Abstract

"POLINUCLEOTìDEOS CODIFICADORES DE GENES DO CROMOSSOMO DA BACTéRIA Chromobacterium violaceum, EXPRESSãO E ATIVIDADE DESSES POLINUCLEOTìDEOS E SUAS APLICAçõES". Chromobacterium violaceum desenvolveu a capacidade de adaptação a alterações ambientais bruscas, incluindo a disponibilidade de nutrientes, mudanças na temperatura, presença de elementos tóxicos, entre outras. Diversos genes desse organismos foram identificados e todos os mecanismos de transcrição, como as polimerases do RNA e os fatores sigma foram anotados, tais como <sym>^ 70^ (rpoD), <sym>^ 54^ (rpoN), <sym>^ 32^ (rpoH), <sym>^ 38^ (rpoS), <sym>^ 28^ (fliA), <sym>^ 24^ (rpoE), anti-<sym>^ 28^ fator (flgM), relacionado a <sym>^ 54^ (yhbH) e Ecf-<sym>^ 70^. Além disso, diversos ativadores transcricionais e repressores que interagem com os fatores sigma e envolvidos na resposta das bactérias a estresses ambientais também estão presentes, com diversos genes relacionados às famílias LysR, AraC, TetR, GntR, Mar, Mer, AsnC, DeoR, AsrR, Lacl, Crp, entre outros. Ainda mais expressiva foi a detecção de um número elevado de proteínas regulatórias associadas à adaptação a diversos ambientes, representando quase 9% dos genes de Chromobacterium violaceum. A versatilidade de Chromobacterium violaceum também fica evidenciada nos mais de 300 genes identificados e relacionados ao metabolismo energético, o que permite a obtenção de energia em condições aeróbicas, microaeróbidas e anaeróbicas. Foram detectados, também, genes relacionados à absorção de determinados nutrientes em mecanismos conhecidos por conferir, às bactérias, vantagens na competição contra outros microrganimos e, ainda, diversos genes capazes de promover a sobrevivência em condições de limitação de nutrientes, como genes envolvidos na utilização de peptídeos sob estresse de carbono, as proteínas SspA e SspB, induzidas por deficiência de glicose, nitrogênio, fosfato e aminoácidos , as proteínas de estresse "universal stress proteins" (Usp), que normalmente agem sob estresses de temperaturas elevadas, salinidade e exposição aos raios ultra-violeta, entre outras. Several genes of these organisms were identified and all transcriptional mechanisms such as RNA polymerases and sigma factors were noted, such as <sym> ^ 70 ^ (rpoD), <sym> ^ 54 ^ (rpoN), <sym> ^ 32 ^ (rpoH), <sym> ^ 38 ^ (rpoS), <sym> ^ 28 ^ (fliA), <sym> ^ 24 ^ (rpoE), anti-<sym> ^ 28 ^ factor (flgM), related to <sym> ^ 54 ^ (yhbH) and Ecf- <sym> ^ 70 ^. In addition, several transcriptional activators and repressors that interact with sigma factors involved in the response of bacteria to environmental stresses are also present, with several genes related to the LysR, AraC, TetR, GntR, Mar, Mer, AsnC, DeoR, AsrR families. , Lacl, Crp, among others. Even more significant was the detection of a large number of regulatory proteins associated with adaptation to various environments, representing almost 9% of the Chromobacterium violaceum genes. The versatility of Chromobacterium violaceum is also evident in the more than 300 genes identified and related to energy metabolism, allowing energy to be obtained under aerobic, microaerobic and anaerobic conditions.
