Verfahren zur Herstellung von neuen Aryloxy-essigsäurederivaten
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Aryloxy-essigsäurederivaten.
Verbindungen der allgemeinen Formel I,

in welcher R1 eine Alkylgruppe mit höchstens 10 Kohlenstoff atomen, R2 Wasserstoff oder die Methylgruppe, Rs Wasserstoff oder eine niedere Alkylgruppe sowie die Alkali- und Erdalkalimetallsalze der unter die allgemeine Formel I fallenden freien Carbonsäuren, sind bis jetzt nicht bekanntgeworden.
Wie nun gefunden wurde, besitzen die Verbindungen der Formel r sowie die Alkali- und Erdalkalimetallsalze der unter die allgemeine Formel I fallenden freien Carbonsäuren, die peroral oder rektal verabreicht werden können, interessante pharmakologische Eigenschaften und einen hohen therapeutischen Index. Sie zeigen insbesondere hypolipämische Wirksamkeit, die sich z. B.
an der Senkung des Cholesterin- und Triglyceridspiegels im Blut und Leber bei mehrmaliger oraler Verabreichung an männlichen Ratten nach Standardmethoden nachweisen lässt. Das Gesamtcholesterin wird nach R. Richterich und K. Lauber [vgl. Klin. Wochenschr. 40, 1252 bis 1256 (1962)] direkt im Serum bestimmt. Ferner werden nach J. Folch et al. [vgl. J. Biol. Chem. 226, 497 (1957)] Serum- sowie Leberlipide extrahiert und Triglyceride und Gesamtcholesterin mit dem Autoanalyzer nach G. Kessler und H. Lederer [vgl. Automation in der analytischen Chemie (1965), Technicon GmbH, Frankfurt/M., Seiten 863-872, bzw. W. D. Block et al.,ibid.
Seiten 970 und 971] bestimmt.
Bei diesen Versuchen zeigen ivlsbesondere die 2-(4b,5,6,7,8,8a¯Hexahydro-fluoren-2-yloxy)- heptans äure,
2-(4b,5,6,7,8,8aHHexahydro-fluoren-2-yloxy)-
5-methyl-hexansäure, 2-(4b, 5,6,7,8,8 a-Hexahydro-fluoren-2-yloxy)- octansäure, und die 2-(4b,5, 6,7,8,8a-Hexahydro-fluoren-2-ylthio)- heptansäure.
Nach dem erfindungsgemässen Verfahren erhält man Verbindungen der allgemeinen Formel I, indem man eine Verbindung der allgemeinen Formel II,

in welcher R1, R2 und R3 die unter Formel I angegebene Bedeutung haben, reduziert und gewünschtenfalls eine erhaltene Carbonsäure der allgemeinen Formel I in ein Alkali- oder Erdalkalimetallsalz überführt.
Die Reduktion wird vorzugsweise durch Hydrierung in Gegenwart eines Katalysators in einem Lösungsmittel vorgenommen. Geeignete Katalysatoren sind Metalle der achten Gruppe des periodischen Systems, z. B. Raney Nickel, Raney-Cobalt, Edelmetallkatalysatoren, z. B.
Ruthenium, Rhodium, Platin und insbesondere Palladium. Es ist vorteilhaft, die Katalysatoren auf Trägern, z. B. auf Kohle, Aluminiumoxid oder Bariumsulfat, einzusetzen. Als Lösungsmittel eignen sich z.B. niedrige Alkanole, wie Methanol, Athanol oder Isopropanol, ätherartige Flüssigkeiten, wie Tetrahydrofuran oder insbesondere Dioxan, Carbonsäureäthylester, wie z. B.
Essigsäureäthylester, Wasser und für Edelmetallkatalysatoren auch Carbonsäuren, wie Eisessig. Verwendet man Raney-Katalysatoren, so setzt man der Reaktionslösung, falls R3 Wasserstoff ist, mindestens 1 Mol äquivalent eines Alkalihydroxids, z. B. Natriumhydroxid, zu. Die Hydrierung kann bei einer Temperatur von etwa 20-150 und einem Druck von 1-200 Atmosphären vorgenommen werden und wird nach Aufnahme von
3 Moläquivalenten Wasserstoff abgebrochen.
Die Ausgangsverbindungen der allgemeinen Formel
II können beispielsweise durch Umsetzung eines Alkali metallsalzes von Fluoren-2-ol mit einem durch R1 und R substituierten Alkalisalz der Bromessigsäure oder einem niederen Alkylester dieser Säure erhalten werden.
Die Verbindungen der allgemeinen Formel I ent halten 3 Assymmetriezentren. Dementsprechend sind
8 enantiomere Formen und 4 Racemate möglich. In den Verbindungen, die durch katalytische Hydrierung des
Fluorensystems bzw. des 5,6,7,8 -Tetrahydrofluoren systems erhalten wurden, liegen in der 4b- und 8a-Stellung cis-ständige Wasserstoffe vor. In diesem Fall exis tieren nur 4 enantiomere Formen und 2 Racemate.
Dieses Auftreten von Isomeren kann bei der Aufarbei tung insofern zu Schwierigkeiten führen, als die gebil deten Racemate der von der allgemeinen Formel I umfassten Carbonsäuren unterschiedliche Kristallisationstendenz besitzen. Aus diesem Grund führt die Bildung von nicht kristallisierenden Racematen zu einer Ausbeuteverschlechterung. In anderen Fällen kann auch ein Gemisch zweier kristalliner Racemate erhalten werden, was sich darin äussert, dass der Schmelzpunkt der so erhaltenen Produkte zwischen den Schmelzpunkten der reinen Komponenten liegt.
Die nicht kristallisierbaren diastereo-isomeren Racemate der von der allgemeinen Formel I umfassten Carbonsäuren können in kristalli- sierbare Diastereoisomere übergeführt werden, indem man sie in einen Ester überführt und diesen Ester durch Erhitzen in einem Lösungsmittel in Gegenwart von starken Basen epimerisiert und anschliessend zur Carbonsäure hydrolysiert. Auf diese Weise wird eine weitere, von der Lage des Epimerisierungsgleichgewichts abhängige Menge an kristallisierbarer Säure erhalten.
Die Behandlung kann nötigenfalls wiederholt werden.
Als Alkali- und Erdalkalimetallsalze von unter die allgemeine Formel I fallenden Carbonsäuren kommen beispielsweise deren Natrium-, Kalium-, Lithium-, Magnesium- und Calciumsalze in Frage. Die Herstellung dieser Salze erfolgt beispielsweise durch Zusammengeben von Säure und Base in einem geeigneten Lösungsmittel, wie z. B. Methanol, Äthanol, Aceton-Wasser. Entstandene, relativ schwer lösliche Salze können durch Abfiltrieren isoliert werden, leichtlösliche durch Eindampfen des Lösungsmittels. Ferner lassen sich Salze, die im verwendeten Lösungsmittel relativ schwer löslich sind, auch durch doppelte Umsetzung eines andern Salzes der Säure mit der Base oder einem geeigneten Salz derselben herstellen.
Die Verbindungen der allgemeinen Formel I und die Alkali- und Erdalkalimetallsalze der unter diese Formel fallenden freien Carbonsäuren werden, wie weiter vorne erwähnt, peroral oder rektal verabreicht. Die täglichen Dosen bewegen sich zwischen 0,5 bis 10 mg/kg für Warmblüter. Geeignete Doseneinheitsformen, wie Dragees, Tabletten, Suppositorien und Kapseln, enthalten als Wirkstoff vorzugsweise 10-250 mg, z. B. 50 oder 100 mg einer Verbindung der allgemeinen Formel I oder eines Alkali- oder Erdalkalimetallsalzes einer von der allgemeinen Formel I umfassten freien Carbonsäure.
Doseneinheitsformen für die perorale Anwendung enthalten als Wirkstoff vorzugsweise zwischen 10 und 90SE; einer Verbindung der allgemeinen Formel I. Zu ihrer Herstellung kombiniert man den Wirkstoff z. B.
mit festen, pulverförmigen Trägerstoffen, wie Lactose, Saccharose, Sorbit, Mannit; Stärken, wie Kartoffelstärke, Maisstärke oder Amylopektin, ferner Laminariapulver oder Citruspulpenpulver; Cellulosederivaten oder Gelatine, gegebenenfalls unter Zusatz von Gleitmitteln, wie Magnesium- oder Calciumstearat oder Polyäthylen glykolen, zu Tabletten oder zu Dragée-Kernen. Die Dragee-Kerne überzieht man beispielsweise mit konzentrierten Zuckerlösungen, welche z. B. noch arabischen Gummi, Talk und/oder Titandioxid enthalten können, oder mit einem Lack, der in leichtflüchtigen organischen Lösungsmitteln oder Lösungsinittelgemischen gelöst ist.
Diesen Überzügen können Farbstoffe zugefügt werden, z. B. zur Kennzeichnung verschiedener Wirkstoffdosen.
Als weitere orale Doseneinheitsformen eignen sich Steckkapseln aus Gelatine sowie weiche, geschlossene Kapseln aus Gelatine und einem Weichmacher, wie
Glycerin. Die Steckkapseln enthalten den Wirkstoff vorzugsweise als Granulat, z. B. in Mischung mit Füllstoffen, wie Mais stärke, und/oder Gleitmitteln, wie Talk oder Magnesiumstearat, und gegebenenfalls Stabilisatoren, wie Natriummetabisulfit (Na2SaO) oder Ascorbinsäure. In weichen Kapseln ist der Wirkstoff vorzugsweise in geeigneten Flüssigkeiten, wie flüssigen Poly äthylenglykolen, gelöst oder suspendiert.
Als Doseneinheitsformen für die rektale Anwendung kommen z. B. Suppositorien in Betracht, welche aus einer Kombination eines Wirkstoffes mit einer Suppositoriengrundmasse bestehen. Als Suppositoriengrundmasse eignen sich z. B. natürliche oder synthetische Triglyceride, Paraffinkohlenwasserstoffe, Polyäthylenglykole oder höhere Alkanole. Ferner eignen sich auch Gelatine Rektalkapseln, welche aus einer Kombination des Wirkstoffes und einer Grundmasse bestehen. Als Grundmasse eignen sich z. B. flüssige Triglyceride, Polyäthylenglykole oder Paraffinkohlenwasserstoffe.
Die folgenden Vorschriften sollen die Herstellung von Tabletten, Dragees, Suppositorien und Kapseln näher erläutern: a) 1000 g 2-(4b,5,6,7,8,Xa-Hexahydro-fluoren-2- yloxy)-heptansäure oder 2-(4b,5,6,7,8,8a-Hexahydro- fluoren-2-ylthio)-heptansäure werden mit 550 g Lactose und 292 g Kartoffelstärke vermischt, die Mischung mit einer alkoholischen Lösung von 8 g Gelatine befeuchtet und durch ein Sieb granuliert. Nach dem Trocknen mischt man 60 g Kartoffelstärke, 60 g Talk und 10 g Magnesiumstearat und 20 g hochdisperses Siliciumdioxid zu und presst die Mischung zu 10000 Tabletten von je 200 mg Gewicht und 100 mg Wirkstoffgehalt, die gewünschtenfalls mit Teilkerben zur feineren Anpassung der Dosierung versehen sein können.
b) 100 g 2-(4b,5,6,7,8,8a-Hexahydro-fluoren-2yloxy)-heptansäure oder a-(4b,5,6,7,8,8a-Hexahydro- fluoren-2-yloxy)-cyclohexanessigsäure werden mit 16 g Maisstärke und 6 g hochdispersem Siliciumdioxid gut vermischt. Die Mischung wird mit einer Lösung von 2 g Stearinsäure, 6 g Äthylcellulose und 6 g Stearin in etwa 70 ml Isopropylalkohol befeuchtet und durch ein Sieb III (Ph. Helv. V) granuliert. Das Granulat wird etwa 14 Stunden getrocknet und dann durch Sieb II-IIIa geschlagen. Hierauf wird es mit 16 g Maisstärke, 16 g Talk und 2 g Magnesiumstearat vermischt und zu 1000 Dragees-Kemen gepresst.
Diese werden mit einem konzentrierten Sirup von 2 g Lacca, 7,5 g arabischem Gummi, 0,15 g Farbstoff, 2 g hochdispersem Siliciumdioxid, 25 g Talk und 53,35 g Zucker überzogen und getrocknet. Die erhaltenen Dragees wiegen je 260 mg und enthalten je 100 mg Wirkstoff.
c) Man bereitet eine Suppositorienmasse aus 10,0 g 2-(4b,5,6,7,8n8a-Hexahydro-fluoren-2-yloxy)- heptansäure oder 2-(4b,5,6,7, 8, 8a-Hexahydro-fluoren-2-ylthio)- heptansäure und 163,5 g Adeps solidus und giesst damit 100 Suppositorien mit je 100 mg Wirkstoffgehalt.
d) Um 1000 Kapseln mit je 75 mg Wirkstoffgehalt herzustellen, mischt man 75 g 2-(4b,5,6,7,8,8a-Hexahydro-fluoren-2-yloxy)- heptansäure oder 2-(4b,5,6,7,8,8a-Hexahydro-fluoren-2-ylthio)- heptansäure mit 198,0 g Lactose, befeuchtet die Mischung gleichmässig mit einer wässrigen Lösung von 20 g Gelatine und granuliert sie durch ein geeignetes Sieb (z. B. Sieb TII nach Ph. Helv. V). Das Granulat mischt man mit 10,0 g getrockneter Maisstärke und 15,0 g Talk und füllt es gleichmässig in 1000 Hartgelatinekapseln der Grösse 1.
Die nachfolgenden Beispiele erläutern die Herstellung der Verbindungen der allgemeinen Formel I und von Salzen derselben näher, sollen jedoch den Umfang der Erfindung in keiner Weise beschränken. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
Man hydriert bei 1000 und unter 100 Atmosphären Druck eine Lösung von 14,5 g (0,046 Mol) 2-(Fluoren2-yloxy)-heptansäure in 101) ml Dioxan innerhalb 15 Stunden in Gegenwart von 3 g Palladiumkohle (5 % Palladium). Dann wird vom Katalysator abfiltriert, mit Dioxan nachgewaschen und die Lösung im Vakuum eingedampft. Man reinigt den Rückstand durch Elutionschromatographie an einer Säule von neutralem Kieselgel (Merck, Korngrösse 0,05-0,2 mm). Als Elutionsmittel verwendet man Benzol-Eisessig (85 : 15). Die vereinigten Fraktionen, welche das Rohprodukt enthalten, werden eingedampft. Der Rückstand wird in Äther aufgenommen, die Ätherlösung mit Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft.
Der Rückstand, den man aus Pentan umkristallisiert, liefert 4,7 g (31,8 % der Theorie) 2-(4b,5,6,7,8,8a- Hexahydro-fluoren-2- yloxy)-heptansäure vom Smp. 81-82 .
Die Ausgangsverbindung, die 2-(Fluoren-2-yloxy)heptansäure, wird wie folgt hergestellt: a) Unter Rühren und Feuchtigkeitsausschluss fügt man zu einer Lösung von 0,945 g (0,041 Mol) Natrium in 40 ml abs. Äthanol unter Stickstoff 6,0 g (0,033 Mol) Fluoren-2-ol. Zu der erhaltenen Lösung des Natriumfluoren-2-olats tropft man 8,2 g (0,0346 Mol) 2-Bromheptansäureäthylester und kocht das Gemisch 3 Stunden unter Rückfluss. Man dampft das Reaktionsgemisch im Vakuum ein-und verteilt den Rückstand zwischen Wasser und Äther. Die Ätherphase wird mit 0,5n Natronlauge ausgeschüttelt, mit Wasser neutral gewaschen, über Magnesiumsulfat getrocknet und eingedampft. Man erhält ein dunkelgelbes Ö1, welches kristallisiert.
Das Rohprodukt wird an einer Säule von 200 g neutralem Kieselgel (Merck, Korngrösse 0,05-0,2 mm) nach der Durchlaufmethode chromatographiert. Als Elutionsmittel verwendet man Benzol. Die Fraktionen, welche das rohprodukt enthalten, dampft man ein. Der Rückstand wird aus Benzin (Kp. 65-100 ) umkristallisiert und man erhält 10,1 g (90,6 % d. Th.) 2-(Fluoren-2-yloxy)-heptansäureäthylester vom Smp. 58-61 .
b) 10,0 g 2-(Fluoren-2-yloxy)-heptans äureäthylester fügt man zu einer Lösung von 4,0 g Kaliumhydroxid in 150 ml Methanol und kocht das Gemisch 30 Minuten unter Rückfluss. Dann dampft man das Reaktionsgemisch ein und verteilt den Rückstand zwischen Äther und Wasser. Die wässrige Phase wird mit konz. Salzsäure angesäuert und ausgeäthert. Man wäscht den Atherextrakt mit Wasser, trocknet ihn über Magnesiumsulfat und dampft ihn ein. Der Rückstand wird aus Methanol-Wasser umkristallisiert und man erhält 8,0 g (78,5 % d. Th.) 2-(Fluoren-2-yloxy)-heptansäure vom Smp. 133-136 .
Beispiel 2

Beispiel 3
19,37 g (0,0558 Mol) 2-Fluoren-2-yloxy)-heptan- säureäthylester werden in 200 ml Essigester gelöst und unter Zusatz von 2 g Palladiumkohle (5 % Palladium) innerhalb von 51/2 Stunden bei einer Temperatur von
800 und einem Druck von 12 atü hydriert. Dann wird vom Katalysator abfiltriert, mit Essigester nachgewaschen und die Lösung im Vakuum eingedampft. Der Rückstand wird einer Vakuumdestillation unterworfen.
Man erhält 10,39 g (52,7 %) des gewünschten 2-(4b,5,6,7, 8 ,8a-Hexahydro-fluoren-2-yloxy)- heptans äureäthylesters vom Siedepunkt 164 bis 1660 bei einem Druck von 0,04 Torr, nD": 1,5121.
Beispiel 4
Man hydriert bei 800 und unter 12 Atmosphären Druck eine Lösung von 150 g (0,483 Mol) 2-(Fluoren2-yloxy)-heptansäure in 1,5 Liter Essigester innerhalb von 5t/2 Stunden in Gegenwart von 15 g Palladiumkohle (5 % Palladium). Dann wird vom Katalysator abfiltriert, mit Essigester nachgewaschen und die Lösung im Vakuum eingedampft. Den vom Lösungsmittel befreiten Rückstand löst man in 700 ml Pentan unter Erwärmung, bis eine klare Lösung entsteht, und kühlt dann den Kolbeninhalt im verschlossenen Kolben mit einer Trockeneis-Aceton-Mischung auf - -300 ab. Innerhalb von 10-15 Minuten kristallisiert das gewünschte Produkt aus, wird auf einer Nutsche abgesogen und mit 250 ml auf - -300 gekühltem Pentan nachgewaschen.
Nach dem Trocknen bei 500 und 60 Torr erhält man 77,1 g Kristalle, die nochmals aus 1,2 Liter Pentan umkristallisiert werden. Zur Kristallisation wird die Lösung etwa 3-4 Stunden bei -10 bis -150 aufbewahrt, abgenutscht und mit 200 ml auf -300 gekühltem Pentan nachgewaschen. Nach dem Trocknen werden 65,1 g 2-(4b, 5,6,7,8, 8a-Hexahydro-fluoren-2-yloxy)- heptansäure vom Smp. 81-820 erhalten. Ausbeute 43 % der Theorie.
Das in der Mutterlauge vorliegende zweite diastereoisomere Racemat, das nicht kristallisierbar ist, kann auf folgende Weise in das vorgenannte diastereoisomere Racemat vom Schmelzpunkt 81-820 umgewandelt werden: a) Die vereinigten Mutterlaugen der Kristallisationen werden im Vakuum eingedampft und der Rückstand (86,5 g/0,274 Mol) in 600 ml Benzol gelöst, mit 50 ml (0,86 Mol) abs. Äthanol und 1 g p-Toluolsulfosäure als Katalysator versetzt und die klare Lösung 15 Stunden mit einem Wasserabscheider azeotrop am Rückfluss verestert. Nach Beendigung wird im Vakuum eingedampft mit 400 ml Äther aufgenommen, einmal mit 100 ml gesättigter Natriumhydrogencarbonat-Lösung gewaschen und mit Wasser bis zur neutralen Reaktion des Waschwassers nachgewaschen.
Die ätherische Lösung wird über MgSO4 getrocknet, abfiltriert und im Vakuum eingedampft. Der Rückstand (93,4 g) wird bei 0,04 Torr destilliert. Die Fraktionen mit dem Siedepunkt von 164-1670 werden zur Weiterverarbeitung verwendet, n2,3": 1,5110, Ausbeute 77,8 g (82,6% der Theorie).
b) 77,65 g (0,226 Mol) des oben erhaltenen Esters werden zu einer Lösung von 0,518 g (0,0226 Mol) Natrium-Metall in 500 ml abs. Äthanol gegeben und 40 Stunden unter Feuchtigkeitsausschluss am Rückfluss erhitzt. Dann wird mit einer Lösung von 28,3 g (0,429 Mol) KOH 85 % puriss. in 150 ml Wasser versetzt und weitere 1¸ Stunden am Rückfluss erwärmt. Nach Beendigung wird das Reaktionsgemisch im Vakuum eingedampft, der ölige Rückstand in etwa 1 Liter Wasser gelöst und mit 250 ml 2n Salzsäure versetzt. Das sich abscheidende öl wird in etwa 400 ml Äther aufgenommen, abgetrennt und die ätherische Lösung mit Wasser aufgenommen, abgetrennt und die ätherische Lösung mit Wasser neutral gewaschen. Nach dem Trocknen über MgSO4 wird abfiltriert, mit Tierkohle entfärbt, nochmals filtriert und im Vakuum eingedampft.
Der Rückstand wird bei 900 und 11 Torr vom Lösungsmittel ganz befreit, und man erhält 72 g des öligen Rohproduktes. Dasselbe wird in 350 ml Pentan gelöst und im verschlossenen Kolben unter Animpfen auf - 300 in einer Aceton-Trockeneis-Mischung gekühlt. Nach 10 bis 15 Minuten, wenn die Kristallisation beendigt ist, wird abgenutscht, mit 150 ml auf -50 gekühltem Pentan nachgewaschen und der Rückstand bei 500 und 60 Torr getrocknet. Die erhaltenen 29 g Kristalle werden nochmals in 500 ml Pentan gelöst und aus Pentan umkristallisiert.
Nach dem Trocknen werden 25 g (16,5% der Theorie bezogen auf die ursprünglich eingesetzte 2 [Fluoren-2-yloxyj-heptans äure) der gewünschten 2-(4b,5,6,7,8,8a-Hexahydro-fluoren-2-yloxy)- heptansäure vom Smp. 81-820 erhalten.
Aus der Mutterlauge werden durch Wiederholung der unter a) und b) beschriebenen Operationen weitere 14,6 g (9,6 % der Theorie bezogen auf ursprünglich eingesetzte 2-[Fluoren-2-yloxy]-heptansäure) erhalten.

Fluorine system or the 5,6,7,8-tetrahydrofluorene system are obtained in the 4b and 8a positions cis hydrogen. In this case there are only 4 enantiomeric forms and 2 racemates.
This occurrence of isomers can lead to difficulties during work-up insofar as the racemates formed of the carboxylic acids encompassed by the general formula I have different tendencies towards crystallization. For this reason, the formation of non-crystallizing racemates leads to a deterioration in the yield. In other cases, a mixture of two crystalline racemates can also be obtained, which is manifested in the fact that the melting point of the products thus obtained is between the melting points of the pure components.
The non-crystallizable diastereoisomeric racemates of the carboxylic acids encompassed by the general formula I can be converted into crystallizable diastereoisomers by converting them into an ester and epimerizing this ester by heating in a solvent in the presence of strong bases and then hydrolyzing it to the carboxylic acid . In this way a further amount of crystallizable acid, which depends on the position of the epimerization equilibrium, is obtained.
The treatment can be repeated if necessary.
As alkali and alkaline earth metal salts of carboxylic acids falling under the general formula I, for example, their sodium, potassium, lithium, magnesium and calcium salts are suitable. These salts are prepared, for example, by combining acid and base in a suitable solvent, such as. B. methanol, ethanol, acetone-water. The relatively sparingly soluble salts formed can be isolated by filtering off, easily soluble salts by evaporating the solvent. The granulate is dried for about 14 hours and then passed through sieve II-IIIa. It is then mixed with 16 g of corn starch, 16 g of talc and 2 g of magnesium stearate and pressed into 1000 coated tablets.
These are coated with a concentrated syrup of 2 g Lacca, 7.5 g gum arabic, 0.15 g color, 2 g highly dispersed silicon dioxide, 25 g talc and 53.35 g sugar and dried. The coated tablets each weigh 260 mg and each contain 100 mg of active ingredient.
c) A suppository mass is prepared from 10.0 g of 2- (4b, 5,6,7,8n8a-hexahydro-fluoren-2-yloxy) - heptanoic acid or 2- (4b, 5,6,7, 8, 8a- Hexahydro-fluoren-2-ylthio) heptanoic acid and 163.5 g of Adeps solidus and pour 100 suppositories with 100 mg of active ingredient each.
d) To produce 1000 capsules with 75 mg active ingredient content each, mix 75 g of 2- (4b, 5,6,7,8,8a-hexahydro-fluoren-2-yloxy) -heptanoic acid or 2- (4b, 5,6 , 7,8,8a-hexahydro-fluoren-2-ylthio) -heptanoic acid with 198.0 g of lactose, moisten the mixture evenly with an aqueous solution of 20 g of gelatin and granulate it through a suitable sieve (e.g. sieve TII according to Ph. Helv. V). The granules are mixed with 10.0 g of dried corn starch and 15.0 g of talc and filled evenly into 1000 size 1 hard gelatine capsules.
The following examples explain the preparation of the compounds of general formula I and salts thereof in more detail, but are not intended to restrict the scope of the invention in any way. The temperatures are given in degrees Celsius.
example 1
A solution of 14.5 g (0.046 mol) of 2- (fluoren2-yloxy) -heptanoic acid in 101) ml of dioxane is hydrogenated at 1000 and under 100 atmospheric pressure in the presence of 3 g of palladium-carbon (5% palladium) over the course of 15 hours. The catalyst is then filtered off, washed with dioxane and the solution is evaporated in vacuo. The residue is purified by elution chromatography on a column of neutral silica gel (Merck, particle size 0.05-0.2 mm). Benzene-glacial acetic acid (85:15) is used as the eluent. The combined fractions, which contain the crude product, are evaporated. The residue is taken up in ether, the ether solution is washed with water, dried over sodium sulfate and evaporated.
The residue, which is recrystallized from pentane, gives 4.7 g (31.8% of theory) of 2- (4b, 5,6,7,8,8a-hexahydro-fluoren-2-yloxy) -heptanoic acid with a melting point of. 81-82.
The starting compound, 2- (fluoren-2-yloxy) heptanoic acid, is prepared as follows: a) While stirring and excluding moisture, it is added to a solution of 0.945 g (0.041 mol) of sodium in 40 ml of abs. Ethanol under nitrogen 6.0 g (0.033 mol) of fluorene-2-ol. 8.2 g (0.0346 mol) of ethyl 2-bromoheptanoate are added dropwise to the resulting solution of sodium fluorene-2-olate, and the mixture is refluxed for 3 hours. The reaction mixture is evaporated in vacuo and the residue is partitioned between water and ether. The ether phase is extracted with 0.5N sodium hydroxide solution, washed neutral with water, dried over magnesium sulfate and evaporated. A dark yellow oil is obtained which crystallizes.
The crude product is chromatographed on a column of 200 g of neutral silica gel (Merck, particle size 0.05-0.2 mm) using the continuous flow method. Benzene is used as the eluent. The fractions which contain the crude product are evaporated. The residue is recrystallized from gasoline (bp 65-100) and 10.1 g (90.6% of theory) of 2- (fluoren-2-yloxy) -heptanoic acid ethyl ester of melting point 58-61 are obtained.
b) 10.0 g of ethyl 2- (fluoren-2-yloxy) -heptane are added to a solution of 4.0 g of potassium hydroxide in 150 ml of methanol and the mixture is refluxed for 30 minutes. The reaction mixture is then evaporated and the residue is partitioned between ether and water. The aqueous phase is diluted with conc. Hydrochloric acid acidified and etherified. The ether extract is washed with water, dried over magnesium sulphate and evaporated. The residue is recrystallized from methanol-water and 8.0 g (78.5% of theory) of 2- (fluoren-2-yloxy) -heptanoic acid with a melting point of 133-136 are obtained.

Example 3
19.37 g (0.0558 mol) of 2-fluoren-2-yloxy) -heptanoic acid ethyl ester are dissolved in 200 ml of ethyl acetate and, with the addition of 2 g of palladium carbon (5% palladium) within 51/2 hours at a temperature of
800 and a pressure of 12 atmospheres hydrogenated. The catalyst is then filtered off, washed with ethyl acetate and the solution is evaporated in vacuo. The residue is subjected to vacuum distillation.
10.39 g (52.7%) of the desired ethyl 2- (4b, 5,6,7, 8, 8a-hexahydro-fluoren-2-yloxy) heptane acid ester of boiling point 164 to 1660 at a pressure of 0 are obtained , 04 Torr, nD ": 1.5121.
Example 4
A solution of 150 g (0.483 mol) of 2- (fluoren2-yloxy) -heptanoic acid in 1.5 liters of ethyl acetate is hydrogenated at 800 and under 12 atmospheric pressure in the presence of 15 g of palladium carbon (5% palladium) . The catalyst is then filtered off, washed with ethyl acetate and the solution is evaporated in vacuo. The residue freed from the solvent is dissolved in 700 ml of pentane with warming until a clear solution is formed, and the contents of the flask are then cooled in the closed flask with a dry ice-acetone mixture to -300. The desired product crystallizes out within 10-15 minutes, is suctioned off on a suction filter and washed with 250 ml of pentane cooled to -300.
After drying at 500 and 60 torr, 77.1 g of crystals are obtained, which are recrystallized again from 1.2 liters of pentane. For crystallization, the solution is stored for about 3-4 hours at -10 to -150, suction filtered and washed with 200 ml of pentane cooled to -300. After drying, 65.1 g of 2- (4b, 5,6,7,8, 8a-hexahydro-fluoren-2-yloxy) heptanoic acid with a melting point of 81-820 are obtained. Yield 43% of theory.
The second diastereoisomeric racemate present in the mother liquor, which cannot be crystallized, can be converted into the aforementioned diastereoisomeric racemate of melting point 81-820 in the following way: a) The combined mother liquors from the crystallizations are evaporated in vacuo and the residue (86.5 g / 0.274 mol) dissolved in 600 ml of benzene, with 50 ml (0.86 mol) of abs. Ethanol and 1 g of p-toluenesulphonic acid were added as a catalyst and the clear solution was esterified azeotropically under reflux using a water separator. After completion, it is evaporated in vacuo, taken up with 400 ml of ether, washed once with 100 ml of saturated sodium hydrogen carbonate solution and rewashed with water until the washing water reacts neutral.
The ethereal solution is dried over MgSO4, filtered off and evaporated in vacuo. The residue (93.4 g) is distilled at 0.04 torr. The fractions with a boiling point of 164-1670 are used for further processing, n2.3 ": 1.5110, yield 77.8 g (82.6% of theory).
b) 77.65 g (0.226 mol) of the ester obtained above are added to a solution of 0.518 g (0.0226 mol) of sodium metal in 500 ml of abs. Added ethanol and refluxed for 40 hours with exclusion of moisture. Then with a solution of 28.3 g (0.429 mol) of KOH 85% puriss. added in 150 ml of water and refluxed for a further 1¸ hours. After completion, the reaction mixture is evaporated in vacuo, the oily residue is dissolved in about 1 liter of water and 250 ml of 2N hydrochloric acid are added. The oil which separates out is taken up in about 400 ml of ether, separated off and the ethereal solution taken up with water, separated off and the ethereal solution washed neutral with water. After drying over MgSO4, it is filtered off, decolorized with animal charcoal, filtered again and evaporated in vacuo.
The residue is completely freed from the solvent at 900 and 11 torr, and 72 g of the oily crude product are obtained. The same is dissolved in 350 ml of pentane and cooled in a closed flask with seeding to -300 in an acetone-dry ice mixture. After 10 to 15 minutes, when the crystallization is complete, suction filtered, washed with 150 ml of pentane cooled to -50 and the residue dried at 500 and 60 torr. The 29 g of crystals obtained are dissolved again in 500 ml of pentane and recrystallized from pentane.
After drying, 25 g (16.5% of theory based on the originally used 2 [fluoren-2-yloxyj-heptanoic acid) of the desired 2- (4b, 5,6,7,8,8a-hexahydro-fluorene- 2-yloxy) -heptanoic acid of m.p. 81-820.
By repeating the operations described under a) and b), a further 14.6 g (9.6% of theory based on the 2- [fluoren-2-yloxy] -heptanoic acid originally used) are obtained from the mother liquor.
