Verfahren zur Herstellung von Vitaminsubstanzen. Gegenstand der Erfindung ist ein Verfah ren zur Herstellung sowohl therapeutisch als für die Ernährung wertvoller Substanzen durch einen mikrobiologischen Prozess, und zwar ein Verfahren zur Herstellung von Vi taminsubstanzen mit wachstumsfördernden <B>E</B>igenschaften für den Mikroorganismus Lacto- baeillus laetis Dorner, wie das Vitamin B12. Dieses Vitamin ist ein wasserlöslicher roter kristalliner Stoff, welcher aus Leber gewon nen werden kann und bei der Behandlung gewisser Arten Anämie beim Menschen, z. B. der pernieiösen Anämle, wertvoll ist.
Ausser dem ist Vitamin B12 als Nahrungsfaktor für Menschen und Tiere wichtig.
Das erfindungsgemässe Verfahren zur Her stellung von Vitaminsubstanzen ist dadurch gekennzeichnet, dass man in einem Nähr medium, welchem Kobalt zugesetzt wurde, einen zu den Pilzen gehörenden Mikroorganis mus züehtet, welcher eine Substanz mit Vi- tamin-B,2-Wirkung erzeugt.
Substanzen mit Vitamin-B12-Wirkung haben die Eigenschaft, das Wachstum des Lactobaeillus laetis Dorner zu fördern. Die Aktivität der nach dem erfindungsgemässen Verfahren gewonnenen Vitaminsubstanzen, z. B. Vitamin B12, kann durch eine mikrobio logische Methode bestimmt werden, wobei der Lactobaeillus laetis Dorner (LLD) als Test organismus verwendet wird.
Die Versuchsbe- dingungen sind von Short <B>(J.</B> Biol. Chem. <B>169,</B> <B>455-6)</B> angegeben worden. Die Wirksamkeit dieser Vitaminsubstanzen, der mit ihnen an- gereieherten Brühen und Konzentrate, wird zweckmässig in LLD-Einheiten ausgedrüekt, und zwar auf der Basis eines willkürlich ge wählten Leber-Standards, welcher eine Aktivi tät von<B>1000</B> LLD-Einh./mg hat.
Es wurde festgestellt, dass reines kristallisiertes Vita min B 12 eine Wirksamkeit aufweist von etwa <B>1.1</B> Millionen LLD-Einh./mg. Anderseits ist die Wirksamkeit von Brühen, welche durch die Vergärung gewöhnlicher Nährmedien durch die fraglichen Organismen erhalten werden, im Vergleich zum reinen Vitamin B12 äusserst gering. Je nach der Art des verwen deten Pilzes besitzen sie eine LLD-Wirksam- keit, die einem Vitamin-B,2-Gehalt in der Grössenordnung von etwa<B>0,00003</B> mg/ein3 ent spricht.
Es ist schwierig, reines Vitamin BI2 oder Konzentrate reich an LLD-Wirksamkeit aus solchen Brühen zu gewinnen.
Es hat sieh gezeigt, dass beim Kultivieren der Pilze in einem Nährmedium mit einem Zusatz von Kobalt erheblich grössere Mengen von LLD-wirksamen Substanzen durch den Mikroorganismus erzeugt werden, so dass die Gewinnung von z. B. reinem Vitamin B12 auts der Gärbrühe erleichtert wird und es möglich ist, die festen Bestandteile der Brühe direkt als Zusatzhahrung für Tiere zu verwenden. Die Wirkung ist überraschend, weil Kobalt als sehr starkes Gift für viele Mikroorganis men bekannt ist.
Versuche haben gezeigt, dass in höheren Konzentrationen Kobalt auch toxisch wirkt auf diejenigen Pilzstämme, welche Substanzen mit LLD-Wirksamkeit er zeugen. Beispielsweise verhindert ein Kobalt- zusatz (in Form des Nitrats) von<B>10</B> bis 20 Teilen pro Million zum Nährmedium die Herstellung von LLD-wirksamen Substanzen durch den Streptomyces griseLis.
Die während der Gärung gegenwärtige Ko- baltmenge kann<B>je</B> nach der Art des Nähr mediums variieren, doch ist es gewöhnlich zweckmässig, Kobalt in einer Menge zwischen etwa<B>0,1</B> und 20 Teilen pro Million Teile Nährmedium zu verwenden. In manchen Fäl len kann es vorteilhaft sein,<B>je</B> nach der Gil- tigkeit von Kobalt für die verwendete Pilzart grössere oder kleinere Mengen Kobalt zu ver wenden.
Bei Verwendung von Streptomyces griseus wurden Gärbrühen von hoher LLD- Wirksamkeit und ausgezeichnete Ausbeuten von kristallisiertem Vitamin Bl? erhalten durch Verwendung von etwa 2 Teilen Kobalt in Form von Kobaltnitrat pro Million Teile Nährmedium.
Das Kobalt kann in metallischer Form der Brühe zugesetzt werden, doch wird zweck mässig eine Kobaltverbindung, vorzugsweise ein Kobaltsalz, z. B. Kobaltnitrat, zugesetzt. Es kann aber auch das Kobalt in der Form eines in der Natur vorkommenden Kobaltsalzes oder einer Verbindung, wie sie in kobalt- reichen mikrobischen Nahrungsstoffen vor kommen, verwendet werden, jedoch sind letz tere schwierig zu beschaffen.
Die Nährmedien können im übrigen die beim Kultivieren von Pilzen übliche Zusam mensetzung aufweisen. Für bestimmte Pilze variiert allerdings die Ausbeute an LLD-wirk- samen Stoff en<B>j</B>e nach dem verwendeten Nähr medium, doch wurde stets gefunden, dass ein an Kobalt armes Medium durch Zugabe einer kleinen Menge Kobalt LLD-wirksame Substan zen in stark erhöhter Ausbeute lieferte.
Die übliehen Nährmedien enthalten eine Quelle von assimilierbarem Kohlenstoff, eine Quelle von assimilierbarem Stickstoff, anor- ganisehe Salze und nötigenfalls auch Wachs tumsfaktoren. Die Kohlenstoffquelle kann ein Kohlehydrat, z. B. Destrose, Maltose, Xylose, Invertzucker, ein Getreideauszug und derglei- ehen sein.
Als Stiekstoffquelle kommen in Betracht: Ammonitimsalze, Aminosäuren oder Proteine, z. B. Sojabohnen, Hafer, Hefe, Hefe extrakte, ferner Tryptinabbauprodukte von Casein, Fleisehextrakt, Blutinehl, proteinreiche Fleisch- und Knochenabfälle, Salmmehl, Fiseh- mehl, Fisehextrakte, lösliche Bestandteile voll Schlempe und dergleichen.
Die Pilze können auch in Medien ohne Kohlehydrate kultiviert werden, in welchem Falle Proteine oder Aminosäuren sowohl den benötigten Kohlen stoff als auch den Stickstoff liefern.
Zur Durchführung des erfindLingsgemässen Verfahrens können Myxomyeeten, Sehizo- myceten und Eumyeeten verwendet werden. Besonders geeignet sind Sehizomyeeten, vor zugsweise gewisse Stämme der Spezies Strep- tomyces griseus, welche auch zur Erzeugung der Antibiotiea Streptomvein und Grisein dienen können.
Andere Spezies wie Strepto- myces albidoflavus, Strept. eolombiensis nov. sp., Strept. roseochromogenus und Strept. anti- biotieus enthalten ebenfalls Stämme, die LLD- wirksame Substanzen in hoher Ausbeute er zeugen.
Weitere verwendbare Sehizomveeten sind Glieder des Genus Clostridium <B>'</B> wie Clo- stridium tetanomorphum, <B>Cl.</B> mehlearium, <B>Cl.</B> flabelliferum und<B>Cl.</B> butyricium. Andere Pilze, welche beim erfindungsgemässen Verfah ren Substanzen mit LLD-Wirksamkeit liefern, sind Torula,
Eremotheeium ashbyii und Esche- richia coll. Das Nährmedium wird zweckmässig sterili siert und das sterile Medium mit dem ge- wünsehten Kobaltzusatz sodann mit einer Kul tur des gewählten Pilzes geimpft, worauf die Brühe bis zur optimalen LLD-Wirksamkeit <B>9</B> entwickelt wird. Die Gärzeit beträgt vorteil haft etwa 2 bis<B>7</B> Tage.
Die Entwicklung er folgt zweckmässig unter der Oberfläche des Mediums und bei einer für den betreffenden Pilz geeigneten Temperatur.<B>9</B> Um kristallisiertes Vitamin B12 aus der Gärbrühe zu isolieren, kann folgendermassen vorgegangen werden. Die Brühe wird filtriert und das Filtrat mit Aktivkohle behandelt, wo bei das Vitamin B 1.2 adsorbiert wird. Das Adsorbat wird sodann mit einer wässrigen Lösung von Pyridin oder von a-Pieolin el-Liiert und das Eluat zur Trockne eingedampft.
Der Rückstand wird mit einem niedri-siedenden aliphatisehen Alkohol, z. B. Methanol, extra hiert und der gewonnene Extrakt durch eine Kolonne mit aktiver Tonerde abgezogen, wobei das Vitamin Bl., an die Tonerde adsorbiert wird. Die Kolonne wird sodann mit frischem Methanol eluiert. Die Eluate mit LLD-Wirk- samkeit werden gesammelt und eingeengt. Die erhaltene konzentrierte methanolische Lösung wird dann mit einer Flüssigkeit versetzt, welche mit der Lösung misehbar ist, aber in der die aktive Substanz unlöslich ist, z.
B. Aceton. Der Niederschlag wird abfiltriert, in Alkohol gelöst, durch Zusatz von Aeeton aus gefällt lind das gereinigte Produkt durch Umkristallisierung aus Wasser, unter Zusatz von Aeeton, in reines kristallisiertes Vita min B12 übergeführt.
<I>Beispiel<B>1:</B></I> Es wurden Nährmedien, bestehend aus einer Suspension von 2% Troekenhefe in dest. Wasser hergestellt und mit variierenden Mengen von Kobaltnitrat versehen, wie in der nachfolgenden Tabelle angegeben. Je<B>50</B> em3 der Nährbrühen wurden in 250-em3-Erlen- meyer-Kolben gegeben und während einer hal ben Stunde bei 12011 <B>C</B> unter ständiger Bewe gung sterilisiert.
Sodann wurde mit etwa <B>1</B> em3 einer 48stündigen vegetativen Kultur eines Grisein erzeugenden Stammes von Streptomyces griseus geimpft. Dieser Stamm war auf einem Gemisch von Fleisehextrakt und einem Tryptinabbauprodukt von Casein gezüchtet worden. Die geimpften Brühen wur den 4 Tage bei 281' <B>C</B> entwickelt, während wel- eher Zeit die Flaschen ständig auf einer Schüttelmasehine -esehüttelt wurden. Nach Beendigung der Gärperiode war die Wirksam keit der Brühen wie folgt-.

Die konzentrierte LÖsung wurde mit Aeeton ver setzt, wobei rohes Vitamin B12 ausgefällt wurde. Das Rohprodukt wurde durch Umfäl- lung aus einer Äthanollösung mit Aceton ge reinigt. Das so erhaltene gereinigte Produkt wurde aus -%vässrigem Aeeton unikristallisiert und so kristallisiertes VitaminB12 erhalten.

The subject of the invention is a method for the production of both therapeutically and nutritionally valuable substances by a microbiological process, namely a method for the production of vitamin substances with growth-promoting properties for the microorganism Lactobaeillus laetis Dorner like the vitamin B12. The method according to the invention for the production of vitamin substances is characterized in that a microorganism belonging to the fungi is grown in a nutrient medium to which cobalt has been added and which produces a substance with vitamin B, 2 action.
Substances with vitamin B12 effect have the property of promoting the growth of the Lactobaeillus laetis Dorner. The activity of the vitamin substances obtained by the process according to the invention, e.g. B. vitamin B12, can be determined by a microbiological method using the Lactobaeillus laetis Dorner (LLD) as a test organism.
The test conditions have been given by Short (J. Biol. Chem. 169, 455-6). The effectiveness of these vitamin substances, the broths and concentrates enriched with them, is expediently expressed in LLD units on the basis of an arbitrarily chosen liver standard, which has an activity of <B> 1000 </B> LLD -Unit / mg has.
It was found that pure crystallized Vita min B 12 has an effectiveness of about 1.1 million LLD units / mg. On the other hand, the effectiveness of broths, which are obtained through the fermentation of ordinary nutrient media by the organisms in question, is extremely low compared to pure vitamin B12. Tests have shown that in higher concentrations cobalt also has a toxic effect on those fungal strains which produce substances with LLD activity. For example, adding cobalt (in the form of nitrate) of <B> 10 </B> to 20 parts per million to the nutrient medium prevents the production of LLD-active substances by Streptomyces griseLis.
The amount of cobalt present during fermentation can vary depending on the type of nutrient medium, but it is usually expedient to use cobalt in an amount between about 0.1 and 20 parts per million parts of culture medium to be used. In some cases it can be advantageous to use larger or smaller amounts of cobalt depending on the validity of cobalt for the type of fungus used.
Using Streptomyces griseus, fermentation broths of high LLD effectiveness and excellent yields of crystallized vitamin Bl? obtained by using about 2 parts of cobalt in the form of cobalt nitrate per million parts of nutrient medium.
The cobalt can be added to the broth in metallic form, but a cobalt compound, preferably a cobalt salt, e.g. B. cobalt nitrate added. However, cobalt can also be used in the form of a naturally occurring cobalt salt or a compound such as those found in cobalt-rich microbial nutrients, but the latter are difficult to obtain.
The nutrient media can also have the usual composition when cultivating mushrooms. For certain fungi, however, the yield of LLD-active substances varies according to the nutrient medium used, but it has always been found that a medium poor in cobalt can be LLD-effective by adding a small amount of cobalt Substan zen delivered in greatly increased yield.
The usual nutrient media contain a source of assimilable carbon, a source of assimilable nitrogen, inorganic salts and, if necessary, growth factors. The carbon source can be a carbohydrate, e.g. B. Destrose, maltose, xylose, invert sugar, a grain extract and so on.
Possible sources of nitrogen are: ammonite salts, amino acids or proteins, e.g. B. soybeans, oats, yeast, yeast extracts, also tryptin breakdown products of casein, meat extract, blood, protein-rich meat and bone waste, salmon meal, fish meal, fish extract, soluble components full of vinasse and the like.
The fungi can also be cultivated in media without carbohydrates, in which case proteins or amino acids provide both the carbon and nitrogen required.
To carry out the method according to the invention, myxomyeetes, sehizomycetes and eumyeetes can be used. Sehizomyeetes are particularly suitable, preferably certain strains of the species Streptomyces griseus, which can also be used to generate the antibiotics Streptomvein and Grisein.
Other species such as Streptomyces albidoflavus, Strept. eolombiensis nov. sp., Strept. roseochromogenus and Strept. Antibiotieus also contain strains that produce LLD-effective substances in high yield.
Further usable Sehizomveetes are members of the genus Clostridium <B> '</B> such as Clostridium tetanomorphum, <B> Cl. </B> mehlearium, <B> Cl. </B> flabelliferum and <B> Cl. < / B> butyricium. Other fungi that provide substances with LLD activity in the process according to the invention are torula,
Eremotheeium ashbyii and Escherichia coll. The nutrient medium is expediently sterilized and the sterile medium with the desired cobalt addition is then inoculated with a culture of the selected fungus, whereupon the broth is up to the optimum LLD effectiveness <B> 9 </ B > is developed. The fermentation time is advantageously around 2 to <B> 7 </B> days.
The development takes place below the surface of the medium and at a temperature suitable for the fungus in question. <B> 9 </B> The following procedure can be used to isolate crystallized vitamin B12 from the fermentation broth. The broth is filtered and the filtrate is treated with activated charcoal, where the vitamin B 1.2 is adsorbed. The adsorbate is then eluted with an aqueous solution of pyridine or of a-pieoline and the eluate is evaporated to dryness.
