Verfahren zur Herstellung von neuen Piperidinderivaten
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Piperidindervate mit wertvollen pharmakologischen Eigenschaften.
Es wurde überraschenderweise gefunden, dass Piperidinderivate der Formel I,

in welcher R, einen gegebenenfalls durch Ersatz von 1-2 nicht be nachbarten, als Ketten-oder Ringglieder vorliegen den Methylen-bzw. Methingruppen durch-O-,-S-, -SO-,-CO-,-CO-O-,-NH-,-CO-NH-bzw.
-N oder-CO-N \ \ abgewandelten und gegebenenfalls durch Hydroxyl-,
Amino-, Nitro-, Carboxyl-, Carbamoyl-, Cyano oder Trifluormethylgruppen, niedere Alkanoyl- aminogruppen oder Halogen substituierten, nicht aromatische gebundenen Kohlenwasserstoffrest mit höchstens 12 Kohlenstoffatomen, R2 Wasserstoff oder den Methylrest, R3 einen Alkylrest mit höchstens 4 Kohlenstoffatomen, den Phenylrest, einen Phenylalkylrest mit 7-9 Koh- lenstoffatomen, den Styrylrest, oder zusammen mit g2 einen gegebenenfalls methylsubstituierten Tri methylen-bis Hexamethylenrest, R4 Wasserstoff oder den Methylrest, und Wasserstoff oder einen Alkanoylrest mit höchstens

und ihre Salze mit anorganischen und organischen SÏuren wertvolle pharmakologische Eigenschaften, insbesondere eine ausgezeichnete analgetische Wirksamkeit bei oraler wie bei parenteraler Applikation und starke antitussive Wirksamkeit besitzen. Im Gegensatz zu anderen Analgetica besitzen sie keine parasympathicolytischen Eigenschaften, sondern wirken eher parasym pathicomimetisch. Sie sind zugleich relativ wenig toxisch und eignen sich deshalb z. B. zur Linderung und Behebung von Schmerzen verschiedener Genese und auch des Hustenreizes.
In den Verbindungen der Formel I und den zuge hörigen, weiter unten genannten Ausgangsstoffen ist R. i beispielsweise durch Wasserstoff, Alkylreste, wie den Methyl-, Athyl-, n-Propyl-, Isopropyl-, n-Butyl-, Isobutyl-, sek.
Butyl-, n-Amyl-, Isoamyl-, n-Hexyl-, n Octyl-, n-Decyl-oder n-Dodecylrest ; durch Alkenylreste, wie den Allyl-, Crotyl-, Methyllyl-oder y, y-Di- methyl-allylrest ; durch Alkoxyalkylreste, wie den ¯ Methoxy-äthyl-, -Äthoxy-äthyl-, ss-Butoxy-äthyl-, y- Methoxy-propyl-, y-Isopropoxy-propyl-oder & Athoxy- butylrest ; durch Alkylthio-alkylreste, wie den ss-Methyl- thio-äthyl-, y-Methylthio-propyl-und y-Isopropylthiopropylrest ;

Benzyl-, o-Methyl-benzyl-, cr-Phenyl-äthyl-, ss-Phenyl- äthyl-, y-Phenyl-propyl-oder Styryl-rest (ss-Phenyl- vinyl-rest). Rs bildet ferner zusammen mit Ra z. B. den
Trimethylen-, Tetramethylen-, 1-Methyl-tetramethylen-, Pentamethylen-oder Hexamethylenrest. Ein niederer
Alkanoylrest R5 ist beispielsweise der Acylrest der
EssigsÏure, PropionsÏure, ButtersÏure, IsobuttersÏure,
ValeriansÏure oder IsovaleriansÏure.
Zur Herstellung der Verbindungen der Formel I setzt man eine Verbindung der Formel II,

in welcher R2, Rs, R4 und R, die oben angegebene Bedeutung haben, mit einem reaktionsfÏhigen Ester einer Verbindung der Formel III, RtOH (III) in welcher R1 die oben angegebene Bedeutung hat, um und behandelt gew nschtenfalls ein Reaktionsprodukt, das als Rest R5 ein Wasserstoffatom enthÏlt, mit einer niederen Alkansäure, mit einem reaktionsfähigen funk tionellen Derivat einer solchen der mit einem entspre- chenten Keten. Als reaktionsfÏhige Ester von Verbindungen der Formel III eignen sich insbesondere HalogenwasserstoffsÏureester, wie Bromide, Chloride und Jodide, weiter ArylsulfonsÏureester, z.
B. p-ToluolsulfonsÏureester, sowie leicht zugÏngliche SchwefelsÏureester, wie Dimethylsulfat und DiÏthylsulfat. Ihre Umsetzung mit Verbindungen der Formel II erfolgt beispielsweise bei Raumtemperatur oder massig erhöhter Temperatur in einem geeigneten organischen Lösungsmittel, wie z. B. Äthanol, Aceton, Äthylacetat oder Dimethylformamid. Gegebenenfalls wird die Umsetzung durch Zusatz säurebindender Mittel, wie z. B. Kaliumcarbonat, und/oder von Kaliumjodid beschleunigt.
Zur Einführung eines niederen Alkanoylrestes R3 behandelt man gewünschtenfalls ein Reaktionsprodukt der Formel I, in dem Rs durch Wasserstoff verkörpert ist, mit einem Halogenid, Cyanid oder Anhydrid einer niederen Alkansäure. Als Reaktionsmedium dient vorzugsweise ein Uberschuss des eingesetzten Anhydrids oder Halogenids ; andere Acylierungsmittel werden in inerten organischen Lösungsmitteln, wie z. B. Diäthyl äther, Tetrahydrofuran oder Benzol, angewendet. Ein säurebindendes Mittel, z. B. eine tertiäre organische Base, wie Pyridin oder Triäthylamin, kann gewünschten- falls zugefügt werden.
Die Acylierungen werden zweck mässig bei Temperaturen von höchstens 60 , vorzugsweise bei 0 bis Raumtemperatur, durchgeführt.
Die Ausgangsstoffe der Formel II lassen sich nach verschiedenen Methoden herstellen. Beispielsweise erhält man derartige Verbindungen, wenn man 4-Piperidon oder 3-Methyl-4-piperidon mit einem Keton der Formel IV, RsCOCHiR2 (IV) in welcher R2 und Rs die oben angegebene Bedeutung haben, in Gegenwart eines in homogener oder heterogener Phase vorliegenden basischen oder sauren Stoffes umsetzt.
Als basische Kondensationsmittel kommen einerseits anorganische Basen, wie Natriumhydroxyd, und organische Basen, wie Piperidin, Piperazin, und in wasserfreiem Medium oder in Abwesenheit von Lösungsmitteln auch Alkalimetallalkoholate, und anderseits basische Inonenaustauscher, vorzugsweise solche mit quatemären Ammoniumgruppen, wie z. B. Amberlite IRA 400 (OH63), aber auch schwächer basische, wie Amberlite-IR 4B, in Frage, welche in ansatzweisem oder gegebenenfalls auch in kontinuierlichem Verfahren angewendet werden können. Als Reaktionsmedium kann je nach der Löslichkeit der Ausgangsstoffe z. B Wasser, ein wasserhaltiges und schliesslich auch ein wasserfreies niederes Alkanol oder ein anderes polares Lösungsmittel dienen.
Als Beispiel von sauren Kondensationsmitteln seien Ammoniumsalze, wie z. B. Ammoniumacetat, allein oder in Kombination mit Eisessig und gegebenenfalls einem inerten Lösungsmittel, wie z. B. Benzol, sowie saure Ionenaustauscher, wie z. B. Amberlite IR 120 (H@-Form), in Wasser oder einem wasserhaltigen niederen Alkanol als Reaktionsmedium genannt.
Die Kondensation erfolgen vorzugsweise bei Raumtemperatur bis massig erhöhter Temperatur, d. h. der bevorzugte Temperaturbereich für die Gewinnung von Ausgangsstoffen der Formel II liegt zwischen ca. 20 und 60 .
Nach einem weiteren Verfahren stelIt man Ausgangsstoffe der Formel II her, in welchen der Rest Rg eine Methylgruppe ist oder an seiner Bindungsstelle mit der Carbonylgruppe eine Methylengruppe aufweist, indem man eine Verbindung der Formel V,

in welcher Rt'Wasserstoff oder Benzyloxycarbonylrest und Rs'Wasserstoff oder einen Rest bedeutet, der bei Zu- fügen der Methylengruppe-CH2-einem unter der allgemeinen Formel I definierten Rest Rg entspricht, und R2 und R4 die oben angegebene Bedeutung haben,
mit einer Quecksilberionen enthaltenden wässrigen Minerasäure beim Raumtemperatur bis massig erhöhter Temperatur behandelt und einen allfälligen Benzyloxycarbonylrest Rt'durch Hydrogenolyse abspaltet. Als wässrige Mineralsäure für die Durchführung der Hydratisierung eignet sich beispielsweise 10-84 /oige Schwe felsäure, ferner kann z. B. auch ca 15 /oige bis 36"/oigne (konzentrierte) Salzsäure verwendet werden. Die Reaktionstemperatur wird bei steigender Säurekonzentration niedriger gewählt, bei Verwendung von 84"/piger Schwe felsäure oder ca. 1 : 1 verdünnter Salzsäure bei 50-60 durchgeführt.
Die Abspaltung eines allfälligen Benzyloxycarbonylrestes Rj'kann beispielsweise durch Behandlung eines entsprechenden Zwischenproduktes mit Wasserstoff in Gegenwart eines Hydrierungskatalysators, wie z. B. Palladium auf Kohle, in einem geeigneten organischen Lösungsmittel, wie z. B. Äthanol, erfolgen.
Gewünschtenfalls kann vor der Hydrogenolyse ein niederer Alkanoylrest Rs nach den weiter oben für die Acylierung von Verbindungen der Formel I bereits genannten Arbeitsweisen eingeführt werden.
Verbindungen der Formel V sind ihrerseits z. B. durch Umsetzung von Propargylbromid oder eines entsprechend der Definition von R2 und Rs substituierten Analogen, wie z. B. 3-Brom-1-butin oder 1-Brom-2- butin, mit amalgamiertem Magnesium, Zink oder Aluminium in einem Gemisch von Tetrahydrofuran und Toluol, und Kondensation der entstandenen metallorganischen Verbindung mit der halbmolaren Menge 4 Piperidon oder 3-Methyl-4-piperidon erhältlich. Anstelle halbmolarer Mengen 4-Piperidon oder 3-Methyl4-piperidon können äquimolare Mengen, bezogen auf die metallorganischen Verbindungen, der entsprechenden 1-Benzyloxycarbonyl-derivate eingesetzt werden.
Gewünschtenfalls werden Verbindungen der Formel I in andere Verbindungen dieser Formel umgewandelt.
Von Interesse ist besonders die Reduktion von Verbindungen, deren Rest Ri eine Nitrogruppe enthält, sowie die Hydrolyse von Verbindungen mit einer niederen Alkanoylaminogruppe als Substituent von R, zu den entsprechenden Verbindungen mit einem durch eine Aminogruppe substituierten Rest Ri.
Als anorganische und organische Säuren, die zur Salzbildung mit den erfindungsgemässen Basen für die Herstellung von kristallisierten analgetischen und antitussiven Wirkstoffen und teilweise auch für die Reinigung der Basen in Frage kommen, seien beispielsweise genannt : Salzsäure, Bromwasserstoffsäure, Schwefel säure, Salpetersäure, Phosphorsäure, Methansulfon säure, Athandisulfonsäure, ss-Hydroxyäthansulfonsäure,
Essigsäure, Propionsäure, Maleinsäure, Fumarsäure,
Milchsäure, Apfelsäure, Weinsäure, Citronensäure, Ben zoesäure, Salicylsäure, Phenylessigsäure und Mandel säure.
In den nachfolgenden Beispielen sind die Tempera turen in Celsiusgraden angegeben.
Beispiel 1 a) Herstellung von 1- (4'-Hydroxy-4'-piperidyl)-2- propanon, erste Methode.
12 g fein gepulvertes Hydrochlorid des 4-Piperidon hydrats (4, 4-Piperidindiol-Hydrochlorid) werden zu 20 g fein gepulvertem Natriumhydroxyd in 80 ml Chloro form bei-20 zugegeben. Dann werden langsam 2 ml Wasser zugegeben, worauf die Temperatur steigt. Durch
Kühlen wird dafür gesorgt, dass sie nicht über 10 steigt.
Dann wird Natriumsulfat zugegeben, das Reaktionsge misch filtriert und das Filtrat im Rotationsverdampfer eingedampft, zuletzt für 5 Minuten unter Hochvakuum, wobei das 4-Peridon zurückbleibt. 9 g Magnesium wer den in 20 ml absolutem Ather vorgelegt, 0, 1 g Queck- silberchlorid zugegeben und bei-10 bis-5 44, 5 g Pro pargylbromid in 100 ml absolutem Ather zugetropft.
Anschliessend wird noch 20 Minuten bei-5 weiter gerührt. Bei5 bis 0 werden hierauf 28, 95 g 4-Piperi don in 250 ml absolutem Benzol zugetropft, wobei sich das Reaktionsgemisch erwärmt. Es ward 30 Minuten bei 0 und anschliessend ca. 14 Stunden bei Raum temperatur weitergerührt. Hierauf gibt man Eis und Eis essig zu, trennt die Benzolphase ab und extrahiert sie mit 2-n. Essigsäure. Die essigsauren Lösungen werden vereinigt, mit Ather geschüttelt und hierauf alkalisch gestellt. Die freigesetzte Base wird mit Chloroform ex trahiert, die Chloroformlösung getrocknet und einge dampft, wobei 9, 3 g eines bls zurückbleiben. Die De stillation liefert ein 61 vom Kp. 100-140 /0, 05 Torr (Luftbad), das kristallisiert.
Das so erhaltene 4- (2'-
Propinyl)-4-piperidinol schmilzt bei 76-78 .
1, 6 g des obigen Produkts werden mit 16 ml 2-n.
Schwefelsäure und 200 mg Quecksilbersulfat 3 Stunden bei Raumtemperatur stehengelassen. Das Reaktions gemisch wird alkalisch gestellt und mit Chloroform aus gezogen, die Chloroformlösung getrocknet und einge dampft, Der Rückstand wird im Kugelrohr destilliert, wobei das 1- (4'-Hydroxy-4'-piperidyl)-2-propanon er halten wird, Kp. 70 /0, 01 Torr, Smp. 98-100 , vgl. auch b). b) Herstellung von 1- (4'-Hydroxy-4'-piperidyl)-2- propanon, zweite Methode.
In einem Sulfierkolben werden 15, 35 g 4-Piperidon
Hydrat-Hydrochlorid in 110 ml 1-n. Natriumbicarbonat lösung gelöst. Unter Kühlung und Vibrieren werden bei 5-10 17, 1 g Benzyloxycarbonylchlorid und 110 ml 1-n.
Natriumbicarbonatlösung so zutropfen gelassen, dass das Reaktionsgemisch immer alkalisch bleibt. Anschlies- send wird es während 1t/2 Stunden weitervibriert und die entstandene milchige Emulsion mit Ather ausge zogen. Die Atherlösung wird zweimal mit 2-n. Salz säure und dann zweimal mit gesättigter Natriumchlorid- lösung durchgeschüttelt, getrocknet und eingedampft und der Rückstand im Hochvakuum destilliert. Das 1
Benzyloxycarbonyl-4-piperidon siedet bei 125-131 /
0, 004 Torr.
In einem Rührkolben werden 4 g Magnesium in 6 ml absolutem Ather vorgelegt und dann unter Rühren bei 10-15 19, 5 g Propargylbromid in 35 ml absolutem Ather zutropfen gelassen. Anschliessend werden 19, 1 g I-Benzyloxycarbonyl-4-piperidon in 80 ml absolutem Benzol bei 20-25 langsam zugetropft, wobei sich ein Klumpen bildet, der sich aber beim weiteren Rühren wieder löst. Hierauf wird das Reaktionsgemisch weitere 3 Stunden gerührt. Dann wird es mit Eis und 2-n.
Schwefelsäure zersetzt und mit Ather ausgezogen. Die Ätherlösung wird mit 2-n. Schwefelsäure gut durchgeschüttelt, getrocknet und eingedampft. Das zurückblei- bende I-Benzyloxycarbonyl-4- (2'-propinyl)-4-piperidin- ol siedet bei 160-163¯/0, 01 Torr.
50 g I-Benzyloxycarbonyl-4- (2'-propinyl)-4-piper- idinol werden in einem Gemisch von 500 ml 20"/t) iger SchwefelsÏure und 350 ml Dioxan, das 5 g Quecksilbersulfat enthÏlt, 1 Stunde auf 35-40¯ erwÏrmt. Hierauf wird das Dioxan im Vakuum abgedampft, der Rückstand mit Chloroform ausgezogen, der Chloroformextrakt mit gesättigter Natriumchloridlösung gewaschen, getrocknet und eingedampft. Bei der Destillation des Rückstandes erhält man das 1-(1'-Benzyloxycarbonyl-4'- hydroxy-4'-piperidyl)-2-propanon vom Kp. 180-190 / 0, 03 Torr.
30,8 g 1-(1'-Benzyloxycarbonyl-4'-hydroxy-4'-piperidyl)- 2-propanon werden in 350 ml ¯thanol gel¯st und in Gegenwart von 3 g und spÏter weiteren 1,5 g 5 %iger Palladiumkohle Wasserstoff durchgeleitet, bis kein Kohlendioxyd mehr nachgewiesen werden kann (ca. 5 Stunden). Dann wird der Katalysator abfiltriert, die Lösung eingedampft, der Rückstand in Methylenchlorid aufgenommen, die Lösung getrocknet und eingedampft und der Rückstand im Hochvakuum destil liert. Das 1- (4'-Hydroxy-4'-piperidyl)-2-propanon geht unter 0, 01 Torr bei 80-100 Luftbadtemperatur unter teilweiser Zersetzung über, Smp. 98-100 , vgl. a).
c) 1 g 1-(4'-Hydroxy-4'-piperidyl)-2-propanon (hergestellt nach a) oder b)) wird in 5 ml ¯thanol gel¯st, mit 3 ml Allylbromid versetzt und 24 Stunden stehengelassen. Hierauf wird das Reaktionsgemisch eingedampft, der R ckstand in 2-n. SalzsÏure gel¯st, mit Ather geschüttelt, die wässrige Phase alkalisch gestellt und mit Chloroform ausgezogen. Die Chloroformlösung wird getrocknet, eingedampft und der Rückstand im Hochvakuum destilliert. Das 1- (1'-Allyl-4'-hydroxy-4' piperidyl)-2-propanon geht unter 0, 01 Torr bei 105 bis 125 Luftbadtemperatur über, Smp. des Citrats 75-78 .
Beispiel 2 2 g 1- (4'-Hydroxy-4'-piperidyl)-2-propanon (vgl.
Beispiel la) und lb)) werden mit 4 g n-Hexylbromid, 1 g Kaliumjodid und 1 g Kaliumcarbonat in 25 ml Aceton 4 Stunden unter Rückfluss gekocht. Dann wird das Reaktionsgemisch eingedampft, mit wenig Wasser versetzt und die alkalische Lösung mit Chloroform extrahiert. Die Chloroformlösung wird getrocknet, im Vakuum eingedampft und der Rückstand im Hoch vakuum destilliert, wobei das 1-(1'-n-Hexyl-4'-hydroxy- 4'-piperidyl)-2-propanon vom Kp. 98 /0, 005 Torr er- halten wird.
In analoger Weise werden z. B.
1- (1'-n-Dodecyl-4'-hydroxy-4'-piperidyl)-2-propanon, Kp. 135-138 /0, 09 Torr, unter Verwendung von
6 ml n-Dodecylbromid ; l- (I'-Cyclopropylmethyl-4'-hydroxy-4'-piperidyl)-
2-propanon unter Verwendung von 5 g Jodmethyl cyclopropanon (vgl. J. Am. Chem. Soc. 85, 1886 [1963]), und l-[l'-(y-Phenyl-propyl)-4'-hydroxy-4'-piperidyl]-
2-propanon, Kp. 117-120 /0, 005 Torr, unter
Verwendung von 5 g y-Phenyl-propyl-bromid, erhalten.
Beispiel 3
9, 3 g rohes 4-(2'-Propinyl)-4-piperidinol, erhalten gemäss erstem Abschnitt von Beispiel la), werden mit 50 ml 2-n. Schwefelsäure und 0, 5 g Quecksilber-II-sulfat 15 Stunden bei Raumtemperatur stehengelassen. Dann wird das Reaktionsgemisch mit Natronlauge alkalisch gestellt und mit Chloroform ausgezogen. Die Chloroforml¯sung wird getrocknet und eingedampft. Der R ckstand von 4, 1 g wird in 20 ml Methanol gelöst, mit 10 g Benzylbromid versetzt und 48 Stunden bei 20 stehengelassen. Dann wird das Gemisch unter Vakuum eingedampft, Wasser zugefügt und die sauer reagierende Lösung mit Äther geschüttelt. Die wässrige Phase wird alkalisch gestellt, mit Chloroform ausgezogen, die Chloroformlösung getrocknet und eingedampft und der R ckstand im Hochvakuum destilliert.

The basic condensing agents used are on the one hand inorganic bases such as sodium hydroxide and organic bases such as piperidine, piperazine, and in an anhydrous medium or in the absence of solvents also alkali metal alcoholates, and on the other hand basic ion exchangers, preferably those with quaternary ammonium groups, such as. B. Amberlite IRA 400 (OH63), but also weaker basic ones such as Amberlite-IR 4B, which can be used in batches or, if necessary, also in continuous processes. 12 g of finely powdered hydrochloride of 4-piperidone hydrate (4, 4-piperidinediol hydrochloride) are added to 20 g of finely powdered sodium hydroxide in 80 ml of chloro form at -20. Then 2 ml of water are slowly added, whereupon the temperature rises. By

c) 1 g of 1- (4'-hydroxy-4'-piperidyl) -2-propanone (prepared according to a) or b)) is dissolved in 5 ml of ethanol, 3 ml of allyl bromide are added and the mixture is left to stand for 24 hours. The reaction mixture is then evaporated, the residue in 2-n. Dissolved hydrochloric acid, shaken with ether, made the aqueous phase alkaline and extracted with chloroform. The chloroform solution is dried and evaporated and the residue is distilled in a high vacuum. Example 3
9.3 g of crude 4- (2'-propynyl) -4-piperidinol, obtained according to the first section of Example la), are mixed with 50 ml of 2-n. Sulfuric acid and 0.5 g of mercury (II) sulfate were allowed to stand for 15 hours at room temperature. The reaction mixture is then made alkaline with sodium hydroxide solution and extracted with chloroform. The chloroform solution is dried and evaporated. The residue of 4.1 g is dissolved in 20 ml of methanol, mixed with 10 g of benzyl bromide and left to stand at 20 for 48 hours. The mixture is then evaporated in vacuo, water is added and the acidic solution is shaken with ether. The aqueous phase is made alkaline, extracted with chloroform, the chloroform solution is dried and evaporated and the residue is distilled in a high vacuum.
The 1- (1'-benzyl-4 'hydroxy-4'-piperidyl) -2-propanone boils in the bulb tube below 0.01 Torr at 115-125¯. The hydrochloride melts at 166-168.
In an analogous manner, using 8 g of methyl iodide instead of benzyl bromide, l- (l'-methyl-4'-hydroxy-4'-piperidyl) -2-propanone is obtained, melting point of the citrate 103-105.
Likewise, using 10.5 g of ethyl β-bromopropionate, 1- [l'- (-Afhoxycarbonyläthyl) -4-hydroxy-4'-pipendyU-propanone (4-acetonyl-4-hydroxypiperidine l-propionic acid- ethyl ester) from bp 200-220 /
0.01 Torr (air bath), using 11 g of a-bromo-acetophenone 1- (I'-phenylacyl-4'-hydroxy-4'-piperidyl) propanone, and using 12 g of a-bromo acetanilide 1- (1'-phenylcarbamoylmethyl-4'-hydroxy-4'-piperidyl) propanone (4-acetoxy-4-hydroxypiperidine-1-acetic acid anilide).
