Verfahren zur Herstellung von Metallkomplexen Gegenstand vorliegender Erfindung ist ein Verfahren zur Herstellung von Metallkomplexsalzen der Formel (MRy)Xn, in welcher M zweiwertiges Zink, zweiwertiges Mangan, zweiwertiges Kupfer oder zweiwertiges Nickel bezeichnet, R 1,10-Phenanthrolin mit Phenyl-, Tolyl- oder Xylyl-Substituenten in einer oder zwei der Stel lungen 3, 4, 7 oder 8 oder mit Alkylresten in sämt lichen genannten Stellungen, die insgesamt 4-l0 Kohlen stoffatome aufweisen, bedeutet und X das Anion einer zur Bildung von Metallkomplexsalzen befähigten Säure d'arstell't,
ferner y 2 oder 3 und n eine durch die Wertig keit von X unter Herstellung elektrischer Neutralität be stimmte ganze Zahl ist, dadurch gekennzeichnet, dass man :ein Salz MXn mit einer entsprechenden Menge des 1,10-Phenanthrolins R in einem eine wesentliche Menge Wasser enthaltenden flüssigen Medium umsetzt und den Komplex aus der Lösung gewinnt.
Beim erfindungsgemässen Verfahren kann das Metallsalz in: einem geeigneten wässrigen Medium suspendiert oder gelöst werden, worauf mit der in einem geeigneten Medium gelösten Base gemischt wird, wobei man. das Gemisch zu homogener Lösung bringt; das Produkt kann durch; Kristallisieren oder andere konven tionelle. Arbeitsweisen abgetrennt werden. Das bevor zugte Lösungsmittel für das Metallsalz ist Wasser, jedoch können auch Gemische aus Wasser, und mit Wasser mischbaren organischen Lösungsmitteln verwendet wer den.
Das bevorzugte Lösungsmittel für die Base ist Ätha- nol, jedoch können auch andere mit Wasser mischbare Lösungsmittel verwendet werden oder wässrige Gemische solcher Lösungsmittel. Gewöhnlich wird durch Wärme einwirkung die Umsetzung zur Vervollständigung ge bracht. Die Reaktionslösung kann durch Eindampfen konzentriert werden, um das gewünschte Produkt zu isolieren. Man kann auf kleines Volumen oder bis zur Trockne eindampfen.
Ferner kann ein eine Ausfällung bewirkendes Lösungsmittel'. oder Salz zugegeben wer den. Gewöhnlich empfiehlt es sich, das Produkt um zukristallisieren. Dis erfindungsgemäss herstellbaren Metallkomplexe sind wertvoll als Therapeutika gegen zahlreiche Organis men, die Tierkrankheiten und Pflanzenschädigungen verursachen. Insbesondere sind diese Komplexe ge eignet zur Behandlung von topischen Infektionen beim Menschen, von Rindermastitis, beim Pilzbefall von Pflan zen und als Wurmmittel. Sie sind besonders aktiv gegen zahlreiche Mikroorganismen, d. h. gram-positive, gram negative. und säurefeste Organismen, pathogene Pilze, Hefe (Saccharomyces cerevisiae) und, Viren (Influenca).
Darüber hinaus entwickeln Organismen wie Staphylo- coccus pyogenes keine merkliche Resistenz gegen diese Komplexe, die auch bei Organismen wirken, die gegen alle anderen üblichen Antibiotika bereits Resistenz ent wickelt haben. Dies trifft besonders für Staphylococcus pyogenes zu. Ferner sind die Komplexe beständig (Lö sungen werden weder durch den bakteriellen noch den Wirts-Metabolismus, ferner nicht durch physikalische Mittel, z.
B. 20 Minuten dauernde Druckeinwirkung von 1,05 atü zwecks Sterilisierung zerstört); und auch bei sehr hohen Konzentrationen (1- bis 2 o ige Lösungen) nicht irritierend für die Haut, Schleimhäute (Auge, Ohr, Nase, Hals), in Körperhöhlen (Vagina, Darm, Blase) und subkutane Gewebe (Muskeln, Knochen).
Die Komplexe: wurden klinisch gegen Infektionen durch gram-positive und gram-negative Mikroorganis men und pathogene Pilze, einschliesslich dermatologi- schen Infektionen (Dermatomycosen, Furunkel, Folli- culitis, Akne vulgares, Pustular-Dermatitis, infektiöse Ekzeme), Wundinfektionen (Geschwüre), gynäkologische Infektionen (Trichomonas vaginalis), Candida albicans und zahlreiche nicht spezifische Organismen, die bei chronischen Nasen-, Hals- und Ohreninfektionen auf treten (Mastoiditis, externe Otitis, nasale Furunkulose), Augeninfektionen (akute und chronische Conjunctivitis)
und bei neonatalen Infektionen (Verhütung der Staphy- loccoceus-Infektion beim Neugeborenen) erprobt. Wund infektionen durch Staphylococcus pyogenes werden schnell beseitigt. Gewisse Komplexe sind aktiv gegen Infektionen in incipienten Furunkeln.
Bei der Verwendung der erfindungsgemäss herge stellten Komplexe als Therapeutica zur Anwendung bei infektiösen Krankheiten kommen diese in Kontakt mit den Wirts-Zellen. Ein wesentlicher Faktor bei ihrer Wirksamkeit ist die differentielle Empfindlichkeit des die Infektion hervorrufenden Agenz und der Wirts Zelle an der Stelle der Infektion gegenüber den Komplexen. Die Empfindlichkeit von. gram-positiven Organismen, wie z. B. Staphylococcus pyogenes gegenüber den Kom plexen ist sehr stark (zwischen 1,5 bis 12,5 microg/ml). Gram-negative Mikroorganismen wie Escherichia coli sind zwar weniger empfindlich (etwa 100 mierog/ml) als die gram-positiven Cocci, jedoch sprechen sie auf die örtlich angewandten Konzentrationen (1 %ige Lösung) ebenfalls an.
Die Gegenwart von Plasma-Gewebeflüssig keit und infektiösen Ausscheidungen beeinträchtigt die Wirkung der Komplexe nicht wesentlich:. Bei der gegen Mikroorganismen wirksamen Konzentration tritt weder eine toxische Wirkung auf das Wirts-Gewebe, auch bei längerer Applikation, noch ein nachteiliger Effekt auf die Regenerierung des Epithels auf.
Bei der Behandlung von Rindermastitis sind die Komplexe wirkam gegen akute und chronische Infek tionen durch Streptococcus pyogenes (Gruppe. B) oder Staph. pyog., die sich gegen die zurzeit gebräuchlichen Antibiotika resistent erwiesen haben. Weder von Strepto- coccen noch von: Staphylococcen wurde eine merkliche Resistenz gegen die Komplexe in: vitro oder in vivo entwickelt. Verschiedene Formen dieser Organismen sind reproduzierbar, die im allgemeinen wesentlich er niedrigte Virulenz zeigen. Die vorliegenden Komplexe scheinen auch verwendbar für chronisch nicht heilende Wunden, die als Infektionsherde wirken können.
Bei der Behandlung von durch Mikroorganismen, insbesondere Pilze und Nematoden, hervorgerufenen Pflanzenkrankheiten wurden Tests mit den Pilzen Ven- turia inaequalis und Phytophthora infestans sowie den Nematoden Panagrellus redivivis und Meloidogune incognita durchgeführt. Vent. in. befällt Äpfel, Phyt. inf. Tomaten und Kartoffeln; die Nematoden verursachen Wurzelschäden. Mit zahlreichen der getesteten Komplexe wurden ausgezeichnete Ergebnisse gegenüber diesen Organismen erhalten.
Es wird angenommen, dass die. erfindungsgemäss herstellbaren Komplexe ihre :therapeutische Wirkung auf Grund ihrer Fähigkeit ausüben, sich an wesentliche biologische Stellen (Enzyme und' Proteine) durch elektro statische und/oder Van der Waalsche Kräfte zu binden oder anzulagern. Wenn sich die aktive Rezeptorstelle an der Oberfläche der biologischen Stelle befindet, genügt eine direkte Adsorption des Metallkomplexes, voraus gesetzt, dass er genügend fest gebunden wird; wenn jedoch die aktive Stelle innerhalb der Zelle liegt, so ist ein Eindringen des Metallkomplexes für die therapeuti sche Wirkung wesentlich.
Sowohl Eindring- als auch Bindefähigkeit der Komplexe wird erhöht durch die selektive Substitution der primären 1,10-Phenanthrolin- Liganden, die als komplex- oder metallbindende Mittel fungieren. Während die Metallkomplexe des primären Ligaiden 1,10-Phenanthrolin verhältnismässig geringe Eindringkraft und Bindefähigkeit gegenüber den aktiven Stellen in oder ausserhalb der Zelle besitzen, werden diese Eigenschaften durch bestimmte Substitutionen wesentlich erhöht.
Die erfindungsgemäss gewählten Sub- stituenten verursachen Bevorzugung der Lipoid-Phase der Zellmembran und geben auf Grund von Faktoren wie der Molekülgrösse dem komplexen Molekül eine genügend grosse Oberfläche für Van der Waalsche Bindung oder Adsorption. Andere Substiuenten haben ungünstige Wirkungen auf die Eindringfähigkeit, wäh rend sie die Oberflächengrösse des Komplexes nicht wesentlich beeinflussen. Der Metallkomplex als Gesamt heit und nicht Spuren von Metall oder abdissoziertem Liganden ist das primär wirksame biologische Agens.
Die Funktion des Metallatoms ist vermutlich zwei fach, indem es eine positive Ladung an den Gesamt komplex abgibt und' zwei oder drei relativ grosse Ligan- denmoleküle substituiertes Phenanthrolin bindet, wobei ein sehr grosses Molekül entsteht. Es wird angenommen, dass die durch das Metallion bewirkte positive Ladung auf die grosse Oberfläche des Komplexes verteilt ist, der somit als sehr grosse positiv geladene Pseudokugel wirkt.
Der Effekt der genannten Substitution der primären Base liegt somit vermutlich darin, das positive Ladung aus der eigentlichen. Kugel an Spitzen oder Enden ver lagert wird, so dass die Bindungsfähigkeit an eine Stelle mit negativer Ladung erhöht wird.
Das Metallatom und dessen Oxydationsstufe sind von entscheidender Be deutung. Höhere oder niedrigere Oxydationsstufen der Metallatome als die vorstehend genannten sind nicht brauchbar, da der Komplex in der therapeutischen Zu sammensetzung gegen Oxydation oder Reduktion unbe ständig und dadurch in seinen Eigenschaften grund legend verändert wird. Höhere oder niedrigere Oxy dationsstufen als die vorstehend genannten bringen auch geringere Beständigkeit der Gesamtkomplexe mit sich;
letztere dissoziieren dann leichter, im allgemeinen unter Verlust der biologischen Eigenschaften, die für den intakten Komplex charakteristisch sind.
Es wurde festgestellt, dass 3,4,7,8-Tetraalkyl- Substitution des 1,10-Phenanthrolins zu Verbindungen mit' hoher Aktivität gegen bakterielle Infektionen bei Menschen führt; symmetrisch substituierte Verbindungen ergeben hochstabile Komplexe. Ferner wird symme trische Substitution bei den Substituenten Phenyl, Tolyl und Xylyl bevorzugt. Die Anionen X können einer be liebigen geeigneten anorganischen oder organischen Säure entstammen. Das Anion kann beispielsweise Chlorid, Sulfat oder Nitrat oder Tartrat, Benzoat oder Stearat sein.
Beispiel 1 Tris-(3,4,7,8-tetramethyl-1,10-phenanthrolin)- nickel(II)-sulfat - H20 27,105 g Nickel(II)-sulfat - 6,5H20 (0,1 Mol) wurden in 200 ml Wasser gelöst und langsam unter heftigem Rühren einer Lösung von 76,297 g 3,4,7,8-Tetramethyl- 1,10-Ph - H20 (0,3 Mol) in 700 ml Äthanol zugegeben. Die Lösung wurde filtriert und in einem Luftstrom auf dem Dampfbad zur Trockne eingedampft. Das hell braune Produkt wurde in 500 ml Wasser von 70 wieder gelöst und mit 50 g Diatomeenerde ( High-Flow ) ge schüttelt, dann durch Whatman No. 542 -Filterpapier filtriert.
Das Filtrat wurde nochmals eingedampft und das Produkt im Trockenschrank bei 90 C getrocknet. Die Formel ist (Ni[C16H16N2]3)SO4 - aq.
Beispiel <I>2</I> Tris-(3,4,7,8-tetramethyl-1,10-phenanthrolin)- kupfer(II)-perchlorat Eine Lösung von 2,49g Kupfer(II)-sulfat - H20 in 500 ml Wasser wird mit einer Lösung von. 8,0 g 3,4,7,8- Tetramethyl-1,10-Ph - H20 in 150 ml Äthanol behan delt.
Das Gemisch wird erhitzt, bis eine einheitlich blaue Farbe entsteht, wobei verdampfter Alkohol von Zeit zu Zeit ersetzt wird Sodann wird der Alkohol bis zur beginnenden Abscheidung des Komplexes abgetrieben; dann gibt man 5 g Natriumperchlorat in wenig Wasser gelöst zu. Der blaue kristalline Niederschlag wird ab filtriert, mit wenig Wasser gewaschen und an der Luft getrocknet. Er besitzt die Formel: (Cu[C16H16N2]3)(C10O4)2 - H20.
Beispiel <I>3</I> Bis-(3,4,7,8-tetramethyl-1,10-phenanthrolin)- kupfer(II)-acetat 4,64 g 3,4,7,8-Tetramethyl-1,10-phenanthrolin und Kupfer(II)-acetat monohydrat wurden unter Rühren in 50 m1 heissem Methanol gelöst. Die resultierende grün blaue Lösung wurde filtriert und dann zur Trockne ein gedampft. Das dabei erhaltene blaue Pulver kann aus einem Gemisch aus Äthanol. und Diäthyläther um kristallisiert werden. Es. besitzt die Formel (Cu[C16H16N2]2)(CH3COO)2 aq. Der Hydratationsgrad hängt vom Trocknungsgrad ab.
<I>Beispiel 4</I> Bis-(3,4,7,8-tetramethyl-1,10-phenanthrolin)- mangan(II)-acetat 710 g 3,4,7,8-Tetramethyl-1,10-phenanthrolin wur den in 2,5<B>1</B> Aceton bei 50 unter Rühren mit einer Lösung von 367 g Mangan(II)-acetat-tetrahydrat in 500 m1! Wasser im Verlauf von 3 Stunden versetzt. Gegen Ende der Zugabe fiel etwas Feststoff aus. Es wurde filtriert, und das Filtrat wurde in 7,51 Aceton eingegeben. Die Acetonlösung wurde auf 5 abgekühlt, und blass cremefarbene Kristalle wurden abfiltriert und bei 50 getrocknet.
Die so erhaltene Verbindung besass die Formel (Mn[C16H16N2]2)(CH3COO)2 aq.
Der Hydratationsgrad hängt von der Trocknungsweise ab. <I>Beispiel 5</I> Bis-(3,4,7,8-tetramethyl-1,10-phenanthrolin)- zinkacetat 54,8 g Zinkacetat-monohydrat wurden mit 118 g 3,4,7,8-Tetramethyl-1,10-phenanthrolin unter den in Beispiel 4 verwendeten Bedingungen umgesetzt. Das blassgraue kristalline Produkt besass die Formel (Zn[C16H16N2]2)(CH3COO)2 aq. <I>Beispiel 6</I> Tris-(3,8-dimethyl-4,7-diäthyl-1,10-phenanthrolin)- nickel(II)-sulfat Eine Lösung von 8,8g Nickelsulfat-Hexahydrat in einem Gemisch aus 10 ml Aceton und 25 ml Wasser wurde unter Rühren bei 50 allmählich mit 26,4 g 3,8- Dimethyl-4,7-diäthyl-1,10 - phenanthrolin versetzt.
Die resultierende Suspension wurde mit 25 ml Aceton ver dünnt und so lange bei 50 gerührt, bis eine klare Lösung erhalten wurde. Die Lösung wunde in: 300 ml Aceton eingegossen, und der sich abscheidende Nickel komplex wurde abfiltriert, mit wenig Aceton gewaschen und getrocknet. Das Produkt besass die Formel (Ni[C18H20N2]3)SO4.
<I>Beispiel 7</I> Bis-(4,7-diphenyl 1,10-phenanthrolin)- kupfer(II)-sulfat Eine Lösung von 12,4 g Kupfer(II)-sulfat-hepta hydrat in 60 ml Wasser wurde unter Rühren zu einer Lösung von 3'3,2 g 4,7-Diphenyl-1,10-phenanthrolin in 100 ml. Methanol von 50 eingegeben. Die resultierende Lösung wurde eine weitere Stunde bei 50 gerührt, dann auf 5 abgekühlt, worauf die blauen Kristalle abfiltriert und getrocknet wurden, Die Verbindung besass die Formel (Cu[C24H16N2]2)SO4 aq. Der Hydra tationsgrad hängt von der Trocknungsweise ab.
<I>Beispiel 8</I> Tris-(4,7-diphenyl-1,10-phenanthrolin)- nickel(II)-sulfat 8,8g Nickelsulfat-Hexahydrat wurden mit 33,2 g 4,7-Diphenyl-1,10-phenanthrolin unter den Arbeits bedingungen von Beispiel 6 umgesetzt. Man erhielt ein Produkt der Formel (Ni[C24H16H2]3)SO4 aq.
Der therapeutische Wert von Tris-3,4,7,8-tetra- methyl-1,10-ph)-nickel(II)-sulfat bei Behandlung von akuter und chronischer, durch Staphylococcen und Streptococcen verursachter Rindermastitis wurde unter sucht. Alle chronischen Infektionen sprachen auf die Penicillintherapie nicht mehr an. Die Behandlung er folgte örtlich (50 bis, 100 mg 2 X täglich in einer Paraffin- und Wachsgrundlage), und der chemische und bakteriologische Verlauf nach der Verabreichung wurde verfolgt bis zur Heilung.
Der genannte Komplex bewirkte eine Heilung.
Der therapeutische Wert von Tris-(3,4,7,8-tetra- methyl-1,10-phenanthrolin)-nickel(II)-sulfat wurde auch gegen Tiere und Pflanzen. befallende Viren untersucht. Der Komplex zeigte eine inhibierende Wirkung zwischen 40 und 99 % bei einer Konzentration von 10---4M gegen Tabakmosaikvirus auf Blättern einer gesunden türkischen Tabakpflanze. Die Isolierung des Tabakmosaikvirus er folgte nach dem Verfahren von Commoner et al. (Arch. Biochem. 27, Seiten 271 bis 286 [1950]).
Der Komplex zeigte ferner bei nichtviroziden Konzentrationen (10-5M) mindestens 80%ige Inhibierung der Infektion durch den Influenza-Virus bei chorioallantoischen Membranen von Kücken nach zweistündiger Inkubation mit dem Virus bei 20 C. Tests wurden durchgeführt, bei denen behandelter Virus in Stücke der Kückenmem- bran in Abwesenheit des Komplexes inokuliert wurde. Die Evaluierung erfolgte: nach der Methode von Fazegas die St. Groth und White (J. Hyg. Cambridge, Bd. 56, Seiten 151 bis 162 [1958]).
Die Anwendung der vorliegenden Komplexe kann mit ,einem Träger oder Verdünnungsmittel erfolgen. Zur Behandlung beim Menschen wird der Komplex z. B. in Form von Salzlösungen, Cremen Lotionen, Nasen-, Augen- oder Ohrentropfen, Pessarien, Suppositorien, Tabletten, Pastillen und dergleichen angewandt. Eine dermatologische oder Vaginal-Creme kann 10 mg/g des Komplexes in der Creme-Grundlage enthalten, ebenso eine Lotion. Eine Salzlösung kann z.
B. 5 bis 10 mg/ml des Komplexes enthalten, Nasentropfen 10 mg/ml, Augentropfen 4 mg/ml, Ohrentropfen 10 mg/ml in Propylenglykol; Suppositorien von 5 g können 50 mg des Komplexes enthalten, und oral zu verabreichende Tabletten von 500 mg können 250 mg enthalten. Auch in der Veterinär-Medizin, wie z.
B. bei der Rinder- mastitis, können die Komplexe mit geeigneten Trägern, wie Paraffin- und Wachsgrundlagen, angewendet werden, während bei Wurmmitteln zweckmässigerweise eine Trinkflüssigkeit, vorzugsweise eine wässrige Lösung oder Suspension, je nach der Wasserlöslichkeit, gegeben wird.
Bei der Anwendung als Fungizide oder Virozide bei Pflanzen werden die wasserunlöslichen Komplexe zweck mässigerweise als dispergierbare Pulver hergestellt, während die wasserlöslichen Komplexe in wässrigen Lö sungen angewendet werden können; jedoch können auch Konzentrate der Komplexe in organischen Lö- sungsmitteln hergestellt werden, aus denen später Emul sionen gebildet werden;
in all diesen Zusammensetzungen können Netzmittel und/oder andere Substanzen, die die Zusammensetzung fördern oder bei der endgültigen Verwendung von Vorteil sind, zugegeben werden.

In the treatment of bovine mastitis, the complexes are effective against acute and chronic infections by Streptococcus pyogenes (group. B) or Staph. pyog. which have proven to be resistant to the antibiotics currently in use. Neither streptococci nor: staphylococci showed any noticeable resistance to the complexes in: vitro or in vivo. Various forms of these organisms are reproducible, which generally exhibit significantly lower virulence. The present complexes also appear useful for chronically non-healing wounds that can act as foci of infection.
In the treatment of plant diseases caused by microorganisms, in particular fungi and nematodes, tests were carried out with the fungi Ven turia inaequalis and Phytophthora infestans and the nematodes Panagrellus redivivis and Meloidogune incognita. It is believed that the. The ability of the complexes to penetrate as well as to bind is increased by the selective substitution of the primary 1,10-phenanthroline ligands, which act as complex- or metal-binding agents. While the metal complexes of the primary leagueid 1,10-phenanthroline have relatively little penetration and binding ability to the active sites in or outside the cell, these properties are significantly increased by certain substitutions.
The substituents selected according to the invention cause the lipoid phase of the cell membrane to be preferred and, based on factors such as the size of the molecule, give the complex molecule a sufficiently large surface for Van der Waals bonding or adsorption. Other substituents have unfavorable effects on the penetrability, while they do not significantly affect the surface area of the complex. The function of the metal atom is probably twofold, in that it gives off a positive charge to the overall complex and binds two or three relatively large ligand molecules of substituted phenanthroline, creating a very large molecule. It has been found that 3,4,7,8-tetraalkyl substitution of 1,10-phenanthroline leads to compounds with high activity against bacterial infections in humans; symmetrically substituted compounds result in highly stable complexes. Symmetrical substitution is also preferred for the phenyl, tolyl and xylyl substituents. The anions X can come from any suitable inorganic or organic acid. <I> Example 8 </I> Tris (4,7-diphenyl-1,10-phenanthroline) - nickel (II) sulfate 8.8 g of nickel sulfate hexahydrate were mixed with 33.2 g of 4,7-diphenyl-1 , 10-phenanthroline implemented under the working conditions of Example 6. A product of the formula (Ni [C24H16H2] 3) SO4 aq was obtained.
The therapeutic value of tris-3,4,7,8-tetra-methyl-1,10-ph) nickel (II) sulfate in the treatment of acute and chronic bovine mastitis caused by staphylococci and streptococci was investigated. All chronic infections no longer responded to penicillin therapy.
