Ce composé fond à l96-197 C. Il est peu soluble dans l'eau, il présente des propriétés bactéricides marquantes et est susceptible d'applications thérapeutiques.
Le procédé selon l'invention est caracté risé en ce que l'on fait réagir du 2-amino- thiazol avec un halogénure de benzène- sulfonyle substitué en para par un reste sus ceptible de donner par réduction le groupe -NH" de manière à former, par élimina tion d'halogénure d'hydrogène, un dérivé du 2-(benzène-sulfamido)-thiazol ayant ledit reste en position para, puis en ce que l'on effectue la réduction de manière à obtenir le 2-(para-amino-benzène-sulfamido)-thiazol. On peut, par exemple,
faire réagir avec le 2-amino-thiazol soit du chlorure de para- nitro-benzène-sulfonyle, soit du chlorure d'un azo-benzène-para-sulfonyle, par exemple du chlorure de para-diéthylamino-azo-benzène- para-sulfongle.
Les exemples suivants montrent comment le procédé selon l'invention peut être mis en pratique: <I>Exemple<B>1.-</B></I> 4,4 gr de chlorure de para-nitro-benzène- sulfonyle sont ajoutés à une solution de 2 gr de 2-amino-thiazol dans 10 cm3 de pyridine anhydre. Le mélange est chauffé à<B>100'</B> C pendant 10 minutes. On laisse reposer 1 h, puis on dilue avec 100 cm' d'eau. Le préci pité obtenu est filtré, puis redissous dans 50 cm' de solution de soude caustique<B>2N.</B> La solution est filtrée, le filtrat neutralisé avec de l'acide acétique.
Le 2,(para-nitro- benzène-sulfamido)-thiazol brut qui préci pite est filtré et redissous dans 10 cm' de solution de soude caustique 2 N chaude. Par refroidissement, le sel de soude qui se sépare sous forme cristalline est filtré, puis redis- sous dans l'eau. Cette solution est traitée par du noir décolorant et filtrée. Par addition d'acide acétique, le 2-(para-nitro-benzène- sulfamido)-thiazol pur précipite. Prismes jaunes fondant à 271 C.
1,5 gr de 2-(para-nitro-benzène-sulf- amido)-thiazol sont mélangés à 1 gr de poudre de fer et le mélange est ajouté peu à peu à 25 cm' d'acide acétique à 10ô bouillant. On maintient l'ébullition pendant encore 1 li, refroidit et filtre. Le résidu est extrait par de la soude caustique 2 N; la solution obte nue est filtrée et neutralisée par de l'acide acétique.
Le 2-(para-amino-benzène-sulf- amido)-thiazol brut précipite. Il est purifié par conversion en sel de sodium et reprécipi- tation. Prismes de P. F. 197 .
Exemple <I>2:</I> 7 gr de chlorure de 4-diéthylamino-azo- benzène-4'-sulfonyle sont ajoutés à une solu tion de 2 gr de 2-amino-thiazol dans 3(t cm:' de pyridine anhydre. Le mélange est alors chauffé quelque temps au bain-marie, puis versé en agitant dans 300 cm' d'eau. Le pré cipité formé est filtré, lavé et séché.
Le 4- diéthyla.mino - azo - benzène - 4'- sulfonyl - 2"- amino-thia.zol est ainsi obtenu sous forme d'une poudre rouge brillant de point de fu sion 196:-198 C.
1 gr de ce produit est dissous dans tin mélange de 111 cm' de soude caustique 2 N, 75 cm' d'eau et 50 cm' d'alcool méthylique. On ajoute 10 cm' d'une suspension alcoolique de 2,5 gr de nickel de Raney et traite le tout, par de l'hydrogène sous une pression de 3,5 kg/cm'. Quand la quantité calculée d'hy drogène a été absorbée, le catalyseur est filtré et on chasse l'alcool par distillation.
La so lution est extraite avec du chloroforme pour éliminer la p-diéthylamino-aniline, puis est neutralisée avec de l'acide acétique, ce qui précipite de 2-(p-amino-benzène-sulfaniido)- thiazol brut; celui-ci est dissous dans l'acide chlorhydrique dilué. La solution est traitée avec du noir décolorant, filtrée et addition née d'acétate de soude en excès.
Le 2-(para-amino-benzène -sulfamido)- thiazol pur cristallise en paillettes fondant à 197 C.
