Diese Verbindung zeigt eine hervorragende di- uretische und natriuretische Wirkung. Dies geht aus Vergleichsversuchen mit dem im Handel erhältlichen Chlorothiazid (6-Chlor-7-sulfamyl-1,2,4-benzothiadi- azin-1,1-dioxyd) hervor. Akute Versuche von sechs stündiger Dauer am Hund zeigen die nachstehenden Ergebnisse, wobei der neuen Verbindung die Be zeichnung Su-5879 zugeordnet wurde.

Aus dieser Tabelle ergibt sich, dass die diuretische Wirkung der neuen Verbindung am Hund wesentlich besser ist als diejenige von Chlorothiazid. Bei einem weiteren Vergleich der diuretischen Wirkung der neuen Verbindung mit Chlorothiazid erhielten Ratten, denen seit 18 Stunden die Nah rung entzogen war, 0,2 o-/o-ige Natriumchloridlösung in einer Menge, die 5 o-/9 ihres Körpergewichtes ent sprach. Unmittelbar anschliessend wurde ihnen je eine dieser Verbindungen, gelöst in 0,1 cms ver dünnter Natronlauge, oral gegeben.
Der spontan ent leerte Urin wurde in graduierten Zylindern während der nächsten drei Stunden gesammelt, sein Volumen gemessen und in Prozenten der Flüssigkeitszufuhr umgerechnet. Die Konzentrationen von Natrium und Kalium wurden flammenphotometrisch bestimmt und die Gesamtausscheidung dieser Elektrolyte während der dreistündigen Versuchsperiode berechnet. Die in der folgenden Tabelle zusammengefassten Werte sind Durchschnittswerte von 8-12 Ratten pro Gruppe.

Die neue Verbindung besitzt auch bei Anwen dung am Menschen eine starke diuretische und natri- uretische Wirkung, während die Kaliumausscheidung nicht in gleichem Masse gefördert wird.
Bei Patien ten mit Flüssigkeits- und Kochsalzretention infolge Erkrankung des Herzens, der Leber (Cirrhose) oder der Nieren (Nephrose) bewirkt die Verbindung eine therapeutisch erwünschte Förderung der Flüssigkeits- und Kochsalzausscheidung, ohne dass gleichzeitig Gefahr einer Kaliumverarmung besteht.
Die für eine derartige Wirkung erforderlichen Dosen bei peroraler Applikation liegen durchschnittlich zwischen 25 und 100 mg pro Tag. Die neue Verbindung ist auch in der Lage, den arteriellen Hochdruck zu senken, vor allem, wenn sie gleichzeitig mit einem andern blut drucksenkenden Mittel gegeben wird.
Die neue Verbindung soll als Heilmittel in Form von pharmazeutischen Präparaten verwendet werden. Das erfindungsgemässe Verfahren zur Herstellung der neuen Verbindung ist dadurch gekennzeichnet, dass man 5-Chlor-2,4-disulfamyl-anilin oder ein Salz davon mit einem Halogenid oder Ester einer Essig säure der Formel Hal-CH.- COOH umsetzt, worin Hal für ein Halogenatom wie Brom oder Jod, oder in erster Linie Chlor, steht.
Als Halogenid der ge nannten Säure kommt in erster Linie das Chlorid zur Verwendung. Als Ester sollen zum Beispiel der Methyl- oder Äthylester genannt werden. Diese Re aktion wird vorzugsweise in Gegenwart eines Äthers wie p-Dioxan durchgeführt, wobei ein sich gegebenen falls bildender Äther-Benzothiadiazinkomplex durch Kochen in Wasser gespalten werden kann. Die Re aktion lässt sich bei erhöhter Temperatur, wie der Siedetemperatur des Lösungsmittels, vervollständi gen. Sie kann, wenn erwünscht, auch in Gegenwart eines inerten Gases, z. B.
Stickstoff, durchgeführt werden.
Je nach den Reaktionsbedingungen erhält man die neue Verbindung in freier Form oder in Form ihrer Salze. Erhaltene Metallsalze können zum Bei spiel durch Reaktion mit wässrigen sauren Mitteln wie Mineralsäure, z. B. Halogenwasserstoffsäure, bei spielsweise Salzsäure, oder Schwefelsäure, in die freie Verbindung übergeführt werden.
Diese wie derum lässt sich in die Metallsalze wie Alkalimetall- salze überführen durch Behandeln zum Beispiel mit einem Metallhydroxyd wie Natrium- oder Kalium hydroxyd, in einem Lösungsmittel wie einem Alkanol, z. B.
Methanol oder Äthanol, oder in Wasser und anschliessendem Abdampfen des Lösungsmittels, oder durch Reagierenlassen der freien Verbindung in einem Äther wie p-Dioxan oder Diäthylenglykol- dimethyläther, mit einem Alkalimetallhydrid oder -amid, z. B. Natrium- oder Kaliumhydrid oder -amid.
Im nachfolgenden Beispiel sind die Temperaturen in Celsiusgraden angegeben. <I>Beispiel</I> Zu einer am Rückfluss kochenden Lösung von 2 g 5-Chlor-2,4-disulfamyl-anilin in 50 cm3 p-Dioxan wird langsam 0,912 g Chloressigsäurechlorid gege ben und die Lösung 3 Stunden am Rückfluss ge kocht. Nach dem Abkühlen und Zufügen von Was ser fällt ein kristalliner Niederschlag aus, der abfil- triert wird. Dieses Produkt wird in heissem Dioxan gelöst und die erhaltene Lösung eingeengt.
Der sich dabei bildende Dioxankomplex wird abgetrennt und mit kochendem Wasser erhitzt. Man erhält so das 6-Chlor-7-sulfamyl-3,4-dihydro-1,2,4-benzothiadi- azin-1,1-dioxyd vom F. 264-265 .
Das Natriumsalz dieser Verbindung erhält man durch Eindampfen einer 6-Chlor-7-sulfamyl-3,4-di- hydro-1,2,4-benzothiadiazin-1,1-dioxyd und eine äquimolare Menge Natriumhydroxyd enthaltenden, wässrigen Lösung.

This table shows that the diuretic activity of the new compound on dogs is significantly better than that of chlorothiazide. In a further comparison of the diuretic effect of the new compound with chlorothiazide, rats which had been deprived of food for 18 hours were given 0.2% sodium chloride solution in an amount corresponding to 5% of their body weight. Immediately thereafter, each one of these compounds, dissolved in 0.1 cms of dilute sodium hydroxide solution, was given orally.
The spontaneously emptied urine was collected in graduated cylinders over the next three hours, its volume measured and converted as a percentage of the fluid intake. The concentrations of sodium and potassium were determined by flame photometry and the total excretion of these electrolytes during the three-hour test period was calculated. The values summarized in the following table are average values from 8-12 rats per group.

The new compound has a strong diuretic and natriuretic effect even when used on humans, while potassium excretion is not promoted to the same extent.
In patients with fluid and saline retention as a result of diseases of the heart, liver (cirrhosis) or kidneys (nephrosis), the compound promotes the excretion of fluids and saline, which is therapeutically desirable, without the risk of potassium depletion.
The doses required for such an effect for oral administration are on average between 25 and 100 mg per day. The new compound is also able to lower arterial high pressure, especially when given at the same time as another blood pressure lowering agent.
The new compound is intended to be used as a remedy in the form of pharmaceutical preparations. The inventive method for the preparation of the new compound is characterized in that 5-chloro-2,4-disulfamyl-aniline or a salt thereof is reacted with a halide or ester of an acetic acid of the formula Hal-CH.-COOH, where Hal is a halogen atom such as bromine or iodine, or primarily chlorine.

In the following example, the temperatures are given in degrees Celsius. <I> Example </I> 0.912 g of chloroacetic acid chloride is slowly added to a refluxing solution of 2 g of 5-chloro-2,4-disulfamyl-aniline in 50 cm3 of p-dioxane and the solution is refluxed for 3 hours . After cooling and adding water, a crystalline precipitate separates out and is filtered off. This product is dissolved in hot dioxane and the resulting solution is concentrated.
The dioxane complex that forms is separated off and heated with boiling water. This gives 6-chloro-7-sulfamyl-3,4-dihydro-1,2,4-benzothiadiazin-1,1-dioxide with a melting point of 264-265.
The sodium salt of this compound is obtained by evaporating an aqueous solution containing 6-chloro-7-sulfamyl-3,4-dihydro-1,2,4-benzothiadiazine-1,1-dioxide and an equimolar amount of sodium hydroxide.
