Verfahren und Einrichtung zur Züchtung von Hefe. Gegenstand des Hauptpatentes ist ein Ver fahren zur Züchtung von Hefe unter Belüf tung durch aufsteigende Luftbläschen, welches dadurch gekennzeichnet ist, dass die hefehal- tige Maische abwärts durch den Bottich be wegt, unten dem Bottich entnommen und von oben her wieder zugeführt wird.
Es hat sich nun gezeigt., da.ss es vorteilhaft ist, wenn die Zuwachszeit der Hefe im Ver- hefungsbottich verlängert wird und im Hin blick auf die Ausnutzung der Nährstoffe von verschiedenen Assimilationszeiten geregelt wer den kann; die unter Verwendung eines ein zigen, nicht unterteilten Bottichs erzielbare Zuwachszeit reicht in der Regel für die Ver wertung schwerer assimilierbarer Nährstoffe nicht aus.
Das den Gegenstand des vorliegenden Pa tentes bildende Verfahren ermöglicht nun, die Umlaufzeit der Maische und den Hefezuwachs in Berücksichtigung der verschiedenen Assi milationszeiten der in der Nährlösung enthal tenen Nährstoffe in einfacher Weise zu regeln.
Das erfindungsgemässe Verfahren zur Züchtung von Hefe nach dem Patentanspruch I des Hauptpatentes ist dadurch gekennzeich net, dass ein senkrecht in mehrere Abteile un terteilter Verhefungsbottich verwendet wird und da.ss die Maische getrennt aus jedem Ab teil in der Nähe seines Bodens entnommen, ausserhalb des Bottichs unter Druck gesetzt, während des Hochdrückens abgekühlt.
und un ter Zusatz von von Nährlösung von oben in einen Verteiler geleitet wird, aus dem das Maische- Nährlösungsgemisch, in Strahlen zerteilt, auf den bei der Verhefung entstehenden Schaum zur Einwirkung gebracht wird, wobei die Maische aus dem einen Abteil in das nächste strömt und aus dem letzten Abteil die dem sekundlichen Hefezuwachs entsprechende Menge der verheften Maische abgezogen wird.
Soll bei einem solchen Hefezüchtungsver- fahren der Hefezuwachs kontinuierlich ent nommen werden, so ist. bei konstant bleiben der Bottichfüllung der hefehaltige Maisehe abfluss gleich der kontinuierlich zugeführten Nährlösungsmenge zu halten, während der günstigste Nährstoffgehalt der Lösung von dem durch die Umlaufzeit des Gesamtumlau fes bestimmten Hefegehalt der Maische und der erzielbaren Ausbeute abhängt.
Die Zufuhr der Nährlösung kann so erfolgen, dass jedem Bottichabschnitt unabhängig von der Grösse seines Inhaltes die gleichen Nährlösungsmen- gen mit gleichem Nährstoffgehalt zugeführt werden, so dass der erste Abschnitt Nährstoffe im Überschuss und der letzte Verhefungs- abschnitt relativ weniger Nährstoffe erhält.
Anderseits können auch die Phosphat- und Stickstoffgehalte der den einzelnen Abteilen zugeführten Nährlösungsmengen so eingestellt werden, dass sie von Abteil zu Abteil abneh men, während die. Zuckergehalte zum Beispiel gleich bleiben oder zunehmen können. Soll der Hefegehalt der Maische im Bottich gleich blei ben, so muss der Inhalt der. einzelneu- Albteile des Verhefungsraiunes des Bottichs im Verhält- nis des jeweiligen Hefezuwachses zunehmen, so dass dann die Durehlaufzeit der Maische für jedes Abteil gleich oder annähernd gleich bleibt.
Dem Verhefimgsraum schliesst. sich zweckmässig ein Ausreifeabschnitt an, dessen Inhalt der der Aasreifezeit der Hefe ange- passten Durchlaufzeit entsprechend zu wählen ist. Die Umlaufzeit der Maische im Gesamt umlauf wird am besten so gewählt, da.ss eine vollständige Ausnutzung der Nährstoffe ge- währleistet ist.
Der Umlauf kann durch eine in die Umlaufleitung eingebaute Pumpe ge regelt werden, während die Einstellung der jeweiligen Füllung des Verhefungs- und Gär bottichs durch eine Höhenverstellung des Maischeabflusses erfolgen kann, wodurch mit der Regelung der Umlaufzeit eine genaue An passung des Hefegehaltes der Maische an die Nährstoffzufuhr ermöglicht wird.
In einen zusätzlichen Gärraum, in welchen die aus dem Ausreifeabschnitt abfliessende Maische geleitet wird, kann bei Alkoholgewin nung eine entsprechende Zuckerlösung zuge führt und ausserhalb des Gesamtkreislaufes vergoren werden,- wobei die Maische zwecks Abfuhr der anfallenden Wärmemengen hier ebenfalls für sich in Umlauf versetzt werden kann. Der Inhalt. des Gärraumes ist dabei durch die erforderliche Vergärungszeit und den Alkoholgehalt der durch die zugeführte Zuckerlösung vermehrten Maischemenge ge geben.
Diese Einrichtung dient zur Trennung der fermentativen von der vegetativen Tätig keit der Hefe und vermindert die zeitliche energetische Belastung derselben sowie den zeitlichen Wärmeanfall. Eine beispielsweise Ausführungsform des erfindungsgemässen Verfahrens sei im folgen den an Hand der Zeichnung näher erläutert:
Fig. 1 zeigt das Schema einer Verhefungs- anlage mit Doppelkreislauf, einer Anlage, bei der die Maische abteilweise getrennt im Kreis lauf II entgegen den aufsteigenden Luftbläs chen geführt wird und die einzelnen Abteile zu einem gemeinsamen Umlauf I hintereinan- dergeschaltet sind, Fig. 2 zeigt im Schnitt nach der Linie x-x in Fig. 1 die Einrichtung zur Durchführung des Gegenstromverfahrens in einem Abteil im so Querschnitt.
Fig. 3 zeigt, in einem Diagramm, wie sich das Hefequantum Q;" in einer CTenerationszeit vermehrt.
In Fig. 1 sind die Inhalte der Einzel abschnitte im Verhefungsraum dieser Vermeh rung proportional vergrössert.
Gemäss Fig.2 ist. in die Umlaufleitung a eine Regelpumpe b eingeschaltet. Am obern Ende des Abteils c befindet sich ein Verteiler d, welcher die vom Boden des Bottichs abge pumpte, luftfreie, in ihn eventuell unter Überdruck eingepumpte Maische in Flüssig keitsstrahlen zerteilt. Diese Flüssigkeitsstrah len zerstören den durch die Belüftung der Maische entstehenden Schaum. Die Umlauf leitung ca führt die Maische durch einen Küh ler e, welcher die bei der Verhefung konti nuierlich anfallenden Wärmemengen abführt.
Zur Aufrechterhaltung einer bestimmten Schaumdeckendicke <I>SH'</I> ist im Bottich c über der Maischeoberfläche ein Sieb f vorgesehen, durch dessen Löcher der Schaum tritt, um von den Maischestrahlen zerstört zu werden. Eine Belüftungseinrichtung g sorgt für die Belüf tung der Maische; mit. L ist. die Zuleitung der sterilen Luft, mit L' die Ableitung für die Ab luft bezeichnet. Die Nährlösung<I>NL</I> wird mit der 'Maische in der Umlaufleitung a vermischt und mit dieser gleichmässig über die Ober fläche der Maische verteilt.
Mit FH ist die Füllhöhe der Maische im Gärbottich, mit SH die Steighöhe der Maische infolge Belüftung bezeichnet. Eine besondere Entlüftung der Maische ist. bei Anwendung des Gegenstrom verfahrens nicht notwendig, da die Luftbläs chen bei diesem Verfahren selbsttätig aus der ihnen entgegenströmenden Maische austreten.

Fig.1 zeigt. mehrere derartige Gegenstrom abteile in einem Gesamtkreislauf hintereinan- dergeschaltet. Der Bottich ist in einen aus drei Teilräumen i1, <I>i2, i3</I> bestehenden Verhe- fungsraum VH und einen Ausreiferaum AR unterteilt.
Jedes der Abteile i1 bis i3 und AR besitzt seinen eigenen Gegenstromumlauf mit den Pumpen b1 bis b4 und den Verteilern d1 bis d4 zur Schaumzerstörung, daneben je einen Kühler e1 bis e4 und eine seiner Grösse ent sprechende Belüftungsapparatur g. Die Füll höhe FH, die durch die Belüftung bedingten Steighöhen<I>SH</I> und die zugelassenen Schaum deckendicken SH' sollen in allen Abteilen gleich sein; die Abteile enthalten Siebe f (Fug. 2), deren Höhe über dem Maischespiegel einstellbar ist.
Regulierventile RV dienen zur Regulierung der zugeführten Nährlösungs- mengen <B>NL,</B> bis NL3 und der durch die Lei tungen L für sterile Luft. den Einzelabschnit ten zugeführten Luftmengen. Der Gesamt umlauf<B>1</B> wird durch eine Pumpe b5 hervor- gerufen und reguliert, während eine zusätz liche Regulierung des Maischeinhaltes durch Änderung der Füllhöhe<I>FH</I> im Bottich mit tels einer Höhenverstellung des Ausflusses der Entnahmeleitung h erfolgen kann.
Wenn die Zufuhr der Nährlösung und die Belüftungsstärke konstant bleiben sollen, ist es notwendig, dass der Hefegehalt der Maische im Bottich gleichbleibt und die Inhalte der Teilabschnitte entsprechend dem aus dem Diagramm der Fig. 3 zu entnehmenden Ver hältnisse bei gleicher Füllhöhe zueinander ver grössert werden.
Die Durchführung des Verfahrens unter Dauerbetrieb geschieht mit der Einrichtung etwa wie folgt Die Inbetriebsetzung der Anlage erfolgt zunächst unter der Verdopplung der halben, dem Bottich zugesetzten Stellhefemenge auf die gewählte Hefekonzentration, zum Beispiel von 50 auf 100 kg pro Kubikmeter Maische. Ist diese Konzentration erreicht, so erfolgt die Entnahme des weiteren Zuwachses kontinuier lich, wobei die Nährlösungsmenge der ablau fenden Maischemenge entspricht, mit der der sekundliche Hefezuwachs abgeführt wird, wor aus sich die Konzentration der Melasse und der Nährsalze in der Nährlösung unter Be rücksichtigung der erreichbaren Hefeausbeute ergibt.
Sind im Bottich zum Beispiel 1000 kg Hefe mit 25 % Trockengehalt enthalten, so er- rechnet sich der durch das Rohr h sekundlich zu entnehmende Hefezuwachs q, und damit die durch das Rohr h zu entnehmendeMaische- menge bzw. die zuzuführende Nährlösungs- menge nach folgender Gleichung: .

Dieser Maischemenge entspricht dann die zu zuführende Nährlösungsmenge, Soll der Hefegehalt imBottichraum Bleich bleiben, so wachsen die Inhalte der drei Ab teile des Verhefungsraumes im Verhältnis von l,26 :1,262 :1,263;
das erste Bottichabteil ent- hält 26 % des gesamten Hefequantums der Gesamtmaische, das zweite 32,8 und das dritte 41,2 %. Obwohl die Hefemenge in den ein- zelnen Abteilen verschieden sind, ist es zweck mässig,
jedem Abteil die gleiche Nährlösungs- menge NLl, NL2, NL3 mit gleichem Nähr stoffgehalt zuzuführen, wodurch das erreicht. wird, dass ein bestimmter Teil der schwerer assimilierbaren Nährsalze eine längere Ver- weilzeit im Bottich erhält, so da.ss also die ge samten Nährstoffe der Nährlösung in der Um laufzeit vollständig assimiliert werden können.
Sollte dies in besonderen Fällen, zum Beispiel wenn man die Umlaufzeit verkürzen will, nicht ausreichen, so kann die Konzentration der den Einzelabschnitten zugeführten Nährlösungs- menge NLl bis NL3 verschieden gestaltet wer den, zum Beispiel so, dass der Phosphat- und Stickstoffgehalt von Abteil zu Abteil abnimmt und dafür der Zuckergehalt gleichbleibt, oder, wenn erforderlich, zunimmt, während die Nährlösungsmengen für jeden Abschnitt untereinander gleichbleibt.
Falls im Anschluss an die Hefezüchtung noch eine Alkoholgewinnung angestrebt wird, so kann ein zusätzlicher Gärbottich an den Ausreifeabsehnitt AR angeschlossen werden, in den die verhefte Maisehe mit voll ausge reiften Hefezellen aus dem Ausreifeabschnitt eingeleitet wird, um unter Zufuhr einer ent sprechenden Zuckerlösung ausserhalb des Ge samtkreislaufes I vergoren zu werden, wobei die Maische in diesem zusätzlichen Gärbottich zwecks Abfuhr der durch die Vergärung an fallenden Wärmemenge für sich im Gegen strom durch einen Kühler gepumpt werden kann;
der Maischeinhalt dieses Gärbottichs ist durch die gewünschte Alkoholausbeute und durch die Gärungsgeschwindigkeit ihres Hefe inhaltes bestimmt.
