Verfahren zur Herstellung eines Heilmittels gegen Leber-, Gallen-und Darmerkrankungen
Das Hauptpatent betrifft ein Verfahren zur Herstellung eines Heilmittels gegen Leber-, Gallen-und Darmerkrankungen, welches dadurch gekennzeichnet ist, dass Bacillus subtilis var. indolasus unter Verwendung eines Nährmediums gezüchtet wird. Hierbei bildet sich sowohl in dem Nährmedium wie auch im Bacillus selbst als Stoffwechselprodukt ein Wirkstoff, der bei den genannten Krankheiten wirksam ist, dessen Konstitution aber noch nicht geklärt ist.
Obwohl der Bacillus nahezu in allen Nährmedien wächst und auch den Wirkstoff produziert, insbesondere soweit ein günstiger pH-Wert eingehalten wird, hat es sich gezeigt, dass nach dem jeweils verwen- deten Nährmedium bei der Züchtung mitunter Stoffe als Nebenprodukte entstehen, die unerwünschte Nebenreaktionen, wie Kopfsehmerzen, tlbelkeit und dergleichen, hervorrufen.
Es wurde nun gefunden, dass sich derartige Stoffe nicht bilden und diese uner wünschten Reaktionen ausbleiben, wenn man zur Kultur ein eiweissfreies Medium verwen det. Man kann dabei so vorgehen, dass man ein synthetisches, eiweissfreies Nährmedium verwendet ; man kann aber auch an sich brauch- bare, jedoch eiweisshaltige Nährmedien, wie beispielsweise Heuaufgüsse, dadurch verwend- bar machen, dass man sie vor der Beimpfung mit den Bazillen von Eiweiss befreit, was zum Beispiel durch Behandlung mit eiweissfällen- den Mitteln, wie Gerbstoffen, und anschlie ssender Filtration möglich ist.
Jedenfalls muss das Nährmedium mindestens eine Koh lenstoff und mindestens eine Stickstoff liefernde Substanz enthalten.
Das erfindungsgemässe Verfahren zur Herstellung eines Heilmittels gegen Leber-, Gal- len-und Darmerkrankungen nach Patentanspruch des Hauptpatentes ist also dadurch gakennzeichnet, dass ein eiweissfreies, mindestens eine Kohlenstoff und mindestens eine Stickstoff liefernde Substanz enthaltendes Nährmedium verwendet wird. Als Kohlen- stoff liefernde Substanzen sind brauchbar Kohlehydrate, wie Traubenzucker, Fruchtzucker, Mannose, Galaktose, Saccharose, Maltose, Xylose, Arabinose, Stärke und Dextrin ; mehrwertige Alkohole mit mindestens 3 Kohlenstoffatomen, wie Cxlycerin, Mannit, Sorbit, Inosit und zitronensaure Salze.
Als Stickstoff liefernde Substanzen erwiesen sich Ammoniumsalze, Aminosäuren und deren Salze, Salze von Aminoalkoholen, wie Äthanolamin, Diäthanolamin und Dioxypropylamin und Salze von Äthylendiamin, als brauchbar.
Bei Verwendung von Aminosäuren und deren Salze zeigte es sich, dass nach Möglichkeit ein Zusatz in der Grossenordnung von 0, 1 O/o nicht überschritten werden soll, da sonst bisweilen auch die bereits erwähnten und zu Nebenwirkungen Anlass gebenden Stoffe entstehen können.
Wie bereits erwähnt, sind auch andere als synthetiseh aus diesen Substanzen hergestellte Nährmedien brauchbar, und zwar Aufg sse von Heu oder Gräsern, soweit sie von Eiweissstoffen befreit sind. Das Wachstum der Bazillen ist in solehen Nährmedien sehr gut. Obwohl sie bereits Kohlenstoff und Stickstoff liefernde Substanzen enthalten, kann durch Zugabe von den zuvor erwÏhnten, Kohlenstoff und Stickstoff liefernde Substan- zen die Brauchbarkeit solcher Nährlösungen noch verbessert werden. Nährmedien, die ei weissfreien Heuaufguss neben andern NÏhrstoffen enthalten, werden daher vorteilhaft venvendet.
F r das Waehstum und die Wirkstoffbildung erwies sich ein Zusatz von anorganischen Salzen, insbesondere auch von'Schwer- metallen in nicht zu grosser Menge, als wertvoll, wie beispielsweise Natriumchlorid, Ka liumphosphat, Zink-, Eisen-, Kupfer- und Mangansalze. Die Zugabe kleiner Mengen Vitamine und Sexualhormone sowie Purinderivate hat auf die Entwicklung des Bacillus und die Produktion des Wirkstoffes ebenfalls eine vorteilhafte Wirkung.
Der pH-Wert des eiweissfreien Nährmediums wird zweckmϯig zwischen 6, 5 und 8, 5 einge- stellt, wobei ein pH-Wert von 7 und etwas hocher im allgemeinen vorzuziehen ist. Um einen derartigen pH-Bereich nach Möglichkeit bei der Ziichtung aufrechtzuerhalten, ist wiederum die Zugabe von Puffersubstanzen angebracht.
Naeh beendeter Kultur können die Ba zillen wie im Hauptpatent besehrieben, bei spielsweise durch Hitzesterilisation, abgetötet werden. Es kann aber auch ein Konservie- rungsmittel zur Haltbarmachung hinzugefügt werden. Da der Wirkstoff nicht dialysierbar ist, ist es möglich, ihn durch Dialyse anzureiehern und niedermolekulare, unwirksame Substanzen so zu entfernen.
Zweckmϯig ist es, die nach der Züchtung entstandene Knltur, die gegebenenfalls zuvor durch Dialyse angereichert wurde, vom Wasser zu befreien, beispielsweise durch Eindampfen, Gefrier-und Sprühtrocknung.
Hierbei kann, wie im Hauptpatent beschrieben, die gesamte Kultur oder auch das Kul turfiltrat benutzt werden. Zur besseren Do sierung erwies es sich als günstig, den Trokkenrüekstand mit physiologisch indifferenten, pulverförmigen Substanzen zu verreiben, beispielsweise mit Rohrzucker, Milchzucker oder anorganisehen Salzen, wie Kochsalz und dergleichen.
Auch die eventuell abgetrennten und getrockneten Bazillen können gleichfalls mit solchen pulverf¯rmigen Substanzen verrieben werden. Solehe Troekenpräparate erweisen sieh ebenfalls für den Vertrieb als besonders vot - teilhaft und können auch zu Tabletten und Dragées usw, verarbeitet werden.
Beispiel 1
Eine sterilisierte Losung von 5 g primÏrem Kaliumphosphat, 0, 6 g Kochsalz, 0, 1 mg Zinksulfat, 0,8 g Ammonchlorid und 3, 2 g Glukose in einem Liter Wasser, die durch Zugabe von Natriumhydroxydiösung auf einen pH-Wertvon 7, 2 eingestellt war, wurde mit Bacillus subtils var. indolasus beimpft. Die Lösung wurde 7 Tage bei einer Temperatur von 35O bebrütet. Dureh Rühren mit Hilfe eines gut wirksamen Schnellrührers wurde die gebildete Oberfläehenhaut feinst in der Lo sung verteilt (eine derartige Verteilung ist auch in einer Schiittelapparatur unter Zugabe von Glaskugeln oder mit Hilfe von Ultra sehall möglich).
Sodann wurden unter Erwärmen in der Lösung 2 g eines Gemisches gleicher Teile p-Oxy-benzoesäuremethyl-und propylester gelöst. Die so entstandene Lösung kann unmittelbar als Heilmittel verwendet werden. Sie enthält in der gesamten Flüssig- keitsmenge annähernd 1 Billion Keime.
Beispiel 2
Eine sterilisierte und auf einen p-Wert von 6, 6 mit Hilfe von Natronlauge eingestellte Lösung von 0, 8 g Ammoniumsulfat, 0, 6 g Kochsalz, 0,8 g Natriumcitrat, 0, 55 g ZitronensÏure, 0, 5 g primärem Natriumphos- phat t und 4 g Fruktose in einem Liter Was- ser, wird mit Bacillus subtilis var. indolasus beimpft und 61/2 Tage bei einer Temperatur von 316 bis 370 bebrütet. Die entstandene Kultur kann entsprechend Beispiel 1 weiterverarbeitet werden.
Beispiel. 3
Eine sterilisierte und auf einen p-Wert von 7,0geingestellteLosungvon 1, 35 g primärem Kaliumphosphat, 5, 615 g sekundärem \atriumphospha. t, 0, 06 g Magnesiumchlorid, 0, 0005 g Calciumchloridhydrat, 0, 45 g Koehsalz, 0.9 g Alanin und 4 g Saccharose in einem Liter Wasser, wird mit Bacillus sub tilis var. indolasus geimpft. Nach einer Be brütungszeit von 5 Tagen, Homogenisierung der ausgebildeten Oberflächeniaut und Ste rilisierung durch Erhitzen auf Ho , wird die entstandene Kulturlösung im Vakuum zur Trockne eingedampft und der Trockenrückstand mit 22, 5 g Milehzueker verrieben.
Die entstandene Verreibung kann unmittelbar verwendet werden.
Beispiel 4
Eine sterilisierte und auf einen pn-'Wert von 7s5 g eingestellte Lösung von 0, 001 g p Amino-benzoesäure, 0, 05 g Adenin, 0, 05g Cytidin, 0, 05 g Mangansulfat, 0, 85 g Asparagin, 0, 5 g Kaliumehlorid, 1, 35 g primärem Kaliumphosphat, 5, 65 g sekundärem Natriumphosphat und 4 g Maltose wird mit Bacillus subtilis var. indolasus geimpft und 41/2Tage bei einer Temperatur von 37, 5 bis 3$ bebrütet. Die entstandene Kulturlosung wird durch ein Bakterienfilter filtriert, das Filtrat zur Trockne eingedampft und mit Milchzucker verrieben.
Die abgetrennten Bakterien wer clen ebenfalls getrocknet und mit Milchzucker verrieben. Beide Milchzuckerverreibungen kön- nen therapeutisch verwendet werden.
Beispiel 5
In einer entsprechend Beispiel 3 hergestellten Nährlosung werden 30 g Agar gelât.
Der sterilisierte Kulturboden wird mit Baeillus subtils var. indolasus geimpft und in Sehalen 71/2 Tage bei einer Temperatur von 38 bebrütet. Die entstandene Oberflächen- haut wird von den Agar-Platten entfernt, getrocknet, die Bakterien durch Erhitzen abget¯tet und sodann mit Rohrzucker verrieben.
Beispiel G 85 g Asparagin, 0, 5 g Aneurin, 0, 5 g Lacto- flavin, 0, 5 g Diäthyldioxystilbenpropionat, 5 g Ferrosulfat, 0, 1 g Kupfersulfat, 5 g Kochsalz, 55 g ZitronensÏure, 430 g sekundärem Natriumphosphat und 400 g Fruktose werden in 100 Liter destilliertem Wasser gelost.
Nach Zugabe von 5 g eines Schaumverhü- tungsmittels (Entsehäumungsmittel Marke E 100 konz. ? von Bayer) wird die Losung durch Erhitzen sterilisiert und nach dem Erkalten mit Bacillus subtils var. indolasus ge impft. Die Lösung wird sodann auf einer Temperatur von etwa 36 bis 37 gehalten und während 4 Tagen unter Einblasen von steriler Luft in das untere Ende des Behälters bebrütet. Sodann wird die entstandene Losung durch Erhitzen sterilisiert und nach Zugabe einer Mischung von gleichen Teilen Kochsalz und Milchzucker auf einem Walzentrockner getrocknet.

It has now been found that such substances do not form and these undesirable reactions do not occur if a protein-free medium is used for culture. One can proceed in such a way that one uses a synthetic, protein-free nutrient medium; But you can also make nutrient media that are useful, but containing protein, such as hay infusions, usable by removing protein from them before inoculating them with the bacilli, for example by treating them with protein-precipitating agents such as tannins , and subsequent filtration is possible.
In any case, the nutrient medium must contain at least one carbon and at least one nitrogen-supplying substance.
The method according to the invention for producing a remedy for liver, gallbladder and intestinal diseases according to claim of the main patent is thus characterized in that a protein-free nutrient medium containing at least one carbon and at least one nitrogen-producing substance is used. Carbohydrates, such as grape sugar, fructose, mannose, galactose, sucrose, maltose, xylose, arabinose, starch and dextrin, can be used as carbon-supplying substances; polyhydric alcohols with at least 3 carbon atoms, such as glycerol, mannitol, sorbitol, inositol and citric acid salts.
Ammonium salts, amino acids and their salts, salts of amino alcohols such as ethanolamine, diethanolamine and dioxypropylamine and salts of ethylenediamine have proven to be useful as nitrogen-supplying substances.
When using amino acids and their salts, it was found that, if possible, an addition in the order of 0.1 O / o should not be exceeded, since otherwise the substances already mentioned and giving rise to side effects can sometimes arise.
As already mentioned, nutrient media other than synthetically produced from these substances can also be used, namely infusions of hay or grass, provided they are freed from protein substances. The growth of the bacilli is very good in such culture media. Although they already contain carbon and nitrogen supplying substances, the usefulness of such nutrient solutions can be further improved by adding the carbon and nitrogen supplying substances mentioned above. Nutrient media that contain egg white-free hay infusion in addition to other nutrients are therefore advantageously used.
For growth and the formation of active ingredients, the addition of inorganic salts, in particular also of heavy metals in not too large quantities, has proven to be valuable, such as sodium chloride, potassium phosphate, zinc, iron, copper and manganese salts. The addition of small amounts of vitamins and sex hormones as well as purine derivatives also has a beneficial effect on the development of Bacillus and the production of the active ingredient.
The pH value of the protein-free nutrient medium is expediently set between 6.5 and 8.5, with a pH value of 7 and slightly higher generally being preferable. In order to maintain such a pH range during breeding as far as possible, the addition of buffer substances is again advisable.
After the culture has ended, the ba cillas can be killed as described in the main patent, for example by heat sterilization. However, a preservative can also be added for preservation purposes. Since the active substance cannot be dialyzed, it is possible to concentrate it by dialysis and thus to remove low-molecular, ineffective substances.
It is expedient to remove the water from the culture produced after cultivation, which may have been enriched beforehand by dialysis, for example by evaporation, freeze-drying and spray-drying.
Here, as described in the main patent, the entire culture or the culture filtrate can be used. For better dosing, it has proven to be beneficial to rub the dry residue with physiologically inert, powdery substances, for example with cane sugar, lactose or inorganic salts such as table salt and the like.
The bacilli, which may have been separated off and dried, can also be rubbed with such powdery substances. Sole dried preparations also prove to be particularly beneficial for distribution and can also be processed into tablets and dragees, etc.
example 1
A sterilized solution of 5 g of primary potassium phosphate, 0.6 g of common salt, 0.1 mg of zinc sulfate, 0.8 g of ammonium chloride and 3.2 g of glucose in one liter of water, which can be brought to a pH of 7.2 by adding sodium hydroxide solution was inoculated with Bacillus subtils var. indolasus. The solution was incubated for 7 days at a temperature of 35O. The surface skin formed was finely distributed in the solution by stirring with the aid of a highly effective high-speed stirrer (such a distribution is also possible in a Schiittel apparatus with the addition of glass balls or with the aid of ultrasound).
2 g of a mixture of equal parts of methyl p-oxy-benzoate and propyl ester were then dissolved in the solution with heating. The resulting solution can be used immediately as a remedy. It contains almost 1 billion germs in the total amount of liquid.
Example 2
A sterilized solution of 0.8 g ammonium sulfate, 0.6 g common salt, 0.8 g sodium citrate, 0.55 g citric acid, 0.5 g primary sodium phosphate, adjusted to a p-value of 6.6 with the help of sodium hydroxide phat t and 4 g of fructose in one liter of water, is inoculated with Bacillus subtilis var. indolasus and incubated for 61/2 days at a temperature of 316 to 370. The resulting culture can be processed further as in Example 1.
Example. 3
A sterilized solution adjusted to a p-value of 7.0 of 1.35 g primary potassium phosphate, 5.615 g secondary \ atrium phosphate. 0.06 g magnesium chloride, 0.005 g calcium chloride hydrate, 0.45 g coal salt, 0.9 g alanine and 4 g sucrose in one liter of water is inoculated with Bacillus sub tilis var. indolasus. After an incubation time of 5 days, homogenization of the surface skin formed and sterilization by heating to Ho, the resulting culture solution is evaporated to dryness in vacuo and the dry residue is triturated with 22.5 g milk sugar.
The resulting trituration can be used immediately.
Example 4
A sterilized solution of 0.001 gp amino-benzoic acid, 0.05 g adenine, 0.05 g cytidine, 0.05 g manganese sulfate, 0.85 g asparagine, 0.5 g, adjusted to a pn-value of 7.5 g Potassium chloride, 1.35 g of primary potassium phosphate, 5.65 g of secondary sodium phosphate and 4 g of maltose are inoculated with Bacillus subtilis var. Indolasus and incubated for 41/2 days at a temperature of 37.5 to 3%. The resulting culture solution is filtered through a bacterial filter, the filtrate is evaporated to dryness and triturated with lactose.
The separated bacteria are also dried and rubbed with milk sugar. Both lactose triturations can be used therapeutically.
Example 5
In a nutrient solution prepared according to Example 3, 30 g of agar are gelatinized.
The sterilized culture soil is inoculated with Baeillus subtils var. Indolasus and incubated in Sehalen for 71/2 days at a temperature of 38. The surface skin formed is removed from the agar plates, dried, the bacteria killed by heating and then rubbed with cane sugar.
Example G 85 g of asparagine, 0.5 g of aneurine, 0.5 g of lactoflavin, 0.5 g of diethyldioxystilbene propionate, 5 g of ferrous sulfate, 0.1 g of copper sulfate, 5 g of table salt, 55 g of citric acid, 430 g of secondary sodium phosphate and 400 g of fructose are dissolved in 100 liters of distilled water.
After adding 5 g of an anti-foaming agent (defoaming agent brand E 100 conc.? From Bayer), the solution is sterilized by heating and, after cooling, inoculated with Bacillus subtils var. Indolasus. The solution is then maintained at a temperature of about 36 to 37 and incubated for 4 days while blowing sterile air into the lower end of the container. The resulting solution is then sterilized by heating and, after adding a mixture of equal parts of table salt and lactose, dried on a drum dryer.
