ethyl acetate each time. The second ethyl acetate extract is washed twice with cold water and then combined with the first extract. The solution is evaporated to dryness and dried to constant weight, the 3-carbamoyloxymethyl-7ss- (2-thienylacetamido) -3-cephem-4-carboxylic acid being obtained. The starting material for this example is prepared by incubating the methoxymethyl ester of 7ss- (2-thienylacetamido) -cephalosporanic acid with citrus acetylesterase according to methods known per se. The methoxymethyl ester is prepared by esterifying the di- (cyclohexyl) amine salt of 7ss- (2-thienylacetamido) -cephalosporanic acid with chloromethyl methyl ether in solution in anhydrous methylene chloride.
The desired product is obtained by evaporating the solvent.
In the process according to the invention, the free acid or a salt thereof can also be used instead of the ester of cephalosporin. Usually, however, it is preferred to use the ester, since with it maximum yields of the desired product are obtained under optimal conditions.
