Verfahren zur Herstellung von Piperazinderivaten des Butadiendioxyds
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung potentieller pharmazeuti- scher Wirkstoffe der Formel

worin n eine Zahl von 0-4, R'H oder CH3 und R H, CH3, -C2H5, -C6H5, -COCH3, -COOCH3, -COOC2H5, -CONH2, -CONHCH3, -CON(CH3) 2,-C (NH)NH2,
PO (OCH3) 2 oder PO (NH2) 2 bedeuten, das dadurch gekennzeichnet ist, dass man ! Butadiendioxyd in einem inerten Lösungsmittel mit unsubstituiertem oder entsprechend substituiertem Piperazin umsetzt.
Es sind bereits zahlreiche N, N'-substituierte Piperazinderivate hergestelllt und f r phairmazeutische Verwendungszwecke vorgeschlagen worden. So ist beispielsweise die antihistaminische und antispasmo- dische Eigenschaft des 1, 4-Dibenzyl-piperazins bekannt (L. Lannoy u. G. Chaboud, C. r. soc. biol. 142, 432 (1948). Ferner wurden Piperazin Abkommli'nge mit sedativer Wirkung beschrieben (DRP Nummern 897 562, 943 771, 943 943).
Wieder andere 1, 4-sub stituierte Piperazind'erivate zeigten eine ausgesprochene Wirkung auf das Zentralnervensystem und die peripheren Blutgefässe (DPA Nr. 1 041 501).
Die meisten dieser Produkte sihd in Wasser schwer löslich. Sie können zwar durch Uberführung in ihre Salze z. B. in die Acetate, Chlorhydrate und Bromhydrate wasserlöslich gemacht werden. Diese Salze sind aber nur bei pH-Werten < 7 beständig, was ihre Verwendung einschränkt. Ausserdem ist f r manche pharmazeutische Zwecke die Anwesenhei't von Ftemdionen unerwünscht.
Dass die Reaktion von 2 Molekülen Piperazin mit 1 Molek l Butadiendioxyd in Ausbeuten von 75-80 % durchführbar ist, war überraschend und in keiner Weise vorauszusehen, denn es ist bisher nur bekannt, dass Butadliendioxyd mit Diaminen unter Harzbildung reagiert (DRP Nr. 731 030, USP Nr. 2 136 928, FP Nr. 881 981). Lediglich mit primären Monoaminen vom Typ des Anilins sind niedermolekulare Additionsverbindungen an Butadendioxyd erhalten wordenr [S. Przybytek, Ber. deutsch. chem.
Ges. 17, 1091 (1884) ; AP Nrn. 2 098 782, 2 098 097] ; hierbei verlläuft aber die Reaktion, nicht eindeutig im Sinne der 1,4-Addition von 2 Molek len Amin, sondern es werden auch ringförmige Produkte aus 1 Molekül Butadiendioxyd und 1 Molekül Aniin erhal- ten (DBP Nrn. 805 522, 896047).
Die neuen Umsetzungsprodukite von unsubstituier- tem Piperazin bzw. von entsprechend substituierten Piperazinen mit Butadiendioxyd lassensich erfindungsgemϯnurinGegenwartvonL¯sungsmittelnherstellen. In Abwesenheit von Verd nnungsmitteln tritt schon bei Zimmertemperatur nach einiger Zeit Verharzung ein, die in manchen Fällen unter Hitzeentwicklung und explosionsartiger Zersetzung verläuft. Als Lösungsmittel eignen sich Wasser, Alkohol, Ather, Kohlen wasserstoffe, Halogenkohlenwasserstoffe und manche Ketone, wie MethylÏthylketon und Cyclohexanon.
Zweckmässig ist die Auswahl solcher Lösungsmittel, die zwar Butadiendioxyd ! und die Piperazinkompo- nente, nicht aber das Additionsprodukt lösen. In solchen Fällen gestaltet sich die Gewinnung des Reak tionsproduktes, das bei lÏngerem Stehen bei Zimmertemperatur im allgemeinen kristallin ausfällt, durch Abfiltrieren oder Zentrifugieren besonders einfach.
Die abgeschleuderte bzw. auf anderem Wege abgetrennte Mutterlauge, die im allgemeinen noch Anteile der im Überschuss verwendeten Piperazinkomponente enthÏlt, kann nach Aufkonzentrierung der verbrauch- ten Bestandteile direkt wieder verwendet werden. Die Anwendung eines Überschusses an unsubstituiertem Piperazin bzw. entsprechend substituiertem Piperazin empfiehlt sich, um Harzbildungen unidi andere unerwünschte Nebenreaktionen zu vermeiden. Man kann aber auch stöchiometrische Verhältnisse, das heisst 2 Mol Piperazinverbindung auf l Mol Butadiendioxyd anwenden, um die erfindungsgemϯen Piperazinaddukte herzustellen.
Die praktische Durchführung des erfindungsgemÏ ¯en Verfahrens erfolgt vorzugsweise bei Temperatu- ren unterhalb etwa 100 C, da oberhalb dieser Temperatur sich auch hochmolekulare Polyaddukte von öligem, manchmal auch gelf¯rmigem Charakter, von denen die niedermolekularen Piperazinaddukte der vorgenannten Formel, selbst wenn sie noch mit verringerter Ausbeute entstehen, nicht mehr ohne wei teres getrennt werden können.
Die untere zur praktischen Durchführung des er findungsgemässen Verfahrens noch in Frage kommende Temperaturgrenze ist durch die Reaktions- geschwindigkeit bestimmt und liegt vorzugsweise ber 0 C. Unterhalb 0¯C erfordert die Addition selbst von den reaktionsfÏhigsten Piperazinderivaten bereits 5 Tage und mehr bis zur Erreichung annehmbarer Umsätze. Eine sehr vorteilhafte Reaktionstemperatur ist Raumtemperatur (20-25 C), wobei mit Piperazin und verschiedenen substituierten Derivaten Reaktionst- zeiten, zwischen 1 und 4 Tagen bei Umsätzen von 70 bis 95% beobachtet wurden.
Butadiendioxyd besitzt zwei asymmetrische Koh l'enstoffatome und wird bei normalen Herstellungsver- fahren als Gemisch von meso-undd,l-Formerhalten.
Beispielsweise kann man zur erfindungsgemϯen Herstellung der Piperazinaddukte von einem solchen Gemisch ausgehen, man kann aber auch die reinen Isomeren einsetzen, die sich durch fraktionierte Destillation des Gemisches erhalten lassen. Man kanh) auch noch weiter gehen und'die d, 1-Form nach be kanntem Verfahren in de optisch aktiven Isomeren trennen ! und diese zur Herstellung von Piperazinaddukten benutzen. Alle diese Butadiendioxyde zeigen gegenüber Piperazin bzw. seinen entsprechend substituierten Derivaten die praktisch gleiche ReaktionsfÏhigkeit.
Zur Addition an eines der vorstehend charakteri- sierten Butadiendioxyde kommt z. B. das Piperazin, das Methyl-piperazin, 2, 5-Dimethyl-piperazin, 2, 6 Dimethyl-piperazin oder das Trimethyl-piperazin in Frage. Bei der Verwendung von am Stickstoff nicht substituierten Piperazinen empfiehlt es sich, um gute Ausbeuten an Butadiendioxydadd'ukt zu erzielen, einen tYberschuss von. 1-10 Mol, vorzugsweise 3-5 Mol an Piperazin anzuwenden.
Als potentielle pharmazeutische Wirkstoffe sind jedoch die Addukte aus Butadiendioxyd und ! 2 Mol eines an einem N-Atom entsprechend substituierten Piperazins von grösserer Bedeutung. In diesem Falle genügt es, die Reaktionskomponenten in äquivalen- ten Mengen, eventuelt mit einem geringen Überschuss an Piperazinderivat, einzusetzen. Zur Herstellung von Addukten dieser Art eignen sich z. B.

N-Butyl-piperazin,
N-Acetyl-piperazin,
N-Carboxymethyl-piperazin,
N-CarboxyÏthyl-piperazin,
Piperazin-N-carbonsÏureamid,
Piperazin-N-carbonsÏure-methylamid,
Piperazin-N-carbonsÏure-dimethylamid,
Piperazin-N-phosphorsÏuremethylester,
Piperazin-N-phosphorsÏure-amid,
Piperazin-N-phosphorsÏure-methyl-amid sowie die entsprechenden einseitig N-substituierten Derivate des Methyl-piperazins, der Dimethyl- piperazine und des Trimethyl-piperazins.
Beispiel 1 1, 4-Dipiperazinobutan-2, 3-diol
Zu einer Lösung von 100Gewichtsteilen wasserfreiem Piperazin (5facher Überschuss) in 220 Volum- teilten reinem Chloroform werden 100 Gewichtsteile Butadiendioxyd zugesetzt. Die klare Lösung wird 6 Tage bei Zimmertemperatur stehengelassen. Sodann wird das Lösungsmittel unter reduziertem Druck voll- ständig verdampft, unter Ausschluss von Feuchtigkeit und Luftsauerstoff. Der farblose, feste Rückstand wird zerrieben und durch Erhitzen auf 100 C bei 12 mm Hg vom überschüssigen Piperazin befreit, das dabe, wegsublimiert. Letzteres lässt sich ohne weiteres für einen neuen Ansatz verwenden.
Das restliche rohe Dipiperazinaddukt wird in kleinen Portionen im Hochvakuum destilliert (Kp. 190 /0, 01 mm Hg). Ein Uberhitzen des Kolbeninhaltes ist wegen der Hitze- empfindlichkeit des Produktes zu vermeiden. Man erhällt 22, 6 Gewichtsteile (76 % der Theorie) eines farb- losen Destillats, das in der Vorlage sofort erstarrt.
Zur weiteren Reinigung kann das destillierte oder sublimierte Dipiperazinaddukt aus der ungefÏhr 10fachen Menge Xylol umkristaHisiert werden. F. 146¯.
Beispiel 2
1, 4-Di-(2,5-dimethylpiperazino)-butan-2,3-diol
Eine konzentrierte Lösung von 114 Gewichtstei- len 2, 5-Dimethyl-piperazin wird mit 8, 6 Gewichts teilen Butadiendlioxyd versetzt. Als Losungsmittel kann Chloroform verwendet werden. Das Gemisch wird 6 Tage bei Zimmertemperatur stehengelassen.
Die nachfolgende Aufarbeitung des entstandenen Di (2, 5-dimethylpiperazino)-adduktes erfolgt analog Beispiel 1).
Beispiel 3 1, 4-Di-(N-acetylpiperazino)-butan-2,3-diol
86 Gewicbtsteile Butadiendioxyd und 256 Ge wichtsteile N-Monoacetylpiperazin [Herstellung : AP Nr. 2 436 685 (1948)] werden in 1000 Volumteilen abs. Ather gelöst. Beim Aufbewahren der Lösung bei Zimmertemperatur wird die Lösung allmählich tr b, und ein Íl fÏllt aus, das beim weiteren Stehentlassen) erstarrt. Nach Ablauf von 41/2 Tagen wird noch 5 Std. unter Rückfluss erwärmt, der farblose Niederschlag zerrieben, abfiltriert und im Vakuum getrock- net.
Er besteht aus 320 Gewichtsteilen (93 % der Theorie) rohem Di-(N-acetylpiperazino)-addukt vom Smp. 136 .
Die Reinigung erfolgt vorteilhaft durch Kristallisation aus Xylol. F. 140 .
Beispiel 4 1, 4-Di-(N-methylpiperazino)-butan-2,3-diol
51, 7 Gewichtsteile 1, 4-Dipiperazinobutan-2, 3diol werden unter Kühlung mit 54 Teilen 90 figer Ameisensäure vermischt. Zum entstandenen Brei fügt man unter Rühren 34 Gewichtsteile 39 % ige wässrige Formaldehydiösung'hinzu. Die entstandene Lösung wird nun dierart erwärmt, dass die einsetzende Koh lendoxydentwicklung nicht zu stürmisch verläuft Nach Abklingen der Reaktion wird noch 12 Std. auf 100 erhitzt, und sodann saugt man im Vakuum die Süchtigen Bestandteile ab.
Das zurückbleibende Öl wird mit 200 Teilen Wasser verdünnt, die Lösung durch eine SÏule von 800 Volumteilen Anionenaustauscher Amberlite IRA-410 gelassen und mit Wasser nacheluiert bis zur nahezu neutralen Reaktion des Eluates. Die gesamte Menge an Eluat wird im Vakuum zur Trockene eingedampft. Man erhält 57 Gewichtsteile veines ötigen Rückstandes, welcher auf Zugabe von etwas Aceton, das hernach wieder abgesaugt wird, vollständig erstarrt.
Zur Reinigung wird das Produkt in 300 Volumteilen Aceton gel¯st und durch langsames Abk hlen auf -5¯ kristallisiert. Man gewinnt derart 41 Ge wichtsteile Di - N - methylpiperazino) - Addukt vom Smp. 90-92¯. Durch Einengen und Abk hlen der Mutterlauge werden noch weitere 3 Gewichtateite dieses Produktes erhalten. Gesamtausbeute 85% der Theorie.
Ein noch reineres Produkt vom Smp. 98¯ wird erhalten durch Cessation des Rohproduktes (Kp.
¯ 150¯/0,01 mm Hg) und nachfolgende Kristalli- satilon.

Numerous N, N'-substituted piperazine derivatives have already been prepared and proposed for pharmaceutical uses. For example, the antihistaminic and antispasmodic property of 1,4-dibenzyl-piperazine is known (L. Lannoy and G. Chaboud, C. r. Soc. Biol. 142, 432 (1948). Furthermore, piperazine agreements were made described as having a sedative effect (DRP numbers 897 562, 943 771, 943 943).
Still other 1,4-substituted piperazine derivatives showed a marked effect on the central nervous system and the peripheral blood vessels (DPA No. 1 041 501).

These salts are only stable at pH values <7, which limits their use. In addition, the presence of foreign ions is undesirable for some pharmaceutical purposes.
The fact that the reaction of 2 molecules of piperazine with 1 molecule of butadiene dioxide can be carried out in yields of 75-80% was surprising and in no way foreseeable, because until now it is only known that butadiene dioxide reacts with diamines to form resin (DRP No. 731 030 , USP No. 2,136,928, FP No. 881,981). Only with primary monoamines of the aniline type have low molecular weight addition compounds to butadendioxide been obtained [S. Przybytek, Ber. German. chem.

The new reaction products of unsubstituted piperazine or of correspondingly substituted piperazines with butadiene dioxide can only be produced according to the invention in the presence of solvents. In the absence of thinners, resinification occurs after a while at room temperature, which in some cases develops with heat and explosive decomposition. Suitable solvents are water, alcohol, ethers, hydrocarbons, halogenated hydrocarbons and some ketones, such as methyl ethyl ketone and cyclohexanone.
It is advisable to choose solvents that contain butadiene dioxide! and the piperazine component, but not the addition product. In such cases, the recovery of the reaction product, which generally precipitates in crystalline form on prolonged standing at room temperature, is particularly easy by filtering off or centrifuging.
The mother liquor centrifuged off or separated off in some other way, which generally still contains proportions of the piperazine component used in excess, can be used again directly after the used constituents have been concentrated. The use of an excess of unsubstituted piperazine or correspondingly substituted piperazine is recommended in order to avoid resin formation and other undesirable side reactions. But one can also use stoichiometric ratios, that is to say 2 moles of piperazine compound to 1 mole of butadiene dioxide, in order to produce the piperazine adducts according to the invention.
The practical implementation of the process according to the invention is preferably carried out at temperatures below about 100 ° C., since above this temperature there are also high molecular weight polyadducts of an oily, sometimes also gel-like character, of which the low molecular weight piperazine adducts of the above formula, even if they are still arise with reduced yield, can no longer be separated easily.
The lower temperature limit still possible for the practical implementation of the process according to the invention is determined by the reaction rate and is preferably above 0 C. Below 0¯C, the addition of even the most reactive piperazine derivatives requires 5 days and more to achieve acceptable conversions . A very advantageous reaction temperature is room temperature (20-25 C), reaction times between 1 and 4 days with conversions of 70 to 95% having been observed with piperazine and various substituted derivatives.
Butadiene dioxide has two asymmetric carbon atoms and is obtained in normal manufacturing processes as a mixture of meso and d, l forms.
For example, one can start from such a mixture for the inventive preparation of the piperazine adducts, but one can also use the pure isomers which can be obtained by fractional distillation of the mixture. One can also go further and separate the d, 1-form into the optically active isomers by a known process! and use these for the production of piperazine adducts. All these butadiene dioxides show practically the same reactivity towards piperazine or its correspondingly substituted derivatives.
For addition to one of the butadiene dioxides characterized above, z. B. piperazine, methyl-piperazine, 2,5-dimethyl-piperazine, 2,6-dimethyl-piperazine or trimethyl-piperazine in question. When using piperazines which are not substituted on the nitrogen, it is advisable to use an excess of in order to achieve good yields of butadiene dioxide adduct. 1-10 moles, preferably 3-5 moles, of piperazine to be used.
However, the adducts of butadiene dioxide and! 2 moles of a piperazine correspondingly substituted on one N atom is of greater importance. Example 1 1, 4-Dipiperazinobutane-2, 3-diol
100 parts by weight of butadiene dioxide are added to a solution of 100 parts by weight of anhydrous piperazine (5-fold excess) in 220 parts by volume of pure chloroform. The clear solution is left to stand for 6 days at room temperature. The solvent is then completely evaporated under reduced pressure, with the exclusion of moisture and atmospheric oxygen. The colorless, solid residue is triturated and freed from excess piperazine by heating to 100 ° C. at 12 mm Hg, which sublimates away. The latter can easily be used for a new approach.
The remaining crude dipiperazine adduct is distilled in small portions in a high vacuum (bp 190/0.01 mm Hg). Overheating of the flask contents should be avoided because of the heat sensitivity of the product. 22.6 parts by weight (76% of theory) of a colorless distillate are obtained, which solidifies immediately in the receiver.
For further purification, the distilled or sublimed dipiperazine adduct can be recrystallized from approximately 10 times the amount of xylene. F. 146¯.
Example 2
1,4-Di (2,5-dimethylpiperazino) -butane-2,3-diol
A concentrated solution of 114 parts by weight of 2,5-dimethylpiperazine is mixed with 8.6 parts by weight of butadiene dioxide. Chloroform can be used as a solvent. The mixture is left to stand at room temperature for 6 days.
The subsequent work-up of the resulting di (2,5-dimethylpiperazino) adduct is carried out analogously to Example 1).
Example 3 1,4-Di- (N-acetylpiperazino) -butane-2,3-diol
86 parts by weight of butadiene dioxide and 256 parts by weight of N-monoacetylpiperazine [manufacture: AP No. 2 436 685 (1948)] are abs in 1000 parts by volume. Ather dissolved. When the solution is kept at room temperature, the solution gradually becomes cloudy, and an oil fills out, which solidifies when it is left to stand. After 41/2 days, the mixture is heated under reflux for a further 5 hours, the colorless precipitate is triturated, filtered off and dried in vacuo.

Example 4 1,4-Di- (N-methylpiperazino) -butane-2,3-diol
51.7 parts by weight of 1,4-dipiperazinobutane-2, 3 diol are mixed with 54 parts of 90% formic acid with cooling. 34 parts by weight of 39% strength aqueous formaldehyde solution are added to the resulting pulp with stirring. The resulting solution is now heated in such a way that the development of carbon oxide does not proceed too stormily. After the reaction has subsided, it is heated to 100 for another 12 hours, and then the addicts components are sucked off in vacuo.
The oil that remains is diluted with 200 parts of water, the solution is passed through a column of 800 parts by volume of Amberlite IRA-410 anion exchanger and eluted with water until the eluate reacts almost neutral. The entire amount of eluate is evaporated to dryness in vacuo. 57 parts by weight of veines ötigen residue are obtained, which solidifies completely on addition of a little acetone, which is then filtered off again with suction.
For cleaning, the product is dissolved in 300 parts by volume of acetone and crystallized by slow cooling to -5¯. 41 parts by weight of di - N - methylpiperazino) adduct of melting point 90-92¯ are obtained in this way. A further 3 parts by weight of this product are obtained by concentrating and cooling the mother liquor. Overall yield 85% of theory.
An even purer product with a melting point of 98¯ is obtained by cessation of the crude product (bp.
¯ 150¯ / 0.01 mm Hg) and the subsequent crystallization.
