Verfahren zur Herstellung von Omegamycin Die Erfindung betrifft die Herstellung von Omegamy ein, eines wertvollen Antibioti kums, das auch unter dem Namen Tetra cyclin bekannt ist.
Während der letzten Jahre wurden eine Anzahl von Produkten isoliert, die das aehstum von Bakterien und Pilzen hemmen mid welche wertvolle therapeutische Eigen seliaften besitzen. Es sind dies unter anderem Penieillin, Streptomycin, Gramieidin, Tyro- cidin, Bacitracin, Subtilin, Streptothriein, :
l.ureomyein (Chlorotetracyclin), Terramycin (Oxytetracyclin). Einige dieser Produkte Haben sich wegen ihrer Wirksamkeit gegen pathogene Organismen als äusserst wertvoll erwiesen. Andere zeigten sich nur von be- sehränktem Wert teilweise wegen ihrer Toxi- iität. Chlorotetra.eyclin und Oxytetraeyclin sind besonders wertvoll wegen ihres breiten Wirkungsspektrums.
Omegamycin, das ebenfalls ein breites Wirkungsspektrum aufweist, ergibt bessere Blutspiegel und weniger Nebenreaktionen als ('lilorotetracyclin und Oxytetracyclin und ist insbesondere stabiler als Chlorotet.ra.cyclin in alkalischem Medium.
Das erfindungsgemässe Verfahren ist da- (lureh f),ekennzeichnet, da.ss in einer wässrigen, Nährstickstoff und Kohlehydrat enthaltenden Nährlösung während zwei bis drei Tagen unter aeroben Bedingungen eine Omegamycin produzierende Species von Streptomyces oder von Mutationsprodukten davon submers ge züchtet wird,
worauf das so erzeugte Omega- mycin aus der Fermentationslösung abge trennt wird. Das so hergestellte Antibiotikum kann gegebenenfalls nachträglich in ein Salz einer Base oder Säure übergeführt werden.
Das in dieser Weise hergestellte Anti biotikum wirkt inhibierend auf das Wachstum gram-positiver und gram-negativer Bakterien. Es ist befähigt zur Salzbildung mit Säuren und Metallen und besteht aus den Elementen Kohlenstoff, Wasserstoff, Stickstoff und Sauerstoff. Ferner weist es ein Spektrum von Rf-Werten auf, wie unten beschrieben.

Das Antibiotikum kann u. a.. hergestellt werden, indem man unter kontrollierten Be dingungen eine bisher unbeschriebene Species von Mikroorganismen kultiviert. Diese vor läufig Streptomyces BL- 567201 genannte Speeies wurde aus einer Bodenprobe isoliert.
Folgendes dient zur Charakterisierung dieser Organismen: BL 567201 Species nova., welche Omega- mycin erzeugt, gehört zu der Gattung Strepto- myces. Ihr Wachstum geht gut vor sich auf Glycerin - Asparagin - Rindfleischextrakt bei 30 C. Auf diesem Medium bilden sich maus graue, luftige Hyphen, während ein gelb. grünes Pigment im Agar abgeschieden wird.
Das l#lycelium besteht. aus verzweigten Hy- phen, deren jüngere Elemente gram-positiv sind. Auf :den Lufthyphen wachsen Conidien. Eine Kultur des lebenden Mikroorganismus BL 567201, die aus einer Bodenprobe isoliert und als Streptomyces viridifaciens identifi ziert worden war, wurde in der American Type Culture Collection, Washington, D. C., deponiert und deren permanenten Sammlung von Mikroorganismen unter dem Zeichen ATCC 11989 einverleibt.
Das Antibiotikum kann ferner u, a. auch durch Kultivieren unter speziellen Bedingtin- gen der einen oder andern bisher unbekannten Speeies von Mikroorganismen erzeugt werden: Streptomyces BL 567714, Streptomyces BL 678033, Streptomyces BL 678035, Streptomy ces BL 678040, Streptomyces BL 678046, Streptomyces BL 678079 und Str eptomyces BL 678110 (vorläufige Bezeichnungen).
Alle diese Orga nismen wurden aus Bodenproben isoliert. Sie alle gehören zu der kürzlich als St.reptomy ces spezifizierten Gattung. Alle diese Organis men wurden auf Glukose-Asparagin-Fleiseh- extrakt-Agar während 14 Tagen bei 32'C wachsen gelassen. Es zeigten sich dabei die folgenden Erscheinungen:

Als Omegamycin produzierende Organis men kommen nicht nur die bisher genannten in Betracht, sondern alle Omegamycinprodu- zierenden Organismen, insbesondere auch die jenigen, welche durch Mutation, z. B. durch Einwirkung von Röntgenstrahlen, U.V.-Strah- lung usw., aus den bisher genannten hervor gehen können.
Omeganiycin ist ein wertvolles Therapeu tikum sowohl in der menschlichen als auch in der Veterinärmedizin. Es besitzt die Vorteile eines breiten Wirkungsspektrums und zeigt einen hohen Blutspiegel und geringe To.Ylzi- tät. Ferner zeigt es hochgradige Widerstands fähigkeit gegen Zersetzung .durch Flitze oder Wasser sowohl in saurem als auch in alkali- seliem Medium.
Gewisse Metall- und Säure- additionsverbindungen des Tetracyclins er wiesen sieh als noch nützlicher als die Tetra- eyelin-Base, -wegen ihrer geringeren Hygro- skopizität. und grösseren Wasserlöslichkeit. Das Antibiotikum Omegamycin erwies sich in vitro aktiv gegen eine Anzahl von gram-positiven und gram-negativen Bakte rien.
Die folgende Tabelle zeigt seine anti biotische Wirksamkeit im Vergleich zu Aureo- myein - HCl, Terramycin - HCl und Tetra eyclin, wenn diese Stoffe in die Furche einer Agar-Infusion bei pu = 7,0 eingebracht wer den.

<tb> <B>3</B> Der Spektraltest wird wie folgt ausge führt:
Ungefähr 30 ml einer sterilen Infu- sionsbrühe ( Difeo> ), mit 2o/oi Agar als Ver- festigungsmittel, werden in eine sterile Petri- schale (etwa 9 cm Durchmesser) eingebracht und erstarren gelassen. Mit einem sterilen Spatel wird hierauf eine Furche von 8 mal -10 mm in die Gallerte gezogen. Der Grund der Furche wird mit einem oder zwei Tropfen flüssigem Aga.r bedeckt.
Hierauf wird aus einer 24stündigen Nährbrühe-Kultur jedes Testbakteriums, vorgängig bei 37 C inku- biert, mit einer schmalen Schlinge ein Strei fen gezogen, der sich vom Ende der Furche zum gegenüberliegenden Rande der Petri- schale erstreckt. Anschliessend wird die Furche mit einer 5 mg/ml Lösung des Anti biotikums gefüllt. Die Schale wird dann 18 bis 24 Stunden bei 37 C gehalten. Hierauf wird die Inhibitionszone vom Ende der Furche zum Punkt, wo das Wachstum der Testorganismen einsetzt, linear ausgemessen.
Die Diffusions-Platten-Testmethode zur Bestimmung der Aktivität des Omegamy eins wird wie folgt ausgeführt: Iirtltur--lledium: Streptomycin-Test-Aga.r (mit. Hefeextrakt) wird in 1 Liter destilliertem Wasser suspen diert, so dass ein pH-Wert von 6,2 oder 8,0 resultiert und eine Mischung von 1,5 g Fleischextrakt, 3 g Hefeextrakt, 6,0 g Pepton und 15 g Agar zugegeben. Die Suspension wird 5 Minuten stehengelassen, zur gleich mässigen Verteilung durchgemischt und unter Rühren leicht erwärmt.
Anschliessend wird 1 oder 2 Minuten bzw. bis alles gelöst ist, zum Sieden erhitzt. Das Kulturmedium wird hierauf ausgebreitet und bei 121 C sterilisiert (etwa -1 atü Dampfdruck, 15 Minuten).
Inoculum: Der -Testorganismus ist Bazillus subtilis, Ameriean Type Culture Collection <B>6633.</B> Eine Sporenaufschlämmung, enthaltend 50 000 000 keimfähige Sporen pro ml wird der oben beschriebenen flüssigen (auf 53 C gekühlten) Agar-Testlösung zugegeben, so dass ein Inoe,i- lat von 21/a entsteht. Herstellung <I>der</I> Platterz:
21 ml der sterilen. oben beschriebenen Aga.r-Testlösnng werden in einer Vielzahl flacher steriler Petriplatten eingebracht und erstarren gelassen. .l ml des inoculierten Agars werden hierauf gleichmässig über die Oberfläche der Basissehicht in jeder Platte verteilt.
Hierauf werden Platten aus rost freiem Stahl, die je eine Serie von Löchern enthalten, auf das Agar-Medium aufgelegt, nachdem letzteres auf Zimmertemperatur aus gekühlt war, worauf das zu prüfende anti- biotisehe Material in die Löcher eingebracht. wird.
<I>Puffer:</I> Es werden Puffer vom pH 6,2 oder 8,0 zur Verdünnung der Agar-Testlösung gebraucht. Für pH 6,2 dient ein Zitratpuffer. Er wird hergestellt, indem man 192,12 g wasserfreie Zitronensäure mit 106,3 g Natriumhydroxy d in einem Liter dest. Wasser löst und die Mi schung auf 1/1o der Konzentration mit. dest. Wasser verdünnt.
Der PH-Wert des Puffers muss potentiometrisch geprüft werden lind wenn nötig, durch Zugabe von Zitronensäure oder Natriumhydroxyd genau eingestellt wer den. Änderungen im pH oder in der Puffer konzentration beeinflussen die Grösse der In- hibitionszone merklieh. Sterilisation des Puf fers ist unnötig. Die Stoeklösung wird mit Chloroform oder Tolu,ol abgedeckt und frische Gebrauehslösungen werden täglich daraus hergestellt.
Für den pH-Wert 8,0 wird ein Phosphat puffer als Verdünnungsmittel gebraucht. Er wird hergestellt, indem man 95 ml molare K21-IP01-Lösung mit 5 ml molarer KH2P0.1- Lösung vermischt und die Mischung mit dest. Wasser 1 zu 10 verdünnt.
Der pH-Wert des Puffers wird pot:entiometrisch kontrolliert und, wenn nötig, durch Zugabe von einem der beiden Phosphate genau eingestellt.. Ände rungen im pH-Wert oder in der Konzentration des Puffers beeinflussen die Grösse der Inhi- bitionszone merklich. Es ist unnötig, den Puffer zu sterilisieren. Die Stoeklösung wird unter Chloroform oder Toluol aufbewahrt und die Gebrauchslösung täglich frisch angesetzt <I>Test:</I> Die zu untersuchenden Proben werden, sofern nötig, mit dem Puffer verdünnt.
In drei Löcher jeder Platte wird eine einzelne Verdünnung der Probe eingebracht. Nach In kubation bei 32 C werden die Zonendurch messer gemessen und ausgemittelt.
St.reptomy ces BL 567201 wurde aus einen Stamm von S. aureofaciens (NRRL 2209) ab- gezweigt, welcher vom Northern Regional I.' e- search Laboratory, Peoria, Illinois, erhalten, wurde, wo er als authentischer Aureomycin produzierender Stamm aufbewahrt war.
Die Abzweigung geschah auf Grund von Beob achtung der Waebstumscharakteristika auf Glycerin-Asparagin-Fleischextrakt-Agar und Czapek-Dox-Agar mit 1% Dextrin. Die dabei verwendeten Agarmischungen und sich erge benden Resultate waren folgende:

Als für den Fermentationsprozess geeig nete Stichstofflieferanten, welche zum Teil waelistumsfördernde Stoffe, organische und anorganische, stickstoffhaltige Verbindungen, insbesondere proteinhaltiges Material, enthal ten, kommen zahlreiche Produkte in Frage, z. B..

Palmölmehl 1i okosölmehl Leinsamenmehl Erdnussölmehl Sonnenblumenölmehl Die proteinhaltigen Substanzen brau,eli, nielit in hohem Reinheitsgrad angewandt werden; auch weniger reines Material, na mentlich solches, das Spuren von Wachs- tumsfaktoren und beträchtliche Mengen von Mineralnährsalzen enthält, ist geeignet.
Wegen dem Rohprodtktcha.rakter mancher dieser Materialien ist es natürlich nicht möglich, genaue Angaben über .die zugesetzten Men genverhältnisse der Stickstoffsubstanzen zu machen. Eine Menge von ungefähr 0,1 bis 5,0 Gewichtsprozent, gerechnet auf Trocken substanz, ist in den meisten Fällen der ge bräuchliche Anteil der Stickstoffsubstanzen im Nährmedium. Insbesondere ergeben holte N-Anteile im Nährmedium Zusätze von Soja- Proteinen und Soja-Peptonen, das heisst teil weise zersetzten Sojaprodukten, wie u. a. hy- drolysiertein Sojabohnenmehl.
Der pH-Wert des Fermentationsmediums ist anfänglich vorteilhaft ungefähr 5,0-6,5, vorzugweise 5,8-6,2. Die Fermentations- temperatur liegt vorzugsweise bei ungefähr 26 bis 280 G. Die höchste Aasbeute ergibt sich normalerweise in 1 bis 3 Tagen, wobei der Optimalwert mit den sonstigen Kultivier verhä,ltnissen variiert.
Omegamycin ist sowohl in vivo als auch in vitro wirksam und zeigt bemerkenswerte chemotherapeutische Wirksamkeit. bei experi menteller Infektion von Mäusen. Die Ergeb nisse solcher Teste zugleich mit der Bestim mung der Giftwirkung sind in .der folgenden Tabelle enthalten:

wobei jede Dosis LD5o für sich allein genügt, innerhalb einer Gruppe von Mäusen 50 /a zu töten. Die Injektion wurde auf einmal verabreicht, und zwar nach der zweiten Dosis der Testdroge. Die Testdroge wurde in zwei gleichen Teilen ungefähr innerhalb eines 18-Stunden-Inter- valles verabreicht. Die Tiere wurden vier Tage lang beobachtet, und die Todesfälle inner halb jeder Gruppe als Prozent der Anzahl Tiere pro Gruppe ausgedrückt. Die Prozent zahl der Todesfälle wird gegen den Loga rithmus der Dosiswerte in mg pro kg Haus- gewicht aufgetragen.
Aus der besten durch die experimental sich ergebenden Punkte ge zogenen Linie ergibt sieh diejenige Konzen tration der Droge, welche die Hälfte der Tiere unter :den angewandten Versuchsbedingungen schützt. Der Antilogarithmus dieses Aus- drLickes wird CD5o-Wert genannt.
<I>Beispiel 1</I> Bei Herstellung von Omegamycin im La boratoriumsmassstab wird die Fermentation in luftoffenen Schüttelflaschen, die gegen Verunreinigung mit Watte oder Gaze ge schützt sind, durchgeführt. Streptomyces BL 567201 wird in einem geeigneten Nähr medium in submersem Zustand gezüchtet, \wobei Bewegung und Belüftung der Kultur mittels einer Schüttelmasehine bewirkt wird, derart, dass ein Zersprühen, Verspritzen oder Durchschütten des Gemisches durch eine sauerstoffhaltige Atmosphäre erfolgt.
500 ml einer Nährlösung aus 10/0 Saccharose 1% :Sojabohnenmehl 1% Sojapepton 1,511/o KH2P04 0,5% (NH4)
2HP04 Wasser q. s. 100% werden in 4-Liter-Flaschen eingefüllt und sterilisiert. Nach der Behandlung im Auto klaven wird das Nährmedium mit etwa 1 Vo- lumprozent einer trüben, wässrigen Sporen- suspension von Streptomyces geimpft. Der pH- Wert wird anfänglich auf 6,0-6,2 eingestellt.
Die Flascheninhalte werden hierauf bei 26 bis 280 C 48 Stunden lang der Inkubation unterworfen, wobei mit 130 Huben pro Mi nute und einer Hubhöhe von 3 cm geschüttelt wird. Am Ende der Inkubationsperiode war die Fermentationsflüssigkeit bläulichgrün und ergab im obenbeschriebenen Test Inhibitions- zonen von etwa. 27 mm in 30facher Verdün nung. Die Lösung enthielt Omezamycin, das auf eine später beschriebene Weise abgetrennt wurde.
<I>Beispiel</I> N Zur Herstellung von Omega.my cin in grösserem Massstab wurde eine Fermentations- lösung folgender Zusammensetzung (Gew.O/o) angesetzt 1% Getreidekornpresssaft 10/a Saccharose 0,5% (NH,4)2HP0,4 1,50/9 KH2P04 0,2% M,-S04 - 7 H20 Wasser 11.s.
10011/o pH '6,2-6>4 2500 ml der Lösung wurden in eine 10-Liter-Flasche eingefüllt. Die Sterilisation erfolgte mit Dampf bei 118-120 C während einer Stunde. Nach Abkühlung wurde mit etwa 0,5 Volumprozent einer trüben, wässri- gen Sporensuspension von Streptomyces ge impft. Der Flascheninhalt wurde 48 Stunden lang bei 26-28 C auf einer Schüttelmasehine der Inkubation unterworfen und sterile Luft über die Oberfläche der Flüssigkeit geblasen.
Aus der Flasche wurde die streptomy ces- haltige Nährlösung in den Fermentationstank (siehe Beispiel 3) unter völlig aseptischen Be dingungen übergeführt. Die Lösung wurde nach der Fermentation, wie unten angegeben, aufgearbeitet. und das Omegamycin isoliert. <I>Beispiel 3</I> Streptomyces BL 567201 kann zur Her stellung von Omegamyein in grossem Massstab als Tiefkultur gezüchtet werden.
Hierfür sind stationäre Fermentationsfässer, ausge rüstet mit Rührern und Belüft.ungsvorrieli- tungen, besonders geeignet. Man verwendet zweckmässig ein Nährmedium, das 56,8 1 Kornpresssa.ft, 56,8 kg, Saccharose, 28,4 kg (NH4)2HP04, 85,2 kg KH2P04, 11,3 kg MgS04 - 7 H20 und Wasser auf 6700 1 ent hält.
Dieses Nährmedium wird zweckmässig in einem 9000 Liter fassenden, mit Glas ausge legten Fermenta.tionsgefäss, ausgerüstet mit einem Wassermantel zum Temperieren, einem Rührei aus rostfreiem Stahl und einer Be lüftungsvorrichtung, verarbeitet. Das Nähr- niedium wird mit. Dampf unter Druck steri lisiert und dann abgekühlt. Nach dem Sterili sieren sollte der pH-Wert bei etwa 6,2 liegen.
Die Nährlösung wird mit 15 Volumprozent einer waehsfähigen Kultur, die entweder in einem. ä.hnliehen Fermentationsgeläss gezüch tet oder in einem der obenbesehriebenen La- boratoriuinsansätze erhalten wurde, inoku liert. Hierauf wird der Inhalt des 9000-Liter- Gefässes bei einer Temperatur von 28 C zwei bis drei Tage lang der Inkubation unterworfen.
Dabei wird mit 90 L'. p. M. gerührt und 3 m pro Minute sterile Luft in das Nährmedium eingeblasen. Am Ende der Inkuba.tionsperiode enthält die Kulturlösung normalerweise ge- nün-erid antibiotische Wirksubstanz, um innerhalb einer Zone von 23 mm Inhibition ge-enüber den Testorganismen zu ergeben, wenn sie in 16facher Verdünnung nach der oben angeführten Methode getestet wird. Das ()me,-@amycin kann, wie unten beschrieben, isoliert werden.

B. indem man das Mycelium abfiltriert, die Brühe (vorzugs iveise beim pH etwa. 8,5) mit Butanol oder Methylisobutylketon umrührt, die Lösungs- mittelschieht, welche das Omegamycin ent hält, abtrennt, sie auf ein kleines Volumen einengt, und hierauf mit einem flüssigen niedrigen Kohlenwasserstoff, z.
B. mit Petrol- äther vom Siedeintervall 85-l00 C, versetzt, wobei das Omegamycin als Base ausfällt, so fern die Brühe alkalisch, z. B. mit dem pH- W ert 8,5, war, oder als Hydrochlorid, sofern die Brühe vor der Extraktion mit Salzsäure angesäuert worden war.
Das so abgetrennte Omegamycin kann weiter gereinigt werden, indem man es in Ammoniaklösung einträgt, oder auch durch Adsorption aus einer Lö sung der freien Base an chromatographischen Adsorbentien (z. B. an Kieselsäure), gefolgt von Auswaschen (z. B. mit. Aceton oder Me thanol) zur Entfernung von Verunreinigun- gen und Eluieren mit einer Säure. Das ge- reinigte Omegamycin-Salz lässt sich aus dem durch Kristallisation, Fällung, Lyophilisation oder dergleichen gewinnen.
Die flüssigen, niedrigeren Kohlenwasserstoffe, die sich als Fällungsrea.genzien verwenden lassen, sind mit Vorteil Petroläther der Siedebereiche 28 bis 38<B>0</B> C, 60-71<B>0</B> C oder 85-100<B>0</B> C.
Durch Zugabe von Säuren zu Omega.niycin in Wasser, bis klare Lösung eintritt, können auf einfache Weise anorganische und orga nische Säuresalze erhalten werden. Feste Salze können hergestellt werden, indem man das PH einer solchen Lösung eines Omega mycinsa.lzes auf einen Wert knapp unter den Punkt. bringt, wo das Antibiotikum auszu fallen beginnt. Die Lösung kann dann ge trocknet werden, indem man sie in gefrorenem Zustand unter Vakuum setzt. Salze des Omegamycins mit Säuren werden erhalten, indem man eine Lösung des Salzes in Wasser bei niedrigem pH verdampft. Mineralsäuren, die Salze ergeben, sind z. B.
Salzsäure, Schwe felsäure und Phosphorsäure. Zur Salzbildung geeignete organische Säuren sind Zitronen säure, Weinsäure, Gluconsäure, Benzoesäure, Essigsäure, Ascorbinsäure, Bernsteinsäure, Nicotinsäure, Ameisensäure, Maleinsäure usw. Da Omegamycin amphoter reagiert, lassen sich auch Salze verschiedener metallischer Elemente mit dem Antibiotikum bilden, z. B. von Natrium, Kalium, Calcium, Magnesium.
Die Alkalisalze des Omega.mycins lassen sich herstellen, indem man eine wässrige Suspen sion des Antibiotikums mit zwei Äquivalenten des Alkalihydroxyds behandelt. Die festen Metallsalze des Antibiotikums erhält man durch Verdampfen einer wässrigen Lösung desselben bei geeignetem pH. Durch Aufarbei ten des Omegamycins aus neutraler Lösung, z.
B. durch Lyophilisieren, erhält man die freie Base, normalerweise als Trihydrat, so fern Wasser zugegeben ist und nicht in der Wärme gearbeitet wird. Omegarnyein-trihy- drat entsteht. ebenfalls, wenn die Base in einem wasserlöslichen, organischen Lösungs mittel, wie Methanol oder Äthanol, gelöst und die Lösung in Wasser gegossen wird. Festes Calciumomegamycin lässt sich in Omega.my- cin-trihydrat überführen durch Lösen in wässriger Säure, wie Schwefel- oder Salzsäure, z. B. bei PH 3 oder weniger, Zugabe eines Agens zur Bindung des Caleiums, z. B.
Na triumversena.t (äthylendiamin-tetraessigsaui-es Natrium), und indem man schliesslich durch Zugabe von Alkali zur Steigerung des pH- Wertes auf wenigstens 6, aber nicht höher als 8, Omega.mycin-trihydrat ausfällt. Omegami-.. ein-trihydrat lässt sich aus wässrigein Alkohol, z. B. Methanol, umkristallisieren und kann in die wasserfreie Form übergeführt werden, indem man mässige Wärme im Vakuum über einem Trocknungsmittel anwendet.
Die Säuresalze des Omeganiyeins können auf verschiedene Weise hergestellt werden; z. B. können stöehiometrische Mengen Säure zu einer Lösung der Omega.my cinbase in Wasser oder einem organischen Lösungsmit- tel, wie Butanol, Aceton, Methanol, zugege ben werden, wobei sieh das Säuresalz bildet;
sofern gewünscht, kann es aus Wasser durch Lyophilisieren abgetrennt oder durch Filtrie ren des ausgefallenen Salzes (aus Äthyl- acetat) oder durch Fällung des Salzes durch Zugabe eines andern organischen Lösungs- mittels (z. B. Petrolätlier zu Butanol oder Cyclohexa.n zu Ä t-hylacetat) oder durch Re duktion der Lösungsmittelmenge im Vakuum, bis das Salz unter Kühlung auszufallen be ginnt (z.
B. aus Bu,tanol), abesehi.eden wer-- n den. Es lassen sich noch viele andere Metho den zur Herstellung der Salze anwenden; z. B. kann man Omegamycin in das Chlor- liydrat überführen, indem man es ehromato- graphiseh adsorbiert (z. B. an Kieselsäure). hierauf mit einer Säure eluiert (z.
B. mit m-ässriger, methy la.lkoholischer oder butanoli- seher Salzsäure). Nach einer andern Methode wird die Omega-myeinbase aus einer Fermen tationsbrühe mit. einem p1., von 8-9 mit.
Bu- ta.nol extrahiert, das Butanol abgetrennt, die Omegamy einbase in das Hydrochlorid über geführt und letzteres mit wässriger Salzsäure in wä.ssrige Phase aufgenommen. Das so iso lierte Omegamycin kann z. B. durch Lyoplii- lisation isoliert werden.
Nach einer bevorzug ten Methode wird die Omeganiycinbase in Salze übergeführt, indem man sie in troeke- izem Aceton oder n-Propanol löst und wasser freie Säure zugibt, z.
B. Chlorwasserstoffgas, Schwefelsäure in trockenem Aeeton, Phos phorsäure, Weinsäure, Zitronensäure, Salpe- tersäure in trockenem Isobutylketon, worauf inaii durch Filtration das ausgefallene Salz der entsprechenden Säuren abtrennt.
Das Onieganiy ein kann aus den Fermenta- tionsbrühen auf verschiedene Weise isoliert werden, wie im folgenden durch Beispiele gezeigt wird: <I>Beispiel 10</I> Omegamycin lädt sieh aus alkalischen Ferinentationslösungen leicht durch unpolare organische Lösungsmittel extrahieren.
1 Liter qtreptomy ees-BL-567201-±'ermen- tationsbrühe vom pH 8,5 wird mit 0,5 Liter 3Ietliyl-isobiitylketon extrahiert.
Das orga- niselie Lösungsmittel wird abgetrennt, mit Wasser gewaschen, durch azeotrope Dest.illa- l ion auf ein Volumen von etwa 20 nil, redu ziert und 250 ml Petroläther (85-100 C) z iuregeben. <B>1</B> '- -)
0 m <B>-</B> Ome- ga myein fallen aus und -erden abfiltriert. Sie zeigen eine Aktivität bei 1 nig/ml, verdünnt 1:6.1, von 19,5 mm (Zone:ndui,chniesser bei B. ssubtilis-Plattentest bei pH <B>6,2</B> auf Agar), 24,2 mm, verdünnt 1 :
l6, und 27,7 mm, verdünnt 1 :4. Die Ori- ginalbrühe ergab im Test 27,3 mm bei der Verdünnung 1 :4.
<I>Beispiel 1.1</I> 4,5 1 Streptoinvees-BL-567201-Fermenta- tionslirühe werden vorn, pH 6,7 auf PH 5,5 ge b raelit und mit 3 1 Butanöl extrahiert. Der Bu- t < inolextrakt wird abgetrennt, mit Wasser ge- a:
schen, durch azeotrope Destillation auf etwa 35 in] eingeengt und 350 ml technisches I leptan zugegeben. Omegamy ein fällt als Testkörper aus und wird abfiltriert [1,2 mg, niit einer Aktivität bei 1 mg/ml, verdünnt I :

Die verbleibende Brühe wird von PH 5,5 auf pH 8,5 gebracht und erneut mit 31 Butanol extrahiert. Die Butanollösung wird a.bge- trennt, mit Wasser gewaschen, auf etwa 30 ml eingeengt und zu 350 ml Petroläther (85 bis 100 C) zugegeben. Rohes Omegamycin fällt als orangegelber Festkörper aus. Es zeigt eine Aktivität bei 1 mg/ml, verdünnt auf 1:16, von 25,2 mm (Zonendurchmesser im Platten- test mit B. subtilis auf Agar bei 2)H = 6,2).
Die Originalbrühe ergab einen Testwert von 27,3 mm bei 1 :4 Verdfnnung.
<I>Beispiel 12</I> 2,5 1 Streptomyces-BL-567201-h'ernienta- tionsbrühe mit einem Testwert. von 25,7 mm bei einer Verdünnung von 1:4 werden auf pH = 6,4 gebracht und mit 2 1 Butanol extra liiert. Die Butanollösung wird abgetrennt, mit Wasser gewaschen, auf 50 ml Volumen einge engt (durch azeotrope Destillation) und zu 1 1 Petroläther (85-1000 Sdp.) zugegeben.
Dunkelgelbes, festes Omegamycin (500 mg) fällt aus, wird abfiltriert und ergibt eine Aktivität bei 1 mg/ml, verdünnt 1 :16, von 18 mm. (Zonendurchmesser beim Plattentest mit B. subtilis auf Agar bei pH 6,2) und von 23 mm bei der Verdünnung 1 :4.
Nach der Extraktion mit B.utanol wird die Brühe bei pH 6,4 drei Tage lang im Kühl raum gelagert, dann mit Natronlauge auf pH 8,5 eingestellt und mit 2 1 Butanol extra liiert. Die Butanollösung wird abgetrennt, mit Wasser gewaschen, durch azeotrope Destilla tion auf etwa 30 ml eingeengt und zu 600 m1 Petrolä:ther (Sdp. 85-100 C) zugegeben.
Dunkelbraunes Omegamy ein (140 mg, Ansatz 25) fällt aus und wird abfiltriert. Es weist eine Aktivität bei 1 mg/ml, verdünnt<B>1:</B> 16, von 28 mm auf (Zonendurchmesser im Plat- tentest mit B. subtilis auf Agar bei pH = 6,2). <I>Beispiel 13</I> 760 ml Streptomyces-BL-567201-Fermen- tationsbrühe werden mit Natronlauge auf pH 9,4 eingestellt und mit 380 ml n-Butanol extrahiert.
Das n-Butanol wird abgetrennt, durch Zugabe von wässriger Salzsäure sein pH-Wert auf 7,35 eingestellt und dann dass Bu- ta.nol im Vakuum abgetrieben.
Das verbleibende wässrige Konzentrat wird mit Wasser auf etwa. 38 ml verdünnt und dann lyophy lisiert. Man erhält 339 mg festes Omegamy ein, das eine Aktivität bei 1 mg pro ml Puffer von 25 mm (Zonendurchmesser im Plattentest auf Agar bei pH = 6,2) und von 18,7 mm bei der Verdünnung 1 : 3 aufweist.
Die ursprüngliche Brühe ergab einen Testwert von ?6,3 mm (und 20,0 mm bei der Verdünnung 1:3), während die verbrauchte Brühe 19,0 mm (12,7 mm bei der Verdünnung 1:3) ergab. Beispiel 14 5,5 1 Streptomyces-BL-567201-Ferm.enta- tionsbrühe wurden auf pH 5,5 eingestellt und mit 3 1 n-Butanol extrahiert.
Die Lösung wird abgetrennt, mit Wasser gewaschen, durch azeotrope Destillation auf etwa 35 ml eingeengt und zu 350 ml Petroläther (Sdp. 85 bis 100 C) zugegeben. 1,2 g goldgelbes festes Omegamyein fallen aus, werden abfiltriert (Substanz A) und ergeben eine Aktivität bei 1 mg/ml, verdünnt<B>1:
</B> 16, von 23,2 mm. (Zo- riendurehmesser im Plattentest mit B. subti- lis auf Agar bei pH 6,2) und 26,9 mm bei der Verdünnung<B>1:</B> 4.
Die nach der Extraktion verbleibende Brühe vom pH = 5,5 wird auf pH 8,5 einge stellt und mit. 3 1 Butanol extrahiert. Die Bu- tanollösung wird abgetrennt, mit Wasser ge waschen, durch azeotrope Destillation auf etwa 30 ml eingeengt und zu 350 ml Petrol- äther (techn. Hepta.n) zugegeben.
0,5 orangegelbes, festes Omegamyein (Ansatz 30) fallen zusätzlich aus, werden abfiltriert (Sub stanz B) und ergeben eine Aktivität bei 1 mg/ml, verdünnt 1:16, von 25,2 mm (Zo nendurchmesser im Plattentest mit B. subtilis auf Agar bei PH = 6,2) und von 28,.5 mm bei der Verdünnung 1:4.
Das so hergestellte Omegamycin kann ein deutig charakterisiert werden, selbst. wenn es mit ähnlichen organischen Substanzen v er unreinigt ist, durch sein Aktivitäts -Spek- trum (das heisst seine Rf-Werte) in verschie denen Lösungsmitteln bei der Papierstreifen- Chromatographie. Diese Methode ist verhält nismässig neu, hat sich aber geit bewährt zur Identifikation organischer Verbindungen.
Wie die Maxima eines Infrarotspektrums, er geben die'Rf-Werte einer Serie von Lösungs mitteln eine eindeutige und reproduzierbare Charakteristik eines Chemikals und dienen als fingerprint .
Die fragliche Methode besteht in folgen dem: Streifen aschefreien, dichten und sehr sa.ugfä.higen Filterpapiers (z. B. Nr. 589, Blau band spezial von Schleicher und Sehüll) etwa 12 mm breit und 58 ein lang, werden bei kon stanter Raumtemperatur in einem verschliess baren Gefäss (z. B. einem. grossen (Tlasgefäss) über den Rand einer Schale aufgehängt.
Das obere Ende jeden Streifens befindet sieh in Kontakt mit einer Lieferquelle eines bestimm ten Systems von Lösungsmitteln (ebenfalls Entwieklerphase _-genannt) in rler Schale. Jeder der Streifen hängt je an einer beson deren Schale, die je ein besonderes Lösungs- mittelsystem enthält. Das untere Ende des Streifens hängt frei und steht nicht in Kon takt. mit den unter den Streifen befindlichen Lösungsmitteln, welche den Luftraum im Ge <B>fäss</B> mit ihren Dämpfen sättigen.
Das zu prüfende Produkt (z. B. in einer Fermentationsbrühe befindlich, oder als iso lierte Substanz) wird auf eine markierte Stelle am obern, freihängenden Ende des Streifens aufgebracht. Falls es eine feste Sub stanz ist, wird diese zuvor in einem geeigne ten Lösungsmittel gelöst. Die angewandten Mengen werden so bemessen, dass geeignete Zonengrössen sich ausbilden. Zum Beispiel eignen sieh 0,005 ml einer Lösung von 1 ing Omegainy ein pro ml für den nachfolgenden Test mit B. subtilis. Der Streifen wird ge trocknet und .dann mit seinem. obern Ende in die Schale mit dem gewählten Lösungsmittel- spstem gegeben.
Die Lösung wandert. abwärts und entwickelt den Streifen, bis die Front der Lösungsmittel zu seinem untern -Ende reicht. Dies erfordert etwa 15 Stunden. Die den Streifen umgebende Atmosphäre wird ouf konstante Temperatur, frei von Luftzug und gesättigt mit den Lösungsmitteldämpfen, gehalten.
Die Streifen werden dann berausgenoin- nien, an der Luft getrocknet und auf eine Schicht Agar mit eingestelltem pfi (hier 6,2), die mit einem Testorganismus B.
subtilis hiokuliert i,st, .aufgelegt. Nach Lagern im Kühl- sclira.nk über Nacht werden die von den ver- sehiedenen Lösungsmittelsysteinen entwickel ten Streifen weggenommen, die Agarscbicliten eiitspreehend markiert, letztere über Nacht o der Inkubation unterworfen und entweder di rekt. beobachtet. oder photographiert, um ein i)leibendes Dokument zu. haben.
Der Umriss des ganzen Streifens ist ge- wöhnlieh auf der Agaroberflä.che und oft auch s aiif der Photographie sichtbar. Die Lage jedes allt.ibiotisehen Agens auf dem Streifen zeich net sich als klarer Bezirk ab, welcher mit den trüben Bezirken des Bakterienwachstums kontrastiert.
Das Streifenbild wird in Zonen eingeteilt, welche 5, 10, 10, 10, 10, 10, 10, 10, 10 und 15 %. der Distanz, zwischen der Auf- bringstelle der Probe und dem untern Strei- fenende ausmachen, und diesen Zonen wer den die RF-Werte von 1 bis und mit 10 zu geteilt.
Ein schmaler Fleck exakt in der Mitte würde dementsprechend den RF-Wert 6 erhal ten, während ein grösserer Fleck, der sich über die anliegenden Zonen erstreckt, die Rf- Werte 5, 6 und 7 erhalten würde, da jeweils die ganze Zone gezählt wird.
Unter Anwendung dieser Technik ergibt sich, folgendes Rf-Spektrum von Omegamycin (Ansatz 30) : Angewendet werden 5 ml einer 1-mg/ml-Lösung des Antibiotikums auf jedem Streifen im Test mit B. subtilis bei PH = 6,2 auf Agar, wobei die Streifen in zwölf Lö- sungsmittelsystemen entwickelt werden.

gesät- tigt mit 10% wässrigem Natriumzitrat; in diesem Falle wurde vor Aufbringen der Probe der Streifen mit 10 %igem wässrigem Natriumzitrat, das mit Ci- tronensäure auf pH = 5,7 eingestellt worden war,
gesättigt und dann ge trocknet.
<I>Beispiel 15</I> . Die Substanz A von Beispiel 14 wird mit. 50 ml wässrigem Ammoniumhydroxyd vom pH-Wert. 11,6 extrahiert, wobei 220 mg einer braunen festen Substanz zurückbleiben. Die Substanz B (0,5 g) von Beispiel 14 wird in gleicher Weise mit 30 ml Ammoniak behan delt, wobei 120 mg feste .Substanz zurück bleiben. Die basischen, wässrigen Lösungen des Omegamycins werden vereinigt und durch eine Chromatographiekolonne (12 auf 100 mm) mit 7,5 g Kieselsäure laufen gelassen. Aus dem braunen Durchlauf erhält man etwa 600 mg einer braunen festen Substanz von geringer Aktivität.
Die Kolonne wird dann mit 125 ml Aceton, hierauf mit 125 ml Alkohol eluiert und das gelbe Ehiat verworfen. Darauf wird die Ko- lonne mit 100 m1 1%iger Salzsäure eluiert. Das saure Eluat, welches noch das restliche in der Kolonne verbliebene Äthanol enthält, wird in Wasser gegeben und im Vakuum ,unter ständiger Wasserzugabe destilliert, bis alles organische Lösungsmittel entfernt. ist.
Hierauf wird mit Ammoniak neutralisiert und dann lyophylisiert. Man erhält 0,5 g Omegamy ein, das eine Aktivität bei 1 mg/ml. verdünnt 1 :16, von 24 mm aufweist. (Zonen durchmesser im Plattentest mit B. subtilis auf Agar bei pH 6,2.) Beispiel <I>16</I> Ein gemäss Beispiel 15 erhaltenes Silica- adsorbat wird bei PH 8,5 mit Butanol extra hiert und das Eluat durch azeotrope Destilla tion getrocknet.
Beim Einbringen in Petrol- äther fällt. Omegamycin aus, das abfiltriert wird (Ansatz 36). Es hat eine Aktivität bei 1 mg/ml, 1 :16 verdünnt, grösser als 30 mm. (Zonendurchmesser im Plattentest mit. B. subtilis auf Agar beim pH 6,2).
<I>Beispiel 17</I> Das Oniega.mycin kann aus Produkten jeden Reinheitsgrades auch durch über führung in sein Natriumsalz abgetrennt. wer den, indem man das unreine Produkt in Wasser mit Natronlauge behandelt, bis der p11-Wert über 9,5 liegt. Die abfiltrierte Lö sung wird dann ausgefroren und im Vakuum getrocknet, wobei das trockene Natriumsalz in Form einer stabilen, wasserlöslichen Sub stanz erhalten wird.
Beispiel <I>18</I> Der Verteilungskoeffizient von Omega- mycin zwischen Butanol und Wasser (Kon zentration in Butanol geteilt durch Konzen tration in Wasser) wurde bei allen p1.1-Werten kleiner als zu 1 befunden, aber er wird durch Zugabe von Caleiumion merklich beeinflusst. So ist der Verteilungskoeffizient.
von Oniega- nwcin zwischen Butanol und ' 0/aiger, wässri-- ger Chlorcalciumlösung kleiner als 2 (olt kleiner als 1) von pH 1 bis etwa pH 6.6, steigt dann mit wachsendem pH scharf an und ist grösser als 5 von etwa pH 7,4- bis PH 11, indem er ein Maximum von 9 bis 10 zwischen pH \.) und 10 erreicht.
Ein sehr ähnliches Verhalten des Omegamycin-Verteilungskoeffizienten in Abhängigkeit vom PH-Wert ergibt sich in Bu- tanol und 0,5% wässriger Ca.leiumchlorid- lösung mit einem Maximum von etwa 20 bei pH 10.
Es ergeben sieh dagegen keine 2-7-össercn Verteilungskoeffizienten des Oniega.mycins als 1,5 bei Verteilung zwischen Butanol und wässriger 5- oder 10%iger Natriumsulfat Lösung oder 2.-, 5- oder 10%iger Kochsalz- lösung bei p11-Werten von 4 bis 9.
Vertei lungskoeffizienten zwischen 2 und 4 ergeben sich mit 2- bis 10%iger Kochsalzlösungen beim stärker sauren PH von 2, wobei aber eine wirtschaftliche Aufarbeitung erschwert, wenn auch nicht v erunmöglieht ist.
Die Extraktion einer wässrig-eri Lösung (Fermenta.tionsbrühe) mit wenigstens 250 y/nil Omegamycin mit. 14 ihres Volumens Butanol beim PH 8,5 in Gegenwart von 0,511./0 Chlor ealcium ergab hohe Ausbringungsgrade (40 bis 50%)
an Omegainycin als teste Substanz von guter Aktivität nach Abtrennung und Einengen der Butanollösung, Rückführung in wässrige Säure, z. B. Salzsäure, und Isolie rung aus letzterer. Hierbei wird das Omeo-a- mycin wie beschrieben extrahiert.
Bei den obigen Arbeitsweisen kann das na türlich in den Fermentationsbrühen v orkom- inende Calciumion (z. B. aus Getreidepress- säften) genügen; es kann aber auch Calcitun- ion zugesetzt werden. Das Caleiumion kann in irgendwelcher Salzform zugegeben werden, z. B. als Chlorid, Karbonat, wobei seine totale Konzentration in der Brühe vorteilhaft auf etwa 0,1 bis '-),01/o, bemessen wird. --Man setzt.
vorteilhafterweise etwa. 0,1% Ca.lciumehlorid zu; man kann auch das stöehiometrische Ca- Äquivalent des in der Brühe vorhandener. Omegamyeins ergänzen, nachdem der Omega- myein- und Calciumgehalt in der Brühe zu- vor ermittelt worden sind.
Cbersehüssige 3Iennen Calciumion sind indessen uner- wünselit, cla sie Caleiumphosphat ausfällen, das seinerseits dazu neigt, Omegamy ein zu adsorbieren. Man kann den adsorbierten An teil zwar zurückgewinnen, jedoch bedingt dies zusätzlichen Arbeitsaufwand.
Ein weiterer Hinweis für eine verbesserte l,:xi Taktion des Omegamy eins mit Butanol aus fermentationsbrülren, welche Caleiumion enthalten, liegt in der Beobachtung, dass die Zugabe eines ca.leiumbindenden Agens zur Brühe die Extraktionsausbeute unter den- jenigen Wert verminderte, der erhalten wird, wenn kein Caleiumchlorid zur Fermentations- brühe zugegeben wurde.
Eine Omega.myein enthaltende Fermenta- tionsbrühe, welche unter Verwendung von Ge- treidepresssaft hergestellt. wurde, wurde auf pH 8,5 gebracht und in drei gleiche Teile ge teilt, zu welchen die angegebenen Mengen Chlorcalcium zugegeben wurden. Die Extrak tion mit Butanol wurde durch die Zugabe von Caleiumion verbessert, wie aus den nach stehenden Resultaten, die im Biotest. erhalten wurden, hervorgeht.

Beispiel <I>20</I> Zu einer Omeg>,amyein enthaltenden Fer- irientationsbrühe werden 0,10/a. CaCI.., zu.Igege- ben und die Brühe z. B. mit Soda. oder Pott- i as( -Wert von <B>-,</B> -lie oder Animoniak auf. einen PH 8,:i bis 9,0 eingestellt. Hierauf wird mit.
n-Butanol extrahiert, wobei für einmalige Extraktion 1/2 des Volumens, für wiederholte Extraktion 1/,1 bis 1/5 des Brühevolumens an Butanol verwendet wird. Die Mischung wird filtriert und die Butanolphase abgetrennt. Das Omegamycin enthaltende Butanol wird durch Vakuumdestillation auf 1/20o des ur sprünglichen Brühevolumens eingeengt.
Das Buta.nolkonzentrat und die darin befindlichen unlöslichen Anteile werden dreimal mit je einem gleichen Volumen Wasser, das mit Salzsäure auf pH 2,5 eingestellt wurde, extra-. liiert. Die wässrigen Extrakte werden ver einigt, filtriert und. durch Vakriumdestilla- tion auf 1/,1oo bis 1/50o des Brühevolumens eingeengt.
Zu diesem Konzentrat wird 10/0 Calciumchlorid (Gewicht/Volum) zugegeben und der pH-Wert mit Ammoniak auf<B>7,8</B> bis 8,5 eingestellt. Die entstehende Fällung von Ca.lcium-Omegamycin wird abfiltriert und nach Waschen mit Aceton an der Luft. oder im Vakuum getrocknet.
Beispiel <I>21</I> Man kann das Omegamyein aus unreinem Omegamycin auch als Hydrochlorin absehei- den. 1 g Omegamycin als freie Base wird in einem minimalen Volumen wasserfreien Acetons gelöst und die zur Salzbildung er forderliche stöchiomtrische Menge Chlor wasserstoff in die Lösung eingeleitet.
Omega- mycin-h. droehlorid fällt in Form gelber Kri stalle aus und wird abfiltriert. <I>Beispiel 22</I> Omegamycin lässt sich von Verunreini gungen durch Chlorotetraeyclin oder Oxy- tetracyclin"reinigen, in dem man es im Craig- apparat einer Gegenstromverteilung unter- wirft,
wobei n-Butanol und 2,5%ige Essig- säure als Lösungsmittel verwendet werden, siehe z. B.: Weisberger, Technique of Orga.nie Chemistry, Vol. III, Interseience Publ. Co. N.
Y. 1950, Seiten 171-311, und Anal. Chem_ Vol. 24, Seiten 66-70 (1952), und Vol. 23, Seiten 41-44 (1951). <I>Beispiel, 23</I> Zur Verbesserung der Extraktion von Omegamy ein mittels Bu.tanol, Amylalkohol oder Methy 1-isobuty lketon aus Fermenta- tionsbrühen gibt man letzteren kationisehe,
anionische oder nichtionisierbare Netzmittel zu. Der pH-Wert wird hierbei in jedem Fall sorgfältig eingestellt, um maximale Aufnahme im organischen Lösungsmittel zu erzielen, wie dies durch einen einfachen Testversuch fest gestellt werden kann. Geeignete Netzmittel und Konzentrationen sind z.
B. folgende: Di- octylnatrium-sulfosuccinat (1,00/0,), Tween 21 (0,5%) und Natriiuntetradeeylsulfat (0,1%i). Es können Konzentrationen bis 5% angewandt werden.
Andere geeignete Netz mittel sind Laurinsäure, Stearinsäure, Schwe- felsäureester, Naphthalinsidfosäuren oder höhere alkylaromatische Sulfosäuren als anionische Netzmittel; als solche kotionischer Art quaternäre Ammoniumsa,lze [z. B.
(CH3)3N+CH2C 0 NI3-C21-140 OCR/Cl- worin OCR die Pettsäuremischung des Coeos- öls darstellt] ;
als nicht ionisierbare Netzmit tel solche wie Alkyl-phenol-polyglykoläther. In einfacher Weise lässt sich so Omega.- myein aus einer Fermentationsbrühe mit. min destens 100 y Omegamy ein pro ml in Methy 1- isobutylketon überführen. Die Lösung wird abgetrennt und durch Zugabe von Chlor wasserstoffgas Omegamy ein-hy drochlor id in fester Form abgetrennt.
Man kann z. B. nach dieser Methode Omegamycin aus der Fermentbrühe in Me- thy l-isobuty lketon überführen, indem man der Brühe beim PH 2-4 etwa. 0,21/o eines anionischen Netzmittels wie Natriumtetra, decylsulfat, Natriumpeptadeeylsulfat oder Dioctyl-na.triumsulfosuccinat zugibt.
<I>Beispiel</I> 2-1 Das folgende Verfahren stellt eine sehr einfache, billige und wirksame Methode zur Abtrennung rohen, festen Omegamy eins aus grossen Volumina. Fermentationsbrühen dar. Die Brühe, die wenigstens 250 "/ml Omega mycin, vorzugsweise aber 1000 y/ml oder mehr enthält, wird auf einen sauren pH-Wert ein gestellt, worauf das Myeelium abfiltriert wird. Die filtrierte Brühe wird dann auf einen pH-Wert. von 8 oder höher gebracht, indem man z. B. Soda. oder Ammoniak zugibt.
Das rohe Omegamyein fällt in fester Form als freie Base oder als Calciumsa.lz aus, zusam men mit Caleiumphosphaten und andern Ver unreinigungen. Es wird abfiltriert. Das rohe Produkt kann gegebenenfalls auf eine der oben beschriebenen Weisen weiter gereinigt werden.
Zum Beispiel kann man es in Salz- säure vom pH-Wert 2, die 10% ent hält, lösen, hierauf mit Butanol extrahieren, worauf der Butanolextrakt abgetrennt, im Vakuum eingeengt und das Omegamy cin- hy.droehlorid daraus ausgefällt wird.
<I>Beispiel 25</I> Calciumomega.myein wird gereinigt, indem man es mit. Methanol, worin es unlöslich ist, auszieht. Hierauf wird es in Methanol, das Ca.lciumehlorid enthält, aufgelöst. Die un gelösten Verunreinigungen werden abfiltriert, Ammoniak bis zum pH-Wert 8 zugegeben. worauf gereinigtes Calciumomega.myein aus fällt.
<I>Beispiel</I> 26 Das Omegamycin kann auch dadurch (vgl. Beispiel ?1) aus unreinen Produkten als FIy- drochlorid abgeschrieben werden, da.ss man das unreine Produkt in Wasser mit Salzsäure behandelt, bis eine klare Lösung erhalten wird und der pH-Wert unter 3--4 gefallen ist.. Die Lösung wird dann ausgefroren und im Va- kuum getrocknet, wobei ein leicht lösliehes Pulver anfällt.
Das Omega.mycin, hergestellt nach vorlie- grender Erfindung, stellt dieselbe Substanz dar, die heute gemeinhin als Tetracyclin bezeieh- ?iet wird und deren Eigenschaften im Journal of the American Chemical Society, Bd. 75, Seiten 1621-1623, <B>1953,</B> beschrieben wird.

I. Rl=H; R2 =H 1I. R1 = H; R2 = OH III. Rl=Cl; R2 =H Es wurde eine Probe Tetracy clin als freie Base nach Literaturangaben aus Chlorotetra- eyelin hergestellt.
Das erhaltene Produkt schmolz bei 170-175 C und enthielt 0,71% Chlor, woraus auf eine Verunreinigung mit etwa 10 /o nicht umgesetztem Ohlorotetra.cy- clin geschlossen werden kann.
Diese Probe und eine andere chlorfreie Probe von reinem Tetra.cyclin (vom Smp. 169-171 C unter Zersetzung) erwiesen sich im wesentlichen als dieselbe Substanz, wie eine Probe von Omega- mycin (Ansatz 30, hergestellt gemäss Beispiel 14).
Alle darin enthaltenen Verunreinigungen wurden durch Prüfung dieser Proben und Vergleich mit Proben von Chlorotetracyclin und Oxy tetracyclin sowohl in reiner Form als auch in hl.iseht@ng durch Papierstreifenchro- matographie ermittelt (speziell unter Ver wendung der Lösungsmittelsysteme D und L).
Dass Omega.mycin (Ansatz 30) identisch ist mit Tetracyclin lässt sich gleichfalls nach weisen, dadurch, dass beide Substanzen in 48 Stunden bei 37 C in einem Puffer vom pH 8,0 gleiche Aktivitätsverluste von etwa 0,7 mg/ml (= 28%) erleiden;
Chlorotetra- eyclin verlor etwa 50'0/0 seiner Aktivität in 11 Stunden und Oxy tetracyclin 50 % in 26 Stunden unter den gleichen Bedingungen.
Ferner lässt sich Omegamycin von Chloro- tetracyclin und Oxytetracy clin eindeutig durch den Plattentest auf Agar bei pH 8,0 unterscheiden, wobei Chlorotetracy clin und Oxytetracyclin viel weniger aktiv erscheinen als Omegamycin.
Es wurden zu diesem Zweck Versuche auf Agarplatten mit B. subtiles beim pH 6,2 und 8,0 durchgeführt, wobei die oben beschrie benen Puffer als Verdünnungsmittel vom ent sprechenden pH-Wert verwendet wurden.

Omegamycin, hergestellt nach der vor liegenden Erfindung kann in Form der freien Base, sowohl in wasserfreier als auch der<B>hy-</B> dratisierten Form, speziell aber als Trihydrat, zur Bekämpfung von mancherlei menschli chen und tierischen, durch Bakterien verur sachten Infektionen dienen. Besonders wirk sam erweist sich Omega.mycin bei Verabrei chung auf oralem oder intramuskulärem Wege; geeignete Dosen in der Huma.nmedizin liegen zwischen 10 und 1000 mg. Man ver abreicht zweckmässig 1 bis 6 Dosen pro Tag, je nach Art. der Infektion, der Applikation und dergleichen bzw. nach dem Zustande des Patienten.
Für intravenöse Injektion sollten vorzugs weise keine kleineren Konzentrationen als 0,10 0/0 verabfolgt werden, obschon sich auch kleinere Konzentrationen noch als wirksam erwiesen haben. Die Aktivität wächst mit der Konzentration des Omegamycins. Der Pro zentgehalt an aktivem Antibiotikum kann 10 oder 25% betragen oder sogar noch höher sein, z. B. können Tabletten mit einem klei neren Anteil an Füllstoffen und grösserem Anteil an aktiver Substanz bereitet werden.
Es eignen sich insbesondere Tabletten mit<B>10</B> bis 1000 mg Omegamyein.
Die Löslichkeit des Omegamy eins in Wasser ist zwischen den pH-Werten von 5 und 7 ziemlich gering (etwa 0,4 mg/ml). Zu gabe von Calciumchlorid, z. B.1' /0 (Gew./Vol.), erlaubt. es, viel konzentriertere Lösungen herzustellen, z. B. mit einer Löslichkeit von 5 mg/ml bei PH 6 oder im pH-Bereich von 5 bis 7. Solche Lösungen sind hochviskos, ob schon sich die Viskosität innerhalb des Be reiches hoher Löslichkeit durch die p.-Ein- stellung verändern lässt.
Auf diese Weise lassen sich stabile Lösungen oder Suspensio nen mit höheren Gehalten an Omegamyein, wertvoll für pharmazeutische Rezepte leicht herstellen.

The method according to the invention is characterized by the fact that an omegamycin-producing species of Streptomyces or mutation products thereof is submerged in an aqueous nutrient solution containing nutrient nitrogen and carbohydrate for two to three days under aerobic conditions,
