Verfahren zur Herstellung neuer Chinazolinone Die Erfindung betrifft ein Verfahren zur Herstel lung neuer Chinazolinone der Formel 1, worin R, für Wasserstoff oder Halogen steht und R2 Wasserstoff oder niederes Alkyl mit 1-4 Kohlenstoffatomen bedeu tet. Das erfindungsgemässe Verfahren ist dadurch ge kennzeichnet, dass man entweder Verbindungen der Formel II, worin R" R2 und Hai obige Bedeutung be sitzen, oder Verbindungen der Formel III, worin R, und R_ obige Bedeutung besitzen, oxydiert.
Die Umsetzung erfolgt zweckmässigerweise in einem inerten organischen Lösungsmittel, beispiels weise gereinigtem Dioxan oder Aceton, indem man die entsprechender. Lösungen bei Raumtemperatur mit Natrium- oder Kaliumpermanganat, zweckmässiger- weise in Form wässriger Lösungen, versetzt. Das über schüssige Oxydationsmittel wird auf an sich bekannte Weise, beispielsweise durch Zusatz von technischem Dioxan und/oder Ameisensäure und/oder NatriumNa- triumthiosulfat, zersetzt, wobei, falls man von Jodiden der Verbindungen der Formel II ausgeht, das bei der Oxydation gegebenenfalls entstandene Jod gleichzeitig reduziert wird.
Die erhaltenen Verbindungen werden in an sich bekannter Weise isoliert und gereinigt.
Die Ausgangsprodukte der Formel<B>111</B> sind neu. Sie lassen sich herstellen, indem man Verbindungen der Formel II, worin R" R2 und Hal obige Bedeutung be sitzen, reduziert.
Die Reduktion wird vorzugsweise in einem inerten organischen Lösungsmittel, beispielsweise einem niede ren Alkohol, wie Methanol oder Äthanol, oder einem Gemisch aus niederen Alkoholen und anderen inerten organischen Lösungsmitteln, wie Methylenchlorid, Chloroform oder Wasser, bei Temperaturen zwischen 20 und 80 C, vorzugsweise bei Zimmertemperatur, durchgeführt. Als Reduktionsmittel eignen sich insbe- sondere Borhydride, beispielsweise Natriumborhydrid. Nach Beendigung der Reduktion wird der Überschuss an Reduktionsmittel auf an sich bekannte Weise zer setzt. Die erhaltenen Verbindungen werden in bekann ter Weise isoliert und gereinigt.
Die Ausgangsprodukte der Formel II sind ebenfalls neu. Sie lassen sich herstellen durch Umsetzung von Verbindungen der Formel IV, worin R, und R$ obige Bedeutung besitzen, mit Verbindungen der Formel V, worin Hal obige Bedeutung besitzt.
Die Umsetzung wird vorzugsweise entweder in einem inerten organischen Lösungsmittel, beispiels weise Tetrahydrofuran, Dioxan, Chloroform oder Ben zol, oder in einem Überschuss an Verbindungen der Formel V durchgeführt. Die Reaktionstemperatur soll zwischen Raumtemperatur (20 C) und 45 C betra gen. Vorzugsweise arbeitet man jedoch bei Siedetempe ratur des Reaktionsgemisches, falls diese 45 C nicht übersteigt. Wird jedoch ein bei Raumtemperatur gas förmiges Methylhalogenid verwendet, empfiehlt sich die Verwendung eines inerten organischen Lösungsmit tels. Die gewünschten Verbindungen werden in an sich bekannter Weise isoliert und gereinigt.
Die Ausgangsprodukte der Formel IV sind neu. Sie werden hergestellt, indem man Verbindungen der For mel VI, worin R, und R= obige Bedeutung besitzen, oxydiert.
Die Oxydation wird vorzugsweise in einem inerten organischen Lösungsmittel, beispielsweise Dioxan oder Aceton, bei Raumtemperatur (20 C) unter Verwen dung von Kalium- oder Natriumpermanganat, vorzugs weise in wässriger Lösung, durchgeführt. Als Oxyda tionsmittel kann ferner Kalium- oder Natriumferricya- nid verwendet werden.
Die Ausgangsprodukte der Formel VI sind eben falls neu. Sie können hergestellt werden, indem man Verbinduncren der Formel VII, worin R, obige Bedeu- tung besitzt, mit Verbindungen der Formel VIII, worin R2 obige Bedeutung besitzt und P für ein Alkali oder für MgHal steht, worin Hal obige Bedeutung besitzt, umsetzt und die Umsetzungsprodukte hydrolysiert. Als Verbindungen der Formel VIII verwendet man vor zugsweise Lithium-, Natrium- oder Magnesiumbromide oder -jodide.
Die Umsetzung erfolgt in Gegenwart eines inerten organischen Lösungsmittels, beispiels weise Diäthyläther oder Tetrahydrofuran, bei Tempe raturen zwischen Raumtemperatur (20 C) und -40 C (abhängig von der Stabilität des Thiophen-Derivates).
Die Verbindungen der Formeln VII und VIII sind entweder bekannt oder können aus bekannten Aus gangsprodukten nach an sich bekannten Verfahren hergestellt werden. Die erfindungsgemäss hergestellten Verbindungen stellen wertvolle Pharmazeutika dar und zeichnen sich insbesondere durch entzündungshemmende, antipyreti- sche und analgetische Wirkungen aus. Die täglich zu verabreichende Dosis beträgt zwischen 3 und 40 mg pro kg Körpergewicht, vorzugsweise verabreicht in mehreren (2-4) Einzeldosen, oder in Retardform. Für die meisten grösseren Säugetiere ist eine Dosis zwi schen 400 und 800 mg pro Tag genügend. Für kleinere Haustiere genügen Mengen von 200-400 mg täglich.
Die Applikationsformen für interne Verabreichung sol len zwischen 100 und 200 mg an Wirkstoff enthalten.
Die erfindungsgemässen Verbindungen können als Arzneimittel allein oder in entsprechenden Arzneifor men für orale oder parenterale Verabreichung verwen det werden. Zwecks Herstellung geeigneter Arzneifor men werden sie mit anorganischen oder organischen, pharmakologisch indifferenten Hilfsstoffen verarbeitet. Als Hilfsstoffe verwendet man für Tabletten und Dra- gèes beispielsweise Milchzucker, Stärke, Talk oder. Stearinsäure, für Sirupe z. B. Rohrzucker-, Invertzuk- ker- oder Glucoselösungen und für Injektionspräparate beispielsweise Wasser, Alkohole, Glycerin oder pflanz liche Öle.
Zudem können die Zubereitungen geeignete Konservierungs-, Stabilisierungs-, Netzmittel, Lösungs vermittler, Süss- und Farbstoffe oder Aromantien ent halten. Eine geeignete Tablette enthält beispielsweise folgende Bestandteile: 1-3 % Bindemittel (z. B. Tragacanth), 3-10 % Stärke, 2-10 % Talk, 0,25-1 % Magnesiumstearat, entsprechende Menge an Wirksubstanzen und ad 100 /o Füllsubstanz, z.

<I>Beispiel 1</I> I-Methyl-4-(2-thienyl)-1H-chinazolinon-(2) a) 4-(2-Thienyl)-3,4-dihydrochinazolin Ein Gemisch von 8,4 g frisch destilliertem Thio phen in 100 ml Diäthyläther und 68 ml 1,6 N Butylli- thiumlösung in Hexan wird während 4 Stunden am Rückfluss erhitzt und anschliessend auf -30 C abge kühlt. Dem gekühlten Gemisch, das Thiophen-Lithium enthält, wird während 5 bis 10 Minuten tropfenweise unter Rühren eine Lösung von 13 g Chinazolin in 100 ml Diäthyläther zugegeben.
Danach wird während 1'/2 Stunden gerührt, wobei die Reaktionstemperatur auf 0 C ansteigt, und anschliessend 200 ml eiskaltes Wasser hinzugefügt. Die organische Phase wird abge trennt, die wässrige Phase zweimal mit je 500 ml Methylenchlorid extrahiert, die vereinigten organischen Phasen über wasserfreiem Natriumsulfat getrocknet und im Vakuum zur Trockne eingedampft. Nach Um kristallisieren aus Äthylacetat erhält man 4-(2-Thie- nyl)-3.4-dihydrochinazolin vom Smp. 169-171 C.
b) 4-(2-Thienyl)-chinazolin Zu einer Lösung von 7,94 g 4-(2-Thienyl)-3,4- dihydrochinazolin in 400 ml trockenem Dioxan werden tropfenweise unter Rühren und bei Raumtemperatur 81 ml einer wässrigen Kaliumpermanganatlösung (5,27 g auf 100 ml) gegeben. Anschliessend werden dem Gemisch zur Zersetzung überschüssigen Perman ganats einige Tropfen Ameisensäure zugefügt, danach filtriert und das Filtrat im Vakuum bis fast zur Trockne eingedampft. Der so erhaltene Rückstand wird zu einem Gemisch von 200 ml Methylenchlorid und 200 ml Wasser gegeben. Die organische Phase wird abgetrennt, über wasserfreiem Natriumsulfat ge trocknet und eingedampft. Nach Umkristallisieren des Rückstandes aus niedrig-siedendem Petroläther erhält man 4-(2-Thieiiyl)-chinazolin vom Smp. 65-66 C.
c) 1-Methyl-4-(2-tliienyl)-chinazolinium-jodid Eine Lösung von 37 g 4-(2-Thienyl)-chinazolin in 40 ml Methyljodid wird während 15 Stunden am Rückfluss erhitzt und anschliessend mit 100 ml Äthyla- cetat verdünnt. Das hierbei ausgefallene 1-Methyl- 4-(2-thienyl)-chinazolinium-jodid wird abfiltriert. Die Verbindung schmilzt bei 210-220 C.
d) 1-Methyl-4-(2-thienyl)-1,2,3,4-tetrahydrochina- zolin Zu einer Lösung von 55 g 1-Methyl-4-(2-thie- nyl)-chinazoliniumjodid in 1000 ml Äthanol und 200 ml Methylenchlorid werden während 5 Minuten 20g Natriumborhydrid in kleinen Portionen gegeben. Das erhaltene Gemisch wird während I/2 Stunde ge rührt und anschliessend durch vorsichtige Zugabe von 200 ml 2 N Salzsäure angesäuert. Danach wird im Vakuum auf 150 ml eingedampft und 200 ml einer 2 N wässrigen Natriumhydroxidlösung hinzugefügt.
Die wässrige Phase wird dreimal mit je 150 ml Methylen- chlorid extrahiert, die vereinigten Methylenchloridex- trakte über wasserfreiem Natriumsulfat getrocknet, fil triert und im Vakuum eingedampft, wobei 1-Methyl- 4-(2-thienyl)-1,2,3,4-tetrahydrochinazolin als gelbes Öl erhalten wird.
e) 1-Methyl-4-(2-thienyl)-1H-chinazolinon-(2) Zu einer Lösung von 33 g 1-Methyl-4-(2-thie- nyl)-1,2,3,4-tetrahydrochinazolin in 1600 ml trocke nem Dioxan werden tropfenweise unter Rühren und bei Raumtemperatur 800 ml einer wässrigen Kalium permanganatlösung (52,7 g/1000 ml) gegeben. An- schliessend werden dem Gemisch zur Zersetzung über schüssigen Permanganats einige Tropfen Ameisensäure zugefügt, danach filtriert und das Filtrat im Vakuum auf 200 ml eingedampft. Nach Zugabe von 300 ml Wasser wird der sich dabei gebildete kristalline Nieder schlag filtriert. Das hierbei erhaltene 1-Methyl- 4-(2-thienyl)-1H-chinazolinon-(2) schmilzt bei 149-150 C.
<I>Beispiel 2</I> 6-Chlor-I -methyl-4-(2-thienyl)-1 H-chinazolinon- a) 6-ChIor-4-(2-thienyl) )3,4-dihydrochinazolin Ein Gemisch von 10 ml Thiophen in 150 ml Diäthyläther und 86 ml einer 1,6 molaren n-Butyllithi- umlösung in Hexan wird während 4 Stunden am Rück- fluss erhitzt. Die erhaltene Lösung, die Thiophen- Lithium enthält. wird auf 0 C gekühlt und anschlies- send tropfenweise unter Rühren eine Lösung von 20 g 6-Chlorchinazolin in 250 ml Benzol zugegeben.
Das er haltene Gemisch wird während einer Stunde bei Raum temperatur gerührt und dann mit 500 ml Wasser ver setzt. Die organische Phase wird abgetrennt und die wässrige Phase zweimal mit je 100 ml Methylenchlorid extrahiert. Die vereinigten organischen Phasen werden über wasserfreiem Natriumsulfat getrocknet, filtriert und das Filtrat im Vakuum zur Trockne eingedampft. Nach Umkristallisieren des Rückstandes aus Äthylace- tat erhält man reines 6-Chfor-4-(2-thienyl)-3,4-dihy- drochinazolin vom Smp. 160-l61 C.
b) 6-Chlor-4-(2-thienyl)chinazolin Zu einer Lösung von 13 g 6-Chlor-4-(2-thienyl)- 3,4-dihydrochinazolin in 500 ml Dioxan werden rop- fenweise unter Rühren und bei Raumtemperatur 116 ml einer wässrigen Kaliumpermanganatlösung (5,27 g auf<B>100</B> ml)<B>100</B> ml) gegeben. Anschliessend werden dem Gemisch zur Zersetzung überschüssigen Permanganats einige Tropfen Ameisensäure zugefügt, danach filtriert und das Filtrat im Vakuum fast zur Trockne eingedampft. Der so erhaltene Rückstand wird zu einem Gemisch von 200 ml Methylenchlorid und 200 ml Wasser gegeben.
Die organische Phase wird abgetrennt, über wasserfreiem Natriumsulfat ge trocknet und abgedampft. Nach Umkristallisieren des Rückstandes aus Diäthyläther erhält man reines 6-Chlor-4-(2-thienyl)-chinazolin vom Smp. 125-126 C.
c) 6-Chlor-l-methyl-4-(2-thienyl)-chinazolinium- jodid Eine Lösung von 11,5 g 6-Chlor-4-(2-thienyl)-chi- nazolin in 30 ml Methyljodid wird während 24 Stun den am Rückfluss erhitzt und anschliessend mit 100 ml Äthylacetat verdünnt. Das hierbei ausgefallene 6-Chlor- 1-methyl-4-(2-thienyl)-chinazolinium-jodid wird abfil triert.
d) 6-Chlor-l-methyl-4-(2-thienyl)-1,2,3,4-tetrahy- drochinazolin Zu einer Lösung von 6,1 g 6-Chlor-I-methyl- 4-(2-thienyl)-chinazolinium-jodid in 100 ml Äthanol und 100 ml Methylenchlorid werden während 5 Minu ten in kleinen Portionen 3 g Natriumborhydrid gege ben. Das erhaltene Gemisch wird während 1/2 Stunde bei Raumtemperatur gerührt und anschliessend durch vorsichtige Zugabe von 200 ml 2 N Salzsäure angesäu ert. Danach wird im Vakuum auf 150 ml eingedampft und 200 ml einer 2 N wässrigen Natriumhydroxidlö- sung hinzugefügt.
Die wässrige Phase wird dreimal mit je 100 ml Methylenchlorid extrahiert, die vereinigten Methylenchloridextrakte über wasserfreiem Natrium sulfat getrocknet, filtriert und im Vakuum einge dampft, wobei das 6-Chlor-l-methyl-4-(2-thienyl)- 1,2,3,4-tetrahydro-chinazolin als gelbes 0l zurück bleibt.
e) 6-Chlor-l-methyl-4-(2-thienyl)-IH-chinazolinon- (2) Zu einer Lösung von 3,5g 6-Chlor-l-methyl- 4-(2-thienyl)-1,2,3,4-tetrahydro-chinazolin in 200 ml trockenem Dioxan werden tropfenweise unter Rühren und bei Raumtemperatur 75m1 einer wässrigen Kali umpermanganatlösung (5,27g/100m1) gegeben. An- schliessend werden dem Gemisch zur Zersetzung über- schüssi2en Permanganats einige Tropfen Ameisensäure zugefügt, danach filtriert und das Filtrat im Vakuum auf 50 ml eingedampft.
Nach Zugabe von 50 ml Was ser wird der dabei entstandene feste Niederschlag abfil triert und aus Äthylacetat umkristallisiert. Das so erhal tene 6-Chlor-l-methyl-4-(2-thienyl)-1H-chinazoli- non-(2) schmilzt bei 208 C. <I>Beispiel 3</I> 1-Methyl-4-(5-methyl-2-thienyl)-1 H-chinazolinon-(2) a) 4-(5-Methyl-2-thienyl)-3,4-dihydrochinazolin Ein Gemisch von 7,5 g 2-Methylthiophen in 150 ml Diäthyläther und 53 ml 1,6N Butyllithiumlö- sung in Hexan wird während 4 Stunden am Rückfluss erhitzt und dann auf -40 C abgekühlt. Dem gekühlten Gemisch, das 2-Methylthiophen-Lithium enthält, wird während 5 bis 10 Minuten tropfenweise unter Rühren eine Lösung von 10 g Chinazolin in 50 ml Diäthyläther hinzugefügt.
Anschliessend wird das Gemisch noch während 11/z Stunden gerührt, wobei die Reaktions temperatur auf 0 C ansteigt und dann 200 ml eiskaltes Wasser zugegeben. Die organische Phase wird abge trennt, die wässrige Phase zweimal mit je 100 ml Methylenchlorid extrahiert, die vereinigten organischen Phasen über wasserfreiem Natriumsulfat getrocknet und dann im Vakuum zur Trockne eingedampft. Nach Umkristallisieren des Rückstandes aus Äthylacetat er hält man reines 4-(5-Methyl-2-thienyl)-3,4-dihydro-chi- nazolin vom Smp. 140-141' C.
b) 4-(5-Methyl-2-thienyl)-chinazolin Zu einer Lösung von 10,6 g 4-(5-Methyl-2-thienyl)- 3,4-dihydro-chinazolin in 400 ml trockenem Dioxan werden tropfenweise unter Rühren und bei Raumtem peratur 102 ml einer wässrigen Kaliumpermanganatlö sung (5,27 g!100 ml) gegeben. Anschliessend werden dem Gemisch zur Zersetzung überschüssigen Perman ganats einige Tropfen Ameisensäure hinzugefügt, danach filtriert und das Filtrat im Vakuum fast zur Trockne eingedampft.
Der so erhaltene Rückstand wird zu einem Gemisch von 200 ml Methylenchlorid und 200 ml Wasser gegeben, die organische Phase ab getrennt, über wasserfreiem Natriumsulfat getrocknet und eingedampft, wobei 4-(5-Methyl-2-thienyl)-china- zolin als gelbes 01 zurückbleibt, c) 1-Methyl-4-(5-methyl-2-thienyl)-chinazolinium- jodid Eine Lösung von 9,3 g 4-(5-Methyl-2-thienyl)-chi- nazolin in 20 ml methyljodid wird während 16 Stunden am Rückfluss erhitzt.
Das überschüssige Methyljodid wird abgedampft, der Rückstand mit 100 ml Aceton am Rückfluss erhitzt und der ausfallende feste Nieder schlag abfiltriert, wobei 1-Methyl-4-(5-methyl-2-thie- nyl)¯chinazolinium-jodid erhalten wird. d) 1-Methyl-4-(5-methyl-2-thienyl)-1,2,3,4- tetrahydro-chinazolin Zu einer Lösung von 14,6 g 1-Methyl-4-(5-methyl- 2-thienyl)-chinazolinium-jodid in 200 ml Äthanol und 200 ml Methylenchlorid werden während 5 Minuten 7 g Natriumborhydrid in kleinen Portionen gegeben.
Das erhaltene Gemisch wird während einer halben Stunde bei Raumtemperatur gerührt und anschliessend durch vorsichtige Zugabe von 200 ml 2 N Salzsäure angesäuert. Danach wird im Vakuum auf 150 ml ein gedampft und 200 ml einer 2 N wässrigen Natriumhy- droxidlösung hinzugefügt. Die wässrige Phase wird dreimal mit je 100 ml Methylenchlorid extrahiert, die vereinigten Methylenchloridextrakte über wasserfreiem Natriumsulfat getrocknet, filtriert und im Vakuum ein gedampft, wobei 1-Methyl-4-(5-methyl-2-thienyl)- 1,2,3,4-tetrahydrochinazolin als gelbes 01 erhalten wird.
e) 1-Methyl-4-(5-methyl-2-thienyl)-IH-chinazoli- non-(2) Zu einer Lösung von 10g 1-Methyl-4-(5-methyl- 2-thienyl)-1,2,3,4-tetrahydrochinazolin in 400 ml trok- kenem Dioxan werden tropfenweise unter Rühren und bei Raumtemperatur 180 ml einer wässrigen Kalium permanganatlösung (5,27 g/100 ml) gegeben. An- schliessend werden dem Gemisch zur Zersetzung über schüssigen Permanganats einige Tropfen Ameisensäure hinzugefügt. danach filtriert und das Filtrat im Vakuum auf 50 ml eingedampft.
Dem so erhaltenen Material werden 50 ml Wasser zugesetzt. Der hierbei ausfallende kristalline Niederschlag wird abfiltriert und aus Äthylacetat Diäthyläther (1:1) umkristallisiert, wobei reines 1-Methyl-4-(5-methyl-2-thienyl)-1H-chi- nazolinon-(2) vom Smp. l04-106 C erhalten wird.
<I>Beispiel 4</I> 1-Methyl-4-(4-methyl-2-thienyl)-1H-chinazolinon- (2) a) 4-(4-Methyl-2-thienyl)-3,4-dihydrochinazolin Ein Gemisch von 7,5 g 3-Methylthiophen in 150 ml Diäthyläther und 53 ml einer 1,6 N Butyllithi- umlösung in Hexan wird während 4 Stunden am Rück- fluss erhitzt und dann auf -40 abgekühlt. Dem ge kühlten Gemisch, das 3-Methylthiophen-Lithium ent hält, wird während 5 bis 10 Minuten tropfenweise un ter Rühren eine Lösung von 10 g Chinazolin in 50 ml Diäthyläther zugegeben.
Anschliessend wird das Ge misch während 1'/a Stunden gerührt, wobei die Reak tionstemperatur auf 0 C ansteigt und danach 200 ml eiskaltes Wasser zugesetzt. Die organische Phase wird abgetrennt, die wässrige Phase zweimal mit je 100 ml Methylenchlorid extrahiert, die vereinigten organischen Phasen über wasserfreiem Natriumsulfat getrocknet und im Vakuum zur Trockne eingedampft, wobei das 4-(4-Methyl-2-thienyl)-3,4-dihydrochinazolin als gelbes 01 erhalten wird. Bimaleinat Durch Behandlung einer acetonischen Lösung der oben erhaltenen Base mit Maleinsäure wird das 4-(4-Methyl-2-thienyl)-3,4-dihydrochinazolin bimalei- nat vom Smp. 189-193 C gewonnen.
b) 4-(4-Methyl-2-thienyl)-chinazolin Zu einer Lösung von 15,2 g 4-(4-Methyl-2-thienyl- )-3,4-dihydrochinazolin in 500 ml trockenem Dioxan werden tropfenweise unter Rühren und bei Raumtem peratur 135 ml einer wässrigen Kaliumpermanganatlö sung (5,27 g/100 ml) zugegeben. Anschliessend werden dem Gemisch zur Zersetzung überschüssigen Perman ganats einige Tropfen Ameisensäure hinzugefügt, danach filtriert und das Filtrat im Vakuum fast zur Trockne eingedampft. Der so erhaltene Rückstand wird zu einem Gemisch von 200 ml Methylenchlorid und 200 ml Wasser gegeben, die organische Phase ab getrennt, über wasserfreiem Natriumsulfat getrocknet und eingedampft.
Nach Umkristallisieren des Rück standes aus Äthylacetat erhält man das reine 4-(4- Methyl-2-thienyl)-chinazolin vom Smp. 84-86 C, c) 1-Methyl-4-(4-methyl-2-thienyl)-chinazolinium- jodid Eine Lösung von 11 g 4-(4-Methyl-2-thienyl)-chi- nazolin in 20 ml Methyljodid wird während 16 Stun den am Rückfuss erhitzt. Das überschüssige Methyljo- did wird abgedampft und der dabei erhaltene Rück stand mit 100 ml Methanol am Rückfluss erhitzt. Der hierbei ausfallende feste Niederschlag wird abfiltriert.
Das so erhaltene 1-Methyl-4-(4-methyl-2-thienyl)-chi- nazolinium-jodid besitzt einen unbestimmten Schmelz punkt.
