Verfahren zur Herstellung von Diolen
Gegenstand der vorliegenden Erfindung ist ein verbessertes Verfahren zur Herstellung von Diolen aus Ketonen.
Es ist bekannt, dass bei der kathodischen Reduktion von Carbonyl-Verbindungen, wie Aldehyden und besonders aliphatischen, aromatischen oder heterocycli- schen Ketonen, wie auch gemischten Ketonen, z. B. ali phatisch-aromatischen, aliphatisch-heterocyclischen oder aromatisch-heterocyclischen Ketonen, Diole gebildet werden (Allen, Organic Electrode Processes, Seiten 61-67, Chapman & Hall, London, 1958). Die entste- henden Diole sind an sich wertvoll oder können als Zwischenprodukte für die Herstellung anderer wertvoller Verbindungen dienen.
Bisher wurde die elektrolytische Reduktion von Car bonyl-Verbindungen stets so ausgeführt, dass der Katholyt mit der zu reduzierenden Carbonylverbindung, z. B. einem Keton, und der Kathode vom Anolyt, enthaltend die Anode, durch eine semi-permeable Membran, wie z. B. Cellophan, poröses Porzellan (Alundum), gesintertes Glas und ähnliches getrennt war.
Obwohl in der Literatur für die Bildung von Diolen auf diesem Weg bei kleinen Laboratoriumsversuchen befriedigende Ausbeuten angegeben sind, hat sich die elektrolytische Reduktion von Carbonylverbindungen, besonders der oben erwähnten, zu den gewünschten Diolen unter Verwendung dieses Zweikammer-Systems, d. h. unter Tirennung des Katholyts vom Anolyt durch ein Diaphragma als durchaus unzulänglich erwiesen, sobald sie in grösserem Massstab ausgeführt werden musste. Das bei einer solchen elektrolytischen Reduktion direkt anfallende Produkt ist sehr unrein und muss mehreren Reinigungsschritten unterzogen werden, wobei die Ausbeuten be trächtlich sinken.
Sie betragen an gereinigtem Diol, ins- beson, dere wenn es sich um ein heterocyclisches Diol handelt, nur ungefähr 20-30 /o und sind offensichtlich unbefriedigend.
Eine MögLichkeit, die beim Arbeiten in einem Zwei kammer-System erhaltenen unbefriedigenden Ausbeuten zu erhöhen, ist manchmal die Einkammer-Arbeitsweise.
Elektrolytische Einkammer-Verfahren sind bekannt, insbesondere aus dem Gebiet der anorganischen Chemie. Angesichts der Tatsache, dass das Reduktionsprodukt an der Anode leicht wieder oxydiert werden kann, ist diese Methode auf dem Gebiet der organischen Chemie nur in beschränktem Umfang angewendet worden. Sie wurde z. B. dann angewendet, wenn eine Rückoxydation des Reaktionsproduktes aus strulkturelle Gründen nicht möglich war, oder wenn die Rüokoxyda- tion durch die Zugabe bestimmter Reagenzien verhindert werden konnte. Im vorliegenden Fall jedoch, welcher eine gleichzeitige Dimerisierung und Reduktion der Ketone zu Diolen betrifft, war die Möglichkeit einer befriedigenden Verwendung des Einkammer-Systems aus naheliegenden Gründen kaum erfolgversprechend.
Die Reaktionsprodukte, d. h. die Diole werden nämlich leicht in die Ausgangsmaterialien, d. h. die Ketone zu rückoxydiert. Man konnte deshalb annehmen, dass das entstandene Diol von der Anode durch eine Membran zu trennen sei, d. h., dass ein Zweikammer-System verwendet werden müsse. Die Zugabe chemischer Reduk tionsmittel, welche die Rückoxydation der erhaltenen Diole verhindern könnte, erschien nicht erfolgverspre- chend, da die Diole sehr labile Verbindungen sind, welche leicht mit anderen chemischenr Verbindungen Reaktionen eingehen können.
Darüber hinaus wäre auch zu befürchten gewesen, dass ein zur Verhinderung der Rückoxydation zugesetztes Reduktionsmittel das Aus gangsketon zu einem Alkohol reduzieren würde, ohne gleichzeitig die erwünschte Dimerisierung herbeizufüh- ren, wie sie bei Verwendung der Zweikammer-Kathode erfolgt.
Entgegen diesen verschiedenen Befürchtungen wurde nun gefunden, dass Diole durch elektrolytische Reduktion von Ketonen oder Mischungen von Ketonen in vereinfachter Weise und mit befriedigender Ausbeute gewonnen werden können.
Das erfindungsgemässe Verfahren zur Herstellung von Diolen besteht darin, dass man die Ketone in einem Elektrolytmedium ohne Reduktion des Elektrolytme diums der eloktrolytischen Reduktion in einer Einkam mer-Elektrolyse-Zelle, d. h. in Abwesenheit eines Dia phragmas, unterwirft.
Es ist besonders hervorzuheben, dass die Abwesen- heit eines Diaphragmas die Konstruktion des Elektrolyse-Apparates vereinfacht, da ja nun die Reaktion in einem Einkammer-Gefäss ausgeführt werden kann. Die Installation eines Diaphragmas wird besonders mühsam in einem grosstechnischen Arbeitsgerät, in welchem das semi-permeable Diaphragma zwischen den beiden Kammern sehr dicht befestigt werden muss, um jegliches Vermischen von Anolyt mit Katholyt zu verhindern.
Das Wegfallen des Diaphragmas stellt einen augenfälli- gen Vorteil über die beschriebenen Methoden dar. Dazu kommt noch, dass bei der Reduktion gebildete Diole im Verlaufe der Reaktion ausfallen und die Membrane verstopfen können, wodurch ein Anstieg des elektrischen Widerstandes und ein Fall der Stromausbeute (Allen, loc. cit., Seiten 31-33) eintritt und kostspieliges Auswechseln der Membran nötig werden. Darüber hinaus besteht bei dem Einkammer-Verfahren die Möglichkeit, den Strom von Zeit zu Zeit zu alternieren. Damit kann die Verstopfung der Kathode durch ausgefälltes Produkt verhindert werden und die Elektroden können während der Reduktion gereinigt werden.
Nach Beendigung der Einkammer-Elektrolyse können die Elektroden durch einfaches Umkehren des Stromes gesäubert werden, während beim Zweikammer-Verfahren entweder die Elektroden oder das Diaphragma zur Reinigung entfernt werden mussen.
Weiter wurde gefunden, dass in Abwesenheit eines Diaphragmas die Ausbeuten an reinem Diol wesentlich höher liegen als beim alten Einkammer-Verfahren. Insbesondere ist das anfallende Diol von weitaus höherem Reinheitsgrad, so dass es ohne langwierige Reinigungsverfahren verwendet werden kann. Dies ist besonders wichtig, da bei der Pinacolon-Umlagerung erhaltener Diole, insbesondere solcher des Amphenon-Typs, die Ausbeuten unerwarteterweise bemerkenswert höher liegen, wenn von einem sehr reinen Diol ausgegangen wird. Diesen Anforderungen entsprechendienachdem vorliegenden Verfahren erhältlichen Diole in besonderem Masse.
Die erfindungsgemässe Einkammer-Elektrolyse kann in Wasser allein als Elektrolytmedium vorgenom- men werden. Vorzugsweise verwendet man jedoch ein Medium, das aus einer Mischung von Wasser und einem unter Reaktionsbedingungen nicht reduzierbaren, wassermischbaren organischen Lösungsmittel besteht. Als organische Lösungsmittel kommen dabei z. B. niedere Alkanole, vorzugsweise solche mit 1-4 Kohlenstoffatomen, wie Methanol, Athanol, n-Propanol oder Isopropanol oder tert. Butanol in Frage ; femer Formamide, wie N, N-Dimethylformamid und ähnliche, oder irgendein anderes braudhbaresorgamsdbesLösungsmittel, wie Eisessig u. dgl. Bevorzugt sind Lösungsmittel mit einem relativ hohen Siedepunkt.
Daher erscheint Isopropanol oder N, N-Dimethylformamid besonders geeignet. Mit Vorteil kann man eine Misohung von Isopro- panol oder N, N-Dimethylformamid und Wasser verwenden. Allfällige Elektrolytverluste während der Elektrolyse können durch Zugabe von Wasser ausgeglichen werden.
Um die Leitfähigkeit des Elektrolytmediums weiter zu erhöhen ist es vorteilhaft, ein ionisierbares Mittel zuzufügen. Als ionisierbare Mittel kommen vor allem Metallsalze von organischen-Carbonsäuren, wie Alkali metall-, z. B. Kaliumsalze von niederen Fettsäuren, z. B.
Ameisensäure, Essigsäure, Propionsäure usw. in Frage.
Besonders zu nennen sind Alkaliacetate, vor allem Kaliumacetat.
Ein besonders bevorzugtes Elektrolytmedium für das Einkammer-Verfahren der vorliegenden Erfindung ist eine Mischung von Isopropanol oder N, N-Dimethylformamid, zusammen mit Wasser und Kaliumacetat. Die Komponenten dieser Mischung können natürlich durch ihre Aquivalente ersetzt werden.
Die Elektrolyse als solche wird z. B. in an sich bekannter Weise durchgeführt. Die Kathode, an welcher die Reduktion erfolgt, kann z. B. irgendeine entsprechende Metall-Kathode sein. Sie kann z. B. aus Zinn, Nickel, Blei, Kupfer, Kadmium, Zink, Eisen, rostfreiem Stahl oder Quecksilber sein, und wird zusammen mit einer geeigneten Anode, die z. B. aus Nickel, Blei oder Platin besteht, verwendet.
Die Elektrolyse wird vorzugsweise bei einer konstanten Stromdichte durchgeführt. Letztere ist bestimmt durch : a) die Grosse der Elektroden, b) die Art des Gerätes, c) die Möglichkeit von Nebenreaktionen, wie z. B.
Gasentwicklung und ähnlidhes.
Eine bevorzugte Stromdichte liegt in der Gegend von ungefälhr 0, 01 amp/cm2 bis 0, 1 amp/cm2, besonders zwischen 0, 02 und 0, 04 amp/cm2. Man kann. die elektrolytische Reduktion bei Zimmertemperatur aus- führen. Zweckmässig arbeitet man jedoch bei erhöhter Temperatur, z. B. zwischen 40 und 80 , insbesondere bei 60-70 . Die Ausgangsstoffe, die bei dem elektroly- tischen Verfahren der vorliegenden Erfindung mit Vorteil eingesetzt werden, sind Ketone, wie z.
B. aliphatische Ketone, vor allem niedere Alkanone, wie Aceton, Methyl-äthyl-keton, Diäthyl-keton und ähnliche, niedere Alkyl-alkenyl-ketone, wie Äthyl-vinyl-keton, cycloaliphatische Ketone, wie Dicyclopentyl-keton oder Dicy- clohexyl-keton, aliphatisch-cycloaliphatische, wie Cycloalkyl-niederalkyl-ketone, z. B. Cyclopentyl-meth- yl-keton oder Cyclohexyl-äthyl-keton, aliphatisch-cyclo- aliphatisch-aliphatische Ketone wie Cyclopentyl-meth- yl methyl-keton oder 2-Cyclohexyläthyl-äthyl-keton oder andere Ketone ähnlicher Natur.
Bevorzugte Ketone sind jedoch aliphatisch-aromati- sche Ketone, vor allem Niederalkyl-phenyl-ketone, in welchen der Phenylrest unsubstituiert oder substituiert sein kann, z. B. durch Kohlenwasserstoffreste, wie niederes Alkyl, z. B. Methyl-Athyl, Isopropyl oder Butyl, oder durch Hydroxyl, Niederalkoxy, wie Methoxy, Athoxy, Isopropyloxy oder Butyloxy, oder Niederalkenyloxy, wie Allyloxy oder 2-Butenyloxy, oder durch Halogen wie Fluor, Chlor, Brom oder das Pseudohalogen Trifluormethyl. Bevorzugte Substitnenten des Phenylrestes sind jedoch Aminogruppen, wie z.
B. unsubstituierte, mono-oder disubstituierte Aminogruppen, vor allem N-Niederalkylamino, wie N-Methylamino, N Äbhylamino, N-Isopropylamino oder N, N-Diniederalk- ylamino, wie N, N-Dimethylamino, N-lAthyl-N-methyl- amino, N, N-Diäthylamino, oder N-Acylamino, wie z. B.
N-Niederalkanoylamino, wie N-Acetylamino oder N-Propionylamino.
Weitere Ketone, die mit besonderem Vorteil als Ausgangsstoffe im Verfahren der vorliegenden Erfindung verwendet werden können, sind aliphatisch-hete- roxyclische Ketone, wie besonders Niederalkyl-pyridyl- ketone, z. B. 2-Acetylpyridin, 3-Acetylpyridin, 3-Pro pionylpyridin oder 4-Acetylpyridin oder irgendein anderes aliphatisch-heterocyclisches Keton, wie z. B. ein Alkylpyrimidylketon. Weiter kann man auch verwenden Diaryl-ketone wie Diphenylketone (Benzophenone), in welchen die Phenylgruppen unsubstituiert oder wie oben angogeben substituiert sind, z. B. Benzophenon, oder 4-Amino-benzophenon, oder auch diheterocyclische Ketone, wie Dipyridyl-ketone, z.
B. Di-3-pyridyl-keton, 3-Pyridyl-4-pyridyl-keton oder Di-4-pyridyl-keton oder aryl-heterocyclische Ketone, wie Phenyl-pyridyl-ketone, in welchen der Phenylrest gegebenenfalls substituiert sein kann, wie z. B. oben angegeben, vor allem 3-Benz oyl-pyridin, 4-p-Chlorbenzoyl-pyridin, 3p-Aminobenz- oyl-pyridin und ähnliche.
Anstelle eines einzelnen Ketons kann auch eine Mischung von 2 verschiedenen Ketonen eingesetzt werden. So ergibt z. B. eine Mischung eines Niederalkylphenyl-ketons und eines Niederalkyl-pyridyl-ketons bei der verfahrensgemässen Reduktion eine Mischung von Diolen, aus der das entsprechende asymmetrische Diol, nämlich ein Phenyl-pyridyl-keton isoliert werden kann.
Das Verfahren dieser Erfindung ist in erster Linie vorteilhaft für die Herstellung von 2, 3-Bis- (3-pyridyl)2, 3-butandiol durch elektrolytische Reduktion des 3 Acetylpyridins.
Die gemäss dem Verfahren der vorliegenden Erfin dung hergestellten Diole sind nützliche Ausgangsstoffe für die Herstellung von Ketonen mittels der Pinacolon- Umlagerung oder für die Herstellung von Indenen mittels doppelter Dehydratisierung und Ringschluss.
Einige der so erhaltenen Diole, Ketone oder Indene zeigen aussergewöhnliche Hemmeffekte auf die Nebennie renfunktion und können daher als Heilmittel oder für diagnostische Zwecke angewendet werden. Besonders wertvoll für diesen Verwendungszweck sind 3, 3-Bis- (4-amino-phenyl)-butan-2-on (Amphenon B), 1, 2-Bis- (3-pyridyl)-2-methyl-propan-l-on (Methopyrapon), welche durch Inacolon-UmIagerung aus 2, 3-Bis-(4-amino-phenyl)-2, 3-butandiol bzw.
2, 3-Bis (3-pyridyl)-2, 3-butandiol erhalten werden können, und ebenso auch 6-Chloro-2- (3-pyridyl)-inden, welches durch doppelte Dehydratisierung und gleichzeitigen Ringschluss in Gegenwart einer starken Säure, wie Salzsäure aus 2- (4-Chloro-phenyl)-3- (3-pyridyl)-2, 3-butandiol erhalten werden kann.
Beim Verfahren der vorliegenden Erfindung werden vorteilhaft solche Ausgangsmaterialien eingesetzt, welche zu den oben hervorgehobenen Endprodukten führen.
Die Temperaturen sind in den nachfolgenden Beispielen in Celsiusgraden angegeben.
Beispiel 1
Eine Lösung von 24, 23 g 3-Acetyl-pyridin und 30 g Kaliumacetat in 100 cm3 N, N-Dimethylformamid und 45 cm3 Wasser wird als Elektrolyt in eine Einkammer Elektrolyse-Zelle gegeben. Als Kathode wird eine Zinnelektrode mit einer Oberfläche von 100 cm (vorher mit Salpetersäure gereinigt) verwendet und als Anode plaziert man eine Niokelelektrode von 12 cm2 Oberflä- che in den Elektrolyt. Bei einer Temperatur von 67-69 leitet man nun einen Strom von 4, 1-4, 4 amp durch die Losung. Während der Elektrolyse gibt man 18 ml Wasser in Portionen zu, um den Flüssigkeitsspie- gel konstant zu halten.
Nach 80 Minuten entsteht ein Niederschlag, und die Elektrolyse wird unterbrochen, sobald 11, 9 Amp.-Stunden abgelaufen sind.
Hierauf wird der Elektrolyt mit 100 ml Wasser ver dünnt, während 64 Stun, den auf-8 abgekühlt und filtriert. Das feste Material wird viermal mit je 30 ml Wasser gewaschen und bei vermindertem Druck während 16 Stunden bei 100 getrocknet. Man erhält so 12, 65 g 2, 3-Bis-(3-pyridyl)-2, 3-butandiol, F. 218-225 .
Beispiel 2
Man verfährt wie in Beispiel 1, wobei man jedoch als Kathode eine Nickelkathode verwendet und eine Nickelanode von 36 cm2 Oberfläche benützt.
Nachdem die so erhaltene Reaktionsmischung wäh- rend 16 Stunden auf-8 gekühlt worden ist, filtriert man sie und wäscht den Niederschlag viermal mit je 30 ml Wasser und trocknet ihn bei 70 im Vakuum während 16 Stunden. Man erhält so 13, 9 g 2, 3-Bis (3-pyridyl)-2, 3-butan, diol vom F. 224-230 .
Andere Niederalkanoylpyridine, wie z. B. 3-Propionv ylpyridin, 4-Acetylpyridin oder aliphatisdh-aromatische Ketone, vor allem Niederalkyl-phenyl-ketone wie 4-Chlorbacetophenon oder 4-Amino-acetophenon oder Mischungen der genannten Ketone können in gleicher Weise verwendet werden, wobei die entsprechenden Diole erhalten werden.
Beispiel 3
Eine Lösung von 36, 3 g 3-Acetylpyridin und 30 g Kaliumacetat in 62, 5 ml deionisiertem Wasser und 82, 5 ml Isopropanol wird in eine Einkammer-Elektrolyt-Zelle gebracht, die mit einer zylindrischen Nickelthode (100 cm2 Oberfläche) und einer kleineren zylindrischen Nickelanode von 36 cm2 Oberfläche versehen ist, wobei die kleinere Anode innerhalb der Kathode zu stehen kommt. Die Reduktion wird bei 62 und einer Stromdichte von 0, 03 amp./cm2 der Kathode ausgeführt. Nach 6 Stunden wird die Reduktion abgebrochen.
Die Reaktionsmischung wird mit 100 ml Wasser versetzt und 16 Stunden bei Raumtemperatur stehen gelassen.
Hierauf wird abfiltriert und der Filterrückstand mit Wasser gewaschen. Man erhält so 29, 4 g reines 2, 3-Bis (3-pyridyl)-2, 3-butandiol.
Beispiel 4
Eine Lösung von 5030 g 3-Acetylpyridin und 6290 g Kaliumacetat in einer Mischung von 15 200 ml Isoprop- anol und 15 200 ml deionisiertem Wasser wird unter Verwendung einer rechteckigen Nickelelektrode (von 7000 cm2 Oberfläche) und einer Nickelanode von 2000 cm2 Oberfläche, der elektrolytischen Reduktion unterworfen. Die Reaktion wird bei 62 und 0, 04 amp/cm2 der Kathode ausgeführt und nach 9 Stunden abgebrochen. Die Reaktionsmischung wird mit 20 000 ml Wasser versetzt und nach dem in Beispiel 3 beschriebenen Verfahren aufgearbeitet. Man erhält so 3240 g reines 2, 3-13is43-pyridyl)-2, 3-butandiol.
Beispiel 5
Eine Mischung von 66, 6 g 3-Acetylpyridin und 82, 5 g Kaliumacetat in 200 ml deionisiertem Wasser und 200 ml Isopropanol wird in eine rechteckige Zelle gebracht ; es werden identische flache nickelektroden, die je 100 em, Oberfläche aufweisen, verwendet. Sie sind mit dem Stromanschluss durch einen Stromwender verbunden, so dass die Rolle der Kathode und der Anode ohne Unterbrechung der Reaktion ausgewechselt werden kann. Die Elektrolyse wird bei 62 und 0, 04 amp/cm2 der Elektrode während 8 Stunden und 20 Minuten ausgeführt, wobei man die Richtung des Stromflusses alle 30 Minuten umkehrt.
Das 2, 3-Bis (3-pyridyl)-2, 3-butandiol wird durch Versetzen der erhaltenen Lösung mit 275ml Wasser isoliert und nach dem in Beispiel 3 beschriebenen Verfahren aufgearbei tet. Man erhält so 43, 5 g reines Produkt.
Process for the preparation of diols
The present invention relates to an improved process for the preparation of diols from ketones.
It is known that in the cathodic reduction of carbonyl compounds such as aldehydes and especially aliphatic, aromatic or heterocyclic ketones, as well as mixed ketones, e.g. B. aliphatic-aromatic, aliphatic-heterocyclic or aromatic-heterocyclic ketones, diols are formed (Allen, Organic Electrode Processes, pages 61-67, Chapman & Hall, London, 1958). The resulting diols are valuable in themselves or can serve as intermediates for the production of other valuable compounds.
So far, the electrolytic reduction of carbonyl compounds has always been carried out so that the catholyte with the carbonyl compound to be reduced, eg. B. a ketone, and the cathode of the anolyte containing the anode, through a semi-permeable membrane, such as. B. cellophane, porous porcelain (Alundum), sintered glass and the like was separated.
Although the literature gives satisfactory yields for the formation of diols in this way in small laboratory tests, the electrolytic reduction of carbonyl compounds, especially those mentioned above, to the desired diols using this two-chamber system, i.e. H. with the separation of the catholyte from the anolyte by a diaphragm proved to be absolutely inadequate as soon as it had to be carried out on a larger scale. The product obtained directly from such an electrolytic reduction is very impure and has to be subjected to several purification steps, with the yields dropping considerably.
They amount to purified diol, especially if it is a heterocyclic diol, only about 20-30 / o and are obviously unsatisfactory.
One way of increasing the unsatisfactory yields obtained when working in a two-chamber system is sometimes the one-chamber procedure.
Electrolytic single-chamber processes are known, in particular from the field of inorganic chemistry. In view of the fact that the reduction product can easily be reoxidized at the anode, this method has only been used to a limited extent in the field of organic chemistry. She was z. B. used when re-oxidation of the reaction product was not possible for structural reasons, or when re-oxidation could be prevented by adding certain reagents. In the present case, however, which concerns a simultaneous dimerization and reduction of the ketones to diols, the possibility of a satisfactory use of the one-chamber system was hardly promising for obvious reasons.
The reaction products, i.e. H. namely, the diols are easily converted into starting materials, i.e. H. the ketones are reoxidized. One could therefore assume that the diol formed could be separated from the anode by a membrane, i.e. This means that a two-chamber system must be used. The addition of chemical reducing agents, which could prevent the reoxidation of the diols obtained, did not appear to be promising, since the diols are very labile compounds which can easily react with other chemical compounds.
In addition, it would have been feared that a reducing agent added to prevent reoxidation would reduce the starting ketone to an alcohol without simultaneously bringing about the desired dimerization, as occurs when using the two-chamber cathode.
Contrary to these various fears, it has now been found that diols can be obtained in a simplified manner and with a satisfactory yield by electrolytic reduction of ketones or mixtures of ketones.
The inventive method for the production of diols consists in that the ketones in an electrolyte medium without reduction of the Elektrolytme medium of the electrolytic reduction in a single-chamber electrolysis cell, d. H. in the absence of a diaphragm, subject.
It should be emphasized that the absence of a diaphragm simplifies the construction of the electrolysis apparatus, since the reaction can now be carried out in a single-chamber vessel. The installation of a diaphragm becomes particularly troublesome in a large-scale technical device in which the semi-permeable diaphragm must be fastened very tightly between the two chambers in order to prevent any mixing of anolyte with catholyte.
The elimination of the diaphragm is an obvious advantage over the methods described. In addition, the diols formed during the reduction precipitate in the course of the reaction and can clog the membrane, causing an increase in electrical resistance and a decrease in current yield (Allen , loc. cit., pages 31-33) occurs and expensive replacement of the membrane becomes necessary. In addition, the single-chamber method offers the option of alternating the flow from time to time. This can prevent the cathode from becoming clogged by precipitated product and the electrodes can be cleaned during the reduction.
After the end of the single-chamber electrolysis, the electrodes can be cleaned by simply reversing the current, while with the two-chamber method either the electrodes or the diaphragm must be removed for cleaning.
It was also found that in the absence of a diaphragm, the yields of pure diol are significantly higher than with the old one-chamber process. In particular, the resulting diol is of a much higher degree of purity, so that it can be used without lengthy purification processes. This is particularly important since the diols obtained in the pinacolone rearrangement, in particular those of the amphenone type, are unexpectedly considerably higher when starting from a very pure diol. Diols obtainable by the present process meet these requirements to a particular extent.
The single-chamber electrolysis according to the invention can be carried out in water alone as the electrolyte medium. Preferably, however, a medium is used which consists of a mixture of water and a water-miscible organic solvent which is not reducible under the reaction conditions. As organic solvents, for. B. lower alkanols, preferably those with 1-4 carbon atoms, such as methanol, ethanol, n-propanol or isopropanol or tert. Butanol in question; furthermore formamides, such as N, N-dimethylformamide and the like, or any other brewable organic solvent such as glacial acetic acid and the like. The like. Solvents with a relatively high boiling point are preferred.
Isopropanol or N, N-dimethylformamide therefore appear particularly suitable. A mixture of isopropanol or N, N-dimethylformamide and water can advantageously be used. Any electrolyte losses during electrolysis can be compensated for by adding water.
In order to increase the conductivity of the electrolyte medium further, it is advantageous to add an ionizable agent. As ionizable agents are mainly metal salts of organic carboxylic acids, such as alkali metal, z. B. potassium salts of lower fatty acids, e.g. B.
Formic acid, acetic acid, propionic acid, etc. in question.
Particular mention should be made of alkali acetates, especially potassium acetate.
A particularly preferred electrolyte medium for the single chamber process of the present invention is a mixture of isopropanol or N, N-dimethylformamide, along with water and potassium acetate. The components of this mixture can of course be replaced by their equivalents.
The electrolysis as such is z. B. carried out in a known manner. The cathode on which the reduction takes place can, for. B. be any corresponding metal cathode. You can z. B. tin, nickel, lead, copper, cadmium, zinc, iron, stainless steel or mercury, and is used together with a suitable anode, the z. B. made of nickel, lead or platinum is used.
The electrolysis is preferably carried out at a constant current density. The latter is determined by: a) the size of the electrodes, b) the type of device, c) the possibility of side reactions, such as B.
Gas evolution and the like.
A preferred current density is in the region of about 0.01 amps / cm2 to 0.1 amps / cm2, especially between 0.02 and 0.04 amps / cm2. One can. carry out the electrolytic reduction at room temperature. However, it is expedient to work at an elevated temperature, e.g. B. between 40 and 80, especially at 60-70. The starting materials which are used with advantage in the electrolytic process of the present invention are ketones, such as.
B. aliphatic ketones, especially lower alkanones such as acetone, methyl ethyl ketone, diethyl ketone and similar, lower alkyl alkenyl ketones such as ethyl vinyl ketone, cycloaliphatic ketones such as dicyclopentyl ketone or dicyclohexyl -ketone, aliphatic-cycloaliphatic, such as cycloalkyl-lower alkyl-ketones, e.g. B. cyclopentyl-methyl-ketone or cyclohexyl-ethyl-ketone, aliphatic-cyclo-aliphatic-aliphatic ketones such as cyclopentyl-meth- yl methyl-ketone or 2-cyclohexylethyl-ethyl-ketone or other ketones of a similar nature.
However, preferred ketones are aliphatic-aromatic ketones, especially lower alkyl-phenyl-ketones, in which the phenyl radical can be unsubstituted or substituted, eg. B. by hydrocarbon radicals such as lower alkyl, e.g. B. methyl-ethyl, isopropyl or butyl, or by hydroxyl, lower alkoxy, such as methoxy, ethoxy, isopropyloxy or butyloxy, or lower alkenyloxy, such as allyloxy or 2-butenyloxy, or by halogen such as fluorine, chlorine, bromine or the pseudohalogen trifluoromethyl. Preferred substituents of the phenyl radical, however, are amino groups, such as.
B. unsubstituted, mono- or disubstituted amino groups, especially N-lower alkylamino, such as N-methylamino, N-ethylamino, N-isopropylamino or N, N-di-lower alkylamino, such as N, N-dimethylamino, N-ethyl-N-methyl - amino, N, N-diethylamino, or N-acylamino, such as. B.
N-lower alkanoylamino, such as N-acetylamino or N-propionylamino.
Further ketones which can be used with particular advantage as starting materials in the process of the present invention are aliphatic-heteroxyclic ketones, such as, in particular, lower alkyl pyridyl ketones, e.g. B. 2-acetylpyridine, 3-acetylpyridine, 3-propionylpyridine or 4-acetylpyridine or any other aliphatic-heterocyclic ketone, such as. B. an alkyl pyrimidyl ketone. Next you can also use diaryl ketones such as diphenyl ketones (benzophenones), in which the phenyl groups are unsubstituted or substituted as indicated above, z. B. benzophenone, or 4-amino-benzophenone, or diheterocyclic ketones, such as dipyridyl ketones, z.
B. Di-3-pyridyl-ketone, 3-pyridyl-4-pyridyl-ketone or di-4-pyridyl-ketone or aryl-heterocyclic ketones, such as phenyl-pyridyl-ketones, in which the phenyl radical can optionally be substituted, such as z. B. stated above, especially 3-benzoyl-pyridine, 4-p-chlorobenzoyl-pyridine, 3p-aminobenzoyl-pyridine and the like.
Instead of a single ketone, a mixture of 2 different ketones can be used. So z. B. a mixture of a lower alkylphenyl ketone and a lower alkyl pyridyl ketone in the process according to the reduction a mixture of diols from which the corresponding asymmetric diol, namely a phenyl pyridyl ketone can be isolated.
The process of this invention is primarily advantageous for the preparation of 2,3-bis- (3-pyridyl) 2,3-butanediol by electrolytic reduction of the 3-acetylpyridine.
The diols produced according to the process of the present invention are useful starting materials for the production of ketones by means of the pinacolone rearrangement or for the production of indenes by means of double dehydration and ring closure.
Some of the diols, ketones or indenes obtained in this way show extraordinary inhibitory effects on the adrenal function and can therefore be used as remedies or for diagnostic purposes. 3,3-Bis- (4-aminophenyl) -butan-2-one (Amphenon B), 1,2-bis- (3-pyridyl) -2-methyl-propane-1- on (methopyrapon), which by inacolon rearrangement from 2,3-bis- (4-aminophenyl) -2, 3-butanediol or
2,3-Bis (3-pyridyl) -2, 3-butanediol can be obtained, and also 6-chloro-2- (3-pyridyl) -indene, which is obtained by double dehydration and simultaneous ring closure in the presence of a strong acid, how hydrochloric acid can be obtained from 2- (4-chloro-phenyl) -3- (3-pyridyl) -2, 3-butanediol.
In the process of the present invention, those starting materials are advantageously used which lead to the end products highlighted above.

example 1
A solution of 24, 23 g of 3-acetylpyridine and 30 g of potassium acetate in 100 cm3 of N, N-dimethylformamide and 45 cm3 of water is placed as an electrolyte in a single-chamber electrolysis cell. A tin electrode with a surface of 100 cm (previously cleaned with nitric acid) is used as the cathode, and a Niokel electrode with a surface of 12 cm2 is placed in the electrolyte as the anode. At a temperature of 67-69, a current of 4, 1-4, 4 amps is now passed through the solution. During the electrolysis, 18 ml of water are added in portions in order to keep the liquid level constant.
After 80 minutes a precipitate forms and the electrolysis is interrupted as soon as 11.9 amp hours have elapsed.
The electrolyte is then diluted with 100 ml of water, cooled to 8 for 64 hours and filtered. The solid material is washed four times with 30 ml of water each time and dried at 100 under reduced pressure for 16 hours. This gives 12.65 g of 2,3-bis (3-pyridyl) -2, 3-butanediol, mp 218-225.
Example 2
The procedure is as in Example 1, except that a nickel cathode is used as the cathode and a nickel anode with a surface area of 36 cm 2 is used.
After the reaction mixture obtained in this way has been cooled to -8 for 16 hours, it is filtered and the precipitate is washed four times with 30 ml of water each time and dried at 70 in a vacuum for 16 hours. This gives 13.9 g of 2,3-bis (3-pyridyl) -2, 3-butane-diol with a melting point of 224-230.
Other lower alkanoylpyridines, such as. B. 3-Propionv ylpyridin, 4-Acetylpyridin or aliphatisdh-aromatic ketones, especially lower alkyl-phenyl-ketones such as 4-chlorobacetophenone or 4-amino-acetophenone or mixtures of the mentioned ketones can be used in the same way, whereby the corresponding diols are obtained will.
Example 3
A solution of 36.3 g of 3-acetylpyridine and 30 g of potassium acetate in 62.5 ml of deionized water and 82.5 ml of isopropanol is placed in a single-chamber electrolyte cell with a cylindrical nickel electrode (100 cm2 surface) and a smaller one cylindrical nickel anode with a surface area of 36 cm2, the smaller anode being located inside the cathode. The reduction is carried out at 62 and a current density of 0.03 amps / cm2 of the cathode. The reduction is terminated after 6 hours.
The reaction mixture is mixed with 100 ml of water and left to stand at room temperature for 16 hours.
It is then filtered off and the filter residue is washed with water. This gives 29.4 g of pure 2,3-bis (3-pyridyl) -2, 3-butanediol.
Example 4
A solution of 5030 g of 3-acetylpyridine and 6290 g of potassium acetate in a mixture of 15,200 ml of isopropanol and 15,200 ml of deionized water is made using a rectangular nickel electrode (with a surface area of 7000 cm2) and a nickel anode with a surface area of 2000 cm2, the electrolytic Subject to reduction. The reaction is carried out at 62 and 0.04 amp / cm2 of the cathode and terminated after 9 hours. The reaction mixture is mixed with 20,000 ml of water and worked up according to the method described in Example 3. This gives 3240 g of pure 2,3-13is43-pyridyl) -2, 3-butanediol.
Example 5
A mixture of 66.6 g of 3-acetylpyridine and 82.5 g of potassium acetate in 200 ml of deionized water and 200 ml of isopropanol is placed in a rectangular cell; Identical flat nick electrodes, each with a surface area of 100 em, are used. They are connected to the power supply by a commutator so that the role of the cathode and the anode can be changed without interrupting the reaction. The electrolysis is carried out at 62 and 0.04 amp / cm2 of the electrode for 8 hours and 20 minutes, reversing the direction of the current flow every 30 minutes.
The 2,3-bis (3-pyridyl) -2, 3-butanediol is isolated by adding 275 ml of water to the resulting solution and worked up according to the method described in Example 3. 43.5 g of pure product are obtained in this way.
