Lösungsmittel für Hormone. Die Herstellung hochkonzentrierter Hor monlösungen für Injektionszwecke bietet in folge-der schlechten Löslichkeit der Sexual hormone und ihrer Derivate in Wasser grosse Schwierigkeiten. Man musste daher zur thera peutischen Anwendung dieser Hormone, wie z. B. der Follikelhormone, um die Einsprit zung von zu grossen Wassermengen zu ver meiden, Öllösungen verwenden. Es wurde jedoch gefunden, dass die Löslichkeit der Hormone und ihrer Derivate in den dazu vorgeschlagenen pflanzlichen Ölen nicht ge nügend gross war, um die in vielen Fällen notwendigen, hochkonzentrierten Lösungen zu erhalten.
So ist -es zum Beispiel nicht möglich, von der wirksamsten östrogenen Substanz, dem Ostradiolbenzoat Öllösungen herzustellen, welche 10 mg/cm' = 100 000 intern. Benzoat-Einh. enthalten.
Lösungen von Ostradiolbenzoat in Ara.- chis-, Mais-, Oliven- und Sesamöl oder Mi schungen dieser Öle kristallisieren bereits bei einem Gehalt von nur 5 mg pro cm' = 50000 intern. Benzoat-Einh. nach 48stündigem Ste hen bei 8 bis<B>10'.</B> Es ist deshalb nicht mög lich Ampullen herzustellen, die 50 000 in tern. Benzoat-Einh. in einem cm' dieser Öle enthalten, und bei tieferen Temperaturen be ständig und lagerfähig sind.
Es wurde nun gefunden, dass eine Mi schung von Lebertran mit pflanzlichen Ölen ein ausgezeichnetes Lösungsmittel, das die Herstellung höher konzentrierter Hormon lösungen gestattet, darstellt. Das neue Lö sungsmittel erlaubt zum Beispiel die Her stellung von Lösungen von Ostradialbenzoat, die 10 mg/cm' - 100 000 intern. Benzoat- Einh. enthalten und vollkommen beständig sind.
Diese Lösungen scheiden keine Kri stalle der östrogenen Substanz aus, selbst wenn sie lange bei einer nahe dem Gefrier punkt liegenden Temperatur aufbewahrt werden. Die Lösungen erwiesen sich bei kli nischen Versuchen als ausgezeichnet aktiv und wurden ohne jegliche Reizwirkung er tragen.
Ein aus einem pflanzlichen (1l, das 60 Lebertran enthält, hergestelltes Lösungsmit tel kann im Vergleich zu den pflanzlichen Ölen allein die zweifache Menge an männ lichen Hormonen, wie Androsteron, Testo- steron, Testosteronpropionat, an Corpus-lu- teum-Hormon, Progesteron und an Neben nierenhormon Corticosteron, in Lösung brin gen.
Die zur Lösung der Hormone erforder liche Lebertranmenge im Lösungsmittel hängt ab von der Art des zu lösenden Hor mons und der gewünschten Hormonkonzen tration. Diese Konzentration kann gemäss folgender Formel berechnet werden, wobei x die Anzahl cm' Lebertran bedeutet: K = Löslichkeit (im pflanzlichen 01) . (1-x) -f- Löslichkeit (in Lebertran)<B>-X.</B>
Die Löslichkeit des zu lösenden Hormons im Lebertran bezw. im pflanzlichen 01 kann leicht experimentell festgestellt. werden.
Beispiel <I>1:</I> Man stellt eine Ölmischung aus 40 Lebertran und 6()% Sesamöl her. In diesem Gemisch kann man reines trans-Üstradiol- Benzoat durch Erhitzen bis zur vollständigen Lösung des Hormons auflösen. Die Öllösung, die :5 mg Ostradiol-Benzoat im ein", enthält. kann in 1 cm'-Ampullen gefüllt und sterili siert werden.
Beispiel <I>2:</I> Man stellt ein 01gemisch aus<B>60%</B> reinem Lebertran und 40% Arachisöl her. In dieser Mischung kann man reines trans-Östradio- Benzoat erhitzen, bis das Hormon vollkom men gelöst ist. Man erhält eine Lösung, die <B>10</B> mg Hormon im cm' enthält. Sie kann in Ampullen von 1 ein' abgefüllt und sterili siert werden.
<I>Beispiel 3:</I> Man stellt ein Gemisch aus 6 cm' Leber tran und 4 cm' Sesamöl her. Erhitzt man Östradiol in diesem Lösungsmittel, so erhält man eine klare Lösung, die in 1. cm'-Ampul- len abgefüllt werden kann, wobei jede 20 mg des Hormons enthält, das heisst die doppelte Menge die in reinem Sesamöl löslich wäre.
