R . CO. OR'.

Process for preparing a product with an ambrettolide odor. Swiss patent no. <B> 203133 </B> has described a process for the preparation of a macrocyclic lactone, according to which an ester of oxy-16-oxa-12-hexademnoic acid is heated in the presence of a functional group exchange catalyst ,. This process is charac terized in that we start with an ester whose alcohol in its composition distils, in the free state, above <B> 100 '</B> C under a vacuum of 2 mm of mercury, operating in such a way that the lactone formed is removed by entrainment in the form of vapor,
of the reaction medium as it is formed.
The method mentioned above was applied to the. preparation of various other macrocyclic lactones, as described in Additional Swiss Patents Nos. 207444, 207445, 207446 and 207447, subordinate to main patent no 203133.
In accordance with the processes claimed in these patents, the lactones obtained of the general formula
EMI0001.0013
were prepared starting from esters of oxy-aeides of general formula HO. R. CO. GOLD'. In these forms, R represents a chain, branched or not, of more than five carbon atoms, in some cases closing a bridge consisting of an oxygen atom.
In the starting ester formula, R 'always represents the radial of an alcohol (mono- or polyvalent) volatile above <B> 100' </B> C, under a vacuum of 2 mm of mercury, and perfectly stable at the temperature at which the transformation into macrocyclic lactone must be carried out.
The physical and chemical conditions, achieved in the processes described in the cited patents, and necessary for the transformation of said esters of oxy-acids into macrocyclic lactones, are such that it was not possible to predict that these processes can be employed without further ado for the conversion of esters of oxy-acids of the same general formula HO. R. CO.
OR 'as above, but in which R represents a chain, not only branched or not, of more than five carbon atoms, but containing at least one double bond connecting two neighboring carbon atoms.
Now, it has been found that, contrary to all expectations, such unsaturated esters can be transformed normally and without side reactions, due to the presence of a double bond in the chain of carbon atoms R, into macrocyclic lactones.
This is how the esters of formula HO. CHI. (CHI) r. CH = CH. (CHI) y. . CO. OR \, in which x + y = 12 and R 'meets the conditions set out above, prepared by shooting 5,15-dioxy-palmitic acid HO. CHI. <B> (CH-.) ,. </B> CH (OH). (CHI) 3C0. OH.
by converting this acid into a mixture of unsaturated mono-hydroxylated acids of general formula <B> s </B> HO. CHI. (CHI),. CH <B> -: </B> CH. (CHI) ,,. CO. OH, in which x + y = 12, then by esterification of these latter acids using known esterification methods, can be transformed into a product with an ambrettolide odor, consisting of a mixture of lac isomeric unsaturated macrocyclic tones, of general formula:
EMI0002.0023
where x + y = 12.
These lactones correspond to ambrettolide, the worshiping principle of. Ambrette seed, or its isomers. Indeed, as Kerschbaum has already demonstrated, see Ber. Deutsche Chem. Gesellsehaft, vol. 60, page 902 et seq.
(1927), ambrettolide has the following constitution formula:

<tb> 17.8 <SEP> gr <SEP> of <SEP> ce, <SEP> mixture <SEP> of esters <SEP> isomers
<tb> (0.05 <SEP> mol) <SEP> with <SEP> a <SEP> dissolution <SEP> of <SEP> 0.3 <SEP> gr <SEP> of
<tb> sodium <SEP> in <SEP> 30 <SEP> cm '<SEP> of <SEP> glycerin <SEP> and <SEP> on <SEP> distilled under a vacuum of 3 mm,
until the temperature reaches 200 ° C. The distillation is then continued at 3 mm by gradually adding glycerin to the reaction medium so as to maintain the temperature at about 200 ° C. The distillation is interrupted. 'operation after 10 hours. The distillate is taken up in ether, decanted, the ethereal solution is washed with water and the solvent is removed. The residue (10 to 11 g) is a colorless oil which is purified by distillation.
In this way, more than 8 g of a colorless liquid with a strong ambrettolide odor are obtained, consisting of a mixture of non-isomeric macrocyclic lactones of the general formula:
EMI0003.0012
where x '+ y = 12.
This liquid completely distils between 118 and 120 ° C under 0.25 mm of mercury. It is further characterized by its density at 20 ° C which is 0.960 and its refractive index nD: 1.480. Its saponification index is 220.4 (calculated: 222). Its chemical analysis provides the following results: C 76.14% <B> 11 </B> 11.22 calculated for C1.11.02 C 76.12% H <B> 1 </B> 1.19 The product thus obtained finds applications mainly in perfumery.
As can be seen, the process of the invention makes it possible to prepare isomers of ambrettolide, which have the same odor note as ambrettolide itself.
