Verfahren zur Zerlegung der DL-a-Azido-phenylessigsäure in die optisch aktiven Formen
Bei der Herstellung der a-Azido-phenylessigsäure, z. B. nach der Methode von Darapsky, J. prakt. Chem., 2, 99, 221 (1919), erhältt !man gewöhnlich die inak- tive DL-Form. Diese Säure ist wertvoll zur HerstelL lung von a-Amino-benzylpenicillin, das vorzügliche antibiotische Aktivität gegenüber gram-positiven und gram-negativen Bakterien hat.
Als therapeutisches Agenz ist das D (-)-Epimere des a-Amino-benzyl- penicillins von besonderem Interesse, da es besser wasserlöslich bei seinem isoelektrischen Punkt und stärker aktiv gegenüber verschiedenen Bakterienarten in vitro ist, als dies bei der L (+)-Form oder bei der DL-Form der Fall ist. Es ist daher erwiinscht, ein Verfahren zur Herstellung der D @(-)-Form der Säure zu haben.
Es ergeben sich aber bei verschiedenen Metho den zur Herstellung der D (-)-Form Schwierigkeiten.
So erfolgt z. B. bei der Reaktion von optisch aktiver a-Brom-phenylessigsläure mit Natriumazid partielle Racemisierung.
Nach dem Verfahren der vorliegenden Erfindung las, sen sich nun die D (-) und die L (+)-Form der a-Azido-phenylessigsäure sowie die entsprechen- den Salze herstellen.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass die DL-a-Azido-phenylessigsäure in einem geeigneten Lösungsmittel mit einer organs schen Base aus der Gruppe 1-Ephedrin, Dehydroabietylamin, a-(2-Naphtyl) täthylamin, Cinchonidin, Cinchonin, Chinidin, Chinin, Brucin, Strychnin oder Morphin behandelt wird, das. entstandene D (-)-Salz durch fraktionierte Kristallisation abgetrennt und daraus durch Behandlung mit Säure die freie D (-) a-Azido-phenylessigsäure gewonnen wird.
Anschlie- ssend kann die nach der Kristallisation in der Mutterlauge verbliebene L (+)-a-Azido-phenylessigsäure aus der Lauge abgetrennt oder durch Behandlung mit einer anorganischen Base racemisiert und das Racemat erneut nach der vorbeschriebenen Weise in die optisch aktiven Formen zerlegt werden.
Zur Zerlegung der inaktiven Säure in die optisch aktiven Formen verwendet man mit Vorteil als Base das 1-Ephedrin, da die Ausbeuten an D (-)-Säure damit höher ausfallen als mit den übrigen genannten organischen Basen.
Als besonders geeignete Lösungsmittel haben sich niedere Alkanole erwiesen, insbesondere Methanol oder Athanol.
Zur Gewinnung der D (-)-Säure aus dem D (-) Salz eignet sich z. B. die Behandlung des Salzes mit kalter, verdünnter Saltzsäure.
Zur Racemisierung der nach der Kristallisation in der Mutterlauge verbliebenen L (+)-a-Azido-phenyl- essigsäure kann z. B. so verfahren werden, dass man die Lösung 2 Stunden lang mit verdünn'ber Natronlauge behandelt, wonach die erhaltene racemische Säure wieder gelöst werden kann.
Zur Herstellung von D (-)-a-Amino-benzylpenicil- lin kann die D (-)-a-Azido-phenylessigsäure mit Vorteil in das Chlorid übergeführt werden, z. B. durch Behandlung mit Thionylchlorid in Trichloräthylen, oder in ein funktionelles Aquivalent d'es Chlorides, welches hierauf mit 6-Amino-penicillansäure zum D (-)-a-Azido-benzylpenicillin umgestzt wird. Das letztere wird reduziert, z. B. durch katalytische Hydrierung mit Raney-Nickel.
Beispiel 1
Trennung der optisch aktiven Formen
Eine warme Lösung von 82, 5 g = 0, 5 Mol Hiphedrin in 200 ml Äthanol wurde zu einer warmen Lösung von 88, 6 g = 0, 5 Mol racemischer a-Azido-phenylessigsäure hinzugefügt. Das Gemenge wurde zunächst bei Zimmertemperatur und hiernach eine Stunde lang in einem Kühlschrank kristallisie- ren gelasse. Das Salz wurde abfiltriert, getrocknet, gewogen und aus Athanol umkristallisiert.

Die letzte Salzfraktion wurde mit kalter, ver dünnter Salzsäure behandelt und die Azidosäure hierauf rasch mit Ather extrahiert. Die ätherische Lösung wurde gewaschen, getrocknet und dann eingedampft.
Der Rückstand wurde in einer kleinen Menge Benzol bei ungefähr 40 C gelöst, Petroläther zur Lösung hinzugegeben und die Säure zum Kristallisieren gebracht. Ausbeute 15 g, Smp. 55-57 C, [a] 2D4 =-144 (c = 1, 0 in Äthanol). Der Smp. war unscharf, da die Kristalle hartnäckig etwas Lösungsu mittel zurückhielten.
Nach der Behandlung mit 1-Ephedrin wurde aus der Mutterlauge eine Säure mit einer Drehung von -f 40 erhalten. Es wurde daraus mit (-)-α-(2-Naph- thyl)-äthylamin ein Salz hergestellt, welches viermal aus Äthanol umkristallisiert wurde. Aus diesem Salz wurde die Säure freigesetzt, die wie oben angegeben umkristallisiert wurde, wobei die L (+) Form der Säure erhalten wurde. Smp. 56-57 C, [α] 2D4 +146 (c = 1, 0 in Athanol).
Racemisierung
Eine Lösung von 0, 50 g L (+)-a-Azido-phenyl- essigsäure, die in der vorstehend beschriebenen Weise erhalten wurde, wurde in 2n Natronlauge 2 Stunden lang bei Zimmertemperatur belassen und hernach mit 4, 0 ml 2n Salzsäure angesäuert. Die Mischung wurde mit einem kleinen Kristall racemischer a-Azido-phenylessigsäure geimpft, abgekühlt und die ausgefallene Säure abfiltriert.
Erhalten wurden 0, 41 g Säure vom Smp. 101 bis 103 C, die bei Zumischung eines authentischen Musters von racemischer a-Azido-phenylessigsäure keine Depression beim Smp. zeigte.
Bedspiel 2
Eine Lösung von 82, 6 g = 0, 50 Mol 1 Ephedrin in 170 ml Methanol wurde zu einer Lösung von 91, 0 g = 0, 51 Mol a-Azido-phenylessigsäure in 170 ml Methanol hinzugefügt. Die vereinigten Lösungen wurden auf 50 C erwärmt, wobei sie vollkommen klar wurden. Nun wurde die Temperatur der Lösung unter Rühren langsam erniedrigt. Bei 40 C wurde ein Impfkristall von D (-)-a-Azido- phenylessigsaurem l-Ephedrinsalz hinzugefügt. Nach dem Verlauf von 8 Stunden wurde die Temperatur der Lösung langsam auf 15 C erniedrigt.
Das auskristallisierte Salz wurde abfiltriert und mit 25 ml kaltem Methanol gewaschen. Ausbeute 55, 1 g = 65%, [α]D24 =-83, 3 (Athanol).
Die a ; Azido-phenylessigsäure wurde aus dem Salz gemäss der im Beispiel 1 beschriebenen Weise in Freiheit gesetzt. Ihre spezifische Drehung wurde zu-145 (Äthanol) gefunden.
Process for the decomposition of DL-a-azido-phenylacetic acid into the optically active forms

So z. B. in the reaction of optically active a-bromophenylacetic acid with sodium azide partial racemization.
According to the process of the present invention, the D (-) and L (+) - forms of α-azido-phenylacetic acid and the corresponding salts can now be produced.
The inventive method is characterized in that the DL-a-azido-phenylacetic acid in a suitable solvent with an organic base from the group 1-ephedrine, dehydroabietylamine, a- (2-naphthyl) ethylamine, cinchonidine, cinchonine, quinidine, quinine , Brucine, strychnine or morphine is treated, the resulting D (-) - salt is separated off by fractional crystallization and the free D (-) a-azido-phenylacetic acid is obtained therefrom by treatment with acid.
The L (+) - α-azido-phenylacetic acid remaining in the mother liquor after crystallization can then be separated off from the liquor or racemized by treatment with an inorganic base and the racemate can be broken down again into the optically active forms in the manner described above.
To break down the inactive acid into the optically active forms, it is advantageous to use 1-ephedrine as the base since the yields of D (-) acid are thus higher than with the other organic bases mentioned.
Lower alkanols, in particular methanol or ethanol, have proven to be particularly suitable solvents.
To obtain the D (-) - acid from the D (-) salt z. B. the treatment of the salt with cold, dilute hydrochloric acid.
To racemize the L (+) - a-azido-phenylacetic acid remaining in the mother liquor after crystallization, z. B. proceed so that the solution is treated with diluent sodium hydroxide solution for 2 hours, after which the racemic acid obtained can be redissolved.
For the preparation of D (-) - a-Amino-benzylpenicillin, the D (-) - a-Azido-phenylacetic acid can advantageously be converted into the chloride, e.g. B. by treatment with thionyl chloride in trichlorethylene, or in a functional equivalent d'es chloride, which is then converted with 6-amino-penicillanic acid to D (-) - a-azido-benzylpenicillin. The latter is reduced, e.g. B. by catalytic hydrogenation with Raney nickel.

Separation of the optically active forms
A warm solution of 82.5 g = 0.5 mol of hiphedrin in 200 ml of ethanol was added to a warm solution of 88.6 g = 0.5 mol of racemic α-azido-phenylacetic acid. The mixture was left to crystallize first at room temperature and then for one hour in a refrigerator. The salt was filtered off, dried, weighed and recrystallized from ethanol.

The last salt fraction was treated with cold, dilute hydrochloric acid and the acidic acid was then rapidly extracted with ether. The ethereal solution was washed, dried and then evaporated.
The residue was dissolved in a small amount of benzene at about 40 ° C., petroleum ether was added to the solution and the acid was caused to crystallize. Yield 15 g, m.p. 55-57 C, [a] 2D4 = -144 (c = 1.0 in ethanol). The m.p. was fuzzy because the crystals stubbornly held back some solvent.
After treatment with 1-ephedrine, an acid with a rotation of -f 40 was obtained from the mother liquor. A salt was prepared therefrom with (-) - α- (2-naphthyl) ethylamine, which was recrystallized four times from ethanol. The acid was liberated from this salt, which was recrystallized as indicated above, the L (+) form of the acid being obtained. M.p. 56-57 C, [α] 2D4 +146 (c = 1.0 in ethanol).
Racemization
A solution of 0.50 g of L (+) - a-azido-phenylacetic acid, which was obtained in the manner described above, was left in 2N sodium hydroxide solution for 2 hours at room temperature and then acidified with 4.0 ml of 2N hydrochloric acid. The mixture was seeded with a small crystal of racemic α-azido-phenylacetic acid, cooled and the acid which had precipitated out was filtered off.
0.41 g of acid with a melting point of 101 to 103 ° C. were obtained which, when an authentic sample of racemic α-azido-phenylacetic acid was mixed in, showed no depression of the melting point.
Bedspiel 2
A solution of 82.6 g = 0.50 mol of 1 ephedrine in 170 ml of methanol was added to a solution of 91.0 g = 0.51 mol of a-azido-phenylacetic acid in 170 ml of methanol. The combined solutions were heated to 50 ° C., during which time they became completely clear. The temperature of the solution was then slowly lowered while stirring. At 40 C a seed crystal of D (-) - a-azidophenylacetic acid l-ephedrine salt was added. After the passage of 8 hours, the temperature of the solution was slowly lowered to 15 ° C.
The crystallized salt was filtered off and washed with 25 ml of cold methanol. Yield 55.1g = 65%, [α] D24 = -83.3 (ethanol).
The a; Azido-phenylacetic acid was released from the salt in the manner described in Example 1. Its specific rotation was found to be -145 (ethanol).
