Abstract

NUCLEASES CAS9 TERMOSTÁVEIS. A presente invenção refere-se ao campo da engenharia genética e, mais particularmente, à edição de ácido nucleico e modificação do genoma. A presente invenção proporciona uma proteína Cas isolada ou fragmento de polipeptídeo da mesma tendo uma sequência de aminoácidos de SEQ ID NO: 1 ou uma sequência de pelo menos 77% de identidade com a mesma, em que a proteína Cas ou o polipeptídeo é capaz de clivagem de DNA a uma temperatura na variam 50(graus celsius) e 100(graus celsius) inclusive. A invenção proporciona ainda moléculas de ácido nucleico isoladas que codificam as referidas nucleases Cas9, vetores de expressão e células hospedeiras. As nucleases Cas9 aqui descritas fornecem novas ferramentas para engenharia genética a temperaturas elevadas e têm um valor particular na manipulação genética de organismos termofílicos; particularmente microrganismos.
