Verfahren zur Herstellung von neuen Pyrrolidinderivaten
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Pyrrolidin-Derivaten, mit wertvollen pharmakologischen Eigenschaften.
Es wurde überraschenderweise gefunden, dass Verbindungen der Formel I,

Sie zeigen spasmolytische, cardiovas kuläre, hypotensive/anti-hypertensive und/oder antise- kretorische Eigenschaften und können deshalb als Spasmolytika, als hypotensiv/anti-hypertendve und antisekretorische Mittel in geeigneten Dosierungseinheiten zusammen mit geeigneten pharmazeutischen Trägern intravenös oder oral verabreicht werden. Beim Rest R handelt es sich insbesondere z. B. um den Methyl- oder Äthylrest, während der Rest Am vorzugsweise insgesamt höchstens 10 Kohlenstoffatome enthält.

Zu Verbindungen der Formel II gelangt man beispielsweise durch Behandlung von Dialkyl-oder Diaralkylestern von Acylamino-(ss-3,4-dimethoxybenzoyl äthyl)-malonsäuren mit einer Mineralsäur,e wobei Hydrolyse, Cyclisierung und Decarboxylierung zu 2-(3 ,4-Dimethoxyphenyl)-pyrrolin-5-carbonsäure eintritt, welche anschliessend in die entsprechenden Ester umgewandelt werden kann. Diese Carbonsäure und ihre Ester können zunächst in der l-Position alkyliert werden und die entsprechenden Pyrroliniumderivate können durch Hydrierung in die 1-Alkylpyrrolidinderi- vate überführt werden.
Ester von Acylamino-(ss-3 4-dimethoxybenzoyläthyl)- malonsäuren lassen sich beispielsweise aus dem entsprechend substituierten Acetophenon durch Umsetzung mit Paraformaldehyd und Dimethylarain-hydro- chlorid und nachfolgende Kondensation mit Dialkyloder Diaralkylestern der Acylaminomalonsäure herstellen.
Die Umwandlung der 2-(3,4-Dimethyoxyphenyl)pyrrolin-5-carbonsäure und deren Ester in die entspre chende Pyrrolidin-2-carbonsäure und deren Ester kann durch katalytische Hydrierung vorgenomment werden, beispielsweise in einer Parr-Apparatur, in Gegenwart eines Edelmetallkatalysators wie z. B. Platinoxyd oder anderer äquivalenter Metalle wie Palladium, Rhodium, Nickel etc., bei Raumtemperatur oder mässig erhöhter Temperaturen und bei Atmosphärendruck oder mässig erhöhten Drücken oder durch eine chemische Reduktion, z. B. mit einem Borhydrid.
Die vorliegende Erfindung umfasst nicht nur die oben beschriebenen Pyrrolidinderivate in der Form der freien Base, sondern schliesst pharmazeutisch verträgliche Salze ein, welche aus den erfindungsgemässen Verbindungen gemäss dem üblichen Verfahren gewonnen werden. Mit anorganischen oder organischen Säuren, wie Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Athandisulfonsäure, Essigsäure, Bemsteinsäure, Fumarsäure, Maleinsäure, Äpfelsäure, Weinsäure, Citronensäure, Benzoesäure, Salicylsäure und Mandelsäure bilden die Verbindungen der Formel I Salze, welche zum Teil wasserlöslich sind.
Die nachfolgenden Beispiele erläutern die Herstellung der neuen Verbindungen näher. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1 l-Mef : hyl-543, 4-Dimethoxyphenyl)-pyrrolidin-2-car- bonsäureamid Hydrochlorid.
(a) 1-Methyl-5-(3,4-dimethoxyphenyl)pyrrolidin-2carbonsäure Methylester Hydrochlorid, Smp. 182-183 (12,6 g, 0,04 Mol) werden in 50 ml Wasser gelöst.
Diese Lösung wird mit gesättigter Natriumcarbonatlösung alkalisch gestellt bis pH-Wert 9 und mit 3 mal 75 mol Chloroform extrahiert. Das Chloroformextrakt wird gewaschen und über Natriumsulfat getrocknet und zur Trockne eingedampft. Der ölige Rückstand ergibt 11 g, wird in Methanol (100 ml) gelöst, die Lösung im Eisbad mit Ammoniak gesättigt und in einem Druckgefäss bei Raumtemperatur fünf Tage lang stehengelassen. Sodann wird die Lösung zur Trockne eingedampft und der erhaltene Rückstand aus 50 ml Isopropanol umkristallisiert. Ausbeute 10, 4 g, Smp.
135-140 . Ein Teil dieses Produkts (5,3 g, 0,02 Mol) wird in 20 mol Methanol gelöst und 3 ml ätherischer 9,9-n Salzsäure beigegeben. Beim Stehenlassen kristal ligiert das Hydrochlorid aus (5,1 g, Smp. 228-230 Zers.). Umkristallisieren aus Methanol (20 ml) erhöhte den Smp. auf 231-233". (Zers.).
Das Ausgangsmaterial wurde wie folgt hergestellt: b) Zu 3-Dimethylamino-3',4'-dimethoxypropiphenon Hydrochlorid vom Smp. 180-182e (49,2 g, 0,18 Mol) in 150 ml wasserfreiem Äthanol wird unter Rühren 27,3 g (0,22 Mol) Dimethylsulfat tropfenweise bei Raumtemperatur gegeben. Nach drei Stunden werden 35, 4 g (0,16 Mol) Acetamido-malonsäure-diäthylester suspendiert in einer Natriumäthoxydlösung (aus 0,36 Mol Natrium in 150 ml wasserfreiem Äthanol zubereitet) zugegeben (leichter Temperaturanstieg). Das Gemisch wird während einer Stunde bei Raumtemperatur belassen und anschliessend während einer Stunde unter Rückfluss gekocht.
Nach Abkühlen werden 1000 g zerhacktes Eis zugegeben, worauf der Acetamido- [3-(3,4- dimethoxyphenyl)-3-oxopropylj-maloissäure - diäthylester in 89 Olo Ausbeute auskristallisiert (59,5 g, Smp. 1061080). Umkristallisieren aus Isopropanol erhöht den Smp. auf 107-109". c) Die unter b) beschriebene Verbindung (35 g, 0,086 Mol) wird in 175 ml 6-n Salzsäure suspendiert und die Suspension während drei Stunden unter Rückfluss erhitzt. Die so erhaltene Lösung wird filtriert, unter verringertem Druck zur Trockne verdampft und der Rückstand durch Destillation mit Benzol getrocknet.
Bei Zugabe von 150 ml Aceton zum Rückstand erfolgt Kristallisation (Smp. 220-223 , Zers.). Zwei- mal aus 250 ml Aceton-Wasser (4:1) umkristallisiert erhält man 16,8 g 2-(3,4-Dimethoxyphenyl) -pyrrolin5-carbonsäure-hydrochlorid, Smp. 215-216 (Zers.). d) Ein Gemisch von 34,3 g 2-(3,4-Dimethoxyphe- nyl) -pyrrolin-Searbonsäure Hydrochlorid (0,12 Mol), 200 ml absolutem Methanol und 30 ml Schwefelsäure wird während drei Stunden unter Rückfluss erhitzt.
Hierauf wird das Methanol durch Destillation unter vermindertem Druck entfernt, der Rückstand mit gesättigter Natriumcarbonatlösung auf pH 9 gestellt und mit Chloroform (3 X 100 ml) ausgezogen.
Die vereinigten Chloroformextrakte werden mit Wasser gewaschen, über Natriumsulfat getrocknet und unter vermindertem Druck zur Trockne eingedampft.
Der ölige Rückstand wird in Isopropanol (50 ml) gelöst, wobei 22,8 g Kristalle (vom Smp. 78-81 ) erhalten werden. Diese werden aus Isopropanol umkristallisiert und ergeben den 2-(3 ,4-Dimethoxyphenyl)-pyr- rolin-5-carbonsäuremethylester, Smp. 79-81". e) Eine Lösung von 2-(3 ,4-diinethoxyphenyl)-pyr- rolin-5-carbonsäure Methylester (10,5 g, 0,04 Mol) und Methyljodid (56,4 g, 0,4 Mol) in 100 ml wasserfreiem Benzol wird für fünf Stunden unter Rückfluss erhitzt. Das auskristallisierte 1-Methyl-2-(3, 4-dimeth- oxyphenyl)-5-methoxycarbonyl- pyrroliniumjodid wird filtriert und mit Benzol gewaschen. Ausbeute 12,6 g; Smp. 1641660.
Wenn in diesem Verfahren beispielsweise Athyl-, Propyl- oder Butyljodid oder -bromid gebraucht werden, so erhält man die entsprechenden 1-Athyl-, 1-Propyl- oder l-Butyl-substituierten Pyrroliniumjodide oder -bromide. f) Eine Suspension von 1-Methyl-2-(3,4-dimethoxyphenyl)-5-methoxycarbonyl-pyrroliniumjodid (37,0 g, 0,091 Mol) in 200 ml Methanol wird bei Raumtemperatur in Gegenwart eines Adams-Katalysators unter 3 Atm.
Druck während zwei Stunden hydriert. Dann wird filtriert und das Filtrat zur Trockne verdampft. Der ölige Rückstand wird in 100 ml Wasser gelöst, diese Lösung mit gesättigter Natriumcarbonatlösung auf pH 9 gestellt und mit Chloroform (3 x150 ml) extrahiert. Der Chloroformextrakt wird mit Wasser gewaschen, über Natriumsulfat getrocknet, zur Trockne eingedampft und der Rückstand in 60 mol Isopropanol aufgenommen. Nach Zusatz von 10 ml äthanolischer 10-n Salzsäurelösung kristallisiert das Hydrochlorid des 1-Methyl-5-(3,4-di- methoxyphenyl)-pyrrolidin-2-carbonsäure Methylesters, welches nach Umkristallisieren. aus Ispopropanol (150 ml) 22 g farblose Kristalle vom Smp. 182-183 (Zers.) lieferte.
Beispiel 2
1-Methyl-5-(3,4-dimethoxyphenyl)-pyrrolidin-2-carbonsäure-N-äthylamid
Eine Lösung von 1-Methyl-5-(3 4-dimethoxyphe- nyl)-pyrrolidin-2-carbonsäure Methylester (8, 4 g, 0,03 Mol) wird mit wasserfreiem Äthylamin bei 0 gesättigt und bei Raumtemperatur fünf Tage stehen gelassen.
Hierauf wird die Lösung wieder mit Ethylamin gesättigt und bei Raumtemperatur für 7 zusätzliche Tage belassen. Die Lösung wird zur Trockne verdampft, das entstandene Öl (8,3 g) in 20 ml Isopropanol gelöst, 4,5 ml ätherische 9-n. Salzsäure zugegeben, diese Lösung wie, der zur Trockne verdampft und das hygroskopische Produkt (9,5 g, Smp. 84-86 ) aus Isopropanol/Isopropyläther (1:1, 30 ml) auskristallisiert. Das so erhaltene Hydrochlorid wird in 10 mi Wasser gelöst, der pH-Wert mit gesättigter Natriumcarbonatlösung auf 9 eingestellt und das Gemisch viermal mit 40 ml Chloroform extrahiert. Hierauf wird die Chloroformlösung mit Wasser gewaschen, über Natriumsulfat getrocknet und bis zur Trockne verdampft.
Das entstandene Öl destilliert man zweimal unter Vakuum und man erhält die gewünschte Verbindung (4,2 g, Siedepunkt 1650/0,1 mm; nD24 1,5395), welche beim Stehen kristallisiert, Smp. 52-54".
Beispiel 3 l-Methyl-5-(3, 4-dimethoxyphenyl)-pyrrolidin-2sar- bonsäure-N-äthylamid
Eine Lösung von Dicyclohexyl-carbodiimid (0,05 Mol, 10,3 g) in Tetrahydrofuran wird tropfenweise einer Lösung von 1-Methyl-5-(3, 4-dimethoxyphenyl) -pyrrolidin-2-carbonsäure vom Smp. 133-135 (0,025 Mol, 6,6 g) und Äthylamin (0,05 Mol, 2,2 g) in 100 ml Tetrahydrofuran, welche in einem Eisbad auf 0 gehalten wird, zugegeben. Das Reaktionsgemisch wird bei Raumtemperatur über Nacht gerührt, dann unter reduziertem Druck zur Trockne eingedampft. Der Rückstand wird in Methylenchlorid suspendiert und filtriert.
Das Filtrat wird eingedampft und der Rückstand im Hochvakuum fraktioniert. Das gewünschte Produkt siedet bei 162-165 /0,1 mm Hg; nD24=1, 5390.
Das Ausgangsmaterial wurde, in Form seines Hydrochlorids vom Smp. 219-220 (Zers.) aus dem Methylester (vgl. Beispiel lf) durch Hydrolyse mit 3-n.
Salzsäure erhalten. Die freie Aminosäure wurde daraus mittels eines Ionenaustauschers (Dowex 3) gewonnen.
Beispiel 4
1-Methyl-5-(3,4-dimethoxyphenyl)-pyrrolidin-2-carbonsäurehydrazid
Eine Lösung von 1-Methyl-5-(3, 4-dimethoxyphe- nyl)-2-carbonsäure Methylester (5,6 g, 0,02 Mol) und Hydrazinhydrat (4 mi) wird bei Raumtemperatur während 5 Tagen gerührt. Die Lösung wird unter Vakuum zur Trockne verdampft und ergibt das gewünschte Produkt; 3,9 g, Siedepunkt 185 /0,1 mm Hg.
Beispiel 5 l-Methyl-5-(3, 4-dimethoxyphenyl)-pyrrolidin-2war- bonsäure-N-(2-anilinoäthyl)-amid.
8,37 g (0,03 Mol) 1-Methyl-5-(3,4-dimethoxyphe- nyl)-pyrrolidin-2-carbonsäure Methylester werden in 50 ml absolutem Methanol gelöst. Zu dieser Lösung werden 14, 4 g (0,09 Mol) N-Phenyläthylendiamin gegeben. Die Reaktionslösung wird darauf während 5 Tagen am Rückfluss gekocht. Dann wird das Methanol unter vermindertem Druck verdampft und der Rückstand im Hochvakuum fraktioniert. Das gewünschte Produkt destilliert als gelbes Öl bei 252-254 /0, 05 mm Hg; Ausbeute 5,6 g.
Beispiel 6 1-Methyl-5-(3, 4-dimethoxyphenyl)-pyrrolidin-2"car- bonsäure-N-(2-dimethylaminoäthyl)-amid.
8,37 g (0,03 Mol) 1-Methyl-5-(3,4-dimethoxyphenyl)-pyrrolidin-2-carbonsäure Methylester werden in 60 ml absolutem Methanol gelöst. Zu dieser Lösung gibt man 8,8 g (0,1 Mol) 2-Dimethylamino-äthylamin.
Die Reaktionslösung wird dann 5 Tage am Rückfluss gekocht, das Methanol unter vermindertem Druck verdampft und der Rückstand unter Hochvakuum destilliert. Man erhält 5,1 g eines gelben Öls, das bei 1982000 /0,05 mm Hg siedet.
Beispiel 7
1-[1-Methyl-5-(3,4-dimethoxyphenyl)-pyrrolidin-2carbonyl]-4-methyl-piperazin.
6,0 g N-Methylpiperazin werden in 120 ml getrocknetem Äther gelöst. Zu dieser Lösung gibt man 0,06 Mol n-Butyl-lithium. Das Reaktionsgemisch wird dann bei Raumtemperatur während 17 Stunden gerührt. Man kühlt dann auf 0 ab und gibt 8, 37 g (0,03 Mol) in 100 ml Äther gelösten 1-Methyl-5-(3,4- dimethoxy-phenyl)-pyrrolidin-2-carbonsäure-Methylester tropfenweise dazu. Die Zugabe dauert 30 Minuten. Dann kocht man 4 Stunden unter Rückfluss und engt das Reaktionsgemisch in Vakuum ein. Der Rückstand wird in Eiswasser aufgenommen und mit Äther extrahiert.
Die organische Phase wird abgetrennt, über Natriumsulfat getrocknet und das Lösungsmittel unter vermindertem Druck verdampft. Der Rückstand wird unter Hochvakuum destilliert und ergibt 7,0 g gelbes Öl, das bei 2082100/0,05 mm Hg siedet.
Beispiel 8 1-[1-methyl-s-(3,4-dimethoxyphenyl) -pyrrolidin-2- carbonyl]-4-phenyl-piperazin.
6, 48 g (0,04 Mol) N-Phenyl-piperazin werden in 200ml getrocknetem Äther gelöst. Zu dieser auf 0 gekühlten Lösung werden 0,04 Mol N-Butyl-lithium gegeben. Die Reaktionslösung wird bei Raustempera- tur 18 Stunden gerührt und dann auf 0 abgekühlt und eine Lösung von 5, 58 g (0,02 Mol) 1-Methyl-5-(3,4dimethoxyphenyl)-pyrrolidin-2-carbonsäure Methylester in 50 ml Äther wird tropfenweise zugegeben.
Das Zutropfen dauert 15 Minuten, dann wird die Reaktionslösung 8 Stunden am Rückfluss gekocht und schliesslich unter vermindertem Druck zur Trockne eingedampft. Zum Rückstand wird Eiswasser gegeben und die entstandene wässrige Lösung wird mit Chloroform extrahiert. Der Chloroformextrakt wird über Natriumsulfat getrocknet, filtriert und zur Trockne eingedampft. Der Rückstand wird mit Petroläther überschichtet und angerieben, wobei spontane Kristallisation erfolgt. Man kristallisiert aus Äther/Petroläther (1:1) um und erhält 1,6 g Produkt vom Smp. 84850.
Beispiel 9
1-[4-Methyl-5-(3,4-dimethoxyphenyl)-pyrrolidin-2carboxyl]-4-carbobenzoxy-piperazin.
5,6 g 1-Methyl-5-(3,4-dimethoxyphenyl)-pyrrolidin2-carbonsäure Methylester (0,02 Mol) werden in 50 ml trockenem Toluol gelöst. Zu dieser Lösung werden 17, 6 g (0,08 Mol) 4-Carbobenzoxy-piperazin gegeben.
Die Reaktionslösung wird darauf 9 Tage unter Rück fluss gekocht und schliesslich wird unter vermindertem Druck das Toluol und das 4-Carbobenzoxypiperazin verdampft. Der Rückstand wird mit Petroläther überschichtet und kristallisiert nach Stehen bei 5 aus. Smp.
65-68"; Ausbeute 3,0 g.
Beispiel 10 l-Methyl-5-(3, 4-dimethoxyphenyl)-pyrrolidin-2-carbonsäure-N-(2'0anilinoäthyl)-amid.
1,63 g (0,005 Mol) Chlorameisensäure Äthylester werden zu einer auf -10 gekühlten Mishcung von 1, 51 g (0,005 Mol) 1-Methyl-5-(3, 4-dimethoxyphe nyl)-pyrrolidion-2-carbonsäure Hydrochlorid, 1, 01 g (0,01 Mol)Triäthylamin und 15 ml Methylenchlorid gegeben. Man rührt eine Stunde bei -10 und gibt dann 1,36 g (0,01 Mol) N-Phenyläthylendiamin dazu.
Die Reaktion wird über Nacht bei Zimmertemperatur gerührt und dann zur Trockne eingedampft. Der Rückstand wird in Wasser aufgenommen, mit gesättigter Natriumcarbonatlösung bis zum pH 9-10 alkalisch gestellt und dann mit Chloroform ausgezogen. Die Chloroformextrakte werden mit Wasser gewaschen, über Natriumsulfat getrocknet und zur Trockne verdampft. Der Rückstand wird unter Hochvakuum destilliert und gibt ein ÖI, Kochpunkt 250-254 /0,05 mm Hg.
Beispiel 11 1-Methyl-5-(3,4- dimethoxyphenyl)-2 -carbonsäure- [4'-phenyl]-piperazid.
1,4 g 1-Methyl-5-(3,4-dimethoxyphenyl)-pyrrolidin2-carbonsäure-hydrazid (0,005 Mol) werden in 20 ml einer 4-n. Lösung von Salzsäure in absolutem Tetrahydrofuran gelöst. Diese Lösung wird auf 150 abgekühlt und dann eine Lösung von 0,62 g (0,006 Mol) Butylnitrit in 10 ml Tetrahydrofuran zugegeben. Die Lösung wird bei - 15 gerührt, bis vollständige Lösung eingetreten ist und dann noch weitere 5 Minuten. Dann werden 50 ml vorgekühlter Essigsäure-l2ithylester zugegeben und die Lösung verschiedentlich mit kalter gesättigter Natriumbicarbonatlösung geschüttelt, bis alle Säure neutralisiert ist.
Die organische Schicht wird dann über Natriumsulfat getrocknet, filtriert, gekühlt und bei 0 mit einer Lösung von 0,97 g N-Phenylpiperazin (0,006 Mol) in 15 ml Essigsäure-Äthylester versetzt. Die Reaktionslösung wird eine Stunde bei 0 und dann bei Raumtemperatur über Nacht gelassen. Man verdampft das Lösungsmittel unter Vakuum und erhält als Rückstand das rohe Amid (1,9 g), welches nach Umkristallisieren aus Ather/Pentan (1:1) einen Smp. von 84850 hat.

Esters of acylamino (ss-3 4-dimethoxybenzoylethyl) malonic acids can be prepared, for example, from the appropriately substituted acetophenone by reaction with paraformaldehyde and dimethylarain hydrochloride and subsequent condensation with dialkyl or diaralkyl esters of acylaminomalonic acid.
The conversion of 2- (3,4-dimethyoxyphenyl) pyrroline-5-carboxylic acid and its ester into the corresponding pyrrolidine-2-carboxylic acid and its ester can be carried out by catalytic hydrogenation, for example in a Parr apparatus, in the presence of a noble metal catalyst such as B. platinum oxide or other equivalent metals such as palladium, rhodium, nickel, etc., at room temperature or moderately elevated temperatures and at atmospheric pressure or moderately elevated pressures or by chemical reduction, e.g. B. with a borohydride.
The present invention not only includes the above-described pyrrolidine derivatives in the form of the free base, but also includes pharmaceutically acceptable salts which are obtained from the compounds according to the invention according to the usual method. With inorganic or organic acids, such as hydrochloric acid, hydrobromic acid, sulfuric acid, phosphoric acid, methanesulphonic acid, ethanedisulphonic acid, acetic acid, succinic acid, fumaric acid, maleic acid, malic acid, tartaric acid, citric acid, benzoic acid, salicylic acid and mandelic acid, the compounds of the formula I form salts, some of which are water-soluble are.
The following examples explain the preparation of the new compounds in more detail. The temperatures are given in degrees Celsius.
Example 1 l-Mef: hyl-543,4-dimethoxyphenyl) pyrrolidine-2-carboxylic acid amide hydrochloride.
(a) 1-Methyl-5- (3,4-dimethoxyphenyl) pyrrolidine-2carboxylic acid methyl ester hydrochloride, m.p. 182-183 (12.6 g, 0.04 mol) are dissolved in 50 ml of water.
This solution is made alkaline with saturated sodium carbonate solution to pH 9 and extracted with 3 times 75 mol of chloroform. The chloroform extract is washed and dried over sodium sulfate and evaporated to dryness. The oily residue yields 11 g, is dissolved in methanol (100 ml), the solution is saturated with ammonia in an ice bath and left to stand in a pressure vessel at room temperature for five days. The solution is then evaporated to dryness and the residue obtained is recrystallized from 50 ml of isopropanol. Yield 10.4 g, m.p.
135-140. A portion of this product (5.3 g, 0.02 mol) is dissolved in 20 mol of methanol and 3 ml of ethereal 9.9 N hydrochloric acid are added. The hydrochloride ligated out as crystals on standing (5.1 g, m.p. 228-230 dec.). Recrystallization from methanol (20 ml) increased the m.p. to 231-233 "(dec.).
The starting material was prepared as follows: b) To 3-dimethylamino-3 ', 4'-dimethoxypropiphenone hydrochloride of melting point 180-182e (49.2 g, 0.18 mol) in 150 ml of anhydrous ethanol, 27.3 is added with stirring g (0.22 mol) of dimethyl sulfate was added dropwise at room temperature. Recrystallization from isopropanol increases the mp to 107-109 ". C) The compound described under b) (35 g, 0.086 mol) is suspended in 175 ml of 6N hydrochloric acid and the suspension is refluxed for three hours The solution is filtered, evaporated to dryness under reduced pressure and the residue is dried by distillation with benzene.
When 150 ml of acetone are added to the residue, crystallization takes place (melting point 220-223, decomp.). Recrystallized twice from 250 ml of acetone / water (4: 1), 16.8 g of 2- (3,4-dimethoxyphenyl) pyrroline-5-carboxylic acid hydrochloride, melting point 215-216 (decomp.), Are obtained. d) A mixture of 34.3 g of 2- (3,4-dimethoxyphenyl) pyrroline-searboxylic acid hydrochloride (0.12 mol), 200 ml of absolute methanol and 30 ml of sulfuric acid is refluxed for three hours.
The methanol is then removed by distillation under reduced pressure, the residue is adjusted to pH 9 with saturated sodium carbonate solution and extracted with chloroform (3 × 100 ml).
The combined chloroform extracts are washed with water, dried over sodium sulfate and evaporated to dryness under reduced pressure.
The oily residue is dissolved in isopropanol (50 ml) to give 22.8 g of crystals (m.p. 78-81). These are recrystallized from isopropanol and give the 2- (3, 4-dimethoxyphenyl) -pyrroline-5-carboxylic acid methyl ester, melting point 79-81 ". E) A solution of 2- (3, 4-diinethoxyphenyl) -pyr- rolin-5-carboxylic acid methyl ester (10.5 g, 0.04 mol) and methyl iodide (56.4 g, 0.4 mol) in 100 ml of anhydrous benzene is heated under reflux for five hours. The 1-methyl-2 - (3,4-dimethoxyphenyl) -5-methoxycarbonyl-pyrrolinium iodide is filtered and washed with benzene, yield 12.6 g; mp 1641660.
If, for example, ethyl, propyl or butyl iodide or bromide are used in this process, the corresponding 1-ethyl, 1-propyl or 1-butyl-substituted pyrrolinium iodides or bromides are obtained. f) A suspension of 1-methyl-2- (3,4-dimethoxyphenyl) -5-methoxycarbonyl-pyrrolinium iodide (37.0 g, 0.091 mol) in 200 ml of methanol is at room temperature in the presence of an Adams catalyst under 3 atm.
Pressure hydrogenated for two hours. It is then filtered and the filtrate is evaporated to dryness. The oily residue is dissolved in 100 ml of water, this solution is adjusted to pH 9 with saturated sodium carbonate solution and extracted with chloroform (3 × 150 ml). The chloroform extract is washed with water, dried over sodium sulfate, evaporated to dryness and the residue taken up in 60 mol isopropanol. After the addition of 10 ml of ethanolic 10N hydrochloric acid solution, the hydrochloride of the 1-methyl-5- (3,4-dimethoxyphenyl) -pyrrolidine-2-carboxylic acid methyl ester crystallizes, which after recrystallization. from isopropanol (150 ml) gave 22 g of colorless crystals with a melting point of 182-183 (decomp.).
Example 2
1-methyl-5- (3,4-dimethoxyphenyl) -pyrrolidine-2-carboxylic acid-N-ethylamide
A solution of 1-methyl-5- (3 4-dimethoxyphenyl) -pyrrolidine-2-carboxylic acid methyl ester (8.4 g, 0.03 mol) is saturated with anhydrous ethylamine at 0 and left to stand at room temperature for five days.
The solution is then saturated again with ethylamine and left at room temperature for an additional 7 days. The solution is evaporated to dryness, the resulting oil (8.3 g) is dissolved in 20 ml of isopropanol, 4.5 ml of essential 9-n. Hydrochloric acid was added, this solution was like, which evaporated to dryness and the hygroscopic product (9.5 g, melting point 84-86) crystallized from isopropanol / isopropyl ether (1: 1, 30 ml). The resulting oil is distilled twice under vacuum and the desired compound is obtained (4.2 g, boiling point 1650 / 0.1 mm; nD24 1.5395), which crystallizes on standing, mp 52-54 ".
Example 3 l-Methyl-5- (3, 4-dimethoxyphenyl) -pyrrolidine-2sarboxylic acid-N-ethylamide
A solution of dicyclohexyl-carbodiimide (0.05 mol, 10.3 g) in tetrahydrofuran is added dropwise to a solution of 1-methyl-5- (3,4-dimethoxyphenyl) -pyrrolidine-2-carboxylic acid of m.p. 133-135 ( 0.025 mol, 6.6 g) and ethylamine (0.05 mol, 2.2 g) in 100 ml of tetrahydrofuran, which is kept at 0 in an ice bath, was added. The reaction mixture is stirred at room temperature overnight, then evaporated to dryness under reduced pressure. The residue is suspended in methylene chloride and filtered.
The filtrate is evaporated and the residue is fractionated in a high vacuum. The desired product boils at 162-165 / 0.1 mm Hg; nD24 = 1.5390.
The starting material, in the form of its hydrochloride with a melting point of 219-220 (decomp.), Was obtained from the methyl ester (cf. Example If) by hydrolysis with 3-n.
Hydrochloric acid obtained. The free amino acid was obtained therefrom by means of an ion exchanger (Dowex 3).
Example 4
1-methyl-5- (3,4-dimethoxyphenyl) pyrrolidine-2-carboxylic acid hydrazide
A solution of 1-methyl-5- (3,4-dimethoxyphenyl) -2-carboxylic acid methyl ester (5.6 g, 0.02 mol) and hydrazine hydrate (4 ml) is stirred at room temperature for 5 days. The solution is evaporated to dryness in vacuo to give the desired product; 3.9 g, boiling point 185 / 0.1 mm Hg.
Example 5 l-methyl-5- (3, 4-dimethoxyphenyl) pyrrolidine-2war- bonsäure-N- (2-anilinoethyl) -amide.
8.37 g (0.03 mol) of 1-methyl-5- (3,4-dimethoxyphenyl) -pyrrolidine-2-carboxylic acid methyl ester are dissolved in 50 ml of absolute methanol. 14.4 g (0.09 mol) of N-phenylethylenediamine are added to this solution. The reaction solution is then refluxed for 5 days. The methanol is then evaporated off under reduced pressure and the residue is fractionated in a high vacuum. The desired product distills as a yellow oil at 252-254/0.05 mm Hg; Yield 5.6g.
Example 6 1-Methyl-5- (3, 4-dimethoxyphenyl) -pyrrolidine-2 "carboxylic acid-N- (2-dimethylaminoethyl) -amide.
8.37 g (0.03 mol) of 1-methyl-5- (3,4-dimethoxyphenyl) -pyrrolidine-2-carboxylic acid methyl ester are dissolved in 60 ml of absolute methanol. 8.8 g (0.1 mol) of 2-dimethylamino-ethylamine are added to this solution.
The reaction solution is then refluxed for 5 days, the methanol is evaporated off under reduced pressure and the residue is distilled under high vacuum. 5.1 g of a yellow oil are obtained which boil at 1982000 / 0.05 mm Hg.
Example 7
1- [1-methyl-5- (3,4-dimethoxyphenyl) pyrrolidine-2carbonyl] -4-methyl-piperazine.
6.0 g of N-methylpiperazine are dissolved in 120 ml of dried ether. 0.06 mol of n-butyllithium are added to this solution. The reaction mixture is then stirred at room temperature for 17 hours. It is then cooled to 0 and 8.37 g (0.03 mol) of methyl 1-methyl-5- (3,4-dimethoxyphenyl) -pyrrolidine-2-carboxylate dissolved in 100 ml of ether are added dropwise. The addition takes 30 minutes. It is then refluxed for 4 hours and the reaction mixture is concentrated in vacuo. The residue is taken up in ice water and extracted with ether.
The organic phase is separated off, dried over sodium sulfate and the solvent is evaporated off under reduced pressure. The residue is distilled under high vacuum and gives 7.0 g of yellow oil which boils at 2082 100 / 0.05 mm Hg.
Example 8 1- [1-methyl-s- (3,4-dimethoxyphenyl) -pyrrolidine-2-carbonyl] -4-phenyl-piperazine.
6.48 g (0.04 mol) of N-phenyl-piperazine are dissolved in 200 ml of dried ether. 0.04 mol of N-butyl-lithium are added to this solution, which has been cooled to 0. The reaction solution is stirred at room temperature for 18 hours and then cooled to 0 and a solution of 5.58 g (0.02 mol) of 1-methyl-5- (3,4dimethoxyphenyl) pyrrolidine-2-carboxylic acid methyl ester in 50 ml Ether is added dropwise.
The dropwise addition takes 15 minutes, then the reaction solution is refluxed for 8 hours and finally evaporated to dryness under reduced pressure. Ice water is added to the residue and the resulting aqueous solution is extracted with chloroform. The chloroform extract is dried over sodium sulfate, filtered and evaporated to dryness. The residue is covered with a layer of petroleum ether and rubbed, with spontaneous crystallization. It is recrystallized from ether / petroleum ether (1: 1) and 1.6 g of product with a melting point of 84850 are obtained.
Example 9
1- [4-Methyl-5- (3,4-dimethoxyphenyl) -pyrrolidine-2-carboxyl] -4-carbobenzoxy-piperazine.
5.6 g of 1-methyl-5- (3,4-dimethoxyphenyl) -pyrrolidine-2-carboxylic acid methyl ester (0.02 mol) are dissolved in 50 ml of dry toluene. 17.6 g (0.08 mol) of 4-carbobenzoxy-piperazine are added to this solution.
The reaction solution is then refluxed for 9 days and finally the toluene and the 4-carbobenzoxypiperazine are evaporated off under reduced pressure. The residue is covered with a layer of petroleum ether and crystallizes out after standing at 5. M.p.

Example 10 1-Methyl-5- (3, 4-dimethoxyphenyl) -pyrrolidine-2-carboxylic acid-N- (2'0anilinoethyl) -amide.
1.63 g (0.005 mol) of ethyl chloroformate are added to a mixture of 1.51 g (0.005 mol) of 1-methyl-5- (3, 4-dimethoxyphenyl) -pyrrolidione-2-carboxylic acid hydrochloride, cooled to -10 , 01 g (0.01 mol) of triethylamine and 15 ml of methylene chloride were added. The mixture is stirred for one hour at -10 and then 1.36 g (0.01 mol) of N-phenylethylenediamine are added.
The reaction is stirred overnight at room temperature and then evaporated to dryness. The residue is taken up in water, made alkaline to pH 9-10 with saturated sodium carbonate solution and then extracted with chloroform. The chloroform extracts are washed with water, dried over sodium sulfate and evaporated to dryness. The residue is distilled under high vacuum and gives an oil, boiling point 250-254 / 0.05 mm Hg.
Example 11 1-Methyl-5- (3,4-dimethoxyphenyl) -2 -carboxylic acid [4'-phenyl] piperazide.
1.4 g of 1-methyl-5- (3,4-dimethoxyphenyl) pyrrolidine-2-carboxylic acid hydrazide (0.005 mol) are dissolved in 20 ml of a 4-n. Solution of hydrochloric acid dissolved in absolute tetrahydrofuran.
