Bekanntlich erzielt man durch orale Verabreichung von verschiedenen 1,2,3,4-Tetrahydro-4-oxochinazolinen hervorragende ,diuretische Wirkungen, ohne dass diese Verbindungen toxisch wären. 1,2,3,4-Tetrahydro-4-oxo- chinazoline wurden beispielsweise dadurch erhalten, dass man die entsprechenden 3,4-Dihydro-4-oxochintazoline mit Natriumborhydrid und Aluminiumchlorid reduzierte.
Dieses Verfahren ist aber im allgemeinen kompliziert und gibt lediglich schlechte Ausbeuten an erwünschten Produkten.
Es wurde nun ein verbessertes Verfahren für die Her- stellung dieser wertvollen 1,2,3,4-Tetrahydro-4-oxochin- azoline entwickelt Gemäss dem vorliegenden Verfahren lassen sich 1,2,3,4-Tetrahydro-4-oxochinazoline der Formel I in hoher Ausbeute und in einfacher Weise dadurch herstellen, dass man eine Anthranilsäure der Formel

Gemäss diesem neuen Verfahren kann man nicht nur bekannte 4-Oxochinazoline erhalten, sondern ebenfalls neue 4-Oxochinazoline der Formel I.
Die vorliegende Erfindung bezieht sich daher auf ein neues und verbessertes Verfahren zur Herstellung von 1,2,3,4-Tetrahydro-4-oxochinazolinen der Formel I, welche, wie erwähnt, eine hervorragende dinretische Wirkung ausüben.
Bei den obigen Definitionen kommen als Halogen- atome R1, R4 oder R5 tbzw. als Halogenatome in den Halogenalkyl- oder Dihalogenalkylresten insbesondere Fluoratome, Chloratome oder Bromatome in Frage. Die Anzahl der Kohlenstoffatome in der Alkylgruppe der besagten Alkylacetale ist nicht beschränkt; vorzugsweise wird man aber Alkylacetale mit 1 bis 3 Kohlenstoffatomen in der Alkylgruppe verwenden.
Beim Arbeiten gemäss dem vorliegenden Verfahren wird man ein Anthranilanilid der Formel IV vorzugs- weise mit einer äquimolaren Menge eines Aldehyds der Formel V oder eines Alkylacetals davon in Berührung bringen. Die Umsetzung kann im emem Lösungsmittel, wie z.B. Essigsäure, Methanol, Äthanol, Propanol, Te trahydrofuran, Dioxan, Benzol oder Toluol, und vorzugsweise in Gegenwart einer Säure, wie z.B. Schwefelsäure, Salzsäure oder dergleichen, durchgeführt wenden.
Die Reaktion verläuft glatt bei einer Temperatur im Bereiche von 30 bis 1 100C und vorzugsweise von 60 bis 800C.
Nach beendeter Umsetzung wird das Reaktionsgemisch in an sich üblicher Weise einer Nachbehandlung unterworfen. So kann man beispielsweise das Reaktionsgemisch in Eiswasser giessen und den Niederschlag, welcher aus dem gewünschten Chinazolinderivat besteht, durch Filtrieren sammeln.
Gemäss vorliegendem Verfahren erhält man beispielsweise die folgenden 1,2,3,4-Tetrahydro-4-oxochinazoline: 2-Phenyl-3-(o-tolyl)-6-sulfamoyl-7-chlor-1,2,3,4-tetra lhydro-4-oxochinrazolin 2-Benzyl-3 -fo -tolyl) -6ulfamoyl-7-chlor- 1,2,3 ,4-tetra- hydro-4-oxochinazolin 2-Äthyl-3-(o-tolyl)-6-sulfamoyl-7-chlor-1,2,3,4-tetra hydro-4-oxochinazolin 2-Propyl-3-(o-tolyl)-6-sulfamoyl-7-chlor-1,2,3,4-tetrahydro -4-oxochinazolin 3-(o-Tolyl)-6-sulfamoyl-7-chlor-1,2,3,4-tetrahydro-4 -oxochinazolin 2-Äthyl-3-(o-chlorphenyl)-6-sulfamoyl-7-chlor-1,2,3,4 -tetrahydro-4- oxochinazolin 2-Chlormethyl-3 - (o-chlorphenyl)-6-sulfamoyl-7-chlor- -1,2,3,4-tetrahydro-4-oxochinazolin

in denen das Symbol Ra. in der Formel I ein 1 bis 3 Kohlenstoffatome enthaltendes Halogenalkyl oder eine durch ein 3 bis 7 Kohlenstoffatome enthaltendes Cycloalkyl substituierte Methylgruppe bedeutet, handelt es sich bei den nach dem vorliegenden Verfahren erhaltenen 1,2,3,4-Tetrahydro-4-oxochinazoli nen um neue Verbindungen, weiche ausgezeichnete diu- retische Wirkungen haben.
Man kann pharmazeutische Präparate herstellen, indem man die neuen l,2,3,4-Tetrahydro-4-oxochinazoline mit einem pharmazeutisch annehmbaren Trägermittel vermischt, wobei man die Wirksubstanz in einer pharmazeutisch wirksamen Menge anwendet.
Die erste Reaktionsstufe des Verfahrens besteht aber darin, dass man eine Anthranilsäure der Formel II vorzugsweise mit einer äquimolaren Menge eines Anilins der Formel III in Gegenwart von Phosphortrichlorid behang delt.
Bei dieser Umsetzung ist es angezeigt, einen Über- schuss der Anthranilsäure zu verwenden. Die zu verwen- denke Menge an Phosphortrichlorid ist nicht von Bedeutung, doch wird man vorzugsweise 0,5 Mol oder mehr und vorzugsweise ungefähr 1 Mol Phosphortrichlorid pro Mol Anilin verwenden. Dlie Umsetzung kann glatt in einem geeigneten Lösungsmittel, wie z.B. Benzol, Tolnol, Xylol oder dergleichen durchgeführt werden.
Bei dieser Reaktion wird das Anilin der Formel III praktisch in die entsprechende, aktivierte Anilinphosphortrichlorid Komplexverbindung übergeführt, welche man leicht ohne Isolierung mit der Anthranilsäure der Formel II bei einer Temperatur von 70 bis 1400C umsetzen kann.
Nach beendeter Umsetzung wird in an sich üblicher Weise aufgearbeitet, um die gewünschten Produkte zu erhalten. So kann man beispielsweise das Reaktionsgemisch in Eiswasser giessen und den entstandenen Niederschlag durch Filtrieren sammeln und mit einer verdünn- ten wässrigen Natriumbicarbonatlösung waschen.
So erhält man beispielsweise die folgenden Anthranilanilide: 4-Chlor-5-sulfamoylanthranil-p-methylanilid 4-Chlor-5-sulfamoylanthranil-m-methylanilid 4-Chlor-5-culfamoylanthraniul-m-methoxyanilid 4-Chlor-5-sulfamoylanthranil-m-chloranilid 4-Fluor-5-sulfamoylanthranilanilid 4-Fluor-5-methylsulfamoylanthranil-o-methylanilid 4-Chlor-5-äthylsulfamoylanthranil-o-methylanilid 4-Brom-5-sulfamoylanthranil-o-methylanilid 4-Chlor-5-methylsulfamoylanthranil-o-trifluormethylanilid 4-Chlor-5-sulfamoylanthranil-o-äthylanilid 4-Brom-5-butylsulfamoylanthranil-o-chloranilid.
Die Erfindung wird durch die folgenden Beispiele erläutert.
Beispiel 1
Stufe 1
Eine Mischung von 2,5 g o-Toluidin und 0,7 g Phosphortrichlorid in Toluol wird unter Rückfluss während 2 ,Stunden n gerührt. Dann wird das Gemisch mit 2 g 4 -Chlor-5-sulfamoylanthranilsäure versetzt, worauf man das Reaktionsgemisch während 4 Stunden unter Rühren auf 1300C erhitzt. Nach Abkühlen wird das Gemisch in Wasser gegossen, worauf der erzeugte Niederchlag durch Filtrieren gesammelt und mit wässiger Natriumcarbonatlösung gewaschen wird. Der rohe Niederchlag wird aus Äthanol umkristallisiert, wobei man 2 g 4-Chlor-5-sulfa- moylanthranil-o-methylanilid. Smp. 289 bis 290 C, erhält.
Stufe 2
Eine Mischung von 1,7 g 4-Chlor-5-sulfamoylanthranil-o-methylanilid, erhalten gemäss der obigen Stufe 1, und 0,05 cm2 konzentrierter Schwefelsäure in 20 cm3 Essigsäure wird auf 70 C erwärmt. Dann wird das Gemisch mit 1 g Cycloptylacetaldehyd-diäthylacetal versetzt und das erhaltene Gemisch während 30 Minuten auf 700C erhitzt, wobei man eine klare gelbe Lösung erhält.
Nach Abkühlen wird die Lösung in 100 cm3 Wasser gegossen und der erzeugte Niederschlag durch Filtrieren gesam- met und mit Wasser gewaschen. Durch Umkristallisieren aus einer Mischung von Äthylacetat und Benzol l(Mi- schungsverhältnis 2:1) erhält man 1,8 g 2-Cyclopentylmethyl-3-(o-tolyl)-6-sulfamoyl-7-chlor-1,2,3,4-tetrahydro -4-oxochinazolin, Smp. 156 bis 1570C.
Beispiel 2
Eine Mischung von 1,1 g 4-Chlor-5-sulfamoylanthranil-o-methylanilid, erhalten in ähnilicher Weise wie in der Stufe 1 des Beispiels 1, und 0,05 cm3 konzentrierter Schwefelsäure in 150 cm3 Essigsäure wird auf 700C erhitzt, worauf man mit 0,55 g CbloracetalfdiehySdiäthyÅal- acetal versetzt. Das erhaltene Gemisch wird während 30 Minuten auf 700C weiter erwärmt. Nach Abkühlen wird die Lösung in Wasser gegossen und Ider erzeugte Niederschlag durch Filtrieren gesammelt und mit Wasser gewaschen. Durch Umkristallisieren aus Benzol erhält man 0,8 g 2-Chlormethyl-3-(o-tolyl)-6-sulfamoyl-7-chlor- 1,2,3,4-tetrahydro-4-oxochinazolin.
Smp. 170 bis 174 C
Beispiel 3
Eine Mischung von 4,9 g 4-CElor-5-sulfamoylanthra- nil-o-methylanilid, erhalten in ähnlicher Weise wie in Stufe 1 des Beispiels 1, und 2,1 g Acvetaldlehydldiiäthyl- acetal in 50 cm3 Methoxyäthanol wird auf 1000C erhitzt.
Dann wird eine Lösung von 0,5 cm3 konzentrierte Schwefelsäure in 5 cm3 Methoxyäthanol zum Gemisch hinzu gegeben und das Reaktionsgemisch während' 30 Minuten auf 100 C erhitzt. Nach Abkühlen wird das Gemisch in Wasser gegossen und der erzeugte Niederschlag durch Filtrieren gesammelt und mit Wasser gewaschen. Durch Umkristallisieren aus Äthanol erhält man 4 g 2-Methyl -3-(o-tolyl)-6-sulfamoyl-chlor-1,2,3,4-tetrahydro-4-oxo chinazolin, Smp. 253 bis 254,5 C.
Arbeitet man in ähnlicher Weise, wie in Stufe 1 von Beispiel 1 beschrieben worden ist, so gelangt man zu den folgenden Verbindungen.

The present invention therefore relates to a new and improved process for the preparation of 1,2,3,4-tetrahydro-4-oxoquinazolines of the formula I which, as mentioned, have an excellent dinretic effect.
In the above definitions, the halogen atoms R1, R4 or R5 t or. as halogen atoms in the haloalkyl or dihaloalkyl radicals, in particular fluorine atoms, chlorine atoms or bromine atoms in question. The number of carbon atoms in the alkyl group of said alkyl acetals is not limited; however, preference will be given to using alkyl acetals having 1 to 3 carbon atoms in the alkyl group.
When working in accordance with the present process, an anthranilanilide of the formula IV will preferably be brought into contact with an equimolar amount of an aldehyde of the formula V or an alkyl acetal thereof. The reaction can be carried out in a solvent, e.g. Acetic acid, methanol, ethanol, propanol, tetrahydrofuran, dioxane, benzene or toluene, and preferably in the presence of an acid such as e.g. Sulfuric acid, hydrochloric acid or the like.
The reaction proceeds smoothly at a temperature ranging from 30 to 110 ° C, and preferably from 60 to 80 ° C.

step 1
A mixture of 2.5 g of o-toluidine and 0.7 g of phosphorus trichloride in toluene is stirred under reflux for 2 hours. Then 2 g of 4-chloro-5-sulfamoylanthranilic acid are added to the mixture, whereupon the reaction mixture is heated to 130 ° C. for 4 hours while stirring. After cooling, the mixture is poured into water and the precipitate produced is collected by filtration and washed with aqueous sodium carbonate solution. The crude precipitate is recrystallized from ethanol, 2 g of 4-chloro-5-sulfamoyanthranil-o-methylanilide being added. M.p. 289 to 290 ° C.

A mixture of 1.7 g of 4-chloro-5-sulfamoylanthranil-o-methylanilide, obtained in step 1 above, and 0.05 cm2 of concentrated sulfuric acid in 20 cm3 of acetic acid is heated to 70.degree. Then 1 g of cycloptylacetaldehyde diethyl acetal is added to the mixture and the mixture obtained is heated to 70 ° C. for 30 minutes, a clear yellow solution being obtained.
After cooling, the solution is poured into 100 cm3 of water and the precipitate produced is collected by filtration and washed with water. Recrystallization from a mixture of ethyl acetate and benzene I (mixing ratio 2: 1) gives 1.8 g of 2-cyclopentylmethyl-3- (o-tolyl) -6-sulfamoyl-7-chloro-1,2,3, 4-tetrahydro -4-oxoquinazoline, m.p. 156 to 1570C.
Example 2
A mixture of 1.1 g of 4-chloro-5-sulfamoylanthranil-o-methylanilide, obtained in a manner similar to that in step 1 of Example 1, and 0.05 cm3 of concentrated sulfuric acid in 150 cm3 of acetic acid is heated to 70.degree with 0.55 g of CbloracetalfdiehySdiäthyÅal- acetal added. The mixture obtained is heated further to 70 ° C. for 30 minutes. After cooling, the solution is poured into water and the precipitate formed is collected by filtration and washed with water. Recrystallization from benzene gives 0.8 g of 2-chloromethyl-3- (o-tolyl) -6-sulfamoyl-7-chloro-1,2,3,4-tetrahydro-4-oxoquinazoline.

Example 3
A mixture of 4.9 g of 4-CElor-5-sulfamoylanthranil-o-methylanilide, obtained in a manner similar to that in Step 1 of Example 1, and 2.1 g of Acvetaldlehydldiiäthyl- acetal in 50 cm3 of methoxyethanol is heated to 1000C.
Then a solution of 0.5 cm3 of concentrated sulfuric acid in 5 cm3 of methoxyethanol is added to the mixture and the reaction mixture is heated to 100 ° C. for 30 minutes. After cooling, the mixture is poured into water and the generated precipitate is collected by filtration and washed with water.
