Verfahren zur Herstellung von Verbindungen mit pilztötender Wirkung
Es ist bekannt, dass gewisse Verbindungen, welche den Rest SO2 S CC13 enthalten, eine pilztötende Wirkung haben. Beispiele solcher Stoffe sind unter anderem: Trichlormethylbenzolthiolsulfonat und Trichlormethyl-p-toluolthiolsulfonat der Formel:

Es hat sich unter anderem ergeben, dass vorstehend erwähnte Verbindungen 1 und II für den praktischen Gebrauch als pilztötende Stoffe den Nachteil aufwiesen, dass sie eine starke Phytotoxizität haben, was sich besonders in Beschädigung der Blätter oder in Keimhemmung äussert.
Es hat sich nun gezeigt, dass Stoffe mit pilztötender Wirkung erhalten werden, die nahezu keine Phytotoxizität aufweisen, wenn der Phenylrest eine Carboxylgruppe enthält oder ein funktionelles Derivat derselben, z. B. eine Ester- oder eine Salzgruppe. Dementsprechend bezieht sich die Erfindung auf ein Verfahren zur Herstellung von Verbindungen der Formel.

oder von Estern oder Salzen derselben, das dadurch gekennzeichnet ist, dass man Carboxybenzolsulfinsäure, ein Salz oder einen Ester davon mit Perchlor methylmerkaptan umsetzt.
Obgleich im Prinzip die Ortho-, Meta- und Paraisomere die gleiche Wirkung aufweisen, sind die Paraverbindungen für praktische Anwendung am meisten geeignet.
Als Ester kommen z. B. die Alkylester, z. B. Methyl- oder Äthylester, in Betracht. Sowohl organische als anorganische Salze weisen die erwähnte Wirkung auf: Es können z. B. die Alkalisalze oder die durch Reaktion der entsprechenden Säure mit organischen Basen erhaltenen Salze, z. B. Morpholin oder Tri athanolamin, verwendet werden.
Das erfindungsgemässe Verfahren wird vorzugsweise in wässerigem Medium durchgeführt. Die zu erhaltende Verbindung präzipitiert dabei und lässt sich leicht abscheiden.
Beispiel 1 p-Carboxy-benzolsulf insäure
110 g (0,5 Mol) p-Carboxy-benzolsulfochlorid wurde unter Rühren einer Lösung von 252 g (4 Mol) kristallisierten Natriumsulfids (Na2SO3 7aq) in 750 cm3 Wasser zugesetzt. Unter Rühren wurde darauf 50 0/o Natronlauge mit einer solchen Geschwindigkeit zugesetzt, dass das PH des Reaktionsgemisches schwach alkalisch blieb. Dazu brauchts man insgesamt 85 cm3 Lauge. Während der Reak- tion wurde die Temperatur auf 400 C gehalten.
Darauf wurde das Reaktionsgemisch noch während einer Stunde gerührt und darauf unter Abkühlung mit einem Gemisch von 185 g konzentrierter Schwefelsäure und 110 cm3 Wasser angesäuert. Der erhaltene Niederschlag wurde abgesaugt und einige Male mit Wasser gewaschen.
Trichlorrnethyl-p-carboxy-benzolthioirutjonat
Die p-Carboxy-benzolsulfinsäure, die gemäss dem vorangehenden Verfahren erhalten wurde, wurde in 500 cm3 Wasser suspendiert. Diesem Gemisch wurde unter Abkühlung und Rühren langsam 50prozentige Natronlauge zugesetzt, bis die Flüssigkeit gerade schwach alkalisch . reagierte. Nach Abfiltrieren nicht gelöster Verbindungen wurde die wässerige Lösung des Dinatriumsalzes von p-Carboxy-benzolsulfinsäure mit 2,5 Liter Wasser verdünnt, und dieser Lösung wurde eine Menge von 2n-Salzsäure zugesetzt, die der halben Menge der vorerwähnten 50prozentigen Natronlauge äquivalent war. Der auf diese Weise erhaltenen Lösung wurde unter kräftigem Rühren langsam 93 g Perchlormethylmerkaptan (0,5 Mol) zugesetzt. Dabei präzipitiert das Trichlormethyl-pcarboxy-benzolthiolsulfonat.
Das Reaktionsgemisch wurde noch während zwei Stunden gerührt, darauf filtriert und der Niederschlag mit Wasser gewaschen.
Die Ausbeute war 154 g (92 0/o), berechnet nach p-Carboxy-benzolsulfochlorid als Ausgangsmaterial.
Der Stoff schmilzt unter Zersetzung zwischen 190 und 192 C.
Beispiel 2 m-Carboxy-benzolsulfinsäure

30,5 g Benzoesäure (0,25 Mol) wurde 145,6 g Chlorsulfonsäure (1,25 Mol) zugesetzt, worauf das Reaktionsgemisch unter Rühren erhitzt wurde. Bei etwa 1000 C fing unter Wärmeentwicklung eine Reaktion an, bei der die Temperatur bis etwa 1350 C anstieg; darauf wurde während noch einer Stunde auf 125 C erhitzt, worauf das Reaktionsgemisch auf Eis ausgegossen wurde, wobei sich das m-Carboxybenzolsulfochlorid absetzte. Nach Abfiltrieren, Waschen und Trocknen des Niederschlags wurde er einmal aus Benzol umkristallisiert. Die Substanz schmilzt bei 130 bis 133 C. 22 g (0,1 Mol) dieses noch nicht ganz reinen m-Carboxy-benzolsulfochlorids wurde tüchtig mit 21 g Phosphorpentachlorid gemischt.
Nachdem das Reaktionsgemisch während etwa einer halben Stunde auf eine Temperatur von 110" C erhitzt worden war, wurde das hergestellte Phosphoroxychlorid in Vakuum abdestilliert, worauf 24 g braunen Öls zurückbleibt, das aus unreinem m-Carboxy-chloridbenzolsulfochlorid besteht. 23 g dieses Öls wurde unter Rühren und Erhitzen tropfenweise mit einer Lösung von 46,4 g Natriumsulfit (Na2SO3 7aq; 0,184 Mol) in 45 cm3 Wasser bei einer Temperatur von 50 bis 70 C gemischt. Um das Reaktionsgemisch stets schwach alkalisch zu halten, wurde gleichzeitig eine 50 0/o wässerige Natronlaugelösung hinzugetropft (in sge- samt 32 g Lösung).
Nachdem alle Reaktionskompo dienten zugesetzt worden waren, wurde noch während etwa einer halben Stunde gerührt; darauf wurde unter Abkühlung vorsichtig 25 g konzentrierte Schwefelsäure zugesetzt, wobei sich die m-Carboxybenzolsulfinsäure in fein verteiltem Zustand niederschlug; sie wurde dadurch isoliert, dass das Reaktionsgemisch einige Male mit äthyläther extrahiert wurde. Aus den ätherischen Extrakten ergab sich 15 g m - Carboxy - benzolsuifinsäure mit einem Schmelzpunkt von 175 bis 1780 C (Zersetzung) und mit einem äquivalenten Gewicht von 95,5 (berechnet 93,1). Die Säure wurde in das Dinatriumsalz umgewandelt. Die Ausbeute an m-Carboxy-benzolsuifochlorid betrug etwa 700/0.
Triclllornlethyl-n1v-carboxybenzolthiolsulfonat

27 g (0,117 Mol) m-Carboxy-benzolsulfinsäure (Dinatriumsalz) und 3 g Natriumbicarbonat wurden in 250 cm Wasser gelöst; darauf wurde der Lösung 24,3 g (0,131 Mol) Perchlormethylmerkaptan zugesetzt, und das Reaktionsgemisch wurde kräftig geschüttelt. Die erhaltene Lösung wurde zweimal mit Ether extrahiert und darauf mit 200 cm3 2n-Schwefelsäure gesäuert. Das Reaktionsgemisch wurde durch Extrahieren mit Diäthyläther abgetrennt. Aus der ätherischen Lösung ergab sich nach Trocknen und Eindämpfen in Vakuum 36,5 g Trichlormethylm-carboxy-benzolthiolsulfonat mit einem Schmelzpunkt von 167 bis 171" C.
(Ausbeute etwa 93 O/o.)
Beispiel 3 o-Carboxy-benzolsuljinsäure

10 g Thiosalicylsäure (0,065 Mol) wurde mit 40 cm wasserfreien Tetrachlorkohlenstoff gemischt, worauf unter Rühren Chlor eingeleitet wurde, bis alles Thiosalicylsäure unter Wärme- und Salzsäureentwicklung gelöst war. Durch Einleiten von Stickstoff wurde das Übermass an Chlor vertrieben; darauf wurde noch 50 cm3 Tetrachlorkohlenstoff dem Reaktionsgemisch zugesetzt, um auskristallisierten Stoff wieder zu lösen. Nach Filtrierung wurde die Reaktionslösung auf Eis ausgegossen, wobei sich die o-Carboxy-benzolsulfinsäure absetzte. Der Stoff wurde abfiltriert und in Vakuum getrocknet. Ausbeute 7,5 g (70 /o) ; äquivalentes Gewicht: 98,1 (berechnet 93,1).
Trichlormethyl-o-carboxy-benzolthiolslflfonat

Einer wässerigen Lösung von 15 g o-Carboxybenzolsulfinsäure (0,081 Mol) wurden in Reihenfolge 2 äquivalente wässerige Natronlauge und 1 äquivalente 2-N-Salzsäure zugesetzt. Der filtrierten Lösung (Gesamtvolumen 300 cm3) wurde 15,3 g (0,082 Mol) Perchlormethylmerkaptan zugesetzt und das Gemisch wurde während gewisser Zeit kräftig geschüttelt. Das niedergeschlagene Trichlormethylo-carboxy-benzolthiolsulfonat hat nach Filtrieren und Trocknen ein Gewicht von 24,5 g (Ausbeute 90 /a) und schmilzt bei 161 bis 168 C. Nach Umkristallisierung aus Benzol wurden schöne schuppenförmige Kristalle mit Schmelzpunkten zwischen 164 bis 1720 C erhalten.
Die Fungitoxizität der erhaltenen Verbindung wurde an Hand einer Sporenkeimprüfung bestimmt, wobei ein verdünnter Kirschenextrakt (1 Gewichtsteil trockenen Stoffes und 1000 Gewichtsteile Wasser) als Stimulans für den Keimprozess diente.
Die Prüfung wurde mit Sporen von zehn Pilzen durchgeführt. Die Resultate der zweifach ausgeführten Bestimmungen sind in nachstehender Tabelle zu sammengefasst:

Aus dieser Tabelle ist ersichtlich, dass die sogenannte Overall LD 50 0,93 1 0,1 Teile pro 1 Million beträgt.
Diese Pilze können in vier Gruppen untergebracht werden, wobei die maximale Empfindlichkeit der zu einer Gruppe gehörenden Pilze gleich ist. Von Gruppe I bis zu Gruppe IV sinkt die maximale Empfindlichkeit stets um den Faktor 10.
Zu der Gruppe I gehörende Pilze:
Chlomerella cingulata;
Mycosphaerella cannabis;
Fusarium culmorum.
Zu der Gruppe II gehörende Pilze:
Sclerotinia fructicola;
Cladosporium cucumerinum;
Penicillium italicum.
Zu der Gruppe 111 gehörende Pilze:
Botrytis allii;
Alternaria tenuis.
Zu der Gruppe IV gehörende Pilze:
Pythium debaryanum;
Rhizopus nigricans.
Zur Bestimmung der Phytotoxizität wurde eine 100/o Lösung des Stoffes in Aceton über eine Anzahl von Sorten höherer Pflanzen in einer Menge von 10 cms pro 1000 cm2 Grundoberfläche verspritzt.
Nachdem die Pflanzen während 5 Tagen bei einer TL-Belichtung von etwa 3000 Lux, einer relativen Feuchtigkeit von etwa 85 /o und einer Temperatur von 220 C aufbewahrt worden waren, wurde das Mass der Blattverbrennung bei Gartenbohnen, Kapuzinerkresse, Kartoffel, Kriechbohnen, Erbsen, Gurken, Tradescantia (zwei Sorten), Tabak, Begonia (zwei Sorten), Coläus und Capsicum festgestellt. Bei keiner der erwähnten Sorten konnte Blattbeschädigung festgestellt werden.
Die Verbindungen der erwähnten Formel können auf verschiedene Weise zu fungiziden Präparationen verarbeitet werden: a) mischbare Öle mit 15 bis 20 /o wirksamem Bestandteil, 150/0 nichtionogenem Emulsierungsmittel und weiter Lösungsmitteln (besonders Ketonen, z. B. Zyklohexanon); b) Aerosole mit als Lösungsmitteln, z. B. Aceton, Methyläthylketon und Zyklohexanon und als Treibgas Methylchlorid oder Freon; c) Spritzpulver mit 50 bis 808/0 aktivem Stoff und weiter Flussmitteln, z. B. Fettalkoholsulfonaten, sowie Dispergiermitteln und/oder Trägern, z. B.
Kaolin, Kreide, Tonerde, sowie Zusätze zum Verhüten von Klumpung, z. B. kolloidaler Kieselsäure; d) Staubpulver mit z. B. 5 0/o aktivem Stoff in einem Gemisch von Kieselgur und Dolomitmergel; e) Saatschützmittel, z. B. 500/0 des aktiven Stoffes in Vereinigung mit Kaolin unter Zusatz von Haftungsstoffen z. B. Spindelöl; f) Staubpulver, bei dem der wirksame Stoff in Form einer Lösung auf einen staubförmigen absorbierenden Träger, z. B. Kieselgur, angebracht und das Lösungsmittel verdunstet wird.

Example 2 m-Carboxy-benzenesulfinic acid

30.5 g of benzoic acid (0.25 mol) was added to 145.6 g of chlorosulfonic acid (1.25 mol) and the reaction mixture was heated with stirring. At about 1000 C a reaction began with the development of heat, during which the temperature rose to about 1350 C; the mixture was then heated to 125 ° C. for a further hour, after which the reaction mixture was poured onto ice, the m-carboxybenzenesulfonyl chloride settling out. Trichloromethyl-o-carboxy-benzenethiol sulfonate

To determine the phytotoxicity, a 100% solution of the substance in acetone was sprayed over a number of varieties of higher plants in an amount of 10 cms per 1000 cm2 of base surface.
After the plants had been stored for 5 days at a TL exposure of about 3000 lux, a relative humidity of about 85 / o and a temperature of 220 C, the degree of leaf burn was measured in kidney beans, nasturtiums, potatoes, creeper beans, peas, Cucumbers, Tradescantia (two varieties), tobacco, Begonia (two varieties), Coläus and Capsicum were noted. No leaf damage was found in any of the varieties mentioned.
The compounds of the formula mentioned can be processed into fungicidal preparations in various ways: a) miscible oils with 15 to 20% active ingredient, 150/0 nonionic emulsifying agent and other solvents (especially ketones, e.g. cyclohexanone); b) aerosols with as solvents, e.g. B. acetone, methyl ethyl ketone and cyclohexanone and as propellant methyl chloride or freon; c) wettable powder with 50 to 808/0 active substance and further fluxes, e.g. B. fatty alcohol sulfonates, and dispersants and / or carriers, e.g. B.
Kaolin, chalk, clay and additives to prevent clumping, e.g. B. colloidal silica; d) dust powder with z. B. 5 0 / o active substance in a mixture of kieselguhr and dolomite marl; e) Seed protectants, e.g. B. 500/0 of the active substance in association with kaolin with the addition of adhesion substances z. B. spindle oil; f) dust powder, in which the active substance in the form of a solution on a dust-like absorbent carrier, z. B. kieselguhr, attached and the solvent is evaporated.
