dans laquelle RI est un radical alcoylène dont les liaisons de valence sont séparées par 2 à 3 atomes de carbone.
Ces composés sont pharmaceutiquement utiles en tant qu'agents endocriniens causant un déplacement favorable de la réponse androgénique anabolique comme on le mesure par le rapport entre la croissance du muscle releveur de l'anus et la croissance de la prostate. Lesdits composés sont également des com posés de valeur comme intermédiaires chimiques dans la synthèse d'autres agents hormonaux.
Le procédé selon l'invention est caractérisé en ce qu'on chauffe une 17-alcoylène-dioxy-3alpha, 5alpha- cycloandrostan-6-one avec de l'hydroxylamine en excès pour former le 6-oximino-17-alcoylènedioxy- 3alpha, 5alpha-cycloandrostane correspondant, que l'on sépare et réduit en solution alcoolique en présence d'un métal alcalin en cétal de la 6-amino-3alpha,
5alpha-cycloandrostan-17-one. Ce procédé fournit principalement les 6 alpha- amino 17-éthylènedioxy (ou propylènedioxy) -3alpha, 5alpha-cycloandrostanes, mais il se forme également les dérivés 6bêta-amino correspondants en plus faible quantité. Ces derniers peuvent être séparés des dérivés 6alpha-amino de manière connue, par exemple par chromatographie.
Les cétals de la 6-amino-3alpha-5alpha cyclo- androstan-17-one peuvent être acylés pour obtenir les 6-acylamino-cétals correspondants et que l'on peut par la suite hydrolyser avec un acide aqueux pour obtenir les 6-acylamino-17-oxo- 3 alpha, 5alpha- cycloandrostanes. D'une manière différente, on peut hydrolyser directement les 6-amino-17-cétals avec un acide aqueux pour obtenir les sels du 6-amino- 17oxo-3alpha,
5alpha-cycloandrostane à partir duquel on peut obtenir l'amine libre par addition d'un agent alcalin.
<I>Exemple 1</I> <I>A)</I> 6-Oximino-17-éthylènedioxy-3alpha, 5alpha- cycloandrostane On chauffe sur un bain de vapeur pendant 3 heures une solution préparée à partir de 4,$- grammes de 17-éthylène-dioxy-3alpha, 5alpha-cycloandrostan-6- one, 5,0 grammes de chlorhydrate d'hydroxylamine et 200 cm3 de pyridine. Après refroidissement on. dilue le mélange avec 1,2 litre d'eau et on l'extrait avec de l'éther.
On lave l'extrait d'éther avec de l'eau et on le sèche sur du sulfate de magnésium. On évapore le solvant, on fait cristalliser le résidu au sein de l'éthanol aqueux, en obtenant des cristaux de 6-oximino-17- éthylène-dioxy-3alpha, 5alpha-cycloandrostane-. On sépare les cristaux par filtration, on- les lave avec de l'éthanol aqueux et on les soumet à la dessiccation en obtenant 4,18 grammes de ce nouvel intermédiaire, fondant à 182 -186 C.
Une portion de ce produit est recristallisée pour obtenir un échantillon analytique fondant à 186-187 C, dont les valeurs analytiques obtenues correspondent aux valeurs calculées pour le 6-oximino-17-éthylène dioxy-3alpha, 5alpha-cyclo- androstane avec la formule empirique C21H3,.03N. <I>B)</I> 6alpha-Amino-17-éthylènedioxy-3alpha,5alpha- cycloandrostane On chauffe au reflux une solution de 4,1 grammes de 6-oximino-17-éthylènedioxy-3alpha,
5alpha-cyclo- androstane dans 400 ce d'éthanol anhydre tandis que l'on ajoute par portions 36 grammes de sodium métallique pendant 90 minutes. Après 60 minutes supplémentaires de chauffage au reflux, on dilue le mélange réactionnel chaud avec 200 ce d'éthanol anhydre et on le filtre à travers un entonnoir de verre fritté. On refroidit le filtrat en y ajoutant de la glace et 2 litres d'eau froide et on l'extrait par la suite avec de l'éther. On lave l'extrait d'éther avec de l'eau et on le sèche sur du sulfate de magnésium. L'éther est évaporé laissant 4,33 grammes d'une huile visqueuse jaune pâle.
On dissout cette huile dans 80 ce d'éther auxquels on ajoute 2,5 ce d'acide acétique glacial, et on agite la solution résultante jusqu'à ce qu'elle devienne homogène. On dissout alors la solution à 200 cm3 avec de l'eau et on l'extrait 2 fois avec des portions de 200 cm3 d'éther. On lave les solutions d'éther en série avec 2 portions de 150 cm3 d'eau que l'on ajoute à l'extrait aqueux initial. On traite avec 200 cm3 d'ine solution de soude à 5 %, la solution aqueuse résultante contenant le sel d'acide acétique de l'amine et on l'extrait avec de l'éther.
On lave la solution d'éther avec de l'eau et on la sèche sur du sulfate de magnésium. L'éther est évaporé laissant 2,83 grammes de 6a-amino-17-éthylènedioxy-3cc,5oc, cycloandrostane identifié par le spectre infrarouge et caractérisé par sa conversion en d'autres dérivés.
C) (facultatif) 6oc-Acétamide-17-éthylènedioxy-3cc-5a- cycloandrostane On conserve à la température ambiante pendant la nuit une solution préparée à partir de 1,6 gramme de 6x-amino-17-éthylènedioxy-3oc,5cc-cycioandrostane de la partie B, 36 ce de pyridine et 12 ce d'anhydride acétique. On verse ensuite la solution résultante dans 400 ce d'eau et on recueille par filtration sur un entonnoir de verre fritté le solide blanc qui se sépare. Cette substance est absorbée dans 200 ce de chloro forme. On lave la solution de chloroforme avec 2 portions de 100 ce d'eau et on la sèche sur du sulfate de magnésium.
On évapore le chloroforme, on fait cristalliser le résidu et on le recristallise au sein d'une solution d'éther-pentane en formant le & x- acétamido-17-éthylènedioxy-3ac,5a,-cycloandrostane qui se sépare du milieu de cristallisation sous forme d'un gel blanc qui se transforme par filtration en un solide blanc finement divisé. Après séchage, le produit fond à 193 -197 C. On identifie le produit par son spectre d'absorption infrarouge et son analyse élémentaire qui est en accord avec la formule empirique C23H3503N.
D) (facultatif) 6a-amino-3oc, 5ac-cycloandtostan-17-one On agite le sel d'acide acétique de l'amine obtenue en B (505 mg) avec un mélange de 200 ce d'éther et 100 ce d'une solution de carbonate de sodium à 5 %. On sépare la phase aqueuse et on l'extrait avec 200 ce d'éther. On lave les solutions d'éther en série avec 3 portions de 100 ce d'eau, on les combine et on les sèche sur du sulfate de magnésium anhydre. On évapore l'éther et on fait cristalliser le résidu au sein du pentane pour produire 390 mg de la 6ac- amino-3oc,5oc-cycloandrostan-17 one pf: 67 -70 C.
La recristallisation au sein du pentane donne un échantillon analytique fondant à 70 -72 C. L'analyse est en accord avec la formule empirique C19H290N. <I>Exemple 2</I> 6oc-Amino-17 propylènedioxy-3a,5oc-cycloandtostane.
A) On prépare le 3(3-hydroxy-17- (1,3-propylène- dioxy)-androst-5-ène en traitant la 3P-hydroxyandrost- 5-ène-17-one avec du 1,3-propylèneglycol en présence d'une quantité catalytique d'acide p-toluène sul- fonique selon le mode opératoire général pour pré parer les propylènedioxy-stéroïdes qui est décrit dans le brevet américain n 2302636 du 17.11.42.
Le produit est converti en 3(3-p-toluène-sulfonyloxy-17- (1,3-propylènedioxy)-androst-5-ène en le traitant avec le chlorure de p-toluènesulfonyle dans la pyridine. On convertit par hydrolyse le p-toluène sulfonate en composé 6p-hydroxyque que l'on oxyde en 17- (1,3-propylènedioxy)-3oc,5a-cycloandrostan-6-one selon le mode opératoire décrit dans Bull. Soc. Chim. Fr.1960 pages 298-299.
Les valeurs analytiques de cette cétone sont en accord avec la formule empirique C22H3203- B) On convertit la 17-(1,3-propylènedioxy)-3oc, 5ac-cyclo-androstan-6-one par le mode opératoire de la partie A de l'Exemple 1 en 6-oximino-17-(1,3- propylènedrioxy)-3a,5oc-cylonandrostane. Les résultats analytiques sont en accord avec la formule empirique C22H3303N.
C) On réduit par addition par portion de 36 grammes de sodium, une solution de 4 grammes de 6-oximino- 17-propylènedioxy-3oc,5a.-cycloandrostane de la partie B dans 400 ce d'éthanol anhydre. On traite le mélange réactionnel selon le mode opératoire décrit dans la partie B de l'exemple 1 pour obtenir le 6cc- amino-17-propylènedioxy-3ac,5a.-cycloandrostane dont les valeurs analytiques correspondent à celles de la formule empirique C22H3502N.
On convertit cette amine en composés suivants: 6cc-Acétamido-17-propylènedioxy-3cc, 5cc-cycloandro- stane, 6x-p-Nitrobenzamido-17-propylènedioxy-3x,5x- cycloandrostane, 6x-Propionamido-17-propylènedioxy- 3x,5x-cycloandrostane, respectivement.
On peut utiliser les glycols suivants pour préparer les stéroïdes à structure cétal en position 17: L'éthylène glycol (fournissant des structures éthylènedioxy) le 1,2-propylène glycol (voir J.Med. Pharm.Chem. 5 133 (l962); le 1,3-propylène glycol le 2,2-diméthylpropane-1,3-diol (voir Brevet américain n 2960502 du 15.11.60).

Parmi les groupes acyle convenables comme substituants, on peut citer les groupes: acétyle, pr opionyle, butyryle, hexanoyle, benzole, p-nitrobenzoyle, p-méthoxybenzoyle, 3,4- diméthoxybenzoyle, 3,4,5-triméthoxy-benzoyle, cyclo- pentanoyle, p-acéthylaminobenzoyle, différentes halo- génobenzoyles,
p-toluoyle cyclohéxanoyle, phényl- acétyle, cinnamoyle, isobutyryle, etc. Dans la plupart de ces cas, les cétals à groupe éthylènedioxy, 1,3 -propylènedioxy, ou 1,2 -propylènedioxy, sont des intermédiaires dans la préparation des composés à atome d'oxygène en 17.
Cependant il est évident que le groupe amino peut être substitué avant ou après que le groupe alkylène dioxy en position 17 soit hydrolysé en groupe oxo.

Said compounds are also valuable compounds as chemical intermediates in the synthesis of other hormonal agents.
The process according to the invention is characterized in that a 17-alkylene-dioxy-3alpha, 5alpha-cycloandrostan-6-one is heated with excess hydroxylamine to form 6-oximino-17-alcoylenedioxy-3alpha, Corresponding 5alpha-cycloandrostane, which is separated and reduced in alcoholic solution in the presence of an alkali metal to the ketal of 6-amino-3alpha,
5alpha-cycloandrostan-17-one. The ether extract is washed with water and dried over magnesium sulfate. The solvent is evaporated off, the residue is crystallized from aqueous ethanol, obtaining crystals of 6-oximino-17-ethylene-dioxy-3alpha, 5alpha-cycloandrostane-. The crystals were filtered off, washed with aqueous ethanol and subjected to drying, obtaining 4.18 grams of this new intermediate, melting at 182-186 ° C.

5alpha-cyclo-androstane in 400 cc of anhydrous ethanol while 36 grams of metallic sodium are added in portions over 90 minutes. After a further 60 minutes of heating under reflux, the hot reaction mixture is diluted with 200 cc of anhydrous ethanol and filtered through a sintered glass funnel. The filtrate was cooled by adding ice and 2 liters of cold water to it and subsequently extracted with ether. The ether extract is washed with water and dried over magnesium sulfate. The ether is evaporated leaving 4.33 grams of a viscous pale yellow oil.
This oil is dissolved in 80 cc of ether to which is added 2.5 cc of glacial acetic acid, and the resulting solution is stirred until it becomes homogeneous. The 200 cm3 solution is then dissolved with water and extracted twice with 200 cm3 portions of ether. The ether solutions are washed in series with 2 portions of 150 cm3 of water which is added to the initial aqueous extract. Treated with 200 cm3 of 5% sodium hydroxide solution, the resulting aqueous solution containing the acetic acid salt of the amine and extracted with ether.
The ether solution is washed with water and dried over magnesium sulfate. The ether is evaporated leaving 2.83 grams of 6a-amino-17-ethylenedioxy-3cc, 5oc, cycloandrostane identified by the infrared spectrum and characterized by its conversion into other derivatives.
C) (optional) 6oc-Acetamide-17-ethylenedioxy-3cc-5a-cycloandrostane A solution prepared from 1.6 grams of 6x-amino-17-ethylenedioxy-3oc, 5cc- is stored at room temperature overnight. part B cycioandrostane, 36 cc of pyridine and 12 cc of acetic anhydride. The resulting solution is then poured into 400 cc of water and the white solid which separates is collected by filtration through a sintered glass funnel. This substance is absorbed in 200 cc of chloro form. The chloroform solution was washed with 2 100 cc portions of water and dried over magnesium sulfate.

D) (optional) 6a-amino-3oc, 5ac-cycloandtostan-17-one The acetic acid salt of the amine obtained in B (505 mg) is stirred with a mixture of 200 cc of ether and 100 cc of a 5% sodium carbonate solution. The aqueous phase is separated and extracted with 200 cc of ether. The ether solutions were washed in series with 3 100 cc portions of water, combined and dried over anhydrous magnesium sulfate. The ether is evaporated off and the residue crystallized from pentane to give 390 mg of 6ac-amino-3oc, 5oc-cycloandrostan-17 one mp: 67 -70 C.
Recrystallization from pentane gives an analytical sample melting at 70 -72 ° C. The analysis is in agreement with the empirical formula C19H290N. <I> Example 2 </I> 6oc-Amino-17 propylenedioxy-3a, 5oc-cycloandtostane.
A) 3 (3-hydroxy-17- (1,3-propylene-dioxy) -androst-5-ene is prepared by treating 3P-hydroxyandrost-5-ene-17-one with 1,3-propylene glycol in presence of a catalytic amount of p-toluene sulfonic acid according to the general procedure for preparing propylenedioxy-steroids which is described in US Pat. No. 2302636 of 17.11.42.
The product is converted to 3 (3-p-toluene-sulfonyloxy-17- (1,3-propylenedioxy) -androst-5-ene by treating it with p-toluenesulfonyl chloride in pyridine. The p is converted by hydrolysis. -toluene sulfonate in 6p-hydroxy compound which is oxidized to 17- (1,3-propylenedioxy) -3oc, 5a-cycloandrostan-6-one according to the procedure described in Bull. Soc. Chim. Fr. 1960 pages 298 -299.
The analytical values of this ketone are in agreement with the empirical formula C22H3203- B) 17- (1,3-propylenedioxy) -3oc, 5ac-cyclo-androstan-6-one is converted by the procedure of part A of Example 1 in 6-oximino-17- (1,3-propylenedrioxy) -3a, 5oc-cylonandrostane. The analytical results are in agreement with the empirical formula C22H3303N.
C) A solution of 4 grams of 6-oximino-17-propylenedioxy-3oc, 5a.-cycloandrostane of part B in 400 cc of anhydrous ethanol is reduced by addition per portion of 36 grams of sodium. The reaction mixture is treated according to the procedure described in part B of Example 1 to obtain 6cc-amino-17-propylenedioxy-3ac, 5a.-cycloandrostane, the analytical values of which correspond to those of the empirical formula C22H3502N.
This amine is converted into the following compounds: 6cc-Acetamido-17-propylenedioxy-3cc, 5cc-cycloandrostane, 6x-p-Nitrobenzamido-17-propylenedioxy-3x, 5x-cycloandrostane, 6x-Propionamido-17-propylenedioxy-3x, 5x-cycloandrostane, respectively.
The following glycols can be used to prepare the steroids having a ketal structure at position 17: Ethylene glycol (providing ethylenedioxy structures) 1,2-propylene glycol (see J. Med. Pharm. Chem. 5,133 (1962); 1,3-propylene glycol 2,2-dimethylpropane-1,3-diol (see US Patent No. 2960502 of 11.15.60).
2,3-butanediol (see Can. J. Technology 29,243 (1951). From the free 6x-amino or 6pamino steroids, carrying in position 17 the alkylene dioxy structure, the amino group can be substituted by a wide variety of acyl groups.
Among the acyl groups suitable as substituents, there may be mentioned the following groups: acetyl, pr opionyl, butyryl, hexanoyl, benzole, p-nitrobenzoyl, p-methoxybenzoyl, 3,4-dimethoxybenzoyl, 3,4,5-trimethoxy-benzoyl, cyclo - pentanoyl, p-acetaminobenzoyl, various halogenobenzoyl,
p-toluoyl cyclohexanoyl, phenylacetyl, cinnamoyl, isobutyryl, etc. In most of these cases, the ethylenedioxy, 1,3 -propylenedioxy, or 1,2 -propylenedioxy ketals are intermediates in the preparation of the 17-oxygen atom compounds.
However, it is evident that the amino group can be substituted before or after the alkylene dioxy group at position 17 is hydrolyzed to an oxo group.
