,Die erhaltenen basischen Verbindungen können ge- gobenenfalls mit optisch aktiven Säuren umgesetzt, die optisch aktiven Basen daraus in Freiheit ges etzt und die racemischen oder optisch, aktiven Basen in Salze von physiologisch verträglichen Säuren überführt werden.
Man kann solche Oxazolidone beispielsweise erhal ten, wenn man I-Amino-2-hydroxy-3-(2'-cyclopentyl- bzw. cyelopentenyl-phenoxy)-propan mit einem reak- tionsfählgen Derivat der Kohlensäure, wie Diäthylcar- bonat, Chlorkohlensäuremethylester oder Phosgen um setzt oder wenn man ein gogebenonfallls in 3-Stellung entsprechend substibuierteis SHydroxymeihyl-oxazoli- don-(2) in Form eines Halogenwasserstoffsäure-, Schwefelsäure- oder Sulfonsäureesters mit einem Alkali- 2-cyclopentyl- bzw. cycliopontienylphenolat umsetzt.
Ge eignete Oxazolidine kann man z. B. herstellen, indem man I-Amino-2-,hydroxy-3-(2'-cyclopentyl- bzw. cyclo- pentemyl-phenoxy)-propan mit Aldehyden oder Ketonen umsetzt. Am -Stickstoff nicht substituierte Oxazolidone bzw. Oxazolidine können mit den entsprechenden Al kylhalogeniden b zw. -sulfonatien alkylier t werden. Die Hydrolyse dieser Oxazolliidon- bzw.
Oxazolidiniderivate kann in saurem oder alkalischem Milieu, durchgeführt werden, beispielsweise mittels verdünnter Salzsäure, verdünnter Schwefelsäure, verdünnter Natronlauge oder verdünnter Kalilauge. Vorteilhaft wird zur Beschleuni gung der Hydrolyse erwärmt. Die Hydrolyse kann auch in Gegenwart wasserlöslicher Lösungsmittel, z. B. nie- iderer Alkohole durchgeführt werden.
Schliesslich kann man die Cyclopentylphenoläther der Formel 1 erhalten, indem man die nach dem genann ten Verfahren hergestellten basisch substituierten Cy- clopentenylphenoläther der -allgemeinenFormel (V) hy driert. Als Katalysatoren für diese Hydrierung.

Es kann mitunter von Vorteil sein, die Herstellung der Ausgangsverbindungen direkt mit der weiteren Um setzung zu kombinieren, d. h. die Ausgangsstoffe nicht gesondert zu isolieren.
Die Verfahrensprodukte können als freie Basen oder in Form ihrer Salze anfallen, und falls erforder lich, durch die üblichen Aufarbeitungsmethoden, bei spielsweise durch Umkristallisieren oder gegebenenfalls Überführen in die freie Base und anschliessende Be handlung mit einer geeigneten Säure ",gereinigt werden. Die Verfahrensprodukte können gegebenenfalls in die Salze physiolpgisch verträglicher organischer oder an organischer Säuren übergeführt werden.
Als organische Säuren seinen beispielsweise ge nannt: Essigsäure, Malonsäure, Propionsäure, Milchsäure, Bernsteinsäure, Weinsäure, Maleinsäure, Funiarsäure, Zitronensäure, Äpfelsäure, Benzoesäure, Salicylsäure, Oxäthansulifonsäure, Acetursäure, Äthylendiamintetra essigsäure, Embonsäure, sowie saure Gruppen enthal tende synthetische Harze.
Als anorganische Säuren kommen beispielsweise in Betracht: Halogenwasserstoffsäuren wie Chlorwasserstoff säure oder Bromwasserstoffsäure, Schwefelsäure, Phos phorsäure und Amidosulfansäure.
Man kann die optisch aktiven Isomeren der Cyclo- pentylphenoläther erhalten, wenn man die racemischen Verbindungen der allgemeinen Formel (1) mit optisch aktiven Säuren in ihre Komponenten spaltet.
Als Säuren, wie sie für die Herstellung optisch akti- ver Salze nach der Erfindung in Frage kommen, seien beispielsweise genannt:
(+)- und (-)-Weinsäure, (+) und (-)-Dibenzoylweinsäure, (+)- und (-)-Ditoluyl- weinsäure, (+)- und (-)-M andelsäure, (+)- und (-) Camphersäure, (+)-Campher-/>-;sulfonsäure, (+)-u- Bromcanipher-z7-sul,fonsäure und N-(p-Nitrobenzoyl)- (+)-glutaminsäure.
Die Herstellung der optisch aktiven Salze kann in Wasser, wasserhaltigen oder wasserfreien organischen Lösungsmitteln erfolgen. Vorteilhaft erweist sich die Verwendung von Alkoholen oder von Estern organi scher Carbonsäuren.
Zur Herstellung von optisch aktiven Verbindungen setzt man das Racemat :der Base in einem Lösungsmit tel, vorzugsweise in molaren Mengenverhältnissen mit einer optisch aktiven Säure um und isoliert das optisch aktive Salz der Verbindungen der allgemeinen Formel (1). Man kann in bestimmten Fällen auch nur ein halbes Äquivalent der optisch aktiven Säure verwenden, um den einen optischen Antipoden aus dem Racernat zu entfernen, wie man ebenso auch überschüssige !Mengen optisch aktiver Säure einsetzen kann.
Je nach Art der optisch aktiven Säure kann der gewünschte Antipode entweder direkt oder aus der Mutterliauge des ersten Kristallisats erhalten werden. Man kann ansehdmessenid die optisch aktive Base in üblicher Weise au s dem Salz in Freiheit setzen und diese optisch aktive Base in ein Salz einer der genannten physiologisch verträglichen organischen oder anorganischen Säuren überführen.
Man kann zu den optisch aktiven Antipoden der Cyclopentylphenoläther auch gelangen, wenn man den entsprechenden optisch aktiven Cyelopentenylphenol- äther katalytisch hydriert. Die erfindungsgemässen Verbindungen -sind wert volle Heilmittel, die vorteilhafte pharmakologische Ei genschaften besitzen und speziell als Antagonisten der @@-Receptoren des adrenergischen Nervensystems ausser- ,ordentlich wirksam sind. Beispielsweise hat sich das 1-tert:
Butylamino-2-hydroxy-3-(2'-cyclopeutyl- phenoxy)-propan als stark wirksamer Antagonist der /2,-Reccptoren des aderenergischen Nervensystems erwiesen.
Die Alundrin antagonistische Wirkunigsstärke der er findungsgemässen Verbindungen, die sowohl an der iso lierten Trachea vom Meerschweinchen als auch am iso- lierten Vorhof vom Meerschweinchen getestet wurde, lag in der gleichen Grössenordnung wie die des bekann ten I-Isopropylamino-2-hydroxy-3-(, < -naphthoxy)-pro- pan. Auch hinsichtlich der akuten Toxizität entsprachen ,die Verfahrensprodukte der bekannten Verbindung.
Darüber hinaus zeichneten sich die neuen Verbin dungen durch eine gegenüber bekannten Verbindungen wesentlich längere Wirkungsdauer aus, ein Vorteil, der sowohl in vitro als auch in vivo nachgewiesen werden konnte. So wurden beispielsweise bei einem Vergleich des I-tert.-Butylamino-2-hydroxy-3-(2'-cyclopentyl- phenoxy)-propan (Substanz A) mit I-Isopropylamino-2-hydroxy-3- i-naphthoxy)- propan (Substanz B) am narkotisierten Hund in vivo folgende Ergebnisse erhalten:

Die linksdrehenden optisch aktiven Isomeren der Verfahrensprodukte sind den entsprechenden racemi- schen Verbindungen noch d deutlich überlegen.
Auch die entsprechendem Cyclopentenylverbindun- gen sind starke Antagonisten der /l'-Receptoren des ad renergischen Nervensystems, die sich in Wirkungs stärke und -dauen praktisch nicht von den entsprechen den Cyclopentylve@rbind@ungen unterscheiden.
Die können daher als Heil mittel in der Huraantherapie bei Herz- und Kreislauf- erkrankungen, z. B. gegen Angina pectoris und Herz- arrythmie verwendet werden. Sie sind auch zur Ver- hinderung einer Übersteigerung der Herztätigkeit ge <U>eignet.</U>
y Für Injektionszwecke kommt etwa eine Dosifrung von 2-20 mg in Betracht, während die p;rorale Dosie rung etwa zwischen 5 und 150 mg liegt; eine einzelne Tablette oder ein Dragee kann etwa 2-50 mg Wirkstoff enthalten.
Die Verfahrensprodukte können in Form der freien Basen oder deren Sassen oral in Form von Tabletten oder Dragees, gegebenenfalls vermischt mit pharma zeutisch üblichen Trägerstoffen und/oder Stabilisatoren oder parenteral in Form von Lösungen in Ampullen verabreicht werden. Als Trägerstoffe für Tabletten kom men beispielsweise Milchzucker, Stärke, Tragamt und/ oder Magnesiumstearat in Frage.
<I>Beispiel 1</I> 6,06 g (0,02 Mol) 3-Isopropyl-5-(2'-cyclopentyl- phenoxymethyl)-oxazolidion-(2), Fp. 106-107 C, [her gestellt durch Cyclisierung von I-(N-Car#bo@m ethoxy-N-is,opropyl,a m#ino)-2- hydroxy-3-(2'-cycliopen1,yl-Ixhenoxy)-propau] werden mit 5 g fein gepulverter NaOH vermischt und 10 Min.
auf 150 C erhitzt. Das erkaltete Reaktions gemisch wird in Wasser aufgenommen, mit verdünnter Salzsäure angesäuert und mit Äther extrahiert. Die wässerige Schicht wird mit verdünnter Natronlauge al kalisch gestellt und ausgeür_hert. Nach Zugabe von methanolischer Salzsäure zu dem getrockneten Äther extrakt fällt das I-Isopropylaimino-2-hydroxy-3-(2'-cyclopentyl- phenoxy)-.propan-hydrochlorid kristallin aus, das bei 139-140 C schmilzt.
Beispiel <I>2</I> 4 g (-)-I-Isopropylamino-2-hydroxy-3-[2'-(4:2-cy- clopentenyl)-phenoxy]-propan-4hydmoch#lorid vom Fp. I04-I05 C und einem Drehwert von [a] D : 44,4 (c = 1,0; Methanol) werden in Methanol mit Palla- diummoh r bis zur Aufnahme der theoretisch berechne ten Menge Wasserstoff hydriert. Nach dem Abdamp fen des Lösungsmittels verbleibt,als Rückstand (-)-1-Isopropylamino-2-hydroxy-3-(2'-cyclopentyl- phenoxy)-propan-hydrochlorid, das nach dem Umkristallisieren aus Methanol/Äther einen Fp. von 116-117 C und einen Drehwert von [a] ri : -27,8 (c = 1,0; Methanal) hat.
<I>Beispiel 3</I> Zu einer Lösung von 15,2 g 1-Isopropylamino-2- hydroxy-3-(2'-cyclopentylphenoxy)-propan in 75 ml Essigester fügt man eine Lösung von 8,25 g D-(-)- Mandelsäure in 75 ml Essigester und saugt nach ein tägigem Stehen die ausgeschiedenen Kristalle ab. Man erhält 10,9 g (-)-1-isopropylamino-2 -hydroxy-3-(2'- cyclopentylphenoxy)-propan-D-(-)-mandelat, das nach dem Umkristallisierm einen Fp. von 133-13-4 C hat und einen Drehwart von [a] ri - 53,2 (c = 1; Me thanol) zeigt.
Durch Behandeln mit Natronlauge er hältman aus diesem Salz :das (-)-1-Isopropylamino-2-hydroxy-3- (2'-cyclopentyliphenoxy)-propan, dessen Hydrochlorid nach dem Umkristallisieren aus Äthanol/Äther bei 116-117 C schmilzt und einen Drehwert von [a]<B><I>\</I></B>D - 27,4 (c = 1; Methanol) hat.
<I>Beispiel 4</I> Zu einer Lösung von 47,4 a 1-tert.-Butylamino-2- hydroxy-3-(2'-cyclopentylphenoxy)-propan in 90 ml Isopropanol fügt .man eine Lösung von 24,5g D=(-)- Mandelsäure in 90 ml Isopropanol und saugt nach mehrtägigem Stehen die ausgeschiedenen Kristalle ab. Man erhält 27,9 g (-)-1-tert.-Butylamino-2-hydroxy- 3-(2'-cyclopentylphenoxy)-propan-D-(-)-mandelat, das nach dem Umkristallisieren aus Isopropanol einen Fp. von 133-134'C hat und einen Drehwert von [a] D - 5'0,7 (c = 1; Methanol) besitzt.
Durch Behandeln mit verdünnter Natronlauge er hält man aus diesem Salz das (-)-1-tert.-Butylamino-2- hydroxy-3-(2'-cyclopentylphenoxy)-proran, dessen Hy- drochlorid nach dem Umkristallisieren aus Methanol/ Äther bei 92-9.3' C schmilzt und einen Drehwert von [a] ö -24,6 (c = 1; Methanol) zeigt.
<I>Beispiel 5</I> Zu einer Lösung von 24 g 1-Isopropylamino-2-hydroxy-3- [2'-(J'- cyclopentenyl)-phenoxy]-propan in 100 ml Isoprapanol fügt man eine Lösung von 9,6 g D -(-)-Mandelsäure in 100 m,1 Isopropanol und saugt nach eintägigem, Stehen die ausgeschiedenen Kri stalle ab. Man erhält 16,2 g (-)--1-Isoprapylamino-2.-hydroxy-3 [2'-(de-cyclopentenyl)-phenoxy]-propan-D- (-)-mandelat, das nach dem Umkristallisieren aus Isopropanol einen Fp. von 135-136 C und einen Drehwert [.x] D =- 62,5 (c = 1; Methanol) zeigt.
Durch Behandeln mit verdünnter Natronlauge und Äther erhält man aus diesem =Salz das (-)-1-Isopropylaminio-2-hydroxy-3-[2'- (_2-cyclopentenyl)-phenoxy]-propan vorm Fp. 40-42 C (aus n-Heptan). [a] n = - 23,1 (c=l; Methanol). Die Base liefert ein Hydrochlorid vom Fp. 104-105 C (aus Äthanol/Äther.
