Certaines 2, 5-pipérazinediones ont déjà été décrites dans la littérature concernée ("J. Chem. Soc."

Commun."35, 619, 1970). présentent un certain intérêt pharmacologique qui est basé sur les propriétés antitumeurs ou antivirales de certains composés apparentés (certificat d'auteur tchécoslovaque n 210. 383) et elles sont également intervenues dans des études théoriques sur la conformation des molécules.

Les dérivés de 2, 5-pipérazinediones de formule générale I exercent des effets biologiques remarquables et surprenants, en particulier, sur la mémoire d'animaux expérimentaux. Comme on le sait dans la technique, des substances ayant des propriétés semblables sont évaluées pharmacologiquement dans l'essai d'extinction de la réaction d'échappement conditionné, ainsi que dans l'essai de durée de la réponse d'évitement passif chez les rats (R. Walter et al.,"Proc. Natl. Acad.
Sci."E. U. A. 1. i, 2493, 1975 ; ibidem 72, 4180, 1 975 ; J. Krejcí et al.,"Neuropeptides and neural transmission", A. Ajmone Marsal et W. Z. Traczyk (éd. ) Raven Press, New York 1980). Au cours de ces essais, on a observé une activité maximum dans le facteur d'inhibition de l'hormone mélano-stimulante (mélanostatine).
Toutefois, cette substance doit être administrée par voie parentérale (de préférence, par voie sous-cutanée) car, lors d'une application par voie orale, elle subit une inactivation enzymatique rapide dans les voies gastro-intestinales. De façon inattendue, on a trouvé que, par exemple, le cyclo (L-alanyl-l-amino-l-cyclopen- tane-carbonyle), qui est un composé spécifique de la présente invention, était hautement actif dans l'essai d'évitement passif ; la substance a été administrée en une dose de 1 mg/kg par voie sous-cutanée, soit immédiatement après l'entraînement, soit 30 minutes avant l'essai de rétention.
Or, il est des plus surprenants de constater que ce composé possède une activité semblable même lors de l'administration de doses du même ordre de grandeur par voie orale et qu'en outre, son effet (à un dosage de 0, 5 à 10 mg/kg par voie orale) se prolonge au-delà d'une période aussi longue que 20 heures avant l'entraînement ou l'essai.
Dans d'autres essais pharmacologiques, il est remarquable de constater que ce composé inhibe le

développement de la tolérance à l'effet cataleptique après administration répétée de neuroleptiques. Dans le modèle expérimental classique qui est utilisé pour établir une approximation du développement de la dyskinésie tardive, lorsque ce composé a été administré deux fois par jour en une dose de 7, 5 mg/kg par voie orale, il a inhibé sensiblement la tolérance après l'application de l'isofloxythépine, qui est un médicament neuroleptique à action prolongée dont on a administré des dosages répétés pendant 6 jours.
Dans une expérience semblable, à une dose de 10 mg/kg par voie orale en un jour, le même composé de l'invention a inhibé sensiblement la diminution de la teneur en acide homovanillique dans le corps strié de rats, ainsi que l'accroissement du décompte maximum de sièges de fixation de la dopamine (en utilisant la

o 3H-spipérone). Lorsqu'elle a été administrée par voie orale, la substance a également empêché le développement d'une sursensibilité des récepteurs dopaminergiques dans le corps strié de rats. En conséquence, on peut s'attendre à ce que le composé soumis aux essais exerce un effet anti-dyskinétique. Le facteur inhibant l'hormone mélano-stimulante (mélanostatine) a été actif dans des conditions expérimentales identiques uniquement après administration par voie sous-cutanée.
D'après les remarques qui précèdent, il découle que l'avantage principal des dérivés de 2, 5pipérazinediones de l'invention réside dans le fait qu'à un niveau d'activité égal ou supérieur à celui de la mélanostatine, leur action est sensiblement prolongée même lors d'une administration par voie orale. En conséquence, on peut escompter que les composés de l'invention trouveront une application dans le traitement des troubles de la mémoire, de la dyskinésie tardive et de la maladie de Parkinson.

muler sous des formes de dosage pharmaceutiques habituelles et appropriées pour une administration par voie orale ou parentérale, par exemple, des comprimés, des comprimés enrobés, des gélules ou des solutions pour injection en vue d'une application par voie souscutanée, intramusculaire ou intraveineuse.
Les formes de dosage pour administration par voie orale peuvent être obtenues selon la pratique courante en formulant l'ingrédient actif avec des adjuvants habituels de granulation et/ou de formation de comprimés, par exemple, le lactose, le saccharose, l'amidon, la polyvinylpyrrolidone, la stéarine, le stéarate de calcium, l'ultra-amylopectine, la cellulose microcristalline et analogues. La poudre de granulation ou de formation de comprimés ainsi obtenue est ensuite introduite dans des gélules ou pressée en comprimés de la dimension et du poids désirés ayant la teneur requise en ingrédient actif. De même, les formes de dosage et les solutions pour administration par voie parenté- rale peuvent être obtenues selon les procédés classiques pour la formulation des médicaments.
On peut avantageusement préparer les dérivés de 2, S-pipérazinediones de la présente invention par un procédé consistant à faire réagir un composé de formule générale II :

Les exemples suivants illustrent des détails particuliers complémentaires des procédés avantageux décrits ci-dessus pour la préparation des composés de l'invention, ainsi que pour leur formulation sous des formes de dosage pharmaceutiques. On a vérifié l'identité et la pureté des produits notamment par analyse

élémentaire. Les valeurs trouvées correspondent aux valeurs calculées dans des limites de tolérance étroites.

Exemple 1 Cyclo (L-alanyl-l-amino-1-cyclohexane-carbonyle).
On traite une solution de 11, g (50 millimoles) de benzyloxycarbonyl-L-alanine dans 100 ml de diméthylformamide contenant 7 ml de N-éthyl-pipéridine refroidie à avec 5 ml de chloroformiate d'éthyle. Après agitation pendant 20 minutes à on ajoute une solution de 1-amino-1-cyclohexane-carboxylate d'éthyle dans du diméthylformamide (100 ml) et on la libère de son chlorhydrate (10, g, 50 millimoles) par addition de 7 ml de N-éthylpipéridine.
Après agitation pendant une heure à OOC et pendant 2 heures à la température ambiante, on évapore le mélange réactionnel sous pression réduite, on dissout le résidu dans un mélange d'acétate d'éthyle et d'eau, on agite la phase organique successivement avec de l'acide chlorhydrique 1M, de l'eau, de l'hydrogénocarbonate de sodium à 5% et à nouveau de l'eau, on la sèche sur du sulfate de sodium anhydre et on l'évapore pour obtenir un résidu non cristallin.
On achève son séchage par distillation azéotrope avec un mélange de tétrahydrofuranne et de benzène ; ensuite, on traite le résidu avec 50 ml d'une solution de bromure d'hydrogène à 35% dans de l'acide acétique glacial et, après avoir laissé reposer pendant une heure à la température ambiante, on précipite le bromhydrate du L-alanyl-1-amino-1-cyclohexane-carboxylate d'éthyle non cristallin avec 500 ml d'éther de pétrole. Rf = 0, 52/S1, 0, 68/S2. S. : n-butanol-acide acétique-eau 4 : 1 : 1, 82 : n-butanol-acide acétique-pyridine-eau 15 : 3 : 10 : 6.
On met le bromhydrate obtenu en suspension dans 50 ml d'une solution ammoniacale de chloroforme

saturée à OOC et, après avoir laissé reposer pendant 20 minutes à SOC, par filtration, on sépare le bromure d'ammonium précipité, on évapore la solution de chloroforme et on la sèche par voie azéotrope avec un mélange de tétrahydrofuranne et de benzène. On dissout le résidu dans 100 ml de toluène, on ajoute 0, 2 ml d'acide acétique glacial et on chauffe la solution à reflux pendant une heure. Après refroidissement de la solution à la température ambiante et après l'avoir laissé reposer pendant 2 heures à OOC, on sépare le produit cristallin et on le lave avec de l'éther de pétrole.
On obtient le produit sous rubrique avec un rendement de 38%. On cristallise un échantillon dans un mélange de diméthylformamide et de 2-propanol et il a alors un point de fusion de 319-3200C.
De la même manière, on prépare les composés suivants : le N-benzyloxycarbonyl-L-séryl-l-amino-l-cyclopentane- carboxylate de méthyle à partir de l'acide N-benzyloxycarbonyl-l-amino-l-cyclopentane-carboxylique et de

Exemple 2 Cyclo (L-alanyl-1-amino-1-cyclopentane-carbonyle).
On traite une solution (refroidie à-10 C) de 2,25 g (10 millimoles) de benzyloxycarbonyl-Lalanine dans 20 ml de chloroforme avec une solution de 1-amino-l-cyclopentane-carboxylate de méthyle dans du chloroforme (15 ml), qu'on libère de son chlorhydrate (1, 8 g, 10 millimoles) au moyen de 15 ml d'une solution ammoniacale de chloroforme et on ajoute 2, 3 g de N, N'dicyclohexylcarbodiimide.
Après agitation pendant une heure à OOC et après avoir laissé reposer pendant 12 heures à 3 C, par filtration, on sépare la N, N'-dicyclohexylurée précipitée, on évapore le filtrat sous pression réduite, on dissout le résidu dans de l'acétate d'éthyle et on agite la phase organique successivement avec de l'acide chlorhydrique 1M, de l'eau, de l'hydrogénocarbonate de sodium à 5% et à nouveau de l'eau, on la sèche sur du sulfate de sodium anhydre et

The derivatives of 2,5-piperazinediones of general formula I exert remarkable and surprising biological effects, in particular, on the memory of experimental animals. As is known in the art, substances with similar properties are evaluated pharmacologically in the extinction test for the conditioned exhaust reaction, as well as in the duration of the passive avoidance response test in rats (R. Walter et al., "Proc. Natl. Acad.
Sci. "EUA 1. i, 2493, 1975; ibidem 72, 4180, 1 975; J. Krejcí et al.," Neuropeptides and neural transmission ", A. Ajmone Marsal and WZ Traczyk (ed.) Raven Press, New York 1980) During these trials, maximum activity was observed in the inhibitory factor of the melano-stimulating hormone (melanostatin).
However, this substance should be administered parenterally (preferably subcutaneously) because, when applied orally, it undergoes rapid enzymatic inactivation in the gastrointestinal tract. Unexpectedly, it has been found that, for example, cyclo (L-alanyl-1-amino-1-cyclopentane-carbonyl), which is a specific compound of the present invention, was highly active in the test for 'passive avoidance; the substance was administered in a dose of 1 mg / kg subcutaneously either immediately after training or 30 minutes before the retention test.
However, it is most surprising to note that this compound has a similar activity even when administering doses of the same order of magnitude by the oral route and that, in addition, its effect (at a dosage of 0.5 to 10 mg / kg orally) extends beyond as long as 20 hours before training or testing.
In other pharmacological tests, it is remarkable to note that this compound inhibits the

development of tolerance to the cataleptic effect after repeated administration of neuroleptics. In the classical experimental model which is used to approximate the development of tardive dyskinesia, when this compound was administered twice a day in a dose of 7.5 mg / kg orally, it significantly inhibited tolerance after the application of isofloxythepine, which is a long-acting neuroleptic drug for which repeated dosages have been administered for 6 days.
In a similar experiment, at a dose of 10 mg / kg orally in one day, the same compound of the invention substantially inhibited the decrease in the content of homovanillic acid in the striated body of rats, as well as the increase of the maximum dopamine fixation seat count (using the

o 3H-spiperone). When administered orally, the substance also prevented the development of dopaminergic receptor oversensitivity in the striated body of rats. As a result, it can be expected that the test compound will exert an anti-dyskinetic effect. The factor inhibiting the melano-stimulating hormone (melanostatin) was active under identical experimental conditions only after administration by the subcutaneous route.
From the above remarks, it follows that the main advantage of the 2,5piperazine derivatives of the invention lies in the fact that at an activity level equal to or greater than that of melanostatin, their action is substantially prolonged even when administered orally. Consequently, it can be expected that the compounds of the invention will find application in the treatment of memory disorders, tardive dyskinesia and Parkinson's disease.

mulate in standard pharmaceutical dosage forms suitable for oral or parenteral administration, for example, tablets, coated tablets, capsules or solutions for injection for subcutaneous, intramuscular or intravenous application.
Dosage forms for oral administration can be obtained according to current practice by formulating the active ingredient with usual granulation and / or tablet-forming adjuvants, for example, lactose, sucrose, starch, polyvinylpyrrolidone, stearin, calcium stearate, ultra-amylopectin, microcrystalline cellulose and the like. The granulation or tablet-forming powder thus obtained is then introduced into capsules or pressed into tablets of the desired size and weight having the required content of active ingredient. Likewise, dosage forms and solutions for parenteral administration can be obtained by conventional methods for drug formulation.
