Verfahren zur Darstellung von Dihydro-ergotamin. In seiner Untersuchung über die Alkaloide des Mutterkornes (Berichte der Deutschen Chemischen Gesellschaft 65, 557 [1982] gibt A. Soltys an, dass Reduktionsversuche der vier Mutterkornalkaloide Ergotamin, Ergot- aminin, Ergotogin und Ergotinin mit Palla dium als Katalysator negativ verlaufen seien. W. A. Jacobs, und L.
C. Craig (J. Biol. Chem. 108, 595 [1935] sowie weitere Publi kationen) unterwarfen einzelne Mutterkorn alkaloide der Einwirkung von Natrium in siedendem Butylalkohol. Dabei trat aber neben der Reduktion eine vollständige Auf spaltung des Moleküls in die einzelnen Be standteile ein.
Die gleichen Autoren berichten später über Versuche zur Reduktion von Ergotogin, Ergotinin, Ergotamin und Ergotaminin in Eisessiglösung mit Wasserstoff und Platin- oxydkatalysator (J. Biol. Chem. 115, 227 [1936]). Die Reduktionen verliefen aber nicht glatt. Die Lösungen färbten sich purpurblau und die Wasserstoffaufnahme kam nicht zum Stillstand.
Die Hydrierungen wurden willkürlich unterbrochen, wenn 1,6 bis 2 Mol. Wasserstoff verbraucht worden waren. Die Hydrierungsprodukte waren je doch amorph. In keinem Fall liess sich eine kristallisierte, definierte Hydroverbindung isolieren. Nach dem gleichen Verfahren haben Jacobs und Craig auch Ergometrin hydriert, wobei ihnen die Isolierung von kristallisiertem Dihydroergometrin gelang (J. Biol. Chem. 113, 767 [1936]), ferner (U. S. P.
Nr. 2086559 vom 13. Juli 1937). Ergometrin nimmt aber nicht nur chemisch unter .den Mutterkomalkaloiden eine Sonder stellung ein, indem es ein viel kleineres Molekul,argewicht besitzt als die übrigen. Alkaloide, sondern. es unterscheidet sich von den hochmolekularen Mutterkornalkaloiden auch dadurch, dass ihm keine sympathikoly- tische Wirksamkeit zukommt.
Es wurde nun gefunden, dass man Di- hydro-Derivate von sympäthikolytisch wirk samen natürlichen und synthetischen Mut terkornalkaloiden dadurch herstellen kann, dass. man diese Alkaloide bezw. deren Salze in Gegenwart eines geeigneten Katalysators und eines Lösungsmittels, gegebenenfalls bei erhöhter Temperatur, mit Wasserstoff unter Druck behandelt.
Als Katalysatoren kann man übliche Hy- drobenisierunbskataly satoren verwenden, wo bei es sich gezeigt hat, dass Palladium, wie z.
B. Palladiummohr, auf Bariumsulfat oder einen andern Träger niedergeschlagenes Pal ladium oder kolloidales Palladium, ferner M j ckel- und Kupfer -Niel-,c-Ilzatalysatoren zür Durchführung des vorliegenden Verfahren besonders geeignet sind.
Die von Soltys durchgeführten erfolg losen Hydrierunbsversuclie mit Palladium als Katalysator wurden unter gewöhnlichem Druck und Raumtemperatur vorgenommen. Demgegenüber arbeitet man nach dem vor liegenden Verfahren unter Druck und vor teilhafterweise auch bei erhöhter Tempera tur.
Wendet man dabei Wasserstoff unter Druck an, eventuell unter gleichzeitiger Steigerung der Temperatur, dann findet unter dem Einfluss der obenbenannten Kata lysatoren, wie sieh zeigte, selektiv Absätti- gunb nur einer Doppelbindung statt. Die Hydrierung bleibt bei der Dihydrostufe stehen und man gelangt zu kristallisierten, einheitlichen Verbindungen. Man arbeitet z. B. bei einem Wasserstoffdruck von ?0 atü und einer Temperatur von 50 C. Man kann aber auch mit dem Druck höher gehen, z. B.
80 atü und dafür mit der Temperatur bei 25 C 'bleiben oder bei 10 atü und 80 C arbeiten.
Die Durchführung der Hydrierung wird in Gegenwart eines Lösungsmittels oder eines Gemisches von Lösungsmitteln vorge nommen, wobei als solche Wasser, Alkohole, Äther, Kohlenwasserstoffe usw. verwendet werden können.
Für die Erlangung eines einheitlichen Reduktionsproduktes ist es zweckmässig, ein Lösungsmittel zu wählen, in dein sich die llutterkornalkaloide nicht oder nur sehr langsam, umlagern, in dem also z. B. Ergot- amin als solches erhalten bleibt und nicht teilweise in Ergotaminin übergeht.

pharmakologisch viel schwächer wirksam Esnd als die Alka loide der Lysergaäurereihe (Ergotamin, Ergot- oxin usw.). Die Dihydroalkaloide geben mit anorganischen oder organischen Säuren stabile, gut kristallisierte Salze, die je nach der Art des Säurerestes, in Wasser beträcht lich löslich sind.
Pharmakologisch unterscheiden sich die Dihydroverbindungen von den natürlichen Alkaloiden dadurch, dass die ersteren prak- c tisch keine Wirksamkeit mehr auf den Uterus besitzen, während die sympathikoly- tische Wirkung erhalten geblieben ist.
Das vorliegende Patent betrifft ein Ver fahren zur Darstellung von Dihydro-ergot- amin, welches. dadurch gekennzeichnet ist, dass man Ergotamin in einem Lösungsmittel mit Wasserstoff unter Druck behandelt unter Verwendung eines Katalysators, der ohne Anwendung von Wasserstoffüberdruck unter sonst gleichen: Bedingungen unwirksam ist.
Die neue Verbindung kristallisiert aus 90%igem Aceton, genau gleich wie Ergot- amin, in rechtwinklig begrenzten, prächtigen <B>)</B>Platten, die 2 Aceton und 2 H20 als Kristallösungsmittel enthalten und die bei 237 (korr.) unter Zersetzung schmelzen. [al D - -63 (c - 0,6 in Pyridin).
Dihydro-ergotamin gibt mit eisenchlorid- haltigem Eisessig und konzentrierter Schwe- fel-säure die gleiche prächtigblaue Farb- reaktion wie Ergotamin. Ebenso ist die Farb- reaktion nach Van Urk mit alkoholischer p Dimethylaminobenzaldehydlösung in konzen trierter Schwefelsäure unverändert geblie ben.
Die neue Verbindung soll therapeutische Verwendung finden. Beispiel: 10 g Ergotamin-Acetonverbindung, in 150 cm' Diogan gelöst, werden unter Zu satz von 5 g Palladiummohr in einem heiz- baren Schüttelautoklaven mit Glaseinsatz mit Wasserstoff von 20 atü vorerst 4 Stun- den bei 20 C geschüttelt. Dann steigert man die Temperatur auf 60 C, wobei der Druck auf ca. 26 atü steigt, und schüttelt 4 weitere Stunden bei dieser Temperatur.
Nach dem Erkalten wird wie üblich auf gearbeitet. Der grösste Teil des Dihydro- ergotamins ist auskristallisiert. Man nutscht die Kristalle mit samt dem Katalysator unter Nachwaschen mit etwas Dioxan ab. Die aus kristallisierte, praktisch reine Verbindung wird durch Aufnehmen mit einem Gemisch von 3 Teilen Chloroform und 1 Teil Alkohol, worin Dihydro-ergotamin leicht löslich ist, vom Katalysator abgetrennt. Der Rückstand der Chloroform-Alkohollösung, ungefähr 8 g, wird durch Aufläsen in der 10fachen Menge Aceton und Verdünnen mit 1 Teil. Wasser umkristallisiert.
Aus 90 % igem Aceton kristallisiert Dihydro-ergotamin in den glei chen, rechtwinklig begrenzten prächtigen Platten wie Ergotamin, die gleichfalls 2 Aceton und 2 H20 als Kristallösungsmittel enthalten. Der Rückstand der Dioxanmutter- lauge wird gleichfalls aus. Aceton-Wasser umkristallisiert.
Ausbeute an reinem Di- hydro-ergotamin-Acetonkristallisat insgeamt 8,5-9 g. Smp. 237 (korr.) unter Zer setzung.
Für die Elementaranalyse wurde die Verbindung im Hochvakuum bei 120' ge trocknet, wobei sie 20,0 % an Gewicht ver lor. Für den Austritt von 2 Mol. -f- 2 Mol. H20 auf 1 Mol. Dihydro-ergotamin-Aceton- Wasserkristallisat berechnet sich ein Ge wichtsverlust von 20,7%. Die Elementaranalyse ergab folgende s.5 Werte: C<B>67,79%"</B> H<B>6,76%,</B> N 11,86%. Für C33g3705N, berechnen sich C 67,89%, H 6,39%, N 12,01%.

Dihydro-ergotamin ist in Wasser sehr schwer, in Benzol und den Alkoholen mässe-, in Chloroform und Pyridin leicht löslich.
Dihydro-ergotamin gibt mit eisenchlorid- haltigem Eisessig und konzentrierter Schwe felsäure die gleiche prächtigblaue Farb reaktion wie Ergotamin. Ebenso ist die Farbreaktion nach Van Urk mit alkoholi- cher p-Dimethylaminobenzaldehy dlösung in konzentrierter Schwefelsäure unverändert ge blieben.
Einstündiges Kochen in methylalkolioli- scher Lösung verändert die Verbindung in keiner Weise, während Ergotamin durch diese Behandlung zum grössten Teil in Ergotaminin umgelagert wird.
Mit anorganischen und organischen Säu ren bildet Dihydro-ergotamin beständige, schön kristallisierende Salze, die je nach dem Säurerest in Wasser mehr oder weniger lös lich sind. Das neutrale Dihydro-ergotamin- tartrat kristallisiert aus 95, igem Methyl- alkohol in prächtigen, wasserklaren sechs- seitigen Platten, die bei<B>205'</B> (korr.)
unter Zersetzung schmelzen. Das Dilivdi-o-ergot- aminmethans-ulfonat, welches eine beträelit- liche Wasserlöslichlkcit besitzt, kristallisiert aus 95 % igem Äthanol in rechtwinklig abge schnittenen, bei<B>9233'</B> (korr.) unter Zer setzung schmelzenden Prismen.
Würde man die Hydrierung ohne Anwendung eines Wasserstoffüberdruckes durchführen, so würde man alles Ergotamin unverändert zurückerhalten.

Soltys states that attempts to reduce the four ergot alkaloids ergotamine, ergotamine, ergotogine and ergotine with palladium as a catalyst had negative results. WA Jacobs, and L.
C. Craig (J. Biol. Chem. 108, 595 [1935] and other publications) subjected individual ergot alkaloids to the action of sodium in boiling butyl alcohol. However, in addition to the reduction, the molecule was completely split into its individual components.
The same authors later report on attempts to reduce ergotogine, ergotine, ergotamine and ergotamine in glacial acetic acid solution with hydrogen and a platinum oxide catalyst (J. Biol. Chem. 115, 227 [1936]). However, the reductions did not go smoothly. The solutions turned purple blue and hydrogen uptake did not stop.
The hydrogenations were arbitrarily interrupted when 1.6 to 2 moles of hydrogen had been consumed. The hydrogenation products were, however, amorphous. In no case could a crystallized, defined hydro-compound be isolated. Jacobs and Craig also hydrogenated ergometrine using the same procedure, and succeeded in isolating crystallized dihydroergometrine (J. Biol. Chem. 113, 767 [1936]), furthermore (U. S. P.
No. 2086559 of July 13, 1937). Ergometrine not only occupies a special position chemically among the parent alkaloids in that it has a much smaller molecule than the others. Alkaloids, rather. it also differs from the high molecular ergot alkaloids in that it has no sympathetic activity.
It has now been found that dihydric derivatives of sympatholytically active seeds natural and synthetic mother grain alkaloids can be produced by using these alkaloids or. their salts are treated with hydrogen under pressure in the presence of a suitable catalyst and a solvent, if appropriate at elevated temperature.
As catalysts, you can use the usual Hy- drobenisierunbskataly catalysts, where it has been shown that palladium, such as.
B. palladium black, precipitated on barium sulfate or another carrier palladium or colloidal palladium, also Mjckel- and copper-Niel-, c-Ilzatalysts are particularly suitable for carrying out the present process.
The unsuccessful hydrogenation experiments carried out by Soltys with palladium as a catalyst were carried out under ordinary pressure and room temperature. In contrast, you work according to the present process under pressure and before geous enough at elevated tempera ture.
If hydrogen is used under pressure, possibly with a simultaneous increase in temperature, then under the influence of the abovementioned catalysts, as has been shown, only one double bond is selectively saturated. The hydrogenation is carried out in the presence of a solvent or a mixture of solvents, and as such water, alcohols, ethers, hydrocarbons, etc. can be used.
In order to obtain a uniform reduction product, it is advisable to choose a solvent in which the llutterkorn alkaloids do not rearrange, or only very slowly, in which z. B. Ergotamine is retained as such and is not partially converted into ergotamine.

It is much less pharmacologically active than the alkaloids of the lysergic acid series (ergotamine, ergotoxine, etc.). With inorganic or organic acids, the dihydroalkaloids give stable, well-crystallized salts which, depending on the type of acid residue, are considerably soluble in water.
Pharmacologically, the dihydro compounds differ from the natural alkaloids in that the former are practically no longer effective on the uterus, while the sympathetic effect is retained.
The present patent relates to a process for the preparation of dihydro-ergot amine, which. characterized in that ergotamine is treated in a solvent with hydrogen under pressure using a catalyst which, without the use of excess hydrogen pressure, is ineffective under otherwise identical conditions.
