Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen heterocyclischen Carbonsäuren.

X Sauerstoff, Schwefel, die Iminogruppe oder die Methyliminogruppe.
Y Wasserstoff. Halogen bis Atomnummer 35 oder die Methylgruppe und
Z1 sowie Z2 Wasserstoff, Halogen bis Atomnummer 35, eine niedere Alkyl- oder Alkoxygruppe bedeuten, sowie ihre Salze mit anorganischen oder organischen Basen sind bisher nicht bekanntgeworden.
Wie nun gefunden wurde, besitzen die neuen Verbindungen wertvolle pharmakologische Eigenschaften. Sie zeigen diuretische und saluretische Wirkung. Diese Eigenschaften kennzeichnen diese Verbindungen als geeignet zur Behandlung von Störungen, welche durch mangelhafte Ausscheidung von Elektrolyten, insbesondere von Natriumchlorid, bedingt sind. Solche Störungen sind die Ursache von Ödemen und Hypertonien. Diese Verbindungen, z. B. die 4-Chlor-5-(2methylen-butyryl)-indol-2-carbonsäure, die 6-Methoxy-5 (2-methylen-butyryl)-benzofuran-2-carbonsäure und die 6-Methyl-5-(2-methylen-butyryl)-benzofuran-2-carbonsäure vermögen am Hund und am Kaninchen bei einer Dosierung von 10 mg/kg p.o. die Harnausscheidung sowie die Ausscheidung der Natriumionen und der Chlorionen beträchtlich zu erhöhen.
In den heterocyclischen Carbonsäuren der Formel I nimmt Z1 die 4- oder 6-Stellung und Z2 die 6- oder 7-Stellung ein.
R, Zl und Z2 bedeuten als niedere Alkylgruppen beispielsweise die Methyl-, Äthyl-, Propyl-, Isopropyl-, Butyl- oder die tert.-Butylgruppe und Z1 sowie Z2 als niedere Alkoxygruppen beispielsweise die Methoxy-, Äthoxy-, Propoxy-, Isopropoxy-, Butoxy- oder die sek.-Butoxygruppe.
Nach dem erfindungsgemässen Verfahren stellt man die Verbindungen der Formel I her, indem man von einer Verbindung der Formel II,

in welcher R1 eine niedere Alkylgruppe oder die Benzylgruppe bedeutet und R, X, Y, Z1 und Z2 die unter der allgemeinen Formel I angegebene Bedeutung haben, und Am den Rest einer sekundären organischen Base bedeutet, gleichzeitig die Estergruppe und die sekundäre Aminogruppe abspaltet und gewünschtenfalls das erhaltene Reaktionsprodukt mit einer anorganischen oder organischen Base in ein Salz überführt.
Diese Abspaltung kann beispielsweise durch kurzes Aufkochen in wässrigalkanolischer Lösung in Gegenwart einer anorganischen Base wie z. B. einem Alkali- oder Erdalkalimetallhydroxid oder -carbonat vorgenommen werden.
Zu den als Ausgangsstoffe für dieses Verfahren benötigten Verbindungen der Formel II gelangt man beispielsweise, indem man eine Carbonsäure entsprechend der Formel II in an sich bekannter Weise unter milden Bedingungen, bei welchen die sekundäre Aminogruppe nicht abgespaltet wird, mit einem niederen Alkanol oder mit Benzylalkohol verestert.
Man kann sie ebenfalls herstellen. indem man von einem niederen Alkylester oder dem Benzylester einer Verbindung der Formel III,

in welcher X, Y, Z1 und Z2 die unter Formel I angegebene Bedeutung haben, ausgeht, und diesen gemäss Friedel-Crafts mit einer Verbindung der Formel IIIa,
R-CH2-COCl (IIIa) in welcher R die unter Formel I angegebene Bedeutung hat, kondensiert. Der so erhaltene 5-Alkanoylester wird dann mit Hilfe von Paraformaldehyd und einem sekundären Amin in das entsprechende Mannichderivat, entsprechend der Formel II übergeführt.
Die neuen Wirkstoffe oder die pharmazeutisch annehmbaren Salze derselben werden vorzugsweise peroral verabreicht. Zur Salzbildung können anorganische oder organische Basen, wie beispielsweise Alkali- oder Erdalkalihydroxide, Carbonate oder Bicarbonate, Triäthanolamin oder Cholin, verwendet werden. Die täglichen Dosen bewegen sich zwischen 50 und 1000 mg für erwachsene Patienten. Geeignete Doseneinheitsformen, wie Dragees. Tabletten, enthalten vorzugsweise 25-500 mg eines erfindungsgemässen Wirkstoffes, und zwar 20-80% einer Verbindung der Formel I.
Das nachfolgende Beispiel erläutert die Herstellung der neuen Verbindungen der Formel I näher. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel a) Man schmilzt 7,2 g 3,6-Dimethyl-5-butyryl-benzofuran-2-carbonsäure-methylester, 3,3 g Dimethylamin-hydrochlorid und rührt das Ganze während 2 Stunden bei 140".
Die dabei erhaltene Schmelze besteht aus dem rohen 3,6 Dimethyl-5 -(2-dimethylamino-methyl-butyryl)-benzofuran2-carbonsäure-methylester-hydrochlorid, wird mit einer Lösung von 80 ml in Natronlauge und 80 ml Äthanol 10 Minuten unter Rückfluss gekocht, dann mit 300 ml Eiswasser verdünnt und mit konz. Salzsäure auf pH 3 angesäuert. Der ausgefallene Niederschlag wird abgenutscht und nach Trocknen im Vakuum durch Chromatographieren über Silicagel gereinigt. Die aus Benzol-Heptan umkristallisierte 3,6-Dimethyl-5 -(2-methyl-butyryl)-benzofuran-2-carbonsäure schmilzt bei 152-154".
b) Der als Ausgangsmaterial verwendete 3,6-Dimethyl5-butyryl-benzofuran-2-carbonsäure-methylester wird wie folgt hergestellt:
10,4 g 3,6-Dimethyl-5-butyryl-benzofuran-2-carbonsäure werden portionenweise während 30 Minuten in eine kochende Aufschlämmung von 7 g Kaliumcarbonat in 80 ml Aceton eingetragen. Gleichzeitig wird aus einem Tropftrichter eine Lösung von 5 ml Dimethyl-sulfat in 30 ml Aceton zum Reaktionsgemisch getropft. Nach der Zugabe wird das Gemisch noch weitere 3 Stunden unter Rückfluss gerührt, dann erkalten gelassen, der Niederschlag abgenutscht und mit 50 ml Aceton ausgewaschen, Filtrat und Waschflüssigkeit vereinigt eingedampft und der Rückstand aus Tetrachlorkohlenstoff Heptan umkristallisiert. Der entstandene 3,6-Dimethyl-5 butyryl-benzofuran-2-carbonsäure-methylester schmilzt bei 101-105 .
c) Die 3,6-Dimethyl-5 -butyryl-benzofuran-2-carbonsäure wird aus 3,6-Dimethyl-benzofuran-2-carbonsäure (vgl. Fries und Finkewirth, Chem. Ann. 362 [1908] 50) mit Buttersäurechlorid und Aluminiumchlorid in Nitrobenzol erhalten.
Die Verbindung schmilzt bei 185-187 (aus Benzol-Essigester).

The present invention relates to a process for the preparation of new heterocyclic carboxylic acids.

As has now been found, the new compounds have valuable pharmacological properties. They show diuretic and saluretic effects. These properties characterize these compounds as being suitable for the treatment of disorders which are caused by inadequate excretion of electrolytes, in particular sodium chloride. Such disorders are the cause of edema and hypertension. These compounds, e.g. B. 4-chloro-5- (2methylene-butyryl) -indole-2-carboxylic acid, 6-methoxy-5 (2-methylen-butyryl) -benzofuran-2-carboxylic acid and 6-methyl-5- (2 -methylene-butyryl) -benzofuran-2-carboxylic acid can be used in dogs and rabbits at a dose of 10 mg / kg po to considerably increase the excretion of urine and the excretion of sodium ions and chlorine ions.

R-CH2-COCl (IIIa) in which R has the meaning given under formula I, condenses. The 5-alkanoyl ester obtained in this way is then converted into the corresponding Mannich derivative, corresponding to formula II, with the aid of paraformaldehyde and a secondary amine.
The new active ingredients or the pharmaceutically acceptable salts thereof are preferably administered orally. Inorganic or organic bases, such as, for example, alkali or alkaline earth metal hydroxides, carbonates or bicarbonates, triethanolamine or choline, can be used for salt formation. The daily doses range between 50 and 1000 mg for adult patients. Suitable unit dosage forms, such as dragees. Tablets preferably contain 25-500 mg of an active ingredient according to the invention, namely 20-80% of a compound of formula I.
The following example explains the preparation of the new compounds of the formula I in more detail. The temperatures are given in degrees Celsius.
Example a) 7.2 g of 3,6-dimethyl-5-butyryl-benzofuran-2-carboxylic acid methyl ester and 3.3 g of dimethylamine hydrochloride are melted and the whole is stirred for 2 hours at 140 ".
The melt obtained consists of the crude 3,6-dimethyl-5 - (2-dimethylamino-methyl-butyryl) -benzofuran2-carboxylic acid methyl ester hydrochloride, is refluxed with a solution of 80 ml in sodium hydroxide and 80 ml of ethanol for 10 minutes boiled, then diluted with 300 ml of ice water and treated with conc. Hydrochloric acid acidified to pH 3. The deposited precipitate is filtered off with suction and, after drying in vacuo, purified by chromatography over silica gel. The 3,6-dimethyl-5 - (2-methyl-butyryl) -benzofuran-2-carboxylic acid recrystallized from benzene-heptane melts at 152-154 ".
b) The methyl 3,6-dimethyl5-butyryl-benzofuran-2-carboxylate used as starting material is prepared as follows:
10.4 g of 3,6-dimethyl-5-butyryl-benzofuran-2-carboxylic acid are added in portions over 30 minutes to a boiling slurry of 7 g of potassium carbonate in 80 ml of acetone. At the same time, a solution of 5 ml of dimethyl sulfate in 30 ml of acetone is added dropwise to the reaction mixture from a dropping funnel. After the addition, the mixture is stirred under reflux for a further 3 hours, then allowed to cool, the precipitate is suction filtered and washed with 50 ml of acetone, the filtrate and washing liquid are combined, evaporated and the residue is recrystallized from carbon tetrachloride heptane. The resulting 3,6-dimethyl-5-butyryl-benzofuran-2-carboxylic acid methyl ester melts at 101-105.
c) The 3,6-dimethyl-5-butyryl-benzofuran-2-carboxylic acid is obtained from 3,6-dimethyl-benzofuran-2-carboxylic acid (cf. Fries and Finkewirth, Chem. Ann. 362 [1908] 50) with butyric acid chloride and aluminum chloride in nitrobenzene.
The compound melts at 185-187 (from benzene-ethyl acetate).
