Verfahren zur Herstellung von Benzimidazolonderivaten
Die Erfindung betrifft ein Verfahren zur Herstellung neuer, an einem Stickstoff basisch substituierter Benzimidazolone (2-Oxo-benzimidazoline) der Formel:

sowie von Säureadditionssalzen davon. In Formel I bedeutet R einen geraden oder verzweigten Alkylenrest mit 2 bis 4 C-Atomen; R1 bedeutet ein Wasserstoffatom oder zusammen mit R2 einen Alkylenrest mit 2 bis 4 C-Atomen; R2 bedeutet ausserdem einen Alkylrest mit 1 bis 3 C-Atomen, oder zusammen mit R3 einen Alkylenrest mit 4 bis 6 C-Atomen; R3 bedeutet ausserdem einen Alkylrest mit 1 bis 3 C-Atomen; R4, das in 5oder 6-Stellung steht, und R5 sind miteinander vertauschbar, R4 bedeutet ein Halogenatom und R5 ein Wasserstoff- oder Halogenatom oder eine 1 bis 3 C-Atome enthaltende Alkylgruppe.
Die genannten Verbindungen sind pharmakologisch aktive Stoffe, deren Eigenschaften insbesondere auf Verwendbarkeit als Antidepressiva und Antikonvulsiva schliessen lassen. Während diese beiden Wirkungsrichtungen bei einigen Vertretern der Substanzgruppe in ähnlichem Ausmass auftreten, steht bei andern die eine oder die andere der beiden Wirkungsrichtungen im Vordergrund. So ist bei den in 5-Stellung und insbesondere in 6-Stellung substituierten Produkten die antidepressive Wirkung, bei den im l-Phenylrest - vorzugs- weise in p-Stellung-substituierten Verbindungen die antikonvulsivische Wirkung besonders ausgeprägt. Als Substituenten R4 und/oder R5 werden Chloratome bevorzugt.
Die basische Seitenkette in 3-Stellung ist vorzugsweise ein y-Dimethylaminopropyl-, y-Diäthylaminopropyl- oder y-Pyrrolidin-l'-yl-propylrest. Als Beispiele für in antikonvulsivischer Richtung besonders wirksame Verbindungen seien 1 -p-Chlorphenyl-3-y-diäthylaminopropyl-benzimidazolon und l-p-Chlorphenyl-3-y-pyrro lidin-l'-yl-propyl-benzimidazolon und ihre Säureadditionssalze, für in antidepressiver Richtung besonders wirksame Verbindungen l-Phenyl-3-y-dimethylaminopropyl-6-chlor-benzimidazolon und seine Säureadditionssalze erwähnt.
Verbindungen gemäss Formel I werden erhalten, indem man in Benzimidazolonen der Formel:

worin R, Rt, R2 und RQ die genannte Bedeutung haben, R'4, das in 5- oder 6-Stellung steht, und R'5 miteinander vertauschbar sind, R'4 eine Aminogruppe und RtÏ ein Halogenatom oder eine Aminogruppe, ein Wasserstoffatom oder eine 1 bis 3 C-Atome enthaltende Alkyl gruppe bedeutet, die Aminogruppen diazotiert und in die entsprechenden Halogenatome überführt.
Die nach dem beschriebenen Verfahren erhaltenen basisch substituierten Benzimidazolone gemäss Formel I können sowohl als freie Basen als auch in Form ihrer Additionssalze mit geeigneten Säuren, wie Halogenwasserstoffsäuren, Schwefelsäure, Salpetersäure, Phosphorsäure, Essigsäure, Oxalsäure, Malonsäure, Bernsteinsäure, Äpfelsäure, Maleinsäure oder Toluolsulfonsäure, gewonnen und verwendet werden.
Beispiel I
8,7 g g 1-p-Aminophenyl-3-y-pyrrolidin-1'-yl-propyl- benzimidazolon werden in 100 ml 1n Salzsäure gelöst und bei 0 C mit einer Lösung von 1,9 g Natriumnitrit in 10 ml Wasser diazotiert. Die Diazoniumlösung wird innert 10 Minuten zu einer bei 800 C gerührten Kupferchlorürlösung, die in üblicher Weise aus 5,3 g Kupfersulfat und 5,3 g Kochsalz in 25 ml Wasser unter Einleiten von Schwefeldioxydgas hergestellt wurde, zugegeben, und die Temperatur wird bis 30 Minuten nach Abklingen der Gasentwicklung aufrechterhalten. Nach dem Erkalten stellt man das Reaktionsgemisch mit Natronlauge alkalisch, nimmt die ausgefällte Base in Äther auf, wäscht die Ätherphase mit verdünnter Natronlauge und Wasser und arbeitet sie in üblicher Weise auf.
Der Rückstand wird in Petrolätherlösung aufgenommen und an Aluminiumoxyd geklärt, worauf man ihn aus kaltem Petroläther in kristalliner Form erhält.
Man gewinnt so 5,6 g 1-p-Chlorphenyl-3-y-pyrrolidin-1'- yl-propyl-benzimidazolon vom Schmelzpunkt 5P560 C.
Durch gleiches Vorgehen wie im vorstehenden Beispiel erhält man weiterhin z. B. die in der nachfolgenden Tabelle aufgeführten Produkte. In der Tabelle sind R, Rt, R2, R5, R4 und R5 die entsprechenden Gruppen gemäss Formel I mit der früher angegebenen Bedeutung.
In der Kolonne rechts sind in Klammer die Lösungsmittel bzw. Lösungsmittelgemische angegeben, aus welchen die Kristallisation erfolgt; dabei bedeuten: Ac = Aceton, Ae = Äther, Me = Methanol, Pe = Petrol äther und Pn = Pentan.

Process for the preparation of benzimidazolone derivatives
The invention relates to a process for the preparation of new benzimidazolones (2-oxobenzimidazolines) of the formula:

as well as acid addition salts thereof. In formula I, R denotes a straight or branched alkylene radical having 2 to 4 carbon atoms; R1 denotes a hydrogen atom or, together with R2, an alkylene radical having 2 to 4 carbon atoms; R2 also denotes an alkyl radical having 1 to 3 carbon atoms, or together with R3 an alkylene radical having 4 to 6 carbon atoms; R3 also denotes an alkyl radical with 1 to 3 carbon atoms; R4, which is in the 5 or 6-position, and R5 are interchangeable, R4 denotes a halogen atom and R5 denotes a hydrogen or halogen atom or an alkyl group containing 1 to 3 carbon atoms.
The compounds mentioned are pharmacologically active substances, the properties of which suggest, in particular, that they can be used as antidepressants and anticonvulsants. While these two directions of action occur to a similar extent with some representatives of the substance group, one or the other of the two directions of action is in the foreground with others. The antidepressant effect is particularly pronounced in the products substituted in the 5-position and especially in the 6-position, and the anticonvulsant effect is particularly pronounced in the compounds substituted in the 1-phenyl radical - preferably in the p-position. Chlorine atoms are preferred as substituents R4 and / or R5.
The basic side chain in the 3-position is preferably a γ-dimethylaminopropyl, γ-diethylaminopropyl or γ-pyrrolidin-l'-yl-propyl radical. Examples of compounds which are particularly effective in the anticonvulsant direction are 1-p-chlorophenyl-3-y-diethylaminopropyl-benzimidazolone and lp-chlorophenyl-3-y-pyrrolidin-l'-yl-propyl-benzimidazolone and their acid addition salts for antidepressant Direction of particularly effective compounds l-phenyl-3-y-dimethylaminopropyl-6-chloro-benzimidazolone and its acid addition salts mentioned.
Compounds according to formula I are obtained by adding in benzimidazolones of the formula:

wherein R, Rt, R2 and RQ have the meaning given, R'4, which is in the 5- or 6-position, and R'5 are interchangeable, R'4 is an amino group and RtÏ is a halogen atom or an amino group, a hydrogen atom or denotes an alkyl group containing 1 to 3 carbon atoms which diazotizes the amino groups and converts them into the corresponding halogen atoms.
The basic substituted benzimidazolones according to formula I obtained by the process described can be used both as free bases and in the form of their addition salts with suitable acids, such as hydrohalic acids, sulfuric acid, nitric acid, phosphoric acid, acetic acid, oxalic acid, malonic acid, succinic acid, malic acid, maleic acid or toluenesulfonic acid, obtained and used.
Example I.
8.7 g g of 1-p-aminophenyl-3-y-pyrrolidin-1'-yl-propyl-benzimidazolone are dissolved in 100 ml of 1N hydrochloric acid and diazotized at 0 ° C. with a solution of 1.9 g of sodium nitrite in 10 ml of water. The diazonium solution is added within 10 minutes to a copper chloride solution which is stirred at 800 ° C. and which has been prepared in the usual manner from 5.3 g of copper sulfate and 5.3 g of sodium chloride in 25 ml of water while introducing sulfur dioxide gas, and the temperature is up to 30 minutes after the evolution of gas has subsided. After cooling, the reaction mixture is made alkaline with sodium hydroxide solution, the precipitated base is taken up in ether, the ether phase is washed with dilute sodium hydroxide solution and water and worked up in the usual way.
The residue is taken up in petroleum ether solution and clarified on aluminum oxide, whereupon it is obtained in crystalline form from cold petroleum ether.
This gives 5.6 g of 1-p-chlorophenyl-3-y-pyrrolidin-1'-yl-propyl-benzimidazolone with a melting point of 5P560 C.
Using the same procedure as in the previous example, you still get z. B. the products listed in the table below. In the table, R, Rt, R2, R5, R4 and R5 are the corresponding groups according to formula I with the meaning given earlier.
In the column on the right, the solvents or solvent mixtures from which the crystallization takes place are indicated in brackets; where: Ac = acetone, Ae = ether, Me = methanol, Pe = petroleum ether and Pn = pentane.
