Procédé de préparation d'un produit riche en vitamines du groupe B La présente invention concerne un procédé de pré paration d'un produit riche en vitamines du groupe B, et spécialement en vitamine B12.
Ce procédé est caractérisé en ce que ,l'on cultive du Propionib,acterium shermanmü. et au moins un autre micro-organisme choisi parmi les levures lactiques et l'Empedabacter munsterii dans un sous-produit de L'in- dustrie laitière,
les mücro-organismes étant cultivés en symbiose ou au moins. l'un des micro-organismes étant cultivé dans. un milieu séparé, auquel cas on réunit subséquemment les milieux de culture.
Dans les comptes-rendus de: l'Académie des Sciences (tome 256 p. l164-1165, C. R. Soc. Bio:l. <B>1963,</B> 67, 850 853 et J. Inter. Vitamin. 1963, 33, 31l-320) on a décrit l'isolement die l'Emp@edobacter munsterii, en signalant son pouvoir vitaminogène.
Selon une première forme de réalisation du présent procédé, on ensemence très abondamment du lacto sérum avec le germe Empedobacter, éventuellement additionné de 0,05 à 0,1 g % (en poids frais) de levures lactiques,
on maintient pendant 2 à 4 jours sous agita- tion à une température constante comprise entre 28 C et 30o C, on ajoute au mélange ensemencé et agité du germe Propionibacteriuan sh ermannii, on laisse reposer pendant 2 à 3 jours sans agitation en conservant la même température constante et on recueille le produit obtenu.
Le procédé peut être mis en muvre par lots ou en continu.
Selon une autre mise en oeuvre avantageuse du pro- cédé, on prépare séparément trois volumes égaux de culture de chacun des Em,pedobacter munsterii, levure No 8, Propâonibacteriurn shermannii, on transvase sépa rément chacune de ces cultures dans un volume, 10 à 15 fois plus grand, de. lacto-sérum réduit au quart, on laisse incuber pendant 18 à 24 heures, on mélange les trois cultures et on transforme en poudre dans un ato miseur.
(Tour de séchage de Nuro par exemple.) En opérant sur des cuvas de 15 litres de cultures et de 100 litres de lactosérum réduit au quart, la poudre obtenue répondait aux caractéristiques ci-après

On peut opérer comme suit 1p) Le lactosérum est avantageusement du sérum de Gruyère additionné de 0,25 à 1 g %, de préférence 0,5 g %, de nitrate d'ammonium, de 0,0002 à 0,
004 g 0/0, de préférence 0,003 g %, de chlorure de colbalt et 0,001 g'% de sulfate de manganèse, de mélange étant ajusté à -un pH de 7,
15 à 7,2, précipité par chauffage à 1100 C pendant 15 minutes environ, filtré et pasteurisé à 701) pendant 10 minutes (ou stérilisé pendant 15 mi nutes à 119-120 C).
L'Empedobacter utilisé est préparé à la manière con nue par culture sur grandes boîtes de Pétri (diamètre 150 mm) contenant une gélose nutritive. La quantité d'Empedobacter mise en oeuvre pour l'ensemencement est, en poids frais, de 0,05 à 0,1 g% du lacto-sérum.
Le Propion'bacterium shermannii est préparé par culture sur le milieu de Neronova et eoll. (Microbio logie, 1959, 28, 647, U.R.S.S.) ; on en ajoute de 0,1 à 0,15 g'% du mélange lactosérum Empedobacter.
Au lactosérum on -peut ajouter de 0,05 à 0,1 g 0/0 de levures lactiques vivantes telles que le Saccharo myces fragilis ,<B> </B>Saccharomyces steineri , Saccha romyces marxianus et < c Torulopsis .sp. Ly. <B>369 .</B>
Les levures sont préparées par des cultures à 300 pendant 24 heures de Saccharomyces fragilis , Saccharomyces steineri , Saccharomyces marxianus ou Torulopsis 5p. Ly. <B>369 </B> sur boîtes de Pétri conte nant de la gélose nutritive (diamètre 150 mm).
20) Suivant une autre exécution du procédé selon l'invention, on ensemence l'Empedobacter, P. shermannii et une levure lactique en symbiose sur un milieu minéral additionné de glucose. Les trois mêmes m!iero-orga- nismes sont cultivés pendant 5 à 7 jours, entre 28 et 30 sur ce milieu et on recueille la biomasse résultant de cette culture.

On y ajoute aseptiquement avant l'emploi 2 g 0/0 de glucose (5 ml % d'une solution stérile de glucose à 40 0/0).
La culture en association est réalisée comme suit : sur ledit milieu minéral on cultive pendant 5 à 7 jours l'Empedobacter munsterü à raison de 0,05 % à 0,1% du milieu minéral ;
le P. shermannii à raison de 0,1 à 0,15 % de da culture d'Empedobacter et une levure (Saccharomyces) à raison de 0,05 à 0,
1% de la culture en symbiose.
On opère ensuite comme il a été indiqué plus haut pour le lactosérum.
30) Comme indiqué aux paragraphes 1 et 2, on prépare séparément 15 litres de culture respectivement d'Empedobacter munsterh, de levure Na 8 et de Pro- pionibacterium shermannii.
On transfère séparément ces cultures dans des cuves contenant chacune 100 litres de lactosérum préalable- ment réduit au quart. Après une incubation de 18 à 24 heures, on mélange les contenus des trois cuves et on les transforme en poudre dans un appareil atomiseur d'une vitesse de rotation appropriée.
La poudre est ensuite incorporée, s'il y a lieu, dans des quantités appropriées d'aliments. Pour la mise en oeuvre du présent procédé (exemples 111) et 21)) en continu, on peut utiliser un appareil de type connu, par exemple l'appareil de J.
Monod (An nales de l'Institut Pasteur,<B>1950,</B> 79, 390-410) ou celui de Neronova N.M. et Jerousalimski N.D. (Microbio- log:'a U.R.S.S., 1959, 28, 647-657), qui est à toutes fins utiles décrit ci-après.
A la partie supérieure d'un bâti 1 est monté, de manière réglable en hauteur, un récipient 2 destiné à contenir le milieu nutritif 3 ; 'le récipient 2 est muni de deux branchements 4 et 5 reliés à un tube plongeur 6 et d'un tube 7 pour l'évacuation du gaz (air en général) contenu dans le récipient 2, lors de son remplissage. Des fermetures 4', 5' et 7', automatiques ou non, sont prévues sur chaque tube 4, 5, 7.
Le branchement 5 sert à l'entrée d'air quand on ferme la fermeture 7'. A sa partie inférieure le récipient 2 est muni d'une évacua tion 8 pour le milieu nitritif reliée par une tubulure 9 avec fermeture 9' à un dapositif goutte à goutte 10 porté par le bâti 1.
Le goutte à goutte 10 est équipé d'une tubulure 11 d'évacuation des gaz de fermentation et d'une tuyauterie 12, dont les orifices sur le goutte à goutte sont à un niveau supérieur à. celui de l'extrémité inférieure libre de la tubulure 9.
Au-dessous du goutte à goutte 10, le bâti 1 porte un premier cultivateur 13 de forme générale cylindrique et traversé axialement .par un tube capillaire plongeur 14 formant par son extrémité :supérieure un prolongement du goutte à goutte 10. Le cultivateur 13 est relié d'une part au goutte à goutte 10 par une tuyauterie 15 permet tant de faire passer ,les gaz de fermentation du cultiva teur 13 dans 'le goutte à goutte 10 débouchant dans ce goutte à goutte en 12 plus haut que l'extrémité infé rieure de la tubulure 9 et, d'autre part,
à un récipient 17 de recueil du liquide deRTI ID="0002.0219" WI="11" HE="4" LX="1543" LY="1847"> culture par un tube 18 avec fermeture 18' permettant des prises d'échantillons. Enfin, le cultivateur 13 est muni d'une tuyauterie 19 permettant le .reflux d'un excès de liquide de culture dans un second goutte à goutte 20, monté sur le bâti 1 à un niveau inférieur. Le goutte à goutte 20 est équipé, comme le goutte à goutte 10, d'une tubulure 22 d'éva cuation des gaz de fermentation,
d'une tuyauterie 23 et d'un capillaire axial 22.
Le bâti 1 porte, enfin, au-dessous du goutte à goutte 20, un second cultivateur 25 à trois volumes différents (dans l'exemple représenté), traversé axialement par le prolongement plongeur 24 du capillaire réuni au goutte à goutte 20 par la tuyauterie 23 et muni de trois éva cuations deliquide de culture 26, 27, 28, qui correspon dent chacune avec un récipient de recueil 29 et permet tent la prise d'échantillons aux niveaux supérieur,
inter médiaire et inférieur de chaque volume du cultivateur 25. Les évacuat'io'ns 26, 27, 28 comportent chacune une fermeture 26', 27' et 28'. Enfin une tuyauterie 30 avec fermeture 30' relie la partie supérieure du cultivateur 25 à un récipient 16 contenant par exemple du carbonate de sodium. Les divers éléments de l'appareil peuvent être mis séparément en température et/ou agités, .par tous moyens appropriés connus.
Les diverses fermetures 4', 5', 7', 9', 18', 26', 2T, 28', 30' sont du type à vis héli coïdale.
Dans le récipient 2 où le an:lieu nutritif arrive par la tubulure 4 on maintient une :pression hydrostatique constante à l'aide de :la tubulure 5-6 le reliant à l'atmo sphère. La vs qui est sur la tubulure 7 est alors fermée.
La vitesse d'écoulement du milieu dépend de la dif férence de niveau du liquide entre l'extrémité inférieure du tube 6 et l'extrémité du tube 9 d'où le milieu s'écoule goutte à goutte. Pour réaliser la vitesse désirée on établit un niveau convenable d'écoulement du compte-goutte à l'aide d'un dispositif spécial 31 monté sur la tablette 1, ,puis on<B>l</B>e règle définitivement au moyen d'une vis hélicoïdale 9'.
Pour contrôler cette vitesse @on mesure le volume du liquide fermenté qui s'écoule de la tubulure 18. On peut utiliser soit uniquement le premier cultiva- ,tcu:r 13 ou, lorsqu'on veut augmenter le mendement, on peut utiliser en même temps le deuxième cultivateur 25 en faisant passer le 1:quide nutritif à travers la tubu lure 19 et :le compte-goutte 20 dans le cultivateur 25.
Ce cultivateur possède trois tubulures de dérivation 26, 27 et 28 à des niveaux différents. Selon que l'on met en circuit l'une ou l'autre de ces tubulures la capacité utile du cultivateur est plus ou moins élevée.
On a constaté qu'en opérant selon l'invention, l'enri- chissement en vitamines, en particulier du groupe B et spécialement en vitamine Blé, était beaucoup plus élevé qu'en cultivant uniquement l'Empedobacter. Ceci ressort des tableaux suivants :dans lesquels les teneurs en vita mines sont exprimées en Rg par litre.
Pour les exemples 1 et 2, le tableau I donne ales résultats obtenus avec l'Empedobacterseul, les tableaux 11, III et IV concer nant les résultats fournis par des procédures selon l'in vention. Les tableaux comparatifs V et VI donnent les résultats correspondants obtenus respectivement selon l'exemple 30) ci-dessus et selon une méthode usuelle.

On utilise de préférence une quantité desdits produits allant de 3 à 5 % en poids par rapport à l'aliment considéré.
La poudre obtenue est enrichie en vitamines B, notamment en vitamine 1312, d'un goût agréable légère ment sucré, presque totalement soluble dans d'eau, et de couleur blanc légèrement jaunâtre.
Cette poudre, qui ne se prend pas en masse, est défi nie par les caractéristiques suivantes a) protéines totales de 13 à 14'0/0 lactose anhydre de 73 à 74'% lactose hydraté de 76 à 78 % sels (d'après les cendres à 5500) :
4 % environ pH reconstitué à 10 1% : 6,3 à 6,4 acidité 17 à 18 degrés Dornic humidité 2 1% poids sec 98 0/0 b) teneur en vitamines (en maligrammes par 1000 g de poudre)

On constate que l'enrichissement effectué avec du lacto-sérurn réduit au 1/4, sans levure, a un effet plus prononcé sur la teneur finale de la poudre en vita mine Bit.
On peut donc profiter de cette circonstance pour préparer des aliments diététiques à prédominance de vitamine Biz.

Process for preparing a product rich in group B vitamins The present invention relates to a process for preparing a product rich in group B vitamins, and especially vitamin B12.
This method is characterized in that, one cultivates Propionib, acterium shermanmü. and at least one other microorganism selected from lactic yeasts and Empedabacter munsterii in a by-product of the dairy industry,
the microorganisms being cultivated in symbiosis or at least. one of the microorganisms being grown in. a separate medium, in which case the culture media are subsequently combined.
In the reviews of: l'Académie des Sciences (tome 256 p. L164-1165, CR Soc. Bio: l. <B> 1963, </B> 67, 850 853 and J. Inter. Vitamin. 1963, 33, 31l-320) the isolation of Emp @ edobacter munsterii has been described, pointing out its vitaminogenic power.
According to a first embodiment of the present method, very abundantly inoculated whey with the Empedobacter germ, optionally supplemented with 0.05 to 0.1 g% (by fresh weight) of lactic yeasts,
it is maintained for 2 to 4 days with stirring at a constant temperature between 28 ° C. and 30 ° C., the seeded and stirred mixture is added to the seeded and stirred mixture of the germ Propionibacteriuan sh ermannii, the mixture is left to stand for 2 to 3 days without stirring, keeping the same constant temperature and the product obtained is collected.
The process can be carried out in batches or continuously.

The procedure is then carried out as indicated above for the whey.
30) As indicated in paragraphs 1 and 2, 15 liters of culture respectively of Empedobacter munsterh, of yeast Na 8 and of Pro- pionibacterium shermannii are prepared separately.
These cultures are transferred separately to tanks each containing 100 liters of whey previously reduced to a quarter. After an incubation of 18-24 hours, the contents of the three tanks are mixed and powdered in an atomizing apparatus of an appropriate speed of rotation.
