Abstract

La présente invention concerne une analyse par capture permettant de cribler simultanément les acides nucléiques de VHB, VHC et VIH dans des échantillons de plasma, par exemple. Les acides nucléiques contenant de l'ADN et de l'ARN viraux sont purifiés à partir des échantillons de plasma au cours d'une opération d'extraction unique. Dans un mode de réalisation, un mélange d'amorces PCR étiquetées par de la biotine dégénérée et spécifiques à VHB, VHC, VIH-1 de type M et VIH-1 de type O est utilisé pour amplifier n'importe lequel de ces virus susceptible de se trouver dans le plasma. Les produits amplifiés sont capturés par hybridation avec une séquence de capture immobilisée, puis détectés. Un vecteur de contrôle interne contenant un fragment synthétique flanqué de séquences correspondant aux amorces VHB permet de surveiller la récupération de l'échantillon au cours de l'extraction, de l'amplification et de la détection. Tous les sous-types principaux de VHB, VHC et VIH-1, y compris VIH-1 de type O, ont été confirmés et détectés au moyen de cette analyse.
