Abstract

"MéTODO PARA IDENTIFICAR INIBIDORES DE MIELOPEROXIDASE, E, USO DE UM MéTODO". Um método de ensaio para a detecção de inibidores da enzima mieloperoxidase, o citado método compreendendo as etapas de: reagir mieloperoxidase com peróxido de hidrogênio e uma fonte de cloreto para gerar ácido hipocloroso na presença de um inibidor potencial da citada mieloperoxidase; reagir qualquer ácido hipocloroso formado com uma amina adequada para formar a cloramina correspondente; opcionalmente remover qualquer peróxido de hidrogênio não reagido; reagir qualquer cloramina com uma molécula detectora apropriada na presença de iodeto para formar molécula detectora oxidada; determinar a quantidade de molécula detectora oxidada formada pela medição da absorbância ou da fluorescência em um comprimento de onda adequado; identificar como inibidores de mieloperoxidase aqueles compostos que causam a diminuição da quantidade de molécula detectora oxidada formada. E é descrito o uso em testes de diagnósticos para a atividade de mieloperoxidase.
