Matrizes: Fragmento proveniente de reação n° 1 (pb) e fragmento de baixo ponto de fusão proveniente da reação n° 2 (pb)
Produto: DNA porcino proveniente do nucieoiídeo 1742 no domínio A2 até o nucleotídeo 3021 no domínio A3 (SEQ ID N° 29) que inclui OL1212, 1279 pb. O produto da reação foi precipitado em etanol.
O linker 1212 foi inserido em PSQ/ReNeo por corte do produto de SOE (inserção) e PSQ/ReNeo (vetor) com BsaB I. O vetor e a inserção foram ligados usando-se T4 ligase e o produto foi usado para transformar células XL1-Blue de E. coli. Foi preparado DNA de plasmídeo partindo de diversas colônias e a seqüência do linker 1212 e outra seqüência gerada em PCR foi verificada por análise de seqüência de DNA.
- A) 48 pl (10 µg) de Miniprep POL1212/ReNeo DNA mais µl de meio sem soro (DMEM/F12) mais 10 µL de Lipofectin ™ (Life Technologies, Gaithersburg, MD).
- B) 10 pL de Lipofectin mais 190 µl de meio (imitação de transfecção) foram misturados cuidadosamente e o DNA e a Lipofectin deixados reagir durante 15 minutos à temperatura ambiente. Durante este período de tempo, as células foram lavadas duas vezes com 2 ml de DMEM/F12. 1,8 ml de DMEM/F12 foi então adicionado às células. O complexo de DNA/ Lipo-fectin foi adicionado gota a gota às células e girado cuidadosamente para efetuar a misturação. As células permaneceram na incubadora durante toda a noite. Removeu-se o DNA/ Lipofectin e adicionaram-se 3 ml de meio com soro às células. As células foram incubadas durante 30 - 48 horas. Geneti-cina foi adquirida de Life Technologies, Gaithersburg, MD). As culturas de célula foram divididas 1:20, 1:50 e 1:100, 1:250, 1:500 sobre placas em 10 ml de meio com soro contendo 535 •g/ml de geneticina. Durante os próximos vários dias, as células que não captaram o plasmídeo POL1212/ReNeo foram mortas em virtude da presença de geneticina. As células restantes continuaram a se replicar em geneticina, formando colônias em monocamada visíveis sobre as placas.

- Fator VIII de porcino modificado, caracterizado pelo fato de que apresenta a seqüência de aminoácido de SEQ ID N° 38.
- Composição terapêutica, caracterizada pelo fato de que compreende um fator VIII de porcino modificado, como definido na reivindicação 5, e um veículo fisiologicamente aceitável.
- Processo para produção de uma proteína fator VIII de porcino modificado apresentando a seqüência de aminoácido da SEQ ID N° 38, caracterizado pelo fato de que compreende expressar em uma célula hospedeira de mamífero um DNA que codifica a seqüência de aminoácido da SEQ ID N° 38.
- Processo, de acordo com a reivindicação 7, caracterizado pelo fato de que o DNA que codifica a seqüência de aminoácido da SEQ ID N° 38 também codifica um peptídeo de sinal, sendo que a proteína fator VIII de porcino modificado é exportada da célula hospedeira de mamífero.
